アデノ随伴ウイルス(AAV)は、パルボウイルス科(Parvoviridae)に属する、ディペンドパルボウイルス属(Dependoparvovirus)の一部である。AAVは、およそ4.7キロベース(kb)から6kbの長さまでの単鎖DNA(ssDNA)ゲノムを有する小さな、無エンベロープの、正二十面体ウイルスである。いくつかの血清型が発見され、これまでに最も詳細に調べられた血清型として、AAV血清型2(AAV2)がある。

組換えAAVベクターに必要なすべての遺伝子エレメントが宿主細胞ゲノムへと安定に組み込まれるプロデューサー細胞株を生成することの主な困難は、細胞傷害性(Yangら、1994年、J. Virol. 68:4847~4856頁)及び細胞増殖抑制性(Schmidtら、2000年、J. Virol. 74:9441~9450頁)であることが周知のRepタンパク質の構成的な発現である。これは、Repタンパク質を安定に発現する細胞が、ラージスケールのバイオリアクターで組換えAAVベクターを産生するのに必要な密度に達するまで生存しないことを意味する。rep遺伝子プロモーター、p5及びp19のE1A媒介活性化のため、HEK293ベースのプロデューサー細胞株を生成する場合、この困難は複雑になる。Rep発現の調節の複雑さのさらなる層は、p19プロモーターの位置であり、それは、p5プロモーターによって発現されるRepタンパク質(Rep78及びRep68)のコード領域内に置かれる。結果として、p19プロモーターの操作は、Rep78及びRep68のコード配列において必然的に変異を引き起こし、これらの必須のRepタンパク質の構造及び機能の崩壊をもたらしやすい。さらに、repのネイティブプロモーターの崩壊は、効率的なAAVベクター産生に必要な様々なRep及びCapタンパク質の正確な化学量論に影響する。

AAVベクター産生に使用される標準的なパッケージングプラスミド(rep及びcap遺伝子)、ヘルパーウイルスプラスミド(E2A、E4及びVA遺伝子)及びトランスファーベクタープラスミド(組換えAAVベクターのDNAゲノムをコードする、ITR配列に隣接する導入遺伝子)に以下の修飾を行った。
インシリコで3つのSV40ポリA配列を再現するため、初期ポリA配列を完全SV40ゲノム(受託番号J02400.1)からコピーし、3つのコピーをタンデムにライゲーションした。
終結イントロンは、rep2cap2及びrep2cap5発現プラスミドへと、野生型AAV2ゲノム配列(GenBank受託番号:J01901)と比較して1022位に挿入した。これは、プロモーターP19の下流のrep遺伝子内である。この位置は、(AAG/G)へと挿入されるイントロンのコンセンサススプライスドナー/アクセプター配列を含有する。
iCreと名付けられたCreのコドン改善バリアントは、Shimshekら(2002年Genesis 32: 19~26頁)によって設計され、CV1-5B細胞へと一時的にトランスフェクトした場合に野生型Creと比較して、「floxed」終止コドンを含有するLacZのほぼ2×のβガラクトシダーゼ活性を回復することが示された(Casanovaら、2002年Genesis 34: 208~14頁)。
Rep発現の調節に加えて、これらも同様に細胞に毒性である可能性があるため、BACにおいてアデノウイルス2ヘルパー遺伝子(HEK293細胞でE2A、E4及びVA)の発現を調節する必要もあり得る(Ferrariら、1996年、Journal Of Virology 70: 3227~3234頁)。したがって、E2A DNA結合タンパク質コード領域及び6個すべてのE4 ORFを含有するE4領域を、PCMV-TO2の下流にそれぞれクローニングした。しかしながら、PolIIIプロモーターから転写されるVAは、PCMV-TO2の下流に置かれた場合、正確に転写されないであろう。したがって、いくつかのTetO配列はネイティブVAプロモーターの上流に置かれ、これらの部位へのTetR-KRABの結合はドキシサイクリンの非存在下では転写を阻害し得る。
Creの発現は条件プロモーター(PCMV-TO2)の制御下にあり、通常の細胞条件下でそれは転写レプレッサーTetR-KRABによって結合される。細胞増殖培地へのドキシサイクリンの添加は、細胞がrAAV産生を開始するのに妥当な密度である場合、TetR-KRABレプレッサーを不安定にし、Cre遺伝子の転写を可能とする。続いて、CreリコンビナーゼはAAV rep遺伝子の組換えイントロンにおいてLoxPに隣接する転写ターミネーターをスプライスアウトし、rep遺伝子の転写を可能とする。この系は、Repが培地へのドキシサイクリンの添加時にのみ細胞において発現され、AAVベクター産生に好適な必要とされる密度に達するまで細胞への毒性を緩和することを意味する。
Tet条件発現系の制御下での任意のリーキーなCre発現は、依然として組換えAAV rep遺伝子から転写ターミネーターを効果的に除去することができ、培養における安定な細胞の生存率を低減させる。したがって、これに対する保護として、Casanovaら(2002年、Genesis 34: 208~214頁)に記載のように、ERT2ドメインと隣接させることによりCre遺伝子を修飾することによってCreリコンビナーゼ制御のさらなる層を追加した。タンパク質のN末端及びC末端のこれらのERT2ドメインは、4-ヒドロキシ-タモキシフェンが細胞培養培地に添加されるまで、Creが核に侵入するのを阻害し、転写レベルでの任意のリーキーな発現の効果を除くことができるタンパク質レベルでCre活性に対する制御レベルを与える。
rep遺伝子から終結イントロンを除去するCreの能力を試験する一時的なトランスフェクション方法
すべてLoxP部位に隣接する3個のSV40ポリA転写終結部位及びハイグロマイシン耐性遺伝子を含有する終結イントロンを、上記のように本発明者らのrep2/cap2及びrep2/cap5発現プラスミドにおいてP19プロモーターの下流のAAV2 rep配列へと挿入した。終結イントロンは、Creも細胞で発現されない限り、これらのプラスミドによってトランスフェクトした細胞におけるRepタンパク質の発現を阻害し得る。Cre発現プラスミドのコトランスフェクションは、2つのLoxP部位を組み換え、転写ターミネーターをスプライスアウトし、RNAポリメラーゼが残りのイントロンをリードスルーすることを可能とする。いくつかのプラスミドが前もって構築され、様々なCreバリアント(野生型Cre、iCre、ERT2-Cre-ERT2及びERT2-iCre-ERT2)がCMVTO2プロモーターの下流にクローニングされ、Tetレプレッサータンパク質の非存在下で細胞において構成的に活性である。Creタンパク質に隣接するERT2も、それらの核への侵入を可能とするリガンドとして作用する4-ヒドロキシ-タモキシフェンを必要とする。

AAVベクター産生を維持する条件プロモーターを有するアデノウイルス2ヘルパー遺伝子の能力を決定する、及びCre依存性rep/capが機能的AAVベクターを産生することができるかどうか決定する試験
アデノウイルス2 E2A及びE4遺伝子のネイティブプロモーターが条件プロモーターCMV-TO2と置き換えられた組換えAAVベクター産生のために、安定なBAC構築物を設計及び構築した。BACは、構成的プロモーターPCMVの下流にDOX感受性「tet-on」転写抑制遺伝子TetR-KRABも有する。この構築物でトランスフェクトした細胞は、E2A及びE4 ORFの上流にCMVTO2プロモーターを結合するTetR-KRABを構成的に発現し、ドキシサイクリンが細胞増殖培地に添加されるまでそれらの転写を遮断する。TetR-KRABも、Tetオペロン配列の近くに結合することによってPol.IIIプロモーターからの転写をブロッキングすることができ、7個のTetオペロンがネイティブPol.IIIプロモーターのVAの上流に置かれ、通常の条件下でこの機能的RNAの発現も遮断され得る。
フラスコのHEK293Tsa細胞(この出願で使用される場合HEK293Tsa細胞は、懸濁適応HEK293細胞を指す)を、公知の陽性対照として一時的なrAAV5産生のための3-プラスミド系の標準的なプラスミドでトランスフェクトした(pG3.Ad2 Helper GSK、pG2.AAV5.R2C5-イントロン及びpG.AAV2.C.GFP.P2a.fLuc.W6.ssb)。
AAV2とAAV5の両方による形質導入に受容性であるCHO細胞を、8×103個の細胞/ウェルで96ウェルプレートに蒔いた(ウェル当たり10% FCS、1×Glutamax及び1×非必須アミノ酸を含有する200μlのDMEM中で増殖する)。次の日、rAAV5を含有する細胞溶解物及び陰性対照溶解物のそれぞれ20μlをCHO細胞のウェルに二連で添加した。CHO細胞のプレートを37℃で5時間インキュベートし、その後、溶解物を含有する培地をウェルから吸引して新しい培地と置き換えた。次いで、プレートを37℃でさらに67時間インキュベートした。次いで、培地を形質導入したCHO細胞から吸引し、細胞を200μlのEDTA溶液によって脱凝集させ、次いでAccuri C6フローサイトメーターで解析してGFP蛍光のレベルを測定し、FlowJoソフトウェアによっても解析した。生細胞集団をゲートにかけ(FSC-A/SSC-A)、次いでゲートを単一細胞に設定した(FSC-A/FSC-H)。非形質導入CHO細胞を使用して、その上であれば細胞はGFP陽性であると考えることができるベースライン蛍光(FL1-A/FSC-A)を設定した。蛍光ベースラインを超える細胞のパーセンテージは、形質導入細胞の各ウェルについて計算し、次いで平均及び標準偏差を二連のそれぞれについて計算した。結果は図3に示す。
AAVバクテリア人工染色体の設計
以下の方法をBACへとエレメントをクローニングするために使用した。
1.各エレメントを、エレメントがBACドナープラスミド、pDonorの多重クローニング部位へとクローニングされるのを可能とするユニークな制限部位を含むプライマーとプルーフリーディングDNAポリメラーゼを使用してPCR増幅する。
2.PCR増幅断片をゲル精製し、TOPOクローニングプラスミド、pCR-BluntII TOPOへとライゲートする。このライゲーションは、化学的にコンピテントな大腸菌を形質転換するために使用する。
3.PCR増幅エレメントを含有するプラスミドを、大腸菌ブロス培養物から抽出し、その部位がPCRプライマーによって導入された2つの制限酵素で消化する。
4.消化したプラスミドをアガロースゲル電気泳動によって分離し、消化したエレメントをゲルから切り出し、精製する。
5.消化及び精製したエレメントをpDonorの多重クローニング部位へとライゲートする。このプラスミドは多重クローニング部位の近位に2個のトリ高感度インスレーター部位(2×cHS4)を含有する。エレメントをpDonorへと方向性クローニングし、その結果2×cHS4はエレメントの3'末端に位置する。メガヌクレアーゼ酵素I-SceIの制限部位は、方向性クローニングしたエレメントの5'末端のpDonor上流に位置するが、メガヌクレアーゼ酵素PI-PspIの制限部位は2×cHS4の3'末端に位置する。このライゲーションは、化学的にコンピテントな大腸菌を形質転換するために使用する。
6.方向性クローニングしたエレメントを有するpDonorプラスミドを、大腸菌ブロス培養物から抽出し、まずメガヌクレアーゼI-SceIで37℃にて消化し、次いでPI-PspIで65℃にて消化する。
7.消化したプラスミドをアガロースゲル電気泳動によって分離し、3'末端に2×cHS4を含む消化したエレメントをゲルから切り出し、精製する。
8.BACは、単一のPI-PspI制限部位を含有する。BAC DNAをPI-PspIで65℃にて消化し、次いで消化反応にFastAPアルカリホスファターゼを添加し、37℃で30分間インキュベートすることによって脱リン酸化する。次いでFastAPを、反応を75℃で5分間インキュベートすることによって不活性化する。
9.メガヌクレアーゼI-SceI及びPI-PspIはDNAを切断し、適合性であるが非対称のオーバーハングを残す。したがって、3'末端に2×cHS4を有するI-SceI及びPI-PspI消化したエレメントは、PI-PspI消化BACへと方向性ライゲートすることができる。エレメントの5'末端のI-SceIオーバーハングはBACのPI-PspIオーバーハングの1つに結合し、もはやI-SceI又はPI-PspIのいずれによっても切断することができない新しい配列を生じる。エレメントの下流の2×cHS4の3'末端におけるPI-PspIオーバーハングは、BACの他のPI-PspIオーバーハングに結合し、ライゲートしたBACの2×cHS4の3'末端に新しいPI-PspI部位の形成をもたらす。このライゲーションは、エレクトロポレーターを使用してエレクトロコンピテントな大腸菌を形質転換するために使用する。
10.3'末端に2×cHS4を有する新しくクローニングしたエレメントを含有するBACを大腸菌ブロス培養物から抽出する。このBACは、PI-PspIによって消化され、脱リン酸化することができ、pDonorへと方向性サブクローニングしたさらなるエレメントをこの部位へとライゲートすることができる。最初にpDonorへとクローニングした任意の新しいエレメントは、BACへとライゲートした場合、先にクローニングした断片からその5'末端に2×cHS4を、それをpDonorへとクローニングした後その3'末端に2×cHS4を常に有する。
AAV5 BAC構築物でトランスフェクトした細胞の安定なプールの生成
rAAV BAC構築物のトランスフェクション及び選択を、接着HEK293T細胞で実施した。振盪培養で、BalanCD培地中で増殖する懸濁適応HEK293Tsa pre-MCB細胞を計数し、10% FCSを含有するDMEM培地中に2×105細胞/mlで再懸濁し、それらの接着行動を戻した。細胞をウェル当たり2mlで6ウェルプレートに蒔き、次いで37℃で終夜インキュベートした。次の日、EGFPトランスファーベクターを含有する安定なAAV BAC構築物:TetR-KRAB iCreBAC9b-GFP及びTetR iCreBAC9b-GFPのマキシプレップを蒔いた細胞へとトランスフェクトした。各トランスフェクションは、300μlのOPTI-MEMに添加したBACマキシプレップからの5μgのDNAを含有し、それに5μlのPEI-proを添加した。チューブを短くボルテックスし、10分間インキュベートした。この後、トランスフェクション混合物を各ウェルに添加した。48時間後、ウェルを吸引し、培地を10% FCS及び300μg/mlのゼオシンを含有するDMEMと置き換えた。プレートは数日間インキュベートし、その間にほとんどの非トランスフェクト細胞が死滅し、ウェルの表面に浮いた。培地を10% FCS及び300μg/mlのゼオシンを含有する新しいDMEMと数回置き換えた。7日後、ウェルの細胞はほとんどEGFP陽性であり、正常速度で倍増した。
TetR-KRAB iCreBAC9b-GFP及びTetR iCreBAC9b-GFP構築物で安定にトランスフェクトしたHEK293T細胞のプールを計数し、10% FCS及び50μg/mlのゼオシンを含有する25mlのDMEM中、フラスコ当たり1×107細胞でT175フラスコに蒔いた。すべてのフラスコの細胞を37℃で終夜インキュベートした。次の日、細胞におけるベクター産生を、培地を2μg/mlのDOX及び5mMの酪酸ナトリウムと10% FCSを含有する新しいDMEMと置き換えることによって誘導した。2つの安定なプールのそれぞれのフラスコは、陰性対照として未誘導のままにした。1つの誘導したフラスコ及び未誘導対照の細胞をDOXの添加の72時間後に回収した。安定なプールのそれぞれの別の誘導したフラスコは、DOXの添加の96時間後に回収した。
AAV5による形質導入に受容性であるCHO細胞を、8×103個の細胞/ウェルで96ウェルプレートに蒔いた(ウェル当たり10% FCS、1×Glutamax及び1×非必須アミノ酸を含有する200μlのDMEM中で増殖する)。次の日、rAAV5を含有する細胞溶解物及び陰性対照溶解物のそれぞれ20μlをCHO細胞のウェルに二連で添加した。CHO細胞のプレートを37℃で5時間インキュベートし、その後、溶解物を含有する培地をウェルから吸引し、新しい培地と置き換えた。次いで、プレートを37℃でさらに67時間インキュベートした。次いで、培地を形質導入したCHO細胞から吸引し、細胞を200μlのEDTA溶液によって脱凝集させ、次いでAccuri C6フローサイトメーターで解析してGFP蛍光のレベルを測定した。生細胞集団をゲートにかけ(FSC-A/SSC-A)、次いでゲートを単一細胞に設定した(FSC-A/FSC-H)。非形質導入CHO細胞を使用して、その上であれば細胞はGFP陽性であると考えることができるベースライン蛍光(FL1-A/FSC-A)を設定した。蛍光ベースラインを超える細胞のパーセンテージは、形質導入細胞の各ウェルについて計算した。誘導した安定なプールからの溶解物によって形質導入した細胞は、ベースラインを超えるいずれのGFP蛍光も示さなかった。
構築物が細胞においてメカニズム上機能的であるか決定するため、DOX誘導及び未誘導細胞からのすべてのポリアデニル化RNAのRNAシークエンシング解析を比較した。TetRKRAB iCreBAC9b-GFP及びTetR iCreBAC9b-GFP構築物で安定にトランスフェクトしたHEK293T細胞のプールを計数し、10% FBSを含有するDMEM中で3×105細胞/mlに希釈し、それぞれウェル当たり2mlで6ウェルプレートに蒔いた。プレートを37℃で終夜インキュベートした。次の日、各安定なプールの3ウェルを、培地を10% FBS、2μg/mlのDOX、及び5mMの酪酸ナトリウムを含有するDMEMに交換することによって誘導した。各プレートの残りの3ウェルでは、培地を新しいDMEM+10% FCSと単に置き換えた。プレートを37℃で5日間インキュベートし、その後未誘導細胞をトリプシンによってそれらを脱凝集することによって回収し、それらを1mlの培地中に再懸濁し、それらを300×gでスピンダウンして、細胞をペレットにした。培地を吸引し、細胞ペレットを-80℃で保存した。誘導細胞の培地は、10% FBS、2μg/mlのDOX、及び5mMの酪酸ナトリウムを含有する新しいDMEMと置き換え、プレートを37℃でさらに2日間インキュベートした。この後、誘導細胞も回収し、ペレットにし、-80℃で保存した。RNA seq解析を、Illuminaを使用するGeneWizによって細胞ペレットから抽出したポリアデニル化RNAで実施した。リードはGSKに戻り、そこでIGVソフトウェアを使用してそれらをBAC配列とアラインした(データは示さない)。
repプロモーターの下流の転写ターミネーターがトランスポゾンITRに隣接し、iCreがトランスポザーゼと置き換えられるさらなるBAC構築物を試験した。
repにおいて組換えトランスポゾンを除去する、及びAAVベクター産生を開始するトランスポザーゼの能力を試験するため、成分を一時的トランスフェクションで試験した。フラスコの懸濁適応HEK293細胞を、以下のプラスミドでトランスフェクトした:
1. rep2/cap5+ヘルパープラスミド + EGFPトランスファーベクタープラスミド
2. rep2/cap5-トランスポゾン3xpA + ヘルパープラスミド + EGFPトランスファーベクタープラスミド

フラスコ当たり60mlのBalanCD HEK293培地、2% Glutamax、0.1% Pluronic F-68中、HEK293Tsa細胞をml当たり2×106細胞で250mlの振盪培養フラスコに播種した。プラスミドは、ヘルパープラスミド対rep/capプラスミド対トランスファーベクターを1.6:1:1のモル比で使用した。プラスミドを、58.5μlのPEI Proを含有する6mlのOpti-MEM培地に添加した。トランスフェクションミックスをボルテックスし、15分間室温でインキュベートした後、細胞を含有する振盪フラスコに添加した。細胞を振盪しながら37℃でインキュベートした。次の日、最終濃度が5mMになるように各フラスコに1Mの酪酸ナトリウムを添加した。
このデータは、予測されるように、標準的な3-プラスミド系でトランスフェクトした細胞が、高レベル(約42%)にレシピエント細胞を形質導入するのに十分な量の組換えAAV5ベクターを産生することができることを示す。

- 1つ以上のヘルパーウイルス遺伝子が転写制御下にある、請求項1~11のいずれか一項に記載のAAVベクタープロデューサー細胞。
- 誘導プロモーター又は転写制御が、それぞれpCMV-TO2プロモーターを含む、請求項6~12のいずれか一項に記載のAAVベクタープロデューサー細胞。
- インスレーターをさらに含み、任意選択で、インスレーターが各核酸配列の間に存在する、請求項1~13のいずれか一項に記載のAAVベクタープロデューサー細胞。
- 選択可能マーカーをさらに含み、任意選択で、選択可能マーカーが増幅可能な選択マーカーである、請求項1~14のいずれか一項に記載のAAVベクタープロデューサー細胞。
- 非哺乳動物の複製起点及び少なくとも25キロベース(kb)のDNAを保持する能力を含む核酸ベクターであって、前記核酸ベクターが、
AAV rep及びcap遺伝子、
ヘルパーウイルス遺伝子、並びに
AAVベクターのDNAゲノム
をコードする核酸配列を含むことを特徴とし、
rep遺伝子がイントロンを含み、前記イントロンが転写終結配列の上流に位置する第1の組換え部位及び下流に位置する第2の組換え部位を有する転写終結配列を含み、
AAV rep及びcap遺伝子、ヘルパーウイルス遺伝子のそれぞれ並びにAAVベクターのDNAゲノムをコードする核酸配列が、核酸ベクター内に個々の発現カセットとして配置される、核酸ベクター。 - イントロンが組換え部位の間に遺伝子をさらに含み、任意選択で、遺伝子が選択可能マーカー遺伝子であり、さらに任意選択で、選択可能マーカーがハイグロマイシンである、請求項16に記載の核酸ベクター。
- リコンビナーゼ遺伝子をコードする核酸配列をさらに含み、核酸ベクター内に個々の発現カセットとして配置される、請求項16又は17に記載の核酸ベクター。
- リコンビナーゼ制御エレメントをさらに含む、請求項16~18のいずれか一項に記載の核酸ベクター。
- リコンビナーゼ制御エレメントが、リコンビナーゼ遺伝子に作動可能に連結した誘導プロモーター及び/又は前記リコンビナーゼ遺伝子と融合したステロイドホルモン受容体リガンド結合ドメインを含む、請求項19に記載の核酸ベクター。
- ステロイドホルモン受容体リガンド結合ドメインがエストロゲン受容体リガンド結合ドメイン(ER)であり、任意選択で、ERがERT2である、請求項20に記載の核酸ベクター。
- ERが前記リコンビナーゼ遺伝子の上流及び下流に作動可能に連結される、請求項21に記載の核酸ベクター。
- 組換え部位がLoxP部位であり、リコンビナーゼ遺伝子がcreリコンビナーゼ遺伝子であり、任意選択で、creリコンビナーゼ遺伝子がコドン最適化creリコンビナーゼ遺伝子(icre)である、請求項18~22のいずれか一項に記載の核酸ベクター。
- 組換え部位がトランスポゾンITRであり、リコンビナーゼ遺伝子がトランスポザーゼ遺伝子であり、任意選択で、トランスポザーゼ遺伝子がコドン最適化トランスポザーゼ遺伝子である、請求項18~22のいずれか一項に記載の核酸ベクター。
- トランスポゾンITR及びトランスポザーゼ遺伝子が真核生物のものである、請求項24に記載の核酸ベクター。
- トランスポゾンITR及びトランスポザーゼ遺伝子が、イラクサギンウワバ(T. ni)、オオキクギンウワバ(M. crassisigna)、バクトロセラ・ミヌタ、オオミノガ、又はオオタバコガ由来である、請求項25に記載の核酸ベクター。
- 個々の発現カセットとして、1つ以上のヘルパーウイルス遺伝子が転写制御下にある、請求項16~26のいずれか一項に記載の核酸ベクター。
- インスレーターをさらに含み、任意選択で、インスレーターが各発現カセットの間に存在する、請求項16~27のいずれか一項に記載の核酸ベクター。
- 選択可能マーカーをさらに含み、任意選択で、選択可能マーカーが増幅可能な選択マーカーである、請求項16~28のいずれか一項に記載の核酸ベクター。
- ベクターが、バクテリア人工染色体、酵母人工染色体、P1由来人工染色体、フォスミド又はコスミドの1つから選択される、請求項16~29のいずれか一項に記載の核酸ベクター。
- 安定なAAVベクタープロデューサー細胞株を産生する方法であって、
(a)請求項16~30のいずれか一項に記載の核酸ベクターを哺乳動物宿主細胞の培養物へと導入すること、及び
(b)培養物内で、哺乳動物宿主細胞の内因性染色体へと組み込まれたベクターにコードされた核酸配列を有する哺乳動物宿主細胞を選択すること
を含む、方法。 - 請求項31に記載の方法によって得られるAAVベクタープロデューサー細胞。
- 複製欠損AAVベクターを産生する方法であって、
(a)請求項16~30のいずれか一項に記載の核酸ベクターを、哺乳動物宿主細胞の培養物へと導入すること、及び
(b)培養物内で、哺乳動物宿主細胞の内因性染色体へと組み込まれたベクターにコードされた核酸配列を有する哺乳動物宿主細胞を選択すること、及び
(c)複製欠損AAVベクターが産生される条件下で、選択された哺乳動物宿主細胞をさらに培養すること
を含む、方法。 - 請求項33に記載の方法によって得られる複製欠損AAVベクター。
- 哺乳動物宿主細胞がHEK293細胞である、請求項31又は33に記載の方法。
