Abstract
Description
本発明は、医学及び細胞生物学の分野に関する。特に本発明は、前原始線条及び/又は中内胚葉細胞を含む組成物、並びにこのような細胞の産生、単離及び使用方法に関する。
ヒト多能性幹細胞、例えば胚性幹(ES)細胞及び胚性生殖(EG)細胞は、1994年に線維芽細胞フィーダーを含有しない培養物中(Bongso et al., 1994)、そして線維芽細胞フィーダーを含有する(Hogan,1997)培養物中で最初に単離された。後に、Thomson、Reubinoff及びShamblottは、有糸分裂不活性化マウスフィーダー層を用いたヒトES及びEG細胞の連続培養を確立した(Reubinoff et al., 2000;Shamblott et al.,1998; Thomson et al., 1998)。

分化因子を提供する前であるか又はそれとほぼ同一時点である。その他の実施形態では、第1の時点は、候補分化因子の提供の後である。本明細書中に記載されるスクリーニング方法のいくつかの実施形態では、ヒト中内胚葉細胞は、候補分化因子に応答して、中胚葉細胞及び/又は胚体内胚葉細胞のような細胞に分化する。いくつかの実施形態では、中胚葉細胞は、FOXF1、FLK1、BMP4、MOX1及びSDF1のようなマーカーの発現により示される。他の実施形態では、胚体内胚葉細胞は、CXCR4及び/又はSOX17のようなマーカーの発現により示される。

原腸形成と呼ばれる初期ヒト発達におけるきわめて重大な段階は、受精後2~3週間で起こる。原腸形成は、3つの一次胚葉が最初に特定化され、そして器官形成されるのがこの時期であるため、非常に有意である(Lu et al., 2001;Schoenwolf and Smith, 2000)。外胚葉は、身体の外被及び全神経系の終局的形成に関与するが、一方、心臓、血液、骨、骨格筋及びその他の結合組織は中胚葉に由来する。胚体内胚葉は、食道、胃並びに小腸及び大腸を含む全腸管、並びに腸管に由来する器官、例えば肺、肝臓、胸腺、上皮小体及び甲状腺、胆嚢及び膵臓の形成に関与する胚葉として定義される(Grapin-Botton and Melton, 2000;Kimelman andGriffin, 2000;Tremblay et al., 2000;Wells and Melton, 1999;Wells and Melton,2000)。非常に重要な区別は、胚体内胚葉と原始内胚葉と呼ば
れる細胞の完全に別個の系統との間でなされるべきである。原始内胚葉は主として、胚体外組織、主に胎盤卵黄嚢の壁側及び臓側内胚葉部分、並びにライヘルト膜の細胞外マトリックス物質である。

図1は、in vitroでのヒト胚性幹細胞(hESC)の初期移行を要約するモデルを示す。原腸形成を厳密に発達反復するプロセスを通してのhESCの分化は、低血清補充の状況での高用量アクチビンAの適用により編成され得る。FGF8の発現及びβ-カテニンの核局在化(原始線条形成前の近位の原外胚葉原に起こる事象である)は、約24時間前に明らかである(前原始線条細胞)。原始線条発現遺伝子(ブラキュリ及びFGF4)の高レベルの発現は、約24時間で起こる。高用量のアクチビンA中に保持される場合、原始線条細胞(中内胚葉細胞)は胚体内胚葉に効率的に転換される。逆に、アクチビンの非存在下では、これらの中内胚葉前駆体は中胚葉になる。BMP4及びSU5402によるhESCの処理は、原始内胚葉及び栄養外胚葉に関連した遺伝子発現を誘導する。
本明細書中に記載される実施形態は、多能性細胞、例えば幹細胞を前原始線条細胞及び/又は中内胚葉細胞に分化させることによる培養物中での前原始線条細胞及び/又は中内胚葉細胞の産生のための新規の限定プロセスに関する。上記のように、前原始線条細胞は、中内胚葉細胞に、並びにそれに由来する細胞、組織及び/又は器官に分化し得る。中内胚葉細胞は、中胚葉細胞及び/又は胚体内胚葉細胞、並びにこれらの系統のいずれかに由来する細胞、組織及び/又は器官に分化し得る。いくつかの実施形態では、前原始線条細胞は、内胚葉系中胚葉中間体を介して、中胚葉又は胚体内胚葉系統のいずれかの最終分化細胞に転換される。以下でさらに詳細に記載されるように、このようなプロセスは、種々のヒト内胚葉及び中胚葉由来組織の効率的産生のための基礎を提供し得る。例えばこのよう
なプロセスは、ヒト内胚葉由来組織、例えば膵臓、肝臓、肺、胃、腸、甲状腺、胸腺、咽頭、胆嚢及び膀胱の効率的産生のための基礎を提供し得る。重要なことに、前原始線条細胞及び/又は中内胚葉細胞の産生が、機能性インスリン産生β細胞への幹細胞の分化における初期段階である。別の例として、前原始線条細胞及び/又は中内胚葉細胞分化は、ヒト中胚葉由来組織、例えば血球細胞、心臓血管性組織、骨格組織、並びにその他の構造及び結合組織の効率的産生のための基礎を提供し得る。有用量の上記細胞又は組織型のいずれかを得るために、分化細胞の最終形に到達する前に起こる分化工程の各々に関して高効率性分化が望ましい。前原始線条細胞及び/又は中内胚葉細胞への幹細胞の分化は中胚葉及び胚体内胚葉細胞系統の機能的最終分化細胞の産生に向かう極初期段階を表わすため(図1に示すように)、この工程での高効率性分化が特に望ましい。

前原始線条細胞を産生するためのいくつかのプロセスにおいて、出発材料として用いられる多能性細胞は、幹細胞である。或る特定のプロセスでは、前原始線条細胞培養物、及び前原始線条細胞を含む富化細胞集団は、胚性幹細胞から産生される。前原始線条細胞を得るための好ましい方法は、前原始線条細胞産生のための出発材料としてヒト胚性幹細胞を利用する。このような多能性細胞は、桑実胚、胚の内部細胞塊、又は胚生殖隆起から得られるものから生じる細胞であり得る。ヒト胚性幹細胞は、当該技術分野で既知である方法を用いて、実質的分化を伴わない多能性状態で培養物中に保持され得る。このような方法は、例えば米国特許第5,453,357号、同第5,670,372号、同第5,690,926号、同第5,843,780号、同第6,200,806号及び同第6,251,671号に記載されている(これらの開示内容は参照により本明細書中に完全に援用される)。
前原始線条細胞へのhESC培養の進行は、前原始線条細胞に特徴的なマーカーの一過性発現を測定することによりモニタリングされ得る。いくつかのプロセスでは、或る特定のマーカーの発現は、マーカーの存在又は非存在を検出することにより決定される。あるいは或る特定のマーカーの発現は、分化因子の添加後の1つ又は複数の時点でマーカーが細胞培養物又は細胞集団の細胞中に存在するレベルを測定することにより決定され得る。このようなプロセスでは、マーカー発現の測定は、定性的又は定量的であり得る。マーカー遺伝子により産生されるマーカーの発現を定量する一方法は、定量的ポリメラーゼ連鎖反応(Q-PCR)の使用による。Q-PCRを実施する方法は、当該技術分野で既知である。当該技術分野で既知である他の方法は、マーカー遺伝子発現を定量するためにも用いられ得る。例えばマーカー遺伝子産物の発現は、当該マーカー遺伝子産物に特異的な抗体を用いることにより検出され得る。或る特定のプロセスでは、前原始線条細胞に特徴的なマーカー遺伝子の発現並びにhESC及び他の細胞型に特徴的なマーカー遺伝子の有意な発現の欠如が決定される。さらに他のプロセスでは、分化因子の添加後の1つ又は複数の時点での前原始線条細胞に特徴的なマーカー遺伝子の発現の時機及び量が測定される。
本明細書中に記載されるプロセスのさらなる態様に関しては、前原始線条細胞は富化され、単離され、且つ/又は精製され得る。いくつかの実施形態では、このような細胞を細胞培養物から単離することにより、前原始線条細胞に関して富化された細胞集団が産生される。

。いくつかの実施形態では、ブラキュリ、FGF4及び/又はSNAI1mRNAは中内胚葉細胞中で実質的に発現されるが、しかしhESC中では発現されない。ほぼピークレベルへのブラキュリ、FGF4及び/又はSNAI1 mRNAの実質的なアップレギュレートは、hESCを適切な分化因子、例えばアクチビンAと接触させた後24時間までに分化中hESC培養物中で観察され得る。この時点で、他の細胞型、例えば原始内胚葉、胚体内胚葉細胞、中胚葉及び外胚葉を示す或る特定のマーカーの発現(表1参照)は、依然として比較的低い。いくつかの実施形態では、原始内胚葉、胚体内胚葉細胞、中胚葉及び外胚葉を示す或る特定のマーカーは、hESCを分化因子と接触させた後、24時間までに実質的に発現されない。いくつかの実施形態では、ブラキュリ、FGF4及び/又はSNAI1mRNA発現は、hESCを分化因子と接触させた後、約24時間後に減少し始める。いくつかの実施形態では、ブラキュリ、FGF4及び/又はSNAI1 mRNA発現は、hESCを分化因子と接触させた後、約30時間後、約36時間後、約42時間後、約48時間後又は約48時間より長い時間の後に減少し始める。
本明細書中に記載されるプロセスのさらなる態様に関しては、中内胚葉細胞は富化され、単離され且つ/又は精製され得る。いくつかの実施形態では、このような細胞を細胞培養物から単離することにより、中内胚葉細胞に関して富化された細胞集団が産生される。
上記の方法により産生される細胞組成物としては、中内胚葉細胞を含む細胞培養物、並びに中内胚葉細胞を富化された細胞集団が挙げられる。例えば、培養中の細胞の少なくとも約5~90%が中内胚葉細胞である中内胚葉細胞を含む細胞培養物が産生され得る。分化プロセスの効率は、或る特定のパラメーター、例えば細胞成長条件、成長因子濃度及び培養工程の時機(これらに限定されない)を修正することにより調整され得るため、本明細書中に記載される分化手法は、中内胚葉細胞への多能性細胞の約5%、約10%、約15%、約20%、約25%、約30%、約35%、約40%、約45%、約50%、約55%、約60%、約65%、約70%、約75%、約80%、約85%、約90%、約95%、又は約95%より多い転換を生じ得る。中内胚葉細胞の単離が用いられるプロセスでは、実質的に純粋な中内胚葉細胞集団が回収され得る。
なくとも約35%、少なくとも約40%、少なくとも約45%、少なくとも約50%、少なくとも約55%、少なくとも約60%、少なくとも約65%、少なくとも約70%、少なくとも約75%、少なくとも約80%、少なくとも約85%、少なくとも約90%、少なくとも約95%、少なくとも約98%、少なくとも約99%又は99%より多くが中内胚葉細胞であるヒト細胞から成る細胞培養物に関する。細胞培養物又は細胞集団がヒトフィーダー細胞を含む実施形態では、上記のパーセンテージは、細胞培養物又は細胞集団中のヒトフィーダー細胞を考慮せずに算出される。
胚体内胚葉を含む細胞培養物及び富化細胞集団を産生するよう多能性細胞を分化させるためのプロセスは、以下で簡単に、そして2004年12月23日に出願された胚体内胚葉(DEFINITIVE ENDODERM)という表題の米国特許第11/021,618号(この開示内容は参照により本明細書中に完全に援用される)で詳細に記載される。これらのプロセスのうちのいくつかでは、出発材料として用いられる多能性細胞は幹細胞である。或る特定のプロセスでは、胚体内胚葉細胞培養物及び胚体内胚葉細胞を含む富化細胞集団は、胚性幹細胞から産生される。胚体内胚葉細胞を得るための好ましい方法は、胚体内胚葉産生のための出発物質としてヒト胚性幹細胞を利用する。このような多能性細胞は、桑実胚、胚の内部細胞塊、又は胚生殖隆起から得られるものから生じる細胞であり得る。ヒト胚性幹細胞は、当該技術分野で既知である方法を用いて、実質的分化を伴わない多能性状態で培養物中に保持され得る。このような方法は、例えば米国特許第5,453,357号、第5,670,372号、第5,690,926号、第5,843,780号、第6,200,806号及び第6,251,671号に記載されている(これらの開示内容は参照により本明細書中に完全に援用される)。
胚体内胚葉細胞へのhESC培養の進行は、胚体内胚葉細胞に特徴的なマーカーの発現を決定することによりモニタリングされ得る。いくつかのプロセスでは、或る特定のマーカーの発現は、マーカーの存在又は非存在を検出することにより決定される。あるいは或る特定のマーカーの発現は、マーカーが細胞培養物又は細胞集団の細胞中に存在するレベルを測定することにより決定され得る。このようなプロセスでは、マーカー発現の測定は、定性的又は定量的であり得る。マーカー遺伝子により産生されるマーカーの発現を定量する一方法は、Q-PCRの使用による。当該技術分野で既知である他の方法は、マーカー遺伝子発現を定量するためにも用いられ得る。例えばマーカー遺伝子産物の発現は、当該マーカー遺伝子産物に特異的な抗体を用いることにより検出され得る。或る特定のプロセスでは、胚体内胚葉細胞に特徴的なマーカー遺伝子の発現並びにhESC及び他の細胞型に特徴的なマーカー遺伝子の有意の発現の欠如が決定される。
る胚体内胚葉細胞はSOX17マーカー遺伝子を発現し、それによりSOX17遺伝子産物を産生する。胚体内胚葉の他のマーカーは、MIXL1、GATA4、HNF3b、GSC、FGF17、VWF、CALCR、FOXQ1、CMKOR1及びCRIP1である。胚体内胚葉細胞はSOX17マーカー遺伝子を、原始及び臓側内胚葉に特徴的であるSOX7マーカー遺伝子より高いレベルで発現するため、いくつかのプロセスでは、SOX17及びSOX7の両方の発現がモニタリングされる。他のプロセスでは、hESCに特徴的であるSOX17マーカー遺伝子及びOCT4マーカー遺伝子の両方の発現がモニタリングされる。さらに、胚体内胚葉細胞はSOX17マーカー遺伝子を、AFP、SPARC又はトロンボモジュリン(TM)マーカー遺伝子より高いレベルで発現するため、これらの遺伝子の発現もモニタリングされる。
上記のプロセスのいずれかにより産生される胚体内胚葉細胞は、このような細胞に特異的であるアフィニティー・タグを用いることにより、富化され、単離され且つ/又は精製され得る。胚体内胚葉細胞に特異的なアフィニティー・タグの例は、抗体、リガンド、あるいは胚体内胚葉細胞の細胞表面に存在するが、しかし本明細書中に記載される方法により産生される細胞培養物中に見出される他の細胞型上には実質的に存在しないポリペプチド等のマーカー分子に特異的なその他の結合剤である。いくつかのプロセスでは、CXCR4と結合する抗体は、胚体内胚葉細胞の富化、単離又は精製のためのアフィニティー・タグとして用いられる。他のプロセスでは、ケモカインSDF-1又はSDF-1を基礎にしたその他の分子もアフィニティー・タグとして用いられ得る。このような分子としては、SDF-1断片、SDF-1融合体、又はSDF-1模倣体が挙げられるが、これらに限定されない。
胞は無作為分化を受ける。しかしながら好ましい方法では、細胞は主として胚体内胚葉に分化するよう指図される。いくつかの好ましい富化、単離及び/又は精製方法は、ヒト胚性幹細胞からの胚体内胚葉のinvitro産生に関する。本明細書中に記載される方法を用いて、細胞集団又は細胞培養物は、非処理細胞集団又は細胞培養物と比較して、少なくとも約2倍~約1000倍、胚体内胚葉細胞内容物を富化され得る。
上記の方法により産生される細胞組成物としては、胚体内胚葉を含む細胞培養物、並びに胚体内胚葉を富化された細胞集団が挙げられる。例えば、細胞培養物又は細胞集団中の細胞の少なくとも約50~99%が胚体内胚葉細胞である胚体内胚葉細胞を含む細胞培養物が産生され得る。分化プロセスの効率は、或る特定のパラメーター、例えば細胞成長条件、成長因子濃度及び培養工程の時機(これらに限定されない)を修正することにより調整され得るため、本明細書中に記載される分化手法は、胚体内胚葉への多能性細胞の約5%、約10%、約15%、約20%、約25%、約30%、約35%、約40%、約45%、約50%、約55%、約60%、約65%、約70%、約75%、約80%、約85%、約90%、約95%、約98%、約99%又は約99%より多い転換を生じ得る。胚体内胚葉細胞の単離が用いられるプロセスでは、例えばCXCR4受容体と結合する親和性試薬を用いることにより、実質的に純粋な胚体内胚葉細胞集団が回収され得る。細胞培養物又は細胞集団がヒトフィーダー細胞を含む実施形態では、上記のパーセンテージは、細胞培養物又は細胞集団中のヒトフィーダー細胞を考慮せずに算出される。
本明細書中に記載される或る特定のスクリーニング方法は、前原始線条細胞及び/又は中内胚葉細胞の分化を促進し得る少なくとも1つの分化因子を同定する方法に関する。これらの方法のいくつかの実施形態では、前原始線条細胞及び/又は中内胚葉細胞、例えばヒト前原始線条細胞及び/又は中内胚葉細胞を含む細胞集団が得られる。次に細胞集団は、候補分化因子を提供される。候補分化因子の提供の前であるか又はほぼ同時である第1の時点で、マーカーの発現が測定される。あるいはマーカーの発現は、候補分化因子の提供後に測定され得る。第1の時点の後であり、候補分化因子を細胞集団に提供する工程の後である第2の時点で、同一マーカーの発現が再び測定される。候補分化因子が胚体内胚葉細胞の分化を促進し得るか否かは、第1の時点でのマーカーの発現を、第2の時点でのマーカーの発現と比較することにより決定される。第2の時点でのマーカーの発現が、第1の時点でのマーカーの発現と比較して増大されるか又は減少される場合には、候補分化因子が胚体内胚葉細胞の分化を促進し得る。
供後、ヒト前原始線条細胞及び/又は胚体内胚葉細胞は、腸管に由来する細胞に分化する。このような細胞としては、膵臓、肝臓、肺、胃、腸、甲状腺、胸腺、咽頭、胆嚢及び膀胱の細胞、並びにこのような細胞の前駆体が挙げられるが、これらに限定されない。さらに、これらの細胞は、より高度な構造、例えば組織及び/又は器官にさらに発達し得る。

物の特定の一過性発現を有する細胞培養物にも関する。このような実施形態では、E-カドヘリンmRNAの発現は、参照時点から約12時間までにダウンレギュレートされ始める。他の実施形態では、E-カドヘリンmRNAの発現は、参照時点から約12時間未満までにダウンレギュレートされ得る。好ましい実施形態では、E-カドヘリンmRNAの発現は、参照時点から約48時間までに実質的にダウンレギュレートされる。
ヒトES細胞
初期発達についてのわれわれの研究のために、多能性であり、そして正常核型を保持しながら培養中に外見上無限に分裂し得るヒト胚性幹細胞を用いた。単離のために免疫学的又は機械的方法を用いて、5日齢胚内部細胞塊からES細胞を得た。特にヒト胚性幹細胞株hESCyt-25を、患者によるインフォームドコンセント後に、invitro受精サイクルからの過剰凍結胚から得た。解凍時に、ES培地(DMEM、20%FBS、非必須アミノ酸、β-メルカプトエタノール、ITSサプリメント)中のマウス胚線維芽細胞(MEF)上に孵化胚盤胞をプレート化した。胚が培養皿に接着し、そして約2週間後、未分化hESCの領域を、MEFとともに新たな皿に移した。機械的に切断し、ディスパーゼで手短に消化し、その後、細胞クラスターを機械的に取り出して、洗浄し、再プレート化することにより移動を成し遂げた。誘導以来、hESCyt-25を、100回にわたって逐次継代した。胚体内胚葉の産生のための出発物質として、hESCyt-25ヒト胚性幹細胞株を用いた。
hESCyt-25特徴づけ
ヒト胚性幹細胞株hESCyt-25は、培養中に18ヶ月にわたって、正常形態学、核型、増殖及び自己再生特性を保持した。この細胞株は、OCT4、SSEA-4及びTRA-1-60抗原(これらはすべて、未分化hESCに特有である)に対する強免疫反応性を示し、そしてアルカリ性ホスファターゼ活性、並びに他の確立されたhESC株と同一の形態学を示す。さらにヒト幹細胞株hESCyt-25は、懸濁液中で培養される場合、胚様体(EB)も容易に形成する。その多能性の実証として、hESCyT-25は、3つの主要胚葉を表わす種々の細胞型に分化する。ZIC1に関するQ-PCR、並びにネスチン及びより成熟したニューロンマーカーに関する免疫細胞化学(ICC)により、外胚葉産生を実証した。β-IIIチューブリンに関する免疫細胞化学染色を、初期ニューロンに特徴的な伸長細胞のクラスター中で観察した。予め、レチノイン酸で懸濁液中のEBを処理して、臓側内胚葉(VE)、胚体外系統への多能性幹細胞の分化を誘導した。処理細胞は、高レベルのα-フェトタンパク質(AFP)及びSOX7(VEの2つのマーカー)を54時間の処理により発現した。単層中で分化された細胞は、免疫細胞化学染色により実証されるように、散在性パッチ中でAFPを発現した。以下で記載するように、hESCyT-25細胞株は、AFP発現の非存在下でのSOX17に関する実時間定量的ポリメラーゼ連鎖反応(Q-PCR)及び免疫細胞化学により立証されるように、胚体内胚葉も形成し得た。中胚葉への分化を実証するために、分化中のEBを、いくつかの時点でのブラキュリ遺伝子発現に関して分析した。ブラキュリ発現は、実験経過中に漸進的に増大した。上記に鑑みて、三胚葉を表わす細胞を形成する能力により示されるように、hESCyT-25株は多能性である。
胚体内胚葉細胞
2004年12月23日に出願された胚体内胚葉(DEFINITIVE ENDODERM)という表題の同時係属中の共有米国特許第11/021,618号は、ヒト胚体内胚葉細胞を含む細胞培養物及び富化細胞集団を記載する。そこには、分化因子の存在下での分化によるhESCからの胚体内胚葉を産生する方法、並びに混合細胞培養物及び/又は細胞集団からこれらの胚体内胚葉細胞を富化、単離及び/又は精製するための方法も記載されている。さらにそこには、胚体内胚葉細胞の同定及び/又は検出のために有用であるマーカー、並びにこのような細胞の精製に有用なマーカーが記載されている。2004年12月23日に出願された胚体内胚葉(DEFINITIVE ENDODERM)という表題の米国特許出願第11/021,618号の開示内容は、全体が参照により本明細書中に完全に援用される。
胚体内胚葉前駆体細胞における遺伝子発現の時系列
胚体内胚葉への分化中に起こる遺伝子発現のダイナミクスを査定するために、2つの異なる4日間分化プロトコール中の種々の時点での分化中の細胞培養物において、多数の遺伝子の発現をモニタリングした。
始めた。BMP4/SU5402による処理は、FGF8発現のレベルの非常に少ない変化を引き起こした(図3A)。これらの結果は、アクチビンAがFGF8発現細胞へのhESCの迅速な移行を媒介する、ということを実証する。FGF8発現は原外胚葉における後方パターン形成の第一指標の1つであるので、分化中のhESCにおけるFGF8の急速なアップレギュレートは、これらの初期段階の細胞がhESC細胞型から分化して、「前線条」(前原始線条)細胞集団を形成した、ということを示す。
ヒト胚性幹細胞上皮-間充織転換
脊椎動物原腸形成中、原始線条での上皮-間充織転換(EMT)を受けている原外胚葉細胞は、細胞表面でのE-カドヘリンタンパク質の発現パターンを変える。FGF及びTGFβシグナル伝達の複合作用は、E-カドヘリンの直接転写抑制因子であるジンクフィンガー転写因子SNAI1を誘導するのに役立つ。本実施例は、原腸形成中にinvivoで起こるEMTにおけるのとまさに同様に、in vitroでアクチビン曝露hESC培養物の初期分化段階中に、E-カドヘリンの転写が抑制される、ということを示す。
疫細胞化学を用いて、ブラキュリ、E-カドヘリン及び活性化(非リン酸化)B-カテニンの発現を測定した。
胚体内胚葉への中内胚葉の分化
本実施例は、胚体内胚葉に分化する中内胚葉前駆体細胞の応答能を実証する。
て12~24時間までに遅延された。分化の48時間後、SOX17mRNA及びタンパク質発現が急速に増大しつつあり、そしてブラキュリ発現が急速に減少しつつあった場合、SOX17陽性細胞のほとんどがブラキュリを同時発現した(図6A~図6D)。これらのデータは、100ng/mlのアクチビン及び低血清の存在下で、大多数のSOX17陽性細胞がブラキュリ陽性中内胚葉細胞に由来した、ということを明らかに実証する。さらに、これらの知見は、ヒト発達における中内胚葉中間体の存在を高度に示唆する。さらに、ブラキュリ発現は発達中の任意の時点で原始内胚葉系統の細胞中で起きないため、これらの知見は最終的に、アクチビン産生内胚葉の決定的性質を立証する。
中内胚葉細胞からの中胚葉及び胚体内胚葉の形成
上記の実施例に加えて、本明細書中に記載される中内胚葉中間体が中胚葉又は胚体内胚葉細胞への分化を受け得る、ということを実証するために実験を実行した。
0.5%血清を含有する培地中での胚体内胚葉産生
胚体内胚葉細胞の産生に及ぼす血清低減の作用をさらに実証するために、hESCを、種々の血清濃度を含有する培地中で胚体内胚葉細胞に分化させた。
クチビンAに応答して減少されるマーカー(非胚体内胚葉細胞型のマーカー、例えばブラキュリ、MOX1、SOX7、SOX1及びZIC1)の場合、血清濃度の低減はこれらのマーカーに対応するmRNAの産生の実質的低減を生じた(図10E~図10I)。
SOX17発現のアップレギュレートはFOXA2発現のアップレギュレートに先行する本実施例は、胚体内胚葉へのhESCのアクチビン媒介性分化において、SOX17mRNA発現はFOXA2 mRNA発現の前にアップレギュレートされる、ということを実証する。
8つの異なるhESC株中での胚体内胚葉の産生
本実施例は、本明細書中に記載される方法を用いて、8つの独立して得られたhESC株から胚体内胚葉細胞が産生され得る、ということを示す。
マウス腎臓被膜下のヒト胚体内胚葉細胞の移植
本明細書中に記載される方法を用いて産生されたヒト胚体内胚葉細胞が、腸管由来細胞を産生するよう分化因子に応答し得ることを実証するために、このようなヒト胚体内胚葉細胞をinvivo分化プロトコールに付した。
FGF8プロモーター-EGFP及びブラキュリプロモーター-EGFPトランスジェニックヒト胚性幹細胞株の生成
細胞単離のためにFGF8及びブラキュリマーカーを使用するために、発現可能レポーター遺伝子と融合されたFGF8又はブラキュリ遺伝子プロモーターを有するhESCを構築する。特に本実施例は、FGF8調節領域の制御下のレポーター遺伝子を含むレポーターカセットを含むベクターの構築を記載する。さらに、ブラキュリ調節領域の制御下のレポーター遺伝子を含むレポーターカセットを含むベクターの構築が記載される。本実施例は、1つ又は複数のこれらのベクターでトランスフェクトされた細胞、例えばヒト胚性幹細胞、並びにそのゲノム中に組み込まれたこれらのレポーターカセットのこの一方又は両方を有する細胞の調製も記載する。
いてGFP又はEGFP以外のレポーター遺伝子を用い得るが、但しレポーターはFACSにより細胞分離を可能にする、と理解される。
前原始線条細胞に富んだ細胞集団の産生
以下の実施例は、FGF8プロモーター/EGFPカセットを含むhESCが前原始線条細胞に分化し、次にその後、蛍光標示式細胞分取器(FACS)により単離し得る、ということを実証する。
中内胚葉細胞に富んだ細胞集団の産生
以下の実施例は、ブラキュリプロモーター/EGFPカセットを含むhESCが原始線条(中内胚葉)細胞に分化し、次にその後、蛍光標示式細胞分取器(FACS)により単離し得る、ということを実証する。
ヒト前原始線条細胞及び/又は中内胚葉細胞のin vitroでの分化を促進し得る分化因子の同定
本明細書中に記載される分化因子スクリーニング方法を例示するために、種々の時点で或る特定のマーカー遺伝子産物の正規化発現レベルを測定しながら、本明細書中に記載される方法を用いて産生されるヒト前原始線条細胞及び中内胚葉細胞の集団に、別々にいくつかの候補分化因子を提供する。
初期胚細胞型におけるマーカー発現の要約
本実施例は、hESCの初期分化中に得られる細胞型の同定及び/又は検出に有用なマーカー遺伝子の発現を要約する表を提供する。下記の表1において、各マーカーに関する正
式名及び略号を最初の2つの縦列に提示する。次の縦列の各々は、遺伝子が特定組織型中で発現されるか否かを記載する。細胞型に関する以下の略号を用いる:ICM/ESCはhESCを指す;PSは原始線条(中内胚葉)細胞を指す;Ectoは外胚葉細胞を指す;Mesoは中胚葉細胞を指す;DEは胚体内胚葉細胞を指す;PrEは原始内胚葉を指す;VEは臓側内胚葉を指す;そしてPEは壁側内胚葉を指す。
前原始線条細胞及び中内胚葉細胞を同定及び/又は検出するために有用な付加的マーカー本実施例は、前原始線条細胞及び/又は中内胚葉細胞を同定及び/又は検出するのに有用な付加的マーカーを記載する。本実施例中に記載される前原始線条マーカーは、前原始線条細胞に関する上記のマーカーのいずれかの代わりに又はそのほかに用いられ得る、と理解される。本実施例中に記載される中内胚葉マーカーは、中内胚葉細胞に関する上記のマーカーのいずれかの代わりに又はそのほかに用いられ得る、とも理解される。
前原始線条段階(原始外胚葉細胞)前に胚性幹細胞分化を同定及び/又は検出するために有用なマーカー
本実施例は、前原始線条段階前に幹細胞分化を同定及び/又は検出するのに有用であるマーカーを記載する。このような細胞型は、ここでは、原始外胚葉細胞と呼ぶ。本実施例中に記載される原始外胚葉細胞マーカーは、原始外胚葉細胞組成物と関連付けられ、そして前原始線条細胞及び/又は中内胚葉細胞を産生し、スクリーニングするためにすでに記載した方法で用いられ得る、と理解される。

Claims (20)
- 培地の5%(v/v)未満が血清である、培地及びヒト中内胚葉細胞を含む細胞培養物。
- TGFβスーパーファミリー成長因子のメンバーをさらに含む、請求項1に記載の細胞培養物。
- TGFβスーパーファミリー成長因子がノーダル、アクチビンA及びアクチビンBから成る群より選択される、請求項2に記載の細胞培養物。
- TGFβスーパーファミリー成長因子がアクチビンAである、請求項3に記載の細胞培養物。
- TGFβスーパーファミリー成長因子がアクチビンA及びアクチビンBである、請求項3に記載の細胞培養物。
- TGFβスーパーファミリー成長因子がアクチビンA及びノーダルである、請求項3に記載の細胞培養物。
- Wntファミリーのメンバーをさらに含む、請求項1~6のいずれか1項に記載の細胞培養物。
- WntファミリーのメンバーがWnt3aである、請求項7に記載の細胞培養物。
- 培地の2%(v/v)未満が血清である、請求項1~8のいずれか1項に記載の細胞培養物。
- 培地の1%(v/v)未満が血清である、請求項1~8のいずれか1項に記載の細胞培養物。
