材料および方法
細胞の培養:Hep-3B肝細胞癌細胞およびLX-2肝星細胞を、MeM-Eagle培地で増殖させた。すべての培地には、ペニシリン(10ユニット/ml)、ストレプトマイシン(10μg/ml)、L-グルタミン(2mM)および10%ウシ胎児血清(FBS)が含まれていた。細胞は、37℃、5%のCO2のインキュベータにおいてT-75フラスコ内で維持し、週に2回、新しく調製した培地に移した。すべての実験において、血清飢餓細胞を使用した。FBSは、18時間培養から省き、実験は、37℃、5%のCO2のインキュベータにおいて、1%のFBSを添加したDMEM培地中の細胞の単層で実施した。異なる比率(C15/T3、C3/T15)のCBD/THCを100pM、1、10、100nMの濃度で培養系に導入した(元のオイルからDMSOで100mMの原液を調製し、さらに培養液で希釈して実験系でnM濃度に到達するようにした。)
図1Aは、CBD/THC含有抽出物が、2つのカンナビノイドの比率に関係なく、ナノモル濃度で星細胞の増殖を抑制する効果があることを示している。
Claims (39)
- A3アデノシン受容体(A3AR)のアゴニストによって治療可能な疾患または障害を治療するための製剤であって、前記疾患または障害を治療または改善するのに有効な量の少なくとも1のカンナビノイドを含むことを特徴とする製剤。
- 請求項1に記載の製剤において、
前記カンナビノイドが、主にA3ARを介してその効果を発揮することを特徴とする製剤。 - 請求項1または2に記載の製剤において、
前記カンナビノイドが、A3ARのアンタゴニストによって低減される前記疾患または障害に対する生理学的効果または治療効果を有することを特徴とする製剤。 - 請求項1~3の何れか一項に記載の製剤において、
少なくとも1のカンナビノイドが、A3AR活性化因子作用を有することを特徴とする製剤。 - 請求項4に記載の製剤において、
少なくとも1のカンナビノイドが、A3ARアゴニスト活性を有することを特徴とする製剤。 - 請求項1~5の何れか一項に記載の製剤において、
前記疾患または障害が、前記疾患または障害に罹患していない対象の同じ系統の細胞または組織と比較した、細胞または組織におけるA3ARの発現レベルの上昇と関連していることを特徴とする製剤。 - 請求項1~6の何れか一項に記載の製剤において、
前記疾患または障害が、癌、炎症性疾患、NAFLDなどの肝疾患または肥満であることを特徴とする製剤。 - 請求項1~7の何れか一項に記載の製剤において、
(i)テトラヒドロカンナビノール酸(THCA)、テトラヒドロカンナビノール(THC)、カンナビジオール酸(CBDA)、カンナビジオール(CBD)、カンナビノール(CBN)、カンナビゲロール(CBG)、カンナビクロメン(CBC)、テトラヒドロカンナビバリン(THCV)、カンナビディバリン(CBDV)と、
(ii)(i)のカンナビノイドの化学誘導体と、
(iii)大麻植物の抽出物と
からなる群のなかから選択される少なくとも1のカンナビノイドを含むことを特徴とする製剤。 - 請求項1~8の何れか一項に記載の製剤において、
