密度アッセイ
最適な6’SLN密度を明らかにするために、3つの異なる6−シアリル−N−アセチルラクトースアミン(6’SLN)密度のNPを合成した(図1)。また、PEG(5)及び3−シアリル−N−アセチルラクトースアミン(3’SLN)でコーティングした金ナノ粒子を用いた対照実験も行った。
ウイルスとNPとの相互作用は、電子顕微鏡観察(TEM)でも示された。H3N2 Vic/11ウイルスをLD 6’SLN NPと1時間インキュベートした。TEMのグリッドを調製した後、メチルタングステン染色を行った。ウイルスの大部分はLD 6’SLN NPで完全に覆われていた(図2B)。PEG(5)NPを用いた対照実験では、NPは全周に渡っていたが、ウイルスのエンベロープには付着していなかった(図2C)。
インフルエンザのパンデミックは、通常、動物のインフルエンザ株とヒトのインフルエンザ株が混じり合うときに現れる。それゆえ、次の目標は、本発明のNPで鳥インフルエンザ株を不可逆的に阻害することである。予備的な研究を、卵馴化ウイルス株であるCAL/09を用いて行った。Neth/09とCAL/09は非常によく似た2種のヒトH1N1株である。LD−6’SLN NPはNeth/09株に対して強い活性を有する。しかしながら、鶏卵を用いて複製されたCAL/09株は、LD 3’SLN NPとより高い親和性で結合する(表4)。この結果は、LD−3’SLN NPが鳥インフルエンザ株を阻害することを示す。
ヒトインフルエンザウイルスは、C15−6’SLNをグラフトした金NPで不可逆的に阻害された。しかしながら、医薬品への応用には、金コアを有機材料に変えることが重要である。ポリマー、デンドリマー、デンドロン等の異なる有機材料の中でも、シクロデキストリン(CD)は、HAを標的にするために6’SLNを持つリガンドをグラフトするのに好ましいコアである。
化学構造と抗ウイルス活性の関係を調べるために、β−シクロデキストリン(β−CD)を、三糖を用いて及び用いずに、異なるリガンドで修飾した。例示的な修飾シクロデキストリンを図4に示す。これらは、1H核磁気共鳴分光法(NMR)で測定したところ、同等の数の三糖を有する(図5及び表5参照)。インフルエンザA/Netherlands/2009(H1N1)株(A/NL/09)に対する用量反応アッセイを行い、これらのNPの阻害活性を比較した(図6)。感染は、感染後24時間(感染後の時間、hpi)に、免疫細胞化学的なアッセイで定量化した。十分に長い疎水性リガンドと6’SLN末端基を持つβ−CD(C6−6’SLN、C11−6’SLN及びC14−6’SLN)は、インフルエンザA/NL/09による細胞への感染に対して強い阻害活性を示し、EC50値はナノモル範囲であった。他方、より短いリガンドC1−6’SLNを有するβ−CDは、感染をほとんど阻害しなかった。十分に長いリガンドを導入することで、末端基の柔軟性が明らかに高まり、その結果、阻害濃度が減少した。疎水性のリガンドを親水性のPEG8リガンドに置き換えた場合(PEG8−6’SLN)、EC50は同等であった(ただし若干高かった)。
ヒトの気道上皮を再構成した3DモデルであるMucilAir(登録商標)を用いて、エキソビボ実験を行った。この気液界面培養物は、ヒトにおけるインフルエンザウイルス複製の主要な部位であるヒトの上気道上皮の偽重層構造(基底細胞、繊毛細胞、杯細胞)とバリアー防御機構(すなわち、粘膜繊毛クリアランス及び上皮細胞免疫)の両方を完全に模倣する。エキソビボ実験は、適応バイアスを排除するため、細胞内で継代培養されていない臨床H1N1パンデミック09株を用いて行った。C11−6’SLN又はP8−6’SLN(50μg/組織)とウイルス(104RNAコピー/組織)を、事前にインキュベートすることなく、組織の頂部(アピカル)表面に最初に同時に添加した。4時間後、接種物を除去し、組織を洗浄し、ナノ粒子を再添加することなく、組織の頂部洗浄液からqPCRを用いて感染の進行を毎日モニタリングした。C11−6’SLNは、実験の全過程を通じてウイルスの複製を完全に阻止したのに対して、P8−6’SLNは、感染後最初の2日間(感染後の日数、dpi)はウイルスの複製をわずかに低下させたが、それ以降は阻止しなかった(図11a)。
BALB/cマウスを用いて、共処置と後処置の両方の条件でインビボ実験を行った。共処置実験では、マウスにC11−6’SLN(25μg/マウス)とA/NL/09(100感染粒子/マウス)を同時に経鼻経路で投与した。C11−6’SLNの投与が感染動物の生理的状態に与える影響を推定するために、マウスの体温と体重を毎日測定した。2dpiにおいて、マウスの半分を無作為に安楽死させ、残りのマウスをC11−6’SLNで再処置した。第2群のマウスは4dpiで安楽死させた。気管支肺胞洗浄液(BAL)からウイルスの力価を定量した(図15a)。処置を受けたマウスでは、感染後2日目及び4日目にウイルス力価の有意な低下が観察された(図15a)。また、C11−6’SLNの抗ウイルス作用により、無処置のマウスに比べて体重と体温の両方が有意に維持され、罹患率が著しく低下した(図15b及び図15c)。総合すると、これらの結果は、C11−6’SLNナノ粒子は、インビボでの肺におけるウイルスの感染と拡散を防ぐ能力があることを実証する。
修飾シクロデキストリンの合成
化学物質:Neu5Acα(2,6)−Galβ(1−4)−GlcNAc−β−エチルアミン及びNeu5Acα(2,3)−Galβ(1−4)−GlcNAc−β−エチルアミンはTCI chemicals(ティーシーアイ・ケミカルズ)から購入した。ヘプタキス−(6−デオキシ−6−メルカプト)−β−シクロデキストリン及びカルボキシメチル−β−シクロデキストリンナトリウム塩は、Cyclodextrin−Shop(シクロデキストリンショップ)から購入した。11−ドデセン酸はabcr GmbH(アーベーツェーエル)から購入した。14−ペンタデセン酸はLarodan AB(ラロダン)から購入した。マレイミド−PEG8−CH2CH2COOHはPurePEG(ピュアーペグ)から購入した。その他の化学物質及び溶媒はすべてSigma−Aldrich(シグマ・アルドリッチ)から購入した。
5mLのDMSO中で、0.04mmolのヘプタキス−(6−デオキシ−6−メルカプト)−β−シクロデキストリンを、UVランプ(250W)の存在下で、0.28mmolのアリル末端基とカルボン酸末端基を持つ2官能分子(リガンド)(6−ヘプテン酸、11−ドデセン酸又は14−ペンタデセン酸等)と一晩撹拌した。得られたヘプタキス−(6−デオキシ−6−アルカン酸チオン)−β−シクロデキストリン誘導体を、DCM−ジエチルエーテル混合物から遠心分離により沈殿させ、真空下で乾燥させた。
それぞれ工程1で得られたシクロデキストリン誘導体0.04mmolを、1.12mmolのN−ヒドロキシスクシンイミド(NHS)、0.56mmolのエチル−3−(3−ジメチルアミノプロピル)カルボジイミド(EDC−hcl)、及び0.02mmolの4−ジメチルアミノピリジン(DMAP)とともに、5mLのDMSO中で一晩撹拌した。得られたNHS活性化シクロデキストリン誘導体を、まず氷冷した水で沈殿させ、アセトニトリル及びジエチルエーテルで洗浄した。生成物を真空下で乾燥させた。
5.6μmolのNeu5Acα(2,6)−Galβ(1−4)−GlcNAc−β−エチルアミン(C11−3’の場合はNeu5Acα(2,3)−Galβ(1−4)−GlcNAc−β−エチルアミン)を、工程2で得られたシクロデキストリン誘導体1.6μmolと混合した。15μmolのトリエチルアミン(TEA)を加え、1mLのDMSO中で一晩反応させた。反応生成物を0.01Mリン酸バッファ(pH:7.5)で希釈し、アミコンフィルター(MWCO:3k)を用いて濃縮した。得られたヘプタキス−(6−デオキシ−6−SLN−エチルカルボキサミド−アルキルチオ)−β−シクロデキストリン誘導体をさらに蒸留水で洗浄し、凍結乾燥によって乾燥した。リガンド修飾シクロデキストリンへの三糖のグラフト化は、1H及びDOSY NMR研究で確認した(図16及び図17)。
β−CDのC15修飾:55mgのチオール修飾β−CDと70mgの14−ペンタデセン酸を5mLのDMSO中で、紫外線下で一晩撹拌した。
PEG(5)NPの合成:15mLのEtOH中の88.6mgのテトラクロロ金(3)酸三水和物(HAuCl4・3H2O)を、5mLのEtOH中の56mgのHS−PEG(5)と10分かけて混合した。この混合物に、37.5mLのEtOH中の94.6mgの水素化ホウ素ナトリウム(NaBH4)を滴下して加えた。完全なNP形成のために、一晩反応させた。結果として得られたNPは、EtOH−H2O溶媒混合物を使用してアミコンフィルターを用いて洗浄した。
材料:DMEM−Glutamax培地はThermo Fischer Scientific(サーモフィッシャーサイエンティフィック)から購入した。洗浄バッファ用のTween 20(登録商標)及び3,3’−ジアミノベンジジン(DAB)錠剤はSigma Aldrichから購入した。一次抗体(A型インフルエンザモノクローナル抗体)は、Light Diagnostics(ライト・ダイアグノスティクス)から購入した。二次抗体(抗マウスIgG、HRP結合抗体)はCell Signaling Technology(登録商標)から購入した。テトラゾリウム化合物[3−(4,5−ジメチルチアゾール−2−イル)−5−(3−カルボキシメトキシフェニル)−2−(4−スルホフェニル)−2H−テトラゾリウム、分子内塩;MTS]と電子結合試薬(フェナジン・エトスルフェート;PES)を含むCellTiter 96(登録商標) AQueous One Solution Cell Proliferation AssayはPromega(プロメガ)から購入した。
MDCK(メイディン・ダービー・イヌ腎臓細胞、Madin−Darby Canine Kidney Cells)細胞株は、ATCC(アメリカ合衆国培養細胞系統保存機関(American Type Culture Collection)、ロックビル(Rockville)、メリーランド州)から購入した。この細胞は、10%ウシ胎仔血清(FBS)及び1%ペニシリン/ストレプトマイシンを含むグルコース添加ダルベッコ改変イーグル培地(DMEM+GlutaMAX(商標))中で培養した。MDCK細胞株は、CO2(5%)を含む加湿空気中で37℃で培養した。
HSV−2の臨床分離株は、もともとはM.Pistello教授(ピサ大学(University of Pisa)、イタリア)から提供されたもので、Vero細胞でのプラークアッセイにより増殖させ、滴定した。H1N1 Neth09及びB Yamagataは、M.Schmolke教授(University of Geneva(ジュネーブ大学))からの寄贈品である。鳥類株NIBRG−23(A/turkey/Turkey/1/2005 H5N1表面タンパク質及びA/PR/8/34(H1N1)バックボーンを用いて逆遺伝学的に調製したもの)は、イギリス国立生物学的製剤研究所(National Institute for Biological Standards and Controls)、ポッターズ・バー(Potters Bar)、英国から入手し、10日齢の胚化鶏卵でさらに増殖させた後、ウイルスの精製と特性評価を行った。臨床サンプルは、University Hospital of Geneva(ジュネーブ大学病院)から匿名の患者から提供された。すべてのインフルエンザ株は、TPCK処理したトリプシン(0.2mg/ml)の存在下、MDCK細胞上でICCにより増殖、滴定した。
細胞生存率は、MTS[3−(4,5−ジメチルチアゾール−2−イル)−5−(3−カルボキシメトキシフェニル)−2−(4−スルホフェニル)−2H−テトラゾリウム]アッセイにより測定した。96ウェルプレートに播種したコンフルエントな細胞培養物を、抗ウイルスアッセイについて記載したのと同じ実験条件で、異なる濃度のナノ粒子又はリガンドと三重にインキュベートした。CellTiter 96 Proliferation Assay Kit(Promega、マディソン(Madison)、ウィスコンシン州、米国)によって、製造者の使用説明書に従って細胞生存率を測定した。吸光度は、マイクロプレートリーダー(Model 680、BIORAD(バイオ・ラッド))を用いて490nmで測定した。ナノ粒子又はシクロデキストリンの異なる濃度における細胞生存率への影響は、処理した細胞の吸光度を培養液のみでインキュベートした細胞の吸光度と比較することにより、パーセントで表した。50%細胞毒性濃度(CC50)及び95%信頼区間(CI)は、Prismソフトウェア(Graph−Pad Software(グラフパッド・ソフトウェア)、サンディエゴ、カリフォルニア州)を用いて求めた。
MDCK細胞を96ウェルプレートに24時間前にプレプレーティングした。濃度を上げた材料をインフルエンザウイルス(MOI:H5N1では0.02又は0.01、その他のウイルスでは0.1)と37℃で1時間インキュベートした後、この混合物を細胞に添加した。ウイルスの吸着(37℃で1時間)後、接種したウイルスを除去し、細胞を洗浄して新鮮な培地を加えた。37℃で24時間インキュベートした後、免疫細胞化学(ICC)アッセイで感染を分析した。細胞を固定し、メタノールで透過処理した。その後、一次抗体(1:100希釈)を加え、37℃で1時間インキュベートした。細胞を洗浄バッファ(DPBS+Tween 0.05%)で3回洗浄した後、2次抗体(1:750希釈)を加えた。1時間後、細胞を洗浄し、DAB溶液を加えた。感染細胞を数え、処理した条件と未処理の条件での感染細胞の数を比較して感染率を算出した。
ウイルス(フォーカス形成単位(ffu):105/mL)と試験材料(EC99濃度、表7)を37℃で1時間インキュベートした。ウイルス−材料複合体の連続希釈を、未処理の対照とともに行い、細胞上に移した。1時間後、上記混合物を除去し、新鮮な培地を加えた。翌日、ウイルスの力価をICCアッセイで評価した。ICCアッセイでは、上述の同じ手順に従った。
すべての結果は、二重に行った3回の独立した実験の平均値として提示されている。阻害曲線のEC50値は、GraphPad Prismバージョン5.0(GraphPad Software、サンディエゴ、カリフォルニア州、米国)というプログラムを用いて回帰分析を行い、可変勾配−シグモイド用量反応曲線に当てはめて算出した。
共処理番号1:H1N1 Neth/09株(pfu:104〜105)及びCD−C15−6’SLN(400μg/mL)をMucilAir(ヒト気道上皮の再構築物)に同時に添加した。4時間後、上清を除去し、新鮮な培地を加えた。上清のウイルス力価を24時間ごとにqPCRで追跡した。
共処理番号2:インビトロで再構築したヒト気道上皮、MucilAir組織(Epithelix Sarl(エピセリクス)、ジュネーブ、スイス)を、健康なドナーに由来する鼻ポリープ上皮細胞の混合物から気液界面で培養した。インフルエンザH1N1 pdm 2009臨床株(104rnaコピー/組織)及びC11−6’(50μg/組織)を、未処理の対照とともに、事前にインキュベートすることなく組織に移した。4時間のインキュベーション後、組織を2回洗浄した。1日ごとに基礎培地を交換した。毎日qPCR測定を行うために、200μLの培地を組織に加え、20分後に回収した。EZNAウイルス抽出キット(Omega Biotek(オメガ・バイオテック))で抽出したRNAを、StepOne ABI Thermocycler中でQuantiTectキット(番号204443;Qiagen(キアゲン)、ヒルデン(Hilden)、ドイツ)を用いてqPCRを使用することにより定量した。
インフルエンザ感染細胞は、A型インフルエンザ抗体(Light Diagnostic)を用いて直接検出し、βチューブリン一次ウサギ抗体(Abcam(アブカム))を繊毛細胞のマーカーとして使用した。二次抗体としてAlexa 488−ヤギ抗ウサギ抗体及びAlexa 594−ヤギ抗マウス抗体(Life Technologies(ライフ・テクノロジーズ))を使用し、核をDAPIで染色した。画像は、Zeiss LSM 700 Meta共焦点顕微鏡で取得し、Imarisで処理した。
基礎培地中のLDH放出は、Cytotoxicity Detection Kit(Roche(ロシュ) 04744926001)を用いて測定した。
MTT溶液をMucilAir培地(1mg/ml)で希釈し、300μlを24ウェルプレートに基礎培地として加えた。37℃で4時間培養した後、組織を新しいプレートに移し、1mlのDMSOで溶解した。上清を570nmで読み取った。処理した組織と未処理の組織を比較して、生存率を算出した。
インターロイキン−6(IL−6)、CXCモチーフケモカイン10(CXCL10又はIP−10)、CCモチーフケモカイン5(CCL5又はRANTES)、インターロイキン−8(IL−8又はCXCL−8)、及びインターフェロンλ(IL−29/IL−28B)を、異なる濃度のCDで毎日処理した後に、ELISA法(R&D DY206−05、DY266−05、DY278−05、DY208−05、DY1598B−05)により基礎培地中で測定した。
前処置:5匹のBALB/cマウスの4群を、0日目に50μlのPBS又はC11−6’(50μl中25μg)で処置し、直ちにA/NL/09(102ffu)を接種した。毎日マウスの体温と体重を測定した。感染後2日目に2群のマウスを犠牲にした(1群はpbs、もう1群はC11−6’処置)。肺ホモジネートと鼻粘膜、及び気管支肺胞洗浄液を採取し、qPCR測定によりウイルスの力価を定量した。C11−6’処置群は、同量のナノ粒子で再処置した。2日後、残ったすべてのマウスを犠牲にし、肺ホモジネートと鼻粘膜を採取した。組織を破壊した後、RNAをTrizolで抽出し、qPCRを用いて定量し、BALはプラークアッセイに供した。2回の独立した実験を行った。
一定ウイルス濃度(ffu:103)を、用量の異なるナノ粒子と一緒に、37℃で1時間インキュベートした。その後、この混合物を細胞に移した。1時間後、混合物を除去し、細胞を洗浄した。翌日、免疫細胞化学(ICC)アッセイで感染を分析した。

Claims (18)
- コアと、前記コアに共有結合された複数のリガンドとを含む殺ウイルス性ナノ粒子であって、前記リガンドの少なくとも一部分は三糖部分を含み、
前記コアはシクロデキストリンであり、
前記リガンドは、同じであるか又は異なり、置換されていてもよいアルキルに基づくリガンドであり、
各三糖部分は、3−シアリル−N−アセチルラクトースアミン(3’SLN)及び6−シアリル−N−アセチルラクトースアミン(6’SLN)から選択される殺ウイルス性ナノ粒子。 - シクロデキストリンが、α−シクロデキストリン、β−シクロデキストリン、γ−シクロデキストリン又はこれらの組み合わせを含む群から選択される請求項1に記載の殺ウイルス性ナノ粒子。
- 前記リガンドが、置換されていてもよいC4〜C30アルキルに基づくリガンドである請求項1又は請求項2に記載の殺ウイルス性ナノ粒子。
- 前記リガンドが、置換されていてもよいC6〜C15アルキルに基づくリガンド化合物である請求項1から請求項3のいずれか1項に記載の殺ウイルス性ナノ粒子。
- 前記リガンドの一部又はすべてが3’SLNを含み、前記リガンドの一部又はすべてが6’SLNを含み、前記リガンドのすべてではなく一部が三糖部分を含まない請求項1から請求項4のいずれか1項に記載の殺ウイルス性ナノ粒子。
- シクロデキストリンが、α−シクロデキストリン、β−シクロデキストリン及びγ−シクロデキストリンを含む群から選択される請求項6に記載の殺ウイルス性ナノ粒子。
- mが3又は4である請求項6又は請求項7に記載の殺ウイルス性ナノ粒子。
- 式(II)によって表される殺ウイルス性ナノ粒子、又はその薬学的に許容できる塩である殺ウイルス性ナノ粒子であって、
各Rは、独立に、置換されていてもよいアルキルに基づくリガンドであり、前記リガンドのうちの少なくとも2つは、3−シアリル−N−アセチルラクトースアミン(3’SLN)及び6−シアリル−N−アセチルラクトースアミン(6’SLN)を含む群から選択される三糖部分を有するか、又は前記リガンドのうちの少なくとも1つは3’SLNを有しかつ別のリガンドは6’SLNを有し、
各R’は、独立に、H、−(CH2)y−COOH、−(CH2)y−SO3 −、ポリマー又は水可溶化部分であり、
xは6、7又は8であり、
yは4〜20の整数である

- 前記アルキルに基づくリガンドが、置換されていてもよいC4〜C30アルキルに基づくリガンドである請求項9又は請求項10に記載の殺ウイルス性ナノ粒子。
- 前記アルキルに基づくリガンドが、6’SLNを含む置換されていてもよいC6〜C15アルキルに基づくリガンドである請求項9から請求項11のいずれか1項に記載の殺ウイルス性ナノ粒子。
- 有効量の1種以上の請求項1から請求項12のいずれか1項に記載の殺ウイルス性ナノ粒子と、少なくとも1種の薬学的に許容できる賦形剤、担体及び/又は希釈剤とを含む医薬組成物。
- インフルエンザウイルス感染症及び/若しくはインフルエンザウイルスと関連する疾患を治療並びに/又は予防することにおいて使用するための、請求項1から請求項12のいずれか1項に記載の殺ウイルス性ナノ粒子。
- 有効量の1種以上の請求項1から請求項12のいずれか1項に記載の殺ウイルス性ナノ粒子と、任意に少なくとも1種の好適なエアロゾル担体とを含む殺ウイルス性組成物。
- 請求項13若しくは請求項15に記載の殺ウイルス性組成物、若しくは請求項1から請求項12のいずれか1項に記載の殺ウイルス性ナノ粒子を使用する工程を備える消毒及び/又は殺菌の方法。
- 請求項13若しくは請求項15に記載の殺ウイルス性組成物、又は1種以上の請求項1から請求項12のいずれか1項に記載の殺ウイルス性ナノ粒子と、前記殺ウイルス性組成物又は前記殺ウイルス性ナノ粒子を付与又は分注するための手段とを含む装置。
- 殺菌のため、及び/若しくは消毒のための、請求項1から請求項12のいずれか1項に記載の殺ウイルス性ナノ粒子又は請求項13若しくは請求項15に記載の殺ウイルス性組成物の使用。
