Abstract
Description
(1.マウス及び免疫化過程)
雌BALB/cマウス(約25g、生後7週齢)をOrient−Bio社(大韓民国、ソウル)から購買し、生後8週齢になったとき実験に用いた。マウスは、グループ当たり5匹のマウスで構成される4個のグループに無作為に分けた。
HIV−1 p24 Gagを発現するrBCG菌株の3個の相異なる類型、すなわち、pMyong2−p24プラスミドを有するBCG(rBCG−pMyong2−p24)、pAL−p24プラスミドを有するBCG(rBCG−pAL−p24)及びpMV306−p24プラスミドを有するBCG(rBCG−pMV306−p24)を生成するために、3個で構成されたプラスミド、すなわち、pMV306−p24、pAL−p24及びpMyong2−p24をコンピテントBCG菌株(Tokyo 172)にGene PulserIIエレクトロポレーション装置(Bio−Rad、Hercules、CA、USA)を用いてエレクトロポレーションした。形質転換体は、カナマイシン(100μg/ml)及びOADCが含有されたMiddlebrook 7H10培地(Difco Laboratories、Detroit、MI、USA)上で選択した。典型的に、形質転換体コロニーをプレートから選択し、0.5%グリセロール、0.05% Tween−80、10% ADC及びカナマイシンが補充された7H9培地(Difco Laboratories、Detroit、MI、USA)に移して3週〜4週間培養した。rBCG菌株の成長率を600nmでの光学密度(OD)により測定した。
組換えp24タンパク質は、上述したものから多少変形させて大腸菌から精製した。融合タンパク質の発現及び精製のために、大腸菌BL21菌株(RBC Bioscience、Taipei City、台湾)をpET23a−p24に形質転換させた。タンパク質の発現は、0.4mMのイソプロピルβ−D−チオガラクトピラシド(IPTG、DuchefaBiochemie、Haarlem、オランダ)を添加することで誘導した。バクテリア細胞を収得して氷で10分間超音波処理により破壊した。超音波処理された溶解物を1600×gで20分間4℃で遠心分離し、p24タンパク質を含有したペレットを4M尿素(Urea、Sigma Aldrich、St.Louis、MO、USA)を含有した結合緩衝液に再懸濁した。タンパク質をNi−NTA His結合樹脂(Merck、Darmstadt、ドイツ)を用いて精製し、4M尿素を含有した溶出緩衝液(300mM NaCl、50mM リン酸ナトリウム緩衝液、250mMイミダゾール)で溶出した。精製されたタンパク質を溶出緩衝液に対して連続的に透析してイミダゾール、尿素及び残留塩を除去した。p24タンパク質の純度をドデシル硫酸ナトリウム−ポリアクリルアミドゲル電気泳動(SDS−PAGE;12%ゲル)により評価した。ゲルは、クマシーブリリアントブルー染色法を用いて視覚化した(図1)。
骨髓由来樹状細胞(BMDCs)を上述したように8週齢〜12週齢のBALB/cマウスの骨髓(BM)から製造した。簡単に説明すれば、BM細胞を血清不含有のIscove’s modified Eagle medium(IMDM;Gibco Invitrogen、UK)を用いて大腿骨及び硬骨から除去した。単一細胞懸濁液を24ウェルプレートに10% FBS(Gibco Invitrogen)、組換えマウスGM−CSF(1.5ng/ml;PeproTech、Rocky Hill、NJ、USA)及びマウスIL−4(1.5ng/ml;PeproTech、USA)、ペニシリン(100units/ml;Gibco Invitrogen)、ストレプトマイシン(100μg/ml;Gibco Invitrogen)、ゲンタマイシン(50μg/ml;Gibco Invitrogen)、L−グルタミン(2mM;Gibco Invitrogen)及びβ−メルカプトエタノール(50nM;Gibco Invitrogen)が補充された最終体積2mlの完全IMDMにウェル当たり1×106細胞の濃度で播種した。培地の半分を6日間一日おきに同等な体積の完全IMDMに交替した。BMDCsを用いたそれぞれの実験を準備するために5匹のマウスを用い、BMDCsを分化させるために5個の24ウェルプレートを用いた。
マウスのマクロファージ細胞株J774.1(American Type Culture Collection、ATCC TIB−67)を37℃及び5%CO2で10%(v/v)ウシ胎児血清(FBS)、2mMグルタミン及び必須アミノ酸が補充されたDulbecco’s modified Eagle’s medium(DMEM;Thermo Scientific、Rockford、IL、USA)で維持させた。上述したように、マウス骨髓からBMDCsを生成させ、37℃及び5%CO2で10%FBS(Gibco Invitrogen)、組換えマウスGM−CSF(1.5ng/ml;PeproTech、Rocky Hill、NJ、USA)、マウス IL−4(1.5ng/ml;PeproTech、USA)、ペニシリン(100units/ml;Gibco Invitrogen)、ストレプトマイシン(100μg/ml;Gibco Invitrogen)、ゲンタマイシン(50μg/ml;Gibco Invitrogen)、L−グルタミン(2mM;Gibco Invitrogen)及びβ−メルカプトエタノール(50nM;Gibco Invitrogen)が補充されたIscove’s modified Eagle medium(IMDM;Gibco Invitrogen、UK)で維持させた。
rBCG菌株でp24 Gag発現レベルを測定するために、ウエスタンブロッティング及びELISA分析を実行した。簡単に説明すれば、培養されたrBCG菌株のペレットをリゾチーム(100μg/ml)、DNase(5U/ml)及びプロテアーゼインヒビターが補充されたB−PER緩衝液(Thermo scientific、Rockford、IL、USA)に懸濁させた。その後、懸濁液を氷で5分間超音波処理し(パルス:0.3秒、中断:0.7秒)、13,000rpmで4℃で15分間遠心分離した。水相の同一の量のタンパク質をウエスタンブロッティング分析に用いた。各rBCG菌株でp24の発現レベルは、マウス抗p24モノクローナル抗体(Abcam、Cambridge、USA;1:1,000希釈)を用いて測定した。タンパク質濃度が全てのサンプルで同等であったかを確認するために、マイコバクテリアHsp65(Abcam、1:1,000希釈)を内部対照群として用いた。p24の安定的な発現を評価するために、多様な継代地点(1、4、6、8、10及び12回継代後)でrBCG−pMyong2−p24菌株のp24発現レベルも測定した。継代過程は、プレートからプレートに(カナマイシンがあるかない7H10寒天プレート)実行し、各継代から得たコロニーを7H9 broth培地で3週間培養した後にそれぞれの実験を実行した。追加的に、p24 ELISAキットを用いて(製造社から提案された方法で)同一の量のタンパク質でp24レベルの検出のために用いた(3個一セットのウェルで)(ABL、Rockville、USA)。全てのグループを2個の独立的な実験で分析した。
rBCG感染のために、J774.1細胞及びBMDCsをウェル当たり5〜10×105個の細胞で(24ウェルプレート、3個一セット)播種して18時間の間培養した。3個の相異なるrBCG菌株を細胞に10の感染多重性(multiplicity of infection、M.O.I)で感染させた。また、相異なるM.O.I(1及び10M.O.I)のrBCG−pMyong2−p24菌株をBMDCsに感染させて相異なるM.O.Iによるp24発現の差を比較した。J774.1細胞及びBMDCsを4時間の間インキュベーションしてバクテリアの食作用を許容し、細胞外バクテリアをPBSで3回洗浄して除去した。感染されたJ774.1細胞及びBMDCsは、24時間及び/又は72時間の間インキュベーションした。
T細胞増殖の分析のために次のような実験を実行した。マウス2匹にp24タンパク質(30μg/マウス)を静脈内注射した。7日後、脾臓細胞を極低温FACS緩衝液[1%ウシ血清アルブミン(BSA)及び1mM EDTAを含有したPBS]で洗浄し、氷で30分間10%ラット血清(Sigma Aldrich)、10%塩素血清(Gibco Invitrogen)、10%マウス血清(Sigma Aldrich)及び2.4G2モノクローナル抗体(10μg/ml;BD Biosciences、San Diego、CA、USA)を含有したスーパーブロック(super block)溶液でブロッキングした。細胞を続いてBV421−コンジュゲートされた抗−CD4(Clone GK1.5、BD Biosciences)及びPE−コンジュゲートされた抗−CD8a(Clone 53−6.7、eBioscience、San Diego、CA、USA)で30分間4℃で染色して極低温FACS緩衝液で3回洗浄した。FACS AriaIII器機(BD Biosciences)を用いてCD4及びCD8 T細胞集団を分類した。また、共培養する前日、未成熟BMDCsを野生型、2個のrBCG(rBCG−pMyong2−p24及び−pAL−p24)又はrSmeg−pMyong2−p24菌株で10のM.O.Iで24時間の間感染させた。増殖分析は、蛍光細胞質追跡染料であるCFSE(Invitrogen、Carlsbad、USA)を用いて実行した。分類されたCD4及びCD8 T細胞を5μM CFSEで4分間37℃で、及び4分間氷で染色した。その後、CFSEが標識されたT細胞及び感染されたBMDCsを4日間共培養した。
前記T細胞増殖分析過程で共培養された上層液(3個一セットのウェル)のマウスIL−2の量をELISAを用いて製造社のマニュアル(BioLegend、USA)によって測定した。全ての実験を2回実行した。
野生型及びrBCG菌株で免疫化されたマウス(5匹/グループ)から得た脾臓細胞を用いて次のようにELISPOT検定を実行した。簡単に説明すれば、96−well ELISPOTプレート(PVDF膜)をPBS中のマウスIFN−γ(3μg/ml、クローン:AN−18)捕捉抗体(BD−Biosciences、San Diego、CA、USA)でコーティングして一晩4℃でインキュベーションした。捕捉抗体を捨て、プレートを0.05% Tween−20(PBST)を含有したPBS及びPBS(それぞれ3回)で洗浄し、プレートを10% FBSを含んだ200μlのRPMI 1640培地で3時間の間37℃でブロッキングした。ブロッキングした後、予防接種されたマウスから採取した脾臓細胞を各ウェル当たり5×105個ずつローディングした。それぞれの治療グループに対して、細胞を3個一セットで総200μlの体積のうち5μg/mlの精製されたp24抗原又は培地単独で刺激した。その後、プレートを37℃で24時間の間インキュベーションした。細胞を陽性対照群として5ng/mlのホルボール12−ミリステート13−アセテート(PMA)(Sigma−Aldrich、St.Louis、USA)及び500ng/mlのイオノマイシン(Sigma−Aldrich)で刺激した。PBST及びPBS(それぞれ3回)で洗浄した後、各ウェルをビオチンが標識されたマウスIFN−γ(3μg/ml、クローン:XMG1.2)検出抗体(BD−Biosciences)で処理してプレートを一晩4℃でインキュベーションした。細胞をさらに洗浄してホースラディッシュ・ペルオキシダーゼ(HRP)−コンジュゲートされたストレプトアビジンを各ウェルに添加した。HRP反応は、3−アミノ−9−エチルカルバゾール(AEC)基質試薬セット(BD−Biosciences)を用いて展開した。ウェル当たりスポット形成単位(SFUs)の数をELISPOT判読機(AID ELISPOT判読機、Strasburg、ドイツ)を用いて自動で計数した。全てのグループを3個一セットで分析し、2回の独立的な実験を実行した。
免疫化されたマウス(5匹/グループ)から得た脾臓細胞を10% FBSを含んだRPMI 1640培地で1×106細胞/ウェル(96ウェルマイクロプレート、200μl体積、3個一セットで)の濃度に調整し、精製されたp24タンパク質を試験管内刺激のために5μg/mlの濃度で添加した。細胞を培養し、上層液をELISAキットを用いてIL−2(BioLegend、San Diego、CA、USA)、IL−6(eBioscience)及びIFN−γ(BioLegend)サイトカイン測定のために収得した。全てのグループを3個一セットで分析し、2回の独立的な実験を実行した。
血清抗体の割合を検出するために、免疫化されたマウス(5匹/グループ)からCO2の過呼吸を通じた安楽死後に心臓穿孔方法を用いて血清サンプルを収集した。96ウェルプレートを一晩4℃で0.05M炭酸塩−重炭酸塩緩衝液(pH9.6)中の精製されたp24タンパク質(5μg/ml)でコーティングした。プレートをPBST及びPBSで3回洗浄して常温(RT)で1時間の間5%ウシ血清アルブミン(PBST中のBSA)でブロッキングした。血清サンプルをPBSで1:10の割合で希釈し、100μlをウェルに添加した(3個一セットで)。プレートを2時間の間常温でインキュベーションし、PBST及びPBSで3回洗浄し、1時間の間ビオチン化ラット抗−マウスIgG2a、IgG1(BD Biosciences、1:1,000希釈)及び総IgG(eBioscience、1:1,000希釈)抗体とともにインキュベーションした。その後、プレートを再び洗浄し、HRPコンジュゲートされたストレプトアビジン(eBioscience))とともに30分間常温でインキュベーションした。最終洗浄段階後に、全てのウェルをBD OptEIA基質(BD Biosciences)と10分間反応させた後、1N H2SO4を用いて反応を中断させた。分光計を用いて450nmの波長で光学密度(OD)を測定した。
誘導されたCTL反応は、上述したものから多少変形を加えて測定した。簡単に説明すれば、エフェクター細胞の場合、それぞれの免疫化されたグループのマウスから得た脾臓細胞を主要組織適合性複合体(MHC)class I−制限p24ペプチドA9I(AMQMLKETI)(10μg/ml;Peptron、大田、大韓民国)を用いてパルスし、6日間IL−2(30U/ml;PeproTech、Rocky Hill、USA)とともに37℃で5% CO2インキュベーターでインキュベーションした。標的細胞、すなわち、P815細胞(H−2d)をA9Iペプチド(10μg/ml)とともに2時間の間インキュベーションした後、エフェクター細胞及び標的細胞の共培養により製造した。細胞の細胞毒性をU字底96ウェルプレートで乳酸脱水素酵素(LDH)分析法により製造社のプロトコル(CytoTox 96非放射性細胞毒性分析;Promega、Madison、USA)によって評価した。簡単に説明すれば、エフェクター細胞(抗原により刺激された脾臓細胞)を標的細胞(p24パルスされたP815細胞)に3個一セットで相異なるエフェクター/標的(E/T)割合(10:1、20:1〜50:1の範囲)で6時間の間添加した;その後、培養された上層液から放出されたLDH値を490nmで分光計を用いて検出した。特異的細胞溶解の百分率を次の式を用いて計算した:[{実験(Experimental)−自然発生的エフェクター(Effector spontaneous)−自然発生的標的(Target spontaneous)}]/{最大標的(Target maximum)−自然発生的標的(Target spontaneous)}×100(%)。全てのグループを3個一セットで分析し、2回の独立的な実験を実行した。
提供された全てのデータは、平均±標準偏差として表示した。スチューデントt−テストは、変量を比較するためにマイクロソフトエクセルソフトウェアを用いて実行し、p値が0.05より少ないときの差を統計学的に有意味なものとみなした。
[実施例1.rBCG−pMyong2−p24菌株はバクテリア及び感染細胞でHIV−1 p24 Gag発現向上をもたらす]
本発明では、HIV−1 p24 Gag予防接種のためのrBCGの製造でpMyong2ベクターシステムの有用性を調べるために、相異なるマイコバクテリウム−大腸菌シャトルベクター、すなわち、pMyong2−TOPO、pAL−TOPO及びpMV306をそれぞれ用いてp24を発現する三つの類型のrBCG菌株、すなわち、rBCG−pMyong2−p24、rBCG−pAL−p24及びrBCG−pMV306−p24を生成した(図2A)。7H9 broth(100μg/mlのカナマイシン含む)で30日間3個のrBCG菌株の成長率を比較した。その結果、rBCG及び野生型BCG菌株がほとんど同一の成長率を示した(図2B)。追加的に、マクロファージ及びDCsにおいてこのようなrBCG菌株の生存を調査するために、感染された細胞を0.05% Triton X−100(in PBS)で溶解させて7H10寒天プレート上にプレーティングした。二つの細胞で全て、向上されたp24発現によるバクテリア負荷(bacterial burden)によりrBCG−pMyong2−p24菌株は、他の菌株(すなわち、rBCG−pAL−p24、−pMyong2−p24及び野生型BCG菌株)より少ないCFUを示した(図2C)。
本発明では、向上されたp24タンパク質生成を示すrBCG−pMyong2−p24がT細胞増殖能力を改善させることができるか否かを測定するために、4個の相異なる菌株、すなわち、野生型BCG(対照群として)、2つの類型のrBCG(rBCG−pMyong2−p24及びrBCG−pAL−p24)及びrSmeg−pMyong2−p24でそれぞれ感染されたBMDCsでのT細胞増殖を混合されたリンパ球反応(mixed lymphocyte response、MLR)を通じたCFSE希釈方法を用いて分析を実行した。同一のpMyong2ベクターシステム、すなわち、rBCG−pMyong2−p24及びrSmeg−pMyong2−p24を用いて2個の相異なる種の間のHIV−1 p24 Gag特異的免疫反応を誘導する能力を比較するために、rSmeg−pMyong2−p24菌株をやはり含ませた。T細胞増殖分析の概略的な内容は、図4Aに示した。
rBCG−pMyong2−p24の予防接種後にT細胞反応が改善されたか否かを測定するために、3個の相異なる菌株、すなわち、2つの類型のrBCG菌株(rBCG−pMyong2−p24及び−pAL−p24)、rSmeg−pMyong2−p24(図5A)及び対照群として野生型BCG菌株(〜106CFU)で皮下免疫化されたBALB/cマウス(5匹/グループ)の脾臓から脾臓細胞を分離し、IFN−γ ELISPOT分析法を用いてHIV−1 p24 Gag特異的T細胞反応に対して分析した。3個の組換え菌株で皮下免疫化されたマウスから得た脾臓細胞は、野生型BCG菌株で免疫化されたマウスから得た脾臓細胞よりさらに高いSFUsを示した。特に、rBCG−pMyong2−p24(987.78±195.11 SFUs/106脾臓細胞)で免疫化されたマウスから採取した脾臓細胞は、他の2個の菌株、すなわち、rBCG−pAL−p24(479.56±213.90 SFUs/106脾臓細胞)及びrSmeg−pMyong2−p24(647.00±151.01 SFUs/106脾臓細胞)で免疫化されたマウスから採取した脾臓細胞よりさらに高いSFUsを誘発した(図5B)。しかし、予防接種されたマウスからのp24特異的IFN−γ SFUsでrBCG−pAL−p24とrSmeg−pMyong2−p24菌株の間に有意な差は観察されなかった(図5B)。
rBCG菌株及びrSmeg−pMyong2−p24で二回免疫化した後4週後に得られた脾臓細胞(5匹/グループ)(図5A)を精製されたp24タンパク質(5μg/ml)で3個一セットで試験管内で刺激させ、細胞培養上層液中のIL−2、IFN−γ及びIL−6の誘導されたサイトカイン生成を検出した。二つの種類のTh1型サイトカイン、すなわち、IL−2及びIFN−γ、そして一つの前炎症性サイトカイン、すなわち、IL−6で、rBCG−pMyong2−p24菌株は、野生型又は他の2個の組換え菌株でよりも常に、全ての時点(1日目及び3日目)で、予防接種されたマウスから得た脾臓細胞でよりもさらに高いレベルのサイトカインを生成した(図5C及び表1)。
本発明では、rBCG−pMyong2−p24が免疫化されたマウスでTh1に傾斜した体液性反応を誘導するか否かを測定するために、それぞれTh1及びTh2反応の公知されたマーカーであるHIV−1 p24 Gag特異的IgG2a及びIgG1のレベルを分析した。図6に示したように、2個のrBCG及びrSmeg−pMyong2−p24菌株は、野生型よりさらに高いレベルのIgG2aアイソタイプを誘導した。IgG1アイソタイプと関連して3個の組換え菌株が類似したレベルのIgG1を誘導した;しかし、その結果は、統計学的有意性には到逹しなかった。総IgGの場合に、rBCG−pMyong2−p24は、他の二つの組換え菌株(すなわち、rBCG−pAL−p24及びrSmeg−pMyong2−p24)より相当にさらに高いレベルの総IgGを示した(図6)。
本発明では、rBCG−pMyong2−p24が免疫化されたマウスで向上されたHIV−1 p24 Gag−特異的細胞傷害性Tリンパ球(CTL)反応を誘導するか否かを測定するために、2個のrBCG(rBCG−pMyong2−p24、−pAL−p24)、rSmeg−pMyong2−p24又は野生型BCG菌株で免疫化されたマウスからの脾臓細胞でCTL活性を、LDH細胞毒性分析を通じて評価した。免疫化過程は、図5Aで記述した。A9Iペプチドで2時間の間パルスされたP815細胞(H−2d)が標的細胞として用いられ、エフェクター/標的割合は、上述したように10:1、20:1及び50:1であった。図7に示したように、50:1のE:T割合で、rBCG−pMyong2−p24で免疫化されたマウスのCTLが他の菌株で免疫化されたものより相当にさらに高いレベルのHIV−1 p24 Gag特異的標的細胞溶解を誘導できることを示す(図7)。しかし、rBCG−pMyong2−p24とrSmeg−pMyong2−p24菌株の間で有意な差は観察されなかった(図7)。
本発明では、p24タンパク質とrBCG−pMyong2−p24菌株の相異なるCFU間のp24特異的免疫反応を比較するために、次のようなグループで独立的な生体内実験を実行した。
i)PBS対照群
ii)p24タンパク質(30μg/マウス)注射、iii)rBCG−pMyong2−p24(1×106CFU)注射、及びiv)rBCG−pMyong2−p24(1×107CFU)注射(1週間隔、2回皮下注射)群。
免疫化過程は、実験方法の段落で記述した。最後の免疫化後に、p24−特異的IFN−γ ELISPOT、IgG亜型分析及びCTL分析を実行した。IFN−γ ELISPOT分析の場合、p24特異的IFN−γ SFUがCFU依存的方式で増加した。しかし、p24タンパク質が注射されたマウスから得た脾臓細胞は、p24特異的IFN−γ SFUを誘導することができなかった(図8A)。これと同様に、それぞれの免疫化されたマウスから得た血清サンプルでp24特異的IgG2a抗体もCFU依存的方式で増加した。しかし、p24タンパク質が注射されたマウスの血清でp24特異的IgG2a抗体は、rBCG−pMyong2−p24注射グループのそれと比較してさらに低いレベルを示した(図8B)。
Claims (12)
- 配列番号1のアミノ酸配列で示すヒト免疫不全ウイルスタイプ1由来のp24タンパク質を発現する組換えマイコバクテリウム・ボビス(Mycobacterium bovis)BCGであって、前記p24タンパク質は、図2Aに開示されたpMyong2−p24ベクターシステムにより発現されることを特徴とする、組換えマイコバクテリウム・ボビスBCG。
- 前記p24タンパク質は、配列番号2の塩基配列で示すヒト免疫不全ウイルスタイプ1由来のGag遺伝子がコーディングされたものであることを特徴とする、請求項1に記載の組換えマイコバクテリウム・ボビスBCG。
- 前記マイコバクテリウム・ボビスBCGは、Tokyo 172菌株であることを特徴とする、請求項1に記載の組換えマイコバクテリウム・ボビスBCG。
- 請求項1〜3のいずれか一項に記載の組換えマイコバクテリウム・ボビスBCGを有効成分として含むことを特徴とする、HIV−1ワクチン組成物。
- 請求項1〜3のいずれか一項に記載の組換えマイコバクテリウム・ボビスBCGを有効成分として含むことを特徴とする、HIV感染症又はHIV及び結核菌同時感染症に対するワクチン組成物。
- 前記組換えマイコバクテリウム・ボビスBCGは、生きているものであることを特徴とする、請求項5に記載のワクチン組成物。
- 前記ワクチンは、さらに弱毒化(attenuation)されないものであることを特徴とする、請求項5に記載のワクチン組成物。
- 前記ワクチンは、プライム−ブーストワクチン接種法においてプライムワクチンで用いられるものであることを特徴とする、請求項5に記載のワクチン組成物。
- 前記感染症は、エイズ又は結核であることを特徴とする、請求項5に記載のワクチン組成物。
- 請求項1〜3のいずれか一項に記載の組換えマイコバクテリウム・ボビスBCGの治療学的有効量を有効成分として含むワクチン組成物を個体に投与してエイズ及び/又は結核を治療又は予防する方法。
- エイズ及び/又は結核の予防又は治療用ワクチンの製造のための請求項1〜3のいずれか一項に記載の組換えマイコバクテリウム・ボビスBCGの用途。
- エイズ及び/又は結核の予防又は治療のための請求項1〜3のいずれか一項に記載の組換えマイコバクテリウム・ボビスBCGの用途。
