本開示は、がん免疫療法で使用される、好ましくは、I型IFN(IFN-1)アクチベーター/インデューサーと組み合わせた、粘膜組織におけるIL-12の局所発現のための方法及び組成物に関する。

ポリアニオン含有ブロックコポリマー、例えば、二元ブロック及び/または三元ブロックコポリマーコーティングと複合体化されたキトサン誘導体核酸ナノ粒子(ポリプレックス)を含むキトサン組成物が本明細書で提供され、個々のポリマー分子は、負荷電アンカー領域及び1つ以上の非荷電親水性尾部領域を含む。本発明の方法及び組成物において有用な例示的なポリマー分子は、ポリエチレングリコール(PEG)部分及びポリアニオン(PA)部分を含む「PEG-PA」ポリマー分子である。

ポリマーの親水性非荷電部分は、ポリアルキレンポリオールもしくはポリアルキレンオキシポリオール部分、またはそれらの組み合わせであり得るか、またはそれらを含み得る。ポリマーの親水性非荷電部分は、ポリアルキレングリコールまたはポリアルキレンオキシグリコール部分であり得るか、またはそれらを含み得る。特定の実施形態では、ポリアルキレングリコール部分は、ポリエチレングリコール部分及び/またはモノメトキシポリエチレングリコール部分であるか、またはそれらを含む。特定の好ましい実施形態では、ポリマーの非荷電部分は、ポリエチレングリコールであるか、またはそれを含む。ポリマーの親水性非荷電部分は、ポリ乳酸などの他の生物学的に適合性のあるポリマー(複数可)であり得るか、またはそれを含み得る。
ポリマーのアニオンポリマー部分は、生理学的pHで負電荷を有する複数の官能基を含み得る。多種多様なアニオンポリマーは、本明細書に記載の方法及び組成物での使用に適する。但し、そのようなアニオンポリマーは、親水性非荷電ポリマー部分を有するポリマーの構成成分として提供することができ、正荷電(例えば、二重)誘導体化キトサン-核酸ナノ粒子との(例えば、可逆的)電荷:電荷複合体を形成可能である。

本発明の核酸ポリプレックスは、ヌクレオチドを凝縮し、酵素分解からヌクレオチドを保護するように機能する。キトサン及びその誘導体に加えて、この目的のためにも有利に使用することができる代替材料は、他の正荷電(すなわち、カチオン性)ポリマー及び/または脂質を含む。

本発明のポリプレックス:ポリマー組成物は、粉末を含む。好ましい実施形態では、本発明は、乾燥粉末ポリプレックス:ポリマー組成物を提供する。好ましい実施形態では、乾燥粉末ポリプレックス:ポリマー組成物は、本発明のキトサン-核酸ポリプレックス分散液の脱水(例えば、噴霧乾燥または凍結乾燥)により生成される。
本発明は、また、本発明のポリプレックス:ポリマー組成物を含む「薬学的に許容される」または「生理学的に許容される」製剤を提供する。そのような製剤は、処置方法を実施するために、対象にin vivoで投与することができる。
一実施形態では、ポリプレックス:ポリマー組成物の使用は、生理学的pHでのポリプレックスの長期安定性を提供する。これは、有効な粘膜投与を提供する。

過剰増殖性疾患
本組成物及び方法は、過剰増殖性障害の処置において有利な使用を見出す。特に興味深いのは、粘膜組織または粘膜組織の近位の組織の過剰増殖性障害を処置するための組成物及び方法である。本発明の方法及び組成物は、限定されないが、口腔癌、食道癌、胃癌、膵臓癌、肝臓癌、結腸直腸癌、及び直腸癌を含む、胃腸癌の処置に使用されてもよい。本発明の方法及び組成物により処置され得る鼻癌及び肺癌は、副鼻腔癌、口腔咽頭癌、気管癌、及び肺癌を含むが、これらに限定されない。本発明の方法及び組成物により処置され得る泌尿生殖器癌は、膀胱癌、尿路上皮癌、尿道癌、精巣癌、腎臓癌、前立腺癌、陰茎癌、副腎癌、子宮癌、子宮頸癌、及び卵巣癌が含まれるが、これらに限定されない。

JetPEI及びアミン対リン(NP)比6の最適化Luc2を含む候補プラスミド20μgを混合することにより、JetPEI-プラスミドDNAポリプレックスを調製した。ポリプレックスを室温で最低15分間インキュベートし、4時間以内に使用した。雌マウス(12~16週間)に、イソフルランを用いる麻酔下、曝露時間60分間で膀胱内点滴注入により、80μlのJet PEI-DNA製剤を投与した。投与の24時間後、膀胱組織を採取し、mRNA発現及びルシフェラーゼ酵素活性アッセイを使用して、トランスフェクション効果を評価した。溶解緩衝液(Qiagen RNeasy)中で均質化した後、採取された膀胱組織からRNAを抽出した。1μgの投入RNA及びLuc2を認識するTaqManプライマー/プローブを使用して、RT-qPCRを実施した。Luc2 RNAの標準曲線を使用して、絶対定量を実施した。図2Aに見られるように、RNAアッセイの結果は、in vitroルシフェラーゼ活性アッセイの結果と一致しており、NTC9385Luc2は、絶対コピー数に基づいてトランスフェクション後に最高のmRNAレベルを示した。ルシフェラーゼアッセイのために、ルシフェラーゼ細胞培養溶解試薬(IX、Promega)の存在下で均質化することにより、採取された膀胱組織を溶解させた。酵素基質であるルシフェリンを、組織溶解物に添加し、生物発光を、EnVisionプレートリーダー(Perkin-Elmer)で直ちに測定した。図2Bに見られるように、gWIZ-Luc2及びNTC9385Luc2-プラスミドの両方が、pVAX-Luc2プラスミドよりもin vivoで非常に高いルシフェラーゼ酵素活性を示した。
in vitroでのマウス膀胱上皮細胞株におけるプロモーター/エンハンサースクリーニング
様々なプロモーター/エンハンサー配列を含有するプラスミドの一団でマウス尿路上皮細胞株をin vitroでトランスフェクトした後に、緑色蛍光レポーター遺伝子(GFP)の用量依存的発現を測定することにより、プロモーター/エンハンサーの組み合わせの有効性を評価した。アッセイで使用されたプロモーター/エンハンサー配列は、CAG、EF1a、CMV/EF1a/HTLV、CMV/UbC、EF1a/HTLV、2xCMV/EF1a、CMV、UbC、CMV/EF1a、CMV/UbB、PGK、UbB、及びCBAであった。SnapFastバックボーン(pSF)の示されたプラスミドでトランスフェクトする24時間前に、MB49細胞(マウス膀胱細胞)を96ウェルプレートに播種した(35000細胞/ウェル)。

ヒト初代膀胱上皮細胞のin vitroでのトランスフェクトした後の緑色蛍光レポート遺伝子(GFP)の用量依存的発現を測定することにより、プロモーター/エンハンサーの組み合わせの有効性を評価した。SnapFastバックボーン(pSF)の代替プロモーター/エンハンサーの組み合わせを含有するプラスミドでトランスフェクトする24時間前に、ヒト初代膀胱上皮細胞(ATCC)を96ウェルプレート(25,000細胞/ウェル)に播種した。研究されたプロモーター/エンハンサーの組み合わせは、CAG、UbB、EF1a、CMV-EF1a-HTLV、2XCEF、PGK、CBA、CMV-Ubb、EF1a-HTLV、CMV、CEF、及びUBCを含んでいた。Avalanche Transfection Reagent(EZ Biosystems)及び漸増用量のプラスミドDNAを使用して、トランスフェクションを実施した。トランスフェクションの24時間後に、Envisionプレートリーダーで、蛍光を測定した。AlamarBlue Cell Viability Reagent(Invitrogen)を使用して、細胞生存率を測定した。データは、細胞生存率に対して正規化されたGFP相対蛍光単位(RFU)として表される。図4Aに見られるように、MB49細胞株で良好に実施されたプロモーター/エンハンサーの組み合わせは、また、ヒト初代細胞でも高レベルの発現を示す。同様に、MB49細胞株における低い発現体は、初代細胞においても低い発現を示す。2つの独立したアッセイにわたってプラスミドのそれぞれについて、総蛍光強度をランク付けした(図4B)。EF1a/HTLV、CAG、及び2xCMV/EF1aを含むプラスミドは、最高レベルの発現を示し、UBB及びPGKプロモーター/エンハンサーを含むプラスミドは、ヒト膀胱細胞で最低の発現を示した。
初代ヒト膀胱上皮細胞における改造プラスミドの測定

インキュベーション期間後にポリプレックスの排尿がある場合及びない場合のin vivoでの膀胱トランスフェクション効率に対するPEG化の影響
マウス膀胱にDDX及びPEG化DDX製剤を投与した後に、ヒトPD-L1-FcのmRNA発現をアッセイした。7:1:17.5(PEG-DDX)または7:1:7(PEG-DDX)または7:1:0(DDX対照)のNPA比で、ポリプレックスを調製した。雌マウス(12~16週間)に、イソフルランを用いる麻酔下、膀胱内点滴注入(曝露時間=60分)により、80μLのポリプレックス(20μgのプラスミドDNA)を投与した。ポリプレックスへの曝露後及び麻酔から覚醒前に膀胱から排尿させるか(尿道を通してシリンジ及びカテーテルを使用)、または排尿させなかった。投与の24時間後に、膀胱組織を採取し、溶解緩衝液中で組織を均質化した後、RNAを抽出した(Qiagen RNeasyキット)。1μgの投入RNA及びコドン最適化ヒトPD-L1-Fcを認識するTaqManプライマー/プローブを使用して、RT-qPCRを実施した。ヒトPD-L1-FcRNAの標準曲線を使用して、絶対定量を実施した。データは、RNAコピー数で表される(図6)。
種々のプラスミド濃度でのPEG対非PEGを使用したin vivoでのmRNA発現の評価
ヒトPD-L1-Fcを含むDDX及びPEG-DDXポリプレックスを、7:1:9(PEG-DDX)または7:1:0(DDX)のNPA比で調製した。雌マウス(12~16週間)に、イソフルランを用いる麻酔下、膀胱内点滴注入(曝露時間=60分)により、80μLのポリプレックス(0.25mgのDNA/mL=20μgのプラスミドDNA;1.0mgのDNA/mL=80μgのプラスミドDNA)を投与した。投与の24時間後に、膀胱組織を採取し、溶解緩衝液中で組織を均質化した後、RNAを抽出した(Qiagen RNeasyキット)。1μgの投入RNA及びコドン最適化ヒトPD-L1-Fcを認識するTaqManプライマー/プローブを使用して、RT-qPCRを実施した。ヒトPD-L1-FcRNAの標準曲線を使用して、絶対定量を実施した。データは、RNAコピー数で表される(図7A)。2番目の実験では、示されたポリプレックスを、7:1:3.5(PEG-DDX)または7:1:0(DDX対照)のNPA比で調製した。雌マウス(12~16週間)を同じレジメンにかけた:イソフルランを用いる麻酔下、膀胱内点滴注入(曝露時間=60分)による、80μLのポリプレックスの投与(0.25mgのDNA/mL=20μgのプラスミドDNA;1.0mgのDNA/mL=80μgのプラスミドDNA)。投与の24時間後に、膀胱組織を採取し、溶解緩衝液中で組織を均質化した後、RNAを抽出した(Qiagen RNeasyキット)。1μgの投入RNA及びコドン最適化ヒトPD-L1-Fcを認識するTaqManプライマー/プローブを使用して、RT-qPCRを実施した。ヒトPD-L1-FcRNAの標準曲線を使用して、絶対定量を実施した。データは、RNAコピー数で表される(図7B)。0.25mgのDNA/mlを投与されたものよりも、1.0mg/DNA/mlを投与された動物で、より高い発現が見られた。PEG化により、非PEG化製剤と比較して、投与の24時間後に、膀胱でのmRNA発現が減少しなかった。
マウス膀胱にPEG化DDX製剤を投与した後のヒトPD-L1-Fcの発現
PEG-DDXポリプレックスを7:1:3.5のNPA比で調製した。雌マウス(12~16週間)に、イソフルランを用いる麻酔下、膀胱内点滴注入(曝露時間=60分)により、80μLのポリプレックス(0.125mgのDNA/mL=10μgのプラスミドDNA)を投与した。投与の48時間後に、膀胱組織を採取し、タンパク質溶解緩衝液及びプロテアーゼ阻害薬の存在下で均質化することにより、組織を溶解させた。カスタム設計のイムノアッセイ(Mesoscale Discovery)を使用して、ヒトPD-L1-Fcタンパク質を定量した。データは、溶解物中のpg/mLのタンパク質として表される(図8)。PEG-DDX(商標)ポリプレックスの膀胱内点滴注入により、膀胱内での強力な定量可能なタンパク質発現が得られた。
マウス膀胱内の尿中のポリプレックス安定性に対するPEG化の影響。
80μLの非PEG化(NPA 7:1:0)ならびにPEG化(NPA 7:1:7及び7:1:17.5)DDX/DNAポリプレックス製剤を、0.25mgのDNA/mL(n=4)でマウス膀胱に投与した。分析のために(膀胱の内容物を収集する)膀胱排尿の前に、製剤を膀胱内で1時間インキュベートした。外観(図9A)及び動的光散乱(図9B)によるナノ粒子のサイジングについて試料を試験した。非PEG化ポリプレックスは、排尿後に大幅に凝集し、凝集を示さなかったPEG化ポリプレックスと比較して、目に見える白い凝血塊があった。
in vivoでのhPD-L1-Fc産生に対するDDX-I対DDX-IIの影響
雌マウス(12~16週間)に、イソフルランを用いる麻酔下、膀胱内点滴注入(曝露時間=60分)により、80μLのポリプレックス(1.0mgのDNA/mL=80μgのプラスミドDNA)を投与した。投与の48時間後に、膀胱組織を採取し、プロテアーゼ阻害薬を含有するタンパク質溶解緩衝液の存在下で均質化することにより、組織を溶解させた。カスタム設計のイムノアッセイ(Mesoscale Discovery)を使用して、ヒトPD-L1-Fcタンパク質を定量した。データは、溶解物中のpg/mLのタンパク質として表される。データは、平均+/-SD;*p<0.05;**p<0.005-Kruskal-Wallis検定を用いる一元ANOVAである。ポリプレックスを、(示されるように)10:1または30:1のNP比及び1.0mg/mLのDNA濃度で調製した。DDX-Iの組成は、14%のR及び3%のGAであった。DDX-II(RXG)の組成は、(示されるように)13%のR/13%のGまたは28%のR/9%のGであった。図10に示されるように、DDX-II(RXG)は、DDX-Iよりもin vivoで有意に高いタンパク質発現をもたらす。
RXG製剤をマウス膀胱に投与した後のタンパク質発現の動態
ポリプレックスを、10:1:5(PEG化)または20:1:0(非PEG化)のNPA比で調製した。PEG化製剤を、5%のトレハロース+5%のマンニトールで行った。非PEG化製剤を、5%のトレハロースで行った。%R及び%GのRXG(DDX-II)組成が示される。雌マウス(12~16週間)に、イソフルランを用いる麻酔下、膀胱内点滴注入(曝露時間=60分)により、80μLのポリプレックス(1.0mgのDNA/mL=80μgのプラスミドDNA)を投与した。投与の24、48、72、及び96時間後の時点で、膀胱組織を採取し、プロテアーゼ阻害薬を含有するタンパク質溶解緩衝液の存在下で均質化することにより、組織を溶解させた。カスタム設計のイムノアッセイ(Mesoscale Discovery)を使用して、ヒトPD-L1-Fcタンパク質を定量した。データは、溶解物中のpg/mLのタンパク質として表される(図11)。タンパク質の発現は、高く(ng/mlの範囲内)、投与の96時間後まで持続する。
RIG-Iアゴニストカセットを含まない及び含む、マウスIL-12を含むプラスミドDNA構築物
RIG-Iアゴニストカセットを含まない、マウスIL-12を含むプラスミド構築物(図12A)及びRIG-Iアゴニストカセットを含む、マウスIL-12を含むプラスミド構築物(図12B)。マウスIL-12導入遺伝子は、短いエラスチンリンカーを含むIL-12 p40及びp35サブユニットの遺伝子の単一のオープンリーディングフレームを含む(図12A)。mIL-12導入遺伝子を含有するプラスミドでHEK293T細胞にトランスフェクトし、上清をトランスフェクションの48時間後に採取した。イムノアッセイにより細胞の上清中のマウスIL-12p40p35を定量した。構築物は、生物活性IL-12を生じ(図13A)、脾細胞を96ウェルプレートに播種し、漸増用量のmIL-12を含有する上清とともに、抗CD3及び抗CD28で刺激した。刺激の48時間後に、ELISAにより、脾細胞培養上清中のIFNγを測定した(図13A)。HEKBlue細胞を96ウェルプレートに播種し、mIL-12を含有する漸増用量の上清で刺激した。組み換えSEAPの標準曲線と比較して、HEKBlue上清中のIL-12媒介性SEAP産生を定量し、データを細胞数に対して正規化した。
RIG-Iアゴニストを含むプラスミドでトランスフェクトされた膀胱癌上皮細胞におけるin vitroでのIFNβ産生。(図14A及び14B)
RIG-Iアゴニストを含むまたは含まないプラスミド(NTC9385R-mIL12)でトランスフェクトする前に、MB49細胞を96ウェルプレート(35,000細胞/ウェル)に播種した。リポフェクタミン2000(Thermofisher)及び漸増用量のプラスミドDNAを使用して、トランスフェクションを実施した。トランスフェクションの48時間後に、細胞培養上清を回収し、IFNβ産生をELISAで測定した。データは、総細胞タンパク質に対して正規化され、pg IFNβ/mgの総タンパク質として表された。
マウス膀胱にRXG製剤を投与した後のin vivoでのmRNA発現
コドン最適化配列を有する一本鎖マウスIL-12p40p35オープンリーディングフレームを、NTC9385RまたはNTC9385R-eRNA41Hベクターバックボーンにクローニングし、MB49細胞においてin vitroで発現を確認した(データは示さず)。RXGポリマーを使用して、ポリプレックスを調製した(NP20、非PEG化;25%のR、10%のG、賦形剤として5%のトレハロース)。
ポリマー構造及びポリプレックス形成
凍結、または凍結乾燥のために、後続のタンジェンシャルフロー濾過(TFF)濃度に対し様々な安定剤中で、3~30のアミン対リン酸塩(N:P)比でpDNAとDDXを複合体化することにより、ポリプレックスを形成した。二重誘導体化キトサン(DDX)の構造:Σ(q+p+n)=1、q=0.03~0.35、p=0.12~0.28(図16)。
DDX-DNA製剤の物理化学的特性
12~28%のアルギニン及び3~35%のポリオールとコンジュゲートされたDDXを使用して、ポリプレックスを3~30の範囲のN:P比で調製した。10mMのNaCl中、動的光散乱(DLS)により、ポリプレックス流体力学的直径(Z平均)及び多分散度指数(PDI)を測定した。100Vに1時間かけられた、pH8(0.5XのTBE)の0.8%アガロースゲルでのポリプレックスからのDNA放出の視覚的評価により、DNA捕捉を決定した。過剰な競合するポリアニオン(ポリ-(α,β)-DL-アスパラギン酸)とのインキュベーションによるポリプレックスからのDNA放出に続いて、アガロースゲル電気泳動(Quantity One v4.6.7、Bio-Rad Laboratories)により、DNAスーパーコイルを定量した。ポリプレックスのゼータ電位は、また、レーザードップラー速度計で測定された(図17)。
ポリマーのin vitroスクリーニング
in vitroスクリーニングを使用して、DDX(3%ポリオール、14%アルギニン(R))と比較してトランスフェクション効果が改善された新規ポリマー。示された製剤を用いるトランスフェクションの24時間前に、マウス尿路上皮癌細胞(MB49)を96ウェルプレートに播種した(35000細胞/ウェル)。示されているように、漸増用量のプラスミドDNAで、トランスフェクションを実施した。トランスフェクションの48時間後に、Envisionプレートリーダーで、蛍光を測定した。AlamarBlue Cell Viability Reagentを使用して、細胞生存率を測定した。データは、細胞生存率に対して正規化されたGFP相対蛍光単位として表される(図18A)。B.ヒートマップは、DDX(3%のポリオール、14%のR)に対する最大発現の倍数変化を表す(図18B)。DDXのハイコンテントイメージングにより、GFP陽性細胞のパーセンテージを決定し、最大効果と比較した(図18C)。
DDXのIVI投与後のin vivoでのタンパク質発現

非ヒト霊長類に、麻酔下、膀胱内点滴注入(曝露時間=60分)により、IL-12p40p35を含む0.25mg/mlのEG-70を投与した。動物は、10mlまたは20mLのEG-70または20mLの空ベクター対照を含有するポリプレックスを投与される(n=1/群、雌(EG-70)または雄(空ベクターDNAを含むポリプレックス))。投与の48時間後に、膀胱組織を採取し、RNA抽出に使用される緩衝液(図20A)またはプロテアーゼ阻害薬を含有するタンパク質溶解緩衝液(図20B)の存在下で均質化することにより、組織を溶解させた。1μgの投入RNA及びコドン最適化ヒトIL-12p40p35を認識するTaqManプライマー/プローブを使用して、RT-qPCRを実施した。in vitro転写により生成されたヒトIL-12p40p35 RNAの標準曲線を使用して、絶対定量を実施した(図20A)。市販のヒトIL-12p70イムノアッセイ(Mesoscale Discovery)を使用して、ヒトIL-12タンパク質を定量した。データは、溶解物中のpg/mLのタンパク質として表される(図20B)。検出可能なレベルのmRNA及びタンパク質は、10ml及び20mlの処置の膀胱内投与の両方で見られた(それぞれ、2.5mgまたは5mgのプラスミドDNAに対応)。個々のデータポイントは、個々の動物内の膀胱組織の割合を示す。

非ヒト霊長類に、麻酔下、膀胱内点滴注入(曝露時間=60分)により、IL-12/eRNA11a/VA1を含む20mlのC250 EG-70ポリプレックス、または空ベクター対照DNAを含有するポリプレックスを投与した。動物は、0.0625mg/mL、0.25mg/mL、もしくは1mg/mL(それぞれ、c62.5、c250、もしくはc1000)のEG-70、または、1mg/mL(c1000)の対照ナノ粒子(RXG-PEG-N9;n=1/群、雌(EG-70、低用量及び中用量)または雄(EG-70、高用量もしくは空ベクターDNAを含むポリプレックス))の投与を受けた。投与の48時間後に、膀胱組織を採取し、RNA抽出緩衝液の存在下で均質化することにより、組織を溶解させた。1μgの投入RNA及びeRNA11aまたはVA1を認識するTaqManプライマー/プローブを使用して、RT-qPCRを実施した。in vitro転写により生成されたヒトeRNA11a RNAの標準曲線(図21)、及びヒトVA1 RNA(図22)を使用して、絶対定量を実施した(個々のデータポイントは、個々の動物内の膀胱組織画分を示す)。
膀胱癌のマウスモデルにおける抗腫瘍活性の評価
mEG-70プロトタイプナノ粒子の抗腫瘍活性を評価するために、マウス膀胱癌の同所性モデルを使用した。要約すると、表在性尿路上皮層の落屑を促進し、がん細胞の着床を促進するために、ポリ-L-リジンでマウスの膀胱を前処理することにより、疾患を確立した。続いて、ルシフェラーゼ遺伝子(MB49-Luc)を安定して過剰発現する尿路上皮癌細胞をマウス膀胱に点滴注入し(マウス1匹当たり100,000細胞)、in vivoイメージングシステム(IVIS)を使用して、点滴注入後12日目に、ルシフェラーゼ発現を確認した。生物発光シグナルの強度を使用して、動物を処置群にランダム化した。さらに、陽性の生物発光シグナルを有しない動物を研究から除外した。マウスは、点滴注入後の13日目及び20日目に、週次で2回のナノ粒子の膀胱内投与を受けた。この投与レジメンを、タンパク質発現動態の評価に基づいて選択した。動物の追加の群は、ナノ粒子ビヒクル、トレハロース(5%)の投与によるシャム手順を受けた。膀胱内の腫瘍量の関数として膀胱重量を測定するために、実験を29日目に終了した。
マウス膀胱におけるマウスIL-12 mRNA発現の動態
マウス膀胱におけるコドン最適化マウスIl12p40p35遺伝子発現の動態を評価するために、健常雌C57Bl/6Jマウス(12~16週間)は、麻酔下、膀胱内点滴注入(曝露時間=60分)により、IL-12/eRNA11a/VA1を含むRXGナノ粒子、または空ベクター対照DNAを含有するポリプレックスの1回の膀胱内点滴注入(IVI)を受けた。動物は、0.25mg/mL、c250でmEG-70プロトタイプまたは対照ナノ粒子(RXG-PEG-N9)の投与を受けた。投与後の指示時間に、膀胱組織を採取し、RNA抽出緩衝液の存在下で均質化することにより、組織を溶解させた。1ugの投入RNA及びコドン最適化mIL-12p40p35を認識するTaqManプライマー/プローブを使用して、RT-qPCRを実施した。in vitro転写により生成されたマウスIl12p40p35 RNAの標準曲線を使用して、絶対定量を実施した(図23)。

マウス膀胱におけるマウスIL12p70タンパク質発現の用量依存的発現があるかどうかを評価するために、健常雌C57Bl/6Jマウス(12~16週間)は、0.1~80μgのプラスミドDNAの用量でmEG70プロトタイプナノ粒子の単回IVIを受けた。陰性対照は、最大20μg投与された。投与の48時間後に膀胱組織を採取し、MSDプラットフォームでのマウスIL12p70イムノアッセイのために組織溶解物を調製した。
in vivoでのPEG化ナノ粒子の発現

NMIBCにおけるヒト臨床研究
膀胱癌は、米国(US)の男性及び女性で、それぞれ、4番目及び10番目に最も一般的な悪性腫瘍である(American Cancer Society 2019)。非筋肉浸潤性膀胱癌(NIMBC)は、通常、再発率を35%低減させるために、外科的切除(TURBT)で、続いて、多くの場合、24時間以内の膀胱内化学療法(ゲムシタビンまたはマイトマイシン)の単回投与により、管理される(Sylvester et al、2016)。

Claims (32)
- 誘導体化キトサン核酸ポリプレックスを含む組成物であって、アミノ官能基化キトサン及びインターロイキン12(IL-12)をコードする少なくとも1つの治療用核酸構築物を含み、前記誘導体化キトサン核酸ポリプレックスが、さらに、少なくとも1つのポリアニオン性アンカー領域及び少なくとも1つの親水性テール領域を有する1つ以上のポリアニオン含有ブロックコポリマーを含む可逆的なコーティングを含む、前記組成物。
- 前記アミノ官能基化キトサンが、さらに、親水性ポリオールを含むか、またはそれで官能基化されている、請求項1に記載の組成物。
- 前記アミノ官能基化キトサンが、アルギニンを含む、請求項1に記載の組成物。
- 前記親水性ポリオールが、グルコースまたはグルコン酸である、請求項2に記載の組成物。
- 前記ポリアニオン含有ブロックコポリマーが、線状二元ブロックまたは三元ブロックコポリマーである、請求項1に記載の組成物。
- 前記ポリプレックスが、さらに、IFN-1アクチベーター及び免疫チェックポイント阻害薬からなる群より選択される追加の免疫刺激分子をコードする核酸を含む、請求項1~5のいずれか1項に記載の組成物。
- 前記治療用核酸構築物が、さらに、IFN-1アクチベーター及び免疫チェックポイント阻害薬からなる群より選択される追加の免疫刺激分子をコードする核酸を含む、請求項1~5のいずれか1項に記載の組成物。
- 前記IFN-1アクチベーターが、RIG-Iアゴニスト、STINGアゴニスト、及びTLR7/9アゴニストからなる群より選択される、請求項6または7に記載の組成物。
- 前記RIG-Iアゴニストが、eRNA11a、VA RNA1、eRNA41H、MK4621、SLR10、SLR14、及びSLR20からなる群より選択され、より好ましくは、eRNA41HまたはeRNA11aからなる群より選択される、請求項8に記載の方法の組成物。
- 前記治療用核酸構築物が、gWIZ、pVAX、NTC8685、またはNTC9385Rからなる群より選択されるプラスミド内に含まれ、任意に、前記治療用核酸構築物が、さらに、CMV、EF1a、CMV/EF1a、CAG、及びCMV/EF1a/HTLVプロモーターからなる群より選択される発現制御エレメントを含む、請求項1に記載の組成物。

- IL-12をコードする前記治療用核酸構築物が、配列番号7を含む、請求項1~12のいずれか1項に記載の組成物。
- 方法を必要とする患者の粘膜組織におけるIL-12の局所発現のための方法であって、請求項1~13のいずれか1項に記載の治療的有効量の医薬組成物を前記患者に投与することを含む、前記方法。
- 方法を必要とする患者の粘膜癌細胞の成長を抑制するための方法であって、請求項1~13のいずれか1項に記載の治療的有効量の医薬組成物を前記患者に投与することを含む、前記方法。
- 方法を必要とする患者の膀胱癌を処置するための方法であって、請求項1~13のいずれか1項に記載の治療的有効量の医薬組成物を前記患者に投与することを含む、前記方法。
- さらに、IFN-1アクチベーター及び免疫チェックポイント阻害薬からなる群より選択される追加の免疫刺激分子の同時または逐次投与を含む、請求項14~16のいずれか1項に記載の方法。
- 前記IFN-1アクチベーターが、RIG-Iアゴニスト、STINGアゴニスト、及びTLR7/9アゴニストからなる群より選択される、請求項17に記載の方法。

- 前記IFN-1アゴニストが、RIG-Iアゴニストであり、好ましくは、前記RIG-Iアゴニストが、eRNA11a、VA RNA1、eRNA41H、MK4621、SLR10、SLR14、及びSLR20からなる群より選択され、より好ましくは、eRNA41HまたはeRNA11aからなる群より選択される、請求項18に記載の方法。
