ある実施形態では、本開示は、様々な細胞型、及び、様々な増殖因子、細胞分化因子、及び、細胞増殖の研究に有用なその他の薬剤、例えば、腫瘍またはがんの成長の阻害剤において増強し得るバイオロジカルスキャフォールドを提供する。様々な実施形態では、当該バイオロジカルスキャフォールドは、少なくとも2つの成分を含む。第1の成分は、哺乳動物血小板溶解物である。用語「哺乳動物血小板溶解物は、ヒトを含むあらゆる哺乳動物供給源から単離した血小板から形成した溶解物を含むことができる。
細胞分化のためのキット
幾つかの実施形態では、本明細書に記載のバイオロジカルスキャフォールドを、医師向けの出発成分として使用して、特定の分化した細胞及び組織を調製し得る。幾つかの例示的な実施形態では、これらのキットは、研究目的に使用し得る。例えば、例示的なキットは、哺乳動物血小板溶解物、マルチウェル基材、及び、試薬A(Dulbecco’s Modified Essential Mediumからなり、1%の抗生物質/抗真菌剤、そして、細胞培養、及び、白色または褐色の脂肪細胞分化のために必要な栄養素、アミノ酸、及び、増殖因子を含有するその他の関連する細胞培養培地を補充したもの)が入った容器を含み得る。特定の実施形態では、使用者が、三次元(3D)細胞培養物で分化細胞の調製を希望する場合には、使用する当該3D組織培養物は、以下の例示的な工程に従って調製し得る。1%抗生物質/抗真菌剤、及び、74%Dulbecco’s Modified Essential Medium、または、細胞培養に必要な栄養素、アミノ酸、及び、増殖因子を含有する、その他の関連する細胞培養培地に対して、25%(v/v)の当該哺乳動物血小板溶解物を加えることで、哺乳動物血小板溶解物補充培地を調製する。混合後は、水浴で温めない。室温に維持する。氷が消える瞬間まで、これらの細胞(SVF細胞、及び/または、ASC、及び/または、BM−MSC)を、37℃の水浴で、液体窒素から解凍する。各クライオバイアルを、1mLの標準的なStromal Mediumで1回洗浄する。チューブを、300×g(1200rpm)で、5分間、遠心分離する。この細胞ペレットを、5×105細胞/mLの濃度で、hPL補充培地に再懸濁した。直ちに、細胞/hPL混合液を、プレーティングする。CO2インキュベーターに入れると、混合物は、直ちにゲル化する。蒸発した培地を7日ごとに交換して、3D培養を維持する。培養物の古い培地を、1mLのマイクロピペットで吸引して除去する。当該ゲルの上隅に、半量のhPL−培地を加えて、新しい培地と交換する。
幾つかの実施形態では、哺乳動物血小板溶解物は、ヒト脂肪組織由来間質血管画分(SVF)細胞、脂肪由来間質/幹細胞(ASC)、及び/または、骨髄由来間葉系間質/幹細胞(BM−MSC)と組み合わせて、脂肪組織、または、骨と骨髄の生物学の研究に適した組み合わせ製品を作製し得る。当該組み合わせ製品は、脂質生成(AdipoQual)、軟骨形成(ChondroQual)、または、骨形成(OsteoQual)を促すことが公知のカクテル剤の存在下、または、非存在下で、インビトロで培養することができる。AdipoQualの存在下で、哺乳動物血小板溶解物を、SVF細胞、ASC、または、BM−MSCと組み合わせると、遺伝子、タンパク質、及び、小分子発現、ならびに、グルコース取り込み、脂肪分解、代謝活性、及び、その他の脂肪組織機能アッセイに基づいて評価できる三次元脂肪組織構造、及び、脂肪組織機能を形成する。ChondroQualの存在下で、哺乳動物血小板溶解物を、SVF細胞、ASC、または、BM−MSCと組み合わせると、遺伝子、タンパク質、及び、小分子発現、ならびに、グリコサミノグリカン産生、代謝活性、及び、その他の軟骨組織機能アッセイに基づいて評価できる三次元軟骨組織構造、及び、機能を形成する。OsteoQualの存在下で、哺乳動物血小板溶解物を、SVF細胞、ASC、または、BM−MSCと組み合わせると、遺伝子、タンパク質、及び、小分子発現、ならびに、細胞外マトリックス脱塩、カルシウム取り込み、代謝活性、及び、リンパ造血などのその他の骨組織機能アッセイに基づいて評価できる三次元の骨及び骨髄組織構造、及び、機能を形成する。
骨髄間葉系幹細胞(BM−MSC)の単離
BM−MSCを単離するために、まず、凝固を防ぐために、ヘパリンまたはクエン酸塩で抗凝固処理をした骨髄穿刺物を得る。使用前に、骨髄穿刺物を4℃の湿った氷上で維持し、そして、BSL2生物学的安全キャビネット内の開封容器で、すべての工程を実行する。FicollPaque溶液(GE Healthcare(商標)Ficoll−Paque(商標)PREMIUM、1.078g/mL)を完全に混合する。20mlのFicoll Paque溶液を、滅菌50mlコニカルチューブに分注する。10mlの骨髄穿刺物を、Ficoll Paque層の上に慎重に重層し、そして、混和をせずにおく。チューブを密閉した後、卓上型Sorvall(商標)Legend(商標)遠心分離機を使用して、ブレーキをかけずに、室温で、500×gで、30分間、遠心分離する。コニカルチューブを、BSL2生物学的安全キャビネットに戻した後に、上部の透明な血漿層を吸引除去し、そして、新たな滅菌50mlコニカルチューブに分注する。次に、Ficoll Paque層の上部から単核球のバフィーコート層を吸引する。単核球を、滅菌15mlコニカルチューブに移し、そして、3倍量の滅菌Phosphate Buffered Saline(PBS)溶液に懸濁する。このチューブを、室温で、200×gで、10分間、遠心分離する。汚染物質とFicoll−Paqueを洗い流した後に、ペレット状の単核細胞から上清を吸引する。ペレット状の単核細胞を、1〜10mlのLaCell Stromal Medium(商標)に再懸濁する。10μlを除去し、そして、アクリジンオレンジ/エチジウムブロマイドを含有する等量のLaCellのLive Dead Assay Solution(商標)と混合し、そして、蛍光顕微鏡(Motic AE30 Epifluorescent Microscope)で、生存(緑色)している、及び、死滅(赤色)した有核細胞を計数する。生細胞のパーセンテージと、細胞の総数/ミリリットルに基づいて、次の式を使用して、有核細胞の総数を計算する。
別の実験例では、SVFとシルクバイオスキャフォールドとを含む脂肪組織装置を記載している。白色脂肪組織(WAT)の間質血管画分(SVF)由来の前駆細胞は、クローン集団を形成し、そして、複数の系統経路に沿って分化する能力を有する。しかしながら、文献は、脂肪細胞前駆体を「間質」または「幹」細胞として定義することに躊躇している。最近の研究は、表現型CD45−/CD31−/CD146−/CD34+を有する脂肪間質細胞の非周皮細胞亜集団が、間葉性であることを実証しており、そして、このものが、脂肪組織内の内因性前駆体亜集団であることを示唆する。Frazier et al.は、GFP導入遺伝子(GFP−Tg)を偏在的に発現する間質血管画分(SVF)細胞、及び、培養増殖脂肪間質/幹細胞(ASC)を、フローサイトメトリーによって分画できることを実証した。新たに単離したSVFと、培養増殖したASCの双方を、3Dシルクスキャフォールドに播種し、野生型宿主に皮下移植し、そして、1度の初期移植と、2度の連続移植(2°移植)とを首尾良く行った。6週間のWAT構築物を取り出し、そして、GFP−Tg脂肪細胞及び幹細胞の存在について評価した。フローサイトメトリー、定量的ポリメラーゼ連鎖反応、及び、共焦点顕微鏡検査は、それぞれ、GFP−Tg細胞の持続性、増殖、及び、増殖を示した。グリセロール分泌、及び、グルコース取り込みアッセイは、GFP−Tg脂肪が、代謝的に機能的であることを明らかにした。GFP−Tg SVF細胞、または、GFP−Tg ASCを播種した構築物は、播種していないコントロールと比較して、消化組織からのSVF収量が高く、また、構築物重量も大きかった。CD146−CD34+を豊富に含有するGFP−Tg ASC集団由来の構築物は、未選別、または、CD29+ GFP−Tg ASC対応物よりも、ヘモグロビン飽和度が大きく、また、高頻度でGFP−Tg細胞を示した。これらのデータは、脂肪組織を固形臓器として再構成可能な非周皮性脂肪由来前駆細胞の連続移植の成功を実証した。
材料及び方法
マウス
動物研究は、連邦、州、及び、National Institute of Healthの政策及び規制に従って、the Institutional Animal Care and Use Committeeによる審査を受け、かつ、承認がされたプロトコルの下で、the Department of Comparative Medicine of the Tulane University School of Medicineの監督下で行った。SVF細胞、及び、ASCは、公開された方法に従って、8〜12週齢のオスのC57BL/6−Tg(UBC−GFP)30cha/Jマウス(ヒトユビキチンCプロモーター駆動緑色蛍光タンパク質(GFP)導入遺伝子、Jackson Laboratory,Bar Harbor,ME,https://www.jax.org)から単離した。GFP導入遺伝子(GFP−Tg)ASCは、単離した時にGFP導入遺伝子を発現すること、それに、少なくとも10継代に向けてのインビトロでの増殖を維持し、そして、2〜2.5日の細胞倍加時間を示す。最初の特徴決定のために、これらの細胞を、マーカーCD11b(Mac−1α;インテグリンアルファM)、CD29(β1インテグリン)、CD34(ムコシアリン)、CD45(白血球共通抗原;Ly5)、CD90(Thy−1)、及び、Sca−1(幹細胞抗原1;Ly6A/E)の発現について調べた。
細胞懸濁液を、Supporting Information Tableに記載の細胞表面抗原に対する抗体と共に、室温(RT)で、30分間、遮光して、インキュベートした。PBSで2回洗浄した後、Beckman−Coulter Galiosフローサイトメーター(BD Biosciences,San Jose,CA,www.bdbiosciences.com)を用いて、フローサイトメトリー分析を行った。これらのGFP−Tg SVF細胞内の免疫表現型、及び、相対亜集団を、Supporting Information Tableに示したスキームを用いて、内皮、造血、間葉、及び、幹細胞関連抗原の以下のパネルを検出する蛍光色素結合モノクローナル抗体を用いて、プラスチック接着培養の継代2まで決定した。
GFP−Tg C57BL/6マウス由来のGFP−Tg細胞を利用した2つの研究を行った(図8)。これらは、GFP−Tg未分画SVF細胞の連続移植、及び、生細胞の選別をして、培養増殖したGFP−Tg ASC亜集団の連続移植を含む。最初の研究では、GFP−Tg SVF細胞を、GFP−Tg集団に対してフロー選別して選択し、そして、未分画GFP−Tg SVF細胞を、非GFP−TgマウスのGFP−Tg SVF連続移植用のシルクスキャフォールドに直ちに導入した。第2の研究では、GFP−Tg CD146−SVF亜集団を選択し、そして、(a)未分画コントロール、または、(b)CD29陽性、及び、(c)CD34陽性のいずれかとしてプレートした。これらの培養増殖集団(a〜c)を、免疫表現型で分類し、そして、GFP−Tg ASC連続移植のためにシルクスキャフォールドに導入した(ASC連続移植研究;後述するHexaFluoroIsoPropanolシルクスキャフォールドの導入、培養、及び、移植を参照されたい)。
GFP−Tg 1°SVF細胞の生細胞選別を、2つのレーザーと、8つの検出器を装備したGalios収集ソフトウェア(BD Biosciences,San Jose,CA)を搭載した、BD Biosciences蛍光活性化セルソーター(FACS)Beckman−Coulter Galiosフローサイトメーターを用いて行った。選択した全蛍光色素と、細胞選別目的のためのトランスジェニックGFPの継続的使用との適合性を保った。マウス(mu)CD3、CD45R、CD116、LyG6、Ter−119(eBioscience;88−7772)を検出するeFluor結合抗体のカクテルを用いて、系統除去を行った。生SVF細胞選択を、APCコンジュゲート抗muCD29(17−0291)と、PEコンジュゲート抗muCD34(56−0341)を用いて行った。プラスチック接着性培養増殖をした後に、かつ、International Federation of Adipose Therapeutics(IFATS)及びISCTが確立した表現型及び機能的基準を用いて、ASCを単離した。要するに、継代0°(P0)のASCを、5mlの0.25%トリプシン(Life Technologies,Grand Isle,NY)で、5分間、トリプシン処理した。等量のASC培地を添加して、トリプシン消化を停止した。トリパンブルー染料排除法を用いてP0 ASCを計数し、そして、0.4×103個の細胞/平方センチメートルの密度で、新たなT175フラスコに継代した。これらの細胞を、それらが70%のコンフルエントに達するまで培養した。これらの集団を、(a)コントロールとして未分画のもの、または(b)CD29+を豊富に含有した集団(Lin−CD29+ CD146−)、または、(c)CD34+を豊富に含有した集団(Lin−CD34+ CD146−)に細分化して、P2にまで培養増殖をした。これらの(a)未分画コントロール、(b)CD29+を豊富に含有したASC集団、及び(c)CD34+を豊富に含有したASC集団を、個別に、フラスコに播種し、そして、我々が公表し、かつ、検証をした方法に従って培養増殖した。CD29について選別した後、これらの細胞の95.03±2%が、CD29及びGFPを発現した。播種に向けて相当数の集団を得るために、これらのSVF亜集団を、直ちに、プレーティングし、増殖し、そして、継代した。これらの細胞(CD29+、及び、CD34+亜集団)を個別に選択し、そして、CD29及びCD34双方の共発現に基づいて選別しなかった、ことに留意されたい。加えて、最初に選別したCD34亜集団(GFP及びCD34の双方に関して、99.85±4%が陽性)は、CD34を共発現するその他のSVF前駆体集団を含み、かつ、接着性ASC集団のプラスチック接着性の増殖及び選択をする間に排除されるので、SVF細胞において最初に選別したCD34発現は、接着性及び増殖のプロセスに応じて、表面マーカーの発現を失う可能性がある培養増殖したCD34で選択したASCの発現よりも高発現である。継代2(P2)において、各SVF画分由来のASCのアリコート(1×105 ASC)を、GFP陽性、及び、抗原CD29、CD31、CD34、CD45、CD73、CD90、CD105、CD146、及び、Sca−1についての分析的フローサイトメトリーで特徴決定をして、増殖に起因するプロファイルの変化をモニターした。並行して、生P2 GFP−Tg ASCを、インビトロでの増殖、コロニー形成、及び、脂質生成分化について、または、移植研究について評価をした。
Tissue Engineering Resource Center,(Tufts University,Medford,MA,http://ase.tufts.edu/terc/)Universityから入手した当該HexaFluoroIsoPropanol(HFIP)シルクスキャフォールドを、4℃で、一晩、Stromal Medium(10%ウシ胎児血清、及び、1%ペニシリン/ストレプトマイシンを補充したDulbecco’s modified Eagle’s−Ham’s F−12培地)で、予め湿潤させた。このスキャフォールドを、非分画SVF細胞を用いた研究、ならびに、分画継代2(P2)ASCを用いた研究で使用した。SVF研究のために、新たに単離したGFP−Tg iWAT 1°SVF細胞を、我々が公表した方法[33]に従って、円筒形HFIPシルクスキャフォールドの反対側にある面に対して、総数で2.5×105個の細胞を含む50μlの2つのアリコートを、それぞれ、ピペットで導入した。ASC研究のために、新たに単離したGFP−Tg iWAT 1°SVF細胞の試料を、直接にプレーティングする(未分画)か、または、CD146− CD29+、もしくは、CD146−/CD34+発現に基づいて選別した。これらの副画分をプレーティングし、そして、P2まで培養した。未分画SVFコントロール、CD146−CD29+を豊富に含有した集団、及び、CD146−CD34+を豊富に含有した集団に由来するP2 ASCを、円筒形HFIPシルクスキャフォールドの反対側にある面に対して、2.5×105個の細胞を含む50μlの2つのアリコートを、それぞれ、ピペットで導入した。
細胞播種直後に、各播種スキャフォールド内の代謝的に活性な幹細胞の相対数を、製造業者の指示に従って、AlamarBlue(商標)アッセイで決定した。播種したスキャフォールドを、10%Alamar Blue試薬を補充したStromal Mediumにて、5%CO2、37℃で、2時間、インキュベートした。培養培地のアリコート(100μl)を、96ウェルプレートに移し、そして、560nmの励起波長、及び、590nmの発光波長を利用する、マイクロタイタープレートリーダー(Fluostar Optima)を用いて、蛍光強度を定量した。播種していないスキャフォールド、及び、組織培養ウェルも、上記したようにして培養培地で維持し、そして、バックグラウンド蛍光を調整するためにブランクコントロールとして同様に分析をした。次いで、スキャフォールドを秤量し、そして、相対細胞数を、前述したようにして、スキャフォールド湿潤重量の1ミリグラム当たりにおける、それらの相対蛍光強度(RFU)の程度を計算した。
培養増殖、及び、スキャフォールド接種した非GFP−Tg ASCまたはGFP−Tg ASCの脂質生成分化を、前述したようにして、15日間にわたって行った。要するに、培養したASCを、Stromal Medium(10%ウシ胎児血清、及び、1%ペニシリン/ストレプトマイシンを補充したDulbecco’s modified Eagle’s−Ham’s F−12培地)で増殖させた。次いで、ASCをトリプシン処理し、そして、ASC培養培地で、1平方センチメートルあたり3×104個の細胞で、24時間、プレーティングして、付着させた。1日目(プレーティングから24時間後)に、培地を除去し、そして、細胞を、脂質生成分化培地(10%ウシ胎児血清、15mM HEPES[pH7.4]、ビオチン[33μM]、パントテン酸塩[17μM、Sigma]、ヒト組換えインスリン[100nM、Boehringer Mannheim]、デキサメタゾン[1μM]、1−メチル−3−イソブチルキサンチン[IBMX;0.25mM]、及び、ロシグリタゾン[1μM]を補充したDulbecco’s modified Eagle’s−Ham’s F−12培地)で、3日間、インキュベートした。脂肪細胞分化維持期間の残りの9日間は、当該培地を3日ごとに除去し、そして、IBMX及びロシグリタゾンを含まない同じ培地(維持培地)と交換した。
プラスチック接着培養ASCについては、Oil−Red−O(ORO)(Sigma−Aidrich)を、脂肪細胞分化培地で12日間培養したGFP−Tg ASCの単分子層へ組み込んで脂質形成を評価した。ORO取り込みを定量した。要するに、0.5%(w/v)のOROを、エタノールにおいて調製した。次いで、3部のOROと、2部のPBSを混合して、希釈標準溶液を作製した。12ウェルプレートで培養したGFP−Tg ASCの単分子層を、PBSで3回すすぎ、続いて、10%(v/v)ホルマリン(Sigma−Aldrich)で、15分間、固定した。これらの単分子層を、次いで、PBSで3回すすぎ、そして、室温で、45分間、ORO希釈標準溶液でインキュベートした。組み込まれていないOROの吸引後、単分子層をPBSで4回すすいだ。これらの染色した単分子層を、位相差顕微鏡(Eclipse 800,Nikon,Tokyo,Japan,http://www.nikon.com/)で可視化した。これらの染色した単分子層を、100%イソプロパノールで、10分間、インキュベートして、取り込まれたOROを抽出した。次いで、各アリコートの510nmでの吸光度を、96ウェルプレートリーダーを用いて測定した。
移植を行った動物を、6〜8週間維持し、安楽死させ、そして、無菌解剖技術を用いてスキャフォールドを取り出した。以前のインビボ研究は、4〜6週間が、移植したシルクスキャフォールドにおいて、ヒトASC脂質生成を可能にするのに十分な期間である、ことを実証した。6週間後に、共焦点顕微鏡検査によって、GFP−Tg細胞の急激な増殖及び分化が認められた。従って、我々の研究は、6週目に行った。顕微鏡分析のために、n=2個のスキャフォールドのセットを、OCTにおいて新たに凍結した。残りのn=18個のスキャフォールドを、0.1%コラゲナーゼI型(Worthington Biochemicals,Brunswich,Lakewood,NJ,http://www.worthington−biochem.com)、1%ウシ血清アルブミン、及び、2mMのCaCl2を、断続的に振とうしながら、37℃で、60分間、補充したPBSで、1時間、コラゲナーゼ消化に供した。これらの2°(二次移植)SVF細胞を、HFIP Silk Scaffoldの下で、上記したようにして単離した。
2°SVFでのGFP−Tg細胞の頻度、ならびに、それらの関連抗原の発現(上記のモノクローナル抗体(mAb)のパネル)を、各グループ分けの試料を用いて、分析的フローサイトメトリーで決定した。当初のコホート由来の残りの選別した2°SVF細胞を、上記と同じプロセスを繰り返すことで、新たに調製したHFIPシルクスキャフォールドに導入した。インキュベーション後、これらのスキャフォールドに、2°SVF細胞を導入し、そして、6週間にわたってC57Bl/6マウスの両側に移植した。その時点で、スキャフォールドの採取及び分析を繰り返して、3°SVF細胞集団を得た。細胞採取の制約が故に、スキャフォールドの採取と分析は、3°SVF細胞集団、及び、2°ASC集団の後に終了した。ポジティブコントロールとして、C57Bl/6(UBC−GFP)マウス由来のインタクトな鼠径部デポーを、同系の野生型レシピエントに、同期間にわたって連続移植した。加えて、取り出したシルクスキャフォールド構築物内のインスリン感受性を、機能性を保証するために、我々が公表したインビトログルコース取り込み及び脂肪分解アッセイを使用して、決定をした。本研究では、(細胞を含む、または、含まない)スキャフォールドの外科的移植の日と、動物からそれを外科的に取り出した日との間の生着時間を測定していた、ことに留意すべきである。ネガティブコントロール群については、年齢及び性別に適合した非GFP−Tg C57BL6マウスから組織を採取した。
Zenascope PCI分光計(Zenalux,Durham NC,http://www.zenalux.com)は、広帯域ハロゲン光源(プローブ)を使用して、標的組織を照明する。Zenascopeは、侵襲的技術が無くとも、生存動物のタンパク質レベルの測定を可能にする。直径が0.25”のステンレス鋼硬質共通端部にステンレス鋼ジャケットを有し、かつ、そこで終端する分岐光ファイバープローブを、組織の表面に適用し、標的組織に光を送達し、そして、反射した光信号を収集し、及び、当該組織の光学的特性(波長依存吸収、及び、散乱係数)を、500〜650nmの波長範囲にわたって測定した反射率スペクトルから定量的に抽出する。抽出した吸収係数から、携帯型の標準化した測定ハードウェアは、オキシヘモグロビン(HbO2)、デオキシヘモグロビン(dHb)、総ヘモグロビン濃度(Hb、HbO2+dHbとして定義した)、及び、ヘモグロビン酸素飽和度(sO2、比率HbO2/Hbとして定義した)の迅速リアルタイム定量分析を達成する。光学的に減少した散乱係数もまた、機器が測定及び報告を行い、このことは、組織発色団の測定値を、組織散乱における変化とは独立して正確に定量することを可能にする。積分時間は、検出器の線形応答範囲内に留まりながら反射率信号を最大にするために、各測定についてシステムソフトウェアによって自動的に設定されており、典型的には、100から200msの範囲であった。99%の反射率標準を用いたプローブの反射率較正を、全ての実験の前に、そして、手順の間に定期的に行った。
ASCまたはSVF長期生着時間について報告するために、各コホートから取り出した全移植片の3つの代表的な画像を、TIFファイルに変換し、そして、分析のためにImageJにエクスポートした。未播種のスキャフォールドの画像(コントロール)を、(ImageJ Version 1.46ソフトウェア、NIH)を用いて定量した。画素値は、組織移植片全体からのものであり、切片化した画像ではない。閾値を、コントロールを用いて背景ピクセル数を差し引くことで設定した。組織切片の群からの相対平均ピクセル数を平均し、標準偏差を計算した。
24時間の細胞培養後に、GFP−Tg ASCを播種したスキャフォールドについて、極低温走査電子顕微鏡検査を行った。Cryo−SEM構造研究を、Gatan 2500 alto Cryo−system、及び、Hitachi S−4800走査型電子顕微鏡(SEM)を用いて行った。組織を、7×5×5(mm)に切断し、そして、極低温SEMサンプルホルダー表面から約5mmの高さに置き、ホルダーに固定して接着し、そして、約−210℃で、約30秒間、液体窒素で凍結した。この凍結組織を、真空中で、SEMに取り付けられたプレチャンバーに移し、次いで、破砕し、そして、−95℃で、5分間、昇華させた。−130℃で、88秒間、Pt/Pdでコーティングした後に、当該試料をSEMに移し、そして、−130℃で、3kVで観察した。
新たな凍結組織サンプルを切片にし、親油性蛍光色素(Bodipy)で染色し、そして、GFPと脂質シグナルの共局在について蛍光顕微鏡で評価した。
Qiagen FFPE tissue prep Mini Kit(Qiagen inc.,Valencia,CA,http://www.qiagen.com)を、製造業者の指示書に従って使用して、取り出した組織改変脂肪パッド、または、ポジティブ、もしくは、ネガティブコントロールから全DNAを単離した。SYBRグリーンリアルタイムPCR DNA結合色素を用いて、約500ngの全DNAを使用して、ポリメラーゼ連鎖反応を行った。反応混合物を、GFPに特異的なオリゴヌクレオチドプライマーセット(フォワード:5’−TCGCCTACCAGCTCATGCATAACA−3’;リバース:5’−TGAAGCTCTTCCAGGTGTCAACGA−3’)と共に、リアルタイム熱検出システム(Bio−Rad Laboratories,Hercules,CA,http://www.bio−rad.com)でインキュベートした。全ての結果を、グリセルアルデヒド3’リン酸デヒドロゲナーゼ(GAPDH)発現コントロールと比較して正規化した。
全ての研究を、少なくとも三重に繰り返し、そして、数値は、平均±標準偏差(SD)として報告する。個々の対は、スチューデントT検定を使用して比較し、一方で、大きいグループは、分散分析で分析した。最小p値が、≦.05であれば、所見は、有意であると定義した。
利用した細胞の単離及び特徴決定
図1Aは、8〜12週齢のオスのマウスから単離したiWATのGFP−SVF内で検出した細胞の代表的な亜集団を提供する。初期SVFは、1ミリリットルの脂肪組織当たり、平均3〜5×105個の1°(一次)SVF細胞を産生する。これは、文献で報告されている数値と近似している。表面免疫表現型に基づく3つの最も大きな亜集団は、前脂肪細胞(24.7±6.9%;CD14−、CD36+)、ASC様(20±10.7%;CD146−、CD34+、CD90+)、及び、白血球(19.4±9.7%;CD34−、CD45+)であった。注目すべきことに、当該ペレット化SVF細胞集団は、コラゲナーゼI型組織消化、及び、分化遠心分離による単離のプロセスが故に、浮遊成熟脂肪細胞(全集団の約30%と推定される)を排除する。最初のSVF画分を、まず、CD146−亜集団(80.5±5%)について選別した。このことは、分画したSVF細胞の初期コホートとして役立った。その他のコホートを、CD34及びCD29表面抗原発現、すなわち、(b)CD29+(CD146−集団の18.2±8.5%)、及び、(c)CD34+(CD146−集団の21.4±12.8%)SVF細胞についての選別に基づいて選択した。フローサイトメトリー選別プロトコルを、図2に示す。これらのコホートの特徴[(a)全CD146−SVF細胞、(b)CD146−、CD29+を豊富に含有した細胞、及び(c)CD146−、CD34+を豊富に含有した細胞]を、プラスチック接着細胞培養の免疫表現型、増殖、コロニー形成、及び、継代2(P2)での脂質生成の可能性に基づいて決定した。その他の公表した報告と同様に、間質マーカー(CD29)は、発現が大きくなっており、そして、内皮マーカー(CD31、CD45)、及び、造血マーカー(CD34、CD11b)は、継代して発現が小さくなった。Sca−1発現は、未選別のSVF集団内で平均61.6±10.7%であり、また、継代を通じて増加した(図2A)。CD29及びCD34の濃縮は、Sca−1発現を小さくした(CD29;13.8±2.4%、CD34、13.7±0.9%)。CD34+を豊富に含有した亜集団は、CD29+を豊富に含有した亜集団よりも有意に高い増殖能と脂質生成能とを示した(図2B)。さらに、CD29+亜集団は、未選別の集団よりも倍加時間が遅く、また、細胞質内の脂質蓄積が少なかった(図2C、2D)。
GFP−ASCのインビトロでの播種、及び、増殖特性を、HFIPをベースとしたシルクスキャフォールドに播種した後に調査した。インキュベーションの24時間後に、播種した、及び、未播種のコントロールスキャフォールドに対して、共焦点顕微鏡観察を行った(図9)。GFP検出、及び、Alamar Blue染色は、シルクスキャフォールドハニカム様構造の端部での細胞局在化の優先度を示した(図9B、明視野、及び、図9D、走査型電子顕微鏡)。これらの端部に残存した細胞は、分化(図9C)、及び、増殖(図9D、9E)を始めた。Alamar Blue(図9D)サイトゾル、及び、DAPI(図9E)核陽性の定量では、播種して2時間後に細胞は存在していた(18時間;16±2細胞;2時間、11±8細胞;図9E)が、スキャフォールドに付着し、そして、播種して18時間後にRFUで測定可能な代謝活性を示した(18時間、36RFU;2時間、0.001RFU;図9E)。
6週間の時間経過を用いて、未分画SVF細胞を、増殖及び脂質生成についてモニターした。写真分析は、SVF細胞の存在が、1週目までに生着を促すことを明らかにした(図3A)。パーセントヘモグロビン飽和度の測定値(%Hb;図3B)も、観察したSVF細胞媒介血管新生の促進を支持した。ヘモグロビン測定は、生存しているマウスの皮膚表面で行っており、そして、飽和度の測定は、単なる動脈の血液供給ではなく、全ての組織を評価する、ことに留意すべきである。2週目以降は、視覚的にも、また、ImageJを用いた外植片画像の定量でも、生着に有意差は認められなかった(図3C)。このことは、インビボでの脂質生成を促すスキャフォールドの血管特性及び支持特性を示唆している公表されたデータと符合した。スキャフォールド質量の測定(図3D)、及び、播種したスキャフォールドから回収した総SVF(図3E)は、1週目の間、未播種のコントロールよりも有意に大きかった。SVF細胞計数、及び、スキャフォールド質量は、2週目に最大に達し、そして、4週目までに減少した(パネル3D、3E)。
1週目から6週目までの初期移植での未選別GFP−SVFの毎週の検出(図4A)は、GFP−SVF細胞が、スキャフォールド内で持続するだけでなく、移植をして1週目という早い時期に増殖、及び、分化することを明らかにした。最初のSVF外植片の1、4、5、及び、6週間目についての共焦点顕微鏡画像を示す(図4A)。GFP−Tg SVF細胞を、初期コホート(T0移植)、ならびに、初期移植(T1移植)、及び、二次連続(T2移植;図4B)移植片由来の外植片内で検出した。GFP−Tg SVF細胞のフローサイトメトリーに基づいた検出は、共焦点顕微鏡観察と一致していた(図4C)。最初の移植では、4週目にフローサイトメトリーを介して19%のGFP抗原陽性が認められたが、T1及びT2移植では、移植と比較して同じ時点で、それぞれ、16.2%及び13.1%のGFP陽性が認められた(図4C)。GFPの定量、BODIPY親油性色素染色(構築物の脂質生成性を実証するため)、及び、核DAPI染色の共局在化を、ImageJを用いて行った。定量は、1週目と比較して、T0移植をして6週後までに、GFP−Tg SVF細胞パーセンテージは50%増加していた(図4D)。図4Dに報告したフローサイトメトリーデータは、図4Cに報告した組織から単離したSVF細胞について行っている、ことに留意することが重要である。脂肪組織は、成熟脂肪細胞、及び、組織消化手順に耐えられないその他の細胞を含有しており、そして、フローサイトメトリー数に反映されない。従って、図4Dは、組織試料での全ての細胞を反映していない。フローサイトメトリー、及び、qPCRのそれぞれを介した、タンパク質及びDNAレベルに基づいて、取り出した外植片でのGFP発現をさらに調査した(図4E、4F)。フローサイトメトリーを介した、パーセントGFP−Tg細胞の毎週の検出は、T0移植後の2週目の3.8%から4週目の16%へと発現を増大することを明らかにした(図4E)。T2移植後、GFP DNA発現を、200倍以上へと豊富にした(非トランスジェニックマウスにおける未播種スキャフォールド移植)(図4F)。T2移植集団由来の細胞の表面免疫表現型も、6週間後に、GFP−Tgコントロール、及び、未播種のスキャフォールドコントロールと比較した(図4G)。このT2スキャフォールドは、36.3±2.4%のGFP陽性、67.8±4.1%のCD29陽性、及び、52.4±2.4%のCD45陽性を示した(図4G)。
GFP−Tg SVF細胞を単離し、そして、未分画細胞として分類するか、または、CD146− CD29+を豊富に含有した、または、CD146− CD34+を豊富に含有した群として選別した。これらの細胞を、P2 GFP−Tg ASCまで培養増殖し、シルクスキャフォールドに播種し、そして、T0連続移植まで非トランスジェニックマウスに移植した。移植片の除去の6週目では、脂肪デポーサイズの視覚的差異は認められなかった(図5A、5B)。パーセントヘモグロビン(%Hb)飽和度、及び、外植片生着の測定を行った。CD29−、及び、CD34−を豊富に含有するGFP−Tg ASCの双方を播種した移植片は、未播種の移植片、及び、未選別のASCコントロールを播種した移植片の双方と比較して、移植の1週後に、高い%Hb飽和度を示した(図5C)。さらに、CD34−を豊富に含有する移植片は、CD29−を豊富に含有する移植片よりも有意に高い%Hb飽和度を示した(未播種ASC:コントロール、37.6±9.9%;CD34−を豊富に含有するもの、68.1±5.3%;CD29−を豊富に含有するもの、45±10.8%;図5C)。このことは、顕微鏡画像のImageJソフトウェア分析を用いて、視覚的及び定量的に認められた(図5D)。外植片の質量、及び、周辺脂肪の質量の測定(図5E)は、CD34−を豊富に含有する移植片由来の人工構築物が、CD29−を豊富に含有する、及び、未選別のGFP−Tg ASC、ならびに、未播種のコントロールよりも著しく密度が大きい、ことを明らかにした(図5E)。CD34−を豊富に含有するASCを接種したスキャフォールドから回収した総SVF(図5F)もまた、1週目の間に、CD29−を豊富に含有する未選別ASC、及び、未接種のコントロールよりも有意に高かった(図5F)。組織を改変した脂肪の機能性のさらなる研究は、CD34−を豊富に含有する構築物が、未選別のASC構築物に匹敵するグリセロールレベルで分泌し、その一方で、CD29−を豊富に含有する構築物は、有意に低いレベルのグリセロールを分泌することを明らかにした(図5G)。コホート間で、グルコース取り込みにおいて、有意差は認められなかった(図5H)。
初期コホート(T0移植)、ならびに、初期連続(T1移植;図6A)から6週間以内に、GFP−Tg ASCを検出した。これらの組織移植片を血管で浸潤させ、このことは、GFP−Tg SVF、及び、GFP−Tg ASCを播種したスキャフォールドによる組織生着の早期の観察と符合している。新たに単離したGFP−Tg細胞のフローサイトメトリーをベースとした検出は、移植の6週後の共焦点顕微鏡観察でのGFP陽性を支持した(図6C、6D)。フローサイトメトリーによって、10.9%のGFP陽性を示したCD29−を豊富に含有する構築物と比較して、当初のCD34−を豊富に含有する構築物では、29.4%のGFP抗原陽性が検出された(図6C)。CD34−を豊富に含有する、及び、CD29−を豊富に含有するT1移植片は、それぞれ、20.4%、及び、8.4%のGFP陽性を示した(図6D)。GFP、BODIPY親油性染料、及び、DAPI共局在化染色の定量を、ImageJを使用して行った。外植片内のGFP発現を、qPCR、及び、フローサイトメトリーのそれぞれを介して、DNA、及び、タンパク質レベルに基づいてさらに調べた(図6E−6G)。検出したGFP DNAは、CD29−を豊富に含有する構築物と比較して、CD34−を豊富に含有する構築物において15倍多く、また、T1移植後の未選別のASC構築物よりも10倍以上も多かった(図6E)。フローサイトメトリーを介したパーセントGFP−Tg細胞の検出は、T0移植片(CD29、9.8±1.1;CD34、24.7±6.5;図6F)、及び、T1移植片(CD29、5.4±1.2;CD34、17.5±1.4;図6G)の8週間後のCD29−を豊富に含有する構築物と比較して、CD34−を豊富に含有する構築物由来のGFP−Tg細胞の発現における有意差を支持した。
播種したGFP−Tg SVF、未選別のGFP−Tg ASC、GFP−Tg CD34−を豊富に含有するASC、GFP−Tg CD29−を豊富に含有するASC、ならびに、GFPポジティブ及びネガティブコントロール由来の脂肪周囲から除去した8週間連続移植片を取り出し、そして、GFP陽性(図7A−7C)と機能性(図7D、7E)について分析をした。BODIPY染色して取り出した脂肪周囲の8週間構築物の共焦点顕微鏡画像は、CD29−を豊富に含有した、未選別ASC、及び、SVF播種構築物と比較して、CD34−を豊富に含有するコホートにおけるパーセントGFP−Tg脂肪細胞の有意な増加を明らかにした(図7A、7B)。ImageJを用いた画像の定量は、目視の観察と符合した(SVF,2.5±1.7%;未選別GFP−Tg ASC,15.5±0.2%;CD29−を豊富に含有したもの、6.1±7.0%;CD34−を豊富に含有したもの、31.3±0.4%;図7C)。組織改変した脂肪機能性のさらなる研究は、CD34−を豊富に含有する構築物が、未選別ASC構築物に匹敵するグリセロールレベルを分泌するのに対して、CD29−を豊富に含有する構築物は、有意に低いレベルのグリセロールを分泌することを示した(CD34,70±16.1μM;未選別ASC,75±22.1μM;CD29,40±14.6μM;図7D)。図5での取り出した構築物と同様に、コホート間でグルコース取り込みに有意差は観察されなかった(図7E)。
本研究は、3Dシルクマトリックスを使用して、新たに単離したGFP−Tg SVF細胞を、1回の初期、及び、2回の連続移植(2°移植)で、首尾よく非−GFP−Tgトランスジェニック同系マウス宿主に移植した。初期の時間をベースとした移植片除去研究は、GFP−Tg SVF細胞の生存、増殖、及び、分化を実証しており、代謝的に機能的な脂肪組織の形成に至った。時間をベースとしたGFP−Tg SVF実験はまた、生着を増強し、そして、生着時間を短縮するSVF細胞の能力を明らかにした。増殖したプラスチック接着性CD146−CD29+、及び、CD146−CD34+ P2 GFP−Tg ASC集団を、3Dマトリックスに播種し、そして、初期、及び、連続移植(1°移植)へと首尾良く非GFP−Tgトランスジェニック宿主に移植した。我々の知る限りでは、我々は、間質性血管コンパートメント内の非周皮細胞性ASCの特定の亜集団が、連続移植を通じて機能的脂肪を生成する能力を初めて報告した。
「間質/前駆体」細胞と「幹」細胞との間で、細胞カテゴリーを区別することには、依然として課題がある。それにもかかわらず、現在の連続移植での知見は、成体マウスの鼠径部WATが、フローサイトメトリーと、機能性脂肪組織デポーの再生、及び、連続移植に寄与することができる細胞の選別可能な亜集団とを含む、ことを示唆している。これらの知見は、フローサイトメトリーで選別した脂肪由来細胞が奏する、脂肪異栄養性マウスにおける脂肪デポーの再生能力を試験しているRodeheffer et al.の初期の研究を確認し、また、発展させるものである。加えて、これらは、機能的組織改変のための自家脂肪由来細胞の有用性をさらに検証する。この研究はまた、単離した脂肪細胞前駆細胞の免疫表現型、及び、周皮細胞特性を特徴決定する以前の研究を発展させるものである。
hPLとDMEM F12を組み合わせて、37%で温めると、その生成物は、増殖した(1〜2ヶ月)3D培養物、及び/または、皮下注射によるインビボ移植に資する熱反応性バイオスキャフォールドとなる。
hPLの調製
特に断りがない限り、生物学的安全キャビネット内ですべての手順を行う。所望の量の濃縮した有効期限の過ぎたヒト血小板を、15mLまたは50mLのコニカルチューブに採取する。直ちに3D細胞培養に使用しない場合は、3回の凍結/解凍サイクルで、溶解物調製を行う前に、−20℃で保存する。3回の凍結/解凍サイクルを実施して、浸透作用によって血小板を溶解する。これは、細胞培養のための増殖因子を最大収率で放出する。8,000×gで、20分間、遠心分離する。吸引によって、ヒトの血小板の固体(白色)最上層を除去する。
細胞培養に必要な栄養素、アミノ酸、及び、増殖因子を含有する1%抗生物質/抗真菌剤、及び、91.5%Dulbecco’s Modified Essential Medium、または、その他の関連細胞培養培地に対して、7.5%(v/v)のhPLを添加して、hPL補充培地を調製する。混合した後は、水浴で温めない。室温に維持する。
調製したhPLを、細胞集団、または、細胞集団の混合物と組み合わせることにより、細胞外マトリックスの細胞分泌、増殖、及び、分化を促すゲル化(厚いマトリックスの形成)を活性化する。これらの細胞とhPLとを混合した後に、ゲル化の活性化のために、20分間、プレートを、インキュベーターに加える。増殖、分化、マウスへの移植、または、その他の測定終点へと進む。
皮膚癌を除外すると、乳癌は米国の女性において最も頻繁に認められる腫瘍であり、がん関連の死因の第2位を保っている。乳癌死亡のリスクは、アフリカ系アメリカ人女性で特に高く、この疾患は、しばしば食事/栄養、経済的格差、遺伝、健康へのアクセス、及び、肥満に関連する要因に起因する予後不良と関連している。免疫不全マウスにおいて維持した患者由来の異種移植片(PDX)モデルは、新規の乳癌療法を開発するための前臨床モデルとして有望であるが、受託研究、及び、腫瘍薬理学的開発のために市販されるPDX乳癌の数には限りがある。本発明者ら、すなわち、マイノリティーで、かつ、女性が所有しているバイオテクノロジー会社は、大都市のニューオーリンズ地域と、隣接するルイジアナ州南東部に居住する、民族的に多様な患者の人口統計学から乳房腫瘍標本にアクセスする、というユニークな立地にある。
厳密性と透明性:免疫不全マウス株は、そのマウス株の厳密な分析と品質保証を行い、NIHが支援する受託研究施設であるJackson Laboratoryから直接に入手する。確立した乳癌細胞株は、その製品の厳密な分析を行い、NIHが支援する細胞貯蔵施設であるthe American Type Culture Collectionから直接に供給を受ける。発明者は、SVF細胞ドナーの重要な人口統計(年齢、性別、肥満度指数、民族性)を、身元を伏せた匿名記録で維持する。同社の細胞製品のすべての凍結保存したバイアルは、最新のデータベースに入力され、そして、貯蔵容器の完全性を、同社の技術スタッフが毎日監視する。本発明者は、インビトロ培養、フローサイトメトリー、及び、分化アッセイを使用して、その哺乳動物血小板溶解産物、及び、SVF細胞に対して、日常的に品質保証及び品質管理の試験を実施する。女性の乳癌の発生は、男性のそれを、ほぼ2桁上回っているので、第I相試験は、メスのNSGマウスと、女性のドナーから単離したhSVF細胞を用いて行う。オスのマウス、及び、男性の脂肪ドナーを対象としたその他の試験は、試験セクションで推奨されている場合にのみ、第II相で検討する。
生物学的試薬の認証は、実験1に概説したようにして行われる。加えて、本発明者らは、署名した患者の同意を得て、または、新たに改訂されたFDA規制を満足し、かつ、認可を受けている場合には、権利放棄を謳った署名入りの同意を得て、承認済みIRBプロトコルの下で、協力病院を拠点とするバイオバンクから腫瘍標本を得る。発明者らに提供されたすべての標本は、特定事項については、匿名のままである。本発明者らは、腫瘍及び患者の人口統計(年齢、性別、肥満度指数、民族、過去の病歴、社会歴、腫瘍の組織学)に関連する患者情報だけを受け取る。バイオバンク、及び、そのIRBプロトコルに関して指定された治験医だけが、組織ドナーに関する追加情報にアクセスすることができる。
Claims (47)
- 哺乳動物血小板溶解物、及び、脂肪組織由来細胞画分(ATDCF)細胞を含む、バイオロジカルスキャフォールド。
- 前記ATDCF細胞が、間質血管画分細胞、脂肪由来間質細胞、脂肪由来幹細胞、骨髄由来間葉系間質細胞、及び、骨髄由来間葉系幹細胞を含む、請求項1に記載のバイオロジカルスキャフォールド。
- 前記SVF細胞が、ヒト脂肪組織に由来する、請求項2に記載のバイオロジカルスキャフォールド。
- 前記哺乳動物血小板溶解物が、ヒト血小板に由来する、請求項1に記載のバイオロジカルスキャフォールド。
- 前記バイオロジカルスキャフォールドが、SVF細胞を含む、請求項1〜4のいずれか1項に記載のバイオロジカルスキャフォールド。
- 前記バイオロジカルスキャフォールドが、脂肪由来間質細胞、及び、脂肪由来幹細胞の少なくとも1つを含む、請求項1〜4のいずれか1項に記載のバイオロジカルスキャフォールド。
- 前記バイオロジカルスキャフォールドが、骨髄由来間葉系間質細胞、及び、骨髄由来間葉系幹細胞の少なくとも1つを含む、請求項1〜4のいずれか1項に記載のバイオロジカルスキャフォールド。
- バイオロジカルスキャフォールド、及び、基材を含む治療薬であって、前記基材が、前記生物学的製剤を適用するための少なくとも1つの表面を有する、前記治療薬。
- 前記基材が、組織培養プレート、整形外科用装置、歯科用移植片、脱塩骨スキャフォールド、包帯、合成メッシュ、金属製移植片、または、セラミック製移植片を含む、請求項8に記載の治療薬。
- 前記バイオロジカルスキャフォールドを、混合、播種、灌流、毛管作用による充填、遠心分離による充填、注入、印刷、または、バイオロジカルスキャフォールドのバイオ印刷によって、基材に接触させる、請求項9に記載の治療薬。
- 前記バイオロジカルスキャフォールドが、プレ脂肪細胞、血管平滑筋細胞、内皮細胞、及び、前駆体、B及びTリンパ球、及び、前駆体、造血前駆体、マクロファージ、単球間葉系細胞、ニューロン、ニューロン前駆細胞、内皮細胞、内皮前駆細胞、線維芽細胞、真皮、表皮、がん由来細胞、がん幹細胞、軟骨細胞、筋肉細胞、脂肪細胞、骨芽細胞、破骨細胞、骨髄細胞、血液細胞、ならびに、それらの組み合わせさらに含む、請求項1に記載のバイオロジカルスキャフォールド、または、請求項8に記載の治療薬。
- 前記基材が、骨である、請求項8に記載の治療薬。
- 前記基材が、コラーゲン、脂肪細胞外マトリックスタンパク質、または、hPLゲルを含む、請求項8に記載の治療薬。
- 前記基材が、ゲル、半固体、または、固体基材である、請求項8に記載の治療薬。
- 前記細胞が、哺乳動物細胞である、請求項11に記載のバイオロジカルスキャフォールド、または、治療薬。

- バイオロジカルスキャフォールド含有製剤または装置を製造する方法であって、前記方法が、
脂肪組織を採取し、
前記脂肪組織からATDCF細胞を単離し、
哺乳動物血小板溶解物を調製し、
前記ATDCF細胞と、前記哺乳動物血小板溶解物とを合わせて、バイオロジカルスキャフォールドを形成し、及び、
前記スキャフォールドを、基材の少なくとも1つの表面に適用または接触させる、
工程を含む、前記方法。 - 前記ATDCF細胞が、SVF細胞である、請求項24に記載の方法。
- 前記SVF細胞が、内皮前駆体からなる、請求項24に記載の方法。
- 前記SVF細胞が、造血前駆体からなる、請求項24に記載の方法。
- 前記SVF細胞が、免疫細胞からなる、請求項24に記載の方法。

- 前記哺乳動物血小板を、凍結/解凍プロセスを用いて溶解する、請求項28に記載の方法。

前記バイオロジカルスキャフォールドでの細胞の生存について試験をする、
ことを含む前記方法。 - 前記薬剤が、増殖因子、分化因子、サイトカイン、ケモカイン、アジポカイン、抗生物質、抗真菌薬、抗真菌剤、抗炎症剤、または、抗癌剤を含む、請求項43に記載の方法。
- バイオロジカルスキャフォールドと、ヒト疾患の再現病変とを含む機能性ヒト脂肪パッドを含む免疫障害または免疫不全マウスであって、前記マウスが、異種移植マウスである、前記免疫障害または免疫不全マウス。
- 1つ以上のバイオロジカルスキャフォールドを含有するマイクロ流体装置であって、前記バイオロジカルスキャフォールドが、単離され、かつ、培地、及び/または、薬剤の均一な流れに供することができる、前記マイクロ流体装置。
- 薬剤の有効性または毒性を試験する方法であって、前記方法が、
互いに分離されており、かつ、マイクロ流体装置に入れられている、少なくとも2つのバイオロジカルスキャフォールドを調製し、
前記マイクロ流体装置を薬剤で処理し、
前記マイクロ流体装置を、前記薬剤と共に培養し、及び、
前記マイクロ流体装置での細胞の生存について試験をする、
ことを含む前記方法。
