本明細書で使用される、「a(1つの)」および「an(1つの)」および「the(その)」および同様の指示対象を示す用語は、他に本明細書で示されない限りまたは文脈によって明らかに矛盾とされない限り、単数形および複数形の両方を指す。

本明細書で提供されるポリマーに含まれる任意選択の非乳タンパク質モノマー成分は、任意の供給源由来の非乳タンパク質モノマーを含むことができる。そのような供給源の非限定的な例には、動物、植物、藻類、真菌、および微生物(microbe)が含まれる。
本明細書で提供されるポリマーは、所望の属性を有する。一部の実施形態では、所望の属性は、石油ベースのポリマーの属性である。
他の態様において、本発明は、石油由来のポリマー(即ち、1つまたは複数の石油由来のモノマーを含むポリマー)に1つまたは複数の組換え乳タンパク質モノマーを補うことによって生成された補足ポリマーを提供する。
別の態様において、本明細書では、本明細書で提供されるポリマーを産生するための方法が提供される。一部の実施形態では、本発明は:i)1つまたは複数の天然および/または組換え乳タンパク質モノマーを得るためのステップ;ii)1つまたは複数の天然および/または組換え乳タンパク質モノマーを重合して、ポリマーを得るためのステップ;およびiii)任意選択でポリマーを後処理するためのステップを含む方法を提供する。

方法はさらに、本明細書に開示される天然または組換え非乳タンパク質モノマーを、ポリマーの調製中の任意のステップで添加するステップを含むことができる。
ポリマーの後処理では、例えば、単離(即ち、精製;例えば、超遠心分離を介して)、脱水(例えば、噴霧乾燥、ローラー乾燥、流動床乾燥、凍結乾燥、エタノールによる乾燥、蒸発を介して)、およびコーティング(例えば、その他のポリマー、ヘパリン、ペクチン、アルギネートによる)が行われる。ポリマーを後処理するための方法は、当技術分野では公知である。
別の態様において、本明細書では、本明細書で提供されるポリマーを含む組成物が提供される。

組換えピキア・パストリス宿主細胞は、ピキア・パストリス(Komagaiaella phaffii)株BG12(Biogrammatics,Carlsbad,CA)を、酵母での発現のためにコドンで最適化されかつフレーム内でN末端分泌シグナルに融合させたウシ(Bos tauras)乳タンパク質(例えば、β−ラクトグロブリン)コード配列(サッカロマイセス・セレビシエ(Saccharomyces cerevisiae)のアルファ接合因子のプレまたはプレプロシグナルペプチド領域、天然の乳タンパク質シグナルペプチド、OST1シグナルペプチド)を含むベクターで形質転換することにより、生成された。乳タンパク質コード化配列は、プロモーター(構成的プロモーターpGAPまたはpPGK、またはメタノール誘発性プロモーターpAOX1)とターミネーター(tAOX1 pAシグナル)とによりフランキングされた。ベクターはさらに、細菌および酵母形質転換の選択に関する優性耐性マーカーと、細菌複製起点を含んでいた。ベクターを、ヒートショックプロトコールを使用してピキア・パストリス宿主細胞に形質転換して、乳タンパク質コード配列の一体化されたコピーを含む組換え宿主株を生成させた。形質転換体を、最小培地上に播き、30℃で48時間インキュベートした。最終形質転換のそれぞれからのクローンを、96ウェルブロックで300μLの緩衝最小培地(Buffered Minimal Medua)に接種し、600rpmで撹拌しながら30℃で、2時間または100時間超にわたりインキュベートした。試料を取り出し、組換え宿主細胞を遠心分離によってペレット化し、発酵上澄みを回収した。ELISAアッセイおよびSDS−PAGEゲル分析を、上澄み試料で行って、陽性の形質転換体を同定した。
組換えトリコデルマ・リーセイ宿主細胞は、トリコデルマ・リーセイ(Hypocrea jecorina)株Qm6を、ウシ(Bos taurus)β−ラクトグロブリンコード配列を含むベクターで形質転換することにより産生させた。ELISAアッセイおよびSDS−PAGEゲル分析を、上澄み試料で行って、陽性の形質転換体を同定した。
実施例1から3までのいずれかの組換えβ−ラクトグロブリンを、弱酸(例えば、5%酢酸)もしくは塩基(例えば、5%炭酸水素ナトリウム、水酸化ナトリウム、水酸化カリウム、水酸化カルシウム)と、単独で合わせ、またはα−ラクトアルブミンおよび/またはκ−カゼインおよび/または分泌された宿主細胞タンパク質と組み合わせて、2%から10重量%の間の組換えβ−ラクトグロブリンの最終濃度、2から8の間の最終pH、および10ms/cmから300mS/cmの間の最終伝導度にした。混合物を、少なくとも79℃で22分間加熱した。固体ポリマー凝集体を、遠心分離(例えば、4,000gで20分間)、濾過、溶媒抽出、クロマトグラフィー、またはその他の方法によって捕捉した。固体ポリマー凝集体を、依然として成形可能な所望の含水量まで乾燥させた。成形後、ポリマーをさらに乾燥させて、最終形態にする。
実施例1から4までのいずれかの組換えカゼインの1つまたは複数を、50mLの水に溶解し、そこに重曹(重炭酸ナトリウム;代替例:消石灰、リン酸カルシウム)5gを添加する。水の液滴を、白色カゼイン糊が得られるまで撹拌しながら、ゆっくり添加する。
実施例1から4までのいずれかの、大豆タンパク質単離物(SI)と組換えカゼイン(CAS)の1つまたは複数とを、10重量%のグリセロールおよび30重量%の水の存在下、同時回転2軸押出し機(例えば、Berstorff ZE 25(CL)40D)で、0%、10%、または30重量%のAl2O3と配合する。他の任意選択の成分には、TCPおよび/またはカップリング剤NZ12(例えば、1重量%で)が含まれる。押し出された配合物(compounds)(ペレットの形をとる)を、それぞれの含水量が10〜12%に達するまでコンディショニングした後(60℃で24時間)、ASTM引張り試験棒(24mm2の断面)に成型される。試験片を、例えばDEMAG D25 NC4を使用して、最適化された加工条件下で成型する。射出成型試料を、23℃および60%の相対湿度(RH)で少なくとも1週間コンディショニングする。
実施例1から4までのいずれかの組換えカゼインの1つまたは複数を、25mMの4−(2−ヒドロキシエチル)−1−ピペラジンエタンスルホン酸(HEPES)、および2mMのCaCl2を含有するpH7またはpH10,5の緩衝溶液(より高いpHは、アルギニン部位の鎖をリジン側鎖に加えて反応させる)に、27.5g/Lで溶解する。ゲニピン(クチナシ(Gardenia jasminoides)から抽出された架橋剤)を、74/26(w/w)のHEPES緩衝剤および無水エタノールの混合物に溶解して、ストック溶液200mMを得る。2つの溶液を合わせて、最終濃度5mM、10mM、または20mMのゲニピンを、同じ最終濃度のカゼインで得る(25g/L;ゲニピンの添加によって引き起こされた希釈は、HEPES緩衝剤および/または無水メタノールを添加することによって補正される)。混合物を、50℃で24時間インキュベートして、共有結合されたゲル母材を得る。
20%から40重量%の間の、実施例1から4までのいずれかの組換え乳清タンパク質の1つまたは複数を含む溶液を、60℃以上で加熱し、次いで合成コポリマー(例えば、ポリ酢酸ビニル[PVAC]、ポリビニルアルコール[PVA]、ポリビニルピロリドン[PVP])と合わせる。化学架橋剤(例えば、フェノール−ホルムアルデヒドオリゴマー、ポリマーメチレンビスフェニルジイソシアネート、グルタルアルデヒド)または酵素架橋剤を、添加する。あるいは、乳清タンパク質懸濁液を、120℃から150℃の間でおよび20MPaの圧力で5分間、ホットプレス機で成型し、その後、周囲温度に冷却し、引き続き、一晩、50℃の炉内でアニールして、所望の結合強度のポリマーを得る。
Claims (79)
- 乳タンパク質モノマー成分を含み、所望の属性を有する、ポリマー。
- 前記乳タンパク質モノマー成分は、1種の乳清タンパク質モノマーからなる請求項1に記載のポリマー。
- 前記1種の乳清タンパク質モノマーは、β−ラクトグロブリンモノマーである請求項2に記載のポリマー。
- 前記1種の乳清タンパク質モノマーは、α−ラクトアルブミンモノマーである請求項2に記載のポリマー。
- 前記乳タンパク質モノマー成分は、1種のカゼインモノマーからなる請求項1に記載のポリマー。
- 前記1種のカゼインモノマーは、κ−カゼインモノマーである請求項5に記載のポリマー。
- 前記1種のカゼインモノマーは、β−カゼインモノマーである請求項5に記載のポリマー。
- 前記1種のカゼインモノマーは、γ−カゼインモノマーである請求項5に記載のポリマー。
- 前記乳タンパク質モノマー成分は、2種の乳清タンパク質モノマーからなる請求項1に記載のポリマー。
- 前記2種の乳清タンパク質モノマーは、β−ラクトグロブリンモノマーおよびα−ラクトアルブミンモノマーからなる請求項9に記載のポリマー。
- 前記乳タンパク質モノマー成分は、2種のカゼインモノマーからなる請求項1に記載のポリマー。
- 前記2種のカゼインモノマーは、κ−カゼインモノマーおよびβ−カゼインモノマーからなる請求項11に記載のポリマー。
- 前記2種のカゼインモノマーは、κ−カゼインモノマーおよびγ−カゼインモノマーからなる請求項11に記載のポリマー。
- 前記2種のカゼインモノマーは、β−カゼインモノマーおよびγ−カゼインモノマーからなる請求項11に記載のポリマー。
