本発明はエリスリトールの製造方法に関し、更に詳しくは、微生物を利用して発酵法により工業的に有利にエリスリトールを製造する方法に関する。

【従来の技術】
エリスリトールの製造方法としては、トリゴノブシス属、キャンジダ属の微生物をグリセロールを炭素源とする培地に培養して製造する方法(特公昭47−41549号公報)、キャンジダ属、トルロプシス属、ハンゼヌラ属の微生物を炭化水素などを炭素源とする培地に培養して製造する方法(特公昭51−21072号公報)等が知られている。しかしながら、これらの方法は炭素源として使用される原料が実際の工業的生産において適当でないため未だ工業化されていない。

また、モニリエラ・トメントサ・パール・ポリニスをグルコース等の糖質を炭素源とする培地に培養して製造する方法(特開昭60−110295号公報)も知られている。この方法は安価で安全な原料であるグルコースを使用し、かつ生産性も高いという点ですぐれているが、培養中の発泡が著しく、通常使用されている消泡剤では役にたたないため、高価なキサダンガムなどを多量に添加する必要があり工業的生産においては必ずしも有利な方法とはいえない。

【発明が解決しようとする課題】
本発明の目的は、安価で且つ容易に供給しうる原料から、高収率で安価にエリスリトールを製造する方法を提供することにある。

【課題を解決するための手段】
本発明者らは、安価な原料からエリスリトールを製造する方法について鋭意研究した結果、モニリエラ属及びトリコスポロノイデス属に属する多くの微生物が、グルコース、フルクトースなどの発酵性糖質から多量のエリスリトールを産生することを見出した。なかでもトリコスポロノイデス属のいくつかの微生物がエリスリトールの生産性が特に優れていることを見出し、本発明を完成するに至った。

すなわち本発明は、トリコスポロノイデス・オエドセファリスCBS649.66株、トリコスポロノイデス・メディアCBS240.79株及びトリコスポロノイデス・ニグレッセンスCBS268.81株より成る群から選ばれた微生物を、培地中の発酵性糖質の濃度が20〜60%(w/v)の培地で培養し、培養物からエリスリトールを採取することを特徴とするエリスリトールの製造法を提供するものである。

更に、この発明の好ましい態様によれば、発酵性糖質がグルコース、フルクトース及びグリセロールより成る群から選ばれたものである上記の製造法、培養を25〜37℃で行う上記の製造法、及びpHが3〜7の培地に微生物を接種して培養する上記の製造法が提供される。

【発明の実施の形態】
以下、本発明の実施の態様について説明する。本発明で用いる微生物は、モニリエラ(Moniliella)属又はトリコスポロノイデス(Trichosporonoides)属に属し、且つ本発明者らにより発酵性糖質からエリスリトールを産生する能力を有することが見出されたものである。

モニエラ属に属する微生物としては、例えば、モニリエラ・アセトアブテン(Moniliella acetoabutens)及びモニリエラ・スアヴェオレンス(Moniliella suaveolens)等を挙げることができ、具体的菌株としては、例えば、モニリエラ・アセトアブテンCBS169.66、モニリエラ・スアヴェオレンスCBS126.42及びモニリエラ・スアヴェオレンスCBS120.67等を挙げることができる。

トリコスポロノイデス属に属する微生物としては、例えば、トリコスポロノイデス・オエドセファリス(Trichosporonoides oedocephalis)、トリコスポロノイデス・メガチリエンシス(Trichosporonoides megachiliensis)、トリコスポロノイデス・メディア(Trichosporonoides madida)、トリコスポロノイデス・ニグレッセンス(Trichosporonoides nigrescens)及びトリコスポロノイデス・スパスラタ(Trichosporonoides spathulata)等を挙げることができ、具体的菌株としては、例えば、トリコスポロノイデス・オエドセファリスCBS649.66、トリコスポロノイデス・メガチリエンシスCBS567.85、トリコスポロノイデス・メディアCBS240.79、トリコスポロノイデス・ニグレッセンスCBS268.81及びトリコスポロノイデス・スパスラタCBS241.79等を挙げることができる。

これらの菌株は、国際寄託機関であるオランダ国の Centraal Bureau voor Schimmelcultures(CBS)に寄託されており容易に入手できる。
上記微生物の培養に使用される培地の主炭素源としては、グルコース、フルクトース、グリセロール等の発酵性糖質が利用される。これらの炭素源は単独でも組み合わせても使用できる。使用濃度は特に限定されないが、エリスリトールの産生を阻害しない範囲で可能な限り高くするのが有利である。好ましい濃度は20〜60%(W/V)の範囲内である。

窒素源としてはアンモニア塩、尿素、ペプトン、微生物エキス、コーンステープリカーなどの各種の有機、無機の窒素化合物が用いられる。無機塩としては各種リン酸塩、硫酸塩、マグネシウム、カリウム、マンガン、鉄、亜鉛等の金属塩が用いられる。また、ビタミン、ヌクレオチド、アミノ酸等の微生物の生育を促進する因子を必要に応じて添加することができる。また、培養中の発泡を抑えるために市販の消泡剤を適量添加しておくことが望ましい。

培養に際しては、斜面培養から菌体を直接培地に接種しても構わないが、液体培地で1日〜4日間の培養で得られる前培養物を接種するのが望ましい。
培養の初めの培地はpH3〜7、好ましくはpH3〜4.5に調整する。培養温度は25℃〜37℃、好ましくは27℃〜35℃が適当である。また、培養は通気攪拌、振とう等の好気的条件で行うのが望ましい。培養時間は主炭素源が消費されるまで行われるのが好ましく、通常は3〜8日間行われる。なお、培養液中のエリスリトール生成量はガスクロマトグラフィー、高速液体クロマトグラフィーなどの方法で測定することができる。

このようにして培養液中に蓄積したエリスリトールは常法に従って、培養物より分離・精製される。具体的には、遠心分離、ろ過等により固形物を除去した後、活性炭、イオン交換樹脂により脱色、脱塩し、その溶液から結晶化することによりエリスリトールを分離・精製することができる。

以下、本発明を実施例により更に具体的に説明するが、本発明の範囲は下記の実施例により何等限定されるものではない。
実施例1〜8
グルコース30%(W/V)及び酵母エキス1%を含む培地50mlを、綿栓した500mlの三角フラスコに入れ、120℃で20分間滅菌した。この培地に、モニリエラ・アセトアブテンCBS169.66株、モニリエラ・スアヴェオレンCBS126.42株、モニリエラ・スアヴェオレンスCBS120.67株、トリコスポロノイデス・オエドセファリスCBS649.66株、トリコスポロノイデス・メディアCBS240.79株、トリコスポロノイデス・ニグレッセンスCBS268.81株、トリコスポロノイデス・スパスラタCBS241.79株及びトリコスポロノイデス・メガチリエンシスCBS567.85株をそれぞれ植菌し、27℃で10日間振とう培養した。培養終了後、培養液中のエリスリトール濃度を高速液体クロマトグラフィーで測定した。
