Ki決定のためのインビトロ結合アッセイ AV-133対AV-133-D6(Ia)

ヒトAD脳ホモジネートのAβ凝集体についてのインビトロ結合アッセイ

18F-III-3(18F-FPBM-D12)の放射性標識
インビトロ結合アッセイ
セロトニントランスポーター(共通の親細胞株LLC-PK1で発現したLLC-SERT)の膜ホモジネートを調製し、結合アッセイに使用した。競合結合アッセイを0.25mLの最終容量で実施した。膜懸濁液のアリコートを、50mMのTris-HCl(pH7.4)、120mMのNaCl及び0.1%のウシ血清アルブミン、0.2nMの[125I]DAM、及び8~10濃度(10-13~10-7M)の競合薬物と混合し
た。非特異的結合は10μMのシタロプラムで定義した。インキュベートを室温で60分間行い、結合したリガンドを、1%のポリエチレンイミン(SIGMA,St.Louis,MO)で予備浸漬したガラス繊維フィルターで回収し、ガンマカウンター(Wizard2,Perkin Elmer)でカウントした。非線形最小二乗曲線適合プログラムLIGANDを使用してデータを解析してIC50を決定し、[125I]IDAMのKdとして0.2nMを使用してCheng-Prusoff方程式によってKiを計算した。
ラットにおける生体内分布
各群3匹のラットを各生体内分布研究に使用した。イソフルラン麻酔下で、20μCiの放射性トレーサーを含有する0.2mLの生理食塩水溶液を大腿静脈に注射した。イソフルラン麻酔下で心臓切除によって、示された時点でラットを屠殺した。対象となる臓器を取り出し、計量し、放射能をカウントした。臓器当たりのパーセント線量は、組織カウント数を、同時に測定された初期線量の1%(注射された物質の100倍希釈アリコート)のカウント数と比較することによって計算した。ラットにおける局所的脳分布を放射性トレーサーの静脈注射後に測定した。異なる脳領域(皮質、線条体、海馬、小脳及び視床下部)からの試料を摘出し、計量し、カウントした。各試料のパーセント線量/gは、試料カウント数を上述した希釈初期線量のカウント数と比較することによって計算した。比率は、各領域のパーセント線量/gを小脳のそれで除することによって計算した。小脳にはわずかな量のSERTしか存在しないので、標的対非標的結合の比を計算するための参照領域として小脳を使用した。
ニン取り込みの遮断が指示される特定の療法に有用であり得る。

有効量の上記項1~35のいずれか1項に記載の化合物またはその薬学的に許容可能な塩を対象に投与すること;及び
前記対象中の前記化合物の放射能のパターンを検出すること、を含む前記方法。
Claims (29)
- R2は、メチルであり、1つ以上の重水素原子で任意に置換されており;
mは、3である、請求項1または2に記載の化合物、またはその薬学的に許容可能な塩。 - R2は、CD3である、請求項1~3のいずれか1項に記載の化合物、またはその薬学的に許容可能な塩。
- Yは、-[OCD2CD2]3-である、請求項1~4のいずれか1項に記載の化合物、またはその薬学的に許容可能な塩。
- A1は、Nであり、Xは、18Fである、請求項6に記載の化合物、またはその薬学的に許容可能な塩。
- A1は、CHであり、Xは、18Fである、請求項6に記載の化合物、またはその薬学的に許容可能な塩。
- R3は、1つ以上の重水素原子で置換されたメチルである、請求項9~12のいずれか1項に記載の化合物、またはその薬学的に許容可能な塩。
- R3は、11CD3であり、Xは、水素である、請求項9~13のいずれか1項に記載の化合物、またはその薬学的に許容可能な塩。
- R3は、CD3であり、Xは、18Fである、請求項9~13のいずれか1項に記載の化合物、またはその薬学的に許容可能な塩。
- nは、3である、請求項19に記載の化合物、またはその薬学的に許容可能な塩。
- Yは、-(CD2)3-である、請求項19または20に記載の化合物、またはその薬学的に許容可能な塩。
- R4及びR5は、それぞれ独立して、水素または1つ以上の重水素原子で任意に置換されたメチルである、請求項19~21のいずれか1項に記載の化合物、またはその薬学的に許容可能な塩。
- R4及びR5は、それぞれ独立して、水素またはCD3である、請求項19~22のいずれか1項に記載の化合物、またはその薬学的に許容可能な塩。
- 各々の指定された重水素原子についての重水素濃縮は、少なくとも約50%、好ましくは少なくとも約80%、より好ましくは少なくとも約90%、最も好ましくは少なくとも約95%である、請求項1~25のいずれか1項に記載の化合物、またはその薬学的に許容可能な塩。
