Abstract
Description

免疫系の刺激

キメラ抗原受容体T細胞(CAR−T)療法
実施例は、本発明の組成物の投与が、TLR2の活性化の増大をもたらし得ることも示す。TLR2刺激はCAR−T療法の効力を増強する[41]。したがって、本発明の組成物は、細胞療法、特にCAR−T細胞療法において有用であり得る。一実施形態において、本発明の組成物は、細胞療法で使用するためのものである。一実施形態において、本発明の組成物は、CAR−T細胞療法で使用するためのものである。一実施形態において、本発明の組成物は、慢性リンパ球性白血病の処置に使用するためのものである。好ましくは、本発明は、そのような使用のいずれかのための、アクセッション番号42380でNCIMBに寄託された菌株またはその派生体もしくは生物型を含む組成物を提供する。
間葉系幹細胞(MSC)療法には免疫刺激特性があることが報告されている。MSCをLPSで処理すると、炎症性サイトカインのIL−8が上方制御され、B細胞増殖の増大がもたらされる[43]。したがって、本発明の組成物がB細胞増殖の発現を増大させることが示されたため、本組成物はMSC細胞療法との組み合わせにおいて有用であり得る。
幹細胞移植療法において未分化幹細胞を使用する代わりに、移植前に幹細胞をある程度まで分化させることが有益であり得ることが報告されている。例えば、Hengら[44]は、幹細胞の心筋形成分化が、より高い生着効率、筋細胞の再生の増強、及び心臓機能の回復の向上を伴うことにより、有益であり得ることを報告した。また、研究により、特定の共生細菌株によるGIコロニー形成が、同種造血細胞移植後の生存率を改善できることが示されている[45]。本発明の組成物の投与により細胞が刺激されたため、本発明の組成物は、幹細胞移植療法における幹細胞分化に有用であり得る。

実施例により、B.breve、特に本発明のB.breve菌株が、強力な抗菌活性を有することが示されている。したがって、ある特定の実施形態において、本発明の組成物は、細菌感染症の処置または予防に使用するためのものである。
好ましくは、本発明の組成物は、本発明の細菌株の腸への送達及び/または腸における部分的もしくは全体的なコロニー形成を可能にするために、消化管に投与するように製剤化される。いくつかの実施形態では、「腸の完全なコロニー形成」という用語は、細菌が腸のすべての部分(すなわち、小腸、大腸及び直腸)にコロニー形成したことを意味する。本発明のさらなる実施形態では、「完全コロニー形成」または「部分コロニー形成」という用語はそれぞれ、細菌が永続的または一時的に腸内に保持されることを意味する。一般的には本発明の組成物を経口投与するが、直腸内、鼻腔内、または頬側もしくは舌下経路を介して投与してもよい。
本発明の組成物は、概して、細菌を含む。本発明の好ましい実施形態では、組成物は凍結乾燥形態で製剤化される。例えば、本発明の組成物は、本発明の細菌株を含む顆粒剤またはゼラチンカプセル、例えば硬ゼラチンカプセルを含み得る。
本発明に使用するための細菌株は、例えば、参考文献[58〜60]に詳述されている標準的な微生物学的技術を使用して培養することができる。
本発明者らは、本発明の細菌株が免疫活性の低減に関連する疾患または状態の処置または予防に有用であることを明らかにした。これは、本発明の細菌株が宿主免疫系に対して有する効果の結果である可能性が高い。したがって、本発明の組成物は、ワクチン組成物として投与された場合に、疾患または状態を予防するのに有用であり得る。そのようなある特定の実施形態において、本発明の細菌株は、死滅、不活化または弱毒化され得る。そのような特定の実施形態において、組成物は、ワクチンアジュバントを含み得る。ある特定の実施形態において、組成物は、注射によって、例えば皮下注射によって投与されるためのものである。

要旨
この試験の目的は、MRx0004のインビトロの免疫調節特性を特性評価することであった。さらに、ゲノミクス、トランスクリプトミクス及びプロテオミクスの組み合わせを使用して、MRx0004に対する宿主応答の媒介を担う可能性のある潜在的な主要エフェクターを同定した。
細菌株、プラスミド及びプライマー
本試験において菌株を作製するために使用したすべての細菌株及びプラスミド及びプライマーを表7に示す。特に明記されていない限り、B.breve菌株を、嫌気性ワークステーション(Don Whitley Scientific,Shipley,UK)内で37℃で、酵母抽出物−カゼイン脂肪酸加水分解物(YCFA)ブロス(E&O Labs,Bonnybridge,UK)中で通常通り培養した。E.coli菌株を、Luria Bertani(LB)ブロス[71]中で37℃で攪拌しながら通常通り培養した。必要に応じて、増殖培地に、テトラサイクリン(10μg/ml)、クロラムフェニコール(E.coliの場合は10μg/ml、もしくはB.breveの場合は3μg/ml)、エリスロマイシン(E.coliの場合は100μg/ml、もしくはB.breveの場合は1μg/ml)、スペクチノマイシン(100〜300μg/ml)、またはカナマイシン(50μg/ml)を補充した(すべての抗生物質はSigma−Aldrich,Gillingham,UKからである)。pORI19またはpWSK29を含有する組換えE.coli細胞を、40μg/mlのX−gal(5−ブロモ−4−クロロ−3−インドリル−β−D−ガラクトピラノシド)及び0.1MのIPTG(イソプロピル−β−D−ガラクトピラノシド)(いずれもSigma−Aldrichより供給)を補充したLB寒天培地上で選択した。
HT29−MTX−E12細胞(Public Health England,Salisbury,UK)を、10%(v/v)のウシ胎児血清(FBS)、4mMのL−グルタミン、1倍の非必須アミノ酸溶液及び1倍の抗生物質抗真菌溶液を補充した高グルコース改変ダルベッコ最小イーグル培地(DMEM)中で、通常通り培養した。細胞をアッセイ容器に播種して9日間培養後、ハンクス平衡塩類溶液で2回洗浄し、処理開始前に共培養培地(4mMのL−グルタミン、1倍の非必須アミノ酸溶液、5μg/mlのアポトランスフェリン及び200ng/mlの亜セレン酸ナトリウムを補充したDMEM)に入れた。
共培養実験では、対数期に達するまで細菌を培養した。生菌細胞及び上清を遠心分離により分離後、生菌(LVと表記)をPBS(Sigma−Aldrich)で1回洗浄し、下流で使用するために適切な細胞培養培地に再懸濁させた。上清(SNと表記)を0.22μmフィルターに通し、共培養培地で適切に希釈した。熱不活化細菌(HKと表記)を、80°Cで30分間インキュベートした後、PBSで洗浄し、適切な細胞培養培地に再懸濁させることにより調製した。生存数をプレーティングにより確認した。
HEK−Blue(商標)−hTLR2細胞及びTHP1−Blue(商標)NF−κB細胞を90%の密度まで増殖させ、PBSで1回洗浄し、抗生物質を含まない培地にそれぞれ280,000細胞/ml及び500,000細胞/mlの密度で再懸濁させた。細菌処理物(生菌、加熱死及び上清)を100:1の感染多重度(MOI)で細胞に添加した。アッセイの陽性対照であるPamCS3K4(Invivogen)及び加熱死L.monocytogenes(HKLM)(Invivogen)をそれぞれ、10ng/mlの濃度及び200:1のMOIで使用した。陰性対照、培地及びビヒクルを、各処理に対し同等のものを提供するように調製した。次に、細胞を37℃、5%CO2で22時間インキュベートした。共培養からの培地をQUANTI−Blue(商標)(Invivogen)で10倍に希釈し、1時間(NFκB)または2時間(TLR2)インキュベートし、655nmでの光学濃度を記録した。
HT29−MTX細胞を、前述のように、直径10cmのTranswell(登録商標)(Thermo Fisher Scientific,Waltham,MA,USA)の上部チャンバーで培養した。細菌を後期対数期まで培養し、前述のように洗浄して再懸濁させた。細菌を100:1のMOIで細胞に添加し、共培養物を37℃、嫌気性条件で3時間インキュベートした。トランスウェルの上部チャンバーから細菌を含む培地を採取し、5000×gで5〜10分間遠心分離して、下流の用途のために細菌細胞を採取した。
RNA単離のために、後期対数期及び定常増殖期のインビトロ培養物から、ならびにHT29−MTXとのラージスケールの共培養後の培養物から、細菌を採取した。製造業者の説明書(QIAGEN,Hilden,Germany)に従って、RNAProtect Bacteria Reagentを使用して細菌を採取及び保管した。細菌細胞を、リゾチーム(15mg/ml)(Sigma−Aldrich)及びプロテイナーゼK(6mAU)(QIAGEN)で37℃で30分間インキュベートすることにより溶解させ、続いて、FastPrep 24装置(6m/sで20秒間のサイクルを2回)及びLysing Matrix B(いずれもMP Biomedicals,Santa Ana,CA,USAより)を使用してホモジナイズした。RNeasy Mini Kit(QIAGEN)を使用して全RNAを単離し、オンカラム消化(QIAGEN)でRNase−Free DNaseを使用して、ゲノムDNAを除去した(いずれも製造業者の説明書に従った)。Superscript IVキット(Thermo Fisher Scientific)を製造業者の説明書に従って使用して、cDNAを合成した。プライマーをPrimer3Plusソフトウェア[72]を使用して設計した。qPCR反応を製造業者の推奨に従ってPower SYBR(商標)Green PCR Master Mix(Thermo Fisher Scientific)で設定し、アッセイを7500 Fast Real−time PCR System(Thermo Fisher Scientific)にて、次のサイクル:95℃で10分間の後、95℃で15秒間、60℃で1分間を40サイクル使用して実施した。データ分析をデルタデルタCt分析を使用して実施し、試験遺伝子の発現をハウスキーパーgroELに対して正規化した。
細菌細胞を、後期対数増殖期における遠心分離によって、またはHT29−MTXとの接触後に適宜回収した(詳細については上記の共培養のセクションを参照されたい)。次に、細胞を洗浄し、50mMのTEAB緩衝液pH8.5(Sigma−Aldrich)に1/20の希釈度で再懸濁させた。細胞を、1mMのDTT(Sigma−Aldrich)を補充した50mMのTEAB緩衝液中で、シーケンスグレードの修飾トリプシン(Promega,Madison,WI,USA)と37°Cで30分間インキュベートすることによって、切り取られたタンパク質の画分を生成した。各試料について、トリプシンを含まないチューブを、シェディングされたタンパク質(シェディングされたタンパク質の画分)の対照として並行してインキュベートした。切り取られたタンパク質の画分及びシェディングされたタンパク質の画分を、4000×g、4°Cで15分間遠心分離して回収し、Millex−GV 0.22μm低タンパク質結合膜(Millipore)を通してシリンジ濾過した。Pierce(商標)BCA Protein Assay Kitを製造業者の説明書(Thermo Fisher Scientific)に従って使用して総タンパク質濃度を測定し、SDS−PAGE(Bio−Rad,Hercules,CA,USA)によって試料の性質を評価した。YCFA寒天培地にプレーティングすることにより、トリプシン処理の前後に生存細胞数の測定を実施した。次に、各アッセイについて、3回の生物学的反復試験からの試料をナノLC−MS/MSで分析した。
簡潔に述べると、培養上清を0.5mlまで濃縮して超純水で洗浄し、ReadyPrep 2−D Cleanup Kit(Bio−Rad)を使用してタンパク質を沈降させ、50mMの炭酸水素アンモニウム100μlに再懸濁させた。次に、試料をブタトリプシン(Promega)と37℃で16時間インキュベートして、得られた上清を真空遠心分離で乾燥させ、0.1%のトリフルオロ酢酸に溶解させた。ペプチドをμ−C18 ZipTip(Merck,Keniloworth,NJ,USA)を使用してさらに脱塩し、96ウェルマイクロタイタープレートに溶出させて、真空遠心分離で乾燥させ、LC−MSローディング溶媒(2%アセトニトリル、0.1%ギ酸)10μlに溶解させた。ペプチドを、15cmのPepMapカラム、60分でのLC−MSの取得方法及び5μlの注入量を使用して、ナノLC−MS/MS(Q Exactiveハイブリッド四重極Orbitrap MSシステム)(Thermo Fisher Scientific)により分離して同定した。切り取られたタンパク質の画分及びシェディングされたタンパク質の画分については、50mMの炭酸水素アンモニウム70μlを、30μlの試料に直接添加した。次に、試料をブタトリプシン(Promega)と37℃で一晩インキュベートして、得られた上清を−70℃で凍結し、真空遠心分離で乾燥させ、20μLのLC−MSローディング溶媒に溶解させた。ペプチドを、25cmのPepMapカラム、60分でのLC−MSの取得方法及び2μlの注入量を使用して、ナノLC−MS/MS(Q Exactiveハイブリッド四重極Orbitrap MSシステム、Thermo Scientific)により分離して同定した。Proteome Discoverer(Thermo Fisher Scientific)を使用してデータ分析を実施した。Mascot Serverを、次のパラメータを用いて検索エンジンとして使用した:酵素=トリプシン、最大混合切断部位=2、前駆体の質量許容差=10ppm、動的修飾=酸化(M)、静的修飾=カルバミドメチル(C)。同定したペプチドを、B.breve MRx0004の配列決定したゲノム(2,047配列)に基づいて構築した菌株特異的タンパク質の配列データベースと照合した。3回の生物学的反復試験のすべてにおいて少なくとも5つのペプチドが同定された場合、タンパク質の同定は有効であるとみなされた。
一次グリコシルトランスフェラーゼをコードする遺伝子pGTF及びその推定プロモーターを包含するDNA断片を、Q5 High−Fidelity Polymerase(New England BioLabs,Herefordshire,United Kingdom)ならびにプライマー対:pGTFcompF及びpGTFcompRを使用した、B.breve MRx0004の染色体DNAからのPCR増幅により作製した。得られた断片をHinDIII及びXbaI(いずれもNew England Biolabs,Ipswich,MA,USAより)で消化して、同様に消化したpBC1.2にライゲーションした。ライゲーション混合物を電気的形質転換によりE.coli EC101に導入し、次に、形質転換体をCm耐性に基づき選択した。いくつかのCm耐性形質転換体のプラスミド含有量を、制限分析によってスクリーニングした。いくつかの組換えプラスミドにおけるクローニングされたインサートの完全性を、メチル化を促進するためにそれらをE.coli EC101 pWSK29−MRX−M+Sに導入する前に、配列決定することによって確認した。メチル化pBC1.2またはpBC1.2−pGTFを、エレクトロポレーションによってMRx0004negに形質転換し、Tet及びCmを補充した強化クロストリジウム寒天培地(RCA、Thermo Fisher Scientific)上で選択した。コロニーPCR、プラスミドDNAの制限分析を使用して形質転換体のプラスミド含有量について検査し、配列決定によって確認した。得られた菌株を、B.breve MRx0004−EPS−−pBC1.2及びMRx0004−EPS−−pBC1.2−pGTFと命名した。
生菌(前述の通りに調製)を100:1のMOIで、37℃、5%CO2で3時間、24ウェルプレート中でHT29−MTX細胞と共インキュベートした。次に、ヒト組換えTNFα(PeproTech,Rocky Hill,NJ,USA)を10ng/mlで細胞に添加し、その後、共培養物をさらに24時間インキュベートし、続いて上清を回収して、4℃、12000×gで3分間遠心分離して細胞デブリを除去した。製造業者の推奨に従ってHuman IL−8(CXCL8)Standard ABTS ELISA Development Kit(PeproTech)を使用して、上清中のIL−8レベルを分析した。
健康なヒトの凍結末梢血単核細胞(PBMC)をSTEMCELL Technologies(Cambridge,UK)から購入した。細胞を解凍して、完全増殖培地(10%のFBS、2mMのL.グルタミン及び100U/mlのペニシリン、100μg/mlのストレプトマイシンを含むRPMI 1640)中に37℃、5%CO2で一晩静置した(すべての試薬はSigma−Aldrichからである)。細菌処理物を前述の通りに調製した。共インキュベーションでは、細胞を48ウェルプレートに750,000細胞/ウェルの密度で播種し、10:1のMOIで熱不活化細菌、及び細菌上清と、ならびに対照として適切なビヒクル及び5ug/mlのPHA(Sigma−Aldrich)と、共インキュベートした。共培養物を37℃、5%CO2で72時間インキュベートした後、細胞を採取して、4℃、10000×gで3分間遠心分離した。無細胞上清を回収し、サイトカイン分析のために−80℃で保管した。細胞ペレットを1回洗浄し、次いで、氷上でPBSに再懸濁させた。
1群当たり1.5×106のPBMCとなるように処理ウェルをプールして、150ulのPBSに再懸濁させ、染色の準備がなされている96V字底プレートに移した。まず、細胞をViobility 405/520 Fixable Dye(Miltenyi Biotec Ltd. Bergisch Gladbach,Germany)で染色して、暗所で室温において10分間、生細胞と死細胞とを判別した。次に、細胞を、CD3、CD4、CD8、CD25、CD127及びCD19に対する抗体の混合物で染色して細胞の表現型を決定し(Miltenyi REA抗体)、室温でさらに10分間インキュベートした。次に、細胞を洗浄してPBSに再懸濁し、フローサイトメトリー分析によって直ちに分析した。最初の実験の際に、ゲートの設定を支援するためにすべての抗体に対してアイソタイプを使用し、すべての実験にわたりFMO対照を含めた。すべての実験は、BD FACS Aria IIを使用して実施し、FACSDivaソフトウェア(BD Biosciences,Reading,UK)を使用して、「生(Live)」ゲート内に100,000細胞で設定した取得用のストップゲートを用いた。Flowjoバージョン10.4.2ソフトウェア(FlowJo LLC,Oregon,USA)を使用して分析を実施し、分析は、生死判定用色素で特定した生細胞に基づくものであった。
カスタムProcartaPlexマルチプレックスイムノアッセイ(Thermo Fischer Scientific)を製造業者の推奨に従って使用して、PBMC共培養物の無細胞上清中のサイトカインの定量化を実施した。簡潔に述べると、MAGPIX(登録商標)MILLIPLEX(登録商標)システム(Merck)をxPONENTソフトウェア(Luminex,Austin,TX,USA)とともに使用して、50μlの上清を処理した。5パラメータロジスティック曲線及びバックグラウンド除去を用いた、MILLIPLEX(登録商標)analystソフトウェア(Merck)を使用してデータを分析し、平均蛍光強度(MFI)をpg/ml値に変換した。
統計分析を、Windows用のGraphPad Prismバージョン7.00(GraphPad Software、La Jolla CA USA)を使用して実施した。一元配置分散分析及びテューキーの多重比較検定を使用してデータを分析した。ベン図をInteractivenn[73]を使用して作成した。
MRx0004はNFκBレポーター細胞及びTLR2レポーター細胞を刺激する
炎症誘発性転写因子NFκBの活性化に対するMRx0004の影響を、自然免疫の調節におけるその役割が不可欠であることから、THP−1−NFκBレポーター細胞株を用いて調査した。このアッセイでは、加熱死させたListeria monocytogenes(HKLM)(InvivoGen)を陽性対照として使用した。この菌株の有効画分(複数可)を同定するために、THP−1−NFκB細胞を生菌(MRx0004LV)、細菌培養上清(MRx0004SN)及び熱不活化細菌(MRx0004HK)の処理物と共インキュベートした。3種の細菌処理物はすべて、未処理細胞及び細菌増殖培地(YCFA)の陰性対照と比較してNFκBを有意に活性化させた(すべての比較でp<0.0001)(図7A)。MRx0004LVが最も有効な処理物であり、MRx0004SN及びMRx0004HKよりも有意に刺激した(両方の比較についてp<0.0001)。次に、MRx0004HKは、MRx0004SNよりも有意に活性であった(p=0.006)。
MRx0004における宿主応答の潜在的なエフェクターを同定するために、3つのアプローチを採用した。標的化した転写アッセイを使用して、ビフィズス菌−宿主相互作用の既知のエフェクターとしての同一性が予測される10個のMRx0004遺伝子を分析した(データは示さず)。これらには、推定アドヘシン及びムーンライティングタンパク質(oppA、エノラーゼ、トランスアルドラーゼ、tadA、eftU、プルラナーゼ([74]に広範に概説されている))をコードする遺伝子、ならびにコロニー形成及び免疫調節において役割を果たすと推定されている遺伝子(MRx0004のEPS遺伝子座[75]、luxS[76]及びセルピン[77]の一次グリコシルトランスフェラーゼ(pGTF))ならびに治療効果を有すると推定されている遺伝子(pks[78])が含まれた。これらの遺伝子の発現を、液体培養液中で後期対数期及び定常期まで増殖させたMRx0004から単離したRNA、ならびにラージスケールのトランスウェルで培養したHT29−MTX細胞と3時間接触させた後のMRx0004から単離したRNAにおいて分析した。qPCR分析(図8)により、eftU、エノラーゼ及びpGTFは、後期対数期において定常期よりも著しく上方制御され、一方で、oppA、プルラナーゼ、セルピン及びtadAの発現は、定常期において後期対数期よりも著しく上昇したことが示された。6つの遺伝子(eftU、エノラーゼ、pGTF、oppA、セルピン及びトランスアルドラーゼ)は、後期対数期と比較して、腸管上皮細胞(IEC)に応答して著しく上方制御された。この分析から、MRx0004の遺伝子発現がIECとの接触によって変化したことが明らかであり、MRx0004−宿主相互作用における上方制御された遺伝子の潜在的な役割が推測された。このqPCR分析で著しく上方制御された遺伝子の大部分は、IECへの接着における役割を有することが予測されており、これは、このことがMRx0004の重要な機能特性である可能性があることを示唆している。
MRx0004に対する宿主反応の特性を明らかにするために、さらなる実験を実施した。
適応免疫系に対するMRx0004の影響を調査するために、健康ヒトドナーに由来する末梢血単核細胞(PBMC)を使用して、細胞集団及びサイトカイン分泌プロファイルを特性評価した。このアッセイでは、PHAを陽性対照として使用した(データは示さず)。MRx0004及びその派生菌株に由来する熱不活化細菌細胞及び無細胞培養上清と、PBMCを72時間共インキュベートした。T細胞(CD3+CD4+及びCD3+CD8+)、Treg(CD3+CD4+CD25+CD127−)及びB細胞(CD3−CD19+)の表面マーカーの発現を、フローサイトメトリーによって(活性化マーカーCD25とともに)分析した(ゲーティング戦略については図18を参照されたい)。72時間のインキュベーション期間中に生菌が増殖し、栄養素を求めてヒト細胞を排除する可能性があることから、生菌ではなく熱不活化細菌をこのモデルにおける処理物として使用した。試験したすべての菌株からの細菌上清に応答した細胞表面マーカー及びサイトカインの両方の発現は、ビヒクル(YCFA)に応答して観察されたものと比較して大幅に異なってはいなかった(データは示さず)。EPSvec及びEPScompのデータを、図20及び図21に示す。
要旨
この試験の目的は、MRx0004の免疫刺激及び治療特性におけるMRx0004菌体外多糖(EPS)の役割を明らかにすることであった。
実験を、以下に記載する追加の手順を含めて、実施例1に記載の通りに実施した。
HEK−Blue(商標)−hTLR2細胞(InvivoGen,San Diego,CA,USA)を、10%(v/v)のFBS、4mMのL−グルタミン、4.5mg/mlのグルコース、100U/mlのペニシリン、100μg/mlのストレプトマイシン、100μg/mlのNormocin(商標)(Invivogen)、30μg/mlのブラストサイジン(blastocydin)及び100μg/mlのゼオシンを補充したDMEM中で、密度90%まで増殖させた。THP1−Blue(商標)NF−κB細胞(InvivoGen)を、10%(v/v)の熱不活化FBS、2mMのL−グルタミン、100U/mlのペニシリン、100μg/mlのストレプトマイシン、25mMのHEPES、100μg/mlのNormocin(商標)、10μg/mlのブラストサイジンを補充したRPMI 1640(cRPMI)中で増殖させた。細胞株を、37℃、5%CO2で培養した。特に明記されていない限り、すべての試薬はSigma Aldrichから供給された。
GenBankデータベースで利用可能なBifidobacterium菌株のゲノムを、EPSクラスターのインシリコ分析及びBifidobacterium菌株に由来する推定上の菌体外多糖の遺伝子クラスターの物理的地図のために使用した。
わずかな変更を加えた製造業者のプロトコールに従って、RNAprotect(Qiagen)及びRNeasy Miniキット(Qiagen)を使用して、全RNAをMRx0004菌株の後期対数期培養物から抽出した。機械的細胞溶解を、Lysing MatrixB及びMP Fast−Prep−24組織及び細胞ホモジナイザー(MP Biomedicals,Santa Ana,CA,USA)を使用して、振動を6m/sに設定して実施した。細胞を、20秒間のサイクル2回で破壊し、サイクルの合間には氷上で1分間静置させた。RNAの品質を、Agilent RNA Screentape(Agilent Technologies,Santa Clara,CA,USA)を用いて、Tapestation(Agilent Technologies)で確認した。RNAの分解がないことを確認し、すべての試料の最小RNA完全性数(RNA Integrity Number)は9以上であった。MICROBExpressキット(Thermo Fisher Scientific)を使用してrRNA種を除去した。16S rRNA種及び23S rRNA種が存在しないことを、Agilent RNA RNA Screentape(Agilent Technologies)を用いてAgilent Bioanalyzerで評価し、確認した。rRNAを除去したRNA試料を、ストランド特異的ライブラリー調製のためにGATC Biotechに送り、イルミナシーケンシングで配列決定し150bpのシングルエンドリードを生成した。RNA−Seqライブラリー当たり、平均22,3878,08(後期対数期試料)及び18627178.6(定常期試料)の生の読み取りデータを、それぞれ合計10.07Gbp超及び8.38Gbp超得た。生の読み取りデータをTrimmomatic(1)を使用してトリミングし、Bowtie(2)を使用して、MRx0004ゲノムに対して性質をフィルタリングした(98.36%の後期対数期試料及び98.26%の定常期試料の読み取りデータがQCを通過し、マッピングしたクリーンな読み取りデータの99.11%(LL)及び98.72%(SP)に整列した)。各増殖期の反復試験試料の発現レベルを、XX及びDeSeq2 v X(Love et al,2014)を使用してMRx0004のEPS遺伝子座の各遺伝子について算出し、続いてGeneious R11(Biomatters,Auckland,New Zealand)を使用して可視化した。2つの増殖期の間の差次的発現が表される。2つの試料間の正規化された値の比の、底を2とする対数であり、一方の試料の発現がないかまたは非常に低い場合、log2の比は+/−1,000,000が上限となる。
細菌を固定液(0.1Mリン酸ナトリウム緩衝液中の0.5Mスクロース、2%パラホルムアルデヒド及び0.16%グルタルアルデヒド)で1:5に希釈し、室温で2時間固定した。その後、フォルムバールカーボンでコーティングされた銅グリッドを、B.breve懸濁液の100μlの液滴に1時間浮遊させ、PBS中の0.02Mのグリシンで3回洗浄した。細胞を1.0%のモリブデン酸アンモニウムで陰性に染色した。JEM−1400透過型電子顕微鏡(JEOL Ltd.,Tokyo,Japan)を使用して、グリッドを検査し、顕微鏡写真を可視化した。
生菌(前述の通りに調製し共培養培地に再懸濁させたもの)を、24ウェルプレート中で100:1のMOIでHT29−MTX細胞にアプライし、37℃、嫌気性条件で3時間共インキュベートした。細胞をPBSで2回洗浄して未結合の細菌を除去し、0.1%(v/v)のTriton X−100(Sigma−Aldrich)で溶解させた。溶解物を播種し、回収したコロニー形成単位(CFU)の数を使用して、接着のパーセンテージを決定した。
MRx0004のeps遺伝子座は、B.breveの他の菌株とは遺伝的に異なり、後期対数増殖の際に高度に発現する。
菌株MRx0004のゲノム配列決定によって、菌株MRx0004が、27個の遺伝子をコードすることが判明した28KbのEPS遺伝子座を保有することが示され、これは、B.breveにおけるEPSの生合成に必要であると予想される機能の完全な補完を表している。この領域には、プライミンググリコシルトランスフェラーゼ、4つの追加のグリコシルトランスフェラーゼ、膜貫通タンパク質の下流にコードされるチアミンピロリン酸結合タンパク質、フリッパーゼ及び鎖長決定因子が含まれる(図9)。B.breve EPS遺伝子座の大部分(菌株MRx0004のものを含む)は、仮想タンパク質に隣接しており(MRx0004領域を31.5Kbに拡張している)、これは、図9に表すB.breveのEPS遺伝子座の表示からは除外されている。図9に示す大部分(16/19)の菌株は乳児単離株であり、それらのゲノムは公共データベースにおいて大きな比率を占めている。50Kbを超えるB.breveのEPS領域は、EPS陽性の表現型を生成するために必要なすべての機能をコードする完全な遺伝子座を表すと考えられている[89]。対照的に、これらの領域が30Kb未満の場合、これらは不完全なまたは残余の遺伝子座を表すと考えられる[89]。
MRX004 EPSの宿主−微生物相互作用及び免疫調節における役割を調査し、上記の通り、挿入変異導入を通じてEPS遺伝子座のpGTF遺伝子が不活化されている菌株(EPSneg)を構築した(図16A)。この菌株を、[88]に記載されている方法論を用いて構築したが、II型制限修飾(RM)系を操作するのではなく、MRx0004のI型RM系に由来するメチラーゼ及び特異性サブユニットを発現させ、形質転換前にプラスミドDNAをメチル化するために使用した。補完されたEPSneg菌株(EPScomp)及びEPSneg空ベクター菌株(EPSvec)もまた、対照として作製した。EPSneg及びEPSvecは、MRx0004と比較して、自己凝集性の表現型の増加を示した(図16B)。EPScompは、EPSneg及びEPSvecよりも凝集性が低かったが、その自己凝集はMRx0004と比較して増加しているように思われ、このことは、この菌株が野生型EPSの表現型に完全に復帰していないことを示唆する。
EPSnegのIECに対する接着性の向上は、EPSの除去が、表面結合タンパク質の露出の増加、または「非保護」をもたらし得ることを示唆する。MRx0004及びEPSnegの表面結合タンパク質の組成物を、IECと接触させた後に分析した。HT29−MTX細胞と3時間接触させた後、前述のように細菌細胞をトリプシンで切り取り、得られた切り取られたタンパク質の画分及びシェディングされたタンパク質の画分をLC−MS/MSで分析した。IECとの接触後のMRx0004細胞の切り取りにより、55個の切り取られたタンパク質(そのうち34個は表面に固定されていると予測された)及び24個のシェディングされたタンパク質を得た(図11A)。IECへの接触後のEPSneg細胞から切り取られたものには、MRx0004のものよりもはるかに多くのタンパク質が含まれており、切り取られたタンパク質の画分中に101個のタンパク質、シェディングされたタンパク質の画分中に45個のタンパク質であった(図11B)。MRx0004のシェディングされたタンパク質の画分中で同定された3つのタンパク質(炭水化物、脂質、及びタンパク質の代謝に関与する酵素)を除いて、同定されたすべてのタンパク質は、両方の菌株の切り取られたタンパク質の画分中に存在していた。
比較ゲノム分析により、MRx0004においてEPSの合成を担う遺伝子座は、他のB.breve菌株とは遺伝的に異なることが示された。この領域で観察された遺伝的相違は、MRx0004及び近縁菌株の効力及び治療的有用性の増強に寄与し得る。
要旨
先行実施例に記載されているように、本発明者らは、B.breve、特に菌株MRX004の新しい免疫刺激効果を同定した。上記の新しいデータを踏まえて、B.breve、特に菌株MRX004を含む組成物は、免疫系を刺激して、免疫系の活性の低減に関連する疾患または免疫系の活性の増大からベネフィットを得られる疾患を処置するのに有効であると予想される。
試験物質−細菌株#MRX004、Bifidobacterium breve。
・(調製済の10mL E&Oラボボトルより)10mLのYCFAをピペットで取り、Hungateチューブに入れる
・チューブを密封し、シリンジによる投入でCO2を流して、系を排気する
・Hungateチューブをオートクレーブする
・冷却後、Hungateチューブに1mLのグリセロールストックを接種する
・37℃のインキュベータ内でチューブを約16時間静置する。
・翌日、この継代培養物1mLを取り、10mLのYCFAに接種する(再度予熱してフラッシュしたHungateチューブ、すべて二連)
・37℃のインキュベータ内でチューブを5〜6時間静置する
使用した細胞株を以下の表に詳述する。
体重及び週齢を一致させた健康な雌Balb/C(BALB/cByJ)マウスを、EMT6モデルの実験のために、CHARLES RIVER(L’Arbresles)から入手した。
抗腫瘍活性、EMT6モデル
処理スケジュール−最初の投与の開始を0日目とした。0日目に、Vivo manager(登録商標)ソフトウェア(Biosystemes,Couternon,France)を使用して、非移植マウスをマウスの個々の体重に従って9/8匹の群に無作為化した。0日目に、マウスにビヒクル(培地)または細菌株を投与した。14日目に、以下に記載の通りにすべてのマウスにEMT−6腫瘍細胞を移植した。24日目に陽性対照群のマウスに抗CTLA−4抗体処理を施した。

処理スケジュール−最初の投与の開始を0日目とした。0日目に、Vivo manager(登録商標)ソフトウェア(Biosystemes,Couternon,France)を使用して、非移植マウスをマウスの個々の体重に従って9匹の7つの群に無作為化した。0日目に、マウスにビヒクル(培地)または細菌株を投与した。14日目に、以下に記載の通りにすべてのマウスにHepa1−6腫瘍細胞を移植した。16日目に陽性対照群のマウスに抗CTLA−4抗体処理を施した。
臨床モニタリング−腫瘍の長さ及び幅をノギスで週に2回測定して、腫瘍の体積をこの式によって推定した[98]。
抗腫瘍活性、EMT6モデル
結果を図1に示す。本発明の細菌株による処理によって、両方の陰性対照と比較して腫瘍体積の明らかな減少が生じた。予想されていたように、免疫系を活性化させることが知られている陽性対照によっても腫瘍体積の減少が生じた。
結果を図2に示す。腫瘍体積が陰性対照群よりも陽性対照で処理したマウスにおいて大きかったことから、陰性対照及び陽性対照は予想通りに表示されていない。それにもかかわらず、本発明の細菌株で処理したマウスの腫瘍体積は陽性対照群と同等であり、これは、有用な治療効果及び免疫刺激効果に合致する。
結果を図3に示す。未処理陰性対照群における肝臓重量が他の群より低かったことから、未処理陰性対照は予想通りに表示されていない。しかし、ビヒクルのみで処理したマウスは、抗CTLA4抗体によって処理したマウスより肝臓が大きく、ビヒクル陰性対照群において腫瘍量がより大きいことを反映していることから、ビヒクル陰性対照群及び陽性対照群はいずれも予想通りに表示されている。本発明の細菌株による処理によって、ビヒクル陰性対照群におけるマウスと比較して、肝臓重量(したがって、腫瘍量)の明らかな減少が生じた。
Analytical Profile Index(API(登録商標))試験系は、細菌種における酵素活性に関してアッセイする小型化した生化学的試験を含有するストリップからなる。MRX004(アクセッション番号NCIMB 42380で寄託された細菌)を2つのAPI試験系を使用して特性評価した:Rapid ID 32A−この系は、特に嫌気性種用に設計されており、炭水化物、アミノ酸及び硝酸塩の代謝、ならびにアルカリホスファターゼ活性についての試験を包含する;API(登録商標)50 CH−この系は、49個の炭水化物源の発酵について試験し、嫌気性種の分析のためにAPI(登録商標)CHL Mediumと併せて利用することができる。
要旨
菌株MRX004及び他のいくつかのBifidobacterium breve菌株のヒト細胞への結合レベルを、YCFA培地中で3つの異なる時点において測定した。ヒト細胞に接着した細菌を培地中に再懸濁させ、次に、培地の光学濃度を分析した。光学濃度が高いほど細菌細胞の数が多くなり、したがって、細菌細胞のヒト細胞への結合レベルが高くなる。MRX004菌株が、Bifidobacterium breve参照菌株と比較して、ヒト細胞への接着の減少を示すことが判明した。
この実験の結果を図6に示す。
本明細書に記載の少なくとも1つの細菌株を含有する本明細書に記載の組成物を、25℃または4℃で密封容器の中で保管して、容器を相対湿度が30%、40%、50%、60%、70%、75%、80%、90%または95%である雰囲気中に置く。1か月、2か月、3か月、6か月、1年、1.5年、2年、2.5年、または3年後に、細菌株の少なくとも50%、60%、70%、80%または90%が、標準的なプロトコールによって決定したコロニー形成単位で測定した場合に残るものとする。
概要
この実験の目的は、ヒト乳児に由来するいくつかのB.breve菌株の抗菌活性の潜在能力を様々な指標菌株に対して試験すること、及びそれらがインビトロでバクテリオシンを生成するかどうかを評価することであった。
菌株のパネルを指標菌株として選択し(表5)、これには、Bifidobacterium種によって阻害されることが既に示されている近縁のグラム陽性菌、他のグラム陽性菌及びグラム陰性菌が含まれた[エラー!ブックマーク未定義]。
Research Cell Bankから(B.breve試験菌株及び参照菌株用)またはビーズストックから(指標菌株用)の菌株を、嫌気性条件下(E.coli、B.subtilis及びS.aureusについては好気性条件下)で、37℃(B.subtilisについては30℃)で16時間増殖させた。YCFAプレート(E&O Labs,UK)上に指標菌株の菌叢を作製し、乾燥させ、菌叢の表面に10μlの試験菌株培養物をスポットした。プレートを嫌気性条件下で37℃で48時間(E.coli、B.subtilis及びS.aureusについては嫌気性条件下で37℃で24時間、続いて好気性条件下で37℃で24時間)インキュベートした。各アッセイを三連で実施した(MRx0004、試験1、試験2、試験3、試験4、試験5、試験6、試験7及び試験8を用いたBacillus subtilis NCIMB8045、ならびにすべてのB.breve菌株を用いたBifidobacterium breve DSM20213、Lactobacillus plantarum NCIMB8826、Clostridium sporogenes ATCC3584及びStaphylococcus aureus NCIMB9518に対して二連で実施したことを除く)。
寒天拡散法を使用して、培養上清の潜在的な抗菌能力を試験した。簡潔に述べると、100μlの濾過した無細胞上清を、指標菌株の菌叢を(上記の通り)事前に接種したYCFA寒天培地上にスポットして、寒天培地に打ち抜いたウェルに入れた。プレートを1時間放置して拡散させ、次に、嫌気性条件下で(E.coli、B.subtilis及びS.aureusについては好気性条件で)37℃で48時間インキュベートした。3回の生物学的反復試験を実施した。
共培養
試験した大部分のB.breve菌株は、E.coli、K.pneumoniae、S.Typhimurium及びB.subtilisに対して拮抗活性を示した(表12)。B.breve DSM 20091は、B.breve DSM20213の増殖を阻害した唯一の試験菌株であった。MRx0004及び他のB.breve試験菌株は、E.coli、K.pneumoniae、S.Typhimurium及びB.subtilisに対して抗菌活性を示し(表12)、全体として、B.breve参照菌株で観察されたものよりも強く阻害した。MRx0004、試験1、試験2、試験3、試験7、試験8、試験11及び試験12は、特に強力な抗菌活性を示した。試験した条件では、S.aureus、C.sporogenes及びL.plantarumに対して、拮抗作用は検出されなかった。
無細胞上清の抗菌活性を、同じパネルの指標菌株に対して試験した。いずれの指標菌株に対しても、試験したすべてのB.breve菌株で阻害は観察されなかった(データは示さず、n=3)。これは、共培養アッセイで観察された阻害が、抗菌分子の分泌によるものではなかったことを示唆している。
実施例4に記載されているAPI 32A試験系を使用して、実施例7で試験したB.breve菌株を特性評価した。Rapid ID 32A試験を、製造業者の説明書に従って細菌コロニー上で実施した。簡潔に述べると、嫌気性ワークステーション内で、細菌をYCFA寒天培地上で37℃で24時間培養した。5μlの滅菌白金耳を使用してプレートからコロニーを除去し、McFarland標準液番号4とほぼ等しい濃度が得られるまで、2mlアンプルのAPI(登録商標)Suspension Mediumに再懸濁させた。Rapid ID 32Aストリップの各小チューブに55マイクロリットルの細菌懸濁液を加え、ウレアーゼ試験を2滴の鉱油で覆った。ストリップをプラスチック製の蓋で覆い、37℃で4時間好気的にインキュベートした後、次の試薬を使用して下の列の小チューブを発色させた:NIT:NIT1及びNIT2を各1滴;IND:James試薬を1滴;残りのすべての小チューブ:FastBlue試薬を1滴。ストリップを室温で5分間インキュベートした後、それぞれの小チューブの色を記録して、陰性、中間陽性または陽性の値に割付けた。
パルスフィールドゲル電気泳動(PFGE)を使用して、実施例7で試験したB.breve菌株を特性評価した。結果を図24に示す。B.breve参照菌株よりも高い抗菌活性を示したB.breve試験菌株を、類似のパターンにグループ化し、参照菌株と識別できることが判明した。
MRx004及びB.breve参照菌株に対して、さらなるパルスフィールドゲル電気泳動(PFGE)試験を実施した。PFGEは、臨床実験室における菌株タイピングの「至適基準」として日常的に適用されており[100]、ヒト糞便ビフィズス菌単離株を判別するための有効な方法であると報告されている[101]。実際、多くの試験において、XbaIまたはSpeI制限酵素のいずれかで消化したゲノムDNAのPFGEで分解した断片のフィンガープリント分析を使用して、ビフィズス菌単離株の関係性が調査されてきた[102、103]。SpeIは、Bifidobacterium breveのプラスミドプロファイリング及び種内遺伝子型同定に特に有用であることが証明されている[104、105]。
PFGE用の高分子量DNAのアガロースゲルプラグを、公開されているプロトコール[106]に従って調製した。
単一の薄片(2mm×2mm)を1mlの10mM Tris.Cl、0.1mM EDTA(pH8.0)で15分間、室温で3回洗浄した。各薄片を、酵素に推奨される250μlの制限緩衝液と4℃で30分間プレインキュベートし、次に20ユニットの制限酵素SmaIを含む250μlの新鮮な緩衝液に交換した。供給元(New England Biolabs(UK)Ltd)の推奨に従って、制限消化を25℃で一晩実施した。
ゲノムDNA(gDNA)の処理済(制限酵素)及び未処理のプラグを以下の条件下で検査した。λラダーを、ゲルにロードする前に45℃に加熱した。実施条件は、0.5倍のTPE緩衝液中、14℃、6.0V/cmで20時間であり、パルス時間は1秒から20秒まで増加させた。ラムダDNAラダー(Bio−Rad)をサイズマーカーとして使用した。0.5倍のTBE(1M Tris−ホウ酸塩、0.5M EDTA、pH8.5)で作製した1.0%アガロースゲル(Bio−Rad)のウェルにプラグを入れ、同じアガロースで密封した。DNA断片を、CHEF−DR IIIパルスフィールドシステム(Bio−Rad Laboratories,Hercules,CA)を使用して、14℃に維持した0.5倍のTBE泳動用緩衝液中で6V/cmで18時間分離させた。直線的に増加するパルス時間を選択した。断片の分離には、1秒から15秒の直線的に増加するパルス時間を使用した。光制限条件下で、ゲルを0.5μg/mlの臭化エチジウムを含有する蒸留水で120分間染色した。
バンドパターンを、[107]で概説されているガイドラインを使用して手動で評価した。PFGE画像をBioNumerics7.6(Applied Maths)で処理し、各菌株のバンドのフィンガープリントを生成した。フィンガープリントのクラスター分析を、Jaccard類似度係数(フィンガープリント型の分析に推奨されている)及び算術平均を用いた非加重結合法(UPGMA)を使用して実施した。
この試験で用いたDNA消化及び電気泳動の条件は、B.breve種のサブタイピングに有効であることが既に示されており[104]、B.breveの菌株を亜種レベルで識別するのに十分な解像度(10本を超える観察可能なバンド)を提供することが判明した。4/5の菌株の制限断片は明瞭であり、十分に分離されていた(図25)。クラスター分析(図26)により、菌株MRx0004の遺伝子型は、この試験において分析された他のB.breve菌株よりも、B.breve参照7の遺伝子型により近縁であることが示唆される。
T細胞分化を誘導するMRx0004の能力について、インビトロで末梢血単核細胞(PBMC、Stemcell、カタログ番号70025)を用いて調査した。簡潔に述べると、ウェル当たり50μlのcRPMI培地中、400,000/ウェルで、抗CD3(Ebioscience、CD3モノクローナル抗体(OKT3クローン)、機能的グレード、カタログ番号16−0037−81)を播種した96ウェルプレートに、PBMCを播種した(cRPMIは、RPMI 1640(+L−グルタミン、21875−034)2mM最終濃度ストック200mM、10%のHI FBS(Gibco life technologies、10082−147)、50μmのメルカプトエタノール(Gibco life technologies、21985−023)、及び1%のペニシリン/ストレプトマイシン(P4333、10mg/ml)を含む)。次いで、加熱死MRx0004(80℃で30分間インキュベートし、その後培養物をPBSで洗浄して、適切な細胞培地に再懸濁することにより調製し、生存数をプレーティングによって確認した)を、100μl/ウェル中に4,000,000個で各ウェルに添加した。37℃のインキュベータに3日間入れた後、細胞を取り出し、PMA−(Sigma、カタログ番号P8139)、イオノマイシン(Sigma、カタログ番号I3909)、及びGolgiSTOP(BD、カタログ番号554724)を含む培地に5時間再懸濁させた。PMAストックはDMSO中1mg/mlであり、これを100ug/mlにさらに希釈し(各試料でcRPMI中50ng/mlが必要であった)、イオノマイシンストックはDMSO中1mMであり(cRPMI中1μMを使用した)、GolgiStop濃度は4μl/6mlで使用した。上清を0.22μmフィルターに通し、共培養培地で適切に希釈した。

要旨
この試験の目的は、脾臓におけるMRx0004のインビトロの免疫刺激特性を特性評価することであった。
処理:未処理、10%YCFA及び10%Bifidobacterium breve菌株MRx0004。
脾細胞を、6〜8週齢の雌C57BL/6マウスから単離した脾臓から新たに調製した。簡潔に述べると、脾細胞を、96ウェルプレート中の10%のFBS、2mMのL−グルタミン及び100U/mlのペニシリン、100μg/mlのストレプトマイシン、55μMのβ−メルカプトエタノールを含むRPMI 1640に900,000細胞/ウェルで播種し、10%の細菌培地YCFA+(ブランク培地)またはMRx0518の静置培養物からの10%の無細胞細菌上清に静置またはそれらで刺激して、次に、CO2インキュベータ内で37℃で72時間インキュベートした。その後、無細胞上清を採取し、4℃、500gで5分間スピンダウンした。次に、サイトカイン分析のために試料を採取して−80℃で保管した。
MTTアッセイキットは、Merck Millipore(カタログ番号CT01)から購入した。72時間のインキュベーション後、10μlのMTT溶液を各ウェルに添加し、細胞をCO2インキュベータ内で4時間インキュベートした。その後、100μlのイソプロパノール/0.04M HCL溶液を各ウェルに添加し、波長560nm及び参照波長655nmで吸光度を測定した。
製造業者(Thermo Fischer Scientific)の推奨に従って26−plex Mouse ProcartaPlexマルチプレックスイムノアッセイを使用して、サイトカインの定量化を実施した。簡潔に述べると、MAGPIX(登録商標)MILLIPLEX(登録商標)システム(Merck)をxPONENTソフトウェア(Luminex,Austin,TX,USA)とともに用いて、50μlの無細胞の共培養上清をサイトカインの定量化に使用した。5パラメータロジスティック曲線及びバックグラウンド除去を用いた、MILLIPLEX(登録商標)analystソフトウェア(Merck)を使用してデータを分析し、平均蛍光強度をpg/ml値に変換した。
まず、細胞をViobility 405/520 Fixable Dye(Miltenyi Biotec Ltd.Bergisch Gladbach,Germany)で染色して、暗所で室温において10分間、生細胞と死細胞とを判別した。次に、細胞をCD3、CD4、CD8及びIFN−γに対する抗体の混合物で染色して、細胞の表現型を決定し(Miltenyi REA抗体)、室温でさらに10分間インキュベートした。次に、細胞を洗浄してPBSに再懸濁し、フローサイトメトリー分析によって直ちに分析した。最初の実験の際に、ゲートの設定を支援するためにすべての抗体に対してアイソタイプを使用し、すべての実験にわたりFMO対照を含めた。すべての実験は、BD FACS Aria IIを使用して実施し、FACSDivaソフトウェア(BD Biosciences,Reading,UK)を使用して、「生(Live)」ゲート内に100,000細胞で設定した取得用のストップゲートを用いた。Flowjoバージョン10.4.2ソフトウェア(FlowJo LLC,Oregon,USA)を使用して分析を実施し、分析は、生死判定用色素で特定した生細胞に基づくものであった。
処理後の脾細胞の生存率を、脾細胞の代謝活性を測定するMTTアッセイを使用して評価した。図29は、MRx0004による処理後に脾細胞が生存可能であることを示す。

Claims (30)

- 前記免疫不全症が、原発性免疫不全症または続発性免疫不全症である、請求項2に記載の組成物。
- 前記原発性免疫不全症が選択される、請求項3に記載の組成物。
- 前記続発性免疫不全症が、AIDS、白血病などの免疫系のがん、ウイルス性肝炎、多発性骨髄腫などの免疫複合体疾患から選択される、請求項3に記載の組成物。
- 前記組成物が、ワクチンアジュバントとして使用するためのものである、請求項1に記載の組成物。
- 前記組成物が、免疫老化の処置、予防、または遅延に使用するためのものである、請求項1に記載の組成物。
- 前記組成物が、CAR−Tなどの細胞療法を増強するのに使用するためのものである、請求項1に記載の組成物。
- 前記組成物が、IL−12p70、IL−12p70、IFNγ、IL−4、TNF−αならびに/またはIL−17αの発現レベル及び/もしくは活性の上昇に使用するためのものである、請求項1〜8のいずれかに記載の組成物。
- 前記組成物が、TLR2を刺激するのに使用するためのものである、請求項1〜9のいずれかに記載の組成物。
- 前記組成物が、NFκBを刺激するのに使用するためのものである、請求項1〜10のいずれかに記載の組成物。
- 前記細菌株が、完全な菌体外多糖遺伝子座を含む、請求項1〜11のいずれかに記載の組成物。
- 前記細菌株が、プルラナーゼを発現する、請求項1〜12のいずれかに記載の組成物。
- 細菌感染症の処置または予防に使用するための、Bifidobacterium breve種の細菌株を含む組成物。
- 前記組成物が、胃腸細菌感染症の処置または予防に使用するためのものである、請求項14に記載の組成物。
- 前記組成物が、グラム陰性菌感染症の処置または予防に使用するためのものである、請求項14または請求項15に記載の組成物。
- 前記細菌株が、配列番号1と少なくとも95%、96%、97%、98%、99%、99.5%もしくは99.9%同一である16s rRNA遺伝子配列を有するか、または配列番号1によって表される16s rRNA遺伝子配列を有する、請求項1〜16のいずれかに記載の組成物。
- 前記細菌株が、ラフィノースを発酵させることができる、請求項1〜17のいずれかに記載の組成物。
- 前記細菌株が、α−ガラクトシダーゼ、β−ガラクトシダーゼ、α−グルコシダーゼ及びβ−グルコシダーゼ、α−アラビノース、マンノース及びラフィノースのうちの1または2以上、例えば2、3、4、5、6または7つすべてを発酵させることができる、請求項1〜18のいずれかに記載の組成物。
- 前記細菌株が、アクセッション番号42380でNCIMBに寄託された菌株である、請求項1〜19のいずれかに記載の組成物。
- 経口投与用である、請求項1〜20のいずれかに記載の組成物。
- 前記組成物が、1もしくは2以上の薬学的に許容される賦形剤または担体を含む、請求項1〜21のいずれかに記載の組成物。
- 前記細菌株が、凍結乾燥されている、請求項1〜22のいずれかに記載の組成物。
- Bifidobacterium breve種の細菌株を含む組成物を、それを必要とする患者に投与することを含む、免疫刺激の低減に関連する疾患もしくは状態を処置または予防する方法。
- 請求項1〜19のいずれかに記載された細菌株の細胞を含む組成物であって、前記細胞が1または2以上の異種抗原を発現する、前記組成物。
- 前記細胞が、前記1または2以上の異種抗原を提示する、請求項25に記載の組成物。
- ワクチンとして使用するための、請求項25または請求項26に記載の組成物。
- 請求項1〜23のいずれかに記載された細菌株の細胞であって、前記細胞が1または2以上の異種抗原を発現する、前記細胞。
- 前記細胞が、前記1または2以上の異種抗原を提示する、請求項28に記載の細胞。
- ワクチンとして使用するための、請求項28または請求項29に記載の細胞。
