本発明は、全体として、医薬、より具体的には、前脳基底部コリン作動性神経細胞(BFCN)、特に、プレセニリン2遺伝子(PSEN2)における1つまたは複数の変異に関連する電気生理学的異常が修復されたBFCN、を幹細胞から生成するための方法および組成物、並びにアルツハイマー病を治療するための細胞ベースの治療におけるかかるBFCNの使用の分野に関する。
本発明は、化学的に定義された培地を使用してPSCからBFCNを生成する、頑強、迅速、かつ再現性のある分化プロトコールの発見に基づく。
BFCNは、全ての形態のADにおいて影響を受ける第一の細胞タイプの1つであると考えられ、それらの機能障害は、短期記憶形成および想起の障害と臨床的に相関する。本開示の実施例において詳述される通り、本発明者らは、PSEN2変異のキャリアおよび対照に由来する細胞株を使用してiPSCからヒトBFCNを生成する、最適化されたインビトロプロトコールを提示する。PSEN2N141I変異を有する細胞株は、iPSCに由来するBFCNにおいてAβ42/40の増大を示した。PSEN2N141I株に由来する神経細胞が矩形の脱分極電流注入に応答して生じた活動電位の最大数はより少なかった。基電流注入における第一の活動電位の大きさもまた、PSEN2N141IBFCNにおいて有意に減少した。PSEN2点変異のCRISPR/Cas9修正は、電気生理学的異常を消失させ、対照において記録されたレベルまで活動電位の最大数と活動電位高の両方を回復させる。増大したAβ42/40はまた、PSEN2N141I変異のCRISPR/Casにより仲介される修正後に正常化された。本明細書に記載されているゲノム編集データセットは、電気生理学におけるPSEN2変異関連変化も示しながら、Aβ42/40比の変異関連変化の強い一貫性を立証する。
iPSCに由来するアルツハイマー病のPSEN2N141I変異における、CRISPR/Cas9により修正可能な変異に関連する分子表現型および生理学的表現型

iPSC株の作製および維持
7889(s)B、050643(対照)、948(AD1)、949(f対照)、および950(AD2)iPSC株を、[60]のガイドラインに従いNYSCF貯蔵所を介して得た。Nkx2.1−GFP ESC株の誘導および特徴決定は、既に公開されている[30]。ESおよびiPS細胞株の拡大増殖および維持は、血清を含まないmTeSR1(商標)培地(Stem Cell Technologies)中で行った。StemPro(商標)Accutase(ThermoFisher)を使用して細胞を剥離し、細胞継代中には10μM ROCK阻害剤(Y27632、Stemgent)を培地に追加した。
Aβ42オリゴマーを、既に報告されている通りに調製した[23、78]。簡単に言うと、本発明者らは、1,1,1,3,3,3−ヘキサフルオロ−2−プロパノール(HFIP)(Sigma)中にAβ42(American Peptide Company)1mgを溶解した。この調製物を等分し、SpeedVac(商標)遠心分離機を使用して乾燥させた。次に、ペレットを、DMSO中に再懸濁して、ウォーターバス中で10分間超音波処理した5mM溶液を得た。ここから、アリコートを−20℃で保存し、PBS 100μlで希釈し、オリゴマー形成を進めるために4℃で12時間放置することにより、2週間以内に使用した。この最終溶液を、研究用の細胞培地中で1:16に希釈し、細胞を5μM Aβ42オリゴマーに暴露した。対照ウェルは、1:16PBSを用いて希釈した。細胞を、3日間、培地を変更することなく、オリゴマーまたはPBSに暴露した。
細胞を、96ウェルプレートフォーマットでアッセイした。オリゴマーまたはビヒクル溶液を培地に加え、そのまま3日間インキュベーションした。次に、培地を集め、乳酸脱水素酵素毒性アッセイ(Thermo Fisher Scientific)を用いてアッセイした。培地50μlおよび等量の反応混合バッファーを、30分間インキュベーションした。1回の実験当たりウェルのさらなるセットを、2%トリトン(商標)X−100を用いて5分間インキュベーションして、全ての細胞を溶解し、これらのウェルの培地も集め、記載した通りインキュベーションした。インキュベーション後、吸光度を、それぞれ、490nmおよび680nmにおいてシグナルおよびバックグラウンド吸光度を記録した。シグナル値からバックグラウンドを引き、値を、トリトンX−100で処理したウェルにより決定した総LDH濃度に適合させた。ヨウ化プロピジウム(Thermo Fisher Scientific)を、1μMの最終濃度のため、細胞培地に加え、そのまま5分間インキュベーションした。次に、細胞を2回培地を用いて洗浄し、画像化した。CELIGO(商標)画像血球計算器および添付のソフトウエア(Nexcelom Bioscience)を使用して、画像を取得した。それぞれの生物学的変数を、それぞれの指定した「実験」において技術的トリプリケートで評価し、それぞれの指定した「実験」を、少なくとも3回の完全な「開始から終了までの」反復で行なった。
ヒトESまたはiPSCを、Accutase(商標)(Sigma−Aldrich)を用いて化学的に解離させた後、Cul−trex(商標)(Trevigen)コーティングプレートに、6ウェルプレートまたはペトリディッシュにおける1ウェル当たり4〜8×105個の細胞の密度にて、細胞数を合わせて、単一の細胞としてプレーティングした。細胞を、完全なコンフルエントに達するまで、mTeSR1(商標)培地(Stem Cell Technologies)においてまず維持した。分化の「0日目」に、培地を、多能性を促進する因子、すなわち、bFGF、TGF−β、Li−Cl、GABAおよびピペコリン酸を欠く、カスタムmTeSR1(商標)培地(Stem Cell Technologies)により置き換えた。0日目における二重SMAD阻害剤(10μM SB431542および250nM LDN193189、Selleckchem)の添加により、細胞を神経外胚葉特定化に向かわせた。分化の2日目において、培地を、二重SMAD阻害剤および2種の腹側化因子:500nM SAG(R&D)および2μM プルモルファミン(Stemgent(商標))を含有するカスタムmTeSR1により置き換えた。培地を、B27(Life Technologies)を追加したBrainphys(商標)培地(Stemcell Technologies)に次第にスイッチするとき、9日目まで、細胞に、2日毎にこの培地を与えた[3]。Accutaseを使用して、11日目に神経前駆細胞を回収し、p75+(CD271)NPCをFACSにより精製し、1ウェル当たり80,000個の細胞密度で、非接着性96ウェルV底プレートに、10μM ROCK阻害剤(Y27632、Stemgent)、神経成長因子、NGF、(Alamone labs、50ng/mL)および脳由来神経栄養因子、BDNF、(R&D、50ng/mL)を追加したBrainphys(商標)+B27においてプレーティングした。細胞を凝集させ、神経胚様体(NEB)を形成させ、19日目まで1日置きに加えた。19日目において、Accutase(Sigma−Aldrich)を使用して、NEBを解離させ、単層培養物として、BDNFおよびNGFを追加したBrainphys(商標)培地+B27中の分岐ポリエチニルイミン(.1%、Sigma−Aldrich)およびラミニン(10mg/mL、Life Technology)を用いてコーティングしたプレートにプレーティングした。解析まで、培地を2日毎に交換した。代替として、3D NEBを、拡大増殖のため3〜4個の小片に手動で解体し、さらに成長させるか、または凍結保存した。当初のバージョンのプロトコールは、Brainphys(商標)の代わりに基礎培地としてNeurobasal(商標)を使用した。
製造元の指示に従い、High Pure(商標)PCR鋳型調製キット(Roche)を使用して、PSEN2変異iPSC株または対照iPSC株からゲノムDNAを単離した。増幅に先立ち、ゲノム試料を、RNAse(QIAGEN)を用いて処理した。次のプライマー:

を使用して、PSEN2N141I変異を含有するPSEN2のエクソン5由来のフラグメントを増幅し、遺伝子型に関わらず、173bpのフラグメントを生じさせた。ApoE対立形質バリアントの検出のため、配列決定に先立ち、プライマー:

を使用して、244bpのフラグメントを増幅した。両方のPCRを、次の設定:94℃で10分;40サイクル(94℃で30秒、62℃で20秒、72℃で10秒);72℃で7分を用いて行なった。PCR産物を2%アガロースゲル中で泳動させて、増幅したフラグメントのサイズをチェックした。増幅後、EXOSAP−it(商標)(Thermo Fisher Scientific)を使用して、試料を洗浄し、次に、以下のプライマー:[53]からのPSEN2用プライマー(フォワード:

iAD1対照株およびiAD2対照株は、PSEN2WT/WTに対するPSEN2N141I/WTヘテロ接合性点変異のCRISPR/Cas9により仲介した修正により、948(AD1)および950(AD2)iPSC株から生じた。g1N141IシングルガイドRNA(sgRNA)を、pSpCas9(BB)−2A−GFP(PX458)ベクターにクローニングし、それにより、ボルガ変異が位置するPSEN2のエクソン5における配列に遺伝子編集を指示するためのpSpCas9−g1N141I−GFPベクターを生じる。一本鎖オリゴヌクレオチド(ssODN)は、CRISPR/Cas9修正のための効率的な鋳型である[13、66]。ssODN#A−N141I(配列の詳細は以下のとおり)を、遺伝子修正のためのドナー配列として使用した。本発明者らは、非対称なssODNが、CRISPR/Cas9を使用した相同性により指示される修復のより高い効率を示したので、91bpの長い相同性アームおよび36bpの短い相同性アームを有する非対称なssODN配列を設計した[68]。
分化の12日目における神経前駆細胞を、Accutase(Sigma−Aldrich)を用いて5分間37℃で解離させ、神経用基礎培地中で不活性化させた。細胞を1000rpmにて4分間スピンし、ペレットを、p75またはNGFRとしても公知のPEマウス抗ヒトCD271抗体(クローンC40〜1457、BD)を1:100で含むFACSバッファー(DPBS、0.5%BSA分画V溶液、100U/mLペニシリン−ストレプトマイシン、0.5%EDTAおよび20mMグルコース)中に再懸濁し、20分間、室温(RT)、暗所にてインキュベーションした。インキュベーション時間の後、FACSバッファーを用いて細胞を洗浄し、ペレットを、10μM ROCK阻害剤(Y27632、Stemgent)を含むFACSバッファー2mL中に再懸濁した。p75陽性細胞を、BD FACSAria IIu Cell Sorter(商標)にて精製し、データを、FlowJo(商標)ソフトウエアを使用して解析した。
PSEN2変異患者または対照患者に由来するヒトiPSCを、単層で増殖させ、RLTバッファーを含む細胞培養ウェル中で直接溶解した。総RNAの精製を、RNeasy(商標)マイクロキット(Qiagen)を用いて、製造元の指示に従って行なった。cDNAを、SuperScript(商標)III逆転写酵素(RT)(Invitrogen、Carlsbad、CA)を使用して合成した。半定量的リアルタイムPCRを、以下の表2において挙げるプライマーを使用して、StepOnePlus(商標)リアルタイムPCRシステム(Applied Biosystems、Foster City、CA)において行った。本発明者らは、GAPDHに対して発現レベルを標準化した。PCRサイクルパラメーターは、50℃で2分間、95℃で10分間、続いて95℃で15秒間、および60℃で1分間の40サイクルであった。それぞれの生物学的変数を、それぞれの指定した「実験」において技術的トリプリケートで評価し、それぞれの指定した「実験」を、少なくとも3回の完全な「開始から終了までの」反復で行なった。発現レベルを、対照株に対して標準化し、結果を、AVG±SEMとして表した。
細胞を、二重SMAD阻害の8日目の後に3日間条件付けて、インビトロで神経前駆細胞によるAβの分泌を測定した。ヒト/ラットβアミロイド40ELISAキットおよびβアミロイド42ELISAキット高感度(Wako)を使用して、Aβレベルを定量した。それぞれの生物学的変数を、それぞれの指定した「実験」において技術的トリプリケートを使用して評価し、それぞれの指定した「実験」を、少なくとも3回の完全な「開始から終了までの」反復で行なった。
細胞を、12、48または96ウェルプレートのウェルに直接、4%PFAを用いて20分間固定し、1×DPBS(ThermoFisher)を用いて3回洗浄した。染色のため、細胞を、ブロッキング溶液(0.1%トリトン(商標)X−100プラス5%ロバ血清を含む1×DPBS)において2時間、室温においてインキュベーションした。対応する1次抗体を、ブロッキング溶液中の適当な濃度にて希釈し、一晩4℃にてインキュベーションした。使用した1次抗体を、以下の表において表す。細胞を、DPBST(1×DPBS+0.1%トリトン(商標)X−100)を用いて3回洗浄し、適当な2次抗体を、ブロッキング溶液において1時間室温にて加えた。次に、細胞を、DPBSTを用いて3回洗浄し、核対比染色のため、DRAQ5またはHoescht33,342(1μg/mL、1×DPBSにおいて希釈した)と10分間室温にてインキュベーションした。10×、20×もしくは63×の拡大率下で倒立蛍光顕微鏡(Olympus(商標)IX71顕微鏡)または共焦点顕微鏡(Zeiss(商標)LSM5パスカル顕微鏡)を使用して、細胞を可視化した。
PSEN2変異患者または対照患者に由来するヒトiPSCを、単層で増殖させ、細胞培養物中でタンパク質分解酵素およびホスファターゼ阻害剤を含むRIPAバッファー(Thermo Scientific)を用いて直接溶解した。タンパク質濃度を、BCAタンパク質アッセイキット(Thermo Scientific)を使用して測定した。タンパク質の推定後、細胞溶解物20μgを、4〜12%Bis−Trisゲル(Bolt(商標)タンパク質ゲル)上でのSDS−PAGE電気泳動により分離し、電気泳動ブロッティングによりニトロセルロース膜に移した。膜を、ブロッキングバッファー1X TBST(トリス−緩衝食塩水+0.1%Tween)および5%無脂肪乾燥ミルクを用いて、1時間、室温にて攪拌しながらブロッキングし、TBSTを用いて3回洗浄した。洗浄後、膜を、4℃にて一晩攪拌しながら、NLRP2(1:1000)、PSEN2(1:200)または3−アクチン(1:1000)に対する1次抗体とインキュベーションした。洗浄後、膜を、西洋ワサビペルオキシダーゼ(HRP)結合した適当な2次抗体と1時間、室温にてインキュベーションした。最後に、タンパク質バンドを、製造元の指示に従い化学発光試薬を用いて可視化した。3−アクチンを、添加対照として使用した。
全細胞パッチ−クランプ記録を、分化の38〜55日目の単一神経細胞から得た。細胞を、低密度において、灌流ベースの付属記録チャンバーに置いたプラスチックカバーガラスに播種した。神経細胞を、Hamamatsu Orca(商標)R2CCDカメラを備えたOlympus BX61WI顕微鏡下で微分干渉光学を使用して、局在化した。記録を、室温にてMultiClamp(商標)700B増幅器(Molecular Devices、Sunnyvale、CA、USA)を使用して行なった。シグナルを、10kHzにおいて試料採取し、6kHzにおいて、デジタル変換器(Molecular Devices)に類似のDigidata(商標)1440Aを使用してフィルターをかけた。増幅器制御およびデータ取得を、pClamp(商標)10.0ソフトウエア(Molecular Devices)を使用して行なった。
qPCR遺伝子発現実験およびAβ42/40 ELISAを、スチューデントt検定を使用した統計学的有意性について解析した。LDH放出アッセイを、2元配置ANOVAボンフェローニ事後検定により解析した。チューキー事後比較を用いたANOVA検定を、電気生理の結果の解析のため使用した。実験は、数日から数週間を要する電気生理学解析のため記録された94個の神経細胞のそれぞれを検出する必要があった。それぞれの実験日において、それぞれの遺伝子型の代表を、それぞれの日に研究したそれぞれの遺伝子型由来の少なくとも3つの試料と共に含んだ。*、p<0.05;**、p<0.01;***、p<0.001。
BFCN分化のプロトコールの最適化
BFCN分化のスキームを、図1Aに記載する。対照対象またはAD患者に由来するiPSCをフィーダーのない状態でプレーティングし、mTeSR1基礎培地を使用した分化の前に100%コンフルエントに到達させた。「0日目」に、TGF−βシグナル伝達の両方の分岐を阻害(二重SMAD阻害)して神経外胚葉の運命を誘導した[12]。分化(2〜10日目)を、多能性を支持する因子(bFGF、TGF−β、Li−Cl、GABAおよびピペコリン酸)を欠く改変mTeSR1組成を使用して行なった。これらの細胞を前脳基底部コリン作動性神経細胞に特定化するためには、淡蒼球原基(MGE)誘導のための腹側化が必要とされる[19、85、91]。細胞自体を、2〜8日目に、500nMソニックヘッジホッグ(Shh)類似体(SAG)および2μMプルモルファミンを用いて処理した。SAGは、組換えShhより低コストであり、Shh自体より神経細胞の生存特性においていくつかの利点を有し[7、35]、ChAT+運動神経細胞およびグルタミン酸作動性介在神経細胞の分化中に示された通り、Shhシグナル伝達を活性化するための適当な代替物である[91]。本発明者らは、腹側化因子の組み合わせ、投薬量、およびタイミングを、誘導されたNkx2.1前脳基底部前駆体に由来するBFCNの特定化にとってより有益なものに調節するためのツールとして、Nkx2.1−GFP胚性幹細胞(ESC)レポーター株を使用した。
PSEN2N141I変異iPSCおよび対照株を、新鮮皮膚生検から得た。確立した線維芽細胞株を、プレセニリン2ボルガ家族性AD変異(PSEN2N141I)のキャリア2名および影響を受けていないメンバー1名の家系より寄付された皮膚パンチから成長させた。加えて、本発明者らは、非家族性関連対照を含めた。線維芽細胞を、改変RNA方法を用いてリプログラミングして、山中因子(Oct4、KLF4、SOX2、およびc−Myc)を導入し、得られたiPSC株は、アルカリ−ホスファターゼ(AP)酵素活性、遺伝子発現解析、および多能性マーカーについての免疫染色、並びに[60]により開発された自動化iPSCリプログラミングおよびQC方法に従った、染色体異常の検出のための核型分析を含む、頑強な細胞再生および多能性を確かめるための幾つかの品質管理プロセスに供した。研究に含まれる対象の遺伝子型、性別および年齢を、図2aに示す。また、2種のPSEN2N141I iPSC株はAPOE ε4についてヘテロ接合性(ε3/ε4)であり、一方、対照iPSC株はホモ接合性ε3/ε3であった。iPSC株の特徴決定、多能性マーカーの発現、および品質管理の結果を、図10に示す。簡単に言うと、選択した全てのiPSCクローンは、胚様体形成および3つの胚葉への分化により多能性を示し(図10A)、これは、参照により本明細書に組み込まれる。最後に、株を、フィンガープリンティングして(Cell Line genetics)、それらが、親線維芽細胞株と一致することを確実にした(データを示していない)。全ての親線維芽細胞株およびiPSC株を、サンガー配列決定に供して、PSEN2およびAPOEの遺伝子型を決定した。PSEN2N141I点変異が位置する(Chr1:227,073,304 A>T)領域を囲む、PSEN2のエクソン5由来の173bpのフラグメントを、PCRにより増幅し、[53]において公開されたプライマーを使用して配列決定し;同様に、3つの対立形質バリアントを決定する2つのSNPを含有するAPOE遺伝子座由来の244bpのフラグメントを、ゲノムDNAからPCRにより増幅し、続いて配列決定して、[36]のプライマーを使用して、ε2/ε3/ε4バリアント間を区別した。PSEN2N141I点変異の存在を示す試料のクロマトグラムを図2Bにおいて示し、全ての遺伝子型を図2Aにおいて概説した。
コリン作動性神経細胞の分化の早期段階におけるPSEN2N141I変異の効果を研究するために、本発明者らは、BFCN分化プロトコールに沿ってDIV11〜16において得られる神経前駆細胞(NPC)を解析した。この中間未成熟集団の解析により、本発明者らが、それを実施しない場合、最後まで分化したコリン作動性神経細胞においては検出されない、BFCNの生成において可能性のある早期変化を検出することが可能となる。かかる異常は、成熟神経細胞において潜在的に役割を果たし、ADの病態生理学に関与し得る。本発明者らは、遺伝子発現および免疫蛍光法により、PSEN2N141I変異NPCおよび対照NPCにおける早期神経細胞マーカーの発現を解析した。本発明者らは、分化の11日目の変異NPCにおいて、一般的な神経細胞マーカーであるTuj1(βIII−チューブリン)のRNA発現がより低いことを見出したが、約16〜21日目には、本発明者らは、免疫細胞化学により定量可能な差を検出しなかった(図2CおよびD)。また、11日目のNPC単層培養物を典型的なNPCマーカー:Sox2、およびPax6について免疫染色したところ;Nkx2.1導入と共に予測された通り、Pax6レベルが低下していた(示していない)。本発明者らは、11日目のPSEN2N141I培養物においてSox2およびネスチンの同等の発現を観察した(図2C、上部のパネル)。21日目では、変異NPCは、同等レベルのNkx2.1(MGEマーカー)を発現していたが、qPCRによるとBF1(前脳マーカー)のレベルが低減しており;しかしながら、この分化段階における免疫染色によるとBF1タンパク質の発現は影響を受けていないように見えた(図2Cの下部パネル、およびD)。本発明者らは、陽性細胞の割合(%)または蛍光中央ピーク値に関して、DIV11〜12のPSEN2N141I細胞のNGFR(p75/CD271)表面発現における差を観察しなかった(図2E)。
BFCNの分化、遺伝子発現、機能、およびコミュニケーションに対するPSEN2N141I変異の影響を決定することを目的に、本発明者らは、適当な発現マーカーについて、より後の時間点において細胞を特徴付け;本発明者らの目標は、PSEN2N141IiPSCが、BFCN成熟プロセスを完了することができたかどうか、およびそうであれば、BFCN分化のより後の段階に沿った任意の異常がEOFADの病態生理学を説明するか、を調査することであった(図3)。プロNGFに優先的に結合するp75に加えて、本発明者らは、PSEN2N141IBFCNおよび対照において発現した初代成熟NGF受容体であるTrkAの発現を解析した(図3a)。これは、PSEN2N141IBFCNが、予想通り、NGFの生存促進シグナルおよび分化シグナルを受けて当該シグナルから恩恵を受けることができることを示唆しており、それらの固有のアイデンティティをさらに裏付ける。本発明者らは、PSEN2N141IBFCNおよび対照において、さらなるコリン作動性神経細胞特異的マーカーであるコリンアセチルトランスフェラーゼ(ChAT)および小胞アセチルコリン輸送体(vAChT)の同等の発現を観察した(図3b)。Tuj1、成熟マーカー微小管関連タンパク質2(MAP2)のような他の一般的な神経細胞マーカーは、免疫蛍光により明らかな差を示した(図3b)。
PSEN1変異体において既に観察された通り[77]、APPのプロセシングおよび切断における分子変化ならびに/またはNLRP2インフラマソームの活性化の悪化がPSEN2N141I変異のみに関与するかを決定するために、本発明者らは、CRISPR/Cas9テクノロジーを利用して、本発明者らのiPSC株におけるPSEN2遺伝子座を修飾した。本発明者らは、2つのPSEN2変異iPSC株(AD1、AD2)におけるPSEN2N141I点変異を修正することにより、これを行なった。この目的のため、オンラインのツール(tools.genome−engineering.org)を使用して、PSEN2N141I変異を囲むPSEN2エクソン5の領域(23bp上流のChr1:227,073,304 A>T)にCas9を誘導するための特異的なガイドRNA(g1N141I)を設計した。g1N141Iを、pSpCas9(BB)−2A−GFP(PX458)ベクターにクローニングした。HEK293Tにおいて、pSpCas9−g1N141I−GFPを形質導入し、GFPの蛍光により発現を評価した(図4a)。
従前の報告は、FAD変異を有するiPSC株が、高濃度のAβ42オリゴマーのような有害な刺激に対して増強した感受性を示し得ることを示した[2]。それ故、本発明者らは、PSEN2N141I変異体由来の本発明者らのBFCNが、培地中のAβ42オリゴマーに対する増強した毒性を示すかどうかを試験した(図5)。本発明者らは、死細胞により放出された乳酸脱水素酵素(LDH)の割合(%)を測定する(これは、毒性についての間接的な測定を提供する)ことにより、神経毒性を評価した。2元配置ANOVAによるこの方法を用いて、本発明者らは、72時間の暴露後に、培地に加えた5μM Aβ42オリゴマーによりもたらされる毒性における有意な効果を検出した(***、p<0.01)。事後ボンフェローニ解析により、AD2株とその修正されたアイソジェニック対照(iAD2対照)との間の有意差を明らかにした。しかしながら、Aβ42オリゴマー毒性に対するこの明らかに増強した感受性が、AD1株およびその対応する対照においては観察されなかった。これらの結果は、Aβ42に対する感受性における差が、変異PSEN2遺伝子型のみには結び付けられないこと、および恐らく、AD1およびAD2対象間のさらなる遺伝的因子が、このストレスに対する感受性に影響し、これが、複数のアイソジェニックモデルの重要性をさらに裏付けることを示している。
本発明者らは、PSEN1変異皮質神経細胞の場合(公開していない知見)と同様に、PSEN1変異iPSCおよびNPCにおいてNLRP2 mRNAが上昇したことを既に報告した[77]。それ故、本発明者らは、PSEN2N141I変異との関連でインフラマソームの成分の状態を解析することを望んだ。本発明者らが、DIV12にてNPCにおけるNLRP2のmRNAレベルをqPCRによりアッセイしたところ、本発明者らは、対照株と比較してAD1およびAD2株において100倍を超える増大を観察した(図6a)。これは、11日目のPSEN2変異体由来の全細胞溶解物においてSDS−PAGEにより観察した、NLRP2タンパク質の著しい増大と相関した(図6d)。しかしながら、著しくは、本発明者らは、AD2株溶解物における免疫ブロットによるNLRP2についてのバンドを検出しなかった。さらに、本発明者らは、ミトコンドリアクレアチニンキナーゼの分解を誘導するE3リガーゼをコードするASB9の上昇のような、PSEN1変異iPS神経前駆細胞において既に見られた幾つかの他の転写現象を実証することができなかった。代わりに、本発明者らは、PSEN2変異キャリアにおいてレベルが20〜30%減少する傾向を観察した。
本発明者らは、BFCN分化プロトコールを用いて、分化の35日目から、PSEN2N141I変異AD患者2名、野生型対照、および家族性対照に由来するディッシュにおいて電気生理的に活性なコリン作動性神経細胞を作製することができた。本発明者らは、まず、神経基礎培地中で成長させたBFCNからは、この段階では成熟した活動電位波形を得ることができなかったが、Brainphys(商標)培地へのスイッチにより、培養した神経細胞の電気生理学的特性が有意に改善した[3]。これらの知見は、両方の培地を比較するために使用された他のiPSCから生成された神経細胞の電気生理学的特徴決定と一致する[3]。ChATおよびVAChTのエンドポイント発現並びに電気生理学的応答が同等の2種のさらなる細胞株(H9胚性幹細胞株を含む)におけるBrainphys(商標)培地を含有するプロトコールの恩恵が再現された。BFCNの電気生理学的特性を調べるために、本発明者らは、全細胞パッチクランプ法を用いて、合計94個の神経細胞(22個の野生型対照、21個の家族性対照、18個のAD1、28個のAD2、および5個のiAD1_対照)から記録した。全ての実験群において、記録した神経細胞は、ナトリウムおよびカリウムイオンチャネルを介した電位活性化型電流、活動電位を生じる能力を示し、古典的な神経細胞形態を示した(図7)。実験のサブセットにおいて、記録した神経細胞を、パッチピペットを通じてビオサイチンを用いて標識し、それにより、事後細胞同定およびICH特徴決定を可能にした。本発明者らは、全てのビオチン標識した細胞はChATおよびVAChTについて免疫陽性でもあったことを見出した(n=12、図8a)。
米国において、500万人の人々が、現在、アルツハイマー病の影響を受けており、アルツハイマー病の団体によると、この数は2050年代までに16000000人まで増大する。不運なことに、本発明者らは、これらの患者の3〜5%を占める遺伝性の原因の直接的な証拠のみを有する。このパーセンテージは、γ−セクレターゼ装置を構成するアミロイドタンパク質前駆体(APP)、またはPSEN1、PSEN2における遺伝性完全浸透性常染色体優性遺伝性変異により引き起こされるEOFADバリアントを包含し[87]、それらの機能における変化は、Aβ42オリゴマーの産生および/またはアミロイド斑の沈着を増大させる。
本発明者らは、プレセニリン2(PSEN2)変異キャリアおよび対照に由来するiPSCからヒトBFCNを生成するようにインビトロプロトコールを最適化した。予想通り、PSEN2N141Iは、iPSCに由来するBFCNにおいてAβ42/40の増大と関連し、これを、CRISPR/Cas9により仲介される遺伝子編集により元に戻した。予想外に、PSEN2N141Iキャリア由来のiPSCに由来するBFCNまたは皮質神経細胞は、活動電位頻度と活動電位振幅の両方の低減により定量される、基礎となる興奮性の低下を示した。また、この電気生理学的表現型はPSEN2N141I変異のCRISPR/Cas9による修正後に消失した。遺伝子編集データにより、全ての予想される知見および全ての細胞由来の遺伝子型を特徴付ける変異関連変化の強い一貫性が存在することが立証される。

- 前脳基底部コリン作動性神経細胞(BFCN)を生成する方法であって、
神経外胚葉性分化を誘導するためにトランスフォーミング増殖因子ベータ(TGF−β)シグナル伝達の阻害剤およびソニックヘッジホッグ(Shh)シグナル伝達の活性化因子を含む基礎培地中で多能性幹細胞(PSC)を培養する工程であって、前記基礎培地が、塩基性線維芽細胞増殖因子(bFGF)、TGF−β、塩化リチウム(Li−Cl)、GABA、およびピペコリン酸を欠く、工程
を含み、それによりBFCNが生成される、方法。 - 前記阻害剤が、SMAD阻害剤である、請求項1に記載の方法。
- 前記阻害剤が、SB431542、LDN193189、またはそれらの組み合わせである、請求項2に記載の方法。
