Abstract
Description
a.前述の請求項のいずれかに記載の装置を用意する;
b.細胞、および足場生体材料を含む液体ヒドロゲルを、前記装置の少なくとも1つのトラフに加える;
c.前記足場生体材料を架橋する;
d.移行中間体に分化培地を適用する;
e.前記分化培地中で移行中間体を培養して、少なくとも1つの周方向の溝内のリングに含まれる組織を形成する。
図1を参照すると、本開示の一実施形態による、細胞から組織を製造する装置100の斜視図が示されている。ここに示されるように、装置100は、溝104のような少なくとも1つの周方向の溝を有する細長い本体102を有する。装置100はまた、トラフ106(図2によりよく示されている)のような少なくとも1つのトラフを有する。細長い本体102は、緊密にフィットする関係によって、少なくとも1つのトラフ、すなわちトラフ106を貫通して中央に延びるように動作可能である。さらに示されるように、トラフ106は閉じた経路状に延びる。例えば、示されるように、閉じた経路は円形であり、それにより、細長い本体102を囲む少なくとも1つの円形のトラフ(トラフ106など)を提供する。
Claims (26)
- 細胞から組織を製造するための装置であって、
少なくとも1つの周方向の溝を有し、緊密にフィットする関係によって、少なくとも1つのトラフを貫通して中央に延びるように動作可能な細長い本体を有し、
前記少なくとも1つのトラフは閉じた経路状に延在し、
前記周方向の溝の少なくとも1つはトラフの内側縁に向かって開いていることを特徴とする装置。 - 前記閉じた経路は円形であり、これにより前記細長い本体を囲む少なくとも1つの円形のトラフが形成される請求項1に記載の装置。
- 前記細長い本体は、緊密にフィットする関係によって、少なくとも1つのトラフを貫通して中央に延び、前記周方向の溝の少なくとも1つがトラフの内側縁に向かって開いている請求項1または2に記載の装置。
- 少なくとも1つのトラフが、トラフの低部領域から前記内側縁まで延びる傾斜壁と、前記低部領域からトラフの外側縁まで延びる外壁とを有する請求項1〜3のいずれか一項に記載の装置。
- 少なくとも1つの細長い本体を有し、別個の細長い本体がそれぞれ別個の少なくとも1つのトラフを貫通して延びている請求項1〜4のいずれか一項に記載の装置。
- トラフがその外壁の外側で端に沿って横方向に隣接するトラフに結合され、これにより別個の細長い本体が前記トラフおよび前記横方向に隣接するトラフを貫通して延びている請求項5に記載の装置。
- 端に沿って結合された複数のトラフが一体構成要素として形成され、前記一体構成要素が上面および下面を有する請求項6に記載の装置。
- 前記一体構成要素が前記上面と前記下面の間を連通する少なくとも1つの貫通穴を有し、前記貫通穴がトラフの外壁の外側に位置する請求項7に記載の装置。
- 前記少なくとも1つのトラフが積み重ねられている請求項1〜8のいずれか一項に記載の装置。
- 前記少なくとも1つのトラフは垂直方向に延びるスペーサーを備え、隣接して積み重ねられたトラフ間の距離が確保されている請求項9に記載の装置。
- 端に沿って結合されたトラフの一体構成要素を少なくとも一つ有し、
一体構成要素のトラフの各上面を、その上側に隣接する一体構成要素のトラフの対応する下面に結合することにより、前記一体構成要素が垂直方向に結合され、大きな一体構成要素を形成している請求項7または8に記載の装置。 - 前記細長い本体は、前記少なくとも1つの周方向の溝の深さを少なくとも有する長手方向の溝を有する請求項1〜11のいずれか一項に記載の装置。
- 前記周方向の溝のそれぞれがトラフの内側縁に向かって開いている請求項1〜12のいずれか一項に記載の装置。
- 前記細長い本体と前記少なくとも1つのトラフとの間の緊密にフィットする関係が、スライド式の係合を含む請求項1〜13のいずれか一項に記載の装置。
- 前記細長い本体が可撓性材料から構成されている請求項1〜14のいずれか一項に記載の装置。
- 前記細長い本体が柱状の形態である請求項1〜15のいずれか一項に記載の装置。
- 細胞から組織に移行する移行中間体を介して、細胞から組織を製造する方法であって、下記の工程を有することを特徴とする方法。
a.請求項1〜16のいずれか一項に記載の装置を用意する;
b.細胞、および足場生体材料を含む液体ヒドロゲルを、前記装置の少なくとも1つのトラフに加える;
c.前記足場生体材料を架橋する;
d.移行中間体に分化培地を適用する;
e.前記分化培地中で移行中間体を培養して、少なくとも1つの周方向の溝内のリングに含まれる組織を形成する。 - 前記工程eの組織が筋線維の形態である請求項17に記載の方法。
- 前記細胞が、間葉系細胞、筋芽細胞および筋細胞よりなる群から選ばれる少なくとも1つである請求項17または18に記載の方法。
- 前記移行中間体が、請求項17の工程cの後に、少なくとも1つのトラフから少なくとも1つの周方向の溝に移動する請求項17〜19のいずれか一項に記載の方法。
- 前記細胞が、前記少なくとも1つのトラフに加えられる前に前記ヒドロゲルに加えられる請求項17〜20のいずれか一項に記載の方法。
- 前記分化培地が、前記培養期間の少なくとも一部の後に活性化され、前記活性化が、i)酸素化、ii)少なくとも1つの栄養素の添加、iii)粒子状物質の除去、およびiv)前記分化培地の新しい分化培地との交換のうちの少なくとも1つを含む請求項17〜21のいずれか一項に記載の方法。
- さらに下記の工程を有する請求項17〜22のいずれか一項に記載の方法。
f.それぞれの少なくとも1つのトラフから前記細長い本体を除去することによって少なくとも1つの細長い本体を収集し、前記細長い本体が、少なくとも1つの周方向の溝内に生体材料のリングを有する。 - さらに下記の工程を有する請求項17〜23のいずれか一項に記載の方法。
g.組織のリングを切断して、前記細長い本体から取り外す。 - 前記切断が前記細長い本体の長手方向の溝に沿って行われる請求項24に記載の方法。
- 前記装置が、閉鎖端と、前記閉鎖端の近くにサイドポートとを有する少なくとも1つのパイプをさらに有し、
前記パイプは、前記サイドポートがトラフと流体連通可能に位置合わせされるように、a)前記少なくとも1つのトラフの外壁の外側を、および/または、b)前記少なくとも1つのトラフを貫通して中央に、前記装置を貫通して移動可能に延びるように動作可能であり、
前記方法が、細胞と、足場生体材料を含む液体ヒドロゲルを、前記パイプおよび前記サイドポートを介して前記装置の少なくとも1つのトラフに加えることを含む請求項17〜25のいずれか一項に記載の方法。
