洗剤および試薬
表1にリストされている例示的な洗剤(Seppic Inc.Fairfield,NJ)のストック溶液、および1,2-プロパンジオール(Sigma-Aldrich、St.Louis,MO)を、重量/体積(w/w)パーセントで水中で調製する。
これらの研究で使用する異種指向性マウス白血病ウイルス(XMuLV)、ブタパルボウイルス(PPV)NADL-2株、および仮性狂犬病ウイルス(PRV)は、ATCC、Manassas,VAから購入することができる。表2に、これらのウイルスの特性を示す。
細胞ベースの50%組織培養感染量(TCID50)アッセイは、PG-4指標胞を使用したXMuLV、PK-13指標細胞を使用したPPV、およびVero指標細胞を使用したPRVのウイルス滴定に使用する。96ウェル組織培養プレートに、2.5×104個細胞/mLの指標細胞を、200μL/ウェルで播種し、37℃で一晩または約16~24時間インキュベートする。次いで、プレートに、50μL/ウェルで一連の10倍希釈液でウイルスを、希釈液当たりn=8の最小値で接種する。接種したプレートを37℃で7~9日間(XMuLVおよびPRV)および10~13日間(PPV)インキュベートする。次いで、プレートを検査し、細胞変性効果(CPE)について顕微鏡下でウェルごとに陽性または陰性としてスコア付けし、ウイルス力価を標準的な方法で計算する。
上で例示したTCID50アッセイを使用して、モノクローナル抗体(mAb)、二重特異性抗体、およびFc融合タンパク質の浄化されたバイオリアクター収集材料におけるウイルスの細胞傷害性効果を決定する。バイオリアクターの収集材料を、遠心分離とそれに続く研磨濾過によって浄化する。HCP、脂質、宿主細胞DNAなどの不純物も、遠心分離プロセスの一部として自動的に濃縮する。0.05w/w%~1w/w%の洗剤を含有するさまざまなタンパク質の浄化されたバイオリアクター収集材料を、接種培地で10,50、100倍に希釈する。各希釈液をn=8で96ウェルの指標細胞播種プレートに添加し、TCID50アッセイに従ってインキュベートする。インキュベーション期間の終わりに、各ウェルを顕微鏡下でCPEについて観察し、各洗剤に必要な適切な希釈を決定する。結果は、洗剤による細胞傷害性を克服するには、50倍希釈係数で十分であることを示した。
各洗剤によるさまざまなエンベロープウイルスのウイルス不活化を、mAb、二重特異性、およびFc融合タンパク質を含有する浄化されたバイオリアクター収集材料において、さまざまな濃度、温度、およびpHの範囲で評価する。XMuLV、PPV、またはPRVウイルスを、5w/w%以下で収集材料にスパイクし、洗剤ストック溶液をスパイクされた材料に添加して、指定された最終洗剤濃度を達成する。次に、試料を特定の温度およびpH範囲でインキュベートする。洗剤がウイルス含有材料に添加されるとすぐに、ウイルスの不活化が開始する。ウイルス不活化の動態を、収集試料からさまざまな時点でアリコートを取り出すことによってモニターし、ウイルス力価を、TCID50アッセイによって評価する。ウイルスの不活化は、洗剤処理の結果であり、マトリックス自体ではないことを実証するために、マトリックス対照が研究に含まれている。
表1に記載されている洗剤によるXMuLVのウイルス不活化は、XMuLV不活化について決定された最適条件で試験する。上で例示したウイルス不活化手順に従って、各洗剤を、0.6w/w%でXMuLVスパイクの浄化されたmAbバイオリアクター収集材料に30℃で添加する。ウイルスの不活化を、ウイルス接種後180分の最長不活化時間でさまざまな時点でモニターする。
濁度を、0%、0.05%、0.1%、0.3%、0.5%、および1w/w%のSIMULSOL(登録商標)SL11Wの濃度で、18℃で浄化されたmAbバイオリアクター収集材料にスパイクされたXMuLVにおいて評価する。ウイルスの不活化を180分間モニターし、濁度を、携帯型濁度計2100P(Hach(登録商標))を使用して比濁法濁度単位(NTU)で測定する。
タンパク質の製造方法で使用するための実行可能な環境適合性洗剤としてのSIMULSOL(登録商標)SL11Wの使用を、上記のウイルス不活化手順に従って、浄化されたmAbバイオリアクター収集材料でさらに評価する。簡単に説明すると、SIMULSOL(登録商標)SL11W活性を、4℃、18℃、25℃、および30℃の4つの異なる温度で、XMuLVスパイクの浄化されたmAbバイオリアクター収集材料において、0.05%、0.1%、0.3%、0.5%、および1w/w%の濃度で評価する。ウイルスの不活化を、XMuLVを浄化されたmAbバイオリアクター収集材料を含有するSIMULSOL(登録商標)SL11Wに接種してから0、10、30、60、120、および180分後にモニターする。表5~9に示される結果は、SIMULSOL(登録商標)SL11Wの濃度、インキュベーション時間、および温度がすべてXMuLVの不活化に影響を与える要因であることを実証している。SIMULSOL(登録商標)SL11Wは、評価した4つの温度すべてで、濃度≧0.1w/w%(≧3mM)で120分までに、濃度≧0.3w/w%で10分までに完全なXMuLVウイルス不活化を達成した。
SIMULSOL(登録商標)SL11Wが、水、細胞培養培地、ダルベッコのリン酸緩衝生理食塩水(DPBS)などのさまざまな開始マトリックスで9mM(0.3w/w%)のウイルスを4℃および30℃で不活化する能力を評価する。ウイルスの不活化を、XMuLVの接種後0、60、120、および180分にモニターする。
SIMULSOL(登録商標)SL11Wがより低い時点でウイルスを不活化する能力を評価する。SIMULSOL(登録商標)SL11Wを、9mM(0.3w/w%)で、4℃および30℃で浄化したmAbバイオリアクター収集材料に添加する。ウイルスの不活化を、XMuLVの接種後0、2、4、6、8、10、20分および180分にモニターする。
30℃で浄化されたmAbバイオリアクター収集材料におけるモノプロピレングリコールの0.1%および0.5%の濃度でのウイルス不活化に対する効果を評価する。ウイルスの不活化を、XMuLVの接種後0分および180分でモニターする。SIMULSOL(登録商標)SL11Wは、1%~5%のモノプロピレングリコールを含有する。ここで報告されている不活化手順で見られるモノプロピレングリコールの最大濃度は、0.05%である。表12に示される結果は、0.1%および0.5%の濃度のモノプロピレングリコールが、30℃で浄化されたmAbバイオリアクター収集材料中のXMuLVを不活化しないことを示している。さらに、表11に示されているモノプロピレングリコールの濃度は、SIMULSOL(登録商標)SL11Wで処理された浄化されたmAbバイオリアクター収集物で見られる最大濃度を少なくとも10倍上回っている。これは、SIMULSOL(登録商標)SL11Wが観察されたウイルスの不活化に関与していることを示している。

SIMULSOL(登録商標)SL11Wが、mAb、二重特異性抗体、Fc融合タンパク質などのさまざまな組換えタンパク質を含有する浄化されたバイオリアクター収集材料中でXMuLVを不活化する能力を評価する。SIMULSOL(登録商標)SL11Wを、9mM(0.3w/w%)で、4℃および18℃で異なるタンパク質の浄化されたバイオリアクター収集材料に添加する。ウイルスの不活化を、0、10、30、60、120、および180分のインキュベーション時点でモニターする。表15に示される結果は、SIMULSOL(登録商標)SL11WによるXMuLVの完全な不活化を、3つの浄化されたバイオリアクター収集材料すべてにおいて4℃および30℃の両方で120分までに示している。さらに、Fc融合タンパク質およびmAbタンパク質を含有する収集物では、4℃で10分までに完全な不活化が観察された。二重特異性抗体の不活化動態は、4℃での他の2つの浄化されたバイオリアクター収集材料よりも4℃でわずかに遅かった。しかし、18℃では、SIMULSOL(登録商標)SL11Wによる完全な不活化が、10分以内に3つのタンパク質すべてで観察された。
Fc融合タンパク質、二重特異性抗体、およびmAbの3つの異なる組換えタンパク質のバイオリアクター収集材料における不純物レベルの影響を評価する。
濃縮された浄化されたmAb収集材料中のウイルスの不活化に対するSIMULSOL(登録商標)SL11Wの効果を試験する。mAbおよびその他の不純物を含有する浄化されたmAbバイオリアクター収集材料を、5KDa分子量カットオフタンジェンシャルフロー濾過(TFF)プロセスによって約4倍に濃縮させる。次いで、mAbおよび不純物を含有する濃縮された材料を、直ちに滅菌濾過に供する。次いで、SIMULSOL(登録商標)SL11Wを、浄化した濃縮濾過材に最終濃度0.1w/w%(3mM)で添加する。次いで、SIMULSOL(登録商標)11Wを含有する材料を、4℃、18℃、30℃の3つの異なる温度でインキュベートする。さまざまな温度でのウイルスの不活化を、0分~180分の範囲の時点で測定する。表17に示す結果は、30℃の試料と比較した場合、低温で不活化速度が低下したにもかかわらず、試験した3つの温度すべてで180分でXMuLVが完全に不活化したことを示している濃縮されていないバイオリアクター収集材料と比較した場合、濃縮されたバイオリアクター材料の不活化動態への影響は観察されなかった(データは示さず)。これは、不純物レベルの増加がSIMULSOL(登録商標)SL11Wによるウイルスの不活化に影響を与えないことを示唆している。
液体クロマトグラフィー質量分析(LC-MS)によるウンデシルグリコシドの定量-方法A
LC-MSの定量を、0.03mM~3mMの範囲の濃度でSIMULSOL(登録商標)SL11Wを含有する浄化されたmAbバイオリアクター収集資料で行う。
0.0008mM~0.012mMの範囲の濃度のSIMULSOL(登録商標)SL11Wを含有する標準溶液を、水またはタンパク質Aの主流で調製する。SIMULSOL(登録商標)SL11Wを添加した後、これらの試料をLC-MS/MS(MRM)分析にかけた。インキュベートした試料(100μL)をLCに注入し、1/20ポストカラムをMSに対してスプリットする。分析には、Sciex Triple Quad 5500質量分析計に連結したAgilent 1260 Infinity II HPLCを使用する。定量では、移動相Aとして水中の0.5mMの酢酸ナトリウムおよび移動相Bとして95:5のアセトニトリル:水を使用して、80℃でAgilent Zorbax、300SB-C8逆相カラム(4.6×50mm、300Å、3.5μm)で実施する。カラムは20%の移動相Bで2分間平衡化し、10分間かけて20%から90%まで直線的に増加させ、90%のBで2分間保持し、20%の移動相Bで平衡化する。SIMULSOL(登録商標)SL11Wを、4.5~5.5分の間に1.0mL/分で分離し、エレクトロスプレーイオン化(ESI)ソースを使用して分析した。質量分析計を、遷移イオンをスキャンするポジティブMRMモード(357.0~185.0)、5.5kVのキャピラリ電圧、450℃の脱溶媒和温度、および55L/時の脱溶媒和ガス流量で作動させる。
0%(対照)、0.3%、1.0w/w%を含有する浄化されたmAbバイオリアクター収集材料溶液SIMULSOL(登録商標)SL11Wを、MabSelect(登録商標)タンパク質Aクロマトグラフィーに供する。タンパク質Aクロマトグラフィーを、SIMULSOL(登録商標)SL11Wを含有する濾過されていない浄化されたmAb収集材料で実施する。カラムを装填した後、酢酸/クエン酸緩衝液でmAbを溶出する前に、6カラム容量のTris/NaClを使用してカラムを洗浄する。溶出したmAbの主流を採取し、LC-MSでSIMULSOL(登録商標)SL11Wの存在を分析する。
組換えタンパク質の品質に対するSIMULSOL(登録商標)SL11Wの影響を、20℃で3時間、次いで4℃で16時間インキュベートする、0.3%のSIMULSOL(登録商標)SL11Wを用いる場合および用いない場合の浄化されたmAbバイオリアクター収集試料を比較することによってテストする。試料を最初に、0.2μmガラス繊維フィルターで濾過し、続いて0.45μmのPVDFフィルターで濾過し、次いでタンパク質A精製にかける。試料を、キャピラリ電気泳動(CE)、サイズ排除クロマトグラフィー(SEC)、iCE(登録商標)機器での等電点電気泳動(IEF)測定など、当技術分野において既知の技術を使用して、表18に示す分析特性について分析する。表18に示す結果は、SIMULSOL(登録商標)SL11Wによる処理が、SIMULSOL(登録商標)SL11Wによる処理がない場合と比較して、測定された品質のいずれにも顕著な影響を及ぼさなかったことを示している。
Claims (45)
- ウイルスを供給流中で不活化するための方法であって、環境適合性洗剤を、約0.05w/w%~約1w/w%の最終濃度で前記供給流に添加することを含み、前記環境適合性洗剤が、約40%以上のウンデシルアルキルグリコシドを含む、方法。
- 前記洗剤が、約40%~約60%のウンデシルアルキルグリコシドを含む、請求項1に記載の方法。
- 前記洗剤が、約53%~約57%のウンデシルアルキルグリコシドを含む、請求項1または2に記載の方法。
- 前記洗剤が、約50%以上のウンデシルアルキルグリコシドを含む、請求項1に記載の方法。
- 前記洗剤が、約10%未満のデシルアルキルグリコシドを含む、請求項1~4のいずれか一項に記載の方法。
- 前記洗剤が、SIMULSOL(登録商標)SL11Wである、請求項1~5のいずれか一項に記載の方法。
- 前記洗剤が、ウンデシルアルキルグリコシドである、請求項1~6のいずれか一項に記載の方法。
- 前記洗剤が、約0.1w/w%~約1w/w%の濃度で前記供給流に添加される、請求項1~7のいずれか一項に記載の方法。
- 前記洗剤が、約0.3w/w%~約1w/w%の最終濃度で前記供給流に添加される、請求項1~7のいずれか一項に記載の方法。
- 前記洗剤が、約0.1w/w%~約0.3w/w%の最終濃度で前記供給流に添加される、請求項1~7のいずれか一項に記載の方法。
- 前記洗剤が、約0.3w/w%の最終濃度で前記供給流に添加される、請求項1~7のいずれか一項に記載の方法。
- 前記洗剤を添加する前記工程の間、前記供給流が、約4℃~約30℃の温度にある、請求項1~11のいずれか一項に記載の方法。
- 前記洗剤を添加する前記工程の間、前記供給流が、約15℃~約30℃の温度にある、請求項1~11のいずれか一項に記載の方法。
- 前記洗剤を添加する前記工程の間、前記供給流が、約15℃~約20℃の温度にある、請求項1~11のいずれか一項に記載の方法。
- 前記洗剤を添加する前記工程の間、前記供給流が、約5.5~約8.0のpHにある、請求項1~14のいずれか一項に記載の方法。
- 前記洗剤を、前記供給流中で約1分~約180分間インキュベートする工程をさらに含む、請求項1~15のいずれか一項に記載の方法。
- インキュベートする前記工程後に、前記洗剤が、約1以上、または約2以上の前記供給流中のウイルス対数減少係数(LRF)を有する、請求項16に記載の方法。
- 前記ウイルスが、エンベロープウイルスである、請求項1~17のいずれか一項に記載の方法。
- 前記エンベロープウイルスが、レトロウイルス、ヘルペスウイルス、フラビウイルス、ポックスウイルス、ヘパドナウイルス、肝炎ウイルス、オルトミクソウイルス、パラミクソウイルス、ラブドウイルス、またはトガウイルスである、請求項18に記載の方法。
- 前記供給流と前記洗剤とを、約4℃~約30℃の温度および約5.5~約8.0のpHで、約15分~約180分間インキュベートする工程をさらに含み、前記洗剤が、約0.1w/w%~約1.0w/w%の最終濃度で前記供給流に添加され、前記洗剤が、前記供給流中で約2以上のウイルス対数減少係数(LRF)を有する、請求項1に記載の方法。
- 前記供給流と前記洗剤とを、約4℃~約30℃の温度および約5.5~約8.0のpHで、約6分~約180分間インキュベートする工程をさらに含み、前記洗剤が、約0.3w/w%~約1.0w/w%の最終濃度で前記供給流に添加され、前記洗剤が、前記供給流中で約4以上のウイルス対数減少係数(LRF)を有する、請求項1に記載の方法。
- 前記供給流と前記洗剤とを、約4℃~約30℃の温度および約5.5~約8.0のpHで、約180分間インキュベートする工程をさらに含み、前記洗剤が、約0.1w/w%~約1.0w/w%の最終濃度で前記供給流に添加され、前記洗剤が、前記供給流中で約5以上のウイルス対数減少係数(LRF)を有する、請求項1に記載の方法。
- 前記供給流と前記洗剤とを、約4℃~約30℃の温度でおよび約5.5~約8.0のpHで、約60分~約180分間インキュベートする工程をさらに含み、前記洗剤が、約0.3w/w%の最終濃度で前記供給流に添加され、前記洗剤が、前記供給流中で約5以上のウイルス対数減少係数(LRF)を有する、請求項1に記載の方法。
- 前記供給流が、収集した細胞培養液、捕捉プール、または回収した生成物プールである、請求項1~23のいずれか一項に記載の方法。
