Description
本発明は、Helicobacter pyloriのNAPタンパク質に関する。

(背景)
Helicobacter pyloriは、ヒトの胃に感染するグラム陰性のらせん形の細菌である。世界の50%を超える人口がこの細菌を保有していると考えられている。

H.pyloriの臨床単離株は、上皮細胞における空胞の形成を誘導する空胞形成細胞毒素(VacA)、および免疫的に優勢な細胞毒素関連抗原(CagA)の発現により特徴付けられ得る。I型株は、潰瘍または癌を有する患者において優勢であり、これらのタンパク質両方を発現するが、II型株は、どちらも発現しない。

精製されたNAPが多量体形態へとアセンブルする能力を、サイズ排除クロマトグラフィー(非還元、非変性)を使用して調べた。NAPを、緩衝液(25mM Tris−HCl、150mM NaCl、pH7.8)で平衡化したSepharose 12 HR 10/30カラムにローディングし、そして0.5ml/分で溶出した。そのNAPの供給源に関わらず、そのタンパク質は、酵母のアルコールデヒドロゲナーゼ(分子量150kDa)と同じ保持時間を有する単一ピークで溶出した。このことは、十量体構造[6]を示す。

Claims (7)

- 請求項1に記載のプロセスであって、金属キレートクロマトグラフィーの工程をさらに包含する、プロセス。

- タンパク質混合物からのH.pylori NAPの精製のためのプロセスであって、該プロセスは塩析の工程および金属キレートクロマトグラフィーの工程を包含する、プロセス。
- 実質的に他のタンパク質を含まないH.pylori NAPを含む組成物を生産するためのプロセスであって、
H.pyloriNAPを生産する工程;および
他のタンパク質を塩析する工程であって、最終の塩濃度は、少なくとも50%飽和である、工程、
