Abstract

メラニンに特異的に結合する抗体が、本明細書において提供される。一部の実施形態において、メラニンは、哺乳動物メラニン、例えば、ヒトメラニン、又はマウスメラニンである。他の実施形態において、メラニンは、非哺乳動物メラニンである。

メラノーマの処置のための治療的使用のためのメラニン抗体が、本明細書において提供される。
本開示は、治療用メラニン抗体を含む組成物を提供する。一部の実施形態において、組成物は、無菌である。医薬組成物は、一般に、有効量の治療用抗体を、薬学的に許容される賦形剤において含む。
本明細書において提供されるメラニン抗体は、診断及び画像化の目的で使用することができる。用途に応じて、メラニン抗体は、インビボ又はインビトロで検出及び定量化され得る。

マウス−ヒトキメラ抗体を、8C3マウスモノクローナルIgGメラニン抗体から生成した(NCBI GenBank受託番号KX346264、Uran ME, Nosanchuk JD, Restrepo A, Hamilton AJ, Gomez BL, Cano LE. Detection of antibodies against Paracoccidioides brasiliensis melanin in in vitro and in vivo studies during infection. Clin Vaccine Immunol. 2011 Oct;18(10):1680-8)。キメラ抗体は、ヒト定常領域及びマウス可変領域を有する。キメラ8C3抗体は、本明細書において、「8C3キメラ」又は「キメラ8C3」又は「キメラ8C3 hIgG1」と互換可能に称される。
111インジウムでの放射標識のために、抗メラニン抗体(ヒト化8C3(HE−5、表6を参照されたい)、マウス8C3、及びキメラ8C3)及び対照IgG1抗体を、まず、標準的な方法を使用して、二官能性キレート剤CHXA”{N−[2−アミノ−3−(p−イソチオシアナトフェニル)プロピル]−trans−シクロヘキサン−1,2−ジアミン−N,N’,N”,N’’’,N’’’’−五酢酸}とコンジュゲートさせた。CHXA”リガンドは、抗体に対して2倍モル過剰で使用した。抗体を、次いで、標準的な方法に従って、111インジウムで放射標識した。111インジウムは、2μCi/μgの比放射能を有した。
すべての動物研究は、Animal Research Ethics Board of the University of Saskatchewanによって承認されていた。画像化研究のために、Charles River Laboratories社(USA)から入手した6週齢のC57BL6雌性マウスに、Matrigel(Corning社、USA)中の5×105個のB16−F10マウスメラノーマ細胞を、右側腹部へ皮下注射した。
このフォローアップ実施例は、マウスデータから仮説的に外挿した、ヒトにおけるBi−213及びLu−177の線量の結果を示す。以下に記載される方法は、マウスからヒトへ、放射線量の結果を外挿するための方法である。
外挿は、マウスモデルから、ヒトモデルの滞留時間を再計算し、OLINDA1.1など、MIRDスキームを実装するソフトウェアを使用してヒト線量を計算することによって行った。この方法は、種間における体重の違いに基づく比率性を想定している(Kirschner AS, Ice RD, Beierwaltes WH, “Radiation-dosimetry of I-131-19-iodocholesterol: J Nucl Med. 16:248-249; 1975)。
In−111トレーサー生物動態データ(減衰補正)を使用して、マウスにおけるBi−213及びLu−177からの放射線量を、In−111の代わりにBi−213又はLu−177のいずれかを想定して計算した。Bi−213及びLu−177について再計算した有効データをプロットし、プロットしたデータ点について最適適合数学関数を得、それぞれの源器官又は組織について最適適合関数を積分し、平衡線量定数及び比吸収割合を乗じた。
マウス器官の結果を、表10に示す。
213Bi/225Acジェネレーターを、Oak Ridge National Laboratory社(TN、USA)から購入し、177Lu塩化物については、Radiomedix社(TX、USA)から購入した。h8C3 HE−5抗体を、詳細な生体分布に記載されるように、CHXA”二官能性リガンドにコンジュゲートさせた。抗体を、放射標識する直前に213Biヨウ化物の形態で213Bi/225Acジェネレーターから溶出した213Bi、又は177Luで、放射標識した。213Bi又はLuによる抗体−CHXA”コンジュゲートの放射標識を行って、およそ5μCi/μgの比放射能の抗体を達成した。「高」(400μCi)線量の213Bi標識抗体又は177Lu標識抗体を調製するために、0.15M 酢酸アンモニウム緩衝液中の400μCiの放射性核種溶液を、80μgの抗体−CHXA”コンジュゲートに添加し、「低」(200μCi)の線量の213Bi標識抗体又は177Lu標識抗体を調製するために、0.15M 酢酸アンモニウム緩衝液中の200μCiの放射性核種溶液を、40μgの抗体−CHXA”コンジュゲートに添加した。213Biでの標識については、反応混合物を、37℃で5分間インキュベートし、177Luでの標識については、60分間インキュベートした。インキュベーションに続いて、3μLの0.05M EDTA溶液の添加によって、反応をクエンチした。放射標識の割合を、0.15M 酢酸アンモニウム緩衝液を溶離液として使用して、SG−iTLCによって測定した(上部は、遊離放射性核種を含有し、下部は、放射標識された抗体を含有する)。SG−iTLCを、Perkin Elmer社2470 Automatic Gamma Counterで読み取った。
比較処置と同じマウスメラノーマモデルを使用した。h8C3 HE−5抗体を、比較処置におけるように、213Biで放射標識した。腫瘍保持マウスを、8匹の群にランダム化し、0日目に、単回投与の400μCiの213−h8C3 HE−5、又は0日目及び3日目に、400μCiの213−h8C3 HE−5、又は0日目、3日目、及び7日目に、400μCiの213−h8C3 HE−5のいずれかで処置した。16日目に、単回投与群におけるマウスを、別の400μCiの213−h8C3 HE−5投与で処置した。腫瘍体積における変化を、図16A、16B、及び16Cに示す。マウス体重における変化を、図17に示す。白血球、赤血球、及び血小板の血球数の比較を、それぞれ、図18A、18B、及び18Cに示す。腎臓及び肝臓に対する全身毒性を、図19に示す。
マウスモデル及びh8C3 HE−5抗体の111Inでの放射標識を、記載されるように行った。microSPECT/CT(マイクロ単一光子放射型コンピュータ断層撮影法/コンピュータ断層撮影法)画像を、MILabs VECTor4 (Netherlands)microSPECT/CTスキャナで収集し、包括的画像分析ソフトウェアパッケージPMOD(バージョン3.9、PMOD Technologies, Inc社、Switzerland)を使用して処理した。CHXA”にコンジュゲートしたh8C3 HE−5で5:1mCi/mgの比放射能の200μCiの111Inを使用して、画像化研究を行った。2匹の腫瘍保持マウスに、尾静脈から静脈内注射を行い、注射の1時間後、24時間後、48時間後、72時間後、及び216時間後に、伏臥位置で画像化した(図20)。らせん状の軌道を使用して、20〜350keVの範囲についてExtra Ultra High Sensitivity Mouse(XUHS-M)コリメーターを使用し、SPECTデータを20分間収集した。すべてのSPECT画像は、MILabs再構築ソフトウェアを用いて0.4mmのボクセルグリッドにおいて245keV及び171keVの両方の111Inガンマ放出を使用して、再構築した。
CHO DG44宿主細胞に、h8C3 HE−5抗体をコードするベクターをトランスフェクトした。トランスフェクタントを選択し、限界希釈によって1回のサブクローニングに供した。3つのサブクローンを、研究用細胞バンク(「RCB(Research Cell Bank)」)の生成に選択し、次の通りに表記した:サブクローン−2−3H2、サブクローン−2−20C3、及びサブクローン−2−3H11。
DHFR欠損CHO DG44のトランスフェクション
本明細書に記載される組換え細胞株の宿主として使用するジヒドロ葉酸還元酵素(DHFR)欠損CHO DG44細胞株は、Columbia UniversityのLarry Chasin博士によって開発された、ヒポキサンチン及びチミジン(HT)の栄養要求体である。DHFR−CHO株は、CHO K1細胞株ATCC CCL−61のEMS及びγ−照射に誘導される変異により導出された。ATCC CCL−61細胞株は、1958年にTed Puck博士によって、チャイニーズハムスター卵巣組織から樹立された細胞株のプロリン栄養要求体である。Chasin博士は、2回のγ−照射を使用して、DHFR遺伝子の両方のアレルが完全に欠損した細胞株を産生した。
バルクプールは、比較的短期間(約3〜4週間)以内にCHO由来の材料を得る手段として生成した。1つのバルクプールを、静置フラスコにおいて、G418の漸増(0.25mg/mLから最終濃度0.5mg/mL)及びHT欠損培地での栄養要求性DHFR選択により生成した。バルクプールを、細胞生存率が約90%に回復した後、振盪フラスコに適合させた。
トランスフェクションの3日後に、トランスフェクトしたプールを、200μlの培地において、8mM Glutamaxを補充したCD−OptiCHO培地で栄養要求性DHFR選択下において、1ウェル当たり1,000個の細胞でミニプールに播種することによって(18×96ウェルプレート)、ミニプールを生成し、プレートを、37℃及び5% CO2で培養した。播種の3日後に開始し、ミニプールを、4週間の期間にわたる培地交換による、G418濃度(0.25mg/mLから最終濃度0.5mg/mL)及びメトトレキサート(MTX)(100nMから200nMから最終濃度400nM)の漸増に供した。細胞のコンフルエンスを、この期間中、顕微鏡によってモニタリングし、より高度な選択を、細胞増殖(すなわち、細胞コンフルエンスの増加)の際に適用した。約5週間後に、ヤギ抗ヒトIgG−Fc及びヤギ抗ヒトカッパ鎖−HRPを、それぞれ、コーティング抗体及び検出抗体として使用して、プレートをELISAによってアッセイした(図23)。
スーパープールを、流加培養振盪フラスコにおいて評価した。細胞を、250mlの振盪フラスコにおいて、8mM L−グルタミンを補充したCD−OptiCHO培地50mL中に、5×105個の細胞/mLで播種した。振盪フラスコを、125rpmで回転する25mmのオービタルスローを備えるシェーカープラットフォームにおいて、37℃及び5% CO2で培養した。培養物に、3日目、6日目、8日目、及び10日目の日ごとに、10mg/L Invitrogen社組換えヒトインスリン及び0.5% Cell Boost 7bを補充した5% Cell Boost 7aを供給した。グルコース及びL−グルタミンについて、モニタリング及び調節するために、NOVA読み取りを、3日目、6日目、8日目に、及び採取まで必要に応じて行った。細胞カウント、及び培養物の試料を、3日目、6日目、8日目、10日目に、及びそれ以降は採取まで毎日、取得した。培養物を、80%未満の生存率で採取した。増殖曲線及び生存率を、図26及び27に示す。スーパープール1は、スーパープール2及び3に由来する細胞よりも、振盪フラスコでの懸濁増殖への適合が遅く、結果として、2回の流加培養評価を、スーパープール1に行った。発現プロファイルを、以下の図28に示す。もっとも高い発現である792.3mg/Lは、スーパープール1の反復で得られ、スーパープール2は、462mg/Lを有した。
限界希釈及びELISAスクリーニングクローン
3つのスーパープールを、限界希釈法によってクローニングした。それぞれの培養物を、96ウェルプレートにおいて、0.5個の細胞/ウェルで播種した。それぞれのスーパープールについて、20個の96ウェルプレートに播種した。クローニング培地は、8mM Glutamax、2mMグルタミン、5μg/mLインスリン、1倍HT、及びバルクプール培養物から採取した等体積の条件培地を補充したCD OptiCHOから構成されていた。プレートを、37℃、5% CO2で、14日間、静置インキュベーターでインキュベートし、それぞれのウェルを、0日目、1日目、2日目、5日目、又は7日目、及び13日目又は14日目に、Solentim Imaging Systemによって画像化した。新しい培地100μLを、7日目にそれぞれのウェルに添加し、培地を、14日目に交換した。14日間又は15日間のインキュベーションの後、すべてのプレートを、ヤギ抗ヒトIgG−Fc及びヤギ抗ヒトカッパ鎖−HRPを、それぞれ、コーティング抗体及び検出抗体として使用して、ELISAによってスクリーニングした。
24ウェルプレートのステージにおいて特定された36個の上位発現体サブクローンのうちの35個は、振盪フラスコにおける懸濁増殖への適合が成功した。これらの上位サブクローンを、流加培養条件において、250mLの振盪フラスコにおける発現について評価した。振盪フラスコに、8mM L−グルタミンを補充したCD−OptiCHO培地50mL中に、5×105個の細胞/mLで播種した。振盪フラスコを、125rpmで回転する25mmのオービタルスローを備えるシェーカープラットフォームにおいて、37℃及び5% CO2で培養した。培養物に、3日目、6日目、8日目、及び10日目に、5%のCell Boost 7a及び0.5%のCell Boos 7bを供給した。グルコース及びL−グルタミンについて、モニタリング及び調節するために、NOVA読み取りを、3日目、6日目、8日目に、及び採取まで必要に応じて毎日、行った。一方で、細胞カウント、及び培養物の試料を、3日目、6日目、8日目、10日目、及びそれ以降採取まで毎日、取得した。培養物を、80%未満の生存率で採取した。細胞を、2500rpmで5分間遠心分離し、条件培地を移し、−20℃で保管した。
クローン2−3H11、2−3H3、及び2−20C3を、Solentimから得られた採取時のそれらの発現レベル及びクローン性に基づいて、研究用細胞バンク(RCB)の調製に選択した。
