Abstract
Description
本発明は、化学製薬の分野に属し、薬物の製造における(5R)−5−ヒドロキシトリプトリドの使用に関し、具体的に、エイズの異常な免疫活性化またはエイズの免疫再構築不全を治療および/または予防する薬物の製造における(5R)−5−ヒドロキシトリプトリドの使用に関する。
タイワンクロヅルは、衛矛科クロヅル属の木質藤本の植物で、山地林縁の陰湿地に生え、長江流域より南の各地および西南地区に分布し、その化学成分は主にジテルペン、トリテルペン、セスキテルペン、アルカロイドなどである。最近の20年の研究では、タイワンクロヅルは抗炎症、免疫阻害、避妊、抗腫瘍、抗菌などの活性を有し、特に免疫阻害作用においては、タイワンクロヅルにおけるトリプトリド(triptolide)が強い薬理的活性を有することを表明した。しかし、トリプトリド(triptolide)の毒性・副作用によって、臨床におけるその使用が制限される。中国特許1223595号(特許文献1)の発明者は、トリプトリドの構造と効果の関係に対する鋭意な研究により、トリプトリド(triptolide)の構造に対して改変と修飾を行い、新規な低毒性のトリプトリド誘導体を公開し、当該誘導体は抗炎症免疫阻害剤として自己免疫性疾患、感染性疾患などの予防と治療に使用され、特に(5R)−5−ヒドロキシトリプトリド(LLDT−8、T8と略され、その構造式は下記に示される)は、複数の体外試験システムおよび体内実験動物モデルの研究において、幅広い抗炎症免疫阻害活性を有し、高効率・低毒性で、優れた治療指数を有することが証明された。
本発明の解決しょうとする技術的課題は、既存技術におけるエイズの異常な免疫活性化およびエイズの免疫再構築不全に対する薬物が欠けているという欠陥を克服し、薬物の製造、特にエイズの異常な免疫活性化またはエイズの免疫再構築不全を治療および/または予防する薬物の製造における(5R)−5−ヒドロキシトリプトリドの使用を提供することである。
以下、実施例の形によってさらに本発明を説明するが、これによって本発明を記載された実施例の範囲内に限定するわけではない。以下の実施例において、具体的な条件が記載されていない実験方法は、通常の方法および条件、あるいは商品の説明書に従って選ばれる。
HEK293T細胞(ATCCから購入)にNF−κB駆動含有ルシフェラーゼ発現プラスミド(p3κB−luc、100ng)および内部参照プラスミドpBgal(生物在線www.bioon.com.cnから購入)(pSV−β−Galactosidase Control Vector、β−ガラクトシダーゼ発現プラスミド、5ng)を形質移入させた。24時間後、細胞をTNF−α(20ng/ml)で処理するか、処理せず、そして同時にT8化合物(100nM)を入れたか、入れなかった。さらに24時間後、細胞を収集し、レポーター分解緩衝液(Promegaから購入)で分解させた。ルシフェラーゼアッセイシステム(Promega)によってルシフェラーゼ活性を検出し、β−Gloアッセイシステム(Promega)によってβ−ガラクトシダーゼ活性を検出し、T8によるNF−κB活性に対する阻害作用を分析した。
健常者の末梢血単核球(PBMC)(ATCCから購入)をIL−2(20IU)を含有するRPMI−1640/10%FBS血清の培地で培養し、CFSE(カルボキシフルオレセインジアセテートスクシン イミジルエステル)(10nM)で37℃で10分間染色し、上記培地で2回洗浄した後、PHA−P(フィトヘマグルチニン−P)(5μg/ml)で細胞の増殖を刺激し、同時にT8化合物(100nM)を入れるか、入れずに、48時間培養した後、細胞を収集し、抗ヒトCD4フローサイトメトリー用抗体(Invitrogenから購入)で染色し(4℃、30分間)、フローサイトメトリーによってCD4+T細胞を選択してCFSE染色強度を検出して細胞の増殖を分析した。
健常者の末梢血単核球(PBMC)1×107をPMA(プロピレングリコールメチルエーテルアセタート)(20nM)およびIonomycin(イオノマイシン)(1μM)で37℃で刺激し、同時にT8化合物(100nM)を入れたか、入れなかった。48時間後、抗ヒトCD4、CD38、HLA−DR細胞フローサイトメトリー用抗体(Invitrogenから購入)を入れて染色し、フローサイトメトリーによってCD4+T細胞を選択して細胞表面分子CD38、HLA−DRの発現を検出した。
(5R)−5−ヒドロキシトリプトリド(T8)およびその対照品(トリプトリド)は、上海医薬グループから提供され、白色の粉末で、純度>99%で、4℃で保存して使用に備えた。
HEK293Tを10%ウシ胎児血清、100U/mLペニシリンおよび100μg/mLストレプトマイシンを含有する完全DMEM培地で培養した。
MTT比色法による細胞毒性の検出:HEK293T細胞を96ウェルプレート(1×104/100μL/ウェル)に敷き、100μLの異なる濃度の化合物(化合物の濃度は図面を参照する)を入れ、各希釈度に重複ウェルを3つずつ設け、同時に化合物を含有しない対照ウェルを設けた。37℃、5%CO2細胞インキュベーターにおいて72時間インキュベートした。各ウェルから上清を100μL吸いて捨て、20μLのMTT溶液(5mg/mL)を入れ、37℃、5%CO2細胞インキュベーターにおいて4時間インキュベートした後、各ウェルに100μLのホルマザン溶解液を入れて一晩インキュベートし、通常の光学顕微鏡による観察においてホルマザンが全部溶解したら、595nmにおける吸光度を測定した。CC50値(50% Cytotoxic Concentration、50%細胞毒性濃度)、すなわち、50%のHEK293T細胞に毒性を生成した時の実験薬物濃度を算出した。
HEK293T細胞で2種類の化合物の細胞毒性を検出したところ、(5R)−5−ヒドロキシトリプトリド(T8)によるHEK293T細胞に対するCC50(50% Cytotoxic Concentration、50%細胞毒性濃度)値は約435ng/mlで、対照品(トリプトリド)によるHEK293T細胞に対するCC50値は約17ng/mlであった。これから、トリプトリドと比べ、T8がより低い細胞毒性を示したことがわかる(詳細は図4を参照する)。
HEK293Tを10%ウシ胎児血清、100U/mLペニシリンおよび100μg/mLストレプトマイシンを含有する完全DMEM培地で培養した。
NF−κBレポーター遺伝子プラスミドp3κB−lucをHEK293T細胞に形質移入して発現させ、同時にpSV−β−Galactosidase Control Vectorを形質移入の内部参照として共形質移入し、TNF−αで処理してNF−κBの活性化を誘導し、T8およびトリプトリドを入れ、ルシフェラーゼ活性を検出することによってNF−κBレポーター遺伝子の発現を示し、T8およびトリプトリドによるTNF−αによって誘導されたNF−κB活性化作用に対する影響を評価した。
図5から、(5R)−5−ヒドロキシトリプトリド(T8)およびその対照品(トリプトリド)はいずれも有効にバックグラウンドNF−κBシグナル経路の活性を阻害し、両者は活性が相当し、バックグラウンドNF−kB活性に対する阻害の有意差分析では、P=0.1で、有意差がなかったことがわかる。(5R)−5−ヒドロキシトリプトリド(T8)およびその対照品(トリプトリド)は有効にTNF−αによって活性化されたNF−κBシグナル経路の活性を阻害し、両者は活性が相当し、TNF−αによって活性化されたNF−kB活性に対する阻害の有意差分析では、P=0.056で、有意差がなかったことがわかる。
末梢血単核球(Peripheral blood mononuclear cell、PBMCs)は上海市長海医院から購入され、細胞は10%ウシ胎児血清、100U/mLペニシリン、100μg/mLストレプトマイシン、20IU/mLのIL−2(R&D)を含有する完全RPMI−1640培地で培養した。
新しく分離された健常者の末梢血細胞PBMC(3×107)をIL−2(20IU/mL)を含有する細胞培地で培養した。PMA(100nM)およびイオノマイシン(1μM)で72時間刺激し、同時に被験化合物(100ng/mL)を入れた。FACS緩衝液で2回洗浄し、抗CD3−PerCP、CD4−PE、CD38−PE−Cy7、HLA−DR−APC抗体を入れ、4℃で30分間染色した。FACS緩衝液で2回洗浄し、FACSによって上記分子の発現を検出した。
細胞活性化指標であるCD38およびHLA−DRの発現を検出することにより、(5R)−5−ヒドロキシトリプトリド(T8)およびその対照品(トリプトリド)が顕著にPMA+イオノマイシンまたは抗CD3/CD28抗体によって活性化されたCD4+T細胞亜集団の免疫活性化を阻害することができることが証明された。
末梢血単核球(Peripheral blood mononuclear cell、PBMCs)は上海市長海医院から購入され、細胞は10%ウシ胎児血清、100U/mLペニシリン、100μg/mLストレプトマイシン、20IU/mLのIL−2(R&D)を含有する完全RPMI−1640培地で培養した。細胞を5μg/mLのフィトヘマグルチニン(Phytohemagglutinin−P、PHA−P)を含有する完全培地で72時間培養し、CD4+T細胞の増殖を誘導した。
新しく分離された健常者の末梢血細胞PBMC(1×107)をIL−2(20IU/mL)を含有する細胞培地で培養した。CFSE(10nM)、37℃で10分間染色した。培地を2回洗浄した後、PHA−P(5μg/mL)を入れ、同時に被験化合物T8またはトリプトリド(100ng/mL)を入れた。37℃で72時間後、細胞を収集し、抗CD4−PE抗体で4℃で30分間染色し、FACS緩衝液で2回洗浄し、フローサイトメトリーによって細胞の増殖を分析した。
図7から、(5R)−5−ヒドロキシトリプトリド(T8)およびその対照品(トリプトリド)が顕著に健常者の末梢血CD4+T細胞の増殖を阻害することができ、かつ健常者の末梢血CD4+ T細胞の増殖に対する阻害効果が相当することがわかる。
Claims (9)
- エイズの異常な免疫活性化、またはエイズの異常な免疫活性化に関連する免疫再構築不全を治療および/または予防する薬物の製造における(5R)−5−ヒドロキシトリプトリドの使用。
- 前記のエイズの異常な免疫活性化はNF−κBに関連するエイズの異常な免疫活性化であることを特徴とする請求項1に記載の使用。
- 前記のエイズの異常な免疫活性化はエイズの慢性の異常な免疫活性化であることを特徴とする請求項1または2に記載の使用。
- 前記の薬物の形態は錠剤であることを特徴とする請求項1に記載の使用。
- 前記の(5R)−5−ヒドロキシトリプトリドは前記薬物の有効成分の一つまたは唯一の有効成分であることを特徴とする請求項1〜4の何れか一項に記載の使用。
- NF−κBシグナル経路阻害薬剤の製造における(5R)−5−ヒドロキシトリプトリドの使用。
- 前記の(5R)−5−ヒドロキシトリプトリドは前記薬剤の有効成分の一つまたは唯一の有効成分であることを特徴とする請求項6に記載の使用。
- Tリンパ球活性阻害薬剤の製造における(5R)−5−ヒドロキシトリプトリドの使用。
- 前記の(5R)−5−ヒドロキシトリプトリドは前記薬剤の有効成分の一つまたは唯一の有効成分であることを特徴とする請求項8に記載の使用。
