Abstract
Description
a)不活化全粒子ジカウイルスと、
b)少なくとも約6.5mMの濃度の少なくとも1つの薬学的に許容される緩衝液と、
c)任意選択により、ポリオールと、を含み、
当該液体不活化ウイルス組成物は、好ましくは、アルミニウム塩から選択されるアジュバントを含有せず、当該少なくとも1つの薬学的に許容される緩衝液は、リン酸イオンを含まない、当該液体不活化ウイルス組成物を目的とする。
a)不活化全粒子ジカウイルスと、
b)少なくとも約6.5mMの濃度の少なくとも1つの薬学的に許容される緩衝液と、
c)任意選択により、ポリオールと、を含み、
当該液体不活化ウイルス組成物は、好ましくは、アルミニウム塩から選択されるアジュバントを含有せず、
当該少なくとも1つの薬学的に許容される緩衝液は、リン酸イオンを含まない、不活化全粒子ジカウイルスを安定化するための当該使用を目的とする。
a)不活化全粒子ジカウイルスと、
b)薬学的に許容される緩衝液であって、当該緩衝液は、リン酸緩衝液ではなく、当該緩衝液の濃度は、少なくとも6.5mMである、緩衝液と、
c)任意選択により、ポリオールと、を含み、
当該不活化ウイルス組成物は、アルミニウム塩から選択されるアジュバントを含有せず、当該方法は、次のステップ:
ステップ1.1つ以上の非ヒト細胞から得られた上清からジカウイルス調製物を分離すること、
ステップ2.ジカウイルス調製物を精製すること、
ステップ3.ウイルス調製物を不活化すること、
ステップ4.ジカウイルス調製物を薬学的に許容される緩衝液に移してジカウイルス原薬を得ること、を含む、当該方法を目的とする。
a)本発明に係る不活化ウイルス組成物と、
b)水酸化アルミニウムなどのアジュバントと、
を含む、液体ワクチンに関する。
ステップ1.上記の本発明に係る不活化ウイルス組成物を提供すること、
ステップ2.不活化ウイルス組成物に、アジュバントであって、好ましくはアルミニウム塩であるアジュバントと、任意選択により更なる薬学的に許容される緩衝液とを添加することを含む、方法を目的とする。

本開示の他の態様は、本明細書に記載される配列番号1の位置98、または配列番号1の位置98に対応する位置にトリプトファンからグリシンへの置換である変異を含むジカウイルスなどの精製された不活化全粒子ウイルスを含有する、ジカウイルス不活化ウイルス組成物に関する。いくつかの実施形態では、不活化ウイルス組成物は、配列番号1の位置98、または配列番号1の位置98に対応する位置にTrp98Gly変異を含む、精製された不活化全粒子ジカウイルスを含み、ジカウイルスは、PRVABC59株に由来する。いくつかの実施形態では、不活化ウイルス組成物は、配列番号1の位置98、または配列番号1の位置98に対応する位置にTrp98Gly変異を含む、精製された不活化全粒子ジカウイルスを含み、ジカウイルスは、配列番号2に従うゲノム配列を含むPRVABC59株に由来する。一実施形態では、不活化ウイルス組成物は、プラーク精製されたクローンジカウイルス分離株を含有する。

ジカウイルスの純度は、サイズ排除クロマトグラフィーによって決定することができる。本開示の特定の実施形態は、サイズ排除クロマトグラフィーにおけるジカウイルスのメインピークが曲線下総面積の85%を超えることによって決定される、少なくとも85%の純度の不活化全粒子ジカウイルスを含む、不活化ウイルス組成物に関する。特定のそのような実施形態では、ジカウイルスは、サイズ排除クロマトグラフィーにおけるジカウイルスのメインピークが曲線下総面積の90%を超えることによって決定される、90%の純度であり得る。特定のそのような実施形態では、ジカウイルスは、サイズ排除クロマトグラフィーにおけるジカウイルスのメインピークが曲線下総面積の95%を超えることによって決定される、95%の純度であり得る。

本発明に係る不活化ウイルス組成物は、一般に、ワクチンの製造に使用される中間組成物を指す。本発明の特定の更なる態様は、上記の不活化ウイルス組成物から得られる/得ることができる液体ワクチン(または疾患もしくは状態の治療もしくは予防における使用に好適な組成物、特にジカウイルス感染症の治療もしくは予防における使用に好適な組成物)に関する。ワクチンは、アジュバントを含有し、不活化ウイルス組成物とは異なる緩衝液及び賦形剤の濃度を有し得る。
a)先行請求項のいずれか1項に記載の不活化ウイルス組成物と、
b)水酸化アルミニウムなどのアジュバントと、
を含む、液体ワクチンに関する。
本発明に係るワクチンは、1つ以上のアジュバントと組み合わせた、少なくとも1つのジカウイルスに由来する1つ以上の抗原を含む。
特定の実施形態では、本発明は、本発明に係る液体ワクチンの単位用量に関する。
特定の実施形態では、本発明は、ジカウイルス感染症を治療または予防すること、特に予防することを、それを必要とするヒト対象において行う方法であって、当該ヒト対象に、本発明に係るワクチンの単位用量を投与することを含む、方法に関する。
特定の実施形態では、本発明はまた、液体ワクチンを調製する方法であって、次のステップ:
ステップ1.本発明に係る不活化ウイルス組成物を提供すること、
ステップ2.不活化ウイルス組成物に、アジュバント、好ましくはアルミニウム塩と、任意選択により更なる薬学的に許容される緩衝液とを添加すること、
を含む、方法に関する。
この実施例は、既知の研究歴のある、ジカウイルス(ZIKAV)株の生産を記載する。
Vero細胞の維持
1バイアルのWHO Vero10-87細胞を水浴中で迅速に融解し、T-75cm2フラスコで、ペニシリン-ストレプトマイシン、L-グルタミン40mM、及び10%FBSを含有する、予め温めておいた19mLのDMEM(ダルベッコ改変最小必須培地)中に36℃+/2℃、5%CO2で直接接種した。細胞をコンフルエントになるまで成長させ、TryplEを使用して継代培養した。このフラスコを2つのT-185cm2フラスコに拡張し、コンフルエントになるまで成長させ、31×T-185cm2フラスコまで継代培養し、細胞が100%コンフルエントになるまで成長させた。トリプシン処理によって細胞を回収し、800×gで10分間遠心分離にかけ、10%FBS及び10%DMSOを含有するDMEM中に、1.9×107細胞/mLの濃度で再懸濁した。上記のように、1バイアルのVero細胞を迅速に解凍し、T-75cm2フラスコ内で蘇生させた。これらを2回継代培養して、13×T-185cm2フラスコの細胞バンクを作製した。トリプシン処理後、細胞を800×gで遠心分離にかけ、凍結培地(10%FBS及び10%DMSOを含有するDMEM)中に、4.68×105細胞/mLの濃度で、再懸濁した。この細胞バンクをクライオバイアルに等分した。
6ウェルプレートで成長させたVero細胞の新鮮なコンフルエント単層のウイルス力価をプラーク滴定により決定した。凍結アリコートを解凍し、96ウェルプレートでアリコートの10倍希釈系列をcDMEM-0%-FBSで作製した。希釈したウイルスは、Vero細胞単層の接種前まで、氷上に維持した。アッセイ時に、6ウェルプレートから成長培地を吸引し、各ウイルス希釈液100μLをウェルに加えた。細胞シートの乾燥を防ぐために、プレートを頻繁に(10分ごとに)揺らしながら、ウイルスを36℃±2℃、5%CO2で60分間吸着させた。ウイルス吸着後、40~41℃に維持した第1のアガロースオーバーレイ(1X cDMEM-2%-FBS+0.8%アガロース)4mLを各ウェルに加えた。アガロースを室温で30分間固化させ、次いで、プレートを上下逆さまにして36℃+/2℃、5%CO2で、4~6日間インキュベートした。160μg/mLのニュートラルレッド生体染色色素を含有する第2のアガロースオーバーレイ2mLを4日目に加えた。プラークを5日目及び6日目に可視化した。
96ウェルプレートで成長させたVero細胞の新鮮なコンフルエント単層でのウイルス力価も滴定により決定した。凍結アリコートを解凍し、96ウェルプレートでアリコートの10倍希釈系列をcDMEM-2%-FBS希釈液で作製した。希釈したウイルスは、Vero細胞単層の接種前まで、氷上に維持した。アッセイ時に、96ウェルプレートから成長培地を吸引し、各ウイルス希釈液100μLをウェルに加えた。プレートを36℃+/2℃、5%CO2で5日間インキュベートした。50%組織培養物感染量(TCID50)の力価をReed/Muench計算表の使用により算出した。
ジカウイルス株PRVABC59(ドライアイス上の0.5mLバイアル1本)を疾病予防管理センター(CDC)から受領し、ジカウイルスの同定をRT-PCRによって確認した。この株を試験すると、PCRによるAlphavirus及びマイコプラズマ汚染は陰性であった。この情報を表1にまとめる。
QIAampViral RNA Mini Spinキットを製造元のプロトコルに従って使用して、各分離株の安定化されたウイルス回収物からRNAを抽出した。各分離株から抽出したRNAを使用して、全ジカウイルスゲノムを包含する6つのcDNAフラグメントを作製し、増幅した。増幅したcDNAフラグメントのサイズ及び純度を1%アガロース/TBEゲルで分析し、その後、Qiagen Quick Gel Extraction Kitを使用してゲル精製した。ABI 3130XL Genetic Analyzerシーケンサーを使用して自動シークエンシング反応を行った。Lasergene SeqManソフトウェアを使用してシークエンシングデータを解析した。
アメリカにおける現在のZIKAVアウトブレイクに関連し、既知の研究歴のあるZIKAV株が必要とされた。このため、ZIKAV PRVABC59株が選択された。Vero細胞での成長に適合した十分に特徴付けされたウイルスを生成するために、ZIKAV PRVABC59を最初にVero細胞で増幅させた(P1)。
不活化(COI)アッセイの完全性とも呼ばれる二重感染力アッセイを開発して、ホルムアルデヒド不活化(0.01%ホルムアルデヒド)の有効性と、精製された不活化ジカウイルス(PIZV)バルク原薬(BDS)の潜在的な残留感染性ウイルス活性を決定した。
1.アッセイの第1部:サンプル添加の2日前に、Vero(1.4E+05細胞/mL)細胞及びネッタイシマ蚊C6/36(4E+05細胞/mL)細胞を96ウェルプレートに播種した。Vero細胞は、DMEM+10%最終FBS+2%L-グルタミン+1%ペニシリン/ストレプトマイシン中、37℃で培養した。C6/36細胞は、DMEM+10%FBS+2%L-グルタミン+1%ペニシリン/ストレプトマイシン+1%非必須アミノ酸中、28℃で培養した。
2.200 TCID50対照ウイルス(ウイルス逆滴定コントロール試験で調製)またはDSサンプルの3つの独立したレプリケートを2%FBS含有培地に希釈した(5倍及び10倍希釈)。
3.96ウェルプレート中の細胞にサンプルを接種した。サンプルは、96ウェルプレートの細胞単層への感染前に、ボルテックス処理をし、存在し得るあらゆる凝集を解いた。100μLの各希釈液をそれぞれVero及びC6/36細胞を含有する2つの別個の96ウェルプレートの5つのウェルのそれぞれにアプライした。
4.各細胞種の別のウェルには、陰性CPE対照として、培地のみを含めた。
5.プレートを細胞株に適した温度で6日間インキュベートした。
6.アッセイの第2部:許容細胞株において、生きているウイルスを更に増幅し、CPEによって可視化するために、Vero及びC6/36細胞の両方から、各96ウェル上清の全量をVero細胞の6ウェルプレートの個々のウェルに移した。15分間隔で揺らしながら、接種を90分間進行させた。
7.2%FBS含有培地をウェルに加え、プレートを更に8日間インキュベートし、その後、CPEに関して増幅サンプルの検出を行った。不活化は、DSのレプリケートのいずれかが8日目の終わりにCPEを示した場合、不完全であるとみなした。
7.生きている/複製するビリオンの存在を、6ウェルプレートに移し、8日間インキュベーションして、ウイルスを複製させた後の感受性細胞単層上のプラークまたはCPEの形成によって可視化した。アッセイ終了時の6ウェルプレートの%CPE評価は、次のように算出した:
Vero細胞の各6ウェルプレートを比色変化の可視化によってCPEについて試験し、続いて、倒立光学顕微鏡での検査によってCPEを確認した。
各6ウェルプレートは、上記の5倍及び10倍希釈で調製されたDS希釈液のレプリケートの1つであった(5つのウェル+培地のみを含有するウェル1つ)。
したがって、各レプリケートの%CPEは、1サンプル当たり合計5ウェルのうち、CPEを有するウェルの数を反映するものであった(アッセイごとに合計120個のウェルを使用)。平均%CPE及び標準偏差は、各希釈液の3つのレプリケートに基づいて算出した。
結果:以下の表6~9に示すようにToxロット#1~4のそれぞれで日々のサンプルを分析した。
実施例3では、「ジカウイルスワクチン原薬」という用語は、ジカワクチンの生産の中間体である不活化ウイルス組成物を指すために使用される。
必要品
精製された不活化ジカワクチン原薬を、上記のとおり、Vero細胞における成長によって製造した。毎日の採取を3~9日目に実施し、これらをプールしてから、精製及び不活化した。毎日の採取物から上清を濾過及びクロマトグラフィーによって精製し、濃縮し、ホルムアルデヒドを0.01%の最終濃度まで加えることによって不活化した。不活化を22℃で10日間進行させた後、サンプルをメタ重亜硫酸ナトリウムで中和し、次いで、ジカリン酸緩衝液(6.46mMのリン酸二ナトリウム、1.47mMのリン酸二カリウム、137mMのNaCl、6%スクロース、pH7.4)に緩衝液交換した。
製造したら、ジカウイルスワクチン原薬を+5±3℃/周囲湿度で最大6ヶ月間保存した。
各試験(実施例3A~3F)のはじめに、緩衝液交換後のジカウイルスワクチン原薬(DS)のサンプルを個々の3mLバイアルに分注した。各バイアルは、それぞれの緩衝液中におよそ0.67mLのジカウイルスワクチン原薬(DS)を含有した。次いで、バイアルをETFEラミネートゴム栓で密封した。
シングルカラムを使用するサイズ排除クロマトグラフィー(SEC)を使用して、安定性の分析を実施した。サイズ排除クロマトグラフィーは、分子量に基づいてタンパク質を分離するのに使用される技術である。タンパク質サンプルの分子量が大きければ大きいほど溶出時間が短くなる。インタクトなジカウイルスでは、8分前後の保持時間でSECトレースのピークが見られた。このピークの積分と参照サンプル(0日目)の積分を比較することによって、保管期間後のサンプル中にインタクトなジカウイルスがどの程度存在しているかを決定することができる。更に、より早い保持時間に何らかの更なるピークが現れるかどうかに注目することによって、ジカウイルスが凝集しているかどうかも決定することができる。
BSA参照スタンダード:2mg/mLの明確に定義された濃度を有するBSA原液を水で希釈することによって、200、400、800、及び1,000μg/mLの濃度のウシ血清アルブミン(BSA)の参照サンプルを調製した。次いで、各参照サンプルのアリコートをラベル付きHPLCバイアルに移し、栓をした。以下の実施例3A~3Fに示される全てのタンパク質濃度は、BSA標準曲線に基づいたジカウイルスの濃度に基づく。
SECの実施ごとに、ウシ血清アルブミン(BSA)の標準曲線を作成し、200、400、800、及び1,000μg/mLの濃度のスタンダードを流し、標準曲線を作成した(線形回帰、y=mx+bであり、b=0)。BSAの単量体及び二量体のピーク面積の合計として、BSAのピーク総面積を算出した。
実施例3Aのサンプルの調製は、上記のとおり実施した。ジカウイルスワクチン原薬を調製し、次いで、以下の表14a及び14bに列挙されるそれぞれの各緩衝液に緩衝液交換した。実施例3Aでは、5±3℃と-80℃の両方で10日間の安定性試験を実施した。
実施例3Bのサンプルの調製は、上記のとおり実施した。ジカウイルスワクチン原薬を調製し、次いで、以下の表に列挙されるそれぞれの各緩衝液に緩衝液交換した。実施例3Bでは、-80℃で60日間の安定性試験を実施した。
実施例3Cのサンプルの調製は、上記のとおり実施した。ジカウイルスワクチン原薬を調製し、次いで、以下の表に列挙されるそれぞれの各緩衝液に緩衝液交換した。実施例3Cでは、5±3℃と-80℃の両方で67日間の安定性試験を実施した。
実施例3Dのサンプルの調製は、上記のとおり実施した。ジカウイルスワクチン原薬のサンプルを調製し、次いで、Tris緩衝液及びZPB緩衝液(上の表11に列挙されるもの)に緩衝液交換した。実施例3Dでは、-80℃で3ヶ月間の安定性試験を実施した。
実施例3Eのサンプルの調製は、上記のとおり実施した。ジカウイルスワクチン原薬を調製し、次いで、以下の表に列挙されるそれぞれの各緩衝液に緩衝液交換した。実施例3Eでは、5±3℃で60日間の安定性試験を実施した。
実施例3Fのサンプルの調製は、上記のとおり実施した。ジカウイルスワクチン原薬を調製し、次いで、Tris緩衝液に緩衝液交換した。その後、サンプルにスクロースを加えた(合計濃度がそれぞれ3、5、7及び10%w/vのサンプルになるように)。

- 前記ジカウイルスが、全粒子生ジカウイルスを0.015%w/v未満のホルムアルデヒドで処理することによって得ることができる不活化全粒子ウイルスである、請求項44に記載の液体不活化ウイルス組成物。
- a)先行請求項のいずれか1項に記載の不活化ウイルス組成物と、
b)水酸化アルミニウムなどのアジュバントと、
を含む、液体ワクチン。 - 前記液体ワクチン中の塩化ナトリウムの濃度が、約50mM~約200mM、または約60mM~約150mM、例えば、約84mMである、請求項46に記載の液体ワクチン。
- 前記液体ワクチンが、約8.5mM~約80mMのTrisと、約60mM~約150mMのNaClと、を含み、前記液体不活化ウイルス組成物のpHは、室温で測定したとき、pH約7.0~pH約8.0である、請求項47に記載の液体ワクチン。
- 前記液体ワクチンが、約0.4%(w/v)~4.7%(w/v)のスクロースを含む、請求項48に記載の液体ワクチン。
- アルミニウム元素に基づいて、100μg/ml~800μg/mlの水酸化アルミニウム、または200μg/ml~600μg/mlの水酸化アルミニウム、または300μg/ml~500μg/mlの水酸化アルミニウム、または約400μg/mlの水酸化アルミニウムを含む、請求項49に記載の液体ワクチン。

- 約2μgの前記不活化全粒子ジカウイルスを含む、請求項52に記載のワクチンの単位用量。
- 約5μgの前記不活化全粒子ジカウイルスを含む、請求項52に記載のワクチンの単位用量。
- 約10μgの前記不活化全粒子ジカウイルスを含む、請求項52に記載のワクチンの単位用量。
- 約0.4mL~約0.8mLの薬学的に許容される液体として提供される、請求項51~55のいずれか1項に記載のワクチンの単位用量。
- 不活化ウイルス組成物の使用であって、
a)不活化全粒子ジカウイルスと、
a)少なくとも約6.5mMの濃度の少なくとも1つの薬学的に許容される緩衝液と、
b)任意選択により、ポリオールと、を含み、
前記不活化ウイルス組成物は、アルミニウム塩から選択されるアジュバントを含有せず、前記少なくとも1つの薬学的に許容される緩衝液は、リン酸イオンを含まない、

b)薬学的に許容される緩衝液であって、前記緩衝液は、リン酸緩衝液ではなく、前記緩衝液の濃度は、少なくとも6.5mMである、前記緩衝液と、
c)任意選択により、ポリオールと、を含み、
前記不活化ウイルス組成物は、好ましくはアルミニウム塩から選択されるアジュバントを含有せず、次のステップ:
ステップ1.1つ以上の非ヒト細胞から得られた上清からジカウイルス調製物を単離すること、
ステップ2.前記ジカウイルス調製物を精製すること、
ステップ3.前記ウイルス調製物を不活化すること、
