Description
(I)細胞表面でCD94/NKG2レセプターを発現する細胞を提供し、
(II)試験すべき化合物の存在下にその細胞をHLA−Eと接触させ、そして
(III)その化合物の存在がHLA−Eと細胞の結合に影響を与えるかどうかを決定する
ことを含み、その方法で同定された化合物が、HLA−EとCD94/NKG2レセプターの結合に影響する化合物であるとするものである。
NK細胞は抗腫瘍細胞活性をもつと信じられている。治療の進行のためのマーカーまたは単純な予後は、NK細胞数および任意的にはそれらの活性状態をモニターする事によって提供することができる。これは極度に簡単な試験を提供する。ここに記載する方法は、患者から採取した検体の中のNK細胞の数を決定または見積もるために用いることができる。NK様の活性(CD94/NKG2+)でのNK細胞とT細胞の数は、NK細胞(たとえばCD56もしくはCD16)のマーカーに対する抗体またはT細胞マーカー(たとえばCD3)に対する抗体の使用によって概算することができる。NK細胞の活性の状態は、活性化マーカーへの抗体による共染色で調べることができる。あるいは、NK細胞の活性化状態は、インビトロでの機能的アッセイで評価することができる。NK細胞を検体から単離し、その細胞傷害活性及び/又はサイトカイン生成能力(たとえばインターフェロン−γ及び/又はTNF−α)を直接または培養中の短時間後で評価する。
NK細胞数は、ウイルスまたはそれ以外で感染中は変化し得る。そしてそれらの数を知ることはたとえばHIVに感染した患者において大きな価値がある。サイトメガロウイルス(CMV)感染におけるNK細胞数のモニターは特に重要であろう。CMVはその蛋白の中に、HLA−E発現に影響を与え得る配列をもっている。さらに詳しくは、これらのCMV配列は、ウイルス宿主細胞中のHLA−Eの細胞表面発現を誘発する。
胎児の拒絶反応の予防における胎盤中のNK細胞の役割は重要である。本発明は、胎盤中のNK細胞をモニターする手段を提供する。
NK細胞は、骨髄移植後の移植拒絶反応および移植片対宿主病(GVHD)に関与しているであろう。NK細胞をモニターすることは、患者の管理において価値があろう。
HLA−Eの診断的使用は、正常なNK細胞レベルよりも低いかまたは高い、本態性または後天的な新しい免疫不全症症候群の検出まで広げることができる。若干の治療はNK細胞に毒性的であるか促進的であろう。
たとえば全身的エリテマトーデス、糖尿病、甲状腺疾患、白斑、関節リュウマチ、などの自己免疫疾患におけるNK細胞のモニターは有用であろう。
本発明はまた、CD94/NKG2+NK細胞のアップまたはダウンレギュレーションをもたらし得る治療の二次的効果のモニターをも可能とする。
本発明は、HLA−E結合NK細胞またはT細胞を混合細胞集団から選択する方法を提供する。選択された細胞はインビトロで拡大されそして患者に戻される。そのような処置は、これらの細胞の生長不全が予後不良と関連しているような幾つかの重篤な感染やガンにおいて効果的であろう。
本発明は、たとえば移植においてドナー骨髄として用いられるべき骨髄からのHLA−E結合NK細胞とT細胞の除去のための方法や手段を提供する。他の技術を用いる骨髄のT細胞除去は既知であり効果的である。毒素とカップリングしたHLA−EもまたインビボでHLA−E結合細胞を破壊するために用いることができる。
本発明は、細胞表面へHLA−Eを提供することによるNK細胞活性の阻害または刺激のための方法や手段を提供する。NK細胞は移植の拒絶反応において役割を果たしている。したがってHLA−Eが異種移植片臓器や組織での細胞表面で発現することを確かめることによって、拒絶反応の可能性が減少されるであろうし、また拒絶反応も避けることができるであろう。
− ブタ胎児のニューロン組織の移植によるパーキンソン病の治療、
− カプセル化したブタの膵臓島(pancreatic islet)の移植または浸透による糖尿病の治療、
− ブタの全肝臓を通しての浸透または移植による肝不全の治療

HLA−E多量体複合体の構成
HLA−Eテトラマー複合体を、インビトロで組換えHLA−Eとβ2m分子をHLA−B*0801のシグナル配列の3−11残基から誘導した合成ペプチド(VMAPRTVLL)で再生することで構成した。ビオチン化部位をHLA−E重鎖のC末端でつくり、HLA−E/β2m/ペプチド複合体を大腸菌BirA酵素を用いて酵素的にビオチン化させ、そしてフィコエリスリン(PE)−標識エクストラビジンと複合させてテトラマー複合体を作った。HLA−Aと−Bテトラマー複合体は、インビトロで末梢血液からの抗原特異的なCD8+T細胞上のT細胞レセプターと特異的に結合するのに大変効率的であることが証明された(Altman et al,1996,Science,274:94−96)。
HLA−Eは、HLA−E*0101ホモ接合体個体の単核細胞から抽出したプライマーのCOO7およびCOO6でRT−PCRによってクローン化した。N末端ヌクレオチド配列は、プライマーCO17およびCOO6を用いるPCR変異誘発によって同義語的に変化して、大腸菌のpGMT7ベクターからの蛋白発現を最適化する。HLA−Eの細胞外部分(1−276残基)をコードする配列は、プライマーCO17とCO23を用いて増幅され、そしてpGMT7誘導体の中へ再クローン化されて、HLA−E重鎖の3’末端のフレーム中のBirA認識とビオチン化部位を含むところのプラスミドCOCO92の発現をもたらす。プライマーは次のようである。

HLA−Eテトラマーの結合
9種の正常のドナーからの末梢血液単核細胞(PBMC)を、例1に記載のようにして製造したHLA−Eテトラマーで染色し、エプスタインバーウイルス(EBV)溶菌サイクルBMLF1 259−267ペプチドエピトープ(Steven et al,1997,J.Exp.ed.185:1605−17)で再生したHLA−A2テトラマーで観察される染色と比較した。高い頻度のリンパ系細胞をHLA−Eテトラマーで染色し(2から11%の範囲)(図1A)、一方ではHLA−A2テトラマーは一般にEBV−血清反応陽性のドナー(図1C)中で0から0.8%のリンパ球で染色する。リンパ球結合HLA−Eテトラマー上で電子的ゲートを設定することにより、大部分はNK細胞(典型的には40から80%のCD3−、CD56+)であるが、著しいサブセットはT細胞であり(典型的には15から50%のCD3+)であり、そのうちの若干はまたCD56を発現する(図1B)ことを観察した。リンパ球結合HLA−Eテトラマーの約2%はCD4+T細胞であり、そして約5%はCD19+B細胞であったが、これらのものはHLA−A2テトラマーによる類似の染色の故に非特異的結合を表わす(データは示してない)。HLA−A2テトラマーはCD56+細胞と結合しないが、EBV−血清反応陽性ドナーにおいては、EBV特異的なCD3+、CD8+T細胞と結合し(図1D)、特定のT細胞レセプターをもつT細胞のためのHMC−テトラマー複合体の特異性についてのこれまでの研究を確認するものである(Altman et al,1996)。

NK細胞クローンに対する保護とHLA−E発現を可能とするHLA−E結合リーダー配列での細胞のトランスフェクション
ヒトのNK細胞クローンを確立し、文献記載のようにして培養した(Litwin et al,1993,J.Exp.Med.,178:1321−1336)。細胞毒性試験を文献記載のようにして行なった(Phillips et al,1996,Immunity,5:163−172)。HLA−GのリーダーセグメントならびにHLA−B*5801の細胞外、トランスメンブラン、および細胞質領域キメラのcDNAを含有するキメラのcDNAを、次のオリゴヌクレオチドプライマーを用いPCRによって作製した。すなわちセンスプライマー1,5’−GCGTCTAGAATGGTGGTCATGGCACCCCGA−3’[SEQ ID NO:16];アンチセンスプライマー1,5’−CATGGAGTGGGAGCCGGCCCAGGTCTCGGT−3’[SEQ ID NO:17];センスプライマー2,5’−GGCTCCCACTCCATGAGGTAT−3’[SEQ ID NO:18];およびアンチセンスプライマー2,5’−AAGCTTTCAAGCTGTGAGAGACA−3’[SEQ ID NO:19]。
HLA−Eの表面発現を誘導するHLA−E結合リーダーペプチドの存在がCD94/NKGT2A+NK細胞クローンに対する保護を提供するのに充分かどうかを決定するために、キメラ相補的DNA(GLS−B*5801)を作製した。それは、HLA−Gのリーダーセグメント(それからペプチドがHLA−Eを結合可能である;表2参照)ならびにHLA−B*5801の細胞外、トランスメンブランおよび細胞質領域(CD94/NKG2Aレセプターでの認識に関連しないHLA分子;Phillips et al,1996,Immunity,5:163−172)を含有している。安定な721.221細胞系トランスフェクタントを選択し、CD94/NKG2Aレセプターを発現するNK細胞クローンによる溶菌に対しての受動性について分析した。図4aに示すように、CD94/NKG2Aレセプターを発現するNK細胞クローンは、野生型HLA−B*5801でトランスフェクトした721.221細胞と同じように、トランスフェクトされてない721.221細胞を効果的に死滅させた。しかしながらNK細胞で仲介された溶菌に対する保護はキメラGLS−B*5801分子の発現で付与されたが、CD94またはHLAクラスI分子に対する抗体の存在下では逆となる。HLA−B*0702(ただしHLA−Eと結合可能なペプチドと共に;表2)のリーダーセグメントならびにマウスCD80の細胞外、トランスメンブランおよび細胞質領域を含有するキメラcDNA(B7LS−mCD80)を、721.221細胞の中へトランスフェクトしCD94/NKG2A+NK細胞クローンでの溶菌について試験した。CD80は、活性化したBおよびT細胞ならびにマクロファージで発現する接着細胞表面分子である。この実験でCD80は、MHCクラスI分子のリーダー配列のみがHLA−Eのアップレギュレートと保護効果の誘発に必要なことを示すために、無関係の対照分子として用いられる。B7LS−mCD80分子を発現する721.211細胞の溶菌は少なかったが野生型のCD80を発現する細胞ではそうではなく、そして保護は対照抗体ではなくて抗CD94によって逆となった(図4b)。これらの結果は、HLA−E結合リーダーペプチド単独が、阻害的CD94/NKG2Aタイプのレセプターを発現するNK細胞クローンによる溶菌から721.221細胞を保護するのに充分である。
蛍光活性化細胞ソーティングによるCD94/NKG2+の単離
末梢血液単核細胞(PBMC)を、ドナーからヘパリン含有のチューブ内へ取出した静脈血から得た。簡単に言えば血液検体を、血清を含まないRPMI−1640で1:1に希釈し、そして希釈血液の10mlを5mlのフィコール−ハイパック勾配においた。1200rpmで30分間遠心分離後、界面のPBMCを注意深く除去し、RPMIで2回洗った。最初の遠心分離は2000rpmで10分間行い、次に1200rpmで10分間行なって殆どの血小板を除去した。次いでPBMCをRPMI内で希釈し、処理中は無菌培地に保持した。
HLA−Eで被覆したビーズを用いるCD94/NKG2+細胞の単離
CD94/NKG2レセプターを発現する細胞を、HLA−E−ストレプタビジン被覆のダイナビーズ(dynabeads)で単離した。ダイナビーズM−280ストレプタビジンは、ストレプタビジンで被覆された磁気ビーズである。可溶性のHLA−Eを、HLA−E重鎖のC末端でBirA酵素のビオチン化部位で処理しβ2マイクログロブリン、および若干のHLA分子のシグナル配列の3−11残基からの合成ペプチドで再生した(Braud et al,1998,Nature,391:795に記載されている)。これらのHLA−Eモノマーは、BirA酵素を使ってビオチン化され、そして標準のプロトコールを用いてダイナビーズM−280と複合させた。ビオチン化したHLA−Eは、二方向性の混合(2μgのHLA−E/107ダイナビーズ)で30分間4℃でPBS−洗浄ダイナビーズM280で培養した。これらのビーズを、ダイナル(Dynal)磁気粒子濃縮機(MPC)内のチューブへ入れて採取し、上澄み液を除去した。ビーズを同様の方法で5回洗浄した。次いでHLA−E被覆M−280ダイナビーズを、単離したPBMC(例3記載のようにして得たもの)(107ビーズ/ml)と混合し、緩やかに攪拌しながら4℃で20分間培養した。チューブをダイナルMPCに入れ、2分間放置した。上澄み液を除去し、ビーズに付着した細胞を5回洗浄した。次いで細胞を、例3に記載のようにして生長させた。
NK細胞の標的死滅
組換えビオチン化HLA−Eを、例1に記載のようにして作製した。ビオチン化したHLA−Eと、酵素パーフォリンのようなビオチン化毒性剤との3:1のモル比の混合物をエクストラビジンと結合して、毒性剤と結合した多量体HLA−Eを製造した。ヒトのドナーからのPBMC検体を標準的技術によって製造しHLA−E試薬と接触させたところ、検体に存在するCD94/NKG2+細胞の死滅をもたらした。CD94/NKG2陰性細胞は回収した。
異種移植
HLA−Eのリーダー配列をHLA−B8のリーダー配列で置き換えたHLA−Eを発現する組換えDNAを作製した。このキメラcDNAは、HLA−Bのリーダー配列ならびにHLA−Eの細胞外(α1、α2とα3)、トランスメンブランおよび細胞質領域を含んでいる。これは、次のオリゴヌクレオチドプライマーを使って製造した。

哺乳動物細胞へのCD94およびNKG2の安定なトランスフェクション
マウスの細胞(P815、L細胞)を、CD94 cDNAおよびNKG2AまたはNKG2CとDAP12を含有する哺乳動物発現ベクターpcDNA3(ネオマイシン耐性遺伝子)で、電気穿孔法または燐酸カルシウム沈澱法のそれぞれにより連続してトランスフェクトした。NKG2AとNKG2C cDNAを発現ベクターpcDNA3.1/ヒグロベクター(ヒグロマイシン耐性遺伝子を含有)の中へクローニングし、そしてDAP12発現ベクターpcDNA3.1/ゼオ(ゼオマイシン耐性)の中へクローニングした。P815細胞を500μF、0.25ボルトで電気穿孔し、そして2日後に選択物(G418とヒグロマイシンとゼオマイシン)を添加した。高いレベルのレセプターを発現する細胞を、フローサイトメトリーでソートした。L細胞を燐酸カルシウムDNA沈澱によってトランスフェクトし、G418、ヒグロマイシンおよびゼオマイシンの存在下で選択した。トランスフェクタントを限定希釈によってクローニングし、CD94/NKG2レセプターの細胞表面発現を特定の抗体を使ってモニターした。

- 潜在的標的細胞に対するCD94/NKG2A+NK細胞の活性を阻害する方法であって、標的細胞表面でHLA−Eを発現することを含む方法。
- HLA−Eが細胞内に安定に組み込まれている異種のDNAによって発現されるものである請求項1の方法。
- 細胞がHLA−Eを通常は発現しない請求項2の方法。
- 細胞が非ヒトの哺乳動物細胞である請求項3の方法。
- 細胞が異種個体の器官または組織の中の一群の細胞に存在する請求項1から4までの何れかの方法。
- 潜在的標的細胞に対するCD94/NKG2C+NK細胞の活性を刺激する方法であって、標的細胞表面でHLA−Eを発現することを含む方法。
- HLA−Eが細胞内に安定に組み込まれている異種のDNAによって発現されるものである請求項6の方法。
