「活性化」又は「リンパ球の活性化」という用語は、RNA、タンパク質、及びDNAの合成及びリンホカインの産生をもたらし;その後、様々なエフェクター細胞及びメモリー細胞の増殖及び分化が生じる、特定の抗原、非特異的マイトジェン、又は同族異系細胞によるリンパ球の刺激を指す。T細胞の活性化は、TCR/CD3複合体と、クラスI又はクラスII MHC分子の溝に結合したペプチドであるその同族リガンドとの相互作用に依存する。受容体の結合によって引き起こされる分子事象は複雑である。初期のステップでは、チロシンキナーゼの活性化が、いくつかのシグナル伝達経路を制御する一連の基質のチロシンリン酸化をもたらすと考えられる。これらには、TCRをras経路に連結する一連のアダプタータンパク質、ホスホリパーゼCγ1が含まれ、そのチロシンリン酸化は、その触媒活性を高め、イノシトールリン脂質代謝経路に関与し、細胞内遊離カルシウム濃度の上昇、並びにプロテインキナーゼCと、細胞の成長及び分化を制御する他の一連の酵素の活性化をもたらす。T細胞の完全な応答性には、受容体の関与に加えて、アクセサリー細胞が提供する共刺激活性、例えば、APC上のCD80及び/又はCD86によるT細胞上のCD28の関与が必要である。
適応免疫応答による病原体の認識と根絶に続いて、T細胞の大部分(90〜95%)が、残りの細胞と共にアポトーシスを受け、メモリーT細胞、指定セントラルメモリーT細胞(TCM)、エフェクターメモリーT細胞(TEM)、及び常在メモリーT細胞(TRM)のプールが形成される(Clark,R.A.,“Resident memory T cells in human health and disease”,Sci.Transl.Med.,7,269rv1,(2015))。
本出願人は、鉄依存性細胞分解を誘導する薬剤での細胞の処置が、免疫活性を高める後細胞シグナル伝達因子の産生及び放出をもたらすことを示した。これらの結果は、鉄依存性細胞分解の阻害が免疫活性を低下させることを示唆している。従って、鉄依存性細胞分解を阻害して免疫活性を低下させる薬剤は、炎症、急性臓器損傷、組織損傷、敗血症、虚血、アテローム性動脈硬化症、神経変性障害、及び免疫関連疾患又は状態を含む炎症性障害又は状態などの免疫活性の低下から恩恵を受け得る障害の処置に使用することができる。
いくつかの態様では、本開示は、炎症性疾患又は状態を処置する方法に関し、この方法は、対象の炎症性疾患又は状態を処置するのに十分な量の鉄依存性細胞分解を阻害する薬剤を対象に投与することを含む。いくつかの実施形態では、炎症性疾患又は状態は、鉄依存性細胞分解(例えば、フェロトーシス)が有害である炎症性疾患又は状態である。

当技術分野で公知であり、本明細書に記載される鉄依存性細胞分解を阻害する薬剤に加えて、本開示はさらに、鉄依存性細胞分解を阻害し、且つ免疫活性を低下させる他の化合物を同定するための方法に関する。

HT1080細胞(5,000個の細胞/ウェル)を、DharmaFECT Iトランスフェクション試薬(Catalog # T−2001)及び対照siRNA(Dharmacon Catalog # D−001810−10−05)又はACSL4を標的とするsiRNA[37.5nM](図9A、Dharmacon Catalog # L−009364−00−005)又はACSL4(Thermo Fisher Silencer Select Catalog #'s :s5001、s5001、s5002)若しくはCARS(Thermo Fisher Silencer Select Catalog #'s :S2404、s2405、s2406)に対するsiRNAのプール(図9B/C)を用いて96ウェルフォーマットで逆トランスフェクトした。トランスフェクションの48時間後、細胞培養培地を、様々な濃度のエラスチン(図9A)又は固定濃度のエラスチン(10μM、図9B/C)を含む新鮮な培地に交換した。加えて、50,000個のレポーターTHP1−dual細胞をいくつかのプレートに追加した(図9C)。24時間後、HT1080細胞の生存率を測定するか(図9A/B)、又はTHP1の上清をNFKB活性について評価した(図9C)。HT1080細胞はATCCから入手し、THP1−Dual細胞はInvivoGen(San Diego,CA)から入手した。
ACSL4遺伝子は、細胞内のアラキドン酸のレベルを制御し、細胞死の調節に関与するアシル−CoAシンテターゼである長鎖脂肪酸−CoAリガーゼ4をコードする。CARS遺伝子は、システイニル−tRNAシンテターゼをコードする。CARSのノックダウンは、脂質活性酸素種の誘導を防止することによってエラスチン誘導性フェロトーシスを阻害することが示されている。Hayano et al.,2016,Cell Death Differ.23(2):270−278を参照されたい。図9A及び図9Bに示されているように、HT1080細胞におけるACSL4又はCARSのいずれかの遺伝子ノックダウンは、エラスチンの存在下で培養したHT1080細胞の生存率をある程度助ける。加えて、HT1080細胞におけるACSL4又はCARSのいずれかの遺伝子ノックダウンは、エラスチン処置HT1080細胞と共培養された単球のNFKB活性を無効にする(図9C)。これらの結果は、細胞死がエラスチン処理HT1080細胞によって誘発されるNFKBシグナル伝達の誘導に役割を果たすこと、及び特定の細胞内タンパク質の欠如がフェロトーシスの炎症促進性を低下させることを実証している。加えて、これらの結果は、鉄依存性細胞分解(例えば、フェロトーシス)を阻害する薬剤が免疫活性(例えば、NFKB活性)を低下させ得ることを実証している。
薬剤/治療デザイン:
A20リンパ腫細胞を、1μM フェロスタチン−1の存在下又は非存在下で、様々な用量(例えば、0.002μM、0.005μM、0.014μM、0.041μM、0.123μM、0.370μM、1.111μM、3.333μM、及び10μM)のGPX4阻害剤(RSL3、ML162、又はML210)で24時間処置した。ML162は、Cayman Chemical Company Incから購入し、DMSOに再懸濁した。ML210は、Sigma−Aldrichから購入し、DMSOに再懸濁した。A20リンパ腫細胞も、陰性対照としてDMSOで処置した。DMSO又はGPX4阻害剤での処置の24時間後、A20リンパ腫細胞を、THP1−Dual細胞とさらに24時間共培養した。続いて、THP1の上清を、NFKBレポーターの活性について評価した。A20細胞はATCCから入手し、THP1−Dual細胞はInvivoGen(San Diego,CA)から入手した。両方の細胞型は、アッセイの期間中、96ウェルプレートで培養した。

- 前記対照細胞、対照組織、又は対照対象が、鉄依存性細胞分解を誘導する薬剤と接触されている、請求項56に記載の方法。
- 前記試験薬と接触していない対照細胞、対照組織、又は対照対象における、脂質過酸化反応、イソプロスタン、活性酸素種(ROS)、鉄、PTGS2、及びCOX−2からなる群から選択されるマーカーのレベル又は活性と比較した、試験薬と接触した細胞、組織、又は対象における前記マーカーのレベル又は活性の低下が、試験薬が、鉄依存性細胞分解を阻害する薬剤であることを示している、請求項51〜57のいずれか一項に記載の方法。
- 前記試験薬と接触していない対照細胞、対照組織、又は対照対象における、GPX4、MDA、及びGSHからなる群から選択されるマーカーのレベル又は活性と比較した、前記試験薬と接触した前記細胞、組織、又は対象における前記マーカーのレベル又は活性の上昇が、前記試験薬が鉄依存性細胞分解を阻害する薬剤であることを示している、請求項51〜57のいずれか一項に記載の方法。
- 前記候補免疫抑制剤を評価することが、前記選択された候補免疫抑制剤と接触した細胞と共に免疫細胞を培養するか、又は前記選択された候補免疫抑制剤と接触した細胞によって産生される後細胞シグナル伝達因子に免疫細胞を曝露すること、及び前記免疫細胞のNFκB、IRF、若しくはSTINGのレベル又は活性を測定することを含む、請求項51に記載の方法。
- 前記免疫細胞がTHP−1細胞である、請求項60に記載の方法。
- 前記候補免疫抑制剤を評価することが、前記選択された候補免疫抑制剤と接触した細胞と共にT細胞を培養するか、又は前記選択された候補免疫抑制剤と接触した細胞によって産生される後細胞シグナル伝達因子にT細胞を曝露すること、及び前記T細胞の活性化及び増殖を測定することを含む、請求項60に記載の方法。
- 前記候補免疫抑制剤を評価することが、前記選択された候補免疫抑制剤と接触した細胞と共に培養されたか、又は前記選択された候補免疫抑制剤と接触した細胞によって産生される後細胞シグナル伝達因子に曝露された免疫細胞におけるNFκB、IRF、又はSTINGのレベル又は活性を、前記選択された候補免疫抑制剤と接触しておらず、また前記選択された候補免疫抑制剤と接触した細胞によって産生される後細胞シグナル伝達因子に曝露もされていない対照免疫細胞におけるNFκB、IRF、又はSTINGのレベル又は活性と比較することをさらに含む、請求項60に記載の方法。
- 前記選択された候補免疫抑制剤と接触した前記細胞が、鉄依存性細胞分解を誘導する薬剤にも接触している、請求項63に記載の方法。
- 前記鉄依存性細胞分解を誘導する薬剤が、アンチポーター系Xc−の阻害剤、GPX4の阻害剤、及びスタチンからなる群から選択される、請求項53、57、及び64のいずれか一項に記載の方法。
- 前記アンチポーター系Xc−の阻害剤が、エラスチン又はその誘導体又は類似体である、請求項65に記載の方法。
- 前記エラスチンの類似体がPE又はIKEである、請求項66に記載の方法。
- 前記GPX4の阻害剤が、(1S,3R)−RSL3又はその誘導体又は類似体、ML162、DPI化合物7、DPI化合物10、DPI化合物12、DPI化合物13、DPI化合物17、DPI化合物18、DPI化合物19、FIN56、及びFINO2からなる群から選択される、請求項65に記載の方法。
- 前記スタチンが、アトルバスタチン、フルバスタチン、ロバスタチン、ピタバスタチン、プラバスタチン、ロスバスタチン、セリバスタチン、及びシンバスタチンからなる群から選択される、請求項65に記載の方法。
- 前記鉄依存性細胞分解を誘導する薬剤が、ソラフェニブ又はその誘導体又は類似体、スルファサラジン、グルタミン酸塩、BSO、DPI2、シスプラチン、システイナーゼ、シリカベースのナノ粒子、CCI4、クエン酸鉄アンモニウム、トリゴネリン、及びブルサトール(brusatol)からなる群から選択される、請求項53、57、及び64のいずれか一項に記載の方法。
