Abstract
Description
物品を洗剤組成物で処理する工程と、
物品を本発明の組成物で処理する工程と、を含む。
前駆材料を提供する工程と、複数の孔を有する分配器を提供する工程と、当該前駆材料を当該孔に通過させる工程と、当該分配器の下に移動コンベアを提供する工程と、当該移動コンベア上に当該前駆材料を堆積させる工程と、当該前駆材料を冷却して、複数の粒子を形成する工程とを含み、前駆材料は好ましくはポリエチレングリコールを含み、当該ポリエチレングリコールが、約2000~約13000の重量平均分子量を有し、当該前駆材料は、当該前駆材料の約0.0001重量%~約5重量%の細菌内生胞子を含む細菌組成物を含み、当該前駆材料が、約70℃未満の温度で提供される。驚くべきことに、この方法に供されたときに、内生胞子が発芽しないことが見出された。
一部のグラム陽性細菌は、2段階の生活環を有する。その生活環中、栄養不足状態に応答するなどの特定の条件下で成長している細菌は、胞子又は内生胞子形成につながる精巧な発生プログラムを実行され得る。細菌胞子は、興味深い形態的及び機械的特性を有する、生化学的に複雑な構造として組み立てられた、約60個の異なるタンパク質からなるコートによって保護される。そのタンパク質コートは、強剛性を提供する静的構造と見なされ、溶菌酵素などの外因性の大きな毒性分子を除外するためのふるいとして主に作用する。胞子は、極端な環境条件に対して非常に耐性があるため、種の長期生存において重要な役割を果たす。胞子はまた、長年にわたって、代謝的に休眠状態でありつづけることができる。栄養細胞から細菌胞子を得るための方法は、当該分野で周知である。一部の例では、栄養細菌細胞は液体培地で増殖させられる。後期対数増殖期又は初期定常期から、細菌は胞子形成を開始し得る。細菌が胞子形成を終了すると、例えば遠心分離を使用することにより、それらの胞子を培地から得ることができる。任意の残りの栄養細胞を殺傷又は除去するために、様々な方法が使用され得る。様々な方法を使用して、細胞破片及び/又は他の材料又は物質から、胞子を精製することができる。細菌胞子を産生するための一部の例示的な方法は、本開示の実施例1に記載されている。細菌胞子は、例えば、様々な技術、相造影顕微鏡法、自動走査顕微鏡法、高解像度原子力顕微鏡法、又は耐熱性法を使用して、栄養細胞から分化され得る。
本明細書では、「非発芽性」担体とは、内生胞子の発芽に寄与しない担体を意味する。
粒子は、染料を含んでもよい。染料としては、洗濯洗剤又は布地柔軟剤に典型的に使用される染料が挙げられ得る。布地処理組成物は、布地処理組成物の粒子の約0.1重量%未満、代替的に約0.001重量%~約0.1重量%、代替的に約0.01重量%~約0.02重量%、代替的にこれらの組み合わせ、及び前述の任意の範囲内の任意の百分の1パーセントの又は任意の百分の1パーセントの範囲の染料を含んでもよい。好適な染料の例としては、限定されないが、LIQUITINT PINK AM、AQUA AS CYAN 15、及びVIOLET FL(Milliken Chemical社製)が挙げられる。ユーザーが異なる香りを有する粒子どうしの区別をするのを助けることを目的として、染料を用いることは実用的であり得る。
担体に加えて、粒子は、0.1重量%~約20重量%の香料を更に含み得る。香料は、非カプセル封入香料、カプセル封入香料、香料送達技術によってもたらされた香料、又は何らかの他の方法でもたらされた香料であってよい。香料については、概説的に、米国特許第7,186,680号の第10欄56行~第25欄22行に記載されている。粒子は、非カプセル封入香料を含む場合もあり、例えば香料マイクロカプセルなどの香料担体を本質的に含まない。粒子は、香料担体材料(及びそれに含まれる香料)を含み得る。香料担体材料の例が、米国特許第7,186,680号の第25欄23行~第31欄7行に記載されている。香料キャリア材料の特別な例は、シクロデキストリン及びゼオライトを含んでもよい。
本発明の粒子は、国際公開第2017/156095(A1)号の18頁14行目~21頁3行目に記載されているものと同様の方法を使用して製造することができる。
A.複数の粒子を含む組成物であって、当該粒子が、
当該粒子の約40重量%~約99重量%の担体と、
約0.0001重量%~約5重量%の、細菌内生胞子を含む細菌組成物と、を含み、
当該粒子の各々が、約1mg~約5000mgの質量を有する、組成物。
B.当該粒子の各々が、約5mg~約200mgの質量を有する、段落Aに記載の組成物。
C.その組成物が、洗濯添加剤組成物である、段落A又はBに記載の組成物。
D.その組成物が、22.6mmの公称ふるい開口サイズを有するふるいを通過し、0.841mmの公称ふるい開口サイズを有するふるい上に保持される粒子を含む、段落A~Cに記載の組成物。
E.当該担体が、水溶性ポリマーである、段落A~Dのいずれか1つに記載の組成物。
F.当該粒子が、それらの粒子の45重量%超~約99重量%の上記担体を含む、段落A~Eのいずれか1つに記載の組成物。
G.当該担体及び当該細菌内生胞子が、実質的に均一に互いと混合される、段落A~Fのいずれか1つに記載の組成物。
H.当該内生胞子が粒子形態である、段落A~Gのいずれか一項に記載の装置。
I.当該粒子が、当該粒子の約0.5重量%~3重量%未満の細菌内生胞子を含む、段落A~Hのいずれか1つに記載の組成物。
J.当該粒子が、約0.1重量%~約20重量%の香料を含む、段落A~Iのいずれか1つに記載の組成物。
K.当該担体が、ポリエチレングリコールであり、当該ポリエチレングリコールが、約2000~約13000の重量平均分子量を有する、段落A~Jのいずれか1つに記載の組成物。
L.当該粒子が、約0.1重量%~約10重量%のカプセル封入香料を含む、段落A~Kのいずれか1つに記載の組成物。
M.当該細菌内生胞子が、少なくとも、バチルス属由来の細菌を含む、段落A~Lのいずれか1つに記載の組成物。
N.当該複数の粒子が、約10mg未満の質量しか有さない粒子を実質的に含まない、段落A~Mのいずれか1つに記載の組成物。
O.当該担体が、スクロース、スクロースベントナイト、水溶性有機アルカリ金属塩、水溶性無機アルカリ土類金属塩、水溶性有機アルカリ土類金属塩、水溶性炭水化物、水溶性ケイ酸塩、水溶性尿素、デンプン、粘土、水不溶性ケイ酸塩、クエン酸カルボキシメチルセルロース、脂肪酸、脂肪族アルコール、水素添加獣脂のグリセリルジエステル、グリセロール、ポリエチレングリコール、及びこれらの組み合わせからなる群から選択される、段落A~Nのいずれか1つに記載の組成物。
P.洗濯物品を処理するための方法であって、
粒子を提供する工程であって、それらの粒子が、
当該粒子の約40重量%~約99重量%の担体と、
約0.0001重量%~約5重量%の、細菌内生胞子を含む細菌組成物と、を含む工程と、
ある投入量の当該粒子を投与カップ内に入れる工程と、
その投入量の当該粒子を、洗濯機に供給する工程と、を含み、
当該粒子の各々が、約1mg~約5000mgの質量を有する、方法。
Q.当該担体が、ポリエチレングリコールであり、当該ポリエチレングリコールが、約2000~約13000の重量平均分子量を有する、段落Pに記載の方法。
R.粒子を形成するための方法であって、
前駆材料を提供する工程と、
複数の孔を有する分配器を提供する工程と、
当該前駆材料をその孔に通過させる工程と、
当該分配器の下に移動コンベアを提供する工程と、
当該前駆材料をその移動コンベア上に堆積させる工程と、
当該前駆材料を冷却して複数の粒子を形成する工程と、を含み、
当該前駆材料がポリエチレングリコールを含み、そのポリエチレングリコールは、約2000~約13000の重量平均分子量を有し、
当該前駆材料が、その前駆材料の約0.0001重量%~約5重量%の、細菌内生胞子を含む細菌組成物を含み、
当該前駆材料が、約70℃未満の温度で提供される、方法。
細菌から胞子を調製するにあたって、その細菌を、液体培養で対数的に増殖させた。対数培養物中の炭素、窒素、及び/又はリンが制限していく(例えば、対数増殖の後期)につれて、栄養細胞は胞子形成を開始した。培養物中に追加の胞子が形成されないと推定されるまで培養をインキュベートし続けた。場合によっては、胞子は生産高である培養物から得た。次いで、培養物を遠心分離して胞子をペレット化し、残りの細胞及び破片を得た。これらの胞子ペレットを水に懸濁させて洗浄し、再び水に懸濁し、胞子懸濁液をチューブ内に沈降させ、3つの可視層を概ね形成した。サンプルの顕微鏡検査を使用して、明るい相の胞子の存在を、所望の純度(99%超の明るい相の胞子)で確認した。純度が達成されなかった場合、所望の純度に達するまで水洗浄を繰り返した。
バチルス胞子粉末を含有する少量のバッチのPEG8000粒子を製造するために、以下の手順に従った。約100gのPEG(Pluriol E 8000)を秤量し、オーブン内において80℃で溶融させた。溶融物に、予め秤量しておいた0.01gのバチルス胞子粉末を添加し(組成物1に対して添加し、組成物2に対しては胞子を添加しなかった)、1.0×108コロニー形成単位(CFU)のバチルス胞子を送達した。Flacktek高速ミキサーを使用して、3500rpmで30秒間混合した。滅菌条件下で、かつ安全キャビネット内で、高温PEG溶融物と胞子との混合物を、滅菌スパチュラを使用して粒子鋳型に移し、鋳型上に均一に広げた。組成物混合物を室温まで約5分間かけて冷却して、固体粒子を形成した。固化した粒子を鋳型から掻き取り、必要になるまで保管のためにこれを滅菌容器に移した。
熱ショック法及び胞子数え上げ手順を使用して、機械的高速混合及びビーズ鋳型上で造粒する間の、高温PEG8000溶融物中での作製プロセス中の胞子の生存率及び生存能力を評価した。
リン酸緩衝液中における胞子粉末の直接適用
異なる品目に直接適用された胞子粉末による悪臭除去を、以下の表に示す。
洗濯プロセスで使用される本発明の粒子の、バスタオル上の悪臭除去について、以下の表に示す。
Pyrex結晶粒子、非イオン性界面活性剤粒子、及び酢酸ナトリウム粒子を、実施例2のPEG8000粒子と同様に作製した。洗濯プロセスで使用される粒子の、バスタオル上の悪臭除去について、以下の表に示す。
Claims (14)
- 複数の粒子を含む組成物であって、前記粒子が、
前記粒子の少なくとも40重量%の非発芽性担体と、
前記粒子の約0.0001重量%~約5重量%の、細菌内生胞子を含む細菌組成物と、
を含み、
前記粒子の各々が、約1mg~約5000mg、好ましくは約5mg~約200mgの質量を有する、組成物。 - 前記細菌組成物が、細菌内生胞子を、粒子1gあたり約1×102~約1×109CFU、好ましくは粒子1gあたり約1×103~約1×106CFUのレベルで含む、請求項1に記載の組成物。
- 前記内生胞子がバチルス(Bacillus)属由来の細菌を含む、請求項1又は2のいずれか一項に記載の組成物。
- 前記内生胞子が、バチルス・ズブチリス(Bacillus subtilis)、バチルス・アミロリクエファシエンス(Bacillus amyloliquefaciens)、バチルス・リケニフォルミス(Bacillus licheniformis)、バチルス・メガテリウム(Bacillus megaterium)、バチルス・プミルス(Bacillus pumilus)、及びそれらの混合物からなる群から選択される細菌を含む、請求項1又は2のいずれか一項に記載の組成物。
- 前記担体が、スクロース、ベントナイト、水溶性有機アルカリ金属塩、水溶性無機アルカリ土類金属塩、水溶性有機アルカリ土類金属塩、水溶性炭水化物、水溶性ケイ酸塩、水溶性尿素、デンプン、クレイ、水不溶性ケイ酸塩、クエン酸、グリコール酸、カルボキシメチルセルロース、脂肪酸、脂肪族アルコール、水素添加獣脂のグリセリルジエステル、グリセロール、ポリエチレングリコール、ポリビニルアルコール、及びこれらの組み合わせからなる群から選択される、請求項1~4のいずれか一項に記載の組成物。
- 前記担体が水溶性ポリマーを含む、請求項1~5のいずれか一項に記載の組成物。
- 前記粒子が、約0.1重量%~約20重量%の香料を含む、請求項1~6のいずれか一項に記載の組成物。
- 前記複数の粒子が、約10mg未満の質量を有する粒子を実質的に含まない、請求項1~7のいずれか一項に記載の組成物。
