Abstract
Description
開示される本発明は、生物活性、特に骨伝導特性を示すポリオレフィン繊維に関し、この繊維は、移植後に繊維上又はこのような繊維を含む物品上での骨成長を促進するためにリン酸カルシウムのような生物活性無機粒子をその表面に有する高強度超高分子量ポリエチレン繊維のように、医療用インプラントにおいて使用するのに適している。また本発明は、このような骨伝導性繊維の製造方法と、可撓性組織アンカー、骨固定デバイス、及びテキスタイルベースの骨再生用足場のような前記繊維を含む医療用インプラントとに関する。
合成ポリマーから紡糸された繊維又はフィラメントで作製された編組ケーブル又は編物及び織物テキスタイル構築物のような繊維物品は、外科用縫合糸及びケーブル、人工靱帯及び腱、ヘルニアメッシュ、並びに可撓性組織アンカーなどにおいて、医療用デバイスの構成要素として種々の用途を見出している。
医療用インプラントにおいて使用するために骨伝導特性を示す繊維ベースの物品の製造方法に関する多数の刊行物(上記の引用文献を含む)にもかかわらず、その意図される医療用途において機能するために必要とされる機械特性を保存しながら、移植後の骨結合を可能にする生物活性表面を有する高強度ポリオレフィン繊維などのポリマー繊維が依然として必要とされている。本開示の目的は、このような繊維、このような繊維を含む医療用インプラント、及びこのような繊維の製造方法を提供することである。
・ a)ポリオレフィンと、
b)0.01〜10μmの粒径D50を有する、1〜30質量%のバイオセラミック粒子と、
c)最大で0.05質量%の残留紡糸溶媒と、
d)0〜3質量%の他の添加剤と
からなり、a)〜d)の合計が100質量%である組成物を含む繊維本体を有し、且つ
・ その表面に露出されたバイオセラミック粒子を有すると共に、生物活性を示す。
・ 5〜40dL/gの間の固有粘度(IV)を有するUHMWPE、
・ デカリン中に分散させたときに0.01〜10μmの粒径D50を有するバイオセラミック粒子、
・ 任意選択的に、他の添加剤、及び
・ 紡糸溶媒
を含む紡糸混合物を調製するステップと、
〇 紡糸混合物をマルチオリフィスダイプレートにより紡糸して、溶媒含有複合繊維を形成するステップと、
〇 少なくとも1回の延伸ステップにおいて溶媒含有複合繊維を延伸するステップと、
〇 繊維の延伸前、延伸中、延伸後に、溶媒含有複合繊維から紡糸溶媒を少なくとも部分的に除去して、その表面に露出されたバイオセラミック粒子を有すると共に、生物活性を示す、高強度UHMWPE複合繊維を得るステップと
を含む、高強度UHMWPE複合繊維の製造方法が記載される。
本開示の文脈において、繊維は、その幅及び厚さの横方向寸法よりもはるかに大きい長さ寸法を有する長尺体を指す。繊維は、リボン、テープ、又はフィラメントのような、種々の断面形状を有する長い連続糸であり得る。また繊維は、例えば、連続繊維を種々の長さに切断することにより作製された不連続長さを有していてもよく、当該技術分野では通常ステープル繊維と呼ばれる。繊維は、複数のフィラメントのように、複数の1つ以上のこのような要素を意味する。フィラメントは、一般に50μm未満の直径の略円形又は楕円形の断面を有し、通常は紡糸プロセスにより作製される(単一の)細い糸であると理解される。(マルチフィラメント)糸は、少なくとも5本のフィラメントのような複数のフィラメントを含み、このフィラメントは撚り合わせられていてもよい。高強度繊維は、ASTM D885Mに従う方法で決定したときに、少なくとも15cN/dtexの引張強さ又はテナシティを有する。

を含むゲル紡糸プロセスを用いて製造される、実施形態[21]の方法。
[方法]
・ IV:UHMWPEの固有粘度は、ASTM D1601(2004)の方法に従って、135℃でデカリン溶液において、溶解時間16時間、抗酸化剤として2g/l溶液の量のBHT(ブチルヒドロキシトルエン)を用いて、種々の濃度で測定された粘度をゼロ濃度まで外挿することによって決定される。
[バイオセラミック粒子の分散]
ヒドロキシアパタイト粉末(HA)は、Premier Biomaterials Ltd(PurAtiteTMHA;Ca10(PO4)6(OH)2;0.7μmの粒径D50を有する球体粒子であると示される)から入手した。ベータ−リン酸三カルシウム粉末(bTCP)は、Premier Biomaterials Ltd(βTCP1;Ca3(PO4)2;2.7μmの粒径D50を有する球体粒子であると示される)から入手した。
[・比較実験1]
デカリン中のIV23dl/gの7.7質量%のUHMWPE粉末のスラリーが210℃で加熱したツインスクリュー押出機に供給され、得られた溶液が、直径1mmの50個の孔を有するスピナレットを介して溶液フィラメントに紡糸され、このフィラメントが空隙に通され、水浴で冷却されて、ゲルフィラメントが形成されるゲル紡糸プロセスによって、マルチフィラメントUHMWPE糸を作製した。ゲルフィラメントをさらに熱風オーブンを通過させ、その間に水及びデカリンを蒸発させ、フィラメントを延伸させて、128dtexのタイターを有する50フィラメント糸を得た。
比較実験1と同様のプロセスを用いて複合糸を作製したが、ここでは、得られた複合マルチフィラメント糸において約10、15又は20質量%(UHMWPE及びバイオセラミック粒子の合計質量を基準として)の粒子含量を目標として、デカリンの一部を表1に記載される粒子分散体で置き換えることより、出発UHMWPEスラリーを作製した。紡糸プロセスは安定しており、24時間までの間、糸が破損することなく生成物を作製することができた(そして実験を中止した)。
国際公開第2012/076728号パンフレットのような、そしてバイオセラミックスについて上記したものと同様のゲル紡糸プロセスを用いて、20質量%の三酸化ビスマス粒子を含む複合糸を作製した。比較実験7及び実施例2について表2に記載される引張り特性は同等であり、両方の繊維は、ほぼ同じ体積%の無機粒子も含有する。
[・モルホロジー]
繊維の表面をSEMで研究した。図1及び図2において、いくつかのSEM顕微鏡写真は、例えば、実施例2〜4(図1A〜1B〜1C)及び実施例5〜6(図2A〜2B)を再現しており、ここで、バイオセラミック粒子は、撚った延伸糸の表面に明るい色の特徴物として見ることができる。前記特徴物の数と、繊維の製造に使用された粒子の量との間に明確な関係があるように見える。HAとbTCP粒子との間には有意な差違は見られない。一般に、繊維の表面層の粒子がポリエチレン中に完全には埋め込まれない、又はポリエチレンによって完全には被覆されず、これらが少なくとも部分的にその環境に露出されていることが示される。この結論は、SEM画像形成において加速電圧を2kVから20kVまで変更するさらなるSEM実験によって支持された;恐らくより大きい侵入深さに起因して、10及び20kVにおいてより明確な白色特徴物が明らかにされた。SEM技術による典型的な侵入深さは、使用条件に応じて約1μmであることに注意されたい。
フィラメントの表面層内又は表面層に存在する粒子は、異なる表面粗度のフィラメント及び糸をもたらし得るので、複合繊維及び純粋なUHMWPE繊維の摩擦係数(CoF)を測定した。CoFは無次元スカラーであり、接触域を動かし続けるのに必要な力と、それを押し付けるのに必要とされる力との比を説明する;CoFは、試験材料に応じて0から1超までの範囲である。表3にまとめられた結果は、バイオセラミック粒子の添加が糸対糸(Y−Y)のCoFを増大させることを示し、より粗い表面が示唆される。このような傾向は、糸対固体(Y−S)のCoF値の測定についてはあまり明白ではない。10質量%のHA負荷では、非負荷繊維に対するY−SのCoFの相違は非常に小さく、ほんのわずかな粒子が表面に存在することが示唆される。対照的に、20質量%のHA/bTCPを含有する複合繊維の場合、Y−SのCoF値は著しく増大した。
[編組及び編物]
比較実験1及び実施例2〜6で得られた糸から、Herzog TT78編組装置、及び各ストランドに対して単一の糸、並びに1cm当たり15ステッチのステッチレベルを用いて、16本のストランドを含む編組構築物を作製した。得られた編組体は、標準サイズのUSP2縫合糸と同様のサイズを有した。
[・プラズマエッチング]
EMPA(Swiss Federal Laboratories for Material Science and Technology)において、編組サンプル及び編物サンプルの繊維の外側表面をプラズマエッチングした。パイロットウェブコーターにおいて、リール・トー・リール(reel−to−reel)システムを用いて処理を実施した。繊維物品をRF駆動反応器(幅65cm、直径59cm)内のドラム電極に巻き取った。バイオセラミック負荷UHMWPEサンプルのプラズマエッチングの前に、ポリオレフィンホイル(LDPE及びPP)を用いることにより、適切なエッチング速度を推定した。続いてサンプルに適用した条件(40分、Ar/O2プラズマ、160/40sccm、1000W、1mbar)は、約25nm/分のエッチング速度をもたらすことが見出された。サンプルは、主に、ドラム電極と接触していない面でエッチングされ得ることが予測された。
サンプル表面のエッチングの前及び後に、SEMにより編組体を調べた。編組体を両面接着剤炭素テープでSEMサンプルホルダーに固定し、導電性炭素層で被覆した。FEI Versa 3D FEG SEMにおいて5kVで画像形成を行った。図4には、15質量%のHAが負荷された実施例3の繊維から作製された編組体について、プラズマエッチングの前(図4A)及び後(図4B)の顕微鏡写真の代表的な例が示される。他の編組体についてもプラズマエッチングの前及び後に同様の結果が得られた。エッチング後により多くの粒子が白色スポットとして見えるようである;粒子はより鋭いエッジを有し、より大きいサイズを有し、表面からより大きく突出するように見える。エッチングステップは約1μmの厚さの表面層からのポリエチレンの除去を生じているはずなので、これは、SEMも約1μmの侵入深さを有し、プラズマエッチングの前でもこのような層内のバイオセラミック粒子の少なくとも一部を示すことを確認する。図4Bのエッチングされたサンプルは、恐らくエッチング除去されたポリエチレンに起因して、より細線維状の表面を示すことに注意されたい。
複合バイオセラミック/UHMWPE繊維の化学組成を、XPS及びAFMによって特徴付けした。AFMは、硬い(バイオセラミック)相と、比較的柔らかい(ポリエチレン)相との明確に区別を行うことができるので、繊維の表面に露出したバイオセラミックスの量の直接的な表示を提供する。XPSは、上部層(約7nmと定義される)中のカルシウム及びリンの原子%を測定するために使用される。
アリザリンレッド染色は骨ミネラルを染色するための周知の方法であるため、これを使用してサンプル中のバイオセラミック粒子を可視化した;この色素はカルシウムに対する高い親和性を有する。純粋なUHMWPE繊維(比較実験1)、及び複合15/85HA/UHMWPE繊維(実施例3)から作製された編物構造を、無機材料と親和性があるが、UHMWPEのようなポリオレフィンとは親和性がない赤色着色剤アリザリンレッド溶液で処理した。図7(それぞれ、図7A及び図7B)に示されるように、純粋なUHMWPE繊維から作製された編物構造は実際に注目すべき着色を示さなかったが、実施例3の繊維から作製された構造はいくらかの着色を示した。図7Cには、最初に構造をプラズマエッチングし、次にアリザリンレッドにさらした後に、実施例3の複合繊維から作製された編物構造が示される。この場合、事実上、構造全体が赤色に着色され、プラズマエッチング後に着色剤との相互作用のためにはるかに多くのHA粒子が利用可能であったことが示される。
表7には、種々の材料において28日間培養したhMSCのPresto blueアッセイを用いた細胞生存率研究の結果が示される。一般に、時間と共に、そしてバイオセラミック負荷の増大と共に、細胞生存率の増大(これは、細胞増殖の表示である)が観察されたが、種々のサンプルの結果はややばらつきがあるように見える。
hMSCと共に28日間培養した後、SEM−EDXを用いて編物サンプルも調べた。走査電子顕微鏡法により、バイオセラミック負荷複合繊維サンプルは、細胞生存率データと一致して、非負荷繊維サンプルよりも多くの細胞及び細胞マトリックスによって被覆されることが示された。材料フィラメント間を架橋する長い紡錘状細胞プロセス(フィロポディア)に代表される細胞伸展も、非負荷対照に対して、複合繊維サンプルによって増強されると思われる(比較実験1(図9A〜B)及び実施例3(図9C〜D)の繊維に基づいたサンプル、並びに実施例3の繊維(図9E〜F)に基づいたエッチングサンプルの画像を表す図9A〜Fを参照;倍率2)。また15質量%のHAプラズマエッチングサンプルなどのバイオセラミック負荷サンプルにおいて、電子密度の高い小結節(明るい微粒子様の特徴物)がより広範囲に観察され、これらは、実験の初めは存在せず、繊維に負荷されたバイオセラミック粒子とはモルホロジー的に異なり(図2及び図4を参照)、培養期間中にバイオミネラリゼーションが生じたことが示唆される。
Claims (15)
- 高強度ポリオレフィン複合繊維であって、前記繊維が、
a)ポリオレフィンと、
b)0.01〜10μmの粒径D50を有する、1〜30質量%のバイオセラミック粒子と、
c)最大で0.05質量%の残留紡糸溶媒と、
d)任意選択的に、0〜3質量%の他の添加剤と
からなり、a)〜d)の合計が100質量%である組成物を含む繊維本体を有し、且つ
前記繊維がその表面に露出されたバイオセラミック粒子を有すると共に、生物活性を示す、高強度ポリオレフィン複合繊維。 - 前記ポリオレフィンが超高分子量ポリエチレン(UHMWPE)である、請求項1に記載の高強度ポリオレフィン複合繊維。
- 前記バイオセラミック粒子がリン酸カルシウム、生物活性ガラス、又はこれらの混合物である、請求項1又は2に記載の高強度ポリオレフィン複合繊維。
- 前記バイオセラミック粒子が実質的に球体、楕円体又は立方体であり、最大で5、好ましくは最大で4、3、又は2のアスペクト比を有する、請求項1〜3のいずれか一項に記載の高強度ポリオレフィン複合繊維。
- 前記組成物が、少なくとも2、3、4又は5質量%、且つ最大で22、20、18、16、14、12又は10質量%のバイオセラミック粒子を含有する、請求項1〜4のいずれか一項に記載の高強度ポリオレフィン複合繊維。
- 前記繊維本体が実質的に前記組成物からなる、請求項1〜5のいずれか一項に記載の高強度ポリオレフィン複合繊維。
- 前記繊維が10〜2000dtexのタイターを有するマルチフィラメント糸であり、最大で50、30、25、20、15、12、又は10μm、且つ少なくとも5又は6μmの繊維直径を有するフィラメントを含む、請求項1〜6のいずれか一項に記載の高強度ポリオレフィン複合繊維。
- 前記繊維が少なくとも20cN/dtex、好ましくは少なくとも25、28、又は30cN/dtexのテナシティを有する、請求項1〜7のいずれか一項に記載の高強度ポリオレフィン複合繊維。
- 前記繊維が、XPS及びAFM表面分析で決定したときに0.1〜25面積%、好ましくは0.5〜6面積%である露出バイオセラミック粒子による表面被覆率を有する、請求項1〜8のいずれか一項に記載の高強度ポリオレフィン複合繊維。
- 前記繊維が、バイオセラミック粒子を含有しない対応する繊維と比較して、ALPアッセイにおいて28日後に少なくとも25%高いALP活性を示し、好ましくは、ALPアッセイにおいて28日後に少なくとも30、40、50、60、70、80、90、100、150又はさらに200%高いALP活性示す、請求項1〜9のいずれか一項に記載の高強度ポリオレフィン複合繊維。
- 請求項2〜10のいずれか一項に記載の高強度UHMWPE複合繊維をゲル紡糸プロセスによって製造する方法であって、
・ a)5〜40dL/gの間のIVを有し、任意選択的に、前記複合繊維について上記でさらに定義されたようなUHMWPE、
b)デカリン中に分散させたときに0.01〜10μmの粒径D50を有し、任意選択的に、前記複合繊維について上記でさらに定義されたようなバイオセラミック粒子、
c)任意選択的に、前記複合繊維について上記でさらに定義されたような他の添加剤、及び
d)紡糸溶媒
を含む紡糸混合物を調製するステップと、
・ 前記紡糸混合物をマルチオリフィスダイプレートにより紡糸して、溶媒含有複合繊維を形成するステップと、
・ 少なくとも1回の延伸ステップにおいて前記溶媒含有複合繊維を延伸するステップと、
・ 前記繊維の延伸前、延伸中、延伸後に、前記溶媒含有複合繊維から前記紡糸溶媒を少なくとも部分的に除去するステップと
を含む、方法。 - 好ましくは酸素含有雰囲気中のプラズマエッチングにより前記繊維の表面を後処理するステップをさらに含む、請求項11に記載の方法。
- 請求項1〜10のいずれか一項に記載の複合繊維を含む繊維物品。
- 骨折又は靱帯若しくは腱の損傷を修復するための永久的な高強度整形外科用インプラントのように、医療用インプラントの構成要素として、又は医療用インプラントとしての、請求項1〜10のいずれか一項に記載の複合繊維又は請求項13に記載の繊維物品の使用。
- 請求項1〜10のいずれか一項に記載の複合繊維又は請求項13に記載の繊維物品を含む生物医学的デバイス又はインプラント。
