Toll様受容体(TLR)は、病原体関連分子パターン(PAMP)として知られる、保存された微生物産物を認識するパターン認識受容体である。TLR4はLPSを認識する。TLR4シグナリングは、MyD88およびTRIFの依存性経路を活性化させる。MyD88経路は、NF-κBおよびJNKを活性化して炎症反応を誘導する。TRIF経路は、IRF3を活性化してIFN-αの生産を誘導する。

一実施形態において、哺乳動物における免疫応答を増強するための方法が提供される。
<リポソーム製剤化2B182c、TLR4アゴニスト、および1V270、TLR7アゴニストのアジュバントポテンシー(potency)>
2B182c(200nmol/注射)および1V270(1nmol/注射)のリポソーム製剤単独、または200nmol 2B182cおよび1nmol 1V270の組み合わせを調製した(Inimmune Corp, Missoula, MT)。リポソーム製剤化アジュバントのアジュバントポテンシーをDMSO製剤(10%DMSO)と比較した。製剤化アジュバントを、同じプロトコルを使用して試験した。簡単に述べると、雌BALB/cマウスに、0日目および21日目に、リポソーム製剤化2B182c(200nmol/注射)および/または1V270(1nmol/注射)を用いて不活化インフルエンザウイルスで免疫付与した。ELISAにより、抗HAおよび抗NA抗体(IgM、IgG1およびIgG2a)について血清を評価した。製剤化アゴニストが胚中心B細胞および形質芽球(抗原分泌細胞)集団に影響を及ぼすかどうかを調べるために、鼠径リンパ節を採取し、FACSによりB細胞集団について分析した。
合成小分子であるTLR4アゴニストおよびTLR7アゴニストの組み合わせは、組換えインフルエンザウイルス血球凝集素の強力なアジュバントであり、同種、異種、および異種サブタイプのマウス攻撃モデルにおいてインフルエンザウイルスに対して防御的である迅速かつ持続的な免疫を誘導する。しかしながら、これらの研究で用いられたTLR4アゴニストは、ピリミドインドールクラスの第一世代の主要な合成TLR4アゴニスト、1Z105であり、自然免疫受容体アゴニストとして作用するアジュバントを発見するためのハイスループットスクリーニングキャンペーンで特定されたヒットから最適化されたものであった。1Z105はマウス細胞において良好な免疫活性を有することが分かったが、ヒト細胞において有意な活性はなかった。より最近の研究ではC8-アリール置換基を含む第2世代のシリーズの化合物がマウス細胞において1Z105よりも強力であったが、ヒト細胞においても非常に活性であった。C8-アリール誘導体のこの活性なグループ内で、C8-フラン-2-イル誘導体(2B182C)を、ポテンシーおよび好ましい予備製剤データに基づいて、さらなる研究のために選択した(図34および36)。比較のために、ピリミドインドール2B182Cを1V270と組み合わせて評価した。強力なTLR4アゴニストであるMPLAアナログ(MPLA-1)は、インビボでの同種および異種インフルエンザ攻撃に対して良好な防御を示した。
TLR4/TLR7アゴニスト複合アジュバントによって誘導される、インフルエンザウイルス感染に対する防御免疫応答のプロファイルを評価するために、マウスに低用量(0.2μg/注射)の組換え血球凝集素(rHA)、体液性応答および致死性ウイルス攻撃に対する防御で免疫付与した(図34A-2D)。複合アジュバントを用いてrHAで免疫付与されたマウスは、最小の体重減少およびより高い生存率を示した(図34Bおよび2C)。複合アジュバントおよび、1V270、TLR7アゴニスト単独で、Th1偏向免疫応答を引き起こした。
前述したように、SAR研究では、ヒトおよびマウスの免疫細胞において、1Z105に比べて、インビトロでより高いポテンシーを示す2B182Cが得られた。インビトロの研究で観察されたより高いポテンシーがインビボでも再現性があるかどうかを調べるために、雌Balb/cマウスを、TLR4アゴニスト(1Z105または2B182C、40または200nmol/注射)および1V270(リン脂質TLR7アゴニストコンジュゲート、0.2または1nmol/注射)を用いて0日および21日に、不活化インフルエンザウイルス(A/California/04/2009(H1N1)pdm09、Cat# NR-49450、BEIリソース)で筋内(IM)免疫付与した(図34A)。28日目に血清を採取し、ELISAにより抗血球凝集素(HA)および抗ノイラミニダーゼ(NA)抗体(IgM、IgG1およびIgG2a)を測定した。1V270、1Z105および2B182CをDMSOに溶解し、希釈し、DMSOの最終濃度をビヒクル対照として10%使用した。同様の結果を示す4つの独立した実験からデータをプールした。
単一のアジュバントとして、0.2nmolおよび1nmolの1V270、並びに40nmolおよび200nmolの2B182Cまたは1Z105を比較した(図34B)。TLR4アゴニスト、1Z105および2B182Cは共に、HAおよびNAに対して有意に高いレベルのIgG1を誘導した。IgG2a誘導については、0.2および1nmol/注射の1V270の両方が抗HAを有意に増加させた(p<0.05)。一方、200nmol/注射では、1Z105ではなく、2B182Cのみが抗NA Absを増強した(p<0.01)(図34B)。2B182Cおよび1Z105は類似のレベルのHA特異的IgG2aを誘導した。いずれのアジュバント処置によっても、IgM応答に差は認められなかった。これらのデータはTLR4アゴニストがIgG1の生産量を増加させ、TLR7アゴニストがIgG2aの分泌に効果を発揮し、2B182Cがインビボの1Z105と同等またはやや高いポテンシーを発揮したとの報告を支持する。
次に、DMSO-水製剤を用いて、複合アジュバントのポテンシーを評価した。1V270と1Z105および2B182Cとの複合アジュバントはいずれも、HAおよびNAの両方に対するIgG1の誘導を改善した。2B182Cは40および200nmolの両方で1Z105より有意に高いIgG1を増強した(p<0.05、図35Aおよび35B)。IgG2aの誘導では、2B182Cは抗HAおよび抗NAのAbsのレベルを上昇させたが、1Z105はほとんどのケースで失敗した(図35Cおよび35D)。これらのアジュバントは、IgM放出に対して最小の効果を示した(図35Eおよび35F)。
ナノ粒子製剤中のMPLA-2、MPLAの硫酸アナログ、組み合わせ、およびすべての主要なTLRアゴニストについてのインビボ評価を行う。NIAIDアジュバントの発見・開発契約の中で、強力なTLR4アゴニストが発見された。そこでは、インビトロ(hPBMC)におけるインフルエンザ関連サイトカイン生産の相乗的な増強、マウス及びブタでの1V270を用いたインビボのIgG2A抗体およびHI力価の増強がないにしても、添加剤が実証された。アジュバントとしてのMPLA-1の主な弱点は、水性媒体中で加水分解しやすいので、化学的安定性がないことである。非特異的抵抗の予備的マウス研究において、MPLA-2は等量のMPLA-1よりも致死的なインフルエンザ攻撃からマウスを防御した。したがって、MPLA-2は次世代TLR4アゴニストを代表する。
<TLR4/TLR7の組み合わせの製剤の開発>
<タスク1A:主要な化合物単独および組み合わせのコロイド安定性ナノ粒子製剤の開発>
粒子送達システムは、我々の免疫系がパターン認識受容体(PRR)を介して認識し、闘うために進化したウイルス性および細菌性の病原体のサイズおよび形状を模倣することを介してアジュバントとして作用する。過去30年間にわたる研究により、生分解性であり、ワクチン抗原送達に適した多数のナノ粒子およびマイクロ粒子ベースのシステムがもたらされた。ワクチン送達システムとしてのそれらの有用性はリポソーム、ビロソーム、Iscom、エマルジョン、ウイルス様粒子(VLP)、固体脂質ナノ粒子(SLN)およびポリ乳酸コグリコール酸(PLGA)ポリマーを用いて文献で実証されており、それぞれのタイプの例はヒトの臨床試験に進んでいる。粒子状送達ビヒクルの主要なアジュバント機構は、APCによって取り込まれた粒子または関連する抗原の取り込みの増強であると考えられる。抗原へのPAMPの添加は、自然免疫細胞の活性化につながるTLRおよび他のPRRのライゲーションを介して、強固な自然免疫応答および適応免疫応答を促進することが現在十分に確立されている。多くのPAMP(細菌性リポタンパク質、糖脂質、DNAおよびウイルスRNAなど)が同定され、ウイルス性および細菌性の病原体から分離されている。これらのアゴニストの多くは強力なアジュバントであるが、許容できないレベルの炎症をもたらすか、または臨床開発のために好ましくない物理的/化学的特性を有する。これに応答して、研究者らは改善された安全性および化学的プロファイルを有する合成アナログを首尾よく生産し、そしてこれらの多くは、PRRライゲーションを介してそれらの病原体模倣を増強するために、粒子送達システムに添加されている。粒子送達システムはまた、インビボでのアジュバントの生体内分布動態を改善し、アジュバントの免疫原性を犠牲にすることなくアジュバントの副作用を低減するために使用され得る。

5.3~10mLスケールの主要な製剤のロットを、超音波処理法よりも粒度均一性(多分散指数、PDI)を改善するLipex押出機を用いて調製する。
製剤の安定性は製品の貯蔵、出荷、および貯蔵寿命に影響を及ぼし、すべてが製品コストに直接寄与するので、成功する市販製品の開発に必要である。製剤は特に、さらに追求する主要な候補を選択する場合に、潜在的な製品として、並びに安定性として適切であることが実証される。
1)IM注射用に50uL中に含まれる2B182Cについて200nmol、および1nmolの1V270の標的濃度を有する2mLスケールでの主要なアジュバント製剤スクリーニング(薬学的に許容される共溶媒、賦形剤、リポソーム)。
2)主要な製剤について、基本的な分析法開発と分析を行い、製剤が品質基準を満たしていることを確認する。
3)好ましい製剤の安定性をリアルタイムおよび加速法により評価し、必要に応じて適切な賦形剤を安定剤として添加する。
4)最終製剤のエンドトキシン汚染がないことを確認するためのrFC試験。
信頼性の高いバイオマーカーを明らかにすることは、安全で有効なワクチンの開発を成功させるために必要である。ワクチン開発プログラムには、安全性に問題がなく、効果的に感染を予防するプロファイルを有するワクチン候補を選択することが不可欠である。ワクチンの臨床試験では、反応原性を最小限に抑えて抗原特異的な適応免疫応答を予測するバイオマーカーの同定が必要である。このプロジェクトでは、バイオマーカーが1)抗原の有無にかかわらず、製剤化主要なアジュバントによって誘導される自然免疫バイオマーカー、および2)適応免疫応答に相関するバイオマーカーの2つのステップで同定される。したがって、TLR4およびTLR7/8リガンドの両方の生物学的活性に相関し、かつ反応原性にも関連するバイオマーカー候補を同定するために、インビトロおよびインビボの研究が実施される。
ワクチン接種を介した感染症からの防御の特徴は、有効な抗体生産の誘導である。TLR4アゴニストおよびTLR7アゴニストを併用すると、抗原特異的抗体価が有意に増加した。免疫応答のTh1偏向へ向かう傾向が観察された。製剤化アジュバントおよびそれらの組み合わせの有効性を、単純なDMSO-水調製物と比較する。
主要なアジュバントの製剤は、DMSO-水製剤について以前に完了したのと同様の様式で、単独で、および様々な比率のTLRアゴニストと組み合わせて、免疫付与試験で評価される。HAおよびNAの両方に特異的なIgM、総IgG、並びにIgG1およびIgG2aのレベルを評価する。抗原特異的抗体の最大力価を提供する組み合わせにおけるTLRアゴニストの1つ以上の比率を同定する。この製剤は、研究領域3の下での研究に挑戦するために進められるだろう。
感染症ワクチンは、健常者の集団において予防効果があるように設計されているため、ワクチンの安全性は開発目標の中で最も優先度が高いものでなければならない。したがって、候補製剤の毒性および反応原性を評価するための適切な実験が行われる。これらの実験および一般に、明白な毒性は、初期毒性評価として厳密に評価される。マウスにおける何らかの苦痛の徴候(すなわち、毛づくろいの欠如、可動性の問題、結膜、異常行動、反応性など)に注意する。全体的な観察に加えて、毒性測定は、完全血球数、血清化学評価(AST、ALT、ALP、アミラーゼ、血中尿素窒素、クレアチニン、総タンパク質、グルコース、カリウム、カルシウム、ナトリウム、総ビリルビン)および剖検評価(ヘマトキシリンおよびエオシンで染色した脾臓、肝臓および腎臓の切片)を含む。さらに、注射部位について、炎症の目に見える徴候および他の異常所見がないか評価する。剖検評価の一環として、注射部位の組織も組織学的に評価する。これらの研究を以下の表6に要約する。
前述したように、抗原特異的な適応免疫応答を最小限の反応原性で予測するバイオマーカーの同定は、臨床試験の設計および方法を容易にする。
ケモカインによる局所的ワクチン投与部位への免疫細胞動員は抗原提示細胞(APC)を動員し、その後の適応免疫応答の誘導に影響を与えるために必須である。しかし、注射部位、すなわち筋肉組織は比較的少数の免疫細胞を含んでいるので、効果的なアジュバントは局所への免疫細胞の動員を誘導しなければならない。TLR4はTLR7/8とは異なり、非免疫細胞上に豊富に発現し、炎症細胞を動員するのに十分なケモカインを発現することができる。TLR刺激の後、APCの動員は通常、炎症細胞に付随するため、炎症反応をアジュバント効果と区別することは困難である。これらの免疫活性化の複雑なカスケードは、インビトロアッセイだけでは研究できない。従って、マーカーのパネルは、マウスにおけるインビボ研究から得られたサンプルにおける上記のインビトロ実験から選択される。
適応免疫応答を評価するための実験は、上記のタスク2A.1と併せて実施される。次の基準を満たすバイオマーカー候補が同定される:1)末梢血で検出される、2)各TLRリガンドの作用機序により駆動され、それらの生物学的効果を相関させる、3)長期的な抗原特異的抗体誘導および広範な防御を予測する、4)反応原性を予測する。
組み合わせたTLRアゴニストの1つ以上の比率は、抗原特異的抗体の最大力価を提供する。さらに、2つのクラスのTLRリガンドの組み合わせの使用は、TLR4アゴニスト単独の使用と比較して、適応免疫応答のTh1偏向応答へのシフトをもたらす。したがって、Th1/Th2応答比は増加する可能性が高い。このTh1偏向は、異種ウイルス防御を含むように応答を広げるのに有利であり得る。毒性に関しては、イミダゾキノリンクラスのTLR7/8リガンドの全身および経口投与が高レベルの血清TNFαおよびIL-1βを伴うインフルエンザ様症状、悪心およびリンパ球減少症を含む重篤な副作用を示している。これは、1V270がメンバーであるオキソアデニンクラスにも当てはまる。しかしながら、これらの望ましくない副作用の大部分は、ワクチン接種のための通常の局所投与経路、IMを使用することによって回避することができる。さらに、TLR7/8リガンドは、脂質部分へのコンジュゲーションによって、並びに製剤をカスタマイズすることによって調製され、アジュバント活性を維持しながら全身性サイトカイン放出を首尾よく減少させた。したがって、炎症性サイトカインへの全身暴露が低いため、主要な製剤化の組み合わせに関連する反応原性はほとんどまたは全くないであろう。
マウスを用いた前臨床免疫付与/ウイルス攻撃研究では、安定性(タスク1B)、免疫活性(タスク2A.1)および反応原性プロファイル(タスク2A.2)を含む製剤研究の結果に基づき、バックアップの組み合わせとともに主要な製剤の組み合わせが選択される。研究に使用されるウイルス抗原は、A/Victoria/3/75(H3N2)、A/Michigan/45/2015(H1N1)pdm09様ウイルスおよびA/Hong Kong/4801/2014(H3N2)様ウイルスなどのライセンスされた市販ワクチンに使用されている組換えワクチン抗原または不活化された全ウイルスのいずれかから選択され得る。
主要な選択基準は、1)製剤化組み合わせの安定性、2)所望の抗原特異的抗体レベルを提供するTLRアゴニストの比率、および3)局所および全身の両方での低い反応原性プロファイルに基づく。これらの基準に関連する特定の研究を表7に要約する。
<タスク3B.1:ウイルス攻撃研究のための最小防御用量の判断>
不活化インフルエンザウイルスは自然免疫受容体リガンド(PAMPS)を含むため、十分な抗原のみでマウスに免疫付与した後、一定の低レベルの防御が期待できると思われる。したがって、(もしあれば)不活化ウイルスを用いて、最小防御用量を判断するための研究を実施する。抗原の最小防御用量は、活性ウイルスの適合株によるその後の攻撃時に部分的防御(生存率30%未満)しかもたらさない用量である。この戦略は、選択された主要な製剤化アジュバントの組み合わせで観察されるべき範囲の活性を可能にする。さらに、攻撃ウイルスの量はまた、免疫付与されていないマウスについて完全な死亡率(代表的には、約5LD50の用量)をもたらすことが確認され得る。
抗原用量の範囲を見出す研究に続いて、マウスモデルを用いて、同種インフルエンザワクチン抗原とともに主要なアジュバントの組み合わせの免疫原性を評価する。成功の主な判断因子は、1)血清中の持続性インフルエンザ特異的IgG2aおよびIgG1、2)致死性インフルエンザウイルス攻撃からの防御、3)低い反応原性、4)細胞内IFNγ/TNFα染色で評価した多機能性CD4+対CD8+T細胞の誘導である。副次的評価項目は、ワクチン接種またはインフルエンザウイルス攻撃後の体重増加/減少および観察可能な臨床症状(波状被毛、円背姿勢および努力性呼吸)のモニタリングによる疾患重症度のスコア化である。
免疫学的評価:マウス(雌雄)にワクチン接種(アジュバント+A/Victoria/3/75(H3N2)等のインフルエンザ抗原)を1回又は2回、1回目接種と2回目接種との間に21日間をおいてIM投与する(図12)。細胞性免疫(CMI)は脾臓細胞培養物におけるTh1/Th2サイトカイン誘導(ELISAにより分析)および多機能性CD4+およびCD8+T細胞応答(FACSにより分析、10色細胞内サイトカイン染色)を測定することにより、1群4匹のマウスのサブセットで評価される。さらに、四量体染色および細胞表面表現型検査を行い、インフルエンザ特異的メモリーCD4+およびCD8+T細胞の頻度を測定する。インフルエンザ特異的体液性応答は血清中(IgG1およびIgG2a)で測定され、HI力価は機能的抗体価を測定するために用いられる。ワクチン接種したマウスおよび対照のマウスの各群は5LD50のA/HK/68(H3N2)で攻撃され、生存率、体重増減、及び疾病重症度を21日間評価した。これらのマウス研究における反応原性は、体重減少および症状スコアと注射部位浸潤の評価とによって測定される。p値の差が<0.05であれば、有意であるとみなす。すべてのデータについて分散分析(ANOVA)およびTukey ANOVAを実施し、頑健な統計的有意性を実証する。
同種防御研究に続いて、異種または異種サブタイプ防御のマウスモデルにおける主要なアジュバントの組み合わせを評価するために同じ研究デザインが用いられる。マウスに上記のように免疫付与するが(タスク3B.2)、異なったHA/NAタイプのインフルエンザウイルス株(例えばA/Puerto Rico/8/1934(H1N1))で攻撃する。このような攻撃モデルにおいて観察される防御は、HAタンパク質の茎領域のような、両方の株に共通の抗原を含む抗原特異的応答の広がりを示唆する。このような広がりを確認するために、B細胞受容体(BCR)およびT細胞受容体(TCR)の配列の研究が行われる。
前述したように、増えつつある文献報告では、ワクチンを介した免疫の大きさを相乗的に増大させ、それによって生じた下流の適応免疫応答のタイプを変更させ、それによってこれらのワクチンのエフィカシーを高めることができる様々なTLRアゴニストの組み合わせが引用されている。インフルエンザウイルス攻撃防御のためのアジュバントの組み合わせは、本明細書中に記載される。
<ワクチン抗原としてのインフルエンザ血球凝集素(HA)>
広範に中和茎抗体を増強する戦略には、1)頭部ドメイン全体を除去して茎ドメインをより「利用可能」にし、従って茎ドメインに対する抗体応答を誘導するヘッドレスHAに焦点を当てること、または2)H1、H3またはB型インフルエンザウイルス由来の茎ドメインからなるキメラHAを組み合わせて使用すること、が含まれる。
毎年のインフルエンザワクチンの有効性は、インフルエンザウイルスの抗原浮動のため、依然として10~60%と評価されている。合成TLR4およびTLR7アゴニスト(1Z105および1V270)は、それぞれTh2およびTh1を介した抗血球凝集素抗体生産を増強した。1Z105と1V270との組み合わせは、インフルエンザウイルス感染を防御するために、バランスのとれたTh1/Th2免疫を促進した。アジュバントのエフィカシーを高めるために、1Z105および2B182Cについて構造活性相関研究を実施した。これは、インビトロでよりポテンシーの高い誘導体であった。モデル抗原であるオボアルブミンを用いたインビボワクチン研究では、2B182Cは、より高い血清IgG1レベルを誘導し、1V270によって誘導される抗原特異的IgG2aの放出を追加的に増強した。さらに、2B182Cおよび1V270のリポソーム製剤は、細胞毒性および反応原性を低下させ、Th1を介した抗体生産およびTh2を介した抗体生産の両方を増強する活性を維持した。不活化A/California/04/2009(H1N1)(pdm09)を抗原としたインビボワクチン接種研究において、リポソーム組み合わせアジュバントはT濾胞ヘルパー細胞、胚中心B細胞及び抗体分泌形質細胞の集団を増加させた。流入鼠径リンパ節におけるBおよびTリンパ球の次世代配列分析は、複合アジュバントが免疫グロブリン重鎖(IGH)遺伝子のB細胞クローン型を増加させ、B細胞クローンおよびTCRクローナリティを共有することを示した。これらの知見は、組み合わせが抗原特異的Th1免疫応答を増強することに寄与することを示唆した。最後に、組み合わせアジュバントを有するワクチンは、肺のダメージがより少ない致死的な同種ウイルス攻撃に対して防御した。
<マウス>
雌の6~8週齢のBALB/cマウスを、Jackson labolatory(Bar Harbor, MA)から購入した。カリフォルニア大学サンディエゴ校動物施設において、オボアルブミンまたは不活化インフルエンザウイルスを抗原とし、生ウイルス攻撃を必要としない動物実験を実施した。インフルエンザ攻撃研究はユタ州立大学のAnimal Research Centerにより、6週齢の雌BALB/cマウス(Charles River Laboratories, Wilmington, MA)を用いて行った。すべての動物実験は、UC San Diegoまたはユタ州立大学のInstitutional Animal Care and Use Committee(IACUC)による事前承認を受けた。
TLR4/NF-kBレポーター細胞株HEK-BlueTMヒトTLR4およびHEK-BlueTMマウスTLR4細胞はInvivoGen(カタログ番号、San Diego, CA)から購入した。マウス初代BMDCを、C57BL/6マウスの大腿骨から採取した骨髄細胞から調製した。BMDCを、10%FBS(Omega, Tarzana, CA)およびペニシリン/ストレプトマイシン(100ユニット/mL/100μg/mL、Thermo Fisher Scientific, Waltham, MA)を補充したRPMI中の示された化合物で処理した。モノリン脂質A(MPLA)、AddaVaxは、InvivoGen(カタログ番号San Diego, CA)から購入した。不活化インフルエンザA型ウイルス[A/California/04/2009(H1N1)pdm09](IIAV)は、BEIリソース(#NR-49450, Manassas, VA)から入手した。TLR7アゴニスト1V270、TLR4アゴニスト1Z105および2B182Cを含むその誘導体を合成した。1V270(20μM)、2B182C(4mM)および1V270+2B182C(20μM+4mM)のリポソーム製剤をInnimune corp.(Missoula, MT)で行った。
TLR4/NF-κB活性化を、HEK-BlueTM hTLR4およびHEK-BlueTM mTLR4(InvivoGen)を用いて評価した。細胞を1Z105および2B182C(10μMから始まる2倍連続希釈)で20時間処理した。培養上清中のNF-κB誘導性分泌胚性アルカリホスファターゼ(SEAP)タンパク質を、製造業者のプロトコルに従って測定した。
BALB/cマウスを、0日目および21日目に、IAV(10μg/注射)に加えて指示されたアジュバントを後肢の腓腹筋に、筋肉内(i.m.)で免疫付与した。各処置におけるアジュバントの詳細な濃度および動物の数を、各図の凡例に記載する。28日目に血清を採取し、抗原特異的抗体(抗HA IgG1、抗NA IgG1、抗HA IgG2a、抗NA IgG2a、抗HA IgMおよび抗NA IgM)について評価した。これらの抗体についてのELISAを、以前に記載されたように行った(参考文献)。DMSO製剤を用いた研究のために、10%DMSOをビヒクルとして使用した。リポソーム製剤化アジュバントを使用する実験において、1,2-ジオレオイル-sn-グリセロ-3-ホスホコリンおよびコレステロール(DOPC/Chol、対照リポソーム)をビヒクルとして使用した。
免疫付与プロトコルを図28Aに示した。簡潔には、マウスを28日目に屠殺し、鼠径リンパ節を採取した。RNeasy Mini Kit(Qiagen, Hilden, Germany)を用いてリンパ球(バルク)から全RNAを抽出し、Agilent 4200 TapeStation(Agilent, Santa Clara, CA)によりRNAの品質を確認した。次世代シーケンシングを、偏りのないTCRレパトア分析技術(Repertoire Genesis Inc., 大阪、日本)を用いて行った。
BALB/cマウスに、製剤化1V270および2B182Cを用いてIIAV(3ug/注射)で0日目にi.m.ワクチン接種し、21日目に同種または異種インフルエンザA型ウイルス、A/California/04/2009(pdmH1N1)およびA/Victoria/3/75(H3N2)それぞれを鼻腔内に感染させた。同種ウイルスの攻撃から30~50%の動物を防御するIIAV;3μg/注射の免疫付与用量を予備実験で判断した。インフルエンザウイルス攻撃のために、マウスの群を、ケタミン/キシラジン(50mg/kg/5mg/kg)の腹腔内注射によって麻酔した後、マウス1匹あたり1×105(3×LD50)の細胞培養感染用量(CCID50)のインフルエンザA/California/04/2009(H1N1pdm)ウイルス;マウス1匹あたり5×102(3×LD50)CCID50のインフルエンザA/Victoria/3/75(H3N2)ウイルスを90μL懸濁液中で鼻腔内攻撃した。研究21日目に全てのマウスにウイルス攻撃を行った。株指定175190のインフルエンザウイルス(H1N1pdm)は、Dr.Elena Govorkova(Department of Infectious Diseases, St. Jude Children’s jemResearch Hospital, Memphis TN)から受領した。このウイルスはマウスの9継代によりBALB/cマウスの肺での複製に適応した。ウイルスはMadin-Darbyイヌ腎臓(MDCK)細胞においてプラーク精製され、そしてウイルスストックは胚化ニワトリ卵、次いでMDCK細胞における増殖によって調製された。インフルエンザA/Victoria/3/75(H3N2)ウイルスは、American Type Culture Collection(Manassas, VA)から入手した。このウイルスは当初マウスには致死的ではなかったが、感染動物の肺で7回連続継代すると致死となった。マウス適応後、ウイルスストックを、MDCK細胞中での増殖によって調製した。
ウイルス攻撃6日後、採血直後に気管支肺胞洗浄(BAL)処置を行い、各動物の死亡から5~10分以内に終了した。0.75mLの体積のリン酸緩衝生理食塩水(PBS)を、気管チューブを通して肺にゆっくりと送達した。送達直後に、穏やかな吸引によって流体をゆっくりと引き出し、サンプルを-80℃で保存した。この手順を合計3回繰り返し、各マウスからの洗浄液をプールした。肺ウイルス力価を測定するために、BALサンプルを2000gで5分間遠心分離した。様々な10倍希釈のBAL上清を、MDCK細胞中の感染性ウイルスについて3連でアッセイし、ウイルス力価を計算した。肺サイトカインレベルの測定のために、各々の肺洗浄からのサンプル(200μL)を、製造業者の指示書に従って、化学発光多重ELISAベースのアッセイを使用して、MCP-1およびIL-6について試験した(Quansys Biosciences Q-PlexTM Array, Logan, UT)。
血球凝集抑制(HI)力価については、血清を受容体破壊酵素II(RDE;Vibrio cholerae neuraminidase;YCC-340;Accurate Chemical and Scientific, Westbury, NY)で前処理して、1部の血清を3部の酵素で希釈し、37℃で18時間インキュベートすることによって非特異的阻害剤を除去した。続いて、RDEを56℃で45分間加熱することによって不活性化した。血清サンプルを、96ウェル丸底マイクロタイタープレート(Fisher Scientific, Pittsburg, PA)中のPBS中で希釈した。血清の希釈後、8HAユニット/ウェルのインフルエンザA/CA/04/2009(H1N1pdm)またはインフルエンザA/Victoria/3/75(H3N2)ウイルスに加えて七面鳥赤血球(Lampire Biological Laboratories, Pipersville, PA)を添加し(ウェル当たり50μL)、短時間混合し、室温で60分間インキュベートした。血清サンプルのHI力価は、血球凝集が完全に阻害された最高血清希釈の逆数として示される。
抗インフルエンザウイルス中和抗体アッセイのために、MDCK細胞を、使用の24時間前に、5%FBS(Hyclone, Logan, UT)を含むMEM中、1ウェルあたり1×104細胞で96ウェルプレートに播種した。血清サンプルの連続2倍希釈物を、1:32希釈で開始し、1:4096で終了する、10ユニット/mLトリプシンおよび1μg/mL EDTAを含有する無血清培地中で調製した。各血清希釈物を、約100CCID50/ウェルH1N1pdmまたはインフルエンザA/Victoria/3/75(H3N2)ウイルスを含有する無血清培地(トリプシンおよびEDTAを含む)と1:1(0.1mL)で混合した。室温で1時間インキュベートした後、血清-インフルエンザウイルス混合物(0.2mL)を、MDCK細胞を含むウェルに移し、3日間インキュベートした。抗インフルエンザウイルス中和抗体を細胞変性効果(CPE)阻害として測定した。感染後3日目に光学顕微鏡下でMDCK細胞単層を調べることによって、CPEを二重サンプルからスコア付けした。
インビボ研究で得られたデータは平均標準誤差(SEM)とともに平均値として提示され、インビトロデータは標準偏差(SD)とともに平均値として示される。インビトロデータについては、Welchの両側t検定を用いて2群を比較した。抗原特異的抗体については免疫細胞集団に対するフローサイトメトリー解析、BCR-seq、TCR-seq、肺ウイルス力価、HIエンドポイント力価及びVNエンドポイント力価についてはDunnの事後検定とともにKruskal-Wallis検定を適用した。肺ウイルス力価とサイトカイン/ケモカインレベルとの相関を、Spearman順位相関検定を用いて解析した。体重については、曲線下面積を各マウスについて計算し、一元配置ANOVAを統計解析に使用した。Kaplan-Meier生存曲線間の有意差の検定にはログランク(Mantel-Cox)検定を用いた。Prism 5ソフトウェア(GraphPad Software, San Diego, CA)を使用した。P値が0.05未満の場合、統計的に有意であるとみなした。
<1Z105の構造活性相関の研究から2B182Cが得られた>
小分子ピリミドインドールTLR4リガンド、1Z105に対するポテンシーを改善するために、構造活性相関分析を行った(化学者が補うであろう)。合計56の化合物を合成し、ヒトおよびマウスのHEK TLR4レポーター細胞(それぞれ、HEK-Blue mTLR4およびhTLR4)によってスクリーニングした。これらのSAR化合物の中で、2B182Cは、マウスおよびヒトの両方のレポーター細胞において、より高いTLR4刺激ポテンシーを有する誘導体として発見された。2B182CのEC50を、HEK TLR4レポーター細胞を用いて調べ、1Z105のEC50と比較した(図21B)。マウスおよびヒトのTLR4レポーター細胞における2B182CのEC50は、1Z105のEC50と比較して、それぞれ5.8倍および870倍増加した。これらのデータは、SAR研究がより高いTLR4刺激ポテンシー、特にヒトTLR4ポテンシーを示す誘導体を首尾よく生じたことを示す。
TLR4アゴニスト1Z105はTh2を介したIgG1生産を誘導し、TLR7アゴニスト1V270はインフルエンザウイルスに対するTh1細胞免疫を増強した(Goff et al., J. Virol., 89:3221 (2015); Goff et al., J. Virol., 91:e01050 (2017))。1V270と組み合わせることで、インフルエンザワクチンアジュバントとしてのTLR4アゴニスト2B182Cのエフィカシーを向上できると仮定した。したがって、1V270を伴う2B182Cが、1Z105と1V270とを用いたコンボアジュバントと比較して、インビボでのアジュバント性を改善するかどうかを調べた。
次に、抗体生産に対するこれらのTLR4アゴニストの共アジュバント効果を、1nmol/注射の用量でTLR7アゴニスト1V270と組み合わせた場合に分析した。1V270は、Th1免疫応答を増強するIgG2a生産を誘導することが報告された(Goff et al., 2017)。結果は1V270単独では抗HA IgG2a生産のみを誘導したが、2B182Cと組み合わせると、HAおよびNAの両方に対するIgG1およびIgG2a抗体が有意に誘導されたことを示した。これは、これらの化合物が付加的な様式で作用し得ることを示唆する(図23Aおよび23B)。一方、1Z105は、1V270によって誘導されたIgG2a生産を増強しなかった。1V270+2B182Cグループの動物はより多くの量のIgG1およびIgG2aの両方を生産し、免疫バランスはTh1を介したIgG2a生産に向かう傾向があり、この治療はTh1免疫応答を増強することに寄与することを示唆している(図23C)。1V270と2B182Cとの組み合わせは、抗HA IgM生産に対して中程度の効果を示した(図24B)。
上記の結果を考慮して、1/200の1V270/2B182C比率(TLR4/TLR7)[1nmol/注射(20μmM)の1V270および200nmol/注射(4mM)の2B182C]を使用した。ワクチンに対する応答を維持しながら、望ましくない細胞毒性および反応原性を回避するために、化合物の製剤を調整することは、ワクチンアジュバントの開発において重要であり得る。従って、1V270および2B182cは、Inimmune Corp(Missoula, MT)によってリポソーム中に製剤化。製剤化化合物の活性を、マウス初代BMDCにおいて試験した。これらの製剤化化合物は、DMSO製剤化合物と同様のレベルのIL-12分泌を維持した(図25A)。DMSO-2B182CまたはDMSO-1V270+2B182Cにより誘導された細胞毒性はリポソーム製剤により有意に改善された(図25B)。注射部位における筋肉のH&E染色による組織学的分析を図25Cに示す。化合物の投与後の血清の多重サイトカイン/ケモカイン分析を図25Dに示す。
インビボでの化合物のアジュバント性を、図22Aに記載のプライム-ブーストレジメンを用いて評価した。28日目に採取した血清を、ELISAによって抗原特異的抗体について評価した。結果はリポ-2B182Cがより高いレベルのIgG1を誘導することを示し、これはDMSO-2B182Cと一致した(図26A)。DMSO-1V270とは異なり、リポ-1V270単独ではIgG2a生産を促進しなかった(図26B)。各アゴニストによるIgG2aに対するこれらの最小の効果にもかかわらず、2つのアジュバントを組み合わせた場合、抗原特異的抗体の生産は相乗的に増強された(図26B)。一方、リポソームビヒクル、1V270、2B182Cおよび1V270+2B182Cによって誘導された総IgGレベルは同等であった(図26C)。抗原特異的IgMレベルはいずれの処置によっても影響を受けなかった(図27)。DMSO製剤で観察された傾向と一致して、リポソーム複合アジュバントは、Th1偏向免疫バランスを発現した(図26D)。
製剤化アジュバントが抗原特異的抗体分泌を促進するB細胞の活性化を誘導するかを調査するために、鼠径リンパ節のリンパ球を、フローサイトメトリーを用いてTfh細胞、GC B細胞、形質芽球および形質細胞について調べた。上記の免疫付与プロトコルを使用し、鼠径リンパ節中のリンパ球を28日目に採取し、フローサイトメトリーによって分析した(図S28Aおよび28B)。その結果、CD3+ CD4+ PD-1+ CXCR5+細胞として確認されたTfh細胞の割合は、リポ-1V270+2B182Cによって大幅に増加した(図28Bおよび図29)。さらに、複合アジュバントは、GC B細胞(CD3- CD19+ CD95+ GL7+)のパーセンテージを増加させた。リポ-1V270+2B182Cを接種したマウスでは、形質芽球および形質細胞の増加が観察された。これらの結果は、複合アジュバントが単一の薬剤と比較してGC反応を増強することを示唆する。
複合アジュバントがBCRの多様性に影響するかどうかを調べるために、IGH遺伝子について次世代シークエンス分析を行った(Repertoire Genesis Inc, 大阪、日本による)。プライムブーストIIAVモデルを用い、28日目に鼠径リンパ節のリンパ球を採取した(図30A)。BCR配列分析から、Pielouの指数で示される総リード数に正規化されたBCR多様性は、リポ-1V270+2B182Cによって有意に増加したことが示された(図30A)。同様の分析によってIgG遺伝子のクローン型を分析し、グループ内の2つのマウス間でIGHクローンを比較し、共有クローンがあるかどうかを調べ、Jaccard指数を算出した;Jaccard指数;J(A、B)=(A∩B)/(A∪B)(図30B)。IGH、IGHG1およびIGHG2AのJaccard指数はリポ-1V270+2B182Cにより有意に増加し、このグループ内で2つのマウス間で共有されたクローンが増加したことを示した。さらに、リポ-1V270+2B182cグループでは、6クローン(0.03%)が3つのマウス間で共有された。これらの結果は、リポソーム複合アジュバントが総IGHおよびIGHG2AにおけるBCR多様性を増加させたことを示唆する。これは、複合アジュバントの免疫付与後のより高いIgG2aレベルと一致する。複合アジュバントで免疫付与されたグループで検出された共通のクローンは、抗原の優性エピトープを認識し得る。TCRシーケンシングを行い、製剤化アジュバントが抗原に対するf TCRクローナリティの増加に寄与するかどうかを調べた。予想された通り、複合アジュバントおよびリポ-2B182CはTCRαおよびTCRβのクローナリティを増加させた(図30C)。まとめると、リポ-1V270+2B182Cの動物は、より高いBCR多様性およびTCRクローナリティを示した。これは、Th1応答が複合アジュバントによって増強されるというデータを支持し得る。
複合アジュバントは、多様なBCRおよびTCRの高いクローナリティを伴うTh1偏向免疫応答を誘導した。この多様性がインフルエンザウイルスに対する免疫応答の兆候となり得るかどうかを試験するために、製剤化1V270および2B182cを、同種および異種のインフルエンザウイルス攻撃モデルで試験した。IIAVに加えてリポソーム1V270、2B182Cまたは1V270+2B182Cをワクチン接種したBalb/cマウスは、ワクチン接種(単回接種)後21日目に、同種(H1N1)インフルエンザウイルスで鼻腔内を攻撃した。マウスの体重および生存率をさらに21日間モニターした(図31A)。リポ-2B182Cおよびリポ-1V270+2B182Cは、ウイルス感染後の体重減少を有意に抑制した(図31B)。さらに、同種インフルエンザウイルスに対して、リポ-1V270は90%の防御を示し、リポ-2B182Cおよびリポ-1V270+2B182Cはマウスを完全に防御した(図31C)。マウスの生存率が肺におけるウイルス力価と相関するかどうかを評価するために、気管支肺胞洗浄を、洗浄液中のウイルス力価のために実施した。その結果、リポ-1V270+2B182Cは、6日目において、ウイルス力価を効果的に抑制することを示した(図31D)。ヒトでは、気道上皮細胞(例えばMCP-1、IL-6など)におけるサイトカインおよびケモカインのアップレギュレーションが致死的な肺損傷および肺炎と相関している(Gurczynski et al., Mucosal Immun., 12:518 (2019); Zhou et al., Nature, 499:500 (2013))。そこで、Quansys multiplex ELISAを用いて肺液中の炎症誘発性サイトカイン(IL-6)およびケモカイン(MCP-1)のレベルを評価した。結果は、複合リポソームアジュバントがMCP-1およびIL-6の両方の生産を有意に抑制したことを示した(図31E)。炎症誘発性サイトカインのレベルは、肺ウイルス力価と相関していた[MCP-1(P<0.0001、Spearman r=0.83)、IL-6(P<0.0001、Spearman r=0.79)](図31F)。この傾向は、リポ-1V270+2B182Cグループにおいてさらに増強された。これらの結果は、複合アジュバントが感染後のウイルスの侵入および増殖を阻害することにより肺のダメージを減少させることを示唆した。防御が血球凝集抑制力価(HI)およびウイルス中和力価(VN)に関連しているかどうかを評価するため、免疫付与後21日目に血清を採取し、HIおよびVNについて調べた(図31A)。非免疫付与グループと比較したHI力価の増加は、リポ-1V270、リポ-2B182C、およびリポ-1V270+2B182Cの20匹中19匹のマウスで観察された(図31G)。さらに、リポ-2B182cおよびリポ-1V270+2B182Cは、リポソーム対照と比較して有意に高いVNを誘導した(図31H)。VN力価は肺ウイルス力価と負の相関を示した(P<0.01、Spearman r=-0.59、図27I)。異種インフルエンザウイルスA/Victoria3/75(H3N2)に対する防御は、同種攻撃実験と同じプロトコルを用いて評価した(図31A)。リポソーム対照群と比較して、体重減少、生存率および肺ウイルス力価に有意差はなかった(図32A~C)。まとめると、製剤化複合アジュバントは異種防御には不十分であるが、有害な炎症作用なしに同種H1N1ウイルスに対して有意な防御を示した。
<1V270(TLR7リガンド)および2B182C(TLR4リガンド)のリポソーム共カプセル化は抗体エピトープを広げる>
インフルエンザウイルス感染の普遍的ワクチンは、主要抗原分子である血球凝集素(HA)、ノイラミニダーゼ(NA)の広範なエピトープを認識する抗体の誘導を必要とする。従って、複合アジュバント(1V270および2B182C)によって誘導された抗体のエピトープの広がりおよび交差反応性を調べた。BALB/cマウス(n=5~10)を、1V270(Lipo-1V270)、2B182C(Lipo-2B182C)、共カプセル化リポソーム1V270+2B182C[Lipo-(1V270+2B182C)]のリポソーム製剤と混合した不活化ウイルスで免疫付与し、別々のリポソーム中のLipo-1V270とLipo-2B182Cとを添加混合した。ブランクリポソームを対照として使用し、0日目(プライム)および21日目(ブースト)に免疫付与を行い、28日目に血清を収集した。
Claims (44)
- 哺乳動物における免疫応答を増強するための方法であって、リポソームを含む組成物を、それを必要とする哺乳動物に投与する工程を含み、前記リポソームを含む組成物は、有効な量のTLR4アゴニストおよびTLR7アゴニストを含む、方法。
- 前記TLR4アゴニストおよび前記TLR7アゴニストを同時に投与する、請求項1に記載の方法。
- 前記TLR4アゴニストは式(II)を有する、請求項1または2に記載の方法:
ここで、z1は0~4の整数であり、z2は0~5の整数であり、R5は置換もしくは非置換シクロアルキル、置換もしくは非置換ヘテロシクロアルキル、置換もしくは非置換アリール、または置換もしくは非置換ヘテロアリールであり、R6は置換もしくは非置換アルキル、置換もしくは非置換ヘテロアルキル、置換もしくは非置換シクロアルキル、置換もしくは非置換ヘテロシクロアルキル、置換もしくは非置換アリール、または置換もしくは非置換ヘテロアリールであり、R7は水素、または置換もしくは非置換アルキルであり、およびR8の各々は、独立して、ハロゲン、-CN、-SH、-OH、-COOH、-NH2、-CONH2,ニトロ、-CF3、-CCl3、置換もしくは非置換アルキル、置換もしくは非置換ヘテロアルキル、置換もしくは非置換シクロアルキル、置換もしくは非置換ヘテロシクロアルキル、置換もしくは非置換アリール、または置換もしくは非置換ヘテロアリールである。 - 前記TLR7アゴニストは式(I)を有する、請求項1~3のいずれか1項に記載の方法:

Rcは、水素、C1-10アルキル、もしくは置換C1-10アルキルであり;またはRcおよびR1は、結合する窒素と共に複素環式環もしくは置換複素環式環を形成し;
各R2は、独立して、-OH、(C1~C6)アルキル、置換(C1~C6)アルキル、(C1~C6)アルコキシ、置換(C1~C6)アルコキシ、-C(O)-(C1~C6)アルキル(アルカノイル)、置換-C(O)-(C1~C6)アルキル、-C(O)-(C6~C10)アリール(アロイル)、置換-C(O)-(C6~C10)アリール、-C(O)OH(カルボキシル)、-C(O)O(C1~C6)アルキル(アルコキシカルボニル)、置換-C(O)O(C1~C6)アルキル、-NRaRb、-C(O)NRaRb(カルバモイル)、ハロ、ニトロ、もしくはシアノであるか、またはR2は欠如しており;

- 前記リポソームは、コレステロールを含む、請求項1~4のいずれか1項に記載の方法。
- 1つまたは複数の免疫原を投与する工程をさらに含む、請求項1~6のいずれか1項に記載の方法。
- 前記免疫原は、微生物免疫原である、請求項7に記載の方法。
- 前記微生物は、ウイルスまたは細菌である、請求項8に記載の方法。
- 前記リポソームは、1つまたは複数の免疫原を含む、請求項7~9のいずれか1項に記載の方法。

- 前記TLR7アゴニスト対前記TLR4アゴニストの比率は、約1:10、1:100、1:200、5:20、5:100、または5:200である、請求項1~13のいずれか1項に記載の方法。
- 前記組成物を、注射、筋肉内投与、鼻腔内投与、または静脈内投与する、請求項1~13のいずれか1項に記載の方法。
- 前記リポソームは、DOPCおよびコレステロールを含む、請求項1~15のいずれか1項に記載の方法。
- リポソーム、TLR4アゴニストおよびTLR7アゴニストを含む、医薬製剤。
- 前記リポソームは、1,2-ジオレオイル-sn-グリセロ-3-ホスホコリン(DOPC)、1,2-ジパルミトイル-sn-グリセロ-3-ホスホコリン(DPPC)、1,2-ジステアロイル-sn-グリセロ-3-ホスホコリン(DSPC)、1,2-ジオレオイル-sn-グリセロ-3-[ホスホ-L-セリン](DOPS)、1,2-ジオレオイル-3-トリメチルアンモニウム-プロパン(18:1DOTAP)、1,2-ジオレオイル-sn-グリセロ-3-ホスホ-(1’-rac-グリセロール)(DOPG)、1,2-ジオレオイル-sn-グリセロ-3-ホスホエタノールアミン(DOPE)、1,2-ジパルミトイル-sn-グリセロ-3-ホスホエタノールアミン(DPPE)、1,2-ジオレオイル-sn-グリセロ-3-PE)、1,2-ジパルミトイル-sn-グリセロ-3-ホスホエタノールアミン-N-[メトキシ(ポリエチレングリコール)-2000](16:0PEG-2000PE)、1-オレオイル-2-[12-[(7-ニトロ-2-1,3-ベンゾオキサジアゾール-4-イル)アミノ]ラウロイル]-sn-グリセロ-3-ホスホコリン(18:1~12:0 NBD PC)、1-パルミトイル-2-{12-[(7-ニトロ-2-1,3-ベンゾオキサジアゾール-4-イル)アミノ]ラウロイル}-sn-グリセロ-3-ホスホコリン(16:0~12:0 NBD PC)、およびそれらの混合物;1,2-ジステアロイル-sn-グリセロ-3-ホスホコリン(DSPC)、1,2-ジオレオイル-sn-グリセロ-3-ホスホエタノールアミン(DOPE)、コレステロール、またはそれらの混合物を含む、請求項17に記載の製剤。

- 前記TLR7アゴニスト対前記TLR4アゴニストの比率は、約1:10、1:100、1:200、5:20、5:100、または5:200である、請求項17~21のいずれか1項に記載の製剤。
- 前記TLR7アゴニストは、式(I)の化合物を含む、前記17~22のいずれか1項に記載の製剤:
式(I)中、X1は-O-、-S-、または-NRc-であり;
R1は、水素、(C1~C10)アルキル、置換(C1~C10)アルキル、C6~10アリール、または置換C6~10アリール、C5~9複素環式、置換C5~9複素環式であり;
Rcは、水素、C1-10アルキル、もしくは置換C1-10アルキルであり;またはRcおよびR1は、結合する窒素と共に複素環式環もしくは置換複素環式環を形成し;
各R2は、独立して、-OH、(C1~C6)アルキル、置換(C1~C6)アルキル、(C1~C6)アルコキシ、置換(C1~C6)アルコキシ、-C(O)-(C1~C6)アルキル(アルカノイル)、置換-C(O)-(C1~C6)アルキル、-C(O)-(C6~C10)アリール(アロイル)、置換-C(O)-(C6~C10)アリール、-C(O)OH(カルボキシル)、-C(O)O(C1~C6)アルキル(アルコキシカルボニル)、置換-C(O)O(C1~C6)アルキル、-NRaRb、-C(O)NRaRb(カルバモイル)、ハロ、ニトロ、もしくはシアノであるか、またはR2は欠如しており;
各RaおよびRbは、独立して、水素、(C1~C6)アルキル、置換(C1~C6)アルキル、(C3~C8)シクロアルキル、置換(C3~C8)シクロアルキル、(C1~C6)アルコキシ、置換(C1~C6)アルコキシ、(C1~C6)アルカノイル、置換(C1~C6)アルカノイル、アリール、アリール(C1~C6)アルキル、Het、Het(C1~C6)アルキル、または(C1~C6)アルコキシカルボニルであり;

- 前記R3のリン脂質は、2つのカルボン酸エステルを含み、前記カルボン酸エステルの各々は、不飽和、エポキシ化、ヒドロキシル化、またはそれらの組み合わせの1つ、2つ、3つ、または4つの部位を含む、請求項23~25のいずれか1項に記載の製剤。
- 前記R3のリン脂質は、2つのカルボン酸エステルを含み、前記カルボン酸エステルは、同じであるかまたは異なっている、請求項23~26のいずれか1項に記載の製剤。
- 前記リン脂質のカルボン酸エステルの各々は、C8~C9において不飽和部位を有する、C17カルボン酸エステルである、請求項27に記載の製剤。
- 前記リン脂質のカルボン酸エステルの各々は、C9~C10において不飽和部位を有する、C18カルボン酸エステルである、請求項27に記載の製剤。
- 前記X2は、結合、または鎖中に1~約10個の原子を有する鎖であり、ここで、前記鎖の原子は炭素、窒素、硫黄、および酸素からなる群から選択され、任意の炭素原子はオキソで置換され得、任意の硫黄原子は1個または2個のオキソ基で置換され得る、請求項23~29のいずれか1項に記載の製剤。
- 前記R3は、ジオレオイルホスファチジルエタノールアミン(DOPE)を含む、請求項23~30のいずれか1項に記載の製剤。
- 前記R3は1,2-ジオレオイル-sn-グリセロ-3-ホスホエタノールアミンであり、前記X2はC(O)である、請求項23から31のいずれか1項に記載の製剤。

- 前記TLR4アゴニストは、式(II)を含む、請求項17~36のいずれか1項に記載の製剤:
