Description
サンプルは、Ca2+/カルモジュリン依存性プロテインキナーゼの自己リン酸化部位を含む配列を合成した。すなわち、9merのペプチド(C02059B、配列番号2:Met-His-Arg-Gln-Glu-Thr-Val-Asp-Cys)の6番目のスレオニン残基にリン酸基が導入されたものを用い、合成した。比較対照用としてスレオニン残基にリン酸基が導入されていない非リン酸化ペプチド(C02059A、配列番号1:Met-His-Arg-Gln-Glu-Thr-Val-Asp-Cys)を用いた。なお、ペプチドはペプチド合成機を用いて合成し、リン酸化ペプチドはペプチド合成後に質量分析計(マススペクトル)でリン酸基が導入されていることを確認した。
スレオニン残基リン酸化ペプチドであるC02059B(配列番号2)と、スレオニンの代わりにセリンを導入し、セリンにリン酸基を導入したリン酸化ペプチドであるC02014B(配列番号4:Met-His-Arg-Gln-Glu-Ser-Val-Asp-Cys)及びスレオニンの代わりにチロシンを導入し、チロシンにリン酸基を導入したリン酸化ペプチドであるC02014D(配列番号6:Met-His-Arg-Gln-Glu-Tyr-Val-Asp-Cys)をサンプルとして、実施例1の条件にて顕微FT−IRの測定を行った。測定結果を図1中のスペクトル(d)、(f)に示す。また、比較対照にセリン導入非リン酸化ペプチド(C02014A、配列番号3:Met-His-Arg-Gln-Glu-Ser-Val-Asp-Cys)及びチロシン導入非リン酸化ペプチド(C02014C、配列番号5:Met-His-Arg-Gln-Glu-Tyr-Val-Asp-Cys)を用いた。測定結果を図1中のスペクトル(c)、(e)に示す。
リン酸化を判別するに際し、試料のpHに依存してピーク位置がどの程度変動するかを調べた。実験例1,2の試料をpH2−4に調整し、スペクトルを測定し、ピーク位置の波数の値を算出したところ、図2に示すようにpHによりピーク位置の波数は変動するが、スレオニン、セリン及びチロシンのピーク位置の相関は保たれ、上記手法でアミノ酸の種別まで可能であることを見いだした。

Claims (2)
- 赤外分光光度計によって得られたペプチドまたはタンパク質の赤外吸収スペクトルに関する情報を入力する入力手段と、
前記入力手段で入力された赤外吸収スペクトルに関する情報を記憶する記憶手段と、
少なくとも前記記憶手段に記憶された情報をもとに得られた赤外吸収スペクトルを出力する出力手段と、
を備える解析装置であって、
前記解析装置は、前記記憶手段に記憶された赤外吸収スペクトルに基づいて該ペプチドまたはタンパク質の赤外吸収スペクトルに含まれるピーク値を算出する算出手段と、前記ピーク値における波数値を記憶する手段を有し、
予め記憶されたリン酸化したアミノ酸の赤外吸収スペクトルに含まれるピーク値における波数値と前記ピーク値の波数値とを比較して、該ペプチドまたはタンパク質のリン酸化の有無を判別する判別手段と、を備え、前記判別手段は、被検対象のペプチドまたはタンパク質に含まれるアミノ酸の中で、どのアミノ酸がリン酸化しているかを特定する特定手段をも有することを特徴とするリン酸化判別解析装置。 - 赤外分光光度計によって得られたペプチドまたはタンパク質の赤外吸収スペクトルに関する情報を入力する入力手段と、
前記入力手段で入力された赤外吸収スペクトルに関する情報を記憶する記憶手段と、
少なくとも前記記憶手段に記憶された情報をもとに得られた赤外吸収スペクトルを出力する出力手段と、
を備えるコンピュータを機能させるプログラムであって、
前記コンピュータにおいて、
前記記憶手段に記憶された赤外吸収スペクトルに基づいて該ペプチドまたはタンパク質の赤外吸収スペクトルに含まれるピーク値を算出する算出手段と、前記ピーク値の波数値を記憶する手段を用いて、
予め記憶されたリン酸化したアミノ酸の赤外吸収スペクトルに含まれるピーク値における波数値と前記ピーク値の波数値とを比較し、該数値の相違から該ペプチドまたはタンパク質のリン酸化の有無を判別する手段と、
予め記憶されたリン酸化されたアミノ酸の種類に対応する赤外吸収スペクトルに含まれるピーク値における波数値と前記ピーク値の波数値とを比較して、
被検対象のペプチドまたはタンパク質に含まれるアミノ酸の中で、どのアミノ酸がリン酸化しているかを特定する特定手段と、
前記リン酸化されているかどうかの情報及び、前記どのアミノ酸がリン酸化しているかの情報を出力する出力手段と、
を機能させることを特徴とするリン酸化判別プログラム。
