本開示の主題は、新生物の処置または防止のためのキットを提供する。ある特定の実施形態では、キットは、単位剤形中に、本開示のシアリルルイスA標的化CARを含む免疫応答性細胞の有効量を含む治療用または予防用組成物を含む。特定の実施形態では、細胞は、少なくとも1つの共刺激リガンドをさらに発現する。一部の実施形態では、キットは、治療用または予防用ワクチンを含有する滅菌容器を含み、このような容器は、箱、アンプル、ボトル、バイアル、チューブ、バッグ、パウチ、ブリスターパックまたは当技術分野で公知の他の適した容器形態であり得る。このような容器は、プラスチック、ガラス、ラミネート紙、金属箔または医薬を保持するのに適した他の材料で作製され得る。
序論
抗原提示を改善する戦略は、エピトープ拡大を誘導するか、または現存の抗腫瘍T細胞応答が腫瘍抗原逃避と戦う見込みを保持することを永続させる。例えば、がんワクチンおよび「免疫原性」放射線(RT)は、抗原提示細胞(APC)を活性化させて、内因性T細胞への腫瘍新抗原表示を改善する(Spiotto et al., Sci Immunol (2016); 1)。しかしながら、それでもなお、完全奏効を得るためには、同じ新抗原がすべてではないにせよ大半の腫瘍細胞において発現し、提示されなければならない。遺伝子変異量と相関する既存の腫瘍反応性T細胞を有する患者では、免疫チェックポイント阻害剤はT細胞疲弊を軽減し、持続した応答を実現することができる。しかしながら、チェックポイント阻害は、CARがCAR標的を欠いている腫瘍細胞に対する応答を導くことができないのと同様に、認識された抗原を提示しない腫瘍細胞に対するT細胞応答を回復することができない。
シアリルルイスA(LeA)特異的CAR T細胞はin vitroにおいて膵臓腫瘍細胞に対して活性である
100%の腫瘍細胞に発現し、重要な正常組織には発現しない固形腫瘍標的を同定することは困難である。膵臓がんはこの問題の好例であり、いくつかの魅力的な標的を有するが、そのうちすべての腫瘍細胞に明確に発現するものは1つもない(Zhao et al., Cancer Cell (2015); 28:415-428)。75~90%の膵臓腫瘍に発現し(Viola-Villegas et al., J Nucl Med (2013); 54:1876-1882)、正常ヒト組織では低発現(Viola-Villegas et al., J Nucl Med (2013); 54:1876-1882)の表面抗原であるシアリルルイスA(LeA)は、臨床試験における有効な抗体標的である(NCT03118349、NCT02672917、NCT02687230)。LeAを標的とするヒトモノクローナル5B1抗体は、in vitroおよびin vivoにおける膵臓がんに対する特異性(Viola-Villegas et al., J Nucl Med (2013); 54:1876-1882)、ならびに生物学的活性用量での膵臓がん患者における安全性および忍容性(O'Reilly et al., Journal of Clinical Oncology (2017); 35:4110-4110)を実証している。したがって、このLeA特異的scFvを使用して進行膵管腺癌(APDAC)標的CARを構築した。LeA特異的LBBz CARは、LeAを発現する複数の膵臓がん腫瘍系に対する効果的な細胞傷害性を示したが、LeA陰性PC3前立腺がん細胞に対しては示さなかった(図6A~6C)。Capan2 PDACは中間レベルのLeAを発現し(図6A~6C)、さらなる実験のために選択された。
放射線療法(RT)はLeA発現を誘導し、異種標的抗原発現を有する腫瘍を排除するCAR T細胞の能力を改善し得るという最初の仮説を試験するために、腫瘍細胞に2Gy RTを照射し、2日後、残っている生存細胞に対する細胞傷害性Tリンパ球(CTL)アッセイおよび表面標的抗原発現のFACS解析を実施した。より高いRT線量は、少ないが有意な腫瘍細胞死を誘導したが、2Gyは腫瘍生存率において検出可能な差を生じなかったために、2Gyを選択した(図1A)。2Gy(以降「低線量RT」と称す)は、あらゆるエフェクター:標的比においてCAR T細胞殺滅に対する腫瘍細胞の感受性を増加させるが(図1B)、驚くべきことに標的抗原発現を増加させない(図1C)ことが見出された。
低線量RTが腫瘍細胞をCAR T細胞による殺滅に対して感受性にする潜在的な機構に対する洞察を得るために、RNAseq解析を低線量RTの前後に腫瘍細胞に対して実施した。RT自体は致死量以下であったが、遺伝子セット解析は多くのアポトーシス経路が低線量RTによって有意に影響を受けることを明らかにした(図1D)。特に、TRAIL媒介死に対して感受性である腫瘍細胞とそうでない腫瘍細胞とを識別する遺伝子セット(Hamai et al., Oncogene (2006); 25:7618-7634)1は最も低い偽発見率を有することが明らかとなった(それぞれに関してFDR<0.0000001;492の陽性経路メンバーのうち429が誘導され、128の陰性経路メンバーのうち114が下方調節された)(図1D)。
TRAILとは、2つの異なる受容体、および感受性に影響を与えるいくつかの下流シグナル伝達分子を介して死を誘導する三量体タンパク質であり、腫瘍細胞は一般に正常細胞よりもTRAIL誘導アポトーシスに対して感受性であるが、様々な程度がある(Walczak et al., Nat Med (1999); 5:157-163)。遺伝子セット解析は低線量RTが腫瘍細胞をTRAIL媒介死に対して転写プライミングし得ることを示唆したが、これは死リガンドが局所的に十分なレベルで存在した場合にのみ関連するものであった。LeA特異的CAR T細胞からのTRAIL産生を解析し、CAR T細胞はベースラインでは低レベルのTRAILを産生するが、標的抗原遭遇の際ではTRAIL mRNAおよびタンパク質を有意に誘導することが見出された(図1E)。対照的に、シグナル伝達ドメインを欠く切断型CAR(Ldel)を発現するT細胞による腫瘍認識後にTRAILは誘導されず、CARシグナル伝達へのTRAIL誘導の依存性が確立された(図1F)。
低線量RTに曝露された抗原陰性腫瘍に関する、活性化CAR T細胞によって産生されたTRAILの機能的有意性を試験するために、腫瘍細胞を抗原陽性(Ag+)および抗原陰性(Ag-)集団にFACS選別した。Ag-細胞にホタルルシフェラーゼ(Luc)を形質導入し、Ag-細胞は経時的に安定に抗原陰性のままであった(図7)。75%のAg+腫瘍細胞と25%のAg-Luc+腫瘍細胞とを混合し、低線量RTに曝露させるかまたはRTに曝露させず、TRAILをCRISPRによって破壊したCAR T細胞とともにインキュベートした(図2A~2Bおよび図7)。TRAILwtまたはノックアウトCAR T細胞を、TRAIL産生を誘導する3日前に静止するかまたは標的抗原によって刺激した。Luc活性を使用してAg-細胞殺滅をモニタリングし、RT曝露腫瘍細胞に関して予備刺激したwt CAR T細胞が最大規模のAg-腫瘍細胞死を生じ、このAg-腫瘍細胞死がCAR T細胞におけるTRAILの非存在または腫瘍に対する増感RTの非存在によって有意に減少することが見出された(図2B)。標的細胞を認識するがTRAILを誘導しないL(del)CAR T細胞は、TRAILを構成的に発現させた場合、有意により多くのRT増感Ag-腫瘍細胞を死滅させた(図2C)。
次に、標的抗原を部分的に欠いている異種固形腫瘍の困難だが一般的な臨床シナリオのためにマウスモデルを確立した。25%Ag-細胞からなるPDACをマウス膵臓において定着させ、9日後にCAR T細胞を用いて処置した(図4A)。Ag+同所PDACを一貫して排除したCAR T細胞はいかなる異種腫瘍も完全には排除することができなかった(図4B~4E)。次に、増感RTがin vivoにおいてCAR T細胞を用いて処置した異種腫瘍に対する何らかの有意義な利益をもたらしたかどうかを試験した。増感RTとその後のCAR T細胞とを用いて処置した、定着した異種PDACを有するマウスは、イメージング、剖検検査および病理学によってより多くのCRおよびPRを達成した(図4Bおよび4F)。CAR非依存性T細胞殺滅の主要な公知の機構はT細胞受容体(TCR)を介し、RTは標的細胞にHLA発現を誘導することができるため、TCR依存性腫瘍殺滅が増感RT後に有意な役割を果たすかどうかを試験した。TCRを欠く(TCR-/-)CAR T細胞(図10)は、RT増感異種腫瘍を排除する能力を維持した(図4G)。RTは最初に、初めの2週間にわたって腫瘍内に中程度に増加したT細胞蓄積をもたらした(図4I~4K)。有意な腫瘍流入(図11)にもかかわらず、TRAIL-/-CAR T細胞はRT増感腫瘍保有マウスにおいて完全奏効を一貫して達成することができず、これは死亡(GVHDまたは腫瘍進行のいずれかから生じた)時の応答のウォーターフォールプロット(図4B)と毎週の生物発光イメージング(図4H)の両方によって実証された。再発/進行した腫瘍を有するマウスは、FACSによって評価されるように血液、脾臓および腫瘍にCAR T細胞を依然として所有し、IHCによって腫瘍に侵入する有意なT細胞を呈したが、Ag-腫瘍細胞という副産物を実証した(図4Lおよび図12A~12B)。
全身RTがCAR T細胞による増感に必要とされるかどうか、または腫瘍に対する局所RTが十分であるかどうかを決定するために、同所PDACを所有するマウスを、全身または膵臓腫瘍のみに対するRTと、それに続くCAR T細胞投与とを用いて処置した(図5A)。総身RT処置マウスは早期時点においてより多いT細胞腫瘍浸潤を有する傾向があったが(図13)、両方の戦略は類似した腫瘍応答をもたらした(図5B~5D)。したがって、全身低線量RTと局所低線量RTの間の潜在的に異なる宿主効果にもかかわらず、いずれのアプローチも異種腫瘍をCAR T細胞による殺滅に対して効果的に感受性にする。
RTをCAR T細胞と組み合わせる経験は限定されている。TRAIL媒介殺滅のためにRTによって転写プライミングした腫瘍細胞が細胞培養およびマウス研究においてCAR T細胞に応答した有意により多くの死を呈したのと同様に、類似した増感がRT後の近傍の抗原陰性正常組織細胞において生じ得ることが考えられる。サンプリングした腫瘍塊において大きな割合のCD19-腫瘍細胞を保有する難治性びまん性大細胞型B細胞リンパ腫(DLBCL)を有する患者(図5E~5F)はCD19 CAR療法のために来院した(NCT02631044)。この患者は下腿、特に右側の下腿の皮膚に浸透する痛みを伴う疾患を有していた。緩和的RTを患者の右下肢に与え(4Gy×5回)、次いで患者は計画通りにCD19 CAR T細胞を受けた。CAR T細胞療法の数日および数週間後、患者は照射範囲内に傷害性の徴候も症状も呈しなかった。患者は神経学的症状を伴わずにグレード2のCRSを呈した。CAR T細胞の1か月後、患者はPET-CTイメージングに基づいて優れた応答を有した(図5G)。CAR T細胞注入の2か月後、腫瘍は、緩和的RTとその後のCAR T細胞とを受けた患部を除いて、以前の場所および新たな場所においてCD19低/陰性発現を伴ってリバウンドした。処置の1年後の現在、緩和的RTとそれに続くCAR T細胞とに供した抗原異種腫瘍の領域はCRのままである(図5G)。
B細胞悪性腫瘍においてCD19を標的とする最初の選択は主として、白血病およびリンパ腫におけるCD19の上昇した比較的同種の発現、ならびに正常組織におけるその発現のB細胞系列への制限によって推進された(Brentjens et al., Nat Med (2003); 9:279-286、Maher et al., Nat Biotechnol (2002); 20:70-75)。第I相ALL試験患者中70~90%という注目すべき完全寛解率に基づいて(Sadelain, J Clin Invest (2015); 125:3392-3400)、CAR療法を幅広い範囲のがんに拡大する見通しが立ちつつある。CAR療法は近年やっと固形腫瘍に取り組み始めたが(Zhao et al., Cancer Cell (2015); 28:415-428、Morello et al., Cancer Discov (2016); 6:133-146、Jindal et al., Med Oncol (2018); 35:87)、結果はこれまでのところ控えめであり、大奏効の発生はほとんどない(Louis et al., Blood (2011); 118:6050-6056、Brown et al., N Engl J Med (2016); 375:2561-2569)。抗原陰性腫瘍細胞の逃避および再成長は今では十分に立証されたCAR療法に耐性の機構であるため(Brown et al., N Engl J Med (2016); 375:2561-2569、Gardner et al., Blood (2016); 127:2406-2410、Jackson and Brentjiens, Cancer Discov (2015); 5:1238-1240)、CAR T細胞が抗原逃避を効果的に防止することを可能にする新規なアプローチが必要である。
細胞培養
ホタルルシフェラーゼ-GFPを発現する腫瘍細胞は以前に記載された(Zhao et al., Cancer Cell (2015); 28:415-428)。293T細胞系、H29およびレトロウイルスパッケージング細胞系を、10%FCSを補充したDMEM中で培養した(Zhao et al., Cancer Cell (2015); 28:415-428)。Capan-2細胞はJason S.Lewis(MSKCC)によって厚意により提供され、これを、10%FCSを補充したRPMI中で成長させた。細胞を、MycoAlertマイコプラズマ検出キット(Lonza)を使用してマイコプラズマに関して試験した後に動物に注射した。
放射線量:PDACを使用したすべての実験は、別段の指定がない限り2Gyを使用した。in vitroにおけるRT研究に関して、すべてのRT増感実験は、別段の定めのない限り、RTを腫瘍分析またはT細胞との共培養の2日前に腫瘍細胞に投与して実施した。
CD3(UCHT1)、CD4(S3.5)、CD8(3B5)、DR5(DJR2-4、PEコンジュゲート、BioLegend)、CD95(DX2、PE-Cy7コンジュゲート、BD Biosciences)、LeA(7LE、AF405コンジュゲート(conjuugated)、Novus)、CD19(SJ25C1)、41BBL(5F4)およびグランザイムB(FGB12、Invitrogen)に対する蛍光色素コンジュゲート抗体を使用した。Alexa647コンジュゲートヤギ抗ヒトF(ab)2(ThermoFisher)を使用してCARを検出した。フローサイトメトリーをBD LSRIIにおいて実施し、データをFlowJoソフトウェアバージョン9.5.2(TreeStar)を用いて解析した。ヒトFc受容体結合阻害剤抗体(eBioscience)を使用してFc受容体を遮断した。場合によっては、CountBrightビーズ(Invitrogen)を試料に添加して細胞数を計数した。
RNAおよびELISA実験のために、CAR T細胞を、標的抗原を発現するCapan2に4時間曝露させ、続いてT細胞の除去および単培養を行い、新しい培地に毎日再播種した。細胞を除去し、所与の時点でTRAIL mRNA発現に関して分析し、培地を毎日の終わりにTRAIL ELISA(MyBiosource MBS335491)のために収集した。qPCRを、プライマーHs00921974(TRAIL)、Hs00366278(DR5)およびHs04194366(RPL13Aハウスキーピング)を使用したTaqManシステム(ThermoFisher)を使用して実施した。
総RNAを、RNeasyキット(QIAGEN)を製造業者の指示に従って使用することによって細胞から抽出した。RNA濃度および品質を、NanoDrop分光光度計(Themo Fisher Scientific)を使用したUV分光法によって評価した。100~200ngの総RNAを使用し、1:1の体積比のランダムヘキサマーとオリゴdTとを用いたSuperScript III First-Strand Synthesis SuperMix(Invitrogen)を使用してcDNAを調製した。完了したcDNA合成反応物を、2UのRNアーゼHを用いて37℃で20分間処理した。定量PCRを、ABsolute Blue qPCR SYBR Green Low ROX Mixを使用して実施した。PCRアッセイを、QuantStudio(商標)7 Flexシステムにおいて実行し、Ct値を、QuantStudioリアルタイムPCRソフトウェアを用いて取得した。遺伝子発現の相対変化を、2ΔΔCt法を用いて解析した。
SJ25C1 CD19特異的scFvを含む1928ζおよび19BBζ CARは以前に記載されている(Maher et al., Nat Biotechnol (2002); 20:70-75)。LBBzおよびL28zは、CD19特異的scFvを、LeAを標的とするヒト5B1 scFvに置き換えることによって構築した。すべての構築物は、以前に記載されたように、T細胞イメージングのためにGaussiaルシフェラーゼを発現するように設計した(Santos et al., Nat Med (2009); 15:338-344)。L(del)変異体は、指定した構築物から細胞内共刺激因子およびシグナル伝達ドメインを除去する一方で細胞外および膜貫通部分を保持することによって作出した。TRAILを発現する構築物は、指定したCARおよびP2A配列に続けてTRAIL cDNA配列を付加することによって作出した。
SFGγ-レトロウイルス(RV)ベクターをコードするプラスミド(Riviere et al., Proc Natl Acad Sci USA (1995); 92:6733-6737)を、以前に記載されたように調製した(Maher et al., Nat Biotechnol (2002); 20:70-75)。形質導入されたgpg29線維芽細胞(H29)に由来するVSV-G偽型レトロウイルス上清を使用して、安定なレトロウイルス産生細胞系を以前に記載されたように構築した(Gallardo et al., Blood (1997); 90:952-957)。T細胞は、レトロネクチン(Takara)コーティングプレートにおける遠心分離によって形質導入した。T細胞ノックアウト研究では、CAR形質導入は、記載されたように、Cas9/gRNA電気穿孔の直後に実施した(Eyquem et al., Nature (2017); 543, 113-117)。
100%Ag+腫瘍細胞を使用したCTL:CARを形質導入したT細胞の細胞傷害性を標準的なルシフェラーゼベースアッセイによって決定した。ルシフェラーゼベースアッセイに関して、ホタルルシフェラーゼ-GFPを発現する腫瘍細胞は標的細胞として役立った。エフェクター細胞および腫瘍細胞を、示されたE/T比(ration)で黒壁96または384ウェルプレートにおいて3連で共培養した。標的細胞単独を同じ細胞密度で播種して、ベースラインルシフェラーゼ発現を決定した(T細胞無し対照)。18時間後、ルシフェラーゼ基質(Bright-Glo、Promega)を各ウェルに直接添加した。放出された光を発光プレートリーダー、またはイメージングデータセットの獲得のためのLiving Imageソフトウェア(Xenogen)を備えるXenogen IVISイメージングシステム(Xenogen)によって測定した。溶解を[1-(RLUsample)/(RLUmax)]×100として決定した。アッセイを、前の週以内に形質導入されたCAR T細胞を使用して実施した。
CTV(CellTrace Violet、Fisher C34571)で標識したAg-細胞を、8ウェル顕微鏡検査スライドにおけるCAR T細胞、およびアネキシンV595(Fisher A13203)に加えて、非標識Ag+細胞と75%LeA+の比で混合した。共焦点画像を、LSM880共焦点顕微鏡(Carl Zeiss)を用いて最適なイメージングパラメーターにおいて培養中で18時間にわたって7分ごとに獲得した。データを、Imaris(Bitplane)を使用して3Dレンダリングし、可視化した。LeA-細胞の殺滅パーセントを、総Ag-細胞(青色細胞)ならびに死/瀕死Ag-細胞(赤色および青色二重陽性)を自動的に定量するImageJ/FIJI(NIH)において作成したカスタムマクロを使用してすべての時点で決定した。
T細胞活性化を開始した48時間後、AgilePulse MAXシステム(Harvard Apparatus)を使用したCas9 mRNAおよびgRNAの電気泳動転写によって細胞にトランスフェクトした。3×106個の細胞を5μgのCas9および5μgのgRNAと混合して0.2cmキュベットに入れた。電気穿孔後、細胞を培養培地に希釈し、37℃、5%CO2でインキュベートした。TCR陰性T細胞を取得するために、TCR陽性T細胞を、gRNAトランスフェクションの3~5日後に磁気ビオチン抗TCRαβおよび抗ビオチンマイクロビーズならびにLSカラム(Miltenyi Biotech)を使用して除去した。TRAIL陰性細胞を取得するために、TRAIL陽性T細胞を、磁気PE-抗TRAIL(R&D、FAB687P)および抗PEマイクロビーズをLSカラム(Miltenyi Biotech)中で使用して除去した。DR5陰性細胞を取得するために、FACS選別を、PE-抗DR5染色を使用して実施した。
8~12週齢NOD/SCID/IL-2Rγ-ヌル(NSG)雄マウス(Jackson Laboratory)を、MSKCC動物実験委員会によって承認されたプロトコールに従って使用した。規定の比のLeA+およびLeA- FACS選別Capan2 PDAC腫瘍細胞を、IRB承認マウスプロトコールに従って、マウスを外科的に切開し膵臓を露出させた後、NSGマウスの膵臓に注射した。マウス1匹当たり50%マトリゲル中75,000個の腫瘍細胞を注射した。マウスを処置に対して無作為化し、処置群を、処置および腫瘍評価を実施する人員に対して盲検化した。腫瘍を膵臓において9日間定着させ、次いでマウスを、RTとそれに続くT細胞とを用いて処置した。腫瘍体積を、後眼窩D-ルシフェリン注射とそれに続くIVISイメージングとを使用して、生物発光イメージング(BLI)によって測定した。各マウスの腫瘍負荷を、処置の開始時のそのマウスのベースライン腫瘍BLIと比較して経時的に表した。
Gaussiaルシフェラーゼを含有するCAR T細胞を、後眼窩注射したセレンテラジン(3031-10 Coelenterazine-SOL in vivo、Nanolight)を使用してイメージングした。
細胞をTrizol LS(Invitrogen)に溶解し、次いでRNA抽出のためにMSKCCのIntegrated Genomics Operationに提出した。バイオアナライザーにおけるribogreen定量および品質管理後、500ngの総RNAは、Truseq Stranded Total RNAライブラリー調製化学(Illumina)を使用して6サイクルのPCRによりライブラリー調製を受けた。試料をバーコード化し、TruSeq SBSキットv3(Illumina)を使用して50bp/50bpペアエンドランにおいてHiseq2500 1Tにかけた。試料1つ当たり平均で5100万のペアリードが生成され、mRNA塩基のパーセントは平均して58%であった。
すべての実験データは平均±s.e.mとして表される。統計的方法は、試料サイズを予め決定するためには使用しなかった。群は対応のない両側t検定を使用して比較した。統計分析はGraphPad Prism7ソフトウェアにおいて実施した。
前述の記載から、本開示の主題を様々な使用法および条件に採用するために、本開示の主題に対して変形および修正を行うことができることは明らかである。そのような実施形態もまた以下の特許請求の範囲の範囲内である。

- 前記細胞外抗原結合性ドメインが、配列番号2に示されるアミノ酸配列を含む重鎖可変領域CDR2またはその保存的改変および配列番号5に示されるアミノ酸配列を含む軽鎖可変領域CDR2またはその保存的改変を含む、請求項3に記載のCAR。
- 前記細胞外抗原結合性ドメインが、配列番号1に示されるアミノ酸配列を含む重鎖可変領域CDR1またはその保存的改変および配列番号4に示されるアミノ酸配列を含む軽鎖可変領域CDR1またはその保存的改変を含む、請求項3または4に記載のCAR。
- 細胞外抗原結合性ドメイン、膜貫通ドメインおよび細胞内ドメインを含むキメラ抗原受容体(CAR)であって、前記細胞外抗原結合性ドメインは、シアリルルイスAに特異的に結合し、配列番号1に示されるアミノ酸配列を含む重鎖可変領域CDR1、配列番号2に示されるアミノ酸配列を含む重鎖可変領域CDR2および配列番号3に示されるアミノ酸配列を含む重鎖可変領域CDR3を含む、キメラ抗原受容体(CAR)。
- 細胞外抗原結合性ドメイン、膜貫通ドメインおよび細胞内ドメインを含むキメラ抗原受容体(CAR)であって、前記細胞外抗原結合性ドメインは、シアリルルイスAに特異的に結合し、配列番号4に示されるアミノ酸配列を含む軽鎖可変領域CDR1、配列番号5に示されるアミノ酸配列を含む軽鎖可変領域CDR2および配列番号6に示されるアミノ酸配列を含む軽鎖可変領域CDR3を含む、キメラ抗原受容体(CAR)。
- 前記細胞外抗原結合性ドメインが、配列番号1に示されるアミノ酸配列を含む重鎖可変領域CDR1、配列番号2に示されるアミノ酸配列を含む重鎖可変領域CDR2、配列番号3に示されるアミノ酸配列を含む重鎖可変領域CDR3、配列番号4に示されるアミノ酸配列を含む軽鎖可変領域CDR1、配列番号5に示されるアミノ酸配列を含む軽鎖可変領域CDR2および配列番号6に示されるアミノ酸配列を含む軽鎖可変領域CDR3を含む、請求項3から7のいずれか一項に記載のCAR。
- 細胞外抗原結合性ドメイン、膜貫通ドメインおよび細胞内ドメインを含むキメラ抗原受容体(CAR)であって、前記細胞外抗原結合性ドメインは、シアリルルイスAに特異的に結合し、配列番号7に示されるアミノ酸配列と少なくとも約80%相同(例えば、少なくとも約80%同一)であるアミノ酸配列を含む重鎖可変領域を含む、キメラ抗原受容体(CAR)。
- 前記細胞外抗原結合性ドメインが、配列番号7に示されるアミノ酸配列を含む重鎖可変領域を含む、請求項9に記載のCAR。
- 細胞外抗原結合性ドメイン、膜貫通ドメインおよび細胞内ドメインを含むキメラ抗原受容体(CAR)であって、前記細胞外抗原結合性ドメインは、シアリルルイスAに特異的に結合し、配列番号8に示されるアミノ酸配列と少なくとも約80%相同(例えば、少なくとも約80%同一)であるアミノ酸配列を含む軽鎖可変領域を含む、キメラ抗原受容体(CAR)。
- 前記細胞外抗原結合性ドメインが、配列番号8に示されるアミノ酸配列を含む軽鎖可変領域を含む、請求項13に記載のCAR。
- 細胞外抗原結合性ドメイン、膜貫通ドメインおよび細胞内ドメインを含むキメラ抗原受容体(CAR)であって、前記細胞外抗原結合性ドメインは、シアリルルイスAに特異的に結合し、
a)配列番号7に示されるアミノ酸配列と少なくとも約80%相同(例えば、少なくとも約80%同一)であるアミノ酸配列を含む重鎖可変領域、および
b)配列番号8に示されるアミノ酸配列と少なくとも約80%相同(例えば、少なくとも約80%同一)であるアミノ酸配列を含む軽鎖可変領域
を含む、キメラ抗原受容体(CAR)。 - 前記細胞外抗原結合性ドメインが、配列番号7に示されるアミノ酸配列を含む重鎖可変領域および配列番号8に示されるアミノ酸配列を含む軽鎖可変領域を含む、請求項13に記載のCAR。
- 前記細胞外抗原結合性ドメインが、配列番号7に示されるアミノ酸配列と少なくとも約80%相同(例えば、少なくとも約80%同一)であるアミノ酸配列を含む重鎖可変領域および配列番号8に示されるアミノ酸配列と少なくとも約80%相同(例えば、少なくとも約80%同一)であるアミノ酸配列を含む軽鎖可変領域を含む、請求項13または14に記載のCAR。
- 前記細胞外抗原結合性ドメインが、配列番号7に示されるアミノ酸配列を含む重鎖可変領域および配列番号8に示されるアミノ酸配列を含む軽鎖可変領域を含む、請求項13から15のいずれか一項に記載のCAR。
- 前記細胞外抗原結合性ドメインが、一本鎖可変断片(scFv)を含む、請求項1から16のいずれか一項に記載のCAR。

- 前記細胞外抗原結合性ドメインが、前記細胞外抗原結合性ドメインの重鎖可変領域と軽鎖可変領域の間にリンカーを含む、請求項1から21のいずれか一項に記載のCAR。
- 前記細胞外抗原結合性ドメインが、前記細胞外抗原結合性ドメインの5’末端に共有結合によって繋がれるシグナルペプチドを含む、請求項1から22のいずれか一項に記載のCAR。
- 前記膜貫通ドメインが、CD8ポリペプチド、CD28ポリペプチド、CD3ζポリペプチド、CD4ポリペプチド、4-1BBポリペプチド、OX40ポリペプチド、ICOSポリペプチド、CTLA-4ポリペプチド、PD-1ポリペプチド、LAG-3ポリペプチド、2B4ポリペプチド、BTLAポリペプチド、合成ペプチド(免疫応答と関連するタンパク質をベースとしない)またはそれらの組合せを含む、請求項1から23のいずれか一項に記載のCAR。
- 前記細胞内ドメインが、少なくとも1つの共刺激シグナル伝達領域をさらに含む、請求項1から24のいずれか一項に記載のCAR。
- 前記少なくとも1つの共刺激シグナル伝達領域が、CD28ポリペプチド、4-1BBポリペプチド、OX40ポリペプチド、ICOSポリペプチド、DAP-10ポリペプチドまたはそれらの組合せを含む、請求項25に記載のCAR。
- 前記少なくとも1つの共刺激シグナル伝達領域が、CD28ポリペプチドを含む、請求項26に記載のCAR。
- 前記細胞内シグナル伝達ドメインが、野生型CD3ζポリペプチドまたは改変されたCD3ζポリペプチドを含み、前記改変されたCD3ζポリペプチドが、a)免疫受容体チロシンベース活性化モチーフ(ITAM)のすべてもしくは一部を欠き、前記ITAMが、ITAM1、ITAM2およびITAM3であり、ならびに/または塩基性豊富ストレッチ(BRS)領域のすべてもしくは一部を欠き、前記BRS領域が、BRS1、BRS2およびBRS3である、請求項1から27のいずれか一項に記載のCAR。
- 前記改変されたCD3ζポリペプチドが、
a)ITAM2もしくはその部分を欠き、必要に応じて、i)ITAM3もしくはその部分および/またはii)ITAM1もしくはその部分をさらに欠き、
b)ITAM1もしくはその部分を欠き、必要に応じて、ITAM3もしくはその部分をさらに欠き、
c)ITAM3もしくはその部分を欠き、
d)ITAM2もしくはその部分の欠失を含み、必要に応じて、i)ITAM3もしくはその部分の欠失および/またはii)ITAM1もしくはその部分の欠失をさらに含み、
e)ITAM1もしくはその部分の欠失を含み、必要に応じて、ITAM3もしくはその部分の欠失をさらに含み、ならびに/または
f)ITAM3もしくはその部分の欠失を含む、請求項28に記載のCAR。 - 前記改変されたCD3ζポリペプチドが、
a)BRS2もしくはその部分を欠き、必要に応じて、i)BRS3もしくはその部分および/またはii)BRS1もしくはその部分をさらに欠き、
b)BRS1もしくはその部分を欠き、必要に応じて、BRS3もしくはその部分をさらに欠き、
c)BRS3もしくはその部分を欠き、ならびに/または
d)BRS1もしくはその部分、BRS2もしくはその部分およびBRS3もしくはその部分を欠き、
e)BRS2もしくはその部分の欠失を含み、必要に応じて、i)BRS3もしくはその部分の欠失および/またはii)BRS1もしくはその部分の欠失をさらに含み、
f)BRS1もしくはその部分の欠失を含み、必要に応じて、BRS3もしくはその部分の欠失をさらに含み、
g)BRS3もしくはその部分の欠失を含み、ならびに/または
h)BRS1もしくはその部分、BRS2もしくはその部分およびBRS3もしくはその部分の欠失を含む、請求項28または29に記載のCAR。 - 前記改変されたCD3ζポリペプチドが、ITAM2、ITAM3、BRS2およびBRS3を欠くか、または、ITAM2、ITAM3、BRS2およびBRS3の欠失を含む、請求項28から30のいずれか一項に記載のCAR。
- ヒンジ/スペーサー領域をさらに含み、前記ヒンジ/スペーサー領域が、必要に応じて、CD8ポリペプチド、CD28ポリペプチド、CD3ζポリペプチド、CD4ポリペプチド、4-1BBポリペプチド、OX40ポリペプチド、CD166ポリペプチド、CD166ポリペプチド、CD8aポリペプチド、CD8bポリペプチド、ICOSポリペプチド、ICAM-1ポリペプチド、CTLA-4ポリペプチド、CD27ポリペプチド、CD40/My88ペプチド、NKGD2ペプチドまたはそれらの組合せからなる群から選択される分子の天然または改変されたヒンジ/スペーサー領域であり、前記膜貫通ドメインが、CD8ポリペプチド、CD28ポリペプチド、CD3ζポリペプチド、CD4ポリペプチド、4-1BBポリペプチド、OX40ポリペプチド、CD166ポリペプチド、CD166ポリペプチド、CD8aポリペプチド、CD8bポリペプチド、ICOSポリペプチド、ICAM-1ポリペプチド、CTLA-4ポリペプチド、CD27ポリペプチド、CD40/My88ペプチド、NKGD2ペプチドまたはそれらの組合せからなる群から選択される分子の天然または改変された膜貫通ドメインである、請求項1から31のいずれか一項に記載のCAR。
- 前記ヒンジ/スペーサー領域が、前記膜貫通ドメインが由来するものと同一の分子に由来する、請求項32に記載のCAR。
- a)CD28ポリペプチドのヒンジ/スペーサー領域およびCD28ポリペプチドの膜貫通ドメイン、
b)CD84ポリペプチドのヒンジ/スペーサー領域およびCD84ポリペプチドの膜貫通ドメイン、
c)CD166ポリペプチドのヒンジ/スペーサー領域およびCD166ポリペプチドの膜貫通ドメイン、
d)CD8aポリペプチドのヒンジ/スペーサー領域およびCD8aポリペプチドの膜貫通ドメイン、または
e)CD8bポリペプチドのヒンジ/スペーサー領域およびCD8bポリペプチドの膜貫通ドメイン
を含む、請求項33に記載のCAR。 - CD166ポリペプチドのヒンジ/スペーサー領域およびCD166ポリペプチドの膜貫通ドメインを含む、請求項34に記載のCAR。
- 前記膜貫通ドメインおよび前記ヒンジ/スペーサー領域が、異なる分子に由来する、請求項35に記載のCAR。
- CD28ポリペプチドのヒンジ/スペーサー領域およびICOSポリペプチドの膜貫通ドメインを含む、請求項36に記載のCAR。
- 組換え発現されるか、またはベクターから発現される、請求項1から37のいずれか一項に記載のCAR。
- 前記ベクターが、レトロウイルスベクター(例えば、γ-レトロウイルスベクター)である、請求項38に記載のCAR。
- 前記請求項のいずれか一項に記載のCARを含む免疫応答性細胞。
- 前記CARを含む組成物(例えば、ベクター)を使用して改変される、請求項40に記載の免疫応答性細胞。
- 前記CARが、前記免疫応答性細胞の表面で構成的に発現される、請求項40または41に記載の免疫応答性細胞。

- シアリルルイスAに結合する免疫応答性細胞を生成するための方法であって、前記免疫応答性細胞中に、請求項1から39のいずれか一項に記載のCARをコードする核酸配列を含む核酸分子を導入することを含む、方法。
- 請求項40から45のいずれか一項に記載の免疫応答性細胞を含む組成物。
- 医薬組成物であり、薬学的に許容される担体をさらに含む、請求項52に記載の組成物。
