本発明は、抗レジオネラ属菌組成物、これを含有するレジオネラ属菌用抗菌剤、およびレジオネラ感染源に対するレジオネラ属菌除菌剤に関する。

【従来の技術】
レジオネラ属菌は水系、土壌など自然界に広く分布するブドウ糖非発酵グラム陰性桿菌であり、肺炎を主徴とする細菌感染症を惹起する。本菌は細胞内増殖菌であり、生体の中ではマクロファージなどの貪食細胞の中で殺菌されずに増殖することが可能である。また、β−ラクタマーゼ産生菌であるため、一部のβラクタム剤に耐性を示す。

感染症の多くは外来性に進入した菌により発症する。感染源としては空調用冷却水、温泉水、老朽化した給水設備による水溜まり等が挙げられている。さらに最近ではいわゆる24時間風呂でのレジオネラ菌増殖が問題視されている。歴史的には、1976年米国フィラデルフィア市内で全米在郷軍人大会が開催された際、原因不明の重傷肺炎が集団発生し、そのうち29名が死亡したが、その後の調査によってレジオネラ菌(レジオネラ・ニューモフィラ)が原因菌であることが明らかになった。我が国においては1981年、レジオネラ肺炎の第一例が報告されて以来、病院での院内感染等多数の報告がなされている。

【発明が解決しようとする課題】
レジオネラ属菌にはエリスロマイシン等の抗生物質が有効であるといわれているが、これらの抗生物質を日常的に使用することは耐性菌の出現や副作用の点で好ましくない。空調用冷却水等の感染源に対する殺菌剤として、塩素やアルコール類、界面活性剤等が挙げられるが、これらの殺菌剤は安全性の点で問題があり、飲食物等に配合することはできない。

このような状況にあって、レジオネラ属菌の感染を予防するという考え方が広がりつつあり、手軽に使用でき、安全性に問題のない抗菌活性素材の開発が望まれているところ、柑橘類果物の種子抽出物、好ましくはグレープフルーツの種子抽出物が抗レジオネラ属菌活性を有するということが特開平6−233986号公報に開示されている。しかし、その効果はレジオネラ属菌の増殖を完全に抑えるほど高いものとは認められない。

そこで、発明者らは、日常的に使用しても副作用がなく、飲食物等に含有させても安全性があり、しかも、レジオネラ属菌の増殖を強力に阻止できる抗レジオネラ属菌組成物を求め、古来より副作用がなく安全性が高いとの評価を得て広く利用されている植物に注目して鋭意研究し、それらのうちのある種の植物の抽出物にレジオネラ属菌増殖阻害作用があることを見出し、本発明を完成するに至ったものである。

【課題を解決するための手段】
すなわち、本発明に係る抗レジオネラ属菌組成物は、マンゴスチン果皮から低級アルコールを含む抽出溶媒によって抽出した抽出物をレジオネラ属菌に対する抗菌成分としたものである。

マンゴスチン果皮から低級アルコールを含む抽出溶媒によって抽出した抽出物の安全性については全く問題がないものであり、また、本発明抗レジオネラ属菌組成物は、レジオネラ属菌の増殖を阻害することにより、レジオネラ属菌感染を防ぐのに有効にしてかつ安全なものである。

マンゴスチン果皮からの抽出物の抽出には、水、メタノール、エタノール、ブタノール等の低級アルコール、ヘキサン、エーテル、酢酸エチル等の有機溶媒の1種または2種以上を適宜混合して使用することができる。また、抽出条件は高温、室温、低温のいずれでもよいが、50〜80℃、1〜5時間程度が好ましい。抽出液は、濾過され、抽出溶媒の留去が行われた後、減圧下で濃縮するかあるいは凍結乾燥されて、目的の抽出物となる。

レジオネラ属菌抗菌剤は、マンゴスチン果皮からの抽出物を所要の製剤用担体ないし希釈剤と任意に混合し、たとえば、経口投与剤として、マウスウォッシュ、トローチ剤、吸入剤、噴霧剤等とする。マウスウォッシュやトローチ剤の場合、口腔内に侵入したレジオネラ菌に対して、その感染を予防することが可能である。吸入剤として用いれば、口腔内および気道内におけるレジオネラ菌に対して同様の効果が期待できる。また、噴霧剤として直接感染源や生体に噴霧することにより、その除菌が可能である。レジオネラ属菌抗菌剤における抽出物の添加量は、最終製品の形態によっても異なるが、その効果の発現を考慮すると0.001%以上の添加が好ましい。

また、レジオネラ属菌除菌剤としては、空調用冷却水の除菌剤、除菌効果を有する入浴剤等を例示することができる。空調用冷却水の除菌剤は、抽出物を無機担体に吸着させ、耐水性ポリマーに分散し成型することによって、安定した除菌力を長期間維持し得る製品を製造することが可能である。レジオネラ菌除菌剤における抽出物の濃度については、実際の使用にあたりその濃度が0.001%以上になることが好ましい。

【発明の実施の形態】
前記抽出物を使用し、レジオネラ属菌用抗菌剤としてマウスウォッシュ、トローチ剤を、レジオネラ属菌感染予防用飲食物としてチューインガム、キャンディ、錠菓、飲料を、レジオネラ感染源に対する除菌剤として入浴剤を実施例として示す。ただし、本発明はこれら実施例に限定されるものではない。

〔植物抽出物の調製〕

〔試験1〕 レジオネラ属菌に対する抽出物の抗菌活性試験
供試菌株 レジオネラ・ニューモフィラ(Legionella pneumophila) ATCC 33152
試料 表2に示した抽出物37種類を用いた。
試験方法 供試菌をバッファード・スターチ・イーストエキス(B−SYE)液体培地で好気的条件下、24時間振盪培養を行い、これを菌液とした。
96穴マイクロプレートを用い各ウェル中で試料溶液(100μl)の2倍希釈系列を調製した。2倍濃度で調製したB−SYE液体培地に上記菌液を接種し、その100μlを各ウェルに添加した。好気的条件下で37℃、48〜72時間培養後マイクロプレートリーダーを用いて550nmにおける各ウェルの濁度を測定し、菌の生育を判定し、最小発育阻止濃度(MIC)を求めた。試験結果を表3に示した。

〔試験2〕 抽出物のレジオネラ属菌除菌試験(シェークフラスコ法)

試験方法 供試菌をバッファード・スターチ・イーストエキス(B−SYE)液体培地で好気的条件下37℃、24時間振盪培養を行い、その後、菌数1〜2×105 個/mlとなるように調製し、これを菌液とした。
滅菌したリン酸緩衝液に試料を懸濁し、この緩衝液に、菌液を菌数1〜2×104 個/mlとなるように加え、1時間振盪した。振盪前と振盪後の緩衝液(菌液添加済)をB−SYE寒天培地に塗抹し、好気的条件下37℃、48時間培養した。培地上のコロニーを計数し、試験菌の除菌率を以下の式により求めた。
