合わせたRX−3117の効果は、NSCLC細胞においてFACS分析を使用して調査した。1μMのRX−3117の比較的低い濃度では、S期の小さいがかろうじて有意な(p<0.05)細胞株依存性増大が、4つの細胞株のうちの3つにおいて見られた。すべての細胞株において、G1期における増大が見られ(図19A)、G2/M期の強い減少が見られた。4Gyの放射線は、S期において細胞の量の明らかな減少、SW1573細胞の両方においてはG2/M期における増大を引き起こし、A549細胞においては効果がなかったが、H460細胞においては減少を引き起こした(図19A)。放射線およびRX−3117の組合せは、両方のSW1573変異体においてS期の細胞の数の増大につながったが、H460細胞においては減少した。A549細胞において、細胞死滅(サブG1)は明らかに増大したが、他の細胞においては増大しなかった(図19A)。

た溶液を、18から23℃で、1H NMRによって完了するまで6時間にわたって撹拌した。反応混合物に8%w/wの炭酸水素ナトリウム(3.0体積)を投入し、18から23℃で5から10分間にわたって撹拌し(注意:穏やかな発熱)、相を分離させ、下部の水性相(2×2.0体積)を除去した。第1の8%w/wの炭酸水素ナトリウム(sodium hydrogen carbonated)抽出により、乳白色の水性層が得られ、静置時間の延長でエマルションは取り除かれなかった。調査は、エマルションが水性層に閉じ込められており、低い有機含有量を有することを示し、故に、プロセスを続けた。第1の抽出の全分離は、5時間29分であった。第2の8%w/wの炭酸水素ナトリウム抽出は、52分しかかからず問題なく分離した。水性相を2−メチルテトラヒドロフラン(2.0体積、1.7重量)で抽出し、ライン(line)を2−メチルテトラヒドロフラン(2.0体積、1.7重量)ですすいだ。INT12を含有する合わせた有機相を、40から50℃に加熱し、減圧下、40から50℃で約4体積に濃縮した。分析のために試料採取を実施し、水含有量が0.2%w/w以下となるまで、カール・フィッシャーによって分析した。プロセスは、99.0%の全収率に相当する、15.3%w/wのINT12 ASM11−アルコールを含有する2−メチルテトラヒドロフラン中、158.8kg正味重量のINT12 ASM11−アルコールを生じた。92.83%面積純度のINT12 ASM11−アルコール((3aS,4R,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−オール)を、HPLC分析によって決定した。
固定反応器内における段階4から5の連続反応によるINT14からのRX−3117一水和物の調製
本発明は、例えば、以下の項目を提供する。
(項目1)
腫瘍の治療における使用のための式(I)の化合物であって、式(I)の化合物
またはその水和物、溶媒和物もしくは薬学的に許容される塩を含む有効量の経口剤形が、それを必要とする対象に、約300〜2,000mg/日の投薬量で投与される、式(I)の化合物。
(項目2)
前記投薬量が、約500〜700mg/日である、項目1に記載の使用。
(項目3)
前記投薬量が、約6〜12mg/kg/日である、項目1または2に記載の使用。
(項目4)
前記経口剤形が、週に5から7日投与される、項目1から3のいずれか一項に記載の使用。
(項目5)
前記経口剤形が、連続する4週間にわたって週に5から7日投与されるか、または連続する3週間にわたって週に5から7日投与され、続いて、前記経口剤形が投与されない1週間の休薬となる、項目1から4に記載の使用。
(項目6)
投薬サイクルが、連続する3週間の治療に続く1週間の休薬、または連続する4週間の治療のいずれかからなり、前記経口剤形が、最大12回の投薬サイクルにわたって投与される、項目5に記載の使用。
(項目7)
前記経口剤形が、単回投与後、約700〜1,100ng/mLのCmaxをもたらす、項目1から6のいずれか一項に記載の使用。
(項目8)
前記経口剤形が、単回投与後、約8,000〜10,000時・ng/mLのAUC0〜t(0〜24時間)をもたらす、項目1から7のいずれか一項に記載の使用。
(項目9)
前記腫瘍が、膵臓、膀胱または結腸直腸がんである、項目1から8のいずれか一項に記載の使用。
(項目10)
放射線または抗腫瘍剤を前記対象に投与することをさらに含む、項目1から9のいずれか一項に記載の使用。
(項目11)
代謝拮抗物質、DNA断片化剤、DNA架橋剤、挿入剤、タンパク質合成阻害剤、トポイソメラーゼI毒、トポイソメラーゼII毒、微小管指向剤、キナーゼ阻害剤、ポリフェノール、ホルモン、ホルモンアンタゴニスト、死受容体アゴニスト、免疫チェックポイント阻害剤、抗プログラム細胞死1(PD−1)受容体抗体および抗プログラム細胞死リガンド1(PD−L1)抗体からなる群から選択される抗腫瘍剤を投与することをさらに含む、項目1から10のいずれか一項に記載の使用。

PD−L1抗体を前記対象に投与することをさらに含む、項目1から10のいずれか一項に記載の使用。
(項目13)
PD−1抗体を前記対象に投与することをさらに含む、項目1から10のいずれか一項に記載の使用。
(項目14)
前記経口剤形が、固体である、項目1から13のいずれか一項に記載の使用。
(項目15)
前記経口剤形が、錠剤である、項目1から13のいずれか一項に記載の使用。
(項目16)
前記経口剤形が、カプセル剤である、項目1から13のいずれか一項に記載の使用。
(項目17)
前記対象が、ヒトである、項目1から13のいずれか一項に記載の使用。
(項目18)
治療を必要とする対象の、式(I)の化合物
またはその水和物、溶媒和物もしくは薬学的に許容される塩による治療の有効性を予測する方法であって、
(i)前記対象から腫瘍細胞または組織の試料を収集するステップと、
(ii)前記腫瘍細胞または組織におけるUCK2発現のレベルを測定するステップと
を含み、ここで、UCK2の発現レベルが、式(I)の化合物による治療の有効性の可能性を示す、方法。

4−アミノ−1−((1S,4R,5S)−2−フルオロ−4,5−ジヒドロキシ−3−(ヒドロキシメチル)シクロペンタ−2−エン−1−イル)ピリミジン−2(1H)−オン1H2O(RX−3117−MH)の調製のためのプロセスであって、いかなる中間体も単離せずに、1つを超えるステップを伴う連続プロセスにおいて、tert−ブチル(((3aR,4R,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イル)オキシ)ジフェニルシラン(ASM11)を4−アミノ−1−((3aS,4S,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イル)ピリミジン−2(1H)−オン(INT14)に変換するステップを含むプロセス。
(項目22)
tert−ブチル(((3aR,4R,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イル)オキシ)ジフェニルシラン(ASM11)を2−メチルテトラヒドロフランに溶解するステップと、
フッ化テトラ−n−ブチルアンモニウムを添加して、反応溶液中で((3aS,4R,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−オール)(INT12)を形成するステップと、
有機相中でINT12を回収するステップと
をさらに含む、項目21に記載のプロセス。
(項目23)
前記有機相中でINT12を回収するステップが、
前記反応溶液を水溶液で洗浄するステップと、
INT12を有する有機相から水性抽出物を分離するステップと、
前記水性抽出物を2−メチルテトラヒドロフランで洗浄して、前記水性抽出物からINT12を抽出するステップと、
抽出されたINT12を、INT12を有する有機相と合わせるステップと
を含む、項目22に記載のプロセス。
(項目24)
トリエチルアミン、および2−メチル(methly)テトラヒドロフラン中の塩化メタンスルホニルを、前記有機相中の前記INT12に添加して、第2の反応溶液中で((3aR,4R,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イルメタンスルホネート)(INT13)を形成するステップと、
DMSO中でINT13を回収するステップと
をさらに含む、項目22に記載のプロセス。
(項目25)
DMSO中でINT13を回収する前記ステップが、
DMSOを、INT13を伴う前記第2の反応溶液に添加するステップと、
少なくとも90%w/wの2−メチルテトラヒドロフランを蒸留によって除去するステップと
を含む、項目24に記載のプロセス。

2.5当量の炭酸セシウムおよびシトシンを、DMSO中の前記INT13に添加して、第3の反応溶液中で4−アミノ−1−((3aS,4S,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イル)ピリミジン−2(1H)−オン(INT14)を形成するステップ

酸を、INT14を伴う前記第3の反応溶液に添加して、4−アミノ−1−((1S,4R,5S)−2−フルオロ−4,5−ジヒドロキシ−3−(ヒドロキシメチル)シクロペンタ−2−エン−1−イル)ピリミジン−2(1H)−オン(RX−3117)を形成するステップと、
前記RX−3117を、メチルtert−ブチルエーテルおよび水で洗浄して、有機相およびRX−3117を有する水性相を形成するステップと、
前記RX−3117を精製して、RX−3117−MHを形成するステップと
をさらに含む、項目26に記載のプロセス。
(項目30)
前記洗浄するステップの前に、反応混合物にメタノールを投入し、前記反応混合物を蒸留して、検出されるアセトニドの面積が約1.0%未満になるまで前記アセトニドを除去するステップ
をさらに含む、項目29に記載のプロセス。

前記洗浄するステップが、
前記有機相から、RX−3117を有する水性相を分離するステップと、
RX−3117を有する水性相を、メチルtert−ブチルエーテルで、前記水性相中で検出されるトリチルアルコールが約0.5%w/w未満になるまで洗浄するステップと、塩基性アニオン樹脂を、RX−3117を有する水性相に添加して、スラリーを形成するステップと、
前記スラリーを濾過して、母液を保持するステップと、
前記母液を濃縮して、濃縮物を形成するステップと、
アセトニトリルを前記濃縮物に添加して、精製されたRX−3117−MHを形成するステップと
をさらに含む、項目29に記載のプロセス。
(項目32)
tert−ブチル(((3aR,4R,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イル)オキシ)ジフェニルシラン(ASM11)から4−アミノ−1−((3aS,4S,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イル)ピリミジン−2(1H)−オン(INT14)を調製するための連続プロセスであって、
前記ASM11を2−メチルテトラヒドロフランに溶解するステップと、
フッ化テトラ−n−ブチルアンモニウムを添加して、((3aS,4R,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−オール)(INT12)を形成するステップと、
トリメチルアミン、および2−メチルテトラヒドロフラン中の塩化メタンスルホニルを、前記INT12に添加して、((3aR,4R,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イルメタンスルホネート)(INT13)を形成するステップと、
炭酸セシウムおよびシトシンを、前記INT13に添加して、4−アミノ−1−((3aS,4S,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イル)ピリミジン−2(1H)−オン(INT14)を形成するステップと
を含み、
ここで、前記ステップが、INT12もINT13も単離せずに、1つまたは複数の固定反応器内で実施される、連続プロセス。

4−アミノ−1−((3aS,4S,6aR)−5−フルオロ−2,2−ジメチル−6−((トリチルオキシ)メチル)−4,6a−ジヒドロ−3aH−シクロペンタ[d][1,3]ジオキソール−4−イル)ピリミジン−2(1H)−オン(INT14)から4−アミノ−1−((1S,4R,5S)−2−フルオロ−4,5−ジヒドロキシ−3−(ヒドロキシメチル)シクロペンタ−2−エン−1−イル)ピリミジン−2(1H)−オン1H2O(RX−3117−MH)を調製するための連続プロセスであって、
前記INT14を酸と反応させて、4−アミノ−1−((1S,4R,5S)−2−フルオロ−4,5−ジヒドロキシ−3−(ヒドロキシメチル)シクロペンタ−2−エン−1−イル)ピリミジン−2(1H)−オン(RX−3117)を形成するステップと、
前記RX−3117を、メチルtert−ブチルエーテルおよび水で洗浄して、有機相およびRX−3117を有する水性相を形成するステップと、
前記有機相から、RX−3117を有する水性相を分離するステップと、
RX−3117を有する水性相を、メチルtert−ブチルエーテルで、前記水性相中で検出されるトリチルアルコールが約0.5%w/w未満になるまで洗浄するステップと、強塩基性アニオン樹脂を、RX−3117を有する水性相に添加して、スラリーを形成するステップと、
前記スラリーを濾過して、母液を保持するステップと、
前記母液を濃縮して、濃縮物を形成するステップと、
アセトニトリルを前記濃縮物に添加して、精製されたRX−3117−MHを形成するステップと
前記精製されたRX−3117−MHを単離するステップと
を含み、
ここで、前記ステップが、1つまたは複数の固定反応器内で実施される、連続プロセス。
