Abstract
Description
例1.核酸構築物の合成,クローニングおよび発現
ヒトG-CSFのcDNA配列は、E. coli(Genescript社)における発現に最適化されたコドンであった。この配列は、配列番号: 51のフォワードプライマーIMT1: 5’ GGATCCATGACGCCGCTGGGTCCG3’と、配列番号: 52のリバースプライマーIMT2: 5’ AAGCTTTTACGGCTGTGCCAGGTGAC3’とを使って、発現ベクターpET23a(Novagen社)のBamHIおよびHindIII部位にクローニングした。この構築物を使ってNovagen社のE.coli BL21 (DE3)株を形質転換した。コドン最適化されたrHuG-CSF配列の収量を増加させるため、コドン最適化構築物のDNAおよびRNA配列のin silico解析を行った。この解析によれば、mRNA転写物の5’末端にヘアピンと高安定な二次構造とが形成されていることを示唆しており、mRNA転写物が翻訳を妨害する可能性について指摘している。5’末端で翻訳的にサイレントな突然変異が起こり、(mRNA転写物における二次構造をさらに促進する可能性が高い)GCリッチコドンを適切な位置でATリッチコドンと置換することによって、mRNA転写物の二次構造を破壊したり減少させたりする。一連の翻訳的にサイレントな突然変異を通して、いくつかの配列を生成し、天然配列のものと比較してタンパク質収量における増加について分析した。

5’末端にIMT8配列(配列番号: 58)を組み込んだヒトG-CSFのコドン最適化されたcDNA配列を、発現ベクターpET23aおよびpET9bのNdeIおよびHindIII部位にクローニングした。その後、この遺伝子操作された構築物(配列番号: 1)を用いてE.coliのBL21 (DE3)株を形質転換した。

v.分離したG-CSFを2ステップのリフォールディング処理に供した。
vi.リフォールドされたG-CSFを、陽イオン交換クロマトグラフィーを使って精製した。
vii.陽イオン交換で精製した試料を疎水性相互作用クロマトグラフィー(HIC)やサイズ排除クロマトグラフィー(SEC)に供した。その樹脂とカラムはGEヘルスケアライフサイエンス社から購入した。
viii. G-CSFのシステイン変異体の場合、ステップviの精製G-CSF変異体を用いて様々なサイズのPEGを結合した。
ix.5倍モル過剰のチオール特異的PEGを用いて、リン酸ナトリウムバッファーまたはトリスバッファー中でrHuG-CSF変異体を結合させた。バッファーの好ましいpHは約6.5〜約7.5である。
x.リン酸ナトリウムバッファー、酢酸ナトリウムバッファーまたはクエン酸ナトリウムバッファー中でN末端のアミノ酸特異的なPEGをrHuG-CSFに結合させた。バッファーの好ましい濃度は25mM〜約100mMの範囲であった。この反応は、還元剤であるシアノ水素化ホウ素の存在下で行った(好ましい濃度は約15mM〜約25mMの範囲)。反応にとって好ましいpHは、4.0-5.5の範囲であった。両方の化学反応に用いたPEGは、JenKem Technology社(米国)のものであった。
xi.PEG化した化学種はSPセファロースまたはより好ましくはMacroCap SP樹脂(GEヘルスケアライフサイエンス社)を使った陽イオン交換クロマトグラフィーに続いてHICやSECによって精製した。
xii.その試料をSDSPAGEで泳動し、PEG化G-CSFを可視化するためにヨウ化バリウムで染色した(図2)。
G-CSFは、好中球前駆細胞のエフェクター好中球(effector neutrophils)への増殖、成熟および分化の主要な成長因子である。長年にわたる豊富な構造学的および機能学的研究により、好中球の増殖において重要や役割を果たすシグナル伝達カスケードを開始するための、G-CSF受容体(G-CSFR)との結合において重要な役割を果たすG-CSFの領域についての膨大な情報を蓄積してきた。2:2立体配座において、G-CSF受容体のリガンド結合領域と複合されるG-CSFの構造が解明された(Aritomi, M.ら, Nature, 1999.401(6754): p. 713-7 and Tamada, T.ら, Proceedings of the National Academy ofSciences of the United States of America, 2006. 103(9): p. 3135-40)。
G-CSFはループで接続される4つのらせんを有し、また、NおよびC末端領域は非構造化領域を有する。重要なことは、コンピュータ分析によりG-CSFのこれらのNおよびC末端が溶媒接触性であることを示唆したということである。そのタンパク質のC末端にフレキシブルなアミノ酸リンカー配列を付加することでも、その領域の適応性および溶解性を高めることができた。そのフレキシブルなリンカーは概してグリシンやセリンなどの小さいまたは極性のアミノ酸において豊富であるが、スレオニン、アラニン、リシンおよびグルタミン酸などのアミノ酸で構成することもできる。
PEG化は治療タンパク質の修飾変異体を生成するために用いられる重要な方法の一つであり、invivoでのそれらの全体的な半減期を改善するためのものである。現在使用されているPEG化G-CSFは、還元アルキル化を使ってN末端に20kDaのPEG分子が結合される。システイン変異は、治療タンパク質のPEG化を促進することが示されている。本発明において、部位特異的なPEG化を促進するため、G-CSFのいくつかのシステイン変異体を提供した(表4)。G-CSF受容体との相互作用に関与するタンパク質のコアが変更されていないため、これらのG-CSF変異体は市販のG-CSFに匹敵する生物活性を有することが予想された。しかしながら、それらの設計およびPEG化に起因して、それらはゆっくりと分解するため、生物学的利用能が改善するという利点を備える。そのようなG-CSFのPEG化変異体は、より長い血清半減期を有することが予想される。
rHuG-CSFおよびその変異体の生物活性を分析するため、細胞増殖アッセイを行った。rHuG-CSFの生物活性は、マウス骨髄芽球性NFS-60細胞を増殖させる能力によって測定した。これらのアッセイにおいて、増殖細胞の代謝活性はXTTなどのテトラゾリウム試薬の還元を通して測定される。その細胞を様々な濃度の標準物質(standard)、rHuG-CSFおよびそのPEG化変異体で48時間処理し、その後、それらの代謝活性についてXTT試薬を用いて評価した。市販のフィルグラスチムおよび実験室で生成したrHuG-CSFおよびその変異体の生物活性は同等であることがわかった。この結果は、G-CSFにおける変異により生物活性を損なわないことを示している可能性がある。
本研究において遺伝子操作した新規のG-CSFの生物活性およびin vivoでの半減期を解析するため、本研究に12〜14週齢の雄のBALB/cマウスを用いた。このために、マウスを1週間慣れさせ、標準的な手順に従ってシクロホスファミド(200mg/kg)の腹腔内注射を用いて好中球減少症をマウスに誘発させた。好中球減少症の誘発を確認するため、血液を採取して全白血球数(TLC)を測定した。好中球減少症の誘発の1日後、「シャム(Sham)」またはモック(バッファーのみを含む)、治療効果のあるG-CSF(市販のもの)、および遺伝子操作した変異体を、単回皮下投与として個別に投与するか(最大1 mg / Kg)、または、4〜7日間の期間に125 μg / Kgを複数回投与した。G-CSF処理後、血液試料を採取し、次の5〜10日間、TLC数を測定した。実験室で生成した、天然様のものと、市販のフィルグラスチムとは、同等の比活性度を示した。興味深いことに、我々はPEG化G-CSF変異体(すなわち新たに構築されたPEG化システイン変異体)で好中球減少症を治療した結果、好中球減少症からの回復が早められたことを観察した。同様に、in vivoでの半減期を見積もるため、マウスの独立したグループにシャム試料、市販のG-CSFおよび新規のG-CSF変異体のいずれかを注射した。血液試料を採取し、血清中のG-CSFの存在を市販のG-CSFエライザキットを使って見積もった。これらのデータによれば、変異体が元の未修飾のG-CSFと比べてより高い生物活性およびより長期のin vivo半減期を有することを明確に示した。
● 開示したポリペプチドは結合に対してより感受性が高い。また、開示したポリペプチドにより結合パターンを最適化することができる。開示した結合体は、治療上有利な性質である、有意に長期の血清半減期を示す。それゆえ開示した複合体は、化学療法剤の投与と同じ日に用いることができる。部位特異的なPEG化は、従来用いられたアミンPEG化に起因して発生する、生成物の不均一性と生物活性の減少といった問題を克服する。
● 部位特異的なPEG化により、そのタンパク質が固有の単一、二重またはそれ以上の所定の部位にてPEGと選択的に結合できるようになる。そのような部位は、ポリペプチドの特徴づけをより着実なものとし、生物活性を比較的正確に予測することができ、異なる治療計画に相応しいものとすることができる。さらに、PEG化の部位は溶媒接触可能な残基にあり、PEG化を高効率にしかつ生成物をより均質にする。
● 本発明ではG-CSFのシステイン変異体を提供するとともに、PEG結合変異体を生成するためのそれらの応用を提供する。これは治療上有利な性質である、有意に長期の血清半減期を有する可能性があり、したがって化学療法剤の投与と同日に使用することができる。

Claims (12)
- 顆粒球コロニー刺激因子活性を示すポリペプチドであって、配列番号: 2に記載の配列に対して少なくとも90%の配列相同性を有し、かつ、T1CP2 (配列番号: 25), P2CL3 (配列番号: 26), L3CG4 (配列番号: 27), G4CP5 (配列番号: 28), P5CA6 (配列番号: 29), A6CS7 (配列番号: 30), S96CP97 (配列番号: 31), P97CE98 (配列番号: 32), L99CG100 (配列番号: 33), P101CT102 (配列番号: 34), E122CE123 (配列番号: 35), L124CG125 (配列番号: 36), M126CA127 (配列番号: 37), P138CA139 (配列番号: 39), A143CF144 (配列番号: 40), R146CR147 (配列番号: 41), R169CH170 (配列番号: 42), H170CL171 (配列番号: 43), L171CA172 (配列番号: 44), A172CQ173 (配列番号: 45)およびQ173CP174(配列番号: 46)からなるグループから選択される部位に少なくとも一つの非天然のシステイン残基を有するアミノ酸配列を備え、システインの挿入を含むことを特徴とするポリペプチド。
- 顆粒球コロニー刺激因子活性を示すポリペプチドであって、配列番号: 2に記載の配列に対して少なくとも90%の配列相同性を有するアミノ酸配列を備え、配列番号: 3に記載のセリン/アラニン残基によるC17の置換を含むことを特徴とするポリペプチド。
- 請求項1または2に記載のポリペプチドであって、N末端および/またはC末端に、(G)nC (配列番号: 47), (GGS)nC (配列番号: 48), (GGGGS)nC (配列番号: 49)および(SGGSGG)nC (配列番号: 50)からなるグループから選択される短いリンカー配列をさらに備えることを特徴とするポリペプチド。
- 請求項1〜3のいずれか一項に記載のポリペプチドであって、少なくとも1分子のポリエチレングリコールに結合されていることを特徴とするポリペプチド。
- 請求項4に記載のポリペプチドであって、ポリエチレングリコールがメトキシPEGマレイミド誘導体であることを特徴とするポリペプチド。
- 請求項4または5に記載のポリペプチドであって、ポリエチレン分子のサイズが5,000〜40,000ダルトンの範囲に及ぶことを特徴とするポリペプチド。
- 配列番号: 58 (IMT8), 配列番号: 59 (IMT9)および配列番号: 60 (IMT10)のオリゴデオキシヌクレオチド配列であって、顆粒球コロニー刺激因子(G-CSF)タンパク質およびその変異体の発現を増大させるためにG-CSFの5'プライム配列にあり、核酸構築物が顆粒球コロニー刺激因子活性を示すポリペプチドをコードし、その構築物が配列番号: 1に記載の配列を備えることを特徴とするオリゴデオキシヌクレオチド配列。
- 発現ベクターであって、請求項7に記載の核酸構築物を備えることを特徴とする発現ベクター。
- 宿主細胞であって、請求項8に記載の発現ベクターを備えることを特徴とする宿主細胞。
- 好中球減少症を患う患者を治療する方法であって、請求項1〜6のいずれか一項に記載のポリペプチドの治療量を患者に投与するステップを備えることを特徴とする方法。
- 請求項1〜6のいずれか一項に記載のポリペプチドの使用であって、好中球減少症を治療するための医薬組成物の調製のための使用。
- 医薬組成物であって、請求項1〜6のいずれか一項に記載のポリペプチドと、少なくとも一つの薬学的に許容できる担体または賦形剤とを備えることを特徴とする医薬組成物。
