Claims (25)
- 混合プロトコルを実施する制御回路の制御下で、試薬を複数の異なる試薬容器からキャッシュチャネルに吸引するステップと、
前記試薬を前記キャッシュチャネルから混合容器に排出するステップと、
前記試薬を前記混合容器内で混合して試薬混合物を構成するステップと、
を含む方法であり、
前記制御回路が実施する前記混合プロトコルに基づいて、前記試薬の指定量が、対応するシッパーによって、対応する前記試薬容器から自動的に吸引される、方法。 - 前記試薬混合物をフローセルに送達するステップを更に含み、
前記試薬混合物は前記フローセル上で試料テンプレートと反応して、前記フローセル上でデオキシリボ核酸(DNA)分子のクローン集団を生成する、請求項1に記載の方法。 - 前記試薬を前記キャッシュチャネルから前記混合容器に排出する前に、前記混合容器は前記混合容器の中に試料テンプレートを包含する、請求項1に記載の方法。
- ある量の前記試薬混合物を、前記混合容器内へ延在するノズルシッパーに吸引し、その後前記ある量の試薬混合物を前記ノズルシッパーから前記混合容器内に排出して戻すことによって、前記混合容器の前記試薬を混合する、請求項1に記載の方法。
- 前記ノズルシッパーはバッファ流体を前記ノズルシッパーの中に包含し、
前記方法は、前記ある量の試薬混合物を前記ノズルシッパーに吸引する前に、空気を前記ノズルシッパー内に導入して、前記バッファ流体と前記ノズルシッパーに吸引される前記試薬混合物との間にエアギャップを定めて、前記バッファ流体と前記試薬混合物との間の混合を妨げるステップを更に含む、請求項4に記載の方法。 - 前記試薬を、順序付けられるシーケンスで1度に1つ、前記キャッシュチャネル内へ吸引する、請求項1に記載の方法。
- 前記試薬を前記キャッシュチャネルに吸引するステップに応じて、前記キャッシュチャネルは、指定量の異なる前記試薬からなり、前記キャッシュチャネルの丈に沿う交互のパターンを含む、請求項6に記載の方法。
- 前記キャッシュチャネル内の前記試薬が前記混合容器内に排出される前に、前記試薬を、少なくとも1度繰り返され順序付けられるシーケンスで1度に1つ、前記異なる試薬容器から前記キャッシュチャネル内に吸引する、請求項1に記載の方法。
- 第1体積の前記試薬を前記キャッシュチャネル内に吸引し、より小さな第2体積の前記試薬を前記混合容器に排出して、前記試薬を前記混合容器に排出した後に、上流バッファゾーンを定める前記試薬の残余体積が前記キャッシュチャネルに残る、請求項1に記載の方法。
- 前記対応する試薬容器内へ延在するシッパーを用いて、前記試薬を前記対応する前記試薬容器から吸引し、
前記シッパーは、流体マニホールド上の対応するポート及び流体チャネルを通じて、前記キャッシュチャネルに流体的に接続される、請求項1に記載の方法。 - 界面活性剤を前記試薬に導入して前記試薬間の混和性の差を低減するステップを更に含む、請求項1に記載の方法。
- 前記試薬の少なくとも一部が互いに対して異なる比重を有する、請求項1に記載の方法。
- 10,000ダルトン未満の分子量を有する密集剤を前記試薬に導入して、前記試薬の粘度を低減するステップを更に含む、請求項1に記載の方法。
- ノズルシッパー及び複数試薬シッパーを含む複数シッパーと、
ポンプ端部及び容器端部の間に延在するキャッシュチャネルと、
前記ポンプ及び試薬セレクタバルブに動作可能に接続され、前記ポンプ及び前記試薬セレクタバルブを制御して、対応試薬シッパーを通じて混合プロトコルに基づく指定量の前記試薬を前記キャッシュチャネルに自動的に吸引することで前記混合プロトコルを実施し、続いて前記ポンプ及び前記試薬セレクタバルブを制御して前記試薬を前記キャッシュチャネルから前記ノズルシッパーを通じて混合容器内に排出するとともに、前記混合容器内で前記試薬を混合して試薬混合物を構成する制御回路と、
を備えるシステムであり、
前記複数試薬シッパーは、異なる試薬を前記複数試薬シッパー内に包含する、異なる対応試薬容器内に延在して、前記複数試薬シッパーのそれぞれの先端チップが前記試薬容器の前記試薬に接触し、
前記ノズルシッパーは前記混合容器内へ延在し、
前記キャッシュチャネルの前記ポンプ端部はポンプに動作可能に接続され、前記キャッシュチャネルの前記容器端部は前記試薬セレクタバルブ及び対応流体チャネルを通じて前記シッパーに流体的に接続される、システム。 - 前記制御回路は前記ポンプを制御して、ある量の前記試薬混合物を前記ノズルシッパー内に吸引し、その後ある体積の前記試薬混合物を前記ノズルシッパーから前記混合容器に排出して戻すことによって、前記混合容器で前記試薬を混合する、請求項14に記載のシステム。
- 前記制御回路は前記ポンプ及び前記試薬セレクタバルブを制御して、前記試薬混合物を前記混合容器内に吸引しその後排出することを複数回行って前記試薬を混合する、請求項15に記載のシステム。
- 前記ノズルシッパーは前記ノズルシッパー内にバッファ流体を包含し、
前記試薬混合物を前記ノズルシッパー内に吸引する前に、前記制御回路は前記ポンプを制御して空気を前記ノズルシッパー内に導入して、前記バッファ流体と前記ノズルシッパー内に吸引される前記試薬混合物との間にエアギャップを定めて前記バッファ流体と前記試薬混合物との間の混合を妨げる、請求項15に記載のシステム。 - 前記ノズルシッパーの内径が、前記試薬シッパーのそれぞれの内径よりも小さい、請求項14に記載のシステム。
- 前記制御回路は前記ポンプ及び前記試薬セレクタバルブを更に制御して、前記試薬混合物を、前記ノズルシッパー及び前記試薬セレクタバルブを通じて前記混合容器に流体的に接続されるフローセルに送達し、
前記試薬混合物は前記フローセル上の試料テンプレートと反応してDNA分子のクローン集団を前記フローセル上に生成する、請求項14に記載のシステム。 - 前記制御回路は前記ポンプ及び前記試薬セレクタバルブを制御して、前記異なる試薬を順序付けられるシーケンスで1度に1つ前記キャッシュチャネル内に吸引するとともに、前記キャッシュチャネルの前記試薬が前記混合容器内に排出される前に、前記順序付けられるシーケンスを少なくとも1度繰り返す、請求項14に記載のシステム。
- 前記制御回路は前記ポンプ及び前記試薬セレクタバルブを制御して、第1体積の前記試薬を前記キャッシュチャネル内に吸引し、その後より小さい第2体積の前記試薬を前記キャッシュチャネルから前記混合容器内に排出して、前記試薬を前記混合容器内に排出した後に、上流バッファゾーンを定める前記試薬の残余体積が前記キャッシュチャネルに残る、請求項14に記載のシステム。
- 複数試薬シッパー及びノズルシッパーを含む複数シッパー、並びにキャッシュチャネルを含む流体マニホールドと、
前記キャッシュチャネル及び前記シッパーの間に動作可能に接続される試薬セレクタバルブと、
前記キャッシュチャネルのポンプ端部に動作可能に接続されるポンプと、
を備えるシステムであり、
前記複数試薬シッパーは異なる試薬を中に包含する異なる対応試薬容器内へ延在して、前記複数試薬シッパーの先端チップは前記試薬に接触し、
前記ノズルシッパーは混合容器内へ延在し、
前記キャッシュチャネルは前記ポンプ端部及び容器端部の間に延在し、
前記容器端部は流体マニホールドに沿う対応流体チャネルを通じて前記シッパーに流体的に接続され、
前記ポンプ及び前記試薬セレクタバルブは混合プロトコルに従って自動的に制御されて、前記混合プロトコルに基づく指定量の前記試薬を、試薬容器から対応試薬シッパーを通じて前記キャッシュチャネル内へ吸引し、
前記ポンプ及び前記試薬セレクタバルブは自動的に続いて制御されて、前記試薬を前記キャッシュチャネルから前記ノズルシッパーを通じて前記混合容器内へ排出し、ある体積の試薬混合物を前記混合容器から前記ノズルシッパー内へ吸引するとともにその後前記ある体積の前記試薬混合物を前記ノズルシッパーから前記混合容器内へ排出して戻すことによって、前記混合容器で前記試薬を混合して前記試薬混合物を構成する、システム。 - 前記ポンプ及び前記試薬セレクタバルブは自動的に制御されて、前記試薬を、順序付けられるシーケンスで1度に1つ、前記試薬容器から前記キャッシュチャネルに吸引し、前記試薬を前記キャッシュチャネルから前記混合容器内に排出する前に、前記順序付けられるシーケンスを少なくとも1度繰り返す、請求項22に記載のシステム。
- 前記流体マニホールド、前記試薬セレクタバルブ及び前記ポンプは器械のハウジング内に共通して配置される、請求項22に記載のシステム。
- ハウジングと、
前記ハウジング内に配置されるとともに、異なる対応容器内へ延在するシッパーに流体的に接続される複数チャネルを含む流体マニホールドと、
前記ハウジング内に配置されるとともに、前記流体マニホールドの前記複数チャネルの少なくとも1つに動作可能に接続されるポンプと、
前記ハウジング内に配置されるとともに、前記流体マニホールドの前記複数チャネルの少なくとも2つに動作可能に接続される試薬セレクタバルブと、
前記ハウジング内に配置されるとともに、前記流体マニホールドの前記複数チャネルの少なくとも1つに流体的に接続されるフローセルと、
を備える器械であり、
前記ポンプ及び前記試薬セレクタバルブが混合プロトコルに従って自動的に制御されて、容器の少なくとも一部の中に包含される試薬を、前記混合プロトコルに基づく指定量だけ、前記対応容器から前記シッパーを通じて前記流体マニホールドの前記複数チャネルに輸送し、
前記ポンプ及び前記試薬セレクタバルブは自動的に制御されて、前記流体マニホールドに輸送される前記試薬を混合して試薬混合物を構成し、その後前記試薬混合物を前記マニホールドから前記フローセルに送達する、器械。
