Abstract
Description

図5は、マイクロウェルプレート、マイクロウェルチップ、ナノ液滴アレイ、または個々のアブレーションされたサブサンプルを受容し、異なる空間的に分離された区画に別々のサブサンプルを維持することができる他の構造などの空間的分化した媒体を含むレシーバ416の例を示している。レシーバ416は、少なくとも2つの軸方向において、より好ましくは、3つの軸方向すべてに選択的に移動することができるレシーバステージ417に取り付けられ得る。
上記のように、下流分析器を利用して、レシーバによって収集されたアブレーションされたサブサンプルをさらに処理および/または分析し得る。所望の処理および/または分析のタイプに応じて、下流分析器は、例えば、1つ以上のPCR機、配列決定機、光学分光計、核磁気共鳴(NMR)分光計、質量分析計、クロマトグラフィーデバイス、遠心分離機、電気泳動デバイス、放射性標識および放射性標識検出デバイス、他の分析生化学デバイス、またはそれらの組み合わせを含み得る。
本明細書に記載のデバイスおよびシステムは、様々な画像化および/またはアブレーションプロセスを実行するように構成され得る。図9は、レシーバ上のアブレーションされたサブサンプルの位置を、アブレーションイベントに、およびサンプルスライド上の標的関心領域に、相関させるための例示的な方法800を示している。上記のように、サンプルスライド上の関心領域は、上流の選別/フローデータとさらに相関し得る(例えば、図3および関連する説明を参照)。方法800は、システム100に関連して上述したコントローラ140などの、システムの特定の構成要素に通信可能に結合されたコントローラを使用して実行され得る。

前記レシーバによって受容された前記アブレーションされた材料の少なくとも一部分を受容して分析するように構成された分析器と、を備える、システム。 - 前記分析器が、PCR機、配列決定機、光学分光計、および質量分析計のうちの1つ以上を備える、請求項24に記載のシステム。
- 前記質量分析計が、飛行時間(TOF)質量分析計、オービトラップ質量分析計、線形イオントラップ質量分析計、四重極質量分析計、四重極イオントラップ質量分析計、磁気セクター質量分析計、またはフーリエ変換イオンサイクロトロン共鳴(FTICR)質量分析計として構成されている、請求項25に記載のシステム。

前記レシーバ上の前記アブレーションされた材料の少なくとも一部分を捕捉することと、を含む、方法。 - 前記アブレーションされた材料の少なくとも一部分を前記レシーバから前記分析器に通過させること、をさらに含む、請求項33に記載の方法。
- 前記アブレーションが、周囲雰囲気中で行われる、請求項33又は34に記載の方法。

- 複数のレーザパルスが前記サンプルに印加され、複数のアブレーションされたサブサンプルが、非重複空間的分化された位置で前記レシーバに収集され、好ましくは、レーザパルス周波数、レーザパルスエネルギーレベル、またはレーザパルス深度のうちの1つ以上が、前記複数のレーザパルスにわたって動的に変化する、請求項33~41のいずれか一項に記載の方法。
- レーザパルス強度が、前記関心領域を覆っている前記サンプル中の材料を除去するために初期の高レベルに設定され、次いで、前記関心領域の前記少なくとも一部分のアブレーションのために、より低いレベルに設定される、請求項42に記載の方法。
- 空間分解能値が、前記サンプルの前記画像を取得するために利用され、前記レーザ光が、前記サンプルの上面から測定され、かつ前記空間分解能値のR倍以下である、深度であって、Rが、約5~約30の値である、深度で集束される、請求項33~43のいずれか一項に記載の方法。
