Description
本発明は、哺乳類細胞において分化を調節するための特異的調節プロモーターの使用に関する。
組織や臓器の損傷を治療するための移植療法の重要性の認識および理解が拡がりつつある。臓器移植は広く実施されているが、個々の細胞移植を元にした治療法は未だ比較的臨床開発の初期段階にある。
本発明は、ネスチンの発現を制御する調節因子が、癌遺伝子でトランスフェクトした細胞における発現を調節することに利用できるという理解をもとにしている。
本発明は、移植療法に適し、および移植時まで不死である細胞の調製方法を開示する。
Claims (13)
- ネスチン遺伝子の第2イントロンのエンハンサーエレメント、もしくは該エンハンサーエレメントの機能的な断片と作動可能に結合された、条件的に誘導可能な癌遺伝子をコードするポリヌクレオチドを含む構築物であって、該エレメントが、ヒトネスチン遺伝子の第2イントロンの3’部分の714bpもしくは374bpいずれかのエレメントである構築物。
- さらに癌遺伝子のプロモーターとして働くポリヌクレオチドを含む請求項1に記載の構築物。
- プロモーターがHSV tkプロモーターである請求項2に記載の構築物。
- さらにテロメラーゼ複合体の触媒サブユニットを含む前記請求項のいずれかに記載の構築物。
- 癌遺伝子がSV40大型T抗原の温度感受性変異体である請求項1に記載の構築物。
- 癌遺伝子がc−myc癌遺伝子を含む請求項1〜5のいずれかに記載の構築物。
- 前記請求項のいずれかに記載の構築物を含む細胞。
- 哺乳類の未分化多能性細胞である請求項7に記載の細胞。
- 未分化、多能性のヒト神経上皮細胞である請求項7もしくは請求項8に記載の細胞。
- 治療に使用するための請求項7〜9のいずれかに記載の細胞。
- 条件的に細胞を不死化させるための請求項1〜6のいずれかに記載の構築物の使用。
- 細胞が請求項8もしくは9において定義される、請求項11に記載の使用。
- 細胞の損失もしくは損傷によって引き起こされた疾病を治療するための移植用薬剤の製造における請求項7〜9のいずれかに記載の細胞の使用。
