Abstract
Description
本発明は、製薬技術分野に属し、具体的に、置換ジアリールアミン化合物及びその医薬組成物、製造方法と用途に関する。
パーキンソン病(Parkinson’s disease,PD)は、よく見られる神経変性疾患の1つであり、1~2%の高齢世代に影響を与えている。ゲノムワイド関連研究(Genome-wide association studies,GWAS)は、非家族性PDの24個の部位に、関連する遺伝的リスク変異を28種関連した。そのうち、LRRK2(Park8)の突然変異は遺伝形式においても発見され、家族性と非家族性PDの共通分子経路により駆動される発症機序が確定され、関連疾患を引き起こす最も一般的な原因である(Simon-Sanchez J, Schulte C, Bras JM, Sharma M, Gibbs JR, et al. Genome-wide association study reveals genetic risk underlying Parkinson’s disease. Nat Genet,2009,41,1308-1312.)。PD病原性LRRK2突然変異マップは主にキナーゼ(G2019S、I2020T)及びROC-CORドメイン(R1441C/G/H、Y1699C)に関連していることで、これらの酵素活性が病状に非常に大切であることを意味する。病原性突然変異の頻度は、大体、全体の2%程度と稀であるが、ある人種の人々では、最もよく見られる突然変異G2019Sが最大40%の患者で発見され、当該キナーゼが2~3倍に活性化される(West AB, Moore DJ, Biskup S, Bugayenko A, Smith WW, et al. From The Cover: Parkinson’s disease-associated mutations in leucine-rich repeat kinase 2 augment kinase activity. Proceedings of the National Academy of Sciences,2005,102,16842-16847.)。病原性突然変異の他に、LRRK2の一般的な遺伝的変異は、散発性PDの危険因子である(Tan, E. K. Identification of a common genetic risk variant(LRRK2 Gly2385Arg) in Parkinson’s disease. Ann Acad Med Singapore. 2006,35,840-842.)。
上記技術問題を改善するため、本発明は、式(I)で示される化合物、そのラセミ体、立体異性体、互変異性体、同位体標識、N-酸化物、水和物、溶媒和物、結晶多形物、代謝物、薬学的に許容される塩、薬学的に許容されるエステル又はそのプロドラッグ化合物を提供する:
R1、R2は、それらと接続された原子と共に、非置換又は任意選択的に1、2個又はそれ以上のRbにより置換された3~20員ヘテロシクリル基を形成し、

本発明の実施形態によれば、PGは、アミノ保護基から選ばれてもよい。そのうち、好適なPGは、C1-40アルキル基、C6-20アリールC1-40アルキル基、C1-40アルキルスルホニル基、C1-40アルキルベンゼンスルホニル基、例えば、4-トルエンスルホニル基、tert-ブチル基、イソプロピル基、ベンジル基、tert-ブトキシカルボニル基(Boc)、2-ビフェニル-2-プロポキシカルボニル基、ベンジルオキシカルボニル基、フルオレニルメトキシカルボニル基(Fmoc)、トリフルオロアセチル基から選ばれてもよい。
(i)キナーゼ及び既知基質の存在下で、本発明の化合物を上記キナーゼに接触させるステップと、
(ii)上記キナーゼと上記既知基質との間の相互作用の何れかの変化を検測するステップと、を含む。
本発明により提供される化合物は、良好なLRRK2キナーゼ調節/阻害作用を有すると共に、より優れた脳透過率を有し、LRRK2キナーゼ活性に関連する病症と疾患、特に脳透過率に関連する病症と疾患の治療、及びこのような症状と疾患のための薬物の調製に利用可能である。本発明の化合物は、製造方法が簡単で、良い応用の見通しを有する。
特に説明のない限り、本願の明細書及び特許請求の範囲に記載される基と用語の定義は、実例としての定義、例示的な定義、好ましい定義、表に記載される定義、実施例における具体的な化合物の定義などを含み、互いに任意に組み合わせたり、結合したりすることができる。このような組み合わせと結合後の基の定義及び化合物の構造は、本願の明細書及び/又は特許請求の範囲に記載される範囲内であると理解すべきである。
本発明の別の態様は、分析試験における上記化合物の用途に関し、この分析試験は、1種又は複数種のキナーゼ、より好ましくはLRRK、更により好ましくはLRRK2を抑制可能な他の候補化合物を同定するためである。
(i)キナーゼの既知基質の存在下で、リガンドを上記キナーゼに接触するステップと、
(ii)上記キナーゼと上記既知基質との間の相互作用の何れかの変化を検測するステップと、を含み、
且つそのうち、上記リガンドは本発明の化合物である。

〔クロマトグラフィー〕
Agela Technologies製の機器により高速液体クロマトグラフィーを行い、そして多波長UV検測器によりモニターした。分離工程に用いられる典型的な移動相は、PE/EA、DCM/MeOH又は水/MeCNである。
特に説明のない限り、室温で上記溶媒において、分光器Bruker AV 400により1H核磁気共鳴(NMR)分光を行った。何れの場合にも、NMRデータは、提出された構造と一致している。メインピークを指定するための一般的な略語を使用し、100万分の1単位で特有の化学シフト(δ)を示す:例えば、s、一重線;d、二重線;t、三重線;q、四重線;dd、二重線の二重線;br、幅広線。Agilent 1290 Infinity/6460 triple Quad LCMSにより質量分析を記録した。薄層クロマトグラフィー(TLC)を使用する場合、それはシリカゲルTLCを指す。
製造出発原料が記載されていない場合に、これらの出発原料は、商業的に入手されるもの、文献に既知のもの、又は当業者の使用標準手順で容易に取得可能なものであってもよい。従来のような実施例又は中間体により化合物を製造する場合、当業者には、それぞれの特定の反応の反応時間、試薬の当量数と温度を変更することができ、且つ異なる後処理又は精製技術の採用が必須又は所望のことであり得ると理解される。

材料
Adapta(商標)キナーゼ測定法(LRRK2_IC50測定)では:キナーゼ反応緩衝液に5×キナーゼ緩衝液S(Life Technologies、PV5213)及び2mMのDTT(Life Technologies、P2325)を含んだ。キナーゼLRRK2 G2019Sタンパク質(PV4881)及びERM(LRRKtide、PV5093)は、Life Technologiesから得られた。Adapta(商標)汎用キナーゼ測定試薬キット(Life Technologies、PV5099)は、Adapta(商標)Eu-抗-ADP抗体(PV5097、4μg)、10μMのAlexaFluor(登録商標)647 ADPトレーサー(PV5098、200pmol)、TR-FRET希釈緩衝液(PV3574、100mL)、キナーゼクエンチ緩衝液(P2825、1mL)、10mMのATP(PV3227、500μL)、及び10mMのADP(PV5096、500μL)を含む。LRRK2-IN-1(1234480-84-2、HY-10875)は、MedChem Expressから得られた。
Adapta(登録商標)汎用キナーゼ測定法は、ADPの均一性を検測するための蛍光に基づく免疫測定法である。ATP消費測定法とは逆に、Adapta(登録商標)測定法は、ADPの形成に非常に敏感であり、ほとんどのシグナル変化は、最初の10~20%のATPがADPに転化された時に発生される。これにより、Adapta(登録商標)汎用キナーゼ測定法は、低活性キナーゼに非常に適用される。
本発明の実施例の化合物のLRRK2 G2019Sへの阻害のIC50値は、表3に示す通りである。
試験品:化合物4、純度99%
試験品の調製 22.73mgの化合物4を秤量してすり鉢に入れ(純度99%で換算)、少量の0.5%のCMC-Naを加えて十分に研磨し、容器に移し、更にすり鉢を4回洗浄し、洗浄液を容器に移し、そして0.5%のCMC-Naを30mLになるまで加え、濃度が0.75mg/mLの試験品懸濁液として調製した。サンプリング及び投与に先立ち、磁気撹拌機により1200rpmの回転速度で少なくとも15min撹拌し、用時調製した。
投与頻度と用量:単回投与30mg/kg(40mL/kg)
投与後に、各群の動物はそれぞれ、対応する時点0.25、0.5、1、2、4、6、8、24hに、心臓から0.4mL採血し、腹腔内に50mg/kgのゾレチルを注射して麻酔し、そして安楽死になるまで放血し、脳、肺、腎を取り、生理食塩水により洗浄してからろ紙で水分を取り、凍結保存管に入れ、ドライアイスに暫く保存し、12h以内に-60℃以下で保存した。
