Abstract
Description
ヒト脳微細血管内皮細胞(HBMEC;human brain microvascular endothelial cell)、ヒト脳血管周皮細胞(HBVP;human brain vascular pericyte)及びヒト星状細胞(HA;human astrocyte)の共同培養のための培養条件を最適化するために、6つの培地条件でMTS(3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium)分析法を利用してそれぞれの細胞代謝活性度を比較した。6つの培地条件は次のとおりであり、グリア細胞培地(G、glial cell medium)は、星状細胞培地(A)及び微細にかわ培地(M)の1:1混合物を意味する。
(i)E:内皮培地(endothelial medium)
(ii)A:星状細胞培地(astrocyte medium)
(iii)P:周皮細胞培地(pericyte medium)
(iv)M:微細にかわ培地(microglia medium)
(v)E+G(E+A+M):内皮培地(E)、星状細胞培地(A)及び微細にかわ培地(M)の1:1:1混合物
(vi)E+G+P(E+A+M+P):内皮培地(E)、星状細胞培地(A)、微細にかわ培地(M)及び周皮細胞培地(P)の1:1:1:1混合物
組み立てられた微細流体装置を利用して次のような方法により、血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置を製作した。
実施例2において製作したヒト脳微細血管内皮細胞(HBMEC)、ヒト脳血管周皮細胞(HBVP)及びヒト星状細胞(HA)を含む血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置の3次元組織障壁機能を試験するために、血液-脳障壁の特異的なタンパク質の発現水準をqRT-PCR分析によって確認した。比較のために、ヒト脳微細血管内皮細胞(HBMEC)のみを単一培養した場合のタンパク質発現を共に確認した。
実施例2において製作したヒト脳微細血管内皮細胞(HBMEC)、ヒト脳血管周皮細胞(HBVP)及びヒト星状細胞(HA)を含む血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置の3次元組織障壁機能を試験するためにTEER(Transendothelial electrical resistance)値を測定した。比較のためにヒト脳微細血管内皮細胞(HBMEC)のみを単一培養した場合のTEER値を共に確認した。TEER(Trans-Endothelial Electrical Resistance)は、オームの法則(Ohm’s raw)を利用し、細胞層の電気抵抗を測定する方法であって、細胞層からなった細胞障壁がどれくらいよく機能しているかを示す要素のうちの1つである。細胞層が互いに強く結合するほど、細胞間の結合をなす接合タンパク質(Junction protein)の発現が強くなり、細胞間(Paracellular)の物質輸送が制限されるので、電気抵抗値が高くなる。ヒト血液-脳障壁は物質輸送がきわめて制限されている障壁であって、TEER値が5000Ω.cm2と推定され[Lauschke K、Frederiksen L、Hall VJ(2017)Paving the way toward complex blood-brain barrier models using pluripotent stem cells.Stem Cells Dev 26(12):857-874]、ラット血液-脳障壁は5900Ω.cm2で測定されたことがある[Butt AM、Jones HC、Abbott NJ(1990)Electrical resistance across the blood-brain barrier in anaesthetized rats:a developmental study.J Physiol 429:47-62]。
実施例2において製作した血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置の第1の中心チャンネルに加えられる血液模写の流れのせん断応力による3次元組織障壁の機能の差を試験するために、第1の中心チャンネルにせん断応力が加えられない場合、0.4dyne cm-2のせん断応力を加えた場合、4dyne cm-2のせん断応力を加えた場合のそれぞれのTEER(Transendothelial electrical resistance)値を実施例4と同一の方法で測定した。
実施例2において製作したヒト脳微細血管内皮細胞(HBMEC)、ヒト脳血管周皮細胞(HBVP)及びヒト星状細胞(HA)を含む血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置の3次元組織障壁機能を試験するために、4kDa及び40kDa FITC-dextranの拡散から透過係数(permeability coefficient)を測定した。比較のために細胞を培養しなかった場合及びヒト脳微細血管内皮細胞(HBMEC)のみを単一培養した場合の透過係数を共に確認した。
実施例2において製作したヒト脳微細血管内皮細胞(HBMEC)、ヒト脳血管周皮細胞(HBVP)及びヒト星状細胞(HA)を含む血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置の第2の中心チャンネル内の星状細胞培養において、2D培養と3D培養との間の差異点を比較した。健康な星状細胞の形態学的及び生理学的な特性を保存することは、血液-脳障壁の機能を模写することにおいて重要な役割を行う。
図36ないし38に示したように、ヒト星状細胞(HA)を2Dマトリゲル(Matrigel)コーティングの表面上で培養した場合、短い突起(process)を有する平たい多角形状の拡大した細胞体の形態を示した。これに対し、ヒト星状細胞(HA)を3Dマトリゲルの内部で培養した場合、放射形に分布された薄くて長い突起を有する小さい細胞体の形態を示した。
2Dまたは3D培養されたヒト星状細胞(HA)で反応性神経膠症マーカーの遺伝子の発現水準をqRT-PCR分析によって確認した。2D及び3D培養システム(n=4)でヒト星状細胞(HA)の神経膠反応性を分析するために、標準qRT-PCRをTaqMan Fast Universal PCR Master Mix(Applied Biosystems)を用いてStepOnePlus Real-Time PCRシステム(Applied Biosystems)で行った。それぞれの標的遺伝子は商業的に入手可能なプライマを用いて評価し、実験に用いられたプライマを下記の表2に示した。
実施例2において製作したヒト脳微細血管内皮細胞(HBMEC)、ヒト脳血管周皮細胞(HBVP)及びヒト星状細胞(HA)を含む血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置の第2の中心チャンネル内の星状細胞培養において、2D培養及び3D培養によるアクアポリン-4(AQP-4;Aquaporin-4)及びα-syn(α-syntrophin)の発現を比較した。血管の周囲空間の星状細胞は末端突起の数でタンパク質であるAQP-4によって脳の水恒常性を調節するので、星状細胞末端突起におけるAQP-4の分極化は、血液-脳障壁の恒常性調節及び生理学的な条件を模写することにおいて重要な役割を行う。α-synはAQP-4の星状細胞末端への分極化を調節するアンカー(anchor)として知られている。
実施例2で製作したヒト脳微細血管内皮細胞(HBMEC)、ヒト脳血管周皮細胞(HBVP)及びヒト星状細胞(HA)を含む血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置でAQP-4の発現の分極化水準を確認した。比較のためにヒト星状細胞(HA)のみを培養した場合及びヒト星状細胞(HA)とヒト脳微細血管内皮細胞(HBMEC)を培養した場合のAQP-4の発現を共に確認した。
実施例2において製作したヒト脳微細血管内皮細胞(HBMEC)、ヒト脳血管周皮細胞(HBVP)及びヒト星状細胞(HA)を含む血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置で血液-脳障壁模写の構造を通じたナノ粒子の輸送をモニターして定量化した。中枢神経系における薬物伝達研究において、細胞及び分子水準で薬物を含む化合物の脳への輸送を定量化することは重要な課題である。
微細流体技術を利用して脂質(lipid、DMPC)、アポリポ蛋白A1(apolipoprotein A1)及び蛍光マーカーで構成されたHDL模倣の円盤型ナノ粒子eHNP-A1を生理学的に適切な大きさと構成で合成した。合成されたeHNP-A1の大きさの分布を測定して図45に示した。
尾静脈注射によって1mg/kgのeHNP-A1をマウスに全身投与した。200μLの食塩水を注入したマウスを対照群とした。投与24時間後、マウスを犠牲させて食塩水及び4%のPFAに15分の間保管した。その後、生体内の映像システム(in vivo imaging system、IVIS;Perkin Elmer、Waltham、MA、USA)を用いて各器官(脳、心臓、肺、肝、腎臓及び脾臓)を得てDiRの含量を可視化した。また、脳組織内部のeHNP-A1の分布を視角化するために、得た脳組織を10μm切片で極低温保管し、DAPI-containing antifade mounting medium(H-1200;Vector Laboratories、Burlingame、CA、USA)を用いてDAPIで染色した。これを共焦点顕微鏡(confocal microscope、Zeiss LSM 780)を利用して画像化した。
実施例2において製作した血液-脳障壁(BBB;blood-brain barrier)模写の微細流体装置でeHNP-A1の分布を定量化し、物質運搬のメカニズムを確認するために、第1の中心チャンネル(vascular)にeHNP-A1を2時間にかけて培養した。また、eHNP-A1が脳内皮細胞のHDL受容体であるscavenger receptor class B type 1(SR-B1)を利用してHDLの主な輸送メカニズムのうちの1つであるトランスサイトーシス(transcytosis)によって血液-脳障壁を通過するか否かを試験するために、BLT-1(block lipid transporter-1)を処理してSR-B1を遮断した。その後、第1の中心チャンネル(vascular)及び第2の中心チャンネル(perivascular)のそれぞれからサンプリングされたeHNP-A1を含む培養培地の相対的な蛍光強度を測定した。また、eHNP-A1の陽性を示すヒト脳微細血管内皮細胞(HBMEC)、ヒト星状細胞(HA)、及びヒト脳血管周皮細胞(HBVP)の分布に対するFACS(fluorescence-activated cell sorting)分析を行った。

Claims (27)
- 第1の中心チャンネルを含むインサートと、
第2の中心チャンネル、第3の側面チャンネル、第4の側面チャンネル、前記第2の中心チャンネルと前記第3の側面チャンネルを連結する第5の通路チャンネル、及び前記第2の中心チャンネルと前記第4の側面チャンネルを連結する第6の通路チャンネルを含むベースと、
前記インサートと前記ベースとの間に位置する多孔性膜とを含む、微細流体装置。 - 前記インサートが、前記ベースから脱着可能であることを特徴とする、請求項1に記載の微細流体装置。
- 前記ベースは、前記インサートが挿入できる溝を含むことを特徴とする、請求項2に記載の微細流体装置。
- 前記インサートが、第1の締結ヘッドを含み、
前記ベースが、第2の締結ヘッドを含み、
前記第1の締結ヘッドと前記第2の締結ヘッドが互いに引っ掛けられて前記インサートと前記ベースがスナップフィット(snap fit)脱着されることを特徴とする、請求項2に記載の微細流体装置。 - 前記第3の側面チャンネル及び前記第4の側面チャンネルは、前記多孔性膜に接触しないことを特徴とする、請求項1に記載の微細流体装置。
- 前記第1の中心チャンネル、前記第2の中心チャンネル、前記第3の側面チャンネル及び前記第4の側面チャンネルが、液剤の灌流を誘導且つ調節できる入口及び出口を含むことを特徴とする、請求項1に記載の微細流体装置。
- 前記第5の通路チャンネル及び前記第6の通路チャンネルは、前記第2の中心チャンネル、前記第3の側面チャンネル及び前記第4の側面チャンネルに比べてチャンネルの高さが低いことを特徴とする、請求項1に記載の微細流体装置。
- 前記ベースが、第1の流体リザーバ及び第2の流体リザーバをさらに含むことを特徴とする、請求項1に記載の微細流体装置。
- 前記ベースが、前記第3の側面チャンネルと前記第1の流体リザーバを連結する第7の通路チャンネル、及び前記第4の側面チャンネルと前記第2の流体リザーバを連結する第8の通路チャンネルを含むことを特徴とする、請求項8に記載の微細流体装置。
- 前記第7の通路チャンネル及び前記第8の通路チャンネルは、前記第2の中心チャンネル、前記第3の側面チャンネル及び前記第4の側面チャンネルに比べてチャンネルの高さが低いことを特徴とする、請求項9に記載の微細流体装置。
- 前記第7の通路チャンネル及び前記第8の通路チャンネルが、前記第2の中心チャンネルの方向に垂直に形成されたことを特徴とする、請求項9に記載の微細流体装置。
- 前記微細流体装置が、プラスチック、ガラス、金属またはシリコンで製造されることを特徴とする、請求項1に記載の微細流体装置。
- 前記第1の中心チャンネルが、組織障壁細胞を含むことを特徴とする、請求項1に記載の微細流体装置。
- 前記組織障壁細胞が、血管内皮細胞;皮膚細胞;癌細胞;分泌腺細胞;筋肉細胞;及び気管支、大腸、小腸、膵臓または腎臓の上皮細胞からなる群より選ばれる少なくとも1つであることを特徴とする、請求項13に記載の微細流体装置。
- 前記第2の中心チャンネルが、内部組織細胞及びハイドロゲルを含むことを特徴とする、請求項1に記載の微細流体装置。
- 前記内部組織細胞が、星状細胞、周皮細胞、神経細胞、神経幹細胞、グリア細胞、心筋細胞、平滑筋細胞、腸上皮細胞、角化細胞、皮膚線維芽細胞、足細胞及び糸球体内皮細胞からなる群より選ばれる少なくとも1つであることを特徴とする、請求項15に記載の微細流体装置。
- 前記ハイドロゲルが、コラーゲン、ラミニン、ヒアルロン酸、ミネラル、フィブリン、フィブロネクチン、エラスチン、ペプチド、ポリエチレングリコール及びアルジネートからなる群より選ばれる少なくとも1つであることを特徴とする、請求項15に記載の微細流体装置。
- 前記インサートを前記ベースから分離し、前記第1の中心チャンネルに含まれた細胞、前記第2の中心チャンネルに含まれた細胞、及び前記多孔性膜に付着された細胞からなる群より選ばれる少なくとも1つの細胞をそれぞれ分離できることを特徴とする、請求項1に記載の微細流体装置。
- 第1の中心チャンネルを含むインサートと、第2の中心チャンネル、第3の側面チャンネル及び第4の側面チャンネルを含むベースと、前記インサートと前記ベースとの間に位置する多孔性膜とを含み、前記インサートが、前記ベースから脱着可能であることを特徴とする、微細流体装置を組み立てるステップと、
第2の細胞を前記第2の中心チャンネルに注入し、その後、前記微細流体装置をひっくり返して培養するステップと、
第3の細胞を含むハイドロゲルを前記第2の中心チャンネルに注入し、その後培養するステップと、
前記微細流体装置をひっくり返し、その後第1の細胞を前記第1の中心チャンネルに注入して培養するステップと、を含む、微細流体装置内の細胞培養方法。 - 前記第1の中心チャンネル、前記第2の中心チャンネル、前記第3の側面チャンネル及び前記第4の側面チャンネルが、液剤の灌流を誘導且つ調節できる入口及び出口を含み、
前記第2の細胞及び前記第3の細胞を含む前記ハイドロゲルが、前記第2の中心チャンネルの入口から注入され、
前記第1の細胞が、前記第1の中心チャンネルの入口から注入されることを特徴とする、請求項19に記載の微細流体装置内の細胞培養方法。 - 前記第1の細胞が、血管内皮細胞;皮膚細胞;癌細胞;分泌腺細胞;筋肉細胞;及び気管支、大腸、小腸、膵臓または腎臓の上皮細胞からなる群より選ばれ、
前記第2の細胞が、周皮細胞、グリア細胞、心筋細胞、平滑筋細胞、腸上皮細胞、角化細胞、皮膚線維芽細胞、足細胞及び糸球体内皮細胞からなる群より選ばれ、
前記第3の細胞が、星状細胞、神経細胞、及び神経幹細胞からなる群より選ばれることを特徴とする、請求項19に記載の微細流体装置内の細胞培養方法。 - 第1の中心チャンネルを含むインサートと、第2の中心チャンネル、第3の側面チャンネル、第4の側面チャンネル、第1の流体リザーバ、及び第2の流体リザーバを含むベースと、前記インサートと前記ベースとの間に位置する多孔性膜とを含み、前記インサートが、前記ベースから脱着可能であることを特徴とする微細流体装置内の器官または臓器の模写方法であって、
第1の流体を前記第1の中心チャンネルに灌流させるステップと、
第2の流体を前記第1の流体リザーバに注入し、前記第3の側面チャンネル、前記第2の中心チャンネル、前記第4の側面チャンネル及び前記第2の流体リザーバまで順に到達させるステップと、を含む、器官または臓器の模写方法。 - 前記ベースが、前記第2の中心チャンネルと前記第3の側面チャンネルを連結する第5の通路チャンネル、前記第2の中心チャンネルと前記第4の側面チャンネルを連結する第6の通路チャンネル、前記第3の側面チャンネルと前記第1の流体リザーバを連結する第7の通路チャンネル、及び前記第4の側面チャンネルと前記第2の流体リザーバを連結する第8の通路チャンネルをさらに含むことを特徴とする、請求項22に記載の器官または臓器の模写方法。
- 前記第1の流体が、1~10dyne/cm2のせん断応力で灌流させることを特徴とする、請求項22に記載の器官または臓器の模写方法。
- 前記第2の流体の流速が、前記第1の流体リザーバと前記第2の流体リザーバの静水圧の差を利用し12時間以上にかけて1~100μL/hで維持されることを特徴とする、請求項22に記載の器官または臓器の模写方法。
- 下記ステップを含む物質の生理学的、薬理学的または毒性学的効果の分析方法:
a)請求項1に記載の微細流体装置を提供するステップと、
b)前記微細流体装置に物質を導入するステップと、
c)前記物質の生理学的、薬理学的または毒性学的効果を評価するステップ。 - 前記物質が、薬物、毒素または病原体であることを特徴とする、請求項26に記載の分析方法。
