Abstract
Description
遺伝子物質を標的に送達するために、改善されたウイルスベクターベースのシステムの必要性がある。
ここで、Pepはオリゴペプチドなどのペプチドであり、RはOH、CH3 、またはコレステロール基であり、mは1〜20の範囲であり、nは1〜100の範囲であり、oは1〜10の範囲である。 好ましい実施態様において、ペプチドは、アルギニン(R)、リシン(K)、ヒスチジン(H)、グルタミン酸(E)、アスパラギン酸(D)、システイン(C)からなる群から選択される少なくとも2つ、または少なくとも3つのアミノ酸を含む。 現在の技術で使えるオリゴペプチド誘導体化PBAEの記述についてはWO2014/136100およびWO2016/116887も参照。されたい。 システインは、ペプチドのポリマーへの共有結合点を提供するために含めることができ、pH7でわずかな負の電荷を運ぶが、正に帯電したアミノ酸と一緒に使用しても電荷を著しく変化させない。 代表的なペプチドは、CRRR(配列番号1)、 CHHH(配列番号2)、CKKK(配列番号3)、CEEE(配列番号4)、CDDD(配列番号5)である 。
1.ポリマーまたはポリマー混合物で被覆されたウイルスベクターからなるナノ粒子で、ウイルスベクターは導入遺伝子を含み、そのナノ粒子は真核細胞に導入遺伝子を送達する手段として機能する。

現在の技術では、ウイルスベクターが「外膜タンパク質の欠如」とは、ベクター外皮の外皮、特に脂質二層膜は、外膜ウイルスの表面に典型的に見られる如何なるタンパク質をも欠いていることを意味します。それらは、例えば、Envタンパク質複合体、またはHIV gp120−gp41複合体によって例示される複合体またはウイルスエンベロープ糖タンパク質、水疱性口内炎ウイルス糖タンパク質(VSV−G)、インフルエンザウイルスヘマグルチニン、麻疹ウイルスHおよびFタンパク質、および野生型または修飾シンドビスウイルス外膜タンパク質などの自然発生のまたは修飾した形態におけるものである。このようなタンパク質は、現在の技術のベクターナノ粒子が欠けている場合、もしあれば、ベクター外皮の脂質二層から外側に向かって投影し、外側のポリマー層と脂質二層の間に必要な安定した接触を妨げます。
ここで、Pepはオリゴペプチドであり、RはOH、CH3 、またはコレステロール、リン脂質分子、または脂肪酸またはアシル鎖であり、mは好ましくは1〜20の範囲であり、nは好ましくは1〜100の範囲であり、oは1〜10の範囲である。 他のポリマーおよびその互いのおよびPBAEとの混合物は、ウイルスベクター粒子をカプセル化するために使用することができる。単層または多重層脂質リポソームまたは界面活性剤および/または脂質分子を含むミセル様構造のような形態におけるなどの脂質を用いたさらなるコーティング機構も同様に利用することができる。 このようなコーティングは、エンベロープタンパク質を欠いているウイルスベクター粒子(「はげた」ベクター)をカプセル化するのに特に有用である。
レトロウイルスは、宿主ゲノムに安定した方法で統合する能力を有するRNAウイルスである。 好ましい実施態様において、遺伝子送達手段におけるレトロウイルスベクターはレンチウイルスベクターである。 レンチウイルスベクターは、レトロウイルス科の遅いウイルスの属を表すレンチウスに由来する。レンチウスは、ヒト免疫不全ウイルス(HIV)、シミアン免疫不全ウイルス(SIV)、ウマ感染性脳炎ウイルス(EIAV)、カプリン関節炎脳炎ウイルス(CAEV)、ウシ免疫不全ウイルス(BIV)、ネコ免疫不全ウイルス(FIV)などを含む。レンチウイルスは、分裂細胞と非分裂細胞のゲノムに統合することができる。さらに、ホストに無期限に保持され、継続的に複製される。本明細書に記載されているいくつかの実施態様では、レンチウイルスベクターはカプシドを有するが、外膜タンパク質を欠いている。 好ましくは、ベクターは、意図された宿主または処置対象の細胞において非複製である。好ましくは、ベクターは、非標的細胞の遺伝子発現を伝達または変化させない。 ましくは、ベクターは病原性を引き起こす遺伝子、特に標的でない細胞の意図しない細胞死を誘発するものとして現れる病原性を欠いている。
トランスフェクト細胞にポリマーカプセル化された手段を使用する以前の技術は、PBAEなどのポリマーで直接コーティングされたプラスミドまたは他の核酸分子に関与していた。ここのような技術の中には、核酸分子を用いてトランスフェクションを達成しているものもある (US2016/0145348A1, Mangraviti et al.2015, Anderson et al.2004,WO2016/116887)。pH7におけるPBAEポリマーの安定性の低下が報告されている (Lynn et al. 2000)。このような処方物は、細胞毒性を引き起こし、ポリマーコーティングプラスミドまたは他の核酸分子に曝露すると細胞死を引き起こすことがある。 したがって、in vivoまたはin vitro細胞に投与されるPBAEカプセル化ベクターの用量は、毒性作用を避けるために、有用な形質導入に必要な最小限にとどめるべきである。しかしながら、PBAEカプセル化ウイルスベクターは、低pHでの分解に有利に抵抗する。
ナノ粒子ベクター送達手段は、治療を必要とする特定の標的細胞や組織、またはベクターによるタンパク質発現が望まれる標的をターゲットにすることができる。標的部分の別の実施態様を図3に示す。この中で、送達手段は、ポリマー表面に共有または非共有的に結合された標的部分ーを含有する。標的部分は、in vivoまたはin vitro のいずれかの特定の細胞または細胞クラスを標的とすることができる。標的部分の例としては、タンパク質(例えば、ヒト抗体、マウス抗体、ラクダ類抗体、サメ抗体を含む抗体、単鎖抗体、ナノボディ、ジアボディ、抗体の抗原結合フラグメント)、ペプチド、アプタマーやオリゴヌクレオチドなどの核酸、細胞表面受容体または細胞外マトリックスの成分に結合するリガンドなどを含み得る。
本願明細書に記載された遺伝子送達手段のウイルスベクターには、細胞に入った後に細胞内で発現し、発現生成物を生成する1つ以上の導入遺伝子を含有する。 ウイルスベクターは、1つ、2つ、3つ、またはそれ以上の導入遺伝子を含有し得る。 導入遺伝子は、治療タンパク質またはマーカータンパク質を含む、自然発生または人工的な(すなわち、組換えの)タンパク質、ポリペプチド、またはペプチドをコードする核酸分子である。このようなベクター発現タンパク質は、好ましくは、それらが発現される対象において治療的価値を有し、したがって治療タンパク質である。 治療タンパク質の1つのタイプは、キメラ抗原受容体(CAR)である。本願明細書において説明する技術で使用するCARは、T細胞受容体などの組換え抗原受容体であり、結合するためにペプチドプロセシングやHLA発現を必要としない細胞表面抗原に対する結合特異性を持つ。 抗原結合部は、抗体に由来するscFV、ライブラリーから選択されたFabに由来するscFV、または細胞表面受容体の天然に存在するリガンドであり得る。 それは、ナノボディ(VHH)でもあり得る。CAR分子の残りの部分は、抗原結合ドメインを細胞内信号導入ドメインと結合する。具体的には、この技術のCARは、細胞外抗原結合ドメイン、ヒンジおよび膜貫通ドメイン、および細胞内信号伝達ドメイン(CD3zeta、および/またはCD28または41−BB/OX40)の各ドメインの1つ以上を含む。 遺伝子送達手段での使用に適したCARの詳細およびそれらをレンチウイルスベクターにコードする配列は、WO2016/012623A1に記載されている。
本技術の遺伝子送達手段は、レンチウイルスベクターなどのレトロウイルスベクターによって、哺乳類細胞やヒト細胞などの関心のある細胞に遺伝子物質を効果的に形質導入する必要があるあらゆる適用に使用できる。 送達手段は高効率で細胞を形質導入し、低毒性と、低免疫原性を示す。 それらは、宿主細胞のゲノムに遺伝物質を永続的に統合することができ、該システムを細胞とその子孫の永続的なラベリングに特に適するようにする。 この手段は in vivo でも高い特異性、低い毒性、および高い形質導入能力を発揮するため、ヒトおよび動物における治療および動物モデルの両方における遺伝子移動および遺伝子治療適用に特に適しており、異なる生物学的機能の根底にある分子機構の調査とその治療への翻訳を可能にする。 しかし、ポリマーカプセル化ウイルスベクターナノ粒子の一部の調製は、分割細胞を優先的に形質導入し、非分割細胞を形質導入しないなど、特定の細胞タイプを形質導入することに限定される場合がある。
これらの活動には、関心のある遺伝子や複数の遺伝子を正確かつ安定的に挿入することが不可欠であり、この技術の手段によって一貫して達成することができる。 この手段は、変換細胞(チャイニーズハムスター卵巣(CHO)細胞、HEK293細胞)や原発細胞などの、製造目的に使用される細胞に元々存在しない遺伝子を挿入するために使用し得る。 あるいは、これらの細胞に存在する特定の遺伝子(免疫原性の一般的な原因であるタンパク質糖化に関与する遺伝子など)を「ノックアウト」するために使用することもできる。これらの手段は、例えば、家畜に影響を及ぼす疾病の予防、処置、治癒するために、あるいは成長と成熟を加速させることにより、経済的価値を向上させるために使用し得る。
細胞形質導入研究に用いるポリマー被覆レンチウイルスベクターは、以下の材料と方法を用いて作られた。
材料
伝達ベクタープラスミドは、pARA−CMV−GFPまたはpARA−CMV−mCherryまたはpARAまたはpAra−hUBC−CD19であった。
プロウイルス用にコードされたカナマイシン耐性プラスミド(HIV−1、株NL4−3由来の非病原性および非複製的組換えプロウイルスDNA)であり、そこで発現カセットがクローンされた。 挿入物は、導入遺伝子、導入遺伝子発現のためのプロモータ、および導入遺伝子発現を増加させ、レンチウイルスベクターが非有糸分裂を含むすべての細胞型を形質導入できるようにするために追加された配列を含んでいた。コード配列は、緑色蛍光タンパク質(GFP)またはmCherry(赤色蛍光タンパク質)またはCD19をコードする遺伝子に対応していた。 プロモータはヒトユビキチンプロモータまたはCMVプロモータであった。エンハンサー配列がなく、ユビキタスな方法で高いレベルで遺伝子発現を促進した。 非符号化シーケンスおよび発現信号は、5’LTR(U3−R−U5)のためのシスアクティブ素子全体と3’LTRのために削除された素子とを有するロング端末リピート配列(LTR)に対応し、それ故にプロモータ領域(ΔU3−R−U5)を欠いていた。 転写および統合実験では、エンカプシデイション配列(SDおよび5’Gag)、ベクターの核転座のための中央ポリ尿トラクト/中央終端サイト、およびBGHポリアデニル化サイトが追加された。
LV293細胞は、LVmax生産培地 (Gibco, A3583401) 1000mL中の2x3000mLのエルレンマイヤーフラスコ (Corning, 431252) 中、5x105細胞/mLで播種された。2つのエルレンマイヤーフラスコは、加湿8%CO2の下で37℃、65rpmで培養された。播種の翌日、過渡トランスフェクションが実施された。PEIPro核酸導入試薬(PolyPlus、115−010)は、転送ベクタープラスミド(pARA-CMV-GFPまたはpARA-CMV-mCherryまたはpARAまたはpAra-HubC-CD19)およびパッケージングプラスミド(pARA-Pack)と混合された。室温で培養した後、混合したPEIPro/プラスミドを細胞株に一滴ずつ添加し、加湿8%CO2の下で37℃、65rpmで培養した。3日目に、レンチベクター産生は5mM最終濃度で酪酸ナトリウムによって刺激された。バルク混合物は、加湿された8%CO2の下で37℃、65rpmで24時間培養した。5および0,5μm(Pall、609−122U)の深層ろ過による清澄化後、DNase処理のために室温で1時間培養した。
ポリ(β−アミノエステル)ポリマーは、Dostaらと同様の2段階手順に従って合成された。5−アミノ−1−ペンタノール(Sigma-Aldrich 3.9 g, 38 mmol)と1−ヘキシルアミン(Sigma-Aldrich 3.9 g, 38 mmol)を丸底フラスコで混合し、ジアクリレートとアミンモノマーとの2.1:1モル比で1,4−ブタンジオールジアクリレート(Aldrich 18 g, 82 mmol)と結合させた。混合物を20時間撹拌しながら90℃に加熱した。その後、室温まで冷却して淡黄色の粘性固体油を形成し、さらなる使用まで−20℃で貯蔵した。
15kDaのポリグルタミン酸(PGA)(Alamanda Polymers)をMES緩衝液pH6.0に20mg・ml−1になるように溶解した後、浴超音波装置で10分間超音波照射した。PGA溶液1.8mLをエチル−N’−(3−ジメチルアミノプロピル)カルボジイミド−HCl(4mg・ml−1,16 equiv.)および0.9mlのスルホ−NHS(2,75w/w EDC)の両方ともMES緩衝液中pH6.0に添加し、溶液を室温で15分間混合した。得られた活性化PGAをリン酸塩バッファ生理食塩水 (PBS) 中抗マウスCD3εF(ab’)2フラグメント(InVivoMAb)の溶液に4:1モル比で添加し、室温で2時間混合した。過剰試薬と非連結PGAをろ過(30,000MWCO ビバスピンカラム)で除去し、緩衝液をPBSに対して7回交換した。抗体濃度はナノドロップ2000分光光度計(Thermo Scientific製)を用いて決定された。
10μLの薬物製品擬似型レンチベクターまたははげたレンチベクターを10μLの異なる緩衝液(pH5〜7)で+4℃で異なる時間(0、15、30、60、90、120分)で希釈した。
擬似型レンチベクターに対する滴定は、フローサイトメトリーによって以下のように実施された。 HEK293T細胞(8x105細胞/ウェル)を24ウェルプレートに播種し、5%CO2 加湿雰囲気で37℃で培養した。 4時間後、培地を除去し、各ウェルに10%のFBS、1%のPSおよび10μLを含有する300μLの新鮮なDMEMを添加した。2時間後、各ウェルに0.5mLの新鮮培地を補充した。感染から3日後、各ウェル内のHEK293T細胞をトリプシナイズし、固定(BD CellFix solution #340181)し、フローサイトメトリー (AttunenXT; Invitrogen, Inc.)を用いて蛍光陽性細胞の数を決定した。
TU/mL=(eGFPを発現する細胞の割合/100)x添加ウイルスストック感染/体積時のHEK293T細胞の総数(mL)。
はげたレンチベクターの滴定は、物理的力価定量によって行われた。アッセイは、レンチウイルス関連HIV−1 p24コア蛋白質のみの検出と定量によって行われた(P24 ELISA Cell Biolabs' QUICK TITER Lenus Titer Kit)。前処理サンプルは、遊離p24と破壊されたレンチベクターを区別することを可能にする。
レンチウイルスベクターのカプセル化は以下のように行われた。レンチウイルスベクターを25mMクエン酸緩衝液pH5.0(または異なる定義のpHの適切な緩衝液システム)中で希釈し、反復ごとに75μLの最終容積を調製した。PBAEポリマーは、レンチウイルスベクター(1反復あたり75μL)と同じ緩衝液で希釈し、均質化のために2秒をボルテクスした。希釈したポリマーを希釈したベクターに1:1 の比(v/v) で添加し、混合物を10秒間静かにボルテックスし、+4℃で30分間培養した。最後に、カプセル化反応に同じ容積の培地(150μL)を添加して細胞に移送した。
前述のカプセル化方法を以下のように修飾した。レンチウイルスベクターを25mMクエン酸緩衝液pH5.0(または定義されたpHの適切な緩衝液系)中で希釈し、反復ごとに75μLの最終容積に調製した。PBAEをレンチウイルスベクター(反復ごとに75μL)と同じ緩衝液で希釈し、均質化のために2秒をボルテックスした。希釈したポリマーを希釈したベクターに1:1の比(v/v) で添加した。混合物は、10秒間穏やかにボルテックスし、+4℃で30分間培養した。 25mMのクエン酸塩緩衝液pH5.0(または定義されたpHで適切な緩衝液系)中で希釈したPGA−FaBポリマーをカプセル化混合物に添加し、均質化のために10秒を穏やかにボルテックスした。コーティング反応ミックスを+4℃で30分培養し、培地で最大300μLまで容積を完成させ、細胞に移した。
LV/PBAE/PGA−Fab錯体の粒子表面電荷測定は、波長633nmを用いたゼータサイザーナノZS(英国Malvern Instruments Ltd、4−mWレーザー)を用いた室温での動的光散乱(DLS)によって決定された。測定は、200μLの錯体を800μLのD−PBS 1x緩衝液(pH7.1)と混合することによって実施した。結果は、強度によって分析された3つの測定値の平均と標準偏差としてプロットされ、図6A〜6Fに示されている。 結果は、ポリマーの電荷に応じてベクター粒子をポリマーでカプセル化することによって表面電位変化を示し、ポリマーによるカプセル化に成功したことを示した。
レンチウイルスベクターの形質導入能力に対するポリマーコーティングの効果をテストするために、ペプチド共役ポリ(β−アミノエステル)ポリマーを調製し、レンチウイルスベクターのコーティングとHEK293T細胞の形質導入試験に使用した。 レンチウイルスベクターは緑色蛍光タンパク質(GFP)導入遺伝子を運び、形質導入された宿主細胞におけるGFP発現により形質導入を定量した(図4A)。 異なる帯電アミノ酸基を有するペプチドがポリマー骨格に結合させて、電荷の効果を調べた。外膜にVSV−Gタンパク質を含有するか、含有してないベクター粒子をポリマーコーティングした。ポリマーカプセル化の有効性は、VSV−Gに対する抗体存在下でのベクターの形質導入能力を決定することによって評価され、VSV−Gは、曝露されたVSV−Gタンパク質に依存する形質導入をブロックする能力を有する(図 4B参照)が、効果的にポリマーカプセル化されているベクターによる形質導入をブロックすることはなく、形質導入のために露出したVSV−Gに依存していない(図4C参照)。VSV−Gを欠いたLVでも同様の実験が行われ、この戦略は図5A〜5Cに描かれている。これらの「はげたLV」は、VSV−Gタンパク質の欠如のために形質導入を行うことができなかったが、PBAEポリマーによるカプセル化後に形質導入が可能となった。ポリマー有効化トランスフェクションは、抗VSV−G Abによってブロックされることは予想されなかった。
VSV−GまたはVSV−Gを発現していないPBAE被覆LVを調製し、実施例3で説明したようにJurkat細胞の形質導入を行ったが、細胞が懸濁液で増殖し、コーティング溶液のpHが変化した点を除く。 アセテート緩衝液は4.5〜6の間のpHで、クエン酸塩緩衝液は4.5〜6の間のpHで、ヒスチジン緩衝液は6〜7の間のpHで、トリス緩衝液は7〜8の間のpHとして使用された。 各pH条件において、LV粒子当たりのポリマー分子数は、106〜1010まで変化した。VSV−G+ LVの安定性を図7に示す。 ベクターは4.5〜8.0のpH範囲にわたって安定しており、最大120分までそれらのpH範囲で維持することができた。
Jurkat細胞とヒトリンパ球の形質導入
形質導入の定量のために、Jurkat細胞は、10%のFBSおよび1%のペニシリン/ストレプトマイシンを含有する完全な培地中のウェルあたり8x104細胞の密度で24ウェルプレートに播種した。細胞に300μLのカプセル化ベクターを添加した。37℃で2時間の培養後、5%CO2、500μLの新鮮な完全培地を各ウェルに添加した。GFPまたはmCherry導入遺伝子を発現する細胞の割合は、それぞれBL1またはYL1チャネルを用いたAttune Nxtフローサイトメータ(Thermo Fisher社製)を用いて72時間の後形質導入によって決定された。GFP導入遺伝子を発現する形質導入された細胞の表現型は、製造者(BD Biosciences)の指示に従って以下の細胞タイプに特異的な異なる抗体を以ってフローサイトメトリー染色によって決定された。CD3(Tリンパ球)およびCD19(Bリンパ球)。
ヒトリンパ球の形質導入は、105個の細胞/ウェルが播種されたことを除いて、同じ方法で行われた。
ヒトリンパ球は、10%のFBSおよび1%のペニシリン/ストレプトマイシンを含有する完全な培地において、ウェルあたり5x105個の細胞の密度で24ウェルプレートに播種した。カプセル化されたCAR CD19表現ベクターを細胞に300μL添加した。2時間の培養後、500μLの新鮮な完全培地を各ウェルに添加した。37℃、5%CO2で48時間培養した後、CD19陽性のラモス腫瘍細胞がリンパ球/腫瘍細胞比50:1 または1:1で添加された。37℃、5%CO2 での4時間の培養後、CAR CD19を発現する細胞をプロテインLで染色し、YL1チャネルを用いたATTUNE NXTフローサイトメータ(Thermo Fisher社製)で検出した。CD19陽性細胞およびリンパ球は、製造者(BD Biosciences)の指示に従ってCD19またはCD3特異的抗体によるフローサイトメトリー染色により定量された。
新たに調製したマウスの末梢血単核細胞(PBMC)は、FICOLL-PAQUE PREMIUM 1.084(GEヘルスケア)を充填したLEUCOSEPチューブ(グライナーバイオワン製)を用いてヘパリン化全血から単離された。DPBSで血液希釈した後、PBMCをFICOLL密度勾配で分離し、DPBSで2回洗浄し、培地中の所望の細胞密度まで再懸濁した。

Claims (48)
- ポリマーまたはポリマーの混合物でコーティングされたウイルスベクターを含むナノ粒子であって、前記ウイルスベクターは導入遺伝子を含み、前記ナノ粒子は、導入遺伝子を真核細胞に送達するための手段として機能する。
- 前記ウイルスベクターが外膜タンパク質を欠く、請求項1に記載のナノ粒子。
- 各オリゴペプチドが、アルギニン(R)、グルタミン酸(E)、リジン(K)、アスパラギン酸(D)、ヒスチジン(H)、およびシステイン(C)からなる群から選択される、少なくとも3つのアミノ酸残基を含む、請求項3に記載のナノ粒子。
- 少なくとも1つの前記オリゴペプチドの前記アミノ酸配列が、CRRR(配列番号1)、またはCHHH(配列番号2)、またはCKKK(配列番号3)、またはCEEE(配列番号4)、またはCDDD(配列番号5)である、請求項3または請求項4に記載のナノ粒子。
- 前記ポリマーまたはポリマーの混合物が、pH7で正味の正電荷または正味の負電荷を有する、請求項1〜5のいずれか1項に記載のナノ粒子。
- 前記ナノ粒子がベクターあたり108〜1012個のポリマー分子を含む、請求項1〜7のいずれか1項に記載のナノ粒子。
- 前記ナノ粒子がベクターあたり約109〜約1010個のポリマー分子を含有する、請求項7に記載のナノ粒子。
- 前記ウイルスベクターがレトロウイルスベクターである、請求項1〜8のいずれか1項に記載のナノ粒子。
- 前記レトロウイルスベクターがレンチウイルスベクターである、請求項9に記載のナノ粒子。
- 1つまたは複数のポリマー分子に連結された1つまたは複数の標的部分をさらに含む、請求項1〜10のいずれか1項に記載のナノ粒子。
- 前記標的部分が、抗体、抗体断片、scFv、抗体様タンパク質骨格、オリゴペプチド、アプタマー、L−RNAアプタマー、および細胞表面受容体のためのリガンドからなる群から選択される、請求項11に記載のナノ粒子。

- VSV−G外膜タンパク質を欠いている、請求項1〜15のいずれか1項に記載のナノ粒子。
- VSV−G外膜タンパク質を含む類似のナノ粒子と比較して、in vivo使用のための改善された安全性プロファイルを有する、請求項16に記載のナノ粒子。
- 前記ウイルスベクターがウイルス外膜タンパク質を欠き、そして前記ナノ粒子が、ベクターあたりのポリマー分子の閾値数を超える哺乳動物細胞のみを形質導入することができる、請求項1〜17のいずれか1項に記載のナノ粒子。
- ベクターあたりのポリマー分子の前記閾値数未満では、前記ポリマーの量が前記ベクターを完全にコーティングするには不十分である、請求項18に記載のナノ粒子。
- ベクターあたりのポリマー分子の前記閾値数未満で、前記ナノ粒子が構造的に不安定であるか、または前記ナノ粒子からのポリマー分子の解離を受けやすい、請求項18または請求項19に記載のナノ粒子。
- 前記閾値を欠くベクターまたはポリマーカプセル化ベクターと比較して、in vivo使用のための改善された安全性プロファイルを有する、請求項18〜20のいずれか1項に記載のナノ粒子。
- 請求項1〜21のいずれか1項に記載のナノ粒子を作る方法であって、当該方法は以下の工程を含む:
(a)導入遺伝子を含むウイルスベクターを提供すること;
(b)ポリマーまたはポリマーの混合物、および任意選択で1つまたは複数の標的部分を提供すること;そして
(c)前記ウイルスベクターとポリマーまたはポリマーの混合物、および任意選択で1つまたは複数の標的部分を接触させ、それにより前記ウイルスベクターと前記ポリマーまたはポリマーの混合物が結合して前記ナノ粒子を形成すること。 - 前記ウイルスベクターが外膜タンパク質を欠く、請求項22に記載の方法。
- ステップ(c)が約5.0〜約6.5のpHで行われる、請求項22または請求項23に記載の方法。
- ステップ(c)が約5.5〜6.0のpHで行われる、請求項24に記載の方法。
- 各オリゴペプチドが、R、E、K、D、H、およびCからなる群から選択される少なくとも3つのアミノ酸残基からなる、請求項26に記載の方法。
- 前記オリゴペプチドの少なくとも1つのアミノ酸配列が、CRRR(配列番号1)、またはCHHH(配列番号2)、またはCKKK(配列番号3)、またはCEEE(配列番号4)、またはCDDD(配列番号5)である、請求項26に記載の方法。

- 前記レトロウイルスベクターが1つのレトロウイルスベクターである、請求項22〜31のいずれか1項に記載の方法。
- 前記レトロウイルスベクターが1つのレンチウイルスベクターである、請求項32に記載の方法。
- 前記ベクターを1つまたは複数の標的部分と接触させることをさらに含み、任意選択で前記標的部分が前記ポリマーまたは粒子の混合物に連結されている、請求項22〜33のいずれか1項に記載の方法。
- 前記標的部分が、抗体、抗体断片、scFv、抗体様タンパク質骨格、オリゴペプチド、アプタマー、L−RNAアプタマー、および細胞表面受容体のリガンドからなる群から選択される、請求項34に記載の方法。

- 前記ウイルスベクターがVSV−G外膜タンパク質を欠く、請求項22〜37のいずれか1項に記載の方法。
- ステップ(c)が、前記ポリマーまたはポリマーの混合物が正味の正電荷を有し、前記レトロウイルスベクターが負の表面電位を有するpHで行われる、請求項22〜38のいずれか1項に記載の方法。
- 疾患を治療する方法であって、請求項1〜21のいずれか1項に記載の複数のナノ粒子を含む組成物をそれを必要とする対象に投与することを含む方法であって、それによって、前記対象の細胞が前記レトロウイルスベクターによって形質導入され、前記導入遺伝子が形質導入された細胞で発現される前記方法。
- 前記疾患が癌である、請求項40に記載の方法。
- 前記対象がヒトである、請求項40または請求項41に記載の方法。
- in vitroで細胞を形質導入する方法であって、培養細胞を請求項1〜21のいずれか1項に記載の複数のナノ粒子と接触させることを含み、前記培養細胞の少なくとも一部がレトロウイルスベクターによって形質導入され、前記導入遺伝子が形質導入された細胞で特異的に発現される前記方法。
- 前記ナノ粒子がCD3陽性細胞を標的とし、前記導入遺伝子がCD3陽性細胞で特異的に発現される、請求項43に記載の方法。
- 前記導入遺伝子がキメラ抗原受容体をコードする、請求項43または請求項44に記載の方法。
- 遺伝子治療を実施する方法であって、in vitroまたは生体内の細胞を請求項1〜21のいずれか1項に記載の複数のナノ粒子と接触させることを含み、それにより前記導入遺伝子が前記細胞内で発現される前記方法。
- 前記ポリマーまたはポリマーの混合物でコーティングされたウイルスベクターを含むナノ粒子であって、前記ウイルスベクターが低レベルの外膜タンパク質を含み、導入遺伝子を含み、前記ナノ粒子が前記導入遺伝子を真核細胞に送達するための手段として機能し、前記低レベルの外膜タンパク質は、前記ベクターの細胞侵入および前記ベクターによる細胞の形質導入を促進するには不十分である、前記ナノ粒子。
- 前記外膜タンパク質の低下したレベルが、前記ベクターが由来する機能的ウイルスにおける外膜タンパク質の量の5%以下である、請求項47に記載のナノ粒子。
