本発明では、1つ以上の非天然アミノ酸を有している、抗CD3抗体分子を提供する。本発明の特定の実施形態において、1つ以上の非天然アミノ酸を有する抗CD3抗体は、1つ以上の翻訳後修飾を有している。一実施形態において、1つ以上の翻訳後修飾には、分子の付加が含まれている。このとき付加される分子としては、水溶性ポリマー、ポリエチレングリコール誘導体、薬物、第2のタンパク質またはポリペプチドもしくはポリペプチドアナログ、抗体または抗体断片、生物活性を有する薬剤、小分子、または上記の任意の組み合わせ、または他の任意の望ましい化合物または物質、が挙げられる(ただし、これらに限定されない)。これらの分子は第2の反応基を有しており、第1の反応基を有している上記1つ以上の非天然アミノ酸と反応する。この反応は、特定の反応基に適していると当業者に知られている化学的方法による。例えば、第1の反応基はアルキニル部分であり(例えば、非天然アミノ酸であるp−プロパルギロキシフェニルアラニン(プロパルギル基は、時にアセチレン部分とも呼ばれる)中のものがあるが、これには限定されない)、第2の反応基はアジド部分である。この例では、[3+2]環状付加の化学的方法が利用される。他の例においては、第1の反応基はアジド部分であり(例えば、非天然アミノ酸のp−アジド−L−フェニルアラニン中のものがあるが、これには限定されない)、第2の反応基はアルキニル部分である。本発明の修飾された抗CD3抗体ポリペプチドの特定の実施形態では、1つ以上の翻訳後修飾にある糖部分において、1つ以上の翻訳後修飾を有している1つ以上の非天然アミノ酸(例えば、ケト機能性基を有する非天然アミノ酸が挙げられるが、これには限定されない)が使用されている。特定の実施形態において、翻訳後修飾は、真核細胞または非真核細胞中で、in vivoにてなされる。
抗CD3抗体の活性は、標準的なin vitroまたはin vivoのアッセイによって測定することができる。例えば、抗CD3抗体と結合する細胞または細胞株(例えば、天然の抗CD3抗体抗原または結合対象を含んでいる細胞、または、抗CD3抗体抗原または結合対象を組み換えにより産生する細胞であるが、これらには限定されない)を、抗CD3抗体の結合を評価するために用いることができる。非天然アミノ酸を有している、PEG化されていない抗原結合性ポリペプチドまたはPEG化抗原結合性ポリペプチドに関しては、抗CD3抗体の抗原または結合対象に対する当該抗CD3抗体の親和性を、公知の技術を用いて測定することができる(例えば、BIAcore(商標)biosensor(Pharmacia)など)。
本発明の重要な態様は、抗CD3抗体(抗CD3抗体と水溶性ポリマー部分との複合体化を伴うものも、伴わないものも含む)の構築によって獲得された、生物学的半減期の延長である。抗CD3抗体の血清濃度は急速に低下するため、複合体化もしくは非複合体化抗CD3抗体およびその変異体による処置に対する、生物学的応答の評価が重要となっていた。好ましくは、本発明の複合体化もしくは非複合体化抗CD3抗体およびその変異体は、静脈内投与の後でも血清半減期を延長させる。このため、例えばELISA法または一次スクリーニングアッセイによる測定が可能となる。in vivoにおける生物学的半減期の測定は、本明細書に記載の方法で行われる。
本発明のポリペプチドまたはタンパク質(1つ以上の非天然アミノ酸を有している抗CD3抗体、合成酵素、タンパク質などであるが、これらには限定されない)を、任意で、治療用途に採用してもよい(例えば、適切な薬物学的キャリアと組み合わせて採用するが、これには限定されない)。このような組成物は、例えば、治療上有効な量の化合物と、薬学的に許容可能なキャリアまたは賦形剤とを含んでいる。このようなキャリアまたは賦形剤としては、生理食塩水、緩衝食塩水、デキストロース、水、グリセロール、エタノールおよび/またはこれらの組み合わせが挙げられる(ただし、これらに限定されない)。製剤は、投与の形態に合うように成される。一般的に、タンパク質を投与する方法は本技術分野においてよく知られており、本発明のポリペプチドの投与にも適用することができる。
本発明の抗CD3抗体ポリペプチドは、広範な種類の障害の処置に有用である。抗CD3抗体アゴニストまたはアンタゴニストの使用による効果を受けうる障害に苛まれているヒト患者に対して、種々の手段で投与した際に効果的な強さとなるように、抗CD3抗体を含んでいる薬学的組成物を製剤することができる。抗CD3抗体アゴニストまたはアンタゴニストによる効果としては、抗増殖、抗炎症、抗ウイルスなどが挙げられる(ただし、これらに限定されない)。この効果は、状態または疾患そのものに対するものでもよいし、状態または疾患の一部に対するものでもよい。抗CD3抗体の平均量は変化しうる。とりわけ、適格な医師の推奨および処方に基づいて変化させるべきである。抗CD3抗体の正確な量は、処置する状態の正確な種類、処置する患者の状態、および組成物中の他の成分などの前提要因を満たすならば、選好の問題である。本発明はまた、治療上有効な量の他の有効成分(抗癌用化学治療薬など)の投与も提供する。当業者ならば、抗CD3抗体を用いる治療に基づいて、投与量を容易に決定することができる。
本実施例では、抗CD3抗体中に天然にコードされていないアミノ酸を組み込むにあたって、好適な部位を選択するための、数多くある可能な基準群のうちの1群について説明する。
すなわち、残基は、
(a)抗CD3抗体の結合を三次元構造の構造解析に基づいて妨げてはならず、また、抗CD3抗体の二次構造、三次構造または四次構造に基づいて妨げてもならない;
(b)アラニンまたはホモログを用いたscanning mutagenesisによる影響を受けてはならない;
(c)表面に露出していなくてはならず、周囲の残基との間におけるvan der Waals相互作用または水素結合相互作用は最小限にとどめねばならない;
(d)抗CD3抗体の露出表面の1箇所以上であってもよい;
(e)第2の抗CD3抗体、または他の分子もしくはその断片の近位に位置する、1または複数の抗CD3抗体の部位であってもよい;
(f)抗CD3抗体変異体においては、欠失しているか、可変であるか、のいずれかでなければならない;
(g)天然にコードされていないアミノ酸と置換した結果、保存的に変化しうる;
(h)完全な構造の柔軟性または剛直性を望ましく変化させることによって、抗CD3抗体自体、または1つ以上の抗CD3抗体を含む二量体もしくは多量体の立体配置を調節してもよい;
(i)柔軟性の高い領域(highly flexible regions)または構造上固定された領域(structurally rigid regions)のいずれにあってもよい;
(j)相補性決定領域(CDR)にあってもよいし、なくてもよい。
本実施例では、天然にコードされていないアミノ酸を有している抗CD3抗体の、E. coliを用いたクローニングおよび発現について詳述する。
(パラアセチルフェニルアラニン(pAcF)による抑制)
公知の標準的なプロトコルに則って、E. coliのアンバー変異を抑制した。要約すると、E. coliのペリプラズムにおいて抗体断片(scFvおよびFab)を抑制するために、次の操作を施した。まず、発現ベクターコンストラクト(M. jannaschii由来の直交なチロシルtRNA合成酵素(MjTyrRS)をコードしているプラスミド)により、E. coli宿主細胞を形質転換させた。一晩後、LB(Luria-Bertani)培地またはSuperbrothを入れたフラスコ内で、振盪しながら細菌培地を1:100に稀釈した。そして、37℃にて、ODが0.8程度となるまで培養した。FabおよびscFvの発現を誘導した。また、パラアセチルフェニルアラニン(pAcF)を最終濃度4mMとなるように加えて、アンバーコドンを抑制した。培地を25℃にて一晩インキュベートした。
このアミノ酸に特異的である、M. jannaschii由来の直交なチロシルtRNA合成酵素を用いた以外は、pAcFと同様の方法によって、アンバー変異をpAcF誘導体(aa9. 2)によって抑制した。誘導の際に、aa9.2を4mM加えることによって抑制した。
遠心分離により細胞を回収し、100μg/mLのリゾチームを添加したペリプラズム放出バッファ(50mM NaPO4、20% スクロース、1mM EDTA;pH8.0)中で再懸濁した。その後、氷上で30分間インキュベートした。遠心分離の後、上清中の抗体断片を、当該抗体断片のHisタグを利用してProBindビーズ(Invitrogen; Carlsbad, CA)上に固定した。ビーズを結合バッファでよく洗った後、結合している断片を0.5Mのイミダゾールでビーズから溶離させた。精製した断片は、保存バッファ(50mM HEPES、150mM NaCl、10% グリセロール、5% スクロース;pH7.8)中で透析した。細胞質中で発現しているscFv断片の小規模な分析のために、15mLの培地中にいるE. coliを遠心分離により回収した。その後、10μg/mLのDNAアーゼを添加した1mLの溶解バッファ(B-PER, Pierce Biotechnology; Rockford, IL)中で再懸濁した。混合物を37℃にて30分間インキュベートし、Protein Loadingバッファ(Invitrogen; Carlsbad, CA)で1×に稀釈して、SDS−PAGEにより分析した。
本実施例では、カルボニルを有するアミノ酸の導入について詳述する。そしてこれに引き続く、アミノオキシを有するPEGとの反応についても詳述する。

本実施例では、天然にコードされていないアミノ酸を有している抗CD3抗体、およびPEG化抗CD3抗体の、in vitroおよびin vivoにおける活性の測定方法について説明する。
PBS/1% BSA(100μL)中で、細胞(3×106個)を1組ずつインキュベートする。インキュベーションは、(i)様々な濃度の標識付けしていない抗CD3抗体、抗CD3抗体またはネガティヴ・コントロール(体積:10μL)の存在下または非存在下、ならびに(ii)125I−抗CD3抗体(約100,000cpmまたは1ng)の存在下で、0℃にて90分間行う(総体積:120μL)。次に、細胞を再懸濁し、350μLのプラスチック製遠心チューブ中で氷冷したFCS(200μL)上に積層し、遠心分離する(1000g;1分間)。チューブの末端を切断してペレットを回収し、ペレットと上清とをそれぞれ別々にガンマカウンター(Packard)で測定する。
PEG−抗CD3抗体、非修飾抗CD3抗体およびバッファ溶液を、マウスまたはラットに投与する。本発明のPEG化抗CD3抗体は、非修飾抗CD3抗体と比較して、活性が向上しており、半減期も延長されていることが示されると予期される。
オスのSprague Dawleyラット(約7週齢)を使用する。投与日に、それぞれの動物の体重を測定する。それぞれ3匹ずつのラットに、体重1kgあたり100μgの非複合体化抗CD3抗体または複合体化抗CD3抗体サンプルを、尾静脈から静脈内注入する。注入から1分後、30分後、1時間後、2時間後、4時間後、6時間後および24時間後に、CO2により麻酔した状態で、それぞれのラットから500μLの血液を採取する。血液サンプルを室温で1.5時間保存した後、遠心分離によって血清を分離する(4℃、18,000×gにて5分間)。血清サンプルは、分析の日まで−80℃で保存する。サンプルを氷上で解凍した後、抗CD3抗体in vitro活性アッセイによって、血清サンプル中の活性な抗CD3抗体の量を求める。
天然にコードされていないアミノ酸を有しているPEG化抗CD3抗体の、ヒトにおける安全性および/または効果の臨床試験。
天然にコードされていないアミノ酸を有しているPEG化組み換えヒト抗CD3抗体を皮下投与した際の、同じ標的抗原に対して特異的である市販品と比較した、安全性および薬物動態を比較する。市販品の例としては、Herceptin、Bexxar、Campath、CEA-Scan、Enbrel、Erbitux、Humira、Myoscint、Prostascint、Raptiva、Remicade、ReoPro、Rituxan、Siμlect、Synagis、Verluma、Xolair、Zenapax、ZevalinまたはAvastinがある(すべて登録商標)。
18人の健康な協力者を、本研究に参加させる(年齢20〜40歳;体重60〜90kg)。被験者は、血液学上または血清化学上の臨床的に重要な臨床検査異常値を示さず、尿の毒性スクリーニング、HIVスクリーニングおよびB型肝炎表面抗原のいずれもが陰性である。被験者は、以下の徴候を呈していてはならない:高血圧;あらゆる原発性血液疾患の病歴;重篤な肝疾患、腎疾患、心血管疾患、胃腸疾患、腎尿路生殖器疾患、代謝性疾患、神経疾患の病歴;貧血または発作障害の病歴;細菌もしくは哺乳動物由来の産物、PEGまたはヒト血清アルブミンに対する既知の感受性;カフェイン含有飲料の常習的かつ重度の消費;他のあらゆる臨床試験への参加、または試験開始前30日以内の輸血もしくは献血への参加;試験開始前3箇月以内の抗CD3抗体への曝露;試験開始前7日以内の疾病への罹患;試験前の健康診断、または試験開始前14日以内の臨床試験評価における重篤な異常。被検者全員について、安全性の評価が可能である。また、薬物動態分析のための血液は、スケジュールに沿って採取する。試験の全体が、施設内倫理委員会の承認および患者の同意の下に行われる。
本試験は健康な男性協力者を対象とする、フェーズI、一施設、オープンラベル、ランダム化、2期クロスオーバー試験である。18人の被検者を、2種類の処置シーケンス群の一方にランダムに割り振る(1群あたり9人の被検者)。抗CD3抗体を、2回に分割した投与期間で、上腿に皮下ボーラス注入により投与する。投与するのは、それぞれ等価な投与量の、天然にコードされていないアミノ酸を有しているPEG化抗CD3抗体および選ばれた市販品である。市販品の投与量および投与頻度は、添付文書の指示に従う。市販品を使用する、追加の用量、投与頻度または他の所望のパラメータを、追加の被検者群を含めることによって、本試験に追加してもよい。各投与期間の間には、14日間のウォッシュアウト期間が挟まれている。2回の投与期間のそれぞれについて、投与の12時間以上前〜投与後72時間の間、被験者を試験施設に拘束する。ただし、投与期間の間には拘束しない。PEG化抗CD3抗体についても、追加の用量、投与頻度または他のパラメータを試験するならば、さらなる被検者群を追加してもよい。抗CD3抗体の実験対象となる製剤は、天然にコードされていないアミノ酸を有しているPEG化抗CD3抗体である。
抗CD3抗体の投与前後に、静脈への直接穿刺によって一連の血液を採取する。血清中の抗CD3抗体濃度を検査するために、(i)投与前約30分、20分および10分において(3つのベースラインサンプル)、ならびに、(ii)投与後約30分、1時間、2時間、5時間、8時間、12時間、15時間、18時間、24時間、30時間、36時間、48時間、60時間および72時間において、静脈血サンプル(5mL)を採取する。それぞれの血清サンプルを2等分する。全ての血清サンプルを−20℃で保存する。血清サンプルは、ドライアイス中に入れて輸送する。空腹時の臨床検査試験(血液検査、血清化学検査および尿検査)を、1日目の初回投与直前、4日目の朝、16日目の投与直前、および19日目の朝に実施する。
ラジオイムノアッセイ(RA)法またはELISAキット法により、血清中の抗CD3抗体濃度を決定する。
各投与(1日目および16日目)の直前、ならびに各投与の6時間後、24時間後、48時間後および72時間後に、バイタルサインを記録する。安全性の判定は、有害事象の発生および類型、ならびに臨床検査試験のベースラインからの変化に基づく。さらに、バイタルサイン(血圧を含む)の測定結果および健康診断結果の、試験前からの変化も評価する。
投与後の値のそれぞれから、ベースライン抗CD3抗体濃度の平均値を減じることによって、投与後の血清濃度の値を投与前のベースライン抗CD3抗体濃度で較正する。ベースライン抗CD3抗体濃度の平均値は、投与前30分、20分および10分に採取した3つのサンプルに由来する抗CD3抗体レベルの平均値から求める。投与前の血清中抗CD3抗体濃度がアッセイの定量レベルに満たない場合には、これを平均値の計算に含めない。薬物動態学的パラメータは、ベースライン抗CD3抗体濃度で較正した血清濃度データから決定する。Digital Equipment Corporation VAX 8600コンピュータシステムで最新バージョンのBIOAVLソフトウェアを用い、モデルに依存しない方法によって、薬物動態学的パラメータを計算する。以下の薬物動態学的パラメータを求める:ピーク血清濃度(Cmax);ピーク血清濃度に達した時間(tmax);線形台形則で計算した、時刻0〜最後の血液サンプリング時(AUC0−72)における濃度−時間曲線下面積(AUC);消失速度定数から計算した、消失半減期(t1/2)。消失速度定数は、対数線形な濃度−時間プロットの末端線形領域における連続するデータ点を、線形回帰することによって推定する。それぞれの処置について、薬物動態学的パラメータの平均、標準偏差(SD)および変動係数(CV)を計算する。パラメータの平均値の割合(保存されている製剤/保存されていない製剤)を計算する。
有害事象の発生は、処置群の間で均等に分布している。ベースライン、試験前の臨床検査試験または血圧からの、臨床的に重要な変化は存在しない。また、試験前の健康診断結果およびバイタルサイン測定からの、特筆すべき変化も存在しない。2つの処置群の間で、安全性プロファイルは類似していなくてはならない。
18人の被検者全員について、同じ標的抗原に特異的な1種類以上の市販品を1回投与した後の血清中抗CD3抗体濃度の平均−時間のプロファイル(ベースライン抗CD3抗体レベルでの較正前)を、同じ時点において測定した天然にコードされていないアミノ酸を有しているPEG化抗CD3抗体のものと比較した。全ての被検者の投与前ベースライン抗CD3抗体濃度は、正常な生理的範囲に収まっていなければならない。投与前のベースライン抗CD3抗体濃度の平均から得た血清データによって、薬物動態学的パラメータを決定する。その後、Cmaxおよびtmaxを決定する。選ばれた1または複数の臨床的比較対象のtmaxの平均は、天然にコードされていないアミノ酸を有しているPEG化抗CD3抗体のtmaxよりも有意に短い。試験に供した市販の抗CD3抗体製品の消失半減期の値は、天然にコードされていないアミノ酸を有しているPEG化抗CD3抗体の消失半減期と比較して有意に短い。
FR+細胞株およびFR−細胞株を用いて、抗CD3Fab−葉酸複合体の親和性および特異性を評価した。葉酸を含まない培地中での培養後にフローサイトメトリー測定を行ったところ、上咽頭癌細胞株(KB)および卵巣癌細胞株(OV−90、SKOV−3)は、FRを過剰発現していた。FRを発現していない腎臓癌細胞株(CAKI−1)および肺胞基底上皮細胞腺癌(A549)を、ネガティヴ・コントロールとした。抗CD3Fab−葉酸複合体は、KB細胞(FR+)に対して、葉酸と複合体化していない抗体と同等の親和性での結合能を有していた。このことは、抗CD3Fabとの複合体化によっては、葉酸受容体に対する結合がほとんど失われないことを示している。同様に、抗CD3Fab−葉酸は、Jurkat 細胞(CD3+)に対して、抗CD3と同等の親和性で結合した。一方、A549細胞(FR−)に対する結合は見られなかった。
次に、KB細胞およびOV−90細胞(FR+)を用いたin vitro細胞傷害アッセイによって、抗CD3Fab−葉酸複合体の活性を検討した。CAKI−1細胞およびA549細胞(FR−)を対照群とした。活性化ヒト末梢血単核細胞(PMBC)と共に細胞を培養し(標的細胞:効果細胞=1:10)、様々な濃度の抗CD3Fab−葉酸または抗CD3Fabのみで処置した。溶解した細胞から放出されるLDHレベルを測定し、さらにCellTiter-GloおよびFACSを利用した傷害アッセイを行って、細胞傷害を定量化した。PBMC存在下において、抗CD3Fab−葉酸複合体は、それぞれ10pMおよび100pMのEC50でKB細胞およびOV−90細胞を傷害した(図2A、附録図4)。しかし、CAKI−1細胞(図2A)およびA549細胞(附録図4)は、100nMの複合体によっても影響を受けなかった。抗CD3Fabのみによる処置では、検討した全ての細胞株において無視できる程度の傷害しか誘導されなかった(図2A)。さらに、SKOV−3細胞(図2B)またはKB細胞(附録図5)を、抗CD3Fab−葉酸複合体の存在下で、活性化PBMCと共に葉酸を含まない培地中で16時間インキュベートしたところ(標的細胞:効果細胞=1:10)、ロゼットが形成された。これは、T細胞による標的化について、さらなる証拠を提供するものである。これとは対照的に、抗CD3Fabは、T細胞による攻撃に影響を及ぼさず、PBMCの非存在下またはKB細胞における凝集は認められなかった(附録図5)。
次に、齧歯類において、抗CD3Fab−葉酸複合体および非複合体化抗CD3Fabの薬物動態を検討した。(i)1mg/kgもしくは5mg/kgの抗CD3Fab−葉酸(PBS中)、または(ii)1mg/kgの非複合体化抗CD3Fabを、3匹のラットに静脈内注入により1回投与した。そして、一定間隔で採取した血清を、ELISAで分析した。抗CD3Fab−葉酸も、対応する非複合体化抗CD3Fab変異体も、いずれも同じ速度で血清濃度が減少した(血清半減期:60分間、図3D)。したがって、抗CD3Fab−葉酸は、非複合体化抗CD3Fabと同様の薬物動態であった。このことは、複合体のPKプロファイルが、葉酸による誘導体化の影響を受けていないを示している。
次に、抗CD3Fab−葉酸二重特異性薬剤のin vivoにおける効果を、メスのNOD-SCIDマウスを用いた異種移植モデルで評価した。動物を低葉酸食で飼育して、二重特異性薬剤の競合するおそれのある、循環血清中の葉酸レベルを低下させた。KB細胞(FR+)またはA549細胞(FR−)には、非活性化ヒトPBMC(標的細胞:効果細胞=1:100)または活性化ヒトPBMC(標的細胞:効果細胞=1:10)と混合した際に腫瘍形成能がみられ、この混合物をマウスに皮下注射した。したがって、ヒトPBMCと混合したKB細胞(標的細胞:効果細胞=1:10)をマウスに埋入した。その後、同じマウスに、1.5mg/kgの抗CD3Fab−葉酸またはPBSを静脈内注射した(腫瘍細胞の埋入と同じ日に開始し、10日間毎日続けた)。この投与レジメンは、抗CD3Fab−葉酸複合体の薬物動態学的特性と、二重特異性抗体に関する以前の研究10に基づいて選択されたものであった。PBSで処置したマウスにおいては、腫瘍体積の急激な増加が見られた(約5日間で倍増する速度)。これとは対照的に、抗CD3Fab−葉酸で処置したマウスの腫瘍は、研究の間を通して、辛うじて検出できる程度であった(図3A)。並行研究では、非活性化ヒトPBMC(標的細胞:効果細胞=1:100)と混合したKB細胞をマウスに埋入した。そして、1.5mg/kgの抗CD3Fab−葉酸またはPBSで静脈内処置した(腫瘍細胞の埋入と同じ日に開始し、10日間毎日続けた)。両群のマウスとも、研究の最初の30日間、腫瘍体積が減少した。これは恐らく、注入部位におけるPBMCの高負荷に起因すると思われる。しかし、腫瘍の減少速度は、抗CD3Fab−葉酸で処置したマウスの方が速かった。研究35日目以降、PBSで処置したマウスの腫瘍は、体積が徐々に増加していった。一方、抗CD3Fab−葉酸で処置したマウスの腫瘍は、辛うじて検出できる程度まで減少した。このことは、抗CD3Fab−葉酸複合体によれば、活性化T細胞の非存在下であっても、細胞傷害性T細胞の攻撃によって腫瘍が排除されることを示している。FRを発現している腫瘍に関して、抗CD3Fab−葉酸複合体のin vivoにおける選択性を検討するために、A549細胞(FR−)を非活性化ヒトPBMC(標的細胞:効果細胞=1:100)と混合して、マウスに埋入した。同様の方法でマウスを処置した(1.5mg/kgの抗CD3Fab−葉酸を、10日間続けて静脈内投与)。その結果、処置から20日後において、PBSで処置した腫瘍との有意差は認められなかった。上記の全実験を通じて、抗CD3Fab−葉酸で処置したマウスにも、PBSで処置したマウスにも、体重の減少または明白な毒性は認められなかった(図3C、附録図6B)。さらに、抗CD3Fab−葉酸を注入したマウスおよび注入していないマウスの、組織病理学的な分析を行った。抗CD3Fab−葉酸またはPBSで処置したマウスの腎臓染色像には、T細胞の侵入は認められなかった。また、他の組織(脾臓、肺および肝臓など)に関しても、処置群と対照群との間で、ヘマトキシリンおよびエオジン(H&E)染色像における明確な病理学的な差異も認められなかった。
標準的なペプチド合成装置を用いて、固相ペプチド合成(SPPS)を行った。ペプチドおよびペプチド複合体は全て、調製用逆相高速液体クロマトグラフィ(RP−HPLC)装置(Waters, xTerra C18 10μm; 19 x 250 mm)を用いて精製した。そして、分析用RP−HPLC(Waters, X-Bridge C18 5μm; 3.0 x 50 mm)および液体クロマトグラフィ/質量分析(LC/MS)によって分析した。Varian 400 MHz NMR分光器を用いて、1Hのスペクトルを得た。サンプルは、DMSO−d6/D2O中を移動させた。1Hのシグナルは全て、残留溶媒またはDMSO(2.50ppm)をレファレンスとして、ppmで記録した。スピン結合定数(Hz)と共に、s:一重線、d:二重線、t:三重線、q:四重線、m:多重線または分解不能、b:幅広線と記載した。タンパク質の質量は、Ambrx質量分析施設(La Jolla, CA)で得た。

- (i)第1結合ドメインと、および(ii)第2結合ドメインを有している、二重特異的に結合する分子であって、

(a)配列番号3のアミノ酸配列を有している、VHドメイン、および
(b)配列番号6のアミノ酸配列を有しているVLドメイン、
を有している、二重特異的に結合する分子。 - 上記結合する分子は、1つ以上の天然にコードされていないアミノ酸を有している、請求項1に記載の二重特異的に結合する分子。
- 上記天然にコードされていないアミノ酸は、p−プロパルギロキシフェニルアラニンまたはp−アジド−L−フェニルアラニンである、請求項2に記載の二重特異的に結合する分子。
- 上記天然にコードされていないアミノ酸は、水溶性ポリマーと連結している、請求項2または3に記載の二重特異的に結合する分子。
- 上記水溶性ポリマーは、ポリエチレングリコール部分を含んでいる、請求項4に記載の二重特異的に結合する分子。
- 上記結合する分子は、1つ以上の翻訳後修飾を有している、請求項1〜5のいずれか1項に記載の二重特異的に結合する分子。
- 上記結合する分子は、リンカー、ポリマーまたは生物活性を有する分子と連結している、請求項1〜6のいずれか1項に記載の二重特異的に結合する分子。
- 上記リンカー、ポリマーまたは生物活性を有する分子は、天然にコードされていないアミノ酸と結合しており、
上記天然にコードされていないアミノ酸は、リボソームによって、上記結合する分子の事前に選択した位置に組み込まれる、
