ヒトの体内では、巨核球は、骨髄に主に存在するCD34+造血幹細胞から引き出されるが、初期発生の間には卵黄嚢、胎児肝臓、および脾臓においても見いだされる。MK分化の間、MK前駆体はある期間の核内分裂を受け、それによって、MKは、細胞分裂を伴わないDNA複製の多数のサイクルを通じて倍数体になり、それにより、最大128nコピーのDNAを有する多形化した核がもたらされる。MKは、サイズが増大するにつれ、それらの転写およびプロテオミクスプロファイルの精密化、α顆粒および高密度顆粒を含めた高度に特殊化された顆粒の数の増加、ならびに、高度陥入膜系の発生も生じ、これらは、MKの成熟化および発達の特質である。
図1は、1つまたは複数の多能性幹細胞から巨核球前駆細胞(preMK)、巨核球(MK)、および血小板(PLT)へのスケーラブルな分化についての全体的な概略図を示す。しかし、本プロセスは多能性幹細胞に関連して記載されているが、種々の実施形態では、多能性幹細胞を他の型の幹細胞で置換することまたは他の型の幹細胞を補充することができることに留意するべきである。
マトリクス依存性増大培養
マトリクス依存性増大培養に関しては、臨床グレード多能性幹細胞(PSC)を、多能性幹細胞培養培地中、支持用マトリクス上でフィーダー細胞を伴わずに培養することによってコロニーとして増大させることができる。支持用マトリクスは、2次元表面であっても3次元構造であってもよい。一部の実施形態では、臨床グレードヒト人工多能性幹細胞は、PBG1、PBG2、またはPBG3などのヒト人工多能性幹細胞(iPSC)であってよいが、胚性幹細胞などの他の型の多能性幹細胞、または他の幹細胞を使用することもできる。
マトリクス非依存性3D増大培養に関しては、臨床グレードPSCを自己凝集性スフェロイドとして増大させることができる。一部の実施形態では、これを、単一の細胞を1ml当たり約10万個から約150万個までの密度で播種することによって達成することができる。例えば、一部の実施形態では、単一の細胞を1ml当たり50万個で播種することができる。
一部の実施形態では、分化の準備をするために、マトリクス依存性2D培養物からPSCコロニーを部分的に解離させることによって、マトリクス非依存性3D培養からPSCスフェロイドを部分的に解離させることによって、または当技術分野で公知の任意の方法によって生成された単一のPSCの自己凝集によって、PSC凝集体を生成することができる。一部の実施形態では、分化を開始する前に、これらの凝集体を多能性幹細胞培養培地、例えば、これだけに限定されないが、Essential 8培地(ThermoFisher)、StemFlex培地(Thermofisher)、またはNutriStem培地(Biological Industries)に再懸濁させ、培養することができる。一部の実施形態では、培地は、例えば、これだけに限定されないが、Y27632、H1152、またはこれらの組合せなどのROCK阻害剤を含み得る。一部の実施形態では、細胞を、分化を開始する前に、37℃、5%CO2、20%O2で0時間から72時間の間培養することができる。
フェーズIにおいて、調製されたPSCを造血性内皮細胞に分化させることができる。簡単に述べると、多能性幹細胞培養培地を取り出し、フェーズI分化培地に置き換える。一部の実施形態では、フェーズI分化培地は、StemSpan(商標)−ACF(STEMCELL Technologies、Cat番号09855)を基本培地として含み、例えば、骨形態形成タンパク質4(BMP4)、塩基性線維芽細胞増殖因子(bFGF)、および血管内皮増殖因子(VEGF)を含めた1つまたは複数の増殖因子を補充した、動物構成成分を含まない培地(ACF)であってよい。一部の実施形態では、基本培地に、それぞれ1ng/mlから200ng/mlの間の、BMP4(例えば、50ng/ml)、bFGF(例えば、50ng/ml)、およびVEGF(例えば、50ng/ml)の1つまたは複数を補充する。細胞を、低酸素条件で(例えば、37℃、5%CO2、5%O2)2日間から6日間の間インキュベートし、その後、正常酸素下(37℃、5%CO2、20%O2)で2日間から6日間の間インキュベートすることができる。一部の実施形態では、例えばWNTアゴニストまたはアンタゴニストなどのWNTモジュレーターを分化の最初の期間にわたって添加することができる。一部の実施形態では、GSK3阻害剤を分化の最初の期間にわたって添加することができる。一部の実施形態では、例えば、CHIR9998014、CHIR99021またはこれらの組合せなどのGSK3阻害剤またはWNTアゴニストを、分化の最初の期間にわたって、例えば、1日間から2日間の間などにわたって添加することができる。一部の実施形態では、WNTモジュレーターを、フェーズIの少なくとも一部分にわたって増殖因子の1つまたは複数に置き換えることができる。例えば、一部の実施形態では、WNTモジュレーターが存在する一方で、BMP4を最初の48時間にわたって添加することができ、フェーズIの残りに対しては不必要であり得る。一部の実施形態では、VEGFおよびbFGFは最初の48時間には不必要であり得るが、一方でWNTモジュレーターが存在する。一部の実施形態では、フェーズI全体を通して、毎日の培地全体の交換を、消費された培地を取り出し、新鮮なフェーズI培地に置き換えることによって実施することができる。一部の実施形態では、消費された培地の10〜95%を取り出し、等体積の新鮮なフェーズI培地に置き換えることで、部分的な培地交換を実施することができる。一部の実施形態では、追加的な体積の新鮮な培地を添加することができ、培養物の総体積が増大するという正味の影響が伴う。一部の実施形態では、新鮮なフェーズI培地に置換えるまたはそれを添加する代わりに、特定の培地構成成分を培養物にスパイクする。
一部の実施形態では、4日間から8日間の間のフェーズI後に、巨核球前駆細胞(フェーズII)分化を開始することができる。簡単に述べると、フェーズI培地を取り出し、等体積のフェーズII培地、例えば、STEMdiff(商標)APEL(商標)2基本培地(STEMCELL Technologies、Cat番号05275)などに置き換える。そのようなフェーズII培地に、それぞれ1ng/mlおよび200ng/mlの、幹細胞因子(SCF)(例えば、25ng/ml)、トロンボポエチン(TPO)(例えば、25ng/ml)、Fms関連チロシンキナーゼ3リガンド(Flt3−L)(例えば、25ng/ml)、インターロイキン−3(IL−3)(例えば、10ng/ml)、インターロイキン−6(IL−6)(例えば、10ng/ml)、およびヘパリン(例えば、5単位/ml)のうちの1つまたは複数を補充することができる。一部の実施形態では、フェーズII培地に、UM171、UM729、SR−1、SU6656、またはこれらの任意の組合せをさらに補充することができる。
一部の実施形態では、成熟巨核球の分化を、上記の通り生成したPSC由来preMKを使用して開始することができる。新鮮なまたは解凍した巨核球前駆細胞を、例えば、StemSpan(商標)−ACFを含むフェーズIIIにおける非接着性表面培地に播種することができる。非接着性表面とは、大多数の細胞がそのような表面に固着または粘着することが意図されておらず、その代わりに大部分が懸濁物中に留まるような表面を指す。例えば、そのような表面は、表面への細胞の接着を防止するまたは最小限にするために、「超低粘着性プラスチック」製であってもよく、細胞外マトリクスタンパク質でコーティングされていないものであってもよい。一部の実施形態では、フェーズIII培地に、それぞれ0ng/mlから200ng/mlの間の、TPO(例えば、25ng/ml)、SCF(例えば、25ng/ml)、IL−6(例えば、10ng/ml)、IL−9(例えば、10ng/ml)、およびヘパリン(例えば、5単位/ml)のうちの1つまたは複数を補充することができる。一部の実施形態では、フェーズIII培地にUM171、UM729、SR−1、SU6656、またはこれらの任意の組合せも補充することができる。
充填層バイオリアクター
一部の実施形態では、preMK、巨核球、血小板、または巨核球および血小板のうちの1つまたは複数の産生のために3Dスケーラブル充填層バイオリアクターを使用することができる。一部の実施形態では、フェーズIおよびフェーズII培養のために充填層バイオリアクターを使用することができる。例えば、PSCを造血性内皮細胞に分化させ、その後、preMKを産生させるために充填層リアクターを使用することができる。充填層リアクターキャリアは、マイクロサイズまたはマクロサイズのいずれであってもよく、生体適合性プラスチック、金属、ガラス、またはアルギネートなどの天然材料から形成されたものであってよい。一部の実施形態では、キャリアは、PTFEから形成された、ラシヒリング、例えば、1mmのラシヒリングの形状である。一部の実施形態では、キャリアを上記のマトリクスでコーティングすることができる。一部の実施形態では、キャリアを、ラミニン、例えば、組換えヒトタンパク質ラミニン521などでコーティングすることができる。一部の実施形態では、多能性細胞を凝集塊としてキャリアに播種することができる。一部の実施形態では、毎日の培地交換で、培地を取り出し、フェーズI培地に置き換えることができる。一部の実施形態では、フェーズIの間、多能性細胞は、充填層リアクター内のキャリアの内側に成長領域を示し得る。一部の実施形態では、多能性細胞から造血性内皮への最初の分化(すなわち、定方向分化のフェーズI)、ならびにさらなる分化およびpreMKの放出(すなわち、定方向分化のフェーズII)を同じ容器内で行うことができる。例えば、充填層バイオリアクターは、多能性細胞、例えば、PBG−1 iPSCが播種された、ラミニン−521がコーティングされたマクロキャリアを含み得る。次いで、充填層を培地の連続流に暴露させて、造血性内皮へのフェーズI分化を可能にすることができる。充填層に浸透させた後、培地を、新鮮な培地構成成分を添加することができ、スパージングまたは他の手段によって酸素/CO2濃度を調整することができる馴化チャンバーを循環させた後に、培地を細胞に再循環させることができる。
一部の実施形態では、スケーラブル3D解決策を使用し、撹拌または振とうされた容器内に懸濁させた自己凝集性スフェロイドを使用して分化を実施することを伴い得るある特定のプロセスのステップを行うことができる(図32)。一部の実施形態では、そのような容器は、低接着性表面または非接着性表面、すなわち、表面への特異的および非特異的な細胞固定化を阻害し、それにより細胞を懸濁状態に強制するために親水性または中性に荷電したコーティングでコーティングされた表面を含み得る。多能性細胞を単一の細胞に解離させ、多能性維持培地中に再懸濁させることができる。一部の実施形態では、維持培地に、H1152または他のROCK阻害剤などのRock阻害剤を補充することができる。次いで、多能性細胞を低接着性または非接着性容器内でインキュベートし、標準の培養条件下(例えば、37C、5%CO2、20%O2)で撹拌に供することができる。一部の実施形態では、撹拌をもたらすために、インキュベーション容器を、オービタルシェーカーに置くことができる、または、一定の撹拌を伴う振とうフラスコもしくはスピナーフラスコ、もしくは制御された撹拌槽バイオリアクターを使用することができる。24時間以内に、多能性細胞が自己凝集して、直径およそ50〜150umのスフェロイドが形成し得る。撹拌を休止すると、スフェロイドが容器の底部に静置し得る。
一部の実施形態では、上記の通り、新鮮なまたは解凍した巨核球preMKをフェーズIII培地における低接着性または非接着性表面に播種することができる。一部の実施形態では、そのような非接着性表面は、気体透過性膜(G−Rex(登録商標)など)であってよい。一部の実施形態では、低接着性または非接着性表面は、穏やかな撹拌を伴う細胞培養バッグまたは容器である。いずれの場合も、preMK(フェーズII培養物から新鮮に回収したもの、または凍結保存ストックから解凍したもののいずれか)をフェーズIII培地に1ml当たり50万〜1000万個の密度で懸濁させ、容器に導入する。例えば、preMKは、1ml当たり100万〜150万個、1ml当たり100万〜200万個、1ml当たり100万〜300万個、1ml当たり100万〜400万個、1ml当たり200万〜500万個、1ml当たり200万〜600万個、1ml当たり300万〜700万個、1ml当たり300万〜800万個、1ml当たり500万〜900万個、または1ml当たり800万〜1000万個の密度であってよい。細胞を、合計1〜5日間(例えば、3日間)培養して、成熟MKへの分化を可能にする。一部の実施形態では、消費された培地の10〜95%を取り出し、等体積の新鮮なフェーズIII培地に置き換えることで、毎日の培地半分の交換を実施する。フェーズIII培養の最後に、得られた細胞はサイズおよび倍数性が増大しており、成熟巨核球を示す多くの特色を示す(例として、図34〜42に示されており、また、以下に記載されている)。
一部の実施形態では、本開示は、in vitroにおいてPSC細胞または細胞株から引き出された巨核球前駆細胞、巨核球、プレ血小板、血小板前駆体または血小板を提供する。本開示の態様によると、PSC細胞または細胞株から引き出された巨核球前駆細胞、巨核球、プレ血小板、血小板前駆体または血小板は、それらの全体が参照により本明細書に組み込まれる、米国特許第9,763,984号の方法または国際出願第PCT/US2018/021354号に開示されているバイオリアクターを使用して産生される。
一部の実施形態では、本preMKおよびMKならびにそれらの構成成分は、ヒト増殖因子などの増殖因子の供給源になり得る。一部の実施形態では、そのような増殖因子を、細胞培養物、組織再生、創傷治癒、骨再生、機能性化粧品、および止血用絆創膏のために使用することができる。一部の実施形態では、本巨核球またはそれらの溶解物またはその組成物を細胞培養に使用することができる。一部の実施形態では、本巨核球またはそれらの溶解物またはその組成物を機能性化粧品として使用することができる。一部の実施形態では、本巨核球またはそれらの溶解物またはその組成物を治療剤として使用することができる。例えば、本巨核球またはそれらの溶解物またはその組成物を、ex vivoにおける細胞の増大を増加させるため、in vivoにおける骨髄再生を改善するため、組織再生および血管新生を増大させるため、ならびに放射線試験における動物の生存率を上昇させるために使用することができる。
本開示は、本開示の巨核球または分化細胞を含むキットを提供する。一実施形態では、キットは、単離された巨核球を含む組成物を含む。特定の実施形態では、本開示は、本開示の巨核球またはその前駆体を分化させる、培養する、かつ/または単離するためのキットを提供する。ある特定の実施形態では、本開示は、血小板を産生するためのキットを提供する。
臨床グレードhiPSC細胞株からの巨核球前駆細胞、巨核球および血小板の産生の拡大
臨床グレードhiPSC細胞株を、巨核球に分化する潜在性について、図2の定方向分化プロトコールを使用して試験した。図2は、多能性幹細胞から巨核球への分化の時間経過を示す概略図であり、分化の各フェーズ(フェーズ0、I、II、III)が示されている。フェーズ0〜IIIにおける細胞型および細胞マーカーが時系列の上に示されている。培地組成、マトリクス、温度および気体条件を含めた培養条件が時系列の下に示されている。
多能性PBG−1細胞の大規模増大
高密度シードバンクに必要な多数の細胞を産生させるため、ならびに臨床産生のために妥当な規模で分化を開始させるために十分な細胞数を生成するために、分化の前に多能性PBG−1を増大させることが必要である。PBG−1は、組換えビトロネクチン(VTN)と、それに加えて、動物構成成分を含まない(ACF)cGMP適合試薬、例えばEssential 8、NutriStem、またはStemFlexなどを使用して2D培養下で維持し、増大させることができる。3種の成長条件の全てで多能性マーカーの特徴的なコロニー成長および維持が観察された(図9A〜9C、図10A〜10C)。大規模増大を可能にするために、PBG−1細胞を、TrypLEを使用して2D培養物から単一の細胞として回収し、撹拌3D容器内、この場合は300mlのDasBOX miniバイオリアクター系で自己凝集させた。最初の24時間にわたって、Y27632などのROCK阻害剤を細胞に添加して、撹拌槽における6〜7日間にわたる最初の凝集の間の細胞生存を促進し、得られたスフェロイドは、その期間内で、直径が50ミクロンから250ミクロンまで増大し、全体的な細胞密度が40倍に至るまで増大した(図11A〜11C)。このように成長させたPBG−1細胞は、繰り返し継代することでき、少なくとも4回の連続した増大のラウンドにわたって多能性を維持することができ(図12A〜12B、図13B)、かつ、正常な核型を維持することができた(図14)。
2D培養容器内でIV型コラーゲンマトリクスを使用したpreMKおよびMKへのPBG−1定方向分化の詳細な特徴付け
PBG−1細胞は、0.5mMのEDTAを用いて回収し、小さな凝集塊として4.2ug/cm2のヒトIV型コラーゲンにプレーティングすると、6日間のフェーズI分化の過程中、特徴的な一連の形態学的変化を示す(図15)。フェーズIの最後に、代表的なウェルを、Accutaseを使用して単一の細胞として回収し、フローサイトメトリーによって造血性内皮マーカーであるCD31およびCD34について評価する(図16A)。複数の独立したPBG−1分化(n=41)にわたって、6日目の分化効率の平均はおよそ40%のCD31+(範囲:約20〜60%)およびおよそ30%のCD31+CD34+(範囲、約15%〜45%)であることが決定された(図16B)。
PBG−1からMKへの定方向分化を支持するために、IV型コラーゲンを組換えラミニン521に置き換えることができる。
IV型コラーゲンは、ヒト胎盤材料から精製され、研究用(Research Use Only)試薬としてのみ入手可能である。したがって、IV型コラーゲンは、PBG−1由来巨核球のcGMP生産には適合せず、これらの細胞を臨床使用のために用いることができるようにする前に代替戦略を開発しなければならない。1つの潜在的な解法は、IV型コラーゲンを組換え供給源から産生される組換えマトリクス構成成分に置き換えることである。ここで、組換えラミニン−521をIV型コラーゲンの適切な代替としてiPSCからMKへの定方向分化のために利用できることが示されている。0.5mMのEDTAを用いて回収することによって生成されたPBG−1凝集塊は、0.13ug/cm2の組換えラミニン−521での6日間のフェーズI分化の過程中、4.2ug/cm2のヒトIV型コラーゲンにおけるものと同じ特徴的な一連の形態学的変化を示した(図22A〜22B)。フェーズIIに移行させると、ラミニン−521培養により、対応するIV型コラーゲン培養と同様の収量および純度のpreMKが産生された(図23A〜23C)。3日間の追加的な分化で、フェーズIIIにおける細胞は、IV型コラーゲンで生成されたかラミニン−521で生成されたかにかかわらず、血小板前駆体の産生について同様のサイズ、形態、および傾向を示した(図24A〜24B)。成熟MKマーカーであるCD42aおよびCD42bを同時発現するCD61+(巨核球系列)細胞の割合も測定し、いずれのマトリクスで生成された細胞でも同様であることが見いだされた(図25A〜25B)。
WNTモジュレーターは、フェーズIおよびII分化効率に影響を及ぼし得る。
WNTシグナル伝達は、発生の間に重要なものである。GSK3キナーゼ阻害剤であるCHIR98014およびCHIR99021は、WNTアゴニストとして作用する。本明細書に記載のフェーズI分化条件(ラミニン521マトリクスを使用する)を、分化の最初の48時間だけ0.6uMのCHIR98014または6uMのCHIR99021を用いて強化した場合、CD31およびCD34の免疫蛍光染色によって決定された通り、6日目にフェーズI分化効率の劇的な増大が観察された(図26A〜26C)。次いで、対照およびCHIR98014培養をフェーズIIに移行し、そこで、preMKの産生および放出をCD41およびCD43の免疫蛍光染色によって追跡した。CD41+細胞の数の目測により、最初の48時間にWNTモジュレーターによって生み出されたより高いフェーズI効率が、フェーズIIの間の出力がより高いことに対応し得ることが示唆される(図27A〜27B)。したがって、WNTモジュレーターを短期間添加することが、その後の分化フェーズ全体を通した分化効率に影響を及ぼし得る。
ラミニン521がコーティングされたマクロキャリアを用いた充填層バイオリアクター。
巨核球および血小板の臨床生産に必要な収量を可能にするためには、分化プロセス全体を小規模組織培養プラスチックウェア(2D、マトリクス依存性)から3Dスケーラブル解決策に移行することが極めて重要である。ここで、本発明者らは、1mmのラシヒリングの形状の、ラミニン521がコーティングされたPTFEマクロキャリアにより、PBG−1細胞の分化に対する支持をもたらすことができ、概略図(図28)に例示されている通り、このマクロキャリア材料が充填層バイオリアクターでの使用に適するという証拠を提供する。PTFEリングをまず1.25ug/mlのラミニン−521と一緒に揺動機において4℃で終夜インキュベートした。使用前に、PTFEリングを6ウェルプレート中、Essential 8培地およびROCK阻害剤H1152を用いて平衡化した。多能性PBG−1 iPSCを、0.5mMのEDTAを使用して回収し、Essential 8培地およびH1152中に再懸濁させ、凝集塊としてPTFEリング上に播種した。10分ごとに、プレートをオービタルシェーカーに75rpmで30秒かけた。1時間後、プレートを75rpmで終夜、連続して振とうした。24時間後、培地の90%を取り出し、フェーズI培地に置き換え、毎日培地交換した。フェーズIの間、PBG−1細胞は、ラシヒリングの内側に成長領域を示し(図29)、成長領域では2D培養において見られるものと同様の形態学的特徴が生じた(図22)。これらの細胞のフローサイトメトリー分析により、高い割合の造血性内皮細胞が示され、細胞の約80%がCD31を発現し、これらの細胞の半分よりも多くがCD34+についてダブルポジティブであった(図31A)。フェーズII培地に切り換え、毎日の培地半分の交換を開始すると、概して平らなコロニーから3Dスフェロイド型構造に形態が変化したが、これらの構造はなおリング形状のマクロキャリアの内側のラミニン521コーティングに付着していたことに留意すべきである(図30)。フェーズIIの間に放出された細胞は、6+2日目の早さで高いpreMK含有量を有し、細胞の約75%がCD43とCD41を同時発現しており(図31B)、純度は2Dマトリクス依存性培養に好都合に匹敵するものであった(図18A)。6+3日目に放出された細胞を収集し、超低接着性プレート中、フェーズIII培地でさらに3日間培養し、これらの細胞の約80%がCD61とCD42bを同時発現し(図31C)、これにより、効率的なMK分化が起こったことが示される。そのようなマクロキャリアは、iPSCから造血性内皮への最初の分化(すなわち、定方向分化のフェーズI)、ならびにさらなる分化およびpreMKの放出(すなわち、定方向分化のフェーズII)を同じ容器内で行うことができる充填層バイオリアクターの材料としての使用に適する(図28)。この設計では、充填層バイオリアクターを、多能性PBG−1 iPSCを新鮮に播種した、ラミニン−521がコーティングされたマクロキャリアを用いて設定する。次いで、充填層を、培地の連続流に暴露させて、造血性内皮へのフェーズI分化を可能にする。充填層に浸透させた後、培地は馴化チャンバーを循環し、そこで、新鮮な培地構成成分を添加し、酸素/CO2濃度をスパージングまたは他の手段によって調整した後、培地を細胞に再循環させる。フェーズIの完了時に、培地を切り換えてフェーズII分化ならびにpreMKの産生および放出を可能にする。1mmのラシヒリングなどの、適切なサイズおよび形状にしたマクロキャリア支持体により、収集および凍結貯蔵のために、放出された細胞が充填層を通ってリアクターの外まで浸透することを可能にするために十分な培地の流動およびチャネル幅が可能になる。この設計では、細胞が受けるせん断力が低減し、その灌流に基づく設計に起因して効率的な培地使用が可能になり、また、preMKが放出されるにしたがってそれらを連続的に収集することが可能になる。
撹拌槽バイオリアクターにおける自己凝集性iPSC由来スフェロイド
スケーラブル3D解決策の別の例は、撹拌または振とう超低接着性容器内に懸濁させた自己凝集性スフェロイドを使用した分化の実行を伴う(図32)。本実施例では、多能性PBG−1 iPSCを、TrypLEを使用して単一の細胞に解離させ、多能性維持培地(例えば、Essential 8、Nutristem、StemFlex、他の同様の培地、またはこれらの組合せなど)およびH1152または他のROCK阻害剤中に1ml当たり細胞50万〜100万個で再懸濁させ、90rpmのオービタルシェーカー、または一定の撹拌を伴うスピナーフラスコ(125mlのスピナーフラスコ中50ml体積に対して90rpm)において、6ウェル超低接着性プレート中、37C、5%CO2、20%O2でインキュベートした。いずれかの系においても、24時間以内に、PBG−1細胞が自己凝集して、直径およそ50〜150umのスフェロイドが形成された(図33A、異なる容器での同様の例に関して図11Aも参照されたい)。次いで、撹拌を停止し、スフェロイドを容器の底部に静置させた(およそ5分間)。次いで、培地の50%〜100%を、造血性内皮への分化を促進するためにフェーズI分化培地に交換し、低酸素条件下(37C、5%CO2、5%O2)でのインキュベーションと共に撹拌を再開した。培地交換を毎日、合計で6日間、同様に実施し(37C、5%CO2、5%O2で4日間、その後、37C、5%CO2、20%O2で2日間)、その期間中に、スフェロイドはより大きく成長し、6日目までに特徴的な構造および形状が生じた(図33A)。6日目のこれらのスフェロイドの試料を解離させ、フローサイトメトリーによって評価したところ、細胞の約44%が造血性内皮マーカーであるCD31およびCD34を発現することが見いだされ(図33B)、純度は2Dマトリクス依存性培養と好都合に匹敵した(図16B)。フェーズIIに移行するために、撹拌を停止し、スフェロイドを容器の底部に静置させた(およそ5分間)。次いで、懸濁細胞の分化および放出を促進するために培地の50〜100%をフェーズII分化培地と交換した(図34A)。その後毎日、懸濁細胞を収集し、部分的な培地交換を実施した。これを行うために、撹拌を停止し、造血性内皮スフェロイドを容器の底部に静置させた(およそ5分間)。培地のおよそ80%(懸濁細胞と一緒に)を収集し、遠心分離した。作業体積の半分の新鮮なフェーズII分化培地を、十分な体積の馴化培地(すなわち、遠心分離後の上清)と一緒にスフェロイドに添加して、元の作業体積に戻した。次いで、残りの上清を廃棄し、細胞ペレットの一部をFACS分析のために使用し(図34B)、残りを凍結保存したかまたは成熟MKへの成熟化のためにフェーズIIIに移した。懸濁細胞のフローサイトメトリー分析により、6+2日目から6+6日目の間に放出された大多数の細胞がpreMKマーカーであるCD43およびCD41を同時発現することが明らかになった(図34B、図34C)。3D自己凝集性スフェロイド培養からの全体的なpreMKの純度および収量(図34C、図34D)は、2D培養からの純度および収量に好都合に匹敵した(図18A、図19A)。静的なフェーズIII培養に移行したら、3D自己凝集性スフェロイド培養からのpreMKにより、2D培養系からのpreMKと同様のMK純度が生じた(図35A〜35C)。さらに、3D自己凝集性スフェロイド培養から生成されたフェーズIII分化培養物は、サイズが劇的に増大した細胞を含有し、血小板前駆体を生成することができ(図36)、これは、それらの真正な巨核球としての正体と一致した。
PBG1 iPSC由来巨核球の詳細な特徴付け
本明細書に記載の方法を使用して生成される巨核球は、MK特異的タンパク質ベータ−1−チューブリン(図37)、ならびにアルファ顆粒(PF4およびVWF、図38A〜38F)および高密度顆粒(LAMP1およびセロトニン、図39A〜39F)に関連するタンパク質について免疫蛍光顕微鏡により画像化した場合のものを含め、機能的な成熟MKに関連する多くの特徴を示す。PBG1由来MKの電子顕微鏡画像から、多小胞体、グリコーゲン顆粒、および陥入膜系を含めた特徴的な超微細構造的特性が明らかになる(図40A〜40D)。遺伝子発現解析により、OCT4などの多能性遺伝子の下方制御(図41A)およびNFE2などの巨核球系列遺伝子の上方制御(図41B)が明らかになった。同様の分析を関連性のある遺伝子のパネルに対して実施し、この解析の結果は、多能性幹細胞シグネチャーの喪失および巨核球シグネチャーの獲得と一致する(図41C)。
本明細書に記載の通り、本開示は、巨核球前駆細胞および巨核球を産生するための組成物および方法を特色とする。一態様では、本開示は、臨床グレードhiPSC細胞または細胞株から分化させた巨核球または巨核球前駆細胞を提供する。一部の実施形態では、本開示は、本明細書に描写されている任意の態様による巨核球または巨核球前駆細胞を含む単離された細胞の集団を提供する。一部の実施形態では、本開示は、本明細書に描写されている任意の態様による巨核球または巨核球前駆細胞を含む組成物を提供する。
Claims (48)
- 巨核球を産生するための方法であって、
低接着性または非接着性条件下、かつ撹拌下で多能性幹細胞を増大させるステップであって、増大した多能性幹細胞が自己凝集性スフェロイドを形成する、ステップと、
前記多能性細胞を第1の培養培地中で造血性内皮細胞に分化させるステップと、
前記造血性内皮細胞を第2の培養培地中で巨核球前駆細胞に分化させるステップと
を含む、方法。 - 前記多能性細胞を造血性内皮細胞に分化させるステップを、マトリクス上、接着性条件下で行う、請求項1に記載の方法。
