図1は、本発明によるFETセンサのセンシング部である金電極へのDNAプローブへの固定化法の操作フローを示している。図1(a)はDNA固定化溶液、図1(b)はDNAプローブの固定化密度が制御された様子、図1(c)は固定化密度が制御されたDNAプローブを用いてハイブリダイゼーションを行った様子を示している。

測定装置:ALSモデル611B電気化学アナライザー(Bioanalytical System, Ltd.,)
作用電極:アルカンチオール固定化金電極
対極:Ptカウンター電極,BAS製
参照電極:Ag/AgCl,RE-1C参照電極,BAS製
測定溶液:0.5M水酸化カリウム溶液
測定装置:ALSモデル611B電気化学アナライザー(Bioanalytical System, Ltd.,)
作用電極:アルカンチオール固定化金電極
対極:Ptカウンター電極,BAS製
参照電極:Ag/AgCl,RE-1C参照電極,BAS製
測定溶液:0.1M Na2SO4溶液
Claims (8)
- 電界効果トランジスタのゲートと配線で接続された金電極にDNAプローブを固定化してDNA固定化FETセンサを作製する方法において、
金電極と結合する反応基を有するアルカンチオールと結合したDNAプローブと、金電極と結合する反応基を有するアルカンチオールのみを有する化合物を含む固定化溶液を調製する工程と、
前記固定化溶液を前記金電極表面に接触させる工程とを有し、
前記アルカンチオールのアルキル基が炭素鎖6以上であり、前記固定化溶液中の前記アルカンチオールのみを有する化合物の濃度が0.5mM以上であることを特徴とする電位計測用のDNA固定化FETセンサ作製方法。 - 請求項1記載のDNA固定化FETセンサ作製方法において、前記固定化溶液中のDNAプローブと前記アルカンチオールの分子数比が1:2〜1:100であることを特徴とする電位計測用のDNA固定化FETセンサ作製方法。
- サンプル中のターゲットと結合するDNAプローブとアルカンチオールとが表面に固定化された金電極を有し、
前記アルカンチオールのアルキル基が炭素鎖6以上であり、
前記金電極表面に固定化された前記アルカンチオールの固定化密度が4×1014個/cm2以上であることを特徴とする電位計測用のDNA固定化FETセンサ。 - 請求項3記載の電位計測用のDNA固定化FETセンサにおいて、前記金電極表面に固定化されたDNAプローブの固定化密度が4×1012個/cm2以下であることを特徴とする電位計測用のDNA固定化FETセンサ。
- 表面にDNAプローブとアルカンチオールとが固定化され、試料溶液と接触する金電極を有する電位計測用の電界効果型トランジスタと、
試料溶液と接触する参照電極と、
前記金電極と参照電極との間に交流電圧を印加する手段とを備え、
前記アルカンチオールのアルキル基が炭素鎖6以上であり、
前記金電極表面に固定化された前記アルカンチオールの固定化密度が4×10 14 個/cm 2 以上であることを特徴とするDNA測定装置。 - 請求項5記載のDNA測定装置において、前記金電極表面に固定化されたDNAプローブの固定化密度が4×1012個/cm2以下であることを特徴とするDNA測定装置。
- サンプル中のターゲットと結合するDNAプローブとアルカンチオールとが表面に固定化された電極を有する電位計測用の電界効果トランジスタの前記電極にバッファー溶液を接触させる工程と、
前記電極と、前記バッファー溶液に接触した参照電極との間に交流電圧を印加する工程と、
前記バッファー溶液中にサンプルを注入する工程と、
前記サンプル注入の前後における前記電界効果トランジスタの応答の変化を検出する工程とを有し、
前記アルカンチオールのアルキル基が炭素鎖6以上であり、
前記電極表面に固定化された前記アルカンチオールの固定化密度が4×10 14 個/cm 2 以上であることを特徴とするDNA測定方法。 - 請求項7記載のDNA測定方法において、前記電極表面に固定化された前記DNAプローブの固定化密度が4×1012個/cm2以下であることを特徴とするDNA測定方法。
