Description
(1)高感度(1fmol量の検出が可能)
(2)高安定性(10年前に精製したピリジルアミノ化糖鎖が現在でも蛍光強度変化無し)
(3)高分解能(高速液体クロマトグラフィー(HPLC)との組み合わせにより、1000個の糖鎖を分離可能)
(4)高汎用性(マススペクトロメトリー(MS)との相性もよい)
(5)定量が可能(1つの糖鎖に1ヶ所しか入らない)
このような従来のピリジルアミノ化法の一例は、以下の通りである。まず、糖鎖を含む試料に対して、20μl(マイクロリットル、1μlは10-9m3)の、2−アミノピリジン/酢酸溶液(552mg/200μl)を加え、よく撹拌後、90℃で60分間加熱する。反応後、70μlのボラン・ジメチルアミン錯体/酢酸溶液/水(200mg/80μl/50μl)を加え、よく撹拌後、80℃で35分間加熱する。反応終了後、有機溶媒による抽出、陽イオン交換樹脂による夾雑物の除去によりピリジルアミノ化糖鎖を得ている。
本発明に係る実施の第一形態における糖類の蛍光標識化装置では、図1に示すように、例えば30ml容量の、有底円筒状の容器(密閉容器)1が、その上部の開口部を密閉可能で、着脱自在なスクリューキャップ1bを有して設けられている。容器1の内底部や外底部の形状については、特に限定されないが、平面状または先端に向かって徐々に径が細くなるテーパー形状が好ましく、特に、容器1を試験立てなどに収め易いことから、上記テーパー形状がより好ましい。
本発明に係る実施の第二形態における糖類の蛍光標識化装置においては、例えば図3に示すように、ガラスからなる、なす型フラスコ(密閉容器)の容器1が、その内部をねじ式にて密閉できるスクリューキャップ1bと、容器1内部の圧力を調整(例えば減圧)するための圧力調整口1cと、容器1内部での圧力の調整のための気体出入管1dとを有して設けられている。圧力調整口1cは、スクリューキャップ1bの頂部に取り付けられている。

Claims (7)
- 糖鎖の還元末端をピリジルアミノ化して、上記糖鎖を蛍光標識化する糖類の蛍光標識化方法において、
上記糖鎖に対し、2−アミノピリジンを気体の状態にて接触させることを特徴とする糖類の蛍光標識化方法。 - 密閉容器内に、糖鎖を含む試料と、2−アミノピリジンを含む標識化剤溶液とを、互いに隔離して収納し、
上記標識化剤溶液を加熱し、上記2−アミノピリジンを気体化して上記糖鎖と接触させることを特徴とする請求項1記載の糖類の蛍光標識化方法。 - 前記糖鎖に対し、2−アミノピリジンを気体の状態にて接触させるとき、弱酸性の雰囲気下とすることを特徴とする請求項1または2記載の糖類の蛍光標識化方法。
- 前記弱酸性の雰囲気の形成に、酢酸を用いることを特徴とする請求項3記載の糖類の蛍光標識化方法。
- 前記糖鎖の還元末端と、2−アミノピリジンとの反応中間体の−CH=N−基を還元するための還元剤を、気体の状態にて上記反応中間体に接触させることを特徴とする請求項1ないし4の何れか1項に記載の糖類の蛍光標識化方法。
- 前記還元剤に、ボラン・ジメチルアミン錯体を用いることを特徴とする請求項5記載の糖類の蛍光標識化方法。
- 糖鎖を含む試料と、2−アミノピリジンを含む標識化剤溶液と、前記糖鎖の還元末端と、2−アミノピリジンとの反応中間体の−CH=N−基を還元するための還元剤とをそれぞれ収納する容器と、
前記試料と、前記標識化剤溶液と、前記還元剤とを、互いに溶液の状態での接触を阻害するためにそれぞれ設けられた壁部と、
前記標識化剤溶液と、前記還元剤とを、それぞれ気体の状態にて前記試料に接触させるためにそれぞれ前記壁部に形成された開口部とを有していることを特徴とする糖類の蛍光標識化装置。
