Abstract
Description
i)幹細胞またはナノベシクルで発現するタンパク質でありうる。例えば、前記トランスメンブレンタンパク質は、CD86のようなエキソソームマーカー、CD105およびCD271のような間葉系幹細胞マーカーなどを使用できる。
ii)タンパク質のN末端とC末端が細胞膜を間にして反対方向に向かって位置しなければならない。
(PREY発現のためのプラスミド設計およびクローニング)
細胞膜でPREYを外部に発現させ、局所化するプラスミドは、外部側N末端-プロモーター-シグナルペプチド-PREY-V5-TMP-GFP-内部側C末端で構成される。シグナルペプチドは、Signal peptide F(BKU002587,Korea Human Gene Bank,Republic of Korea)またはSignal peptide R(BKU008396,Korea Human Gene Bank,Republic of Korea)を使用する。プラスミド配列で、i)シグナルペプチドは、PREYペプチドの細胞膜外部への局所化を誘導し;およびii)V5タグおよびGFPは、PREYの位置および発現レベルをモニタリングする。発現ベクターは、NEB Gibson Assembly kit(New England Biolabs,MA)を使って製造メーカーの説明書に従ってクローニングした。シグナルペプチドおよびトランスメンブレンタンパク質の各タイプは、次の鋳型を用いたPCRを通じて別に増幅した:Signal peptide F(BKU002587,Korea Human Gene Bank,Republic of Korea)、Signal peptide R(BKU008396,Korea Human Gene Bank,Republic of Korea)、CD86、CD105および切断されたCD271(LNGFR)に対しては、NGFR(Addgene plasmid #27489;Addgene,MA)。ベクター成分は、プラスミド合成(Macrogen,Republic of Korea)と共にCas9-digested p3S-Cas9-HN(Addgene plasmid #104171)バックボーンに挿入された。この手順に従ってPCR増幅およびギブスンクローニングを行った。すべてのプライマーおよびプラスミドは、表1および2に羅列される。
PREYトランスフェクション効率は、2種のMSCタイプ(ASCおよびBMSC)と比較してトランスフェクション後1日に定量的分析と共にFACSCanto(BD Bioscience,CA)を使ってフローサイトメトリー(Flow cytometry)を通じてトランスメンブレンタンパク質の試験候補物質(CD86、CD105およびCD271)の中で比較した。したがって、細胞は、抗v5タグ1次抗体(ab27671,Abcam,MA)およびAlexa Fluor 647コンジュゲートされた2次抗体(Jackson Immuno Research,PA)で免疫染色された。プラスミド用量依存性細胞死滅を評価するために、トランスフェクションされたASCおよびBMSCをトランスフェクション後30分に採取し、Alexa Fluor 488コンジュゲートされたAnnexin V(Thermo Fisher Scientific,CA)で免疫染色し、フローサイトメトリーを行った。また、トランスフェクション後1日目にトリパンブルー染色を通じて生存細胞数を計数した。
トランスフェクション後3日にヒトASC(Promocell,Germany)、BMSC(Lonza,Switzerland)およびPMSCをPBSで2回洗浄し、0.25%trypsin/EDTAで脱着させた。次に、ナノベシクルを生成するために、PBSに溶かした細胞懸濁液1×106cells/mLを押出キット(Avanti Polar Lipids,AL)を使って10μm、5μmおよび400nm気孔サイズのポリカーボネートメンブレンフィルター(Whatman,UK)を順次に変えながら6回押出させた。15,000gで30分間遠心分離してNVを収集した。次に、ペレットをPBSに再懸濁させ、0.20μm注射器フィルター(Avantec,Japan)を用いてろ過し、使用時まで-70℃で保管した。PMSC-NVのサイズおよび形態は、透過電子顕微鏡(TEM;JEM-F200,JEOL,Japan)および動的光散乱(DLS;ELS-1000ZS,Otsuka Electronics,Japan)により測定された。
RAW264.7細胞を24ウェルプレートにシーディングした(5×105cells/well)。RAW264.7細胞の前炎症性活性化は、24時間の間LPS(Sigma-Aldrich;100ng/mL)処理後に24時間の間ASC-NVまたはBMSC-NV(10μg/mL)処理を通じて誘導された。細胞性吸収を視覚化するために、RAW264.7細胞およびNVをそれぞれDiOおよびDiI(Invitrogen,CA)で標識し、共焦点顕微鏡(LSM780;Zeiss,Germany)で映像化した。qRT-PCR分析のために、NV処理後に24時間に細胞を採取した。IL-10、IL-1β、IL-6およびTNF-αのプライマー配列は、表3に羅列されている。マウス炎症抗体アレイ(ab133999,Abcam)を用いたサイトカイン分析のために、製造メーカーの説明書に従って細胞上澄み液を採取した。MSC-NVの抗食菌効果は、VybrantTM Phagocytosis Assay Kit(V6694,Molecular Probes,OR)を使って製造メーカーの説明書に従って測定した。イメージは、共焦点顕微鏡を通じて得、蛍光強度は、Varioskan(登録商標) LUX multimode microplate reader(Thermo Fisher Scientific,MA)を使って測定した。
iMAEC(ATCC,VA)を24ウェルプレートにシーディングしてから(1×105cells/well)、24時間の間LPS(100ng/mL)を処理した。次に、追加で24時間の間ASC-NV、BMSC-NVまたはPMSC-NV(10μg/mL)を処理した。細胞吸収を視覚化するために、iMAECおよびNVをそれぞれDiOおよびDiIで標識し、共焦点顕微鏡を用いて映像化した。qRT-PCR分析のために、NV処理後に24時間にiMAECを採取した。E-selectin、ICAM-1およびVCAM-1のプライマー配列は、表3に羅列した。iMAECは、VCAM-1抗体(ab134047,Abcam)で免疫染色し、ImageJを用いて定量的分析と共に共焦点顕微鏡を用いて映像化した。NV(10μg/mL)およびCyA(25μg/mL;Santa Cruz Biotechnology,CA)をHUVEC(Lonza)に処理し、マトリゲル(BD Biosciences,MA)でそれぞれ2時間および24時間の間培養して、抗血管形成および脈管破裂に対するプレEC修復効果を測定した。イメージは、共焦点顕微鏡を用いて得、ImageJを用いて定量化した。
すべての動物研究は、延世大学校医科大学の施設内の動物管理および使用委員会(IACUC)が承認した手続き(2018-0044)によって行われた。手術手続きは、6週齢の雄Balb/c(Orient Bio Inc,韓国)またはKOR-ApoE(shl)(SLC,Japan)マウスで行われた。PCL手術のために、ザイラジン(10mg/kg)およびゾレチル(50mg/kg)混合物の腹腔内注射を通じて麻酔させた。首のひげを剃り、ベタジンを用いて消毒した。次に、中間線切開を行った(5mm)。LCA露出後、LCAの4種の枝のうち3個(ECA、ICA、OA)を10-0ポリアミド縫合糸で結紮し、STAは結紮させなかった状態にした。次に、切開部を6-0シルク縫合糸で縫い合わせた。それから、マウスをモニタリングし、動脈硬化性食事(Research Diets,NJ)を供給し、結紮後3日にMSC-NV、PMSCまたはPMSC-NVを静脈投与した。
渦流部位のインビボPMSC-NVターゲッティング効率は、試験群の注射後24時間にIVISイメージング(PerkinElmer,WA)および組織学的分析を通じてマウスPCLモデルで測定した。MSCおよびNV群は、VivoTrack 680(PerkinElmer)で30分間標識され、PCLモデルに注射された。次に、イソフルランで呼吸麻酔下でIVISイメージングを行った。それから、マウスを犠牲にさせ、これらのLCAs、RCAsおよび主要臓器を採取して、エクスビボIVISイメージングおよび組織学的分析を行った。LCAおよびRCAの組織セクションは、抗フィラミンA抗体(ab51217,Abcam)で免疫染色し、相当する蛍光強度は、ImageJソフトウェアを使って定量化した。組織セクションは、また、H&Eで染色したり、抗CD68抗体(ab125212,Abcam)および抗VCAM-1抗体(ab134047,Abcam)で免疫染色した。新生血管内膜(Neointima)構造因子(新生血管内膜対新生血管内膜+ルーメンの面積比率、新生血管内膜対培地の面積比率および新生血管内膜面積)または相当する蛍光強度をImageJを使って定量的に分析した。
PCL手術は、25~30kg重量の雌ヨークシャーブタ(XP bio,Republic of Korea)で以前の研究に従って行った。ブタに前処置(premedication)としてアトロピン(0.04mg/kg)、ザイラジン(2mg/kg)およびアザペロン(2mg/kg)を筋肉内注射した。マウスは、アルファキサン(1mg/kg)で麻酔し、手術中に2%イソフルランの器官内挿管によりこの状態に維持された。首は、ベタジンを使って消毒し、次に、中間線皮膚切開を行った。滅菌されたステンレススチール棒(外径=0.9mm)をLCAにスペーサーとして配置し、5-0シルク縫合糸とともに結紮した。棒を順次に除去し、縫い合わせて、切開を防いで、頸動脈の80%閉塞を誘発した。RCAは、正常群として結紮なしで放置された。血流パターンは、超音波(S22V;SonoScape Medical Corp.,China)により観察された。Vivotrack680標識されたMSC-NVまたはPMSC-NVを結紮後3日に耳静脈(1mg/pig)を通じて静脈内注射し、ブタを注射後1日または21日に犠牲にさせた。エクスビボIVISおよび組織学的分析のためにRCA、LCAおよび大動脈弓を採取した。これらの組織セクションは、抗フィラミンA抗体および抗CD31抗体(sc-1506,Santa Cruz Biotechnology,CA)で免疫染色され、次に、ImageJ分析と共に蛍光イメージングを行った。
以前に記述された通り(Sei,Y.J.et al.Sci Rep 7,10019,2017)、微小流体装置は、ソフトリソグラフィーによりポリジメチルシロキサン(PDMS,Dow Corning,MI)で製造し、ガラスカバースリップ(VWR,PA)と結合させてポリスチレンボックス(Ted Pella Inc.,CA)に入れた。装置は、70%エタノールで滅菌し、PBSで洗浄した。次に、50μg/mLのコラーゲンI(Corning,MA)で37℃で1時間の間チャネルをコーティングした。ヒト冠状動脈内皮細胞(hCAEC;Lonza)またはヒト大動脈内皮細胞(hAEC;Lonza)を12時間の間チャネルに(2×107cells/mLで)シーディングした。各装置の排出口は、PhD Ultraシリンジポンプ(Harvard Apparatus,MA)に連結されて、正常な培地流れおよび渦流が形成されるようにした。正常な層流を形成するために、22.5μl/minの培地を10dyne/cm2のせん断応力で灌流させた。渦流は、それぞれ22.5μl/min(10dyne/cm2)および20μl/min(9dyne/cm2)の流速で培地を注射および除去する反復サイクルを通じて生成した。ヒトECをどちらか一方の流れタイプに露出させた後、NV(10μg/mL)を37℃で1時間の間チャネルに灌流させた。各試験群においてヒトECへのNV吸収は、ImageJを使って定量的に分析された。
トータルRNAは、製造メーカーの説明書に従って1mLのTRIzol試薬(Invitrogen)を使って各サンプルから抽出された。RNAをジエチルピロカーボネート(DEPC)ウォーターに溶かし、AccuPower CycleScript RT Premix(Bioneer,Republic of Korea)を使ってcDNAを合成した。次に、StepOnePlus real-time PCR system(Applied Biosystems,CA)でSYBR Green PCR mix(Thermo Fisher Scientific)でPCRを行った。グリセルアルデヒド3-ホスフェートデヒドロゲナーゼ(GAPDH)をハウスキーピング遺伝子として提供し、各マーカーの遺伝子発現は、相対定量化方法2-ΔΔCtを使って測定した。プライマー配列は、表3に羅列した。溶解したRNAは、miRNAアレイ(GeneChip 4.0 microRNA Microarray;Affimetrix,Japan)を使って製造メーカーの説明書に従ってプロファイリングした。
細胞サンプルを4%パラホルムアルデヒド(Sigma-Aldrich)で10分間固定し、組織サンプルは、10%パラホルムアルデヒドで3日間固定し、両方とも室温で行った。固定されたサンプルは、PBSで洗浄し、パラフィンに固定して、組織セクションを作成した。次に、一連のキシレンおよびエタノール溶液(蒸留水内で100%、95%、80%、70% v/v)を用いてこれらを水和させ、抗原修復のためにペプシン試薬(Sigma-Aldrich)を37℃で30分間処理した。次に、組織セクションにブロッキング溶液(5%ウシ胎児血清アルブミン(Millipore,MD)+0.3%triton X-100(Sigma-Aldrich))を1時間の間室温で処理した。1次抗体は、抗v5タグ抗体(ab27671,Abcam)、抗VCAM-1(ab134047,Abcam)、抗CD68(ab125212,Abcam)、抗Filamin A(ab51217,Abcam)および抗CD31(sc-1506,Santa Cruz Biotechnology,CA,USA)である。これらの抗体は、1:100で希釈してPBSに処理し、次に、後続2次抗体を1:200で希釈してPBSに処理した。2次抗体は、Alexa Fluor 594にコンジュゲートされた抗マウス抗体、Alexa Fluor 594にコンジュゲートされた抗ウサギ抗体、Alexa Fluor 488にコンジュゲートされた抗ウサギ抗体およびAlexa Fluor 488にコンジュゲートされた抗ヤギ抗体(all from Jackson Laboratories)である。次に、サンプルを載置し、4’,6-diamidino-2-phenylindole(DAPI,Vector Laboratories,CA)を含有するマウンティング溶液で対比染色させて、細胞核を視覚化した。
サンプルをRIPAバッファー(Sigma-Aldrich)で溶解させて総タンパク質を収得し、タンパク質の濃度は、ブラッドフォードアッセイ(Sigma-Aldrich)を使って測定した。タンパク質抽出物を10%(w/v)SDS-PAGEゲルで電気泳動した後、ニトロセルロースメンブレン上に移動させた。メンブレンを5%(w/v)スキムミルクとともにTBST(20mM Tris、0.9%NaCl、0.1%ツイン20、pH7.4)で遮断した後、1次抗体である抗v5タグ(ab27671,Abcam)、抗CD271(345102,Biolegend,CA)、抗GFP(ab32146,Abcam)、抗CD9(ab92726,Abcam)および抗β-アクチン(ab8227,Abcam)とインキュベーションした。次に、2次抗体であるヤギ抗マウスIgG(H+L)-HRPコンジュゲートおよびヤギ抗ウサギIgG(H+L)-HRPコンジュゲートされた抗体(全部Vector Laboratories社製)を製造メーカーの説明書に従って適用した。シグナルは、CL Plus Western Blotting Detection Kit(Amersham Biosciences,UK)を使って製造メーカーの説明書に従って視覚化とLAS-3000イメージ リーダー(Fujifilm,Japan)を使って分析した。
定量的データは、平均±標準偏差(stdev)で表示される。結果は、SigmaPlot 12.0(Systat Software,Inc.,California,USA)を使用したTurkey’s significant difference post hoc testで一元分散分析(ANOVA)により統計的に分析された。
NVが渦流部位をターゲッティングできるようにするために、ターゲッティングペプチドであるPREYをMSC膜(PMSC:PREY-発現ヒトMSC)全体にわたって特異的に設計されたプラスミドを発現させることによって外部に提示した。プラスミドは、N末端でシグナルペプチドとともにPREY細胞外ドメインを発現するように設計されることによって、シグナルペプチドは、MSC膜からPREY細胞外ドメインの外部側局所化を誘導し、切断を通じて誘導シグナルを不活性化させることができる。
PMSC-NVを生産するために、気孔直径を10μm~5μmまでおよび最後に0.4μmに次第に低減しながら、一連のポリカーボネート微細気孔膜を通じてPMSCを順次に押出させた(図1b)。TEM結果(図2e)は、47.2±12.1nmの平均を有するPMSC-NV直径の均一な分布を示す。また、トランスフェクションおよびNV押出中にPREY-ペプチド(GFPおよびv5タグ)の完全な保存およびエキソソーム特性(CD9)は、ウェスタンブロット分析を通じて測定した(図2f、図6)。
MSCおよびMSCに由来する細胞内の内容物が強力な抗炎症効果を示すことはよく知られている。したがって、BMSC-NVおよびASC-NVの抗炎症効果は、PREYトランスフェクション前にテストした(図7a)。マウス単核球/マクロファージ(RAW264.7 cells)にMSC-NVを処理してこれらの炎症反応を抑制した。活性化マクロファージによるMSC-NVの効果的な細胞性吸収は、それぞれ、DiOおよびDiIで細胞およびMSC-NVを視覚化して確認した(図7b)。ASC-NVおよびBMSC-NVは、両方とも活性化マクロファージにより類似に吸収された。
LPS処理による不滅化マウス大動脈内皮細胞(iMAEC)の前炎症性機能障害を活性化させて、MSC-NV処理のEC-修復効果をテストした(図8a)。ASC-NVおよびBMSC-NV両方とも活性化したiMAECへの効果的な内在化は、それぞれ、DiOおよびDiIで標識して視覚化した(図8b)。ASC-NVおよびBMSC-NV間のNV吸収での顕著な差異はなかった。ECは、E-セレクチン、ICAM-1およびVCAM-1を優勢に発現して、機能障害活性化時に炎症細胞を動員する。したがって、iMAECでこれらのマーカーの遺伝子発現をqRT-PCRにより測定すると(図8c)、LPS処理により全部上向き調節されるが、2つのMSC-NVタイプで処理し、しかも、処理しなくても、有意的に下向き調節されて、これは、2つのMSC-NVタイプのEC-修復効果を約束するだけでなく、100ng/mLのLPSによるEC機能障害の効果的な誘導を示唆する。その結果は、また、VCAM-1が最初内皮機能障害マーカーの一つであるから、免疫染色からVCAM-1タンパク質発現によっても支持された(図8d)。プレEC修復効果は、ASC-NV、BMSC-NV、およびPMSC-NVのうち有意差がなくて(図9および図10)、PREYトランスフェクションがMSC-NVの治療的効果を損傷させないことを示唆する。
PMSC-NVの診断治療効率は、マウスPCLモデルで測定した。4個の左側頸動脈(LCA)枝のうち3個である、外部頸動脈(ECA)、内部頸動脈(ICA)および喉頭動脈(OA)を結紮させると(図11)、渦流形成がドップラー超音波イメージングにより確認された(図12)。次に、結紮後3日間MSC-NV、PMSC-NV、およびPMSCを尾静脈を通じて静脈内に注射した。インビボイメージングシステム(IVIS)を使って注射後24時間にこれらの生体内分布を視覚化した(図13aおよび図13b、図14)。PMSC-NVのLCA-ターゲッティング効率は、MSC-NVまたはPMSCよりさらに良く、これは、PREYおよびNVが細胞の肺毛細管捕捉を妨害して、渦流部位を効果的にターゲッティングする相乗作用の役割を示唆する。肺毛細管捕捉は、全身性細胞伝達のための主な妨害物である。本発明者らの以前のプロテオミクス分析でPREYのターゲット分子としてフィラミンAを同定した。RCAおよびLCAを採取し、注射後24時間に免疫染色した(図13c)。フィラミンA発現は、RCAよりLACにおいて有意的にさらに高く、これは、PMSC動員のために渦流部位でのそれの過発現を確認させることである。NVをVivotrack680で標識した試験群のうち、PMSC-NVのみが、フィラミンAとともに明白に一緒に内在化した。
プレ臨床の大きい動物モデルで、ブタPCLモデルを使って以前に確立された方法によって渦流部位に対するPMSC-NVのターゲッティング効率を確認した。簡単に言えば、ステンレススチール棒(直径=0.9mm)をLCA(直径=4.5mm)で堅固に結紮させてから除去し(図17aの上段)、LCAの70~80%閉塞を誘導した(図17bの下段)。結紮されたLCAの遠位部分で渦流の明確な形成がドップラー超音波イメージングにより確認された(図17b)。また、フィラミンAは、RCAよりLCAの内皮で高く発現した(図17c)。Vivotrack680標識されたMSC-NVまたはPMSC-NVを耳静脈を通じて静脈内に投与し、結紮後3日間全身循環させた。RCA、LCAおよび大動脈弓は、それぞれ、NV注入後24時間または21日に正常、誘導渦流および自然渦流部位から組織サンプルとして採取した。自然渦流は、曲率と枝構造に起因して大動脈弓で形成される。IVISイメージは、PMSC-NVが病原性リモデリングの指標であるLCAの結紮点の遠位領域にさらに多く蓄積されるが、RCAでは観察されなかった(図17d、図18の左側)。大動脈弓サンプルでPMSC-NVのIVIS蛍光分布は、MSC-NVと比較して渦流部位とさらに高い相関関係を示し、PREYの効果的なターゲッティング機能を示し(図17f、図18の右側)、MSC-NVよりPMSC-NVによるCD31のさらに大きい共存(図17eおよび図17g)により支持された。
Claims (11)
- 渦流(disturbed blood flow)部位ターゲッティングペプチドを表面に提示する幹細胞由来のナノベシクル。
- 渦流部位ターゲッティングペプチドは、配列番号1~5からなる群から選はれる、請求項1に記載のナノベシクル。
- ナノベシクルは、シグナルペプチド-渦流部位ターゲッティングペプチド-トランスメンブレンタンパク質のコーディング配列が順次に挿入されたベクターでトランスフェクションされた幹細胞に由来するものである、請求項1に記載のナノベシクル。
- トランスメンブレンタンパク質は、CD86、CD105、CD271、CD34およびCD22からなる群から選ばれた一つ以上である、請求項3に記載のナノベシクル。
- 幹細胞は、骨髄、臍帯、臍帯血、胎盤、血液、皮膚、脂肪組織、神経組織、肝、膵胆道、筋肉および羊膜からなる群から選ばれた1種以上の組織由来の幹細胞;間葉系幹細胞;胚性幹細胞;および誘導万能幹細胞のうちいずれか一つである、請求項1に記載のナノベシクル。
- シグナルペプチド-渦流部位ターゲッティングペプチド-トランスメンブレンタンパク質のコーディング配列が順次に挿入されたベクターでトランスフェクションされた幹細胞から渦流部位ターゲッティングペプチドを表面に提示するナノベシクルを得る段階を含む請求項1に記載のナノベシクルの製造方法。
