Abstract
Description
文献:R.A.Sperlingらによる“Toward defining the preclinical stages of Alzheimer’s disease: Recommendations from the National Institute on Aging-Alzheimer’s Association workgroups on diagnostic guidelines for Alzheimer’s disease,” Alzheimer’s Dement.,vol.7,no.3,pp.280-92,2011年を参照。 文献:J.L. Price及びJ.C. Morrisによる“Tangles and plaques in nondemented aging and ‘preclinicaT Alzheimer’s disease,”Ann.Neurol.,vol.45,no.3,pp.358-68,1999年;J.C.Morrisらによる“Mild cognitive impairment represents early-stage Alzheimer disease,”Arch.Neurol.,vol.58,no.3,pp.397-405,2001年を参照。
文献:S.E.Counts,M.D. Ikonomovic,N.Mercado,I.E.Vega及びE.J.Mufsonによる“Biomarkers for the Early Detection and Progression of Alzheimer’s Disease,”Neurotherapeutics,vol.14,no.2017,pp. 35-53,2016年を参照。
[0005] AD進行バイオマーカのモデル(文献:C.R.Jackらによる“Hypothetical model of dynamic biomarkers of the Alzheimer’s pathological cascade,”The Lancet Neurology,vol.9,no.1.Lancet Publishing Group,pp.119-28,2010年を参照)によれば、図1に示されるように、脳脊髄液(CSF)、Aβ42アッセイ、又は陽電子放出断層撮影(PET)アミロイド画像化で検出可能なアミロイドβ(Aβ)の変化4は、シナプス損失(文献:J.Jacksonらによる“Targeting the synapse in Alzheimer’s disease,”Front.
文献:B.P.Luceyらによる“Reduced non rapid eye movement sleep is associated with tau pathology in early Alzheimer’s disease,”Sci.Transl.Med.,vol.11,no.474,2019年を参照。
[0007] Aβ負荷に関するNREM睡眠と覚醒との間の相互作用を調整するプロセスに関してMander等により提案されたモデル(文献:B.A.Mander,J.Winer,W.J.Jagust及びM.P.Walkerによる“Sleep: A Novel Mechanistic Pathway, Biomarker, and Treatment Target in the Pathology of Alzheimer’s Disease?”Trends Neurosci.,vol.39,no.8,pp.552-66,2016年を参照)によれば、図2におけるように、覚醒の間において:
[0008] - グリムファティック・アミロイド・クリアランスは低く(文献:L.Xieらによる“Sleep drives metAβolite Clearance from the adult Brain,”Science,vol.342,no.6156,pp.373-77,2013年を参照);
[0009] - 神経代謝及びニューロンのスパイキング活動は高く(文献:M.Buchsbaumらによる“Regional Cerebral Glucose MetAβolic Rate in Human Sleep Assessed by Positron Emission Tomography,”Life Sci.,vol.45,no.15,pp.1349-56,1989年を参照);及び
[0010]- 酸化ストレスは高い(文献:G.Villafuerte,A.Miguel-Puga,E.Murillo Rodriguez,S.Machado, E.Manjarrez及びO.Arias-Carrionによる“Sleep deprivation and oxidative stress in animal models:A systematic review,”Oxid.Med.Cell.
[0012] - 高い酸化ストレス(文献:S.M.Yatin,S.Varadarajan,C.D.Link及びD.AΒutterfieldによる“In vitro and in vivo oxidative stress associated with Alzheimer’s amyloid b-βpeptide (1-42),”Neurobiol.Aging,vol.20,no.3,pp.325-30,1999年を参照);
[0013] - 神経細胞の過興奮性(文献:M.TAβuchiらによる“Sleep interacts with aβ to modulate internal neuronal excitAβility,”Curr.Biol.,vol.25,no.6,pp.702-12,2015年を参照);
[0015] - 恐らく脳アミロイド血管障害を引き起こすことによる、グリファティック・クリアランスの減少(文献:R.O.Weller,S.D.Preston, M.Subash及びR.O.Carareによる“Cerebral amyloid angiopathy in the aetiology and immunotherapy of Alzheimer disease,”Alzheimers.
[0015] - 恐らく脳アミロイド血管障害を引き起こすことによる、グリファティック・クリアランスの減少(文献:R.O.Weller,S.D.Preston, M.Subash及びR.O.Carareによる“Cerebral amyloid angiopathy in the aetiology and immunotherapy of Alzheimer disease,”Alzheimers.
[0016] これらの要因は、Aβがそれ自体の蓄積を促進するように思われる正のフィードバックループを構成する(文献:B.A.Manderらによる“Beta-amyloid destroys human NREM slow wave and related hippocampus-dependent memory conversion,”Nat.Neurosci.,vol.18,no.7,pp.1-10,2015年を参照)。

[0019] - 神経代謝率は低く(文献:A.R Braunらによる“Regional cerebral Blood flow through the sleep-wake cycle.An H2150 PET study,”Brain,vol.120,no.7,pp.1173-97,1997年を参照);及び
Hoggによる「Cell Injury and Repair Resultsing from Sleep Loss and Sleep Recovery in LAβoratory Rats」、Sleep、vol.37、no.12,pp.1929-40,2014年を参照)。
文献:G.Tononi及びC.Cirelliによる“Sleep and the price of plasticity:from synaptic and cellular homeostasis to memory consolidation and integration,”Neuron,vol.81,no.1,pp.12-34,Jan.2014年を参照。 文献:J.Schneider,P.A.Lewis,D.Koester,J.Born,H.-V.V.Ngoによる“Susceptibility to auditory closed-loop stimulation of sleep slow oscillations changes with age,”Sleep,zsaal11,2019年を参照。
文献:E.Bresch,U.Grossekathofer及びG.Garcia-Molinaによる“Recurrent deep neural networks for real-time sleep stage classification from single channel EEG,”Front.Comput.Neurosci.,vol.12,no.2018,pp.1-12,2018年;及びG.Garcia-molinaらによる“Hybrid in-phase and continuous auditory stimulation significantly enhances slow wave activity during sleep,”in 2019 41st Annual International Conference of the IEEE Engineering in Medicine and Biology Society EMBC),2019年,pp.4052-55を参照。 文献:G.Garcia-Molinaらによる“Closed-loop system to enhance slow- wave activity,”J.Neural Eng.,vol.15,no.6,pp.1-11,2018年を参照。
文献:“Targeting the synapse in Alzheimer’s disease,”前出を参照。 文献:“Sleep and the price of plasticity: from synaptic and cellular homeostasis to memory consolidation and integration,”前出を参照。 当該反応の振幅は年齢とともに減少する(“Susceptibility to auditory closed-loop stimulation of sleep slow oscillations changes with age,”前出を参照)が、初期のアミロイド蓄積による減少は、より劇的であると予想される。
文献:G.Garcia-Molinaらによる“Closed-loop system to enhance slow-wave activity,”J.Neural Eng.,vol.15,no.6,pp.1-11,2018年;H.V.Ngo,A.Miedema,I.Faude,T.Martinetz,M.Molle及びJ.Bornによる“Driving Sleep Slow Oscillations by Auditory Closed-Loop Stimulation-A Self-Limiting Process,”J.Neurosci.,vol.35,no.17,pp.6630-38,2015年;N.A.Papalambrosらによる“Acoustic enhancement of sleep slow oscillations and concomitant memory improvement in older adults,”Front.Hum.Neurosci.,vol.11,no.March,pp.1-14,2017年;及びN.A.Papalambros,S.Weintraub,T.Chen,G.Santostasi,K.A.Paller及びP.C.Zeeによる“Acoustic enhancement of sleep slow oscillations in mild cognitive impairment,”pp.1-42,2019年を参照。この効果は、若い被験者(24.2±0.9歳)において(H.V.Ngo、T.Martinetz、J.Born及びM.Molleによる“Auditory Closed-Loop Stimulation of the Sleep Slow Oscillation Enhances Memory”Neuron、vol.78、no.May、pp.1-9、2013年を参照)、高齢者(60~84歳)において(“Acoustic enhancement of sleep slow oscillations and concomitant memory improvement in older adults,”前出を参照)、及び軽度の認知障害を有する患者においても(“Acoustic enhancement of sleep slow oscillations in mild cognitive impairment,”前出を参照)観察され得る。
