Description
(i)金属錯体を酸化する工程、および、
(ii)発生期水素により金属錯体を還元して、化学発光可能な形態の金属錯体を形成させる工程、
(iii)化学発光により検体を検出する工程、
により金属錯体を発光させる、標識基として発光性金属錯体を用いてサンプル中の検体を検出する方法に関する。
(i)金属錯体を酸化する手段、および、
(ii)発生期水素を発生させる手段、
を含む、発光発生剤として発光性金属錯体を用いて化学発光させる装置である。
(i)金属錯体を酸化する手段、
(ii)発生期水素を発生する手段、および、
(iii)発光を検出する手段、
を含んでなる当該装置は、標識基として発光性金属錯体を用いてサンプル中の検体を検出するために用いることを意図している。
[M(L1L2L3)]n-Ym- (I)
[式中、
Mは希土類または遷移金属カチオンから選択される二価または三価の金属カチオンであり、
L1, L2および L3は同一または異なり、少なくとも2個の窒素含有へテロ環を含む配位子であって、
ここでL1, L2および L3は、窒素原子を介して金属カチオンに結合しており、
Yは、これら配位子のうちの1個に結合した結合基であり(これら配位子により錯体が、(a)生物学的物質に結合するかまたは(b)生物学的物質に結合し得る)、
mは1〜10、好ましくは1〜4、特に好ましくは1の整数であり、
nは1〜6、好ましくは1〜3、特に好ましくは1の整数である]
の構造を含む化学発光性金属錯体を用いることである。
ルテニウム(ビピリジル)3錯体(Ru2+カチオン含有)をRu3+錯体に酸化した。このためにLi/ブタノール/H2SO4を含む均一系を用いた。濃硫酸をPbO2(粉末)入りの容器に添加し、ブタノールで覆った。Ru2+錯体を1mmolの濃度にブタノールを用いて溶解した。Ru2+錯体をH2SO4とブタノールの界面で酸化した。Ru2+錯体をRu3+に酸化させた後、リチウムを加えた。この工程において、発生期水素がH2SO4とブタノールの界面で形成される。この界面にて顕著なルテニウム化学発光が観察された。
Claims (8)
- 標識基として発光性金属錯体を用いてサンプル中の検体を検出する方法であって、金属錯体の化学発光が、以下の工程:
(i)金属錯体を酸化する工程、および
(ii)発生期水素により金属錯体を還元して、化学発光可能な形態の金属錯体を形成する工程、
(iii)化学発光により検体を測定する工程、
により起こることを特徴とする、該方法。 - 発生期水素を金属錯体の直接近傍で発生させることを特徴とする、請求項1に記載の方法。
- 発生期水素を電気化学的に発生させることを特徴とする、請求項1または2に記載の方法。
- 発生期水素を化学的に発生させる、請求項1〜3のいずれか1項に記載の方法。
- 発生期水素を、超音波で発生させることを特徴とする、請求項1〜4のいずれか1項に記載の方法。
- 標識基として発光性金属錯体を用いてサンプル中の検体を検出する装置であって、以下の手段:
(i)金属錯体を酸化する手段、
(ii)酸化された金属錯体を還元するために発生期水素を発生させる手段、および、
(iii)金属錯体の還元により発生した化学発光を検出する手段、
を含む、該装置。 - 発光性金属錯体の提供、金属錯体の酸化、発生期水素による金属錯体の還元により、化学発光することが可能な形態の金属錯体を形成させることを含む、化学発光の発生法。
- 発光性金属錯体を用いて化学発光を発生させるために、
(i)金属錯体を酸化する手段、および
(ii)酸化された金属錯体を還元するために発生期水素を発生する手段
を含む装置の使用。
