Description
幹細胞はナイーブ状態幹細胞でありうる。あるいは、ステップ(i)で、幹細胞はナイーブ状態若しくはプライムド状態の幹細胞でありうる。そこではナイーブ状態幹細胞に対する作用薬の効果は、プライムド状態の幹細胞に対する影響と比較され、プライムド状態の幹細胞と比較して、作用薬がナイーブ状態幹細胞により大きな効果があれば、そこでは、該作用薬が抗ガン剤でありうると決定できる。作用薬は、多クローン性抗体、単クローン抗体、分子様抗体、抗体フラグメント融合蛋白質、抗体擬態、ペプチド、ペプチド擬態、小分子あるいは天然産物でありうる。幹細胞は、ヒト由来でありうる。幹細胞は、NME7ABあるいはNME7-X1を含む培地中で培養することにより、ナイーブ状態で維持されうる。癌は、卵巣癌、乳癌、黒色腫、前立腺癌、結腸癌、肺癌、あるいは膵臓癌でありうる。癌は、MUC1陽性あるいはMUCl*陽性癌でありうる。癌は、NME7ABあるいはNME7-X1陽性癌でありうる。作用薬は、一般に細胞毒性がない。作用薬は、繊維芽細胞あるいは繊維芽細胞始原細胞に細胞毒性がない。

そして、他の治療と組み合わせることも可能であるし、組み合わせないこともありえ、それはさらに量、タイミングあるいは期間において異なりうる。癌の別の治療法は外科である。それは単独であるいは前述の治療方法のうちのいずれかと組み合わせで利用することができる。医術技術のうちの通常技術の1つは適切な治療を決定可能である。

私たちは、幹細胞多分化能あるいは増殖を阻害するか、幹細胞分化を引き起こす作用薬が、癌細胞増殖を阻害し、患者に投与された時、癌の予防若しくは治療のための有能な作用薬である作用薬であることを見出した。ナイーブ幹細胞多分化能あるいは増殖を阻害するか、ナイーブ幹細胞分化を引き起こす作用薬は、癌細胞移動を阻害する作用薬である。それは転移癌の特徴で、患者に投与された時攻撃的若しくは転移癌の予防若しくは治療用の効果的な抗癌物質である。多分化能若しくは増殖を阻害するか、プライムド幹細胞ではなくナイーブ幹細胞の幹細胞分化を引き起こすか、プライムド幹細胞に遠いより劣った効果がある作用薬は、攻撃的若しくは転移癌の予防若しくは治療のための有能な作用薬である。
1)多能性の状態で幹細胞を育てる;
2)候補薬と多能性幹細胞の個体群を接触させる;
3) 多能性幹細胞の多分化能若しくは増殖を阻害するか、多能性幹細胞の分化を引き起こす候補薬を同定する;そして、
4) 多能性幹細胞の多分化能若しくは増殖を阻害するか、多能性幹細胞の分化を引き起こす候補薬が、抗ガン剤であると結論を下す。
1)ナイーブ状態の幹細胞を育てる;

また、本発明の別の様相では、新規癌治療あるいは抗転移薬の標的は、プライムド幹細胞でではなくナイーブ幹細胞でアップレギュレートされるmicroRNAsの同定により特定される。

- 化合物が、本願明細書の段落0200に記述と同様に描写されるいずれかである、請求項52に記載の方法。
- 癌が、MUC1陽性若しくはMUC1*陽性癌である、請求項52の方法。
- 癌が、NME7AB陽性癌若しくはNME7-X1陽性癌である、請求項52の方法。
- 次のステップを含む、癌若しくは癌転移を予防若しくは処置する方法;
(1) 患者からの癌サンプルを分析し、MUC1*陽性、NME7AB陽性若しくはNME7-X1陽性であることを決定する;そして
(2) 式1〜17の化合物の有効量を患者に投与する。 - 分析ステップが、PCRによって行われる、請求項56に記載の方法。
- 癌サンプルが、EEF1A1遺伝子のmRNA発現レベルの少なくとも0.5%であるMUC1遺伝子、NME7遺伝子、NME7-X1遺伝子のmRNA発現レベルであるとき、それは、MUC1*陽性、NME7AB陽性若しくはNME7-X1陽性であると決定される。
- 分析ステップが、免疫組織化学によって行われる、請求項56に記載の方法。
- 癌サンプルが、PSMGFRペプチド若しくはN-10ペプチドに結合する抗体と接触され、病理学的標準スコア1-4(+〜++++)で組織が染色され、MUC1*陽性であると決定される、請求項56に記載の方法。
- 癌サンプルが、NME7のB3ペプチドに結合する抗体と接触され、病理学的標準スコア1-4(+〜++++)で組織が染色され、NME7AB陽性若しくはNME7-X1陽性であると決定される、請求項56に記載の方法。
- 次のステップを含む、炎症性疾患及びその状態を予防若しくは治療する薬剤を同定する方法:
(1) 作用薬に、幹細胞を暴露し、そして
(2) 幹細胞多分化能あるいは増殖を阻害するか、幹細胞分化を引き起こす作用薬を同定する、そこでは、作用薬またはそのアナログは、炎症性疾患またはその状態を治療するための作用薬である。 - 炎症性疾患若しくはその状態が、慢性関節リウマチ、炎症性の腸症候群、クローン病、変形性関節症、喘息、皮膚炎、乾癬、嚢胞性繊維症、遅延性移植および慢性の実質器官拒絶、多発性硬化症、全身性紅斑性狼瘡、シェーグレン症候群、橋本甲状腺炎、多発性筋炎、強皮症、アジソン病、白斑、悪性貧血、糸球体腎炎、肺線維症、自己免疫性の糖尿病、糖尿病性網膜症、鼻炎、乏血再潅流傷害、ポスト血管形成術再狭窄、慢性閉塞性肺疾患(COPD)、グレーブス病、消化管アレルギー、結膜炎、アテローム性動脈硬化、冠動脈疾患、狭心症、癌転移、小動脈疾病あるいはミトコンドリアの疾病、である請求項62に記載の方法。
- 幹細胞と接触された時、幹多分化能あるいは増殖を阻害する、あるいは幹細胞分化を引き起こす、作用薬を、その必要のある人に投与することを含む炎症性疾患若しくはその状態を治療する方法。
- 炎症性疾患若しくはその状態が、慢性関節リウマチ、炎症性の腸症候群、クローン病、変形性関節症、喘息、皮膚炎、乾癬、嚢胞性繊維症、遅延性移植および慢性の実質器官拒絶、多発性硬化症、全身性紅斑性狼瘡、シェーグレン症候群、橋本甲状腺炎、多発性筋炎、強皮症、アジソン病、白斑、悪性貧血、糸球体腎炎、肺線維症、自己免疫性の糖尿病、糖尿病性網膜症、鼻炎、乏血再潅流傷害、ポスト血管形成術再狭窄、慢性閉塞性肺疾患(COPD)、グレーブス病、消化管アレルギー、結膜炎、アテローム性動脈硬化、冠動脈疾患、狭心症、癌転移、小動脈疾病あるいはミトコンドリアの疾病、である請求項64に記載の方法。
- 作用薬が、式1〜17の化合物、又は図18A〜18Eに示された化合物、又は本願明細書の段落0200に記述の化合物である、請求項64に記載の方法。
- 癌進展の危険がある患者若しくは癌と診断された患者を治療する方法であって以下の工程を含む:
1)ある化合物の存在下で、ナイーブ幹細胞及び繊維芽細胞を育てる;
2)ナイーブ幹細胞の多能性若しくは増殖を阻害する化合物を特定する;
3)当該化合物が、繊維芽細胞に殆ど若しくは全く効果をもたないことを確認する;
4)該化合物は、癌細胞の増殖若しくは侵襲を阻害すると結論を下し;そして、
5)癌転移の処置若しくは予防のために、該化合物の有効量を患者に投与する。
