Abstract
Description
培養した胎盤、または、その一部から、エキソソームを生成、単離、及び、特徴決定する方法を提供する。本明細書で説明する代替手法は、バイオテクノロジーのツール、及び、治療法として使用することができるエキソソームの生成、分離、特徴決定を容易にする。
エキソソームは、生体細胞から分泌されるナノサイズの二脂質膜小胞であり、細胞間連絡において重要な機能を果たす。ヒトが妊娠している期間、胎盤は、生理的ホメオスタシスの調節と、胎児の発達の支援において中心的な役割を果たす。胎盤が分泌する細胞外小胞、及び、エキソソームは、胎盤と母体組織との間の連絡に寄与して、母体胎児耐性を維持することが知られている。エキソソームは、脂質、サイトカイン、ミクロRNA、mRNA、及び、DNA、ならびに、タンパク質などの活性な生物製剤を含んでおり、これらを、当該エキソソームの表面に提示することができる。エキソソームは、免疫調節、血管形成の促進などの数多くの治療方法、及び、薬剤の送達に有用であると考えられている。大量のエキソソームの分離を可能にするさらなる手法が必要であることは、明らかである。
本発明の態様は、培養した胎盤、または、その一部から、エキソソームを生成、単離、及び、特徴決定する方法に関する。本明細書で説明する手法は、バイオテクノロジーのツール、及び、治療法として使用することができるエキソソームの生成、分離、特徴決定を容易にする。好ましい代替手法は、以下の通りである:
a)胎盤、または、その一部、好ましくは、培養胎盤、または、その一部を、第1の培地と接触させること;及び
b)前記胎盤、または、その一部からエキソソームの集団を含む第1の画分を得ること;
c)任意に、前記胎盤、または、その一部を、第2の培地と接触させ、かつ、前記胎盤、または、その一部に由来するエキソソームの集団を含む第2の画分を得ること;
d)任意に、前記胎盤、または、その一部を、第3の培地と接触させ、及び、前記胎盤、または、その一部に由来するエキソソームの集団を含む第3の画分を得ること;及び
e)任意に、前記第1、第2、及び/または、第3の画分に由来するエキソソームの集団を、好ましくは、連続遠心分離、及び/または、所望のエキソソームの集団に存在するマーカー、または、ペプチドに特異的な抗体、または、その結合部分を使用するアフィニティークロマトグラフィーで単離すること、を含み、前記抗体、または、その結合部分を、膜、樹脂、ビーズ、または、容器などの基質に固定している、前記方法。
(b)(a)で回収した濾過物に対して第1の遠心分離を行って、細胞ペレット、及び、第1の上清を生成すること;
(c)前記第1の上清に関して第2の遠心分離を行って、第2の上清を生成すること;及び
(d)前記第2の上清に関して第3の遠心分離を行って、エキソソームペレットを生成すること;及び、任意に、
(e)前記エキソソームを溶液に再懸濁すること、
を含む方法で、前記エキソソームを、前記第1、第2、及び/または、第3の画分、または、複数の画分から単離する、前出代替手法のいずれか1つに記載の方法。
5.1.胎盤由来のエキソソーム
本明細書に記載した胎盤由来のエキソソームは、後述するように、それらの形態、及び/または、分子マーカーで選択、及び、同定することができる。本明細書に記載した胎盤由来のエキソソームは、当該技術分野で公知のエキソソーム、例えば、絨毛膜絨毛間葉系幹細胞由来のエキソソーム、例えば、Salomon et al.,2013、PLOS ONE、8:7、e68451に記載されたものなど、とは異なる。したがって、本明細書で使用する用語「胎盤由来のエキソソーム」は、絨毛膜絨毛間葉系幹細胞から得た、または、それに由来したエキソソームを含むことを意味しない。
本明細書に記載した胎盤由来のエキソソームは、当該エキソソームを、同定、及び/または、単離するために使用することができるマーカーを含む。これらのマーカーは、例えば、タンパク質、核酸、糖分子、グリコシル化タンパク質、脂質分子とし得るものであり、そして、モノマー、オリゴマー、及び/または、マルチマーの形態で存在し得る。特定の実施形態では、これらのマーカーは、エキソソームが由来する細胞が生産する。特定の実施形態では、当該マーカーは、エキソソームの由来となる細胞が提供するが、当該マーカーは、前記細胞が、高レベルで発現することはない。特定の実施形態では、本明細書に記載したエキソソームのマーカーは、対照マーカー分子と比較した場合に、起源の細胞と比較して、エキソソームの方がより多い。別の特定の実施形態では、本明細書に記載したエキソソームのマーカーは、別の細胞型から得たエキソソームと比較して、当該エキソソームの方で豊富にあり(例えば、Salomon et al.,2013、PLOS ONE、8:7、e6845lに記載の絨毛膜絨毛間葉系幹細胞、及び、前脂肪細胞間葉系幹細胞)、これらのエキソソームは、同一の方法で単離する。
本明細書に記載したエキソソームは、それらの同定を可能にし、かつ、それらの起源の細胞、ならびに、その他の細胞物質、及び、非細胞物質を含まない細胞エキソソームの実質的に純粋な集団を単離/得るために使用できる表面マーカーを含む。エキソソーム表面マーカー組成を決定する方法は、当該技術分野で公知である。例えば、エキソソーム表面マーカーは、蛍光活性化細胞選別(FACS)、または、ウエスタンブロッティングで検出することができる。
本明細書に記載したエキソソームは、本明細書に記載した方法に従って単離し得るものであり、そして、それらの収率を定量し得る。特定の実施形態では、本明細書に記載したエキソソームを、約0.5〜5.0mg/リットルの培養培地(例えば、血清を含む、または、含まない培養培地)の濃度で単離する。別の特定の実施形態では、本明細書に記載したエキソソームを、約2〜3mg/リットルの培養培地(例えば、血清を含む培養培地)の濃度で単離する。別の特定の実施形態では、本明細書に記載したエキソソームは、約0.5〜1.5mg/リットルの培養培地(例えば、血清を含まない培養培地)の濃度で単離する。
本明細書に記載したエキソソームは、保存することができ、すなわち、長期保存を可能にする条件下、または、エキソソームの分解を阻害する条件下に置くことができる。
本明細書では、本明細書で提供するエキソソームを含む組成物、例えば、医薬組成物をさらに提供する。本明細書に記載した組成物は、エキソソームを用いた処置が有益である対象(例えば、ヒト対象)における特定の疾患、及び、障害の処置において有用である。
疾患または病態の処置、及び/または、予防における治療的使用に有効である、本明細書に記載したエキソソームまたは組成物の量は、疾患の性質によって変化し、また、標準的な臨床技術によって決定することができる。対象に投与するエキソソーム、または、その組成物の正確な投与量も、投与経路、及び、処置する疾患または病態の重症度によって変化し、また、開業医の判断と、それぞれの対象の状況に応じて決定すべきである。例えば、有効な投与量は、投与手段、標的部位、患者の生理学的状態(年齢、体重、及び、健康状態など)、患者が人間であるか動物であるか、投与するその他の薬剤、及び、処置が予防的であるか否かによって、変化し得る。安全性と有効性を最適化するために、最適な治療用量に調整する。
5.3.1.血管形成が有効な疾患の処置
本明細書に記載したエキソソーム、及び、その組成物は、血管形成を促すものであり、したがって、血管形成が有効な疾患及び障害を処置するために使用することができる。したがって、本明細書に記載したエキソソーム、または、その組成物を使用して、それを必要とする対象における血管形成を促す方法を、本明細書で提供する。本明細書で使用する用語「処置する」は、対象における疾患、障害、または、病態、または、それらのあらゆるパラメータ、または、症状の治癒、緩和、改善、重症度の軽減、または、経時的抑制を含む。特定の実施形態では、本明細書で提供する方法に従って処置する対象は、哺乳動物、例えば、ヒトである。
特定の実施形態では、本明細書に記載したエキソソームは、本明細書に記載した疾患または病態のいずれかを治療する必要がある対象に対して投与する。別の実施形態では、本明細書に記載した組成物は、本明細書に記載した疾患または病態のいずれかを治療する必要がある対象に対して投与する。特定の実施形態では、当該対象は、ヒトである。
本明細書に記載した医薬組成物、すなわち、本明細書に記載したエキソソームを含む組成物の1つ以上の成分で満たした1つ以上の容器を含む医薬パックまたはキットを、本明細書で提供する。医薬品または生物学的製剤の製造、使用、または、販売を規制する政府機関が規定した形式の通知を、任意に、それら容器(複数可)に明記することができ、このものは、ヒトへの投与のための製造、使用、または、販売の規制機関の承認を示す。
胎盤は、造血幹細胞(HSC)、及び、非造血幹細胞などの幹細胞を含む細胞の貯蔵庫である。本明細書では、バイオリアクターで培養をする胎盤、または、その一部からエキソソームを単離する方法を記載している。培養を行っている間に、細胞からエキソソームは分泌され、そして、培地にエキソソームは分泌され、これにより、エキソソームのさらなる処理と分離が容易になる。エキソソームは、培養の異なる段階で、胎盤、または、その一部から分離することもできる(例えば、異なる時点で、かつ、それぞれの回復ステップで、異なるかん流液を使用し得る)。これらのエキソソームを、一旦、培地に取り込むと、例えば、遠心分離、(例えば、小さなRabファミリーGTPase、アネキシン、フロチリン、Alix、Tsg101、ESCRT複合体、CD9、CD37、CD53、CD63、CD63A、CD81、CD82)、Hsp70、Hsp90、上皮細胞接着分子(EpCam)、パーフォリン、TRAIL、グランザイムB、Fasに特異的な基質に結合した結合剤などの固定化結合剤;Fasリガンド、CD24、EpCAM、EDIL3、フィブロネクチン、Survivin、PCA3、TMPRSS2:ERG、Glypican−1、TGF−β1、MAGE 3/6、EGFR、EGFRvIII、CD9、CD147、CA−125、EpCam、及び/または、CD24などの1つ以上のがんマーカー;または、α−シヌクレイン、HIV、または、HCVタンパク質、タウ、ベータ−アミロイド、TGF−ベータ、TNF−アルファ、フェチュイン−A、及び/または、CD133など、これらに限定されない、ウイルス、真菌、または、細菌タンパク質、または、ペプチドなどの1つ以上の炎症マーカーまたは病原マーカーを利用する)市販のエキソソーム単離キット、レクチンアフィニティー、及び/または、アフィニティークロマトグラフィーを使用してさらに単離することができる。単離したエキソソームは、治療、診断、及び、バイオテクノロジーツールとして使用することができる。
胎盤からエキソソームを回収する例示的な方法を、図1に示す。エキソソーム生成のためのバイオリアクターとして胎盤を使用するのであれば、臍帯血:PRP、胎盤かん流液(PS)、胎盤組織培養物(PTS)、胎盤臓器培養物(PO)、または、胎盤、または、その一部に存在し得る外因性細胞胎盤を、エキソソーム単離の供給源とし得る。ある手法によって、胎盤、または、その一部を回収する(#200010323、2017年9月25日に回収した)。胎盤を培地と接触させ、または、通常のPSC−100 回収方法でかん流させて、PS−1(2017年9月26日)として回収した。胎盤、または、その一部を、フード内で、少なくとも4時間インキュベートする。胎盤、または、その一部を、培地(RPMI培地)と接触させ、または、500mLのRPMIベース培地(1%抗生物質)でかん流させて、PS−2として回収した。次に、胎盤、または、その一部を、フード内で、一晩インキュベートし、そして、カバーをかける。胎盤、または、その一部を、750mLの生理食塩水と接触させ、または、かん流させて、そして、PS−3として回収する。その後、これらの試料を、分析(Warren)のために実験室に発送した。PS1、PS2、及び、PS3を、RBC溶解した後に、同日に、FACSで分析した。
胎盤バイオリアクターの上清の画分を、上記した方法で回収し、そして、これらの画分を濾過した。その後、上清を、400g×10分の遠心分離にかけて、細胞を回収した。最初の遠心分離の後に、2回目の遠心分離を、3000g×30分で行って、細胞破片をペレット化した。3回目の遠心分離を、10,000g×1時間で行って、微小胞をペレット化した。次に、4回目の遠心分離を、100,000g×1.5時間で行って、エキソソームをペレット化した。次に、ペレット化したエキソソームを含む遠沈管を逆さにして紙の上に置いて、残留液体を排出した。次に、当該エキソソームペレットを、適切な容量(例えば、2.0mL)の滅菌PBSに溶解して、ペレットを溶解し、そして、当該エキソソームを含む溶液を、滅菌エッペンドルフチューブに等分し、次に、−20℃/−80℃の冷凍庫で凍結した。そして、エキソソームを、ELISA−63A、及び、タンパク質定量キットを使用して、エキソソーム特異的マーカーCD63Aの存在についてアッセイした。
ここで示したように、PRP、胎盤かん流液、及び、胎盤組織は、CD63+であり、かつ、超遠心によって効率的に単離できるエキソソームの集団を含む。当該エキソソームを単離するために、まず、培養上清を、組織フィルターで濾過し、そして、上記したようにして数度の遠心分離を行ってエキソソームを得て、それを、次に、凍結した。当該エキソソームをELISA検出するために、抗CD63抗体を使用した。アッセイでは、試料を、エキソソーム結合緩衝剤(60uL+60uL)で、1:1に希釈した。この手順によって、CD63+エキソソームを、効率的に単離した。
エキソソームは、タンパク質、ペプチド、RNA、DNA、及び、サイトカインを含み得る。miRNA配列決定、表面タンパク質分析(MACSPlex Exosome Kit、Miltenyi)、プロテオミクス分析、機能研究(インビトロでの酵素アッセイ創傷治癒アッセイ(スクラッチアッセイ)、エキソソーム誘発細胞増殖(ヒトケラチノサイト、または、線維芽細胞)(5つの公知の刺激物質との比較)、エキソソーム誘発コラーゲン生産(ヒトケラチノサイト、または、線維芽細胞):TGFbと比較して、血清及び非血清対照、プロコラーゲン1CペプチドのELISA、炎症性サイトカインのエキソソーム誘発阻害を含んでおり:応答細胞タイプは、ヒトケラチノサイト、または、ヒト線維芽細胞、及び、凍結乾燥した加熱殺菌細菌、または、LPSとの比較を含む)などの方法で実行し得る。
ドナーの完全な同意の下、分娩後のヒト胎盤をかん流した。胎盤の残存血液を、大量の滅菌生理食塩水で洗い流し、次いで、抗生物質を補充した無血清培養培地を含む5−Lバイオリアクターで培養し、そして、37℃インキュベーター(5%CO2)で培養し、次いで、冷蔵条件で、最長で4日間かけて回転させた。培養培地の上清を、3000g、及び、10,000gの連続遠心分離で処理して、組織、細胞、及び、微小胞をペレット化した。エキソソームを、10,000g遠心分離で得た上清から、100,000g超遠心分離によってペレット化し、そして、滅菌PBSで溶解した。エキソソームの収量を、BCAタンパク質アッセイで定量した。
一部の代替手法では、胎盤、または、その一部からエキソソームを単離する方法を提供する。この方法は、a)胎盤、または、その一部を、第1の培地と接触させること;b)当該胎盤、または、その一部から、エキソソームを含む第1の画分を得ること;c)当該胎盤、または、その一部を、第2の培地と接触させること;d)当該胎盤、または、その一部から、エキソソームを含む第2の画分を得ること;e)当該胎盤、または、その一部を、第3の培地と接触させること;f)当該胎盤、または、その一部から、エキソソームを含む第3の画分を得ること、及び、任意に、当該第1、第2、及び/または、第3の画分からエキソソームを単離すること、を含む。一部の代替手法では、当該方法は、胎盤、または、その一部を、さらなる培地と接触させること;及び、当該胎盤、または、その一部から、エキソソームを含むさらなる画分を得る複数のステップをさらに含む。これらの2つのステップは、複数回反復し得る。好ましくは、当該胎盤、または、その一部を、バイオリアクターで、培養及び/または維持する。一部の代替手法では、当該胎盤、または、その一部は、羊膜を含む。一部の代替手法では、当該胎盤、または、その一部は、ヒトの胎盤、または、その一部である。一部の代替手法では、当該第1、第2、及び/または、第3の培地は、当該胎盤、または、その一部と、45分間、または、1、2、3、4、5、6、7、8、9、10、11、または、12時間、または、前出した時点のいずれか2つで定義した範囲内にある長さの時間など、少なくとも45分間かけて接触させる。一部の代替手法では、当該第1、第2、及び/または、第3の培地は、胎盤、または、その一部と、少なくとも7、14、28、35、または、42日間、または、前出した時点のいずれか2つで定義した範囲内にある長さの時間をかけて接触させる。一部の代替手法では、胎盤、または、その一部を、細分化、粉砕、または、コラゲナーゼ、及び/または、プロテアーゼなどの酵素で処理する。
(a)当該第1、第2、及び/または、第3の画分、または、複数の画分を組織フィルターに通すこと;
(b)(a)で回収した濾過物の第1の遠心分離を実行して、細胞ペレット、及び、第1の上清を生成すること;
(c)当該第1の上清について第2の遠心分離を実行して、第2の上清を生成すること;及び
(d)当該第2の上清について第3の遠心分離を実行して、エキソソームペレットを生成すること;及び、任意に、
(e)当該エキソソームを溶液に再懸濁すること、を含む方法で、当該エキソソームを、当該第1、第2、及び/または、第3の画分、または、複数の画分から単離する。
6.1.実施例1:ヒト胎盤の培養
ヒト胎盤を受領し、そして、滅菌PBS、または、生理食塩水で洗浄して血液を除去する。次いで、この胎盤を、抗生物質と2mM EDTAとを補充した500mLまたは1000mLのDMEM培養液を入れた大きな容器で、臓器全体を容器内で培養する。別の方法では、当該胎盤を、様々な大きさに細分し、そして、培養容器に入れることができる。この培養は、37oCで、5%CO2を含む細胞培養インキュベーターで行う。培養時間は、4時間〜8時間であり、そして、培養液の上清を、エキソソームの単離のために使用する。新たな培地を、それぞれの回収時点で(例えば、8時間ごと、または、12時間ごとに)加え、そして、当該胎盤臓器と組織とを、少なくとも5日間培養する。
培養の上清を、3,000gで、30分間、遠心して、細胞と、組織の破片とをペレット化する。次いで、当該上清を、10,000gで、1時間、遠心分離し、そして、ペレット(小細胞破片と細胞小器官)を廃棄する。次いで、当該上清を、100,000gで、2時間、遠心分離する。得られたペレットは、エキソソームである。このエキソソームペレットは、以下の方法でさらに精製できる:異なる量の滅菌PBSで再懸濁し、そして、100,000で、2時間遠心し、次いで、最終ペレットを、滅菌PBSで再懸濁する。この再懸濁したエキソソームを、シリンジフィルター(0.2um)で濾過し、−80℃で、300uL〜1mLまでの異なる量で分注する。
pExoのタンパク質マーカーを、MACSPlex Exosome Kit(Miltenyi Biotec、Cat#130−108−8l3)を用いて、このキットに備え付けのプロトコールに従って分析した。要するに、120μLのpExo単離物を、15μLのエキソソーム捕捉ビーズと、室温で、一晩、インキュベートした。1mLの洗浄液で1度洗浄した後に、当該エキソソームを、エキソソーム検出試薬CD9、CD63、CD81カクテルとインキュベートし、そして、さらに1時間インキュベートした。2回洗浄した後に、これらの試料を、FACS(BD Canto 10)で分析した。このキットは、mIgG1とREA対照とを除いて、合計で37個のタンパク質マーカーを備えている(表1)。

41個の異なるサイトカインを備えたMiltiPlex Luminexキットを使用して、サイトカインの含有量について、pExo試料を分析した。以下の表は、15個の異なるpExo調製物で検出したサイトカインのデータを示す。このデータは、pExoが、FGF2、G−CSF、フラクタルカイン、GDGF−AA/BB、GRO、IL−1RA、IL−8、VEGF、及び、RANTESなどのサイトカインを、有意なレベル(平均>50pg/mL)で含むことを示す。pExoは、EGF、Flt−3L、IFNa3、MCP−3、PDGF−AA、IL−15、sCD40L、IL6、IP−10、MCP−1、MIP−アルファ、MIP−1ベータ、及び、TNF−アルファなどのその他のサイトカインも、検出可能なレベル(5pg/mL〜49pg/mL)のサイトカインとして含む。

3つのpExo試料を、プロテオーム分析に供した。提出を受けた試料を、超音波プローブ(QSonica)を使用して:振幅40%、パルス10×1秒オン、1秒オフに設定して溶解した。そのタンパク質濃度を、Qubitフルオロメトリーで測定した。10ugのそれぞれの試料を、SDSページで処理し、そして、精製したタンパク質を、トリプシン消化した。表5は、それぞれの試料で同定した総タンパク質を示す。これらの試料では、合計で1814のタンパク質を同定している。表6は、pExo試料において同定頻度が大きかったタンパク質の識別子と遺伝子IDを示す。さらなるデータを、図4及び図5に示す。

3つのpExo試料を、配列決定して、それらのRNAプロファイルを分析した。要するに、pExo試料由来のRNAを抽出し、そして、cDNAに変換し、そして、配列決定する。次いで、当該配列データを、データベースと比較して、それぞれの配列データのタイプと実体を同定する。表7に、RNA配列決定結果の全体的なプロファイルを示す。pExoのRNAは、tRNA、ミクロRNA、及び、その他の非コードRNAのカテゴリを含む。ミクロRNAは、pEXO試料の構成において2番目に豊富なRNAである。これら3つのpExo試料では、合計で1500の異なるミクロRNAが同定されている。一部は、3つすべての試料に一般的に存在しており、また、一部は、1つ、または、2つの試料に特異的に認められている。最も高頻度のミクロRNAの遺伝子識別子、及び、相対的頻度、及び、存在量を示す。ミクロRNAは、細胞間連絡の機能において重要な役割を果たすことが知られている。

サイトカインプロファイルは、pExoが、走化性成長因子を含むことを示しており、このことは、pExoが、細胞移動を促す機能を備えていることを示唆している。このことを調べるために、トランスウェル移動アッセイを、次のように設定した:750uLのDMEM基本培地(無血清)を、トランスウェル(24ウェル)プレートの下部チャンバーに置き、pExoを、50uLで加えた。PBSを、対照として、同量を加えた。1×10e5 HDFを、当該トランスウェルの上部チャンバー(8um細孔)に播種した。6〜24時間後に、当該トランスウェルの上部チャンバーの細胞を、綿棒を使って取り除いた。次いで、当該トランスウェルを、1%エタノールを含むPBSを含んだ溶液で固定し、1%エタノール−PBSに溶解した1%クリスタルバイオレットで染色する。移動した細胞を、顕微鏡で可視化する。このデータは、HDFが当該トランスウェルの底面に向けて移動するのに対して、PBS対照トランスウェルでは、ウェルを通って移動する細胞が大幅に少ない、という結果の例を示す。この研究は、pExoが、ヒト皮膚線維芽細胞の移動を促すことができる、ことを実証している。図6を参照されたい。
トランスウェル移動アッセイも、次のように設定した:750uLのDMEM基本培地(無血清)を、トランスウェル(24ウェル)プレートの下部チャンバーに置き、pExoを、50uLで加えた。PBSを、対照として、同量を加えた。2×10e5 HUVEC発現GFPタンパク質を、当該トランスウェルの上部チャンバー(8um細孔)に播種した。6〜24時間後、移動したウェルを倒立蛍光顕微鏡(AMG)で、直接に可視化する。この研究は、試験した3つすべてのpExo試料が、3連のすべてのウェルでHUVECの移動を促すことができる、ことを実証している。陽性対照として使用するHUVECの完全培地では、細胞移動が顕著であり、また、さらなる対照として使用するPBSは、完全培地、または、pExo試験済ウェルと比較して、細胞移動がわずかである。図7を参照されたい。
pExoのサイトカインプロファイルは、それが、HUVECの増殖に関与することが公知である幾つかの増殖因子(PGDF−AA、BB、VEGF)を有することを示す。HUVECの成長と増殖に関するpExoの影響を調べるために、HUVEC発現GFPを、100uLのHUVEC増殖完全培地が入った96ウェルプレート(透明な底部と、不透明な壁)に、1×10e4細胞を播種した。2時間播種した後に、細胞を、ウェルの底に付着させた。次いで、これらのウェルに、25uLの異なるpExo試料(N=6/試料)を加える。次いで、プレートリーダーを使用して、当該プレートを、播種をして0日後と2日後に、蛍光強度で評価する(Synergy H4、励起395nm/発光509nm)。図13に示すように、完全培地は、0日目から2日目で、GFPシグナル(細胞数の指標)が強くなっていることが実証されている。完全培地を50%希釈するPBS対照は、完全培地と比較して、わずかな増殖を示した。8つの異なるpExo試料は、すべて、2日目にGFPの堅実な成長を示した。図8を参照されたい。
造血幹細胞の増殖に関するpExoの効果を試験するために、ヒト臍帯血CD34+細胞(自社で調製した)を解凍し、そして、1×10e4/細胞/ml(N=4)で、10% FCS−IMDMを含む、SCF、Flt−3、KL(培地A)のカクテルを含有する増殖培地で培養した。培養ウェルに、25uLのPBS、または、25uLのpExo試料(試験した2つのpExo試料)を加えた。1週間の培養後に、それぞれのウェルでの総細胞数を計数し、そして、当該培養物でのCD34+細胞の割合を、抗CD34抗体を使用したフローサイトメトリー(FACS)で評価した。CD34+総細胞数を、培養物でのCD34+細胞の%に対するウェル内の細胞総数として計算する。これらの結果は、pExoで処理した両方の培養物が、PBS対照培養物と比較して、有意に多くのCD34+細胞を有することを示した。また、pExoは、コロニー形成単位培養(CFU)でのCD34+細胞への影響についても試験した。CFU培養は、MethoCult H4434培地(Stem Cell Technologies)で確立されたものであり、そして、pExoまたはPBSを、50uL/mLで添加した。2週間の培養後に、それぞれの35mmディッシュでの合計CFU数を計数する(N=3)。このデータは、pExoの存在下で、PBS対照培養と比較して、有意に多くのCFUが存在することを示した。図9及び図10を参照されたい。
ミクロRNAデータ、及び、サイトカインデータは、pExoが、がん細胞増殖を阻害する活性を有することを示唆している。pExo試料を使用して、96ウェルプレートでのSKOV3(ヒト卵巣癌細胞株)の増殖に関する影響を調べた。このSKOV3細胞は、ルシフェラーゼを発現するように遺伝子操作しているので、ルシフェラーゼ活性の測定が、細胞増殖の指標となる。合計で8つの異なるpExo試料を使用した。2000個のSKOV3細胞を、100uLの増殖培地(DMEM−10%FCS)の入った96ウェルプレートに加えた。2時間後に、40uLのpExoを、ウェル(N=6)に加え、そして、60uLの増殖培地を補充した。40μLのPBSを、対照として使用した。完全培地の条件は、ウェルに対して100μLの培地を加えることである。インキュベーターで2日間培養した後に、ルシフェラーゼの活性を、Luciferase Assay Kit(Promega)で、細胞を溶解して測定し、そして、ルシフェラーゼ活性を、プレートリーダー(Synergy H4)を用いてルミネセンス発光で測定した。このデータは、それぞれの細胞濃度で、pExo処理した培養物が、PBS対照と比較して、ルミネックス指標が有意に小さいことを示す。このデータは、pExoが、SKOV3細胞の増殖を阻害したことを示す。図11を参照されたい。
免疫細胞に関するpExoの効果を調べるために、ヒト臍帯血T細胞を、PKH蛍光色素で標識し、次いで、刺激となるpExoまたはPHAとインキュベートした。RPMI+10%FCSで5日間培養した後に、すべてのT細胞、ならびに、CD4、CD8、CD69、CD27などの様々なタイプのT細胞のサブタイプを区別できる抗体を利用するFACSで、細胞を分析する。このデータは、pExoの存在下で、CD3+細胞のMFIが、対照培養と同様であることを示しており、このことは、pExoだけでは、T細胞に関する増殖活性に影響を与えないことを示す。PHA刺激で、MFIは有意に減少しており、PHAとpExoの両方の存在下で細胞が増殖したことを示しているが、当該MFIはPHA単独と同様であるので、細胞増殖が、pExoの存在による影響を受けないことを示す。CD69+細胞は、pExoで処理した細胞で有意に多く、CD69+細胞は、CD3+細胞(T細胞)で有意に増加させており、このことは、T細胞の活性化を、pExoが高めたことを示す。この観察は、臍帯血T細胞と、PBMC細胞の両方で認められた。加えて、pExoが、PBMCでのCD56+細胞(NK)細胞の割合を高めることが認められた。図14、図15、図16、及び、図17を参照されたい。
胎盤かん流液、及び、PRP(臍帯血血清)を、培養したヒト胎盤組織と同じ方法で単離した。以下の表は、胎盤かん流液、及び、PRPに由来するエキソソームの収量が、培養した胎盤よりも有意に少ないことを示す。

本発明の方法は、独特かつ有利な特性を有する大量のエキソソームを生成することができる。これらのエキソソームは、実証したエキソソームの機能によって生物活性であると考えられる数多くのタンパク質、及び、RNAを含むことが明らかになっている。これらのエキソソームは、数多くの細胞表面マーカーを発現しており、それらは、例えば、受容体またはリガンドなどの結合パートナーとして機能し得るものであり、それにより、この生物活性を、所望の細胞型に対する標的とすることができる。
本開示は、本明細書に記載した特定の実施形態によって、範囲が限定されることはない。実際のところ、本明細書で提供する内容の様々な変更は、これまでに説明したものに加えて、前出の説明、及び、添付した図面から当業者に自明である。そのような変更が、添付した特許請求の範囲に属するものである、ことを意図している。
Claims (79)
- 胎盤、または、その一部からエキソソームを単離する方法であって、前記方法は:
a)胎盤、または、その一部、好ましくは、培養胎盤、または、その一部を、第1の培地と接触させること;及び
b)前記胎盤、または、その一部からエキソソームの集団を含む第1の画分を得ること;
c)任意に、前記胎盤、または、その一部を、第2の培地と接触させ、かつ、前記胎盤、または、その一部に由来するエキソソームの集団を含む第2の画分を得ること;
d)任意に、前記胎盤、または、その一部を、第3の培地と接触させ、かつ、前記胎盤、または、その一部に由来するエキソソームの集団を含む第3の画分を得ること;及び
e)任意に、前記第1、第2、及び/または、第3の画分に由来するエキソソームの集団を、好ましくは、連続遠心分離、及び/または、所望のエキソソームの集団に存在するマーカー、または、ペプチドに特異的な抗体、または、その結合部分を使用するアフィニティークロマトグラフィーで単離すること、を含み、前記抗体、または、その結合部分を、膜、樹脂、ビーズ、または、容器などの基質に固定している、前記方法。 - 前記胎盤、または、その一部が、羊膜をさらに含む、請求項1に記載の方法。
