・乳、初乳及び酸性又は甘性ホエーからLF及びLPOを単離する迅速かつ簡単な方法、
・高純度な目的タンパク質(例えば、純度が98%超のLF)を得ることができる、LF及びLPO単離のための線形pH勾配を用いた単一のクロマトグラフィー工程単離プロセス、
・開示された方法によって得られた生産物(LFタンパク質)であり、不飽和鉄結合能(UIBC)を測定するために確立された方法によると、高活性(C値が70%超、LF用C値決定キット、株式会社NRLファーマ製)であり、既に結合した鉄の量(A値が3.9%未満、LF用A値決定キット、株式会社NRLファーマ製)が少なく、これらは市場で入手可能なLFのなかでも優れた特徴である。前記生産物は、特殊な化学物質を使用することなく、オンサイトで(モノリスカチオン交換体において)行われる、経済的で手段的に無理のない方法によって製造される。
・少ない量の鉄(A値が3.9%未満)を含むと同時にアンギオゲニンを含まないLFの製造を可能にする方法、
・高い総生物活性(C値+A値が72%超)を有するLFの製造を可能にする方法、
・製造規模まで容易にスケールアップでき、高純度及び高生物活性であるLF及びLPOのコスト効率の高い製造を可能にする方法、
・全体としてLFの85%超を回収できる、クロマトグラフィー、濃縮/脱塩及び乾燥プロセスを含む、LF/LPO単離の全プロセス、
・タンパク質の高分解能を妨げることなく、移動相の高流速(700L/m2/h)を可能にし、高生産性プロセスをもたらすクロマトグラフィープロセス、
・単離を目的としたタンパク質(例えば、ホエー)を有する媒質に化学物質を導入しないか、あるいは他の方法でその特性を変化させないプロセスであって、これにより他の生物化学産業プロセスにおいて媒質をさらに利用可能にするプロセス、
・既に結合した鉄を所定割合で有するLFの製造を可能にする方法、
を提供する。
(i)原料のpHを7未満の値、特にpH6.5未満に調整する工程、
(ii)工程(i)の原料を強カチオン特性を有するモノリスカラム、特に-SO3修飾モノリスカラムと接触させる工程、次いで、
(iii)勾配緩衝剤をカラムに流し、それによってpH値を増加させる工程、及び
(iv)約8~約11未満のpH範囲、好ましくは8.0~10.0のpH範囲、好ましくは8.2~10.0のpH範囲、特に約8.9~約10のpH範囲において溶出する画分Aを回収する工程、及び/又は

[HPLC分析]
サンプル中のLF及びLPOを特定するために、MWD多波長検出器及びpH測定キットを有する伝導度モニターを備えたクロマトグラフィーシステム(PATfix(商標)、BIAセパレーションズ社製、アイドフシュチナ、スロベニア)を使用した。クロマトグラフィー分離は、3.5mS/cmから152mS/cmに増加させた伝導度勾配モードで、2つのリン酸ナトリウム一塩基緩衝液(25mM、pH=7.5)を使用して、CIMac SO3-0.1(孔1.3μm、BIAセパレーションズ社製、アイドフシュチナ、スロベニア)分析カラムにおいて実行した。分析されたタンパク質の溶出は、226nmにおいてMWD検出器によって観察された。クロマトグラフィー法に関する詳細な情報は表2に示され、いくつかの分析したサンプルのクロマトグラムは図1及び2に示されている。すべてのサンプルは、分析の前に、CHROMAFIL(商標) A-45/25(孔径0.45μm)セルロース混合エステルフィルターを用いてろ過された。
乾燥LFサンプルのC値及びA値を決定するために、株式会社NRLファーマ(日本)によって提供された鉄比色分析キットを使用し(詳細な原理はIto他[33]によって公表された)、乾燥サンプル及び液体サンプルのA値を決定するために、HPLC法と組み合わせて、文献[33、34]に記載されたアプローチと同じものを用いた。
LFのC値を決定するために、LFは公知の過剰量の鉄で飽和された。残留鉄はキレート試薬(最大吸収波長760nm)で着色された。残留鉄は760nmで分光光度法で定量化される。鉄錯体の連続希釈液を調製し、検量線を760nmで分光光度法で決定する。結合した鉄は、添加した鉄から残留鉄を差し引くことで算出される。C値は相対的に示され、LFの算出された理論鉄結合容量(LFの各分子は2個の鉄原子に結合できる)は100%として設定される。
LFのA値を決定するために、LFは変性試薬で変性され、放出された鉄はキレート試薬(最大吸収波長760nm)で着色された。放出された鉄は760nmで分光光度法で定量される。鉄錯体の連続希釈液を調製し、検量線を760nmで分光光度法で決定する。既に結合した鉄(A値)は、相対的に示され、1個のLF分子に結合した2個の鉄原子は100%として定義される。
図3は、LF/LPO単離法における原理に関するフローシート及び1回のクロマトグラフィー実験の規模におけるプロセスの物質収支の概算値を示す。LF/LPO単離の前に、1100Lの酸性ホエーは、0.8μm未満の孔径を有するセラミックTFFフィルターシステムを用いてろ過される。次いで、ろ過したホエー(1000L)は、平衡化されたクロマトグラフィーカラムに送り出される。その後、カラムに結合したLF及びLPOは、様々な緩衝液の組み合わせによって溶出される。次いで、溶出した画分は濃縮され、必要に応じて脱塩される。濃縮タンパク質の乾燥後、60~90gの乾燥LF生産物が得られる。僅かに変化したフロースルーホエー及びホエースラリーは、別個で又は混合され、下流においてさらに使用又は処理され得る。
モノリスカラム、80mL CIMmultus(商標) SO3(BIAセパレーションズ社製)は、導入前に、800mLの緩衝液A(リン酸ナトリウム又はクエン酸緩衝剤:5~50mM、pH=4.6)を用いて平衡化された。平衡化後、ろ過された酸性ホエーは、LF及びLPOのカラム容量に達するまでカラムに送り出された。カラム容量の飽和は、分析の項目で説明されたHPLC法により、カラムの流出側におけるフロースルーサンプルを分析することによって確認された。0.24L/minの流速でカラムに送り出されたホエーの体積は、通常、処理されたホエー中のLF/LPO濃度に主に依存して、10~20Lであった。その後、カラムは緩衝液Aで洗い流された。LF及びLPOの分離は、線形pH勾配モードにおいて、2つの緩衝液(クエン酸ナトリウム緩衝剤、リン酸塩緩衝剤、TRIS緩衝剤及び炭酸塩緩衝剤の混合物、4~50mM、pH=4.6及び12.0)を使用してカラムを洗い流すことによって、開始された。pHは、4.6~12.0の範囲内で徐々に線形的に変化させた。LPO/LF溶出のpH範囲は、それぞれ8.9~10及び11~11.7であった。分離操作の結果は、クロマトグラフ的に非常によく分離されたLPO及びLFの2つの溶出画分であり、これらはさらに別々に容易に処理された。引き続く工程において、単離されたタンパク質の画分は、少量の好適な酸性溶液を用いてpH=6に中和され、1~50kDaの孔径を有するTFF膜を用いて濃縮され、そして噴霧乾燥された。LF及びLPOの最終純度は、それぞれ98%超及び70%超であった。LFのC値及びA値は、それぞれ71%及び3.4%であると決定された。
モノリスカラム、80mL CIMmultus(商標) SO3(BIAセパレーションズ社製)は、導入前に、緩衝液B(リン酸ナトリウム又はクエン酸緩衝剤:5~50mM、甘性ホエーのpHと同じpH=5.0~6.5)を用いて平衡化された。その後、甘性ホエーは、LF及びLPOのカラム容量に達するまでカラムに流された。カラム容量の飽和は、分析の項目で説明されたHPLC法により、カラムの流出側におけるフロースルーサンプルを分析することによって確認された。0.24L/minの流速でカラムに送り出されたホエーの体積は、通常、処理されたホエー中のLF/LPO濃度に主に依存して、10~40Lであった。その後、カラムは緩衝液Bで洗い流された。LF及びLPOの分離は、線形pH勾配モードにおいて、2つの緩衝液(クエン酸ナトリウム緩衝剤、リン酸塩緩衝剤、TRIS緩衝剤及び炭酸塩緩衝剤の混合物、4~50mM、pH=5.0及び12.0)を使用してカラムを洗い流すことによって、開始された。pHは、5.0~12.0の範囲内で徐々に線形的に変化させた。LPO/LF溶出のpH範囲は、それぞれ8.9~10及び11~11.7であった。前述の操作の結果は、クロマトグラフ的に非常によく分離されたLPO及びLFの2つの画分であり、これらはさらに別々に容易に処理された。引き続く工程において、単離されたタンパク質の画分は、少量の好適な酸性溶液を用いてpH=6に中和され、1~50kDaの孔径を有するTFF膜を用いて濃縮され、そして噴霧乾燥された。LF/LPOの最終純度は、98%超又は70%超であった。LFのC値及びA値は、それぞれ70.2%及び3.9%であると決定された。
モノリスカラム、8L CIMmultus(商標) SO3-Strong CEX(BIAセパレーションズ社製)は、導入前に、40~80Lの緩衝液C(NaClを添加した、リン酸ナトリウム又はクエン酸緩衝剤:5~50mM、pH=4.6及び15mS/cmの伝導度)を用いて平衡化された。その後、酸性ホエーは、LF及びLPOのカラム容量に達するまでカラムに流された。カラム容量の飽和は、分析の項目で説明されたHPLC法により、カラムの流出側におけるフロースルーサンプルを分析することによって確認された。8L/minの流速でカラムに送り出されたホエーの体積は、通常、処理されたホエー中のLF/LPO濃度に主に依存して、1000~2000Lであった。その後、カラムは緩衝液Cで洗い流された。LF及びLPOの分離は、線形pH勾配モードにおいて、2つの緩衝液(NaClを添加した、クエン酸ナトリウム緩衝剤、リン酸塩緩衝剤、TRIS緩衝剤及び炭酸塩緩衝剤の混合物、4~50mM、pH=4.6及び12.0)を使用してカラムを洗い流すことによって行われた。pHは4.6~12.0の範囲内で徐々に線形的に変化させ、伝導度(15mS/cm)は、線形pH勾配の全体を通して一定であった。LPO/LF溶出のpH範囲は、それぞれ8.2~9.3及び10.7~11.2であった。前述の操作の結果は、クロマトグラフ的に非常によく分離されたLPO及びLFの2つの溶出画分であり、これらはさらに別々に容易に処理された。引き続く工程において、回収されたタンパク質の画分は、先ず、少量の好適な酸性溶液を用いてpH=6に中和され、1~50kDaの孔径を有するTFF膜を用いて脱塩及び濃縮され、そして噴霧乾燥により乾燥された。LF/LPOの最終純度は、98%超又は75%超であった。LFのC値及びA値は、それぞれ74.2%及び2.5%であると決定された。
モノリスカラム、8L CIMmultus(商標) SO3-Strong CEX(BIAセパレーションズ社製)は、導入前に、緩衝液C(リン酸ナトリウム又はクエン酸緩衝剤:5~50mM、甘性ホエーのpHと同じpH=5.0~6.5)を用いて平衡化された。その後、酸性ホエーは、LF及びLPOのカラム容量に達するまでカラムに流された。カラム容量の飽和は、分析の項目で説明されたHPLC法により、カラムの流出側におけるフロースルーサンプルを分析することによって確認された。8L/minの流速でカラムに送り出されたホエーの体積は、通常、処理されたホエー中のLF/LPO濃度に主に依存して、1000~2000Lであった。IEPが7.5未満であるタンパク質は、先ず、pH=7.5の緩衝液Cを用いてpHステップ溶出モードにより溶出され、その後、LF及びLPOの分離は、実施例2の緩衝液と同じ緩衝液を使用し、線形pH勾配モードにおいてカラムを洗い流すことで行われた。pHは7.5~12.0の範囲内で徐々に線形的に変化させた。LPO/LF溶出のpH範囲は、それぞれ8.9~10及び11~11.7であった。別々に回収されたLPO及びLFの溶出画分は、その後、少量の好適な酸性溶液を用いてpH=6に中和され、1~50kDaの孔径を有するTFF膜を用いて濃縮され、そして噴霧乾燥により乾燥された。LF及びLPOの最終純度は、それぞれ98%超又は75%超であった。LFのC値及びA値は、それぞれ72.6%及び2.8%であると決定された。

Claims (17)
- ラクトフェリンタンパク質及び/又はラクトペルオキシダーゼタンパク質を含む画分をこれらのタンパク質の少なくとも一つを含む原料から製造する方法であって、
前記原料は、乳、初乳、酸性又は甘性ホエーからなる群から選択され、
強カチオン交換体特性を有するモノリスカラムを用いたクロマトグラフィー分離プロセスを利用し、

- 約8~約11未満のpH範囲、特に約8.9~約10のpH範囲、又は約5~55mS/cmの範囲内のより高い伝導度において約6.6~約7.5のpH範囲において溶出する画分Aが回収される、請求項1~4のいずれか一項に記載の方法。
- 約10.4超~約12のpH範囲、特に約11超~約11.7のpH範囲、又は約5~55mS/cmの範囲内のより高い伝導度において約9.6~約10.7のpH範囲において溶出する画分Bが回収される、請求項1~5のいずれか一項に記載の方法。
- 前記クロマトグラフィー分離プロセスが、
(i)前記原料のpHをpH7未満の値、特にpH6.5未満に調整する工程、
(ii)前記工程(i)の原料を強カチオン交換体特性を有するモノリスカラムと接触させる工程、次いで、
(iii)pH勾配緩衝剤を前記カラムに流し、それによって前記pH値を増加させる工程、及び
(iv)約8~約11未満のpH範囲、特に約8.0~約10のpH範囲、好ましくは約8.2~約10のpH範囲、より好ましくは約8.9~約10のpH範囲、又は約5~55mS/cmの範囲内のより高い伝導度において約6.6~約7.5のpH範囲において溶出し、典型的にはラクトペルオキシダーゼを含む、画分Aを回収する工程、及び/又は
(v)約10超~約12.0のpH範囲、好ましくは約10.4超~約12のpH範囲、より好ましくは11.0超~12.0のpH範囲、特に約11超~約11.7のpH範囲、又は約5~55mS/cmのより高い伝導度において約9.6~約10.7のpH範囲において溶出し、典型的にはラクトフェリンを含む、画分Bを回収する工程、
(vi)任意で、前記画分A及び/又は前記画分Bを、特に、中和、濃縮及び保存などの処理によって、さらに処理する工程、
を含む、請求項1~6のいずれか一項に記載の方法。 - 前記原料が、前記工程(ii)の前にろ過され、又は前記モノリスカラムが、前記工程(ii)の前に、約7未満、特に約6.5未満のpH値を有する平衡化緩衝剤を用いて平衡化される、請求項7に記載の方法。
- 前記強カチオン交換体特性を有するモノリスカラムが、-SO3H修飾モノリスカラム、-COOH修飾モノリスカラム、-OSO3H修飾モノリスカラム又は-OPO3H修飾モノリスカラムからなる群から選択される、請求項1~8のいずれか一項に記載の方法。
- 前記塩勾配が塩の濃度によって行われ、特に前記塩勾配は、約5mS/cm~約55mS/cmの範囲内における伝導度に対応する、請求項1~9のいずれか一項に記載の方法。
- 前記工程(iii)又は前記工程(iv)の前に、前記カラムが、請求項8に記載の前記平衡化緩衝剤で洗い流される、請求項1~10のいずれか一項に記載の方法。
- 前記ラクトフェリン及び前記ラクトペルオキシダーゼを含む画分は、乾燥され、特に噴霧乾燥によって乾燥される、請求項1~11のいずれか一項に記載の方法。
- 前記ラクトフェリンの純度は90%超であり、前記ラクトペルオキシダーゼの純度は50%超であり、特に前記ラクトフェリンのC値は50%超であり、前記ラクトフェリンのA値は1%超である、請求項1~12のいずれか一項に記載の方法。
- 前記工程(iv)又は前記工程(v)の後、前記カラムは、pHが約12超の緩衝剤で前記カラムを洗い流すことにより洗浄される、請求項1~13のいずれか一項に記載の方法。
- C値が60%超であり、A値が1%超であるラクトフェリンを含む組成物。
- 前記C値が70%以上であり、前記A値が2%以上である、請求項15に記載の組成物。
- 請求項1~14のいずれか一項に記載の方法により得られる、ラクトフェリン又はラクトペルオキシダーゼを含む組成物。
