a)所定の蛋白質のためにコードするコード配列、b)コード配列の発現を制御するプロモーター配列、c)少なくとも1つの線状配列の末端で分子Bと結合する成分aおよびbを含有して接合した分子A。分子量が500以上、好ましくは1000以上、好ましくは10000以上を有する分子を分子Bと呼ぶ。合目的的には分子Bは蛋白質である。分子Aは少なくとも1つの自由な結合部位を分子Aと対になる分子Bのために有する比較的小さい分子である。分子Bは、さらにまた分子Bと対になる分子Aのために複数の結合部位を有してもよい。分子Aと分子Bとの間の結合は、非共有的でも共有的でもよい。線状配列の末端のヌクレオチドへの分子Aの結合は合目的的に共有的である。線状配列の3'末端にも5'末端にも各々1つの分子Aが取り込まれるときは、3'−および5'−エキソヌクレアーゼに対する本発明による核酸の安定化が行われる。両末端で、二本鎖の核酸のセンス鎖でもアンチセンス鎖でもそれぞれ5'−末端に1つのプライマーを分子Aで非ハイブリダイズすることによって分子Aを接合することができる。
a)核酸塩基配列が3’および5’末端でそれぞれ1つのアダプタープライマーでハイブリダイズされる、b)ステップa)の産物が3’および5’末端で伸長配列を含有するそれぞれ1つの伸長プライマーとハイブリダイズされ、核酸塩基配列から核酸塩基配列の3’および5’末端で伸長配列分だけ伸長かつ増幅された核酸配列が形成され、好ましくは最後の増幅ステップで使用したプライマーが分子Aをもっている。
PCRは、トリス−HCl10mM(20℃でpH8.85)、KCl25mM、(NH4)2SO45mM、MgSO42mM、各dNTP0.25mM、PwoDNAポリメラーゼ3U(ロシュ)ならびに本例で指示した量の核酸塩基配列を含む本例で定量化した反応容積で実施した。サイクルは94℃で0.5分、55℃で1分および72℃で1分で実施した。
インビトロ発現、文献箇所Zubay、g.; Annu. ReV. Genet. 7:267−287(1973)に基づき以下のような変更によって実施した。Esherichia coliS−30溶解物をT7ファージRNAポリメラーゼ750U/ml(Stratagene)および300μM[14C]Leu(15dpm/pmol、アマースハム)で補充した。PCR産物および制御プラスミドは、1nM〜15nMの濃度で使用した。反応は37℃で実施し、その経過は後の時点で部分標本5μlを反応混合物から抽出し、TCA沈降によって[14C]Leuの組込みを評価した。別の部分標本10μlは、合成した蛋白質の分析のためにSDS−PAGEを利用してオートラジオグラフィーに続いてホスホイメージャ−システム(モレキュラーダイナミクス)で抽出した。
ウシ心臓脂肪酸結合蛋白質のためにコードして構成されたプラスミドpET BH−FABP(Specht,B.ら;J.Biotechnol. 33:259−269(1994))の高複製誘導体がpHMFAと呼ばれる。pET BH−FABPのフラグメントはエンドヌクレアーゼSphIとEcoRIによる消化によって生成し、ベクターpUC18の中に挿入した。H−FABPの合成にとり重要である配列に関して、プラスミドpHMFAはオリジナルプラスミドと同一である。ここで線状化プラスミドは環状プラスミドよりも良好に挙動しないことを注記しておく。
プラスミドpHMFAは、プロモーターの下流に種々の配列領域を有する核酸の構造のためのマトリックスとして利用する。プロモーターの上流に塩基対0.5および249を有する構造体(例参照)FA1、FA2およびFA4は、プライマーP1、C1およびP2ならびに下流のプライマーP3によって発生した。プロモーターの上流に15塩基対の配列領域を有する構造体FA3は、エンドヌクレアーゼBglIIによるFA4の消化によって得た。3040塩基対の配列領域を有する制御プラスミドpHMFA(EcoRV)は、EcoRVによるプラスミドの消化によって得た。全産物は、「高純度PCR産物精製キット」によるゲル抽出に続き、アガロースゲル電気泳動によって洗浄した。
Strep−tagIIを含有する脂肪酸結合蛋白質の洗浄(Voss, S.ら;Protein Eng. 10:975−962(1997))は、アフィニティカラム(200μl)の容積を低減したほかは、製造者規則(IBAゲッティンゲン、ドイツ)に基づくアフィニティクロマトグラフィーを利用して実施した。結合した転写/翻訳の反応混合物は短く遠心分離し、次にカラムに取り込んだ。単離した画分はSDS−PAGE(上記参照)によるTCA沈降とオートラジオグラフィーによって分析した。
インビトロ合成H−FABPを含む完全な反応混合物は、オレイン酸の結合の活性について調査した。様々な容積(0〜30μl)にH−FABPを含有しない反応混合物を30μlまで充填し、翻訳緩衝溶液(50mM HEPES pH7.6、70mM KOAc、30mM NH4Cl、10mM MgCl2、0.1mM EDTA、0.002%NaN3)で最終容積120μlに希釈した。比活性1000dpm/pmolを有するオレイン酸(アマースハム)2μl5mM[9.10(n)−3H]の添加後、サンプルを1時間37℃でインキュベートした。サンプル50μlをゲル濾過(マイクロバイオスピンクロマトグラフィカラム;バイオ−ラート)を利用して非結合オレイン酸の除去のために使用した。溶離した画分の3H放射能は、シンチレーションカウンターで測定した。
放射性標識核酸を前記条件に従って、ただしdCTP0.167μCi/μl[α−35S]の存在下に合成した。標識した核酸は、結合した転写/翻訳、反応容積400μlに使用した。部分標本30μlは、連続する時点で抽出した。リボヌクレアーゼA15μg(DNAseなし、ロシュ)の添加後、これを15分間37℃でインキュベートした。さらに37℃で30分間のインキュベートは、SDS0.5%、EDTA20mMおよびプロテイナーゼK500μg/ml(ジブコBRL)を全反応容積60μlで実施した。残留するPCR産物は、さらにエタノール沈降によって洗浄し、その後に減成電気泳動(ポリアクリルアミド5.3%、尿素7M、SDS0.1%、TBE)にかけた。乾燥したゲルは放射能の定量化のためにホスイメージャシステム(モレキュラーダイナミクス)を通過させた。
使用したプライマー配列は図1に示した。
図2に、4プライマーによる本発明による1段階PCRの模式図を示した。中央にまずエンドヌクレアーゼNcolおよびBamHIによる消化によりpHMFAから548bp制限断片として得たH−FABP(「ウシ心臓由来の均質および機能活性脂肪酸結合蛋白質」“homogeneous and functionally active fatty acid binding protein from bovine heart”)のための完全なコーディング配列を含む蛋白質のためにコードする核酸塩基配列が識別される(ならびに翻訳されず、アダプタープライマーまたは伸長プライマーとも相補的ではない3’−末端の150bp配列)。それに核酸塩基配列の末端と均質の末端を包む両アダプタープライマーAおよびBがハイブリダイズされる。アダプタープライマーAは、さらにリボソーム結合配列を含む。アダプタープライマーAおよびBの外部末端に伸長プライマーCおよびDがハイブリダイズされる。伸長プライマーCはT7転写プロモーターを含むT7遺伝子10リーダー配列と、選択により下流に、たとえば5ヌクレオチドの配列とを包む。伸長プライマーDはT7遺伝子10ターミネータ配列を含む。
プロモーターの上流に種々の配列領域(0、5、15、250塩基対)を有する4PCR産物(FA1〜FA4)およびプロモーターの上流に3040bpを有する線状化制御プラスミドpHMFA(EcoRV)は、様々な濃度(1、5、10および15mM)でインビトロ転写/翻訳について調査した。図3は、0(下側曲線)を除き、全配列領域が蛋白質生合成の増加を生ぜしめることを示す。すでに5塩基対で充分である。
ターミネータ配列の状態の影響の調査のために、産物FAstおよびFAast(図2参照)を生成した。両者とも、22bpスペーサー配列がストップコドンとターミネータとの間で異なるプライマーを用いて挿入したこと以外は、FAtおよびFAatと同一である。図5に、スペーサー配列が約2倍の発現の増加を生ぜしめることが識別される。
本発明による方法の効果性および特異性を大量の適格DNAの存在下に調査した。FAstのPCRは、以下を例外として、前記説明に準じて実施した。すなわち核酸塩基配列は0.16〜20pg/50μl反応器容積の濃度で実施し、反応をEsherichia coliからクロモソマールDNA0.83μgを用いて、5分間超音波処理して補充した。500万倍過剰の適格DNAの存在によってPCR産物の質も量も影響されないことが見出された。
放射性標識FAast10μgを含む反応混合物をアフィニティ洗浄にかけ
た。装填物質約81%をカラムから得て、純産物として67%を溶離画分中で得ることができた(アフィニティカラムの画分のTCA沈降から計算)。
H−FABPのサンプルを、プラスミドまたはPCR産物FAastのいずれかを利用して合成し、相互にオレイン酸の結合活性について調査した。転写/翻訳後、非標識H−FABP0〜330pmolを有する種々の容積を結合アッセイ中で前記方法の説明に従って調査した。活性は、製造方法に関係なく同一であることが見出された。
PCR産物の安定性はできる限り発現の効果性を制限しうるかの調査のために、PCR産物FAastの減少を測定した。そのために放射性標識産物を使用した。所定の時間間隔で反応混合物の部分標本から抽出し、減成ポリアクリルアミドゲル電気泳動を利用して調査した。残留するPCR産物の量は、ゲルの放射能のスキャニングによって定量し、SDS−PAGEによる反応混合物の分離後にゲル中のH−FABPの放射能のスキャニングによって測定して、蛋白質生合成の時間的推移と比較した。この結果は図6に示した。PCR産物の半減期は約100分になり、これがプラトー部でH−FABP合成の流入に相当することが識別される。
表1に、反応容積25μl中の4プライマーによるPCR用の最適化条件をまとめた。
例9からの物質を用いて、ただし付加的に2増幅プライマーe(26ヌクレオチド)とf(33ヌクレオチド)ならびに増加したアダプタープライマー濃度0.2μMを用いて、変化させた伸長プライマー濃度を調節した。増幅プライマーに関しては、図1のBIORおよびBIOFを参照されたい。BIOFはビオチン標識フォワードプライマーであり、BIORはビオチン標識リザーブプライマーである。この構造は図7に示している。
基本的に前記のような物質を用いて処理する。初めに反応容積5μl中のプレPCRを核酸塩基配列0.1pg、しかもアダプタプライマー0.3μMおよび伸長プライマー0.5μMで20サイクル以上実施する。次にこのように得た反応溶液をPCR開始容積で25μlに希釈する。次に増幅プライマーを最終濃度0.5μMまで添加する。最後に、さらに30サイクル増幅する。
プライマーBIOFおよびBIORの使用下に6プライマーによるPCRの方法で、前記のように核酸を製造し、その分解を時間の関数ならびに蛋白質合成の改善を調査する。これは図7および図8に示している。ビオチンにより、特にストレプトアビジンによる転換後、著しく改善された安定性が生じることが識別される。これは20%までの高い蛋白質合成をもたらし、これは1つの系で少量の割合のエキソヌクレアーゼを有する。従ってこの例は、さらにこのような系で蛋白質合成効率の改善が達成されることを証明する。より多いエキソヌクレアーゼの量を有する溶解物を含む系内では、係数5まで、およびそれ以上の合成効率の改善が予想されている。
Claims (4)
- 細胞を含有しない蛋白質生合成系または細胞による蛋白質生合成系における、コード配列によってコードする蛋白質の製造方法における核酸の使用であって、
次の構成要素:
a)所定のペプチドまたは蛋白質のためにコードするコード配列、
b)選択的に、コード配列の発現を制御するプロモーター配列、および
c)前記構成要素aおよびbを含有する線状配列の両末端に各々1つ結合された分子Aであって、分子量500以上の分子Bと結合する分子A
をもつエキソヌクレアーゼによる分解に対して安定化した核酸の使用。 - 前記構成要素aおよび/またはbと分子Aとの間にスペーサー配列が配置される、請求項1記載の核酸の使用。
- 各分子Aが各々1つの分子Bと結合するか、または両分子Aがただ1つの、分子Aのために少なくとも2つの結合部位を有する分子Bと結合する、請求項1または2のいずれか1項記載の核酸の使用。
- 分子Aがビオチンまたはジゴキシゲニンであり、分子Bがアビジン、ストレプトアビジンまたは抗ジゴキシゲニン抗体である、請求項1ないし3のいずれか1項記載の核酸の使用。
