Abstract
Description
1) XBP-1 mRNAのIRE-1スプライシング
タンパク質フォールディング需要の増加からの、又はERプロセスの化学的阻害によるERストレスは、シャペロンBiPの解離を経て、ER膜貫通タンパク質IRE1により検出される。このBiPの解離は、タンパク質のオリゴマー化及びリン酸化を可能にすることによりIRE1を活性化し、それにより、ER内腔に面しているRNアーゼドメインが露出される。その後、RNアーゼドメインは、スプライソソーム非依存性様式でXBP-1 mRNAからの非標準イントロンの除去を触媒する。非ストレス条件下で、XBP-1 mRNAは、スプライシングされない(XBP1u(翻訳された場合、非機能性タンパク質である261アミノ酸のORFを形成する))。しかしながら、ERストレスが検出された場合、XBP1uは、IRE1 RNアーゼによりスプライシングされて、機能性376アミノ酸タンパク質をコードするXBP1sを形成する。この機能性タンパク質、XBP1は、シャペロン、ジスルフィドイソメラーゼ、及びリン脂質生合成に関与する酵素等のタンパク質ホメオスタシスに関与するいくつかの遺伝子の発現を調節する転写因子であり、特定の配列、ERストレス応答エレメント(ERSE)又はアンフォールディング型タンパク質応答エレメント(UPRE)と結合し、遺伝子発現を増強することにより、これらのプロセスを制御する。したがって、遺伝子発現の調節は、IRE1によるXBP1uからのmRNAイントロンの除去である。非常に類似した系が、非哺乳類の真核細胞において機能している。
2) ATF6

3) 二本鎖RNA活性化プロテインキナーゼ(PERK)
PERKは、ER内腔ストレスセンサー及び細胞質プロテインキナーゼドメインを含有する1型ER膜貫通タンパク質である。PERKは、ERストレスに応答して活性化され、真核生物開始因子(elF2a)をリン酸化によって不活性化することにより、哺乳類細胞のERにおける正常タンパク質翻訳を阻害する。
- イントロンは、イントロンの下流で(すなわち、3'方向で)フレームシフトを生じる可能性がある。これは、典型的には、イントロンの切除可能な配列が3ヌクレオチド長の倍数ではない場合に生じる。そのようなフレームシフトは、停止コドンの導入を生じる場合が多く、その結果として、切り詰められたタンパク質を生じる。他の場合では、それは単に、イントロンの下流のコードされたアミノ酸配列における全面的な変更を単純に生じ得る。
- タンパク質の機能に対して破壊的であるアミノ酸が、翻訳されたタンパク質に存在することになる、コード配列の導入。この場合、イントロンの下流のアミノ酸配列は変更されないが、スプライシングされていない型において、翻訳されたタンパク質に存在するだろう、イントロンによりコードされたアミノ酸配列が、タンパク質の機能を破壊する。

- 40mlの細胞を、250mlのベント型Erlenmeyerフラスコ(Sigma-Aldrich社 CLS431144)中、37℃°、8%CO2において、100rpmで撹拌しながら、増殖させた。細胞を、製造会社の使用説明書に記載されているように播種した。
- トランスフェクションの1日前に、細胞を、血球計算器を用いて計数し、500,000細胞/mlに分けた。
- トランスフェクションの当日、細胞を24ウェルプレートにおいて500μlの適切な培地中100万細胞/mlで播種する。
- その後、0.625μgのDNA/ウェルを、10μlのOptiMem培地(Thermofisher社; 11058021)に加え、室温で5分間、インキュベートした。
- 同時に、0.625μlのMax試薬を、OptiMemの添加により10μlにし、室温で5分間、インキュベートした。

- GFP構築物からのイントロンスプライシングを、2mM DTTの添加及び1時間後のGFP蛍光のモニタリングにより測定した。DTTは、ER内のジスルフィド結合の形成を阻止することによりERストレスを誘導する強い還元剤である。
- 細胞溶解バッファーを、ルシフェラーゼアッセイ系(Promega社; E1500)から、滅菌水で5分の1に希釈することにより調製した。
- 細胞を900xgで5分間、ペレット化し、ルシフェラーゼキットからの細胞溶解バッファーの100μl/1×106細胞中に再懸濁した。
- その後、これを、溶解を起こすために室温で10〜15分間、インキュベートした。
- 細胞核及びデブリを900xgで5分間、採取した。
- 上清を、96ウェル黒色プレートへ最大100μl/試料で収集した。
- 上清をPBSで2分の1に希釈した(これは、GFPシグナルに影響を及ぼす、溶解バッファー由来のメルカプトエタノールを希釈することである;GFPシグナルは還元剤の存在下で減少する)。
- 試料を室温で5分間、インキュベートした。
- GFP蛍光を、プレートリーダーを用いて測定し、励起及び発光をそれぞれ、485nm及び520nmに設定した。
- GFP蛍光をまた、蛍光顕微鏡下で直接、可視化した。
SYNP-XBP-01実験
HEK293F実験:
- 24ウェルのトランスフェクションを上記のように設定した。各条件を2連/3連で実施した。
- トランスフェクションから24時間後、細胞を2mM DTTによるか又は同量の水でのモック処理によるかのいずれかで処理した。
- 細胞の処理から1時間後、GFP蛍光を上記のように測定した。
- GFP蛍光の対照プラスミドに対する比を、誘導の前と後で計算した。図1は3つの独立した実験の累積結果を示す。
CHO-S細胞を用いる実験を、HEK293F細胞について記載されているように同一方法で実施した。実験の結果は、図2a及び図2bに見ることができる。
ATF6は、ERストレスにより活性化される転写因子である。いったん活性化されたならば、この転写因子はERSE又はUPREと結合し、タンパク質ホメオスタシスの重要な構成要素の遺伝子転写を活性化する(Yoshidaら、Cell、107巻、881〜891頁、2001年12月28日参照)。
1) CMV最小プロモーター(CMV-MP)の上流の6×ATF6エレメント(配列TGACGTGCTを有する)及びEGFP;この構築物は、SYNP-ATF6-01と名付けられた(下記の配列番号30参照)。
2) CMV-MPの上流の6×ATF6エレメント及びXBP1イントロン挿入を有するEGFP;この構築物はSYNP-ATF6-02と名付けられた(下記の配列番31参照)。

24ウェルのトランスフェクションを上記のように設定した。各条件を2連/3連で実施した。トランスフェクションから24時間後、細胞を、UPRを誘導するために2mM DTTでの処理か、又は同量の水でのモック処理によるかのいずれかで処理した。細胞の処理から1時間後、3時間後、5時間後、及び24時間後、GFP蛍光を、前に論じられているように測定した。
この例の目的は、ERストレス応答が、以前に用いられるような化学的ERストレスの代替として(又はそれに加えて)、異種性タンパク質の産生により活性化され得るかどうかを決定することであった。SYNP-ATF6-01及びSYNP-ATF6-02からのGFP発現がAAV産生中に測定される実験を実施した。
いくつかの候補作用物質のUPRを誘導する能力を評価するために実験を実施した。これらの候補誘導剤のUPRを誘導する能力を、本質的に上記のような技術を用いて評価した。試験された追加の候補は、0.5mM パルミチン酸、1uM MF-43(2-メチル-5-(6-(4-(2-(トリフルオロメチル)フェノキシ)ピペリジン-1-イル)ピリダジン-3-イル)-1,3,4-チアジアゾール)(ステアロイル-CoAデサチュラーゼの阻害剤である)、及びこれらの両方の組合せ(同じ濃度のそれぞれが用いられた)であった。

この実験は、制御可能なイントロンからの発現調節が、発現する細胞にとって致死性である毒性発現産物の発現を調節するのに適切に厳密であることを確認するために実施した。制御可能なイントロンを、アポトーシス誘発性タンパク質カスパーゼ9へ組み込んだ。HEK細胞におけるこのタンパク質の過剰発現は、迅速な死を引き起こす。したがって、このタンパク質の非修飾型配列からの少しの発現でも、培養物内で死細胞の有意な増加をもたらす。このことが、本発明者らが前記イントロンにより提供される調節の厳密性を決定するように促している。カスパーゼ9も、GFP-Sparkと融合させた(イントロンが転写産物に存在しているならば、目で見ることができない)。
発現のために、イントロンをCMV-IE構成的プロモーターと連結した。イントロン構築物を、BsaI制限部位及び2つの相補性オリゴを用いて、プラスミドベクターSYNP-CASPSp-001へクローニングした。そのプラスミドを、BsaI(NEB社 R0535S)で消化し、98℃へ5分間、加熱して二次構造を融解することにより、オリゴをアニールし、その後、55℃で20分間、インキュベートした。これは、オリゴが二本鎖DNAを形成することを可能にした。このDNAを、消化されたSYNP-CASPSp-001へのライゲーションを可能にするだろう、5'末端と3'末端の両方においてオーバーハングを有するようにデザインした。その後、二本鎖DNAをプラスミドへライゲーションし、続いて、one shot top ten chemically competent cells (Thermofisher社、C404003)へ形質転換した。単離されたDNAをシーケンシングして、イントロンの存在を確認した。完全なプラスミドは、SYNP-CASP9-INTと呼ばれ、プラスミドマップは図13に示されている。

- 治療的有効量の機能性治療用発現産物を対象の標的細胞において発現する工程。 - 合成のUPR応答性シス制御性エレメントを含む合成UPR誘導性プロモーターであって、前記UPR応答性シス制御性エレメントが、ATF6、XBP1、若しくはbZIP60、又はUPRの一部として遺伝子発現を駆動させるホモログ若しくは別途等価の転写因子に対する少なくとも1つの結合部位を含む、合成UPR誘導性プロモーター。
- 最小プロモーター又は近位プロモーターと作動可能に連結した少なくとも1つの合成UPR応答性シス制御性エレメントを含む、請求項43に記載の合成UPR誘導性プロモーター。
- 以下の転写因子標的配列の少なくとも1つの1つ又は複数のコピーを含むUPR応答性シス制御性エレメントを含む、請求項43又は44に記載の合成UPR誘導性プロモーター:
- TGACGTG;
- TGACGTGCT;
- TGACGTG[TG];
- CCAAT-N9-CCACG(ERSE1部位として知られている)(配列番号18);及び
- ATTGG-N-CCACG(ERSE2部位として知られている)(配列番号19)。 - 転写因子標的配列TGACGTGの1つ又は複数のコピー、好ましくは転写因子標的配列TGACGTGの3つ又はそれ以上のコピー、及び好ましくは転写因子標的配列TGACGTGの5つ又はそれ以上のコピー、例えば、転写因子標的配列TGACGTGの6つ又はそれ以上のコピーを含む、UPR応答性シス制御性エレメントを含む、請求項43〜45のいずれか一項に記載の合成UPR誘導性プロモーター。
- UPR応答性シス制御性エレメントが、配列
TGACGTG-S-TGACGTG-S-TGACGTG-S-TGACGTG-S-TGACGTG-S-TGACGTG(配列番号54)を含み、ただし、Sが任意選択のスペーサー配列を表す、請求項43に記載の合成UPR誘導性プロモーター。 - UPR応答性シス制御性エレメントが、配列TGACGTGCT-S-TGACGTGCT-S-TGACGTGCT-S-TGACGTGCT-S-TGACGTGCT-S-TGACGTGCT(配列番号55)を含み、ただし、Sが任意選択のスペーサー配列を表す、請求項43に記載の合成UPR誘導性プロモーター。
- UPR応答性シス制御性エレメントが、配列TGACGTGCTTGACGTGCTTGACGTGCTTGACGTGCTTGACGTGCTTGACGTGCT(配列番号20)を含む、請求項43に記載の合成UPR誘導性プロモーター。
- UPR応答性シス制御性エレメントが、配列
