Abstract
Description
インビボでの組織及び細胞は、発達及び形態形成において形成される優先配向で大部分が組織化されている。その目的がインビトロ条件で組織機能を複製することである人工組織の場合、特定の細胞組織化を達成することは、人工組織の将来の機能性に影響を及ぼし得る。いくつかの例には、筋線維の平行整列、腱のコラーゲン線維、及び血管平滑筋細胞のらせん整列が含まれる。例えば、組織化された筋繊維の累積的な微小収縮は強い収縮を達成することを可能にし、腱及び心臓弁のような耐荷重性組織において、細胞外マトリックス(ECM)の一部としてのコラーゲン繊維の平行整列は組織の靭性を著しく改善する。さらに、脊髄などの軟組織では、神経索損傷の修復を改善するために、本来の組織化と同様に正しい方向に刺激を伝達するために軸索の整列した組織化が必要とされる。
本明細書の文脈における生分解性ポリマー又は生体吸収性ポリマーという用語は、人体内で、又は細胞培養条件下で能動的に(酵素的加水分解によって)又は受動的に(化学的加水分解によって)、日から年に及ぶ時間枠で、分解するポリマーに関する。生分解性ポリマーは、2つの品質によって特徴付けられる:ポリマーは、生物学的に意味のある時間枠内で生理学的条件下でその構成モノマー部分又はその誘導体にゆっくり溶解され、それらが含まれる生物体に対して、この分解の生成物は少なくとも溶解の条件下で生じる濃度で毒性はない。分解又は吸収に必要な時間は、ポリマーの性質、移植部位及びポリマーの寸法及び多孔度に依存する。典型的な生分解性又は生体吸収性ポリマーは、PLA、PGA、PGLA又はポリ酪酸である。

本発明者らは、本明細書において、市販の生分解性/再吸収性ポリマー、特にPGAを用いてセンチメートルスケールでマイクロファイバー整列を誘導するための簡単な方法を報告する。例示的実施形態として、本発明者らは不織PGAメッシュを使用した。本発明の方法は、基本的に他の任意の微小繊維状足場に適用することができる。
PGAは、35〜45℃の転移温度(Tg)及び220〜230℃の融点(Tm)を有し、一方、ポリ4‐ヒドロキシブチレート(P4HB)のTg及びTmは、それぞれ5〜15℃及び168〜182℃の間で変化する。PGA足場をP4HB溶液(テトラヒドロフラン(THF)中1.75%)に浸漬し、続いて周囲温度で一晩乾燥することによってPGA足場をP4HBで被覆する。P4HBを添加すると、PGA繊維の分解時間が延び、組織工学応用に用いた場合、足場上の保護層として作用し、同時に交差する繊維を橋渡しすることにより足場の一体性を改善する。両方のポリマーのTgを超えて80℃まで足場を加熱することは、足場繊維及びP4HBの軟化をもたらし、それ故、繊維の切断や足場の損傷を誘導することなく一方向のひずみの適用を可能にした。PGAのTg以下の温度を例えば室温まで連続的に低下させると、繊維を相互に連結し、足場の一体性を保持しながら、P4HBとともに、繊維網を新しい配置に固化及び設定することができた(図1a)。マクロスケールでは、一方向性のひずみによって、幅を減少させ、長さを増大することによりポリマー網目構造の長方形の形状が変形した。プラスチックフィルムとは対照的に、PGA網目構造の形状の変化は、弾性フィルムで観察されたものとほぼ同じであり、中央のごく一部に蓄積するのではなく、延伸足場の全長にわたって幅の減少の分布を示した(図1a)。この挙動は、この処理アプローチを、初期にランダムに配向された足場のより大きな面積から整列された足場を得るのに適したものにする(図1)。巨視的測定は、延伸に伴うそれぞれ50%及び100%の長さの増大に関連して、最大53%及び73%の幅の減少を示した(図1b)。足場の厚さは、無ひずみ状態の858±55μmから、50%及び100%延伸PGA状態のそれぞれ1170±172μm及び1036±191μmに増加した。繊維配列を定量化するために、我々は3D蛍光共焦点画像を分析した。100%延伸サンプルでは、53%の繊維が軸ひずみに対して整列(±10度)したが、50%の延伸は34%の繊維整列をもたらした。非延伸PGA網目構造は、軸ひずみに対して整列した繊維がわずか14%であった(図1c及びd)。電子顕微鏡写真はまた、PGA繊維の整列、及び延伸を適用した後のPGA繊維へのP4HB相互接続の存在を確認した(図1e、白い矢印)。
足場に播種したときに細胞が増殖して培地から十分な栄養素を受け取ることを確実にするために、本発明者らは、細胞培養期間中に組織を灌流するために単純な円形シェーカーを使用した。このようなバイオリアクターにより、1mm厚までのかなり厚い組織シートで細胞を増殖させることができた。最初に、ヒツジ血管線維芽細胞をPGA‐P4HB足場を使用して1週間の間に装置内で培養した。その後、アクチン繊維と核整列を分析した(図3a及びb)。分析は、アクチンストレスファイバーの大部分が100%延伸PGA足場に整列していることを示した(図3b)。計数された整列アクチン繊維から、51.1±5.7%が100%延伸足場でPGA繊維の延伸軸に対して±10度、50%延伸足場で37.6±6.5%、及びランダムに配向したPGA足場で6.9%±4.7で整列した(図3b、c)。さらに、細胞核整列分析は、100%、50%及び非延伸PGA足場について、それぞれ、PGA繊維の延伸軸に対して±10度で52.6±10.2%、36.9±13.6%及び14.4±3.7%の整列細胞核を示した(図3d)。これは、アクチン繊維整列分析において得られた結果を支持する。
整列に関する研究の大部分は短時間(数時間〜数日)での細胞核又はアクチン整列を報告しているが、本発明者らは、一週間後の細胞核及びアクチン繊維整列だけでなく、3週間後の主要ECM成分としてフィブロネクチンフィブリル(小線維)束整列も評価した。これを行うために、AlexaFluor(登録商標)488標識フィブロネクチンをウサギ腱線維芽細胞の培地に3週間かけて加えた。培養時間中、標識されたフィブロネクチンは灌流され、ECMに共集合し、足場を横切るフィブロネクチンフィブリル(小線維)ネットワークの可視化を可能にし、フィブロネクチンフィブリル(小線維)の配向角を分析するために使用された(図4)。小線維フィブロネクチン束整列の定量化は、100%延伸サンプル(図4において「整列された足場」と称される)及びランダムPGA足場において行われた。これを行うために、最初に高解像度画像を包含オブジェクト(「しきい値処理済」)について分析し、次にしきい値オブジェクトの長軸の角度を束の配向角度として定義した。整列PGA足場上の集合したフィブロネクチンマトリックスは、ランダムPGA足場と比較した場合、より高い整列を示した(図4a〜g)。しかしながら、ランダムPGA足場におけるわずかに優先的な整列は可視であり、これは整列及び/又は組織収縮の軸に対して平行な流れ方向から生じ得る。断面画像(図4e、f)を用いて、フィブロネクチンがどのように足場を横切って沈着したかを示し、構築物の三次元性及び効率的な細胞播種及び浸潤に対する足場の孔径の適合性を示した。フィブロネクチンは足場を横切って沈着したが、沈着は側面に顕著であり、これは流れに直面し(画像h、k中の矢印)、前面での灌流の増強を示した。3週齢のECMの評価は、この年齢ではECMが安定しているので重要である可能性があり、そのような組織は採取及び動物への移植に適していることが以前に示されている。また、脱細胞化及びさらなるECM足場の移植は、最近注目を集め、動物及びヒトの臨床研究において有望な結果を明らかにした別の選択肢である。通常、操作された組織の脱細胞化は、さらなる処理のために高品質のECMを採取するための適切な範囲と考えられる4〜8週間のインビトロ培養の後に行うことができる。脱細胞化ECMの短期2D再細胞化はまた、細胞が沈着したフィブロネクチン繊維に沿って整列し、繊維の方向性に従うことができることを示した。これを考慮して、ここで行われた3週齢のフィブロネクチンマトリックスの分析は、最終的な細胞整列結果を予測することができ、それは将来の臨床応用のための1つの関心点となるであろう。さらに、3週間後に整列を維持することは、そのような足場のインビボ移植がおそらく新しい組織形成を誘導するのに十分な時間を有するであろう予測指標となり得る。
PGA又は他のポリマー足場からの脱細胞化組織工学マトリックスは、血管及び心臓弁の組織工学において、特に緊急事態及び発展途上国において異種移植片及び同種移植片に対する容易に入手可能な「既製の」代替物として有用であり得る。これに関連して、本発明者らは、脱細胞化プロセス後の整列ECMが細胞整列を誘導することができるかどうかを調べた。これを行うために、ポリジメチルシロキサン(PDMS)表面上のナノパターン化リッジをシリコン‐SU8型から複製した。型は従来のフォトリソグラフィー手法により製造され、Aldo Ferrari博士により寛大に提供された。PDMS表面は、深さ350nm、幅500nm及びリッジ幅500nmの溝を有していた。ナノパターン化PDMS表面をフィブロネクチン(50μg/ml)で被覆し、次いでヒト包皮線維芽細胞(HFF)を被覆表面上に播種した。細胞接着後、AlexaFlour(登録商標)488標識フィブロネクチンを細胞培養培地(5μg/ml)に添加し、細胞に3日間細胞外マトリックスを構築させた。3日後、表面をpH8.5中0.5%のTriton X100を用いて15分間脱細胞化し、かくして脱細胞化ECMマトリックスを得た。その後、無細胞ECM足場にHFFを播種し、24時間インキュベートした。ここでは、イメージング、脱細胞化及び再細胞化のプロセスを容易にするために、3D構築物の代わりに2D表面を使用した。AlexaFlour(登録商標)488標識フィブロネクチンを(培地に標識フィブロネクチンを添加することにより)組み込むと、平坦面と比較してフィブロネクチン小線維束が溝の方向に沿ってどのように整列しているかを容易に視覚化できた。脱細胞化プロセス後のフィブロネクチンECM上への線維芽細胞の再播種は、新しく播種された細胞が組み立てられたマトリックスの方向に沿って整列することができ、古いECMが細胞整列(アクチン整列)を開始し、トポグラフィカルなガイダンスとして役立つことができることを示している。この知見は、さらなる再細胞化及び細胞整列が事前整列脱細胞化マトリックスによって引き起こされ得る既製用途のための代替マトリックスとしての事前整列化脱細胞化マトリックスの適用を支持する。
PLGAをクロロホルムとエタノールの混合物(75:25w/w)に溶解することにより、最終ポリマー濃度6重量%のPLGAポリマー溶液を調製した。ポリマーを室温で一晩溶解し、均質性を得るために使用前に溶液を30分間撹拌した。
足場の調製
市販の不織布PGA足場(厚さ1.0mm;比重70mg/cm3;Biofelt、Bereldange Luxembourg)をテトラヒドロフラン(Sigma‐Aldrich、米国ミズーリ州セントルイス)中の1.75%ポリ‐4‐ヒドロキシブチレート(P4HB;MW:100000;TEPHA Inc.、米国マサチューセッツ州レキシントン)でコーティングした。網目構造の一体性を高め、PGAの分解速度を遅くするためにP4HBを添加した。整列したPGA足場を調製するために、一片のPGA足場を延伸装置に取り付け、オーブン中80℃で2分間加熱した。試験片をオーブンから取り出した後、それらを手動で、それらの元の長さから1.5倍又は2倍に可能な限り速く延伸した。その後、足場を室温に冷却し、将来の分析及び細胞培養研究に使用した。
PGA繊維を可視化するために、405nm又は488nmで励起した際のPGA繊維の自己蛍光特性をレーザー走査共焦点顕微鏡(Olympus、FV1000、日本)で画像化した。PGA足場の画像を得た後、ImageJソフトウェアを用いて繊維配向を決定した。手短に言えば、ImageJの粒子分析器オプションを介して10°刻み(0〜90°)で繊維角度をすべての走査面において計算し、各刻みに対する繊維の数を数え、繊維の総数からの平均百分率(%)として表した。
PGA足場の比重は、重量測定的に決定された、すなわち、足場の重量は、足場の総体積で割られた。足場の長さ及び幅は定規によって測定され、それらの厚さは明視野顕微鏡画像を用いてそれらの断面から測定された。結果はmg/cm 3で表される。さらに、足場の多孔度は、足場の比重をPGAのかさ密度(1530mg/cm3)に割ることによって計算し、百分率として表した。
100N荷重セルを備えた一軸荷重試験機(Instron引張試験機、High Wycombe、Buck、UK:モデル5864)を使用して測定された応力/ひずみ曲線から、異なる延伸PGA足場の機械的性質を得た。長方形の形状及び10×2mm2の試験領域及び800〜1200μmの厚さ範囲を有するサンプルを使用した。破損するまで20mm/分の伸長速度を適用した。ヤング率[MPa]と破断ひずみ[%]はすべての条件で決定された(n=3)。
PGA足場の3D環境で効果的に細胞増殖を可能にするために、我々は単純な拍動流ミニバイオリアクターを開発した。我々の以前の研究は、このバイオリアクターが静的条件(引用)と比較した場合に、工学的に作製されたPGA足場へのECM沈着を改善することを示した。簡潔には、PGA足場を、UV光学接着剤(Norland光学接着剤、米国ニュージャージー州)を使用して、ペトリ皿(直径5cm)の周壁に固定した。足場はオービタルシェーカーの各パルスによって前後に動いていた。足場を取り付けた後、70%エタノール中の10%過酸化水素の混合物及びUV光を1時間使用して装置を滅菌した。全設定を滅菌PBSで少なくとも3回洗浄し、乾燥させ、続いて10%ウシ胎児血清(FBS、Invitrogen、米国)を補ったDMEM‐Glutamax(Invitrogen、米国)、1%ペニシリン/ストレプトマイシン(P/S、Invitrogen、米国)及び50μg/ml L‐アスコルビン酸(Sigma、米国)からなる培地中で細胞播種前に一晩インキュベートした。
ヒト静脈筋線維芽細胞を大伏在大静脈から採取し、血管工学及び細胞整列分析のためのモデルとして増殖した。さらに、アキレス腱由来のウサギ腱細胞を、以前に記載されたように単離して培養したもの(Evrovaら、Macromolecular Bioscience 2016、19、1048)を、工学的に作成された腱を再構築するためのモデルとして使用した。細胞播種は、以前に記載されているように(Moetら、Biomaterials 2005、26、3113)、担体としてフィブリンゲルを使用して行った。手短に言えば、細胞を標準的な細胞培養プロトコルのように解凍し、増殖させ、トリプシン処理した。その後、細胞をPBS(10IUトロンビン/ml)中の滅菌ウシトロンビン(Sigma、USA)溶液中に再懸濁した。10mg/mlの活性フィブリノーゲンに等しい14mgの凍結乾燥ウシフィブリノーゲン粉末(Sigma、USA)を培地に溶解することによってフィブリノーゲン溶液を調製し、この溶液を0.21μmの滅菌フィルターを用いて濾過滅菌した。続いて、トロンビン溶液中の細胞を等容量の滅菌ウシフィブリノーゲン溶液に添加した。短時間混合した後、細胞を含むフィブリン溶液をPGA足場上にピペットで移した。フィブリンゲルの凝固時間は20から40秒の間で変化すると決定された。したがって、培養培地を添加する前に、構築物を37℃及び10%CO2で15分間重合させた。全ての構築物を最初の24時間静的条件下で培養し、その後オービタルシェーカー(VWR、米国)及びミニバイオリアクタープラットフォームを用いて灌流を開始した。シェーカー(振盪機)の回転半径は19mmであり、1Hzの回転速度(60rpm)を試験中の全てのサンプルに適用した。培地の半分を交換し、蒸発した培地を滅菌脱イオン水で調整することによって、培地を3日ごとに交換した。研究中の最適化されたDMEM緩衝化のために、インキュベーションは37℃及び10%CO2で行われた。
特定の培養期間の後、サンプルをアクチン及び核について免疫染色した。免疫染色のために、サンプルをPBSで洗浄し、次いでPBS中の4%パラホルムアルデヒド中で30分間固定した。アルブミンを添加することにより抗体の非特異的吸着を防止した(BSA、2%w/v、30分)。アクチン細胞骨格は、Alexaflour(登録商標)488結合ファロイジン(Invitrogen、米国)を用いて染色した。細胞核をDAPIで染色した(2μg/ml、10分)。サンプルを最後にPBSで3回洗浄し、画像化した。
細胞核整列は、ImageJの共焦点画像及び粒子分析器オプションを使用して分析した。405nmレーザーを使用するとPGA繊維の自己蛍光が干渉するので、核を円形度、サイズによって細分化し、さらにすべてのサンプルにおいて手動でチェックした。核の配向角度を定義した後、繊維の延伸軸に対して±10度の核、すなわち好ましい繊維方向を数え、整列細胞核として提示した。
一元配置分散分析(ANOVA)と事後ボンフェローニ検定(PASW統計 18ソフトウェア)を用いてデータを比較した。エラーバーは標準偏差を表す。0.05以下のp値は統計的に有意であると考えられた。
Claims (15)
- ポリマー足場を調製するための方法であって、
a.第1のポリマーのフィラメントの布地片、特に不織布メッシュ、より詳細にはフェルト様布地を提供するステップ、ここで前記第1のポリマーは生分解性又は生体適合性であり、前記片は長手方向伸長軸によって特徴付けられる;
b.コーティングステップにおいて、前記フィラメントの配列に第2のポリマーのコーティングを施すステップ、ここで前記第2のポリマーは、前記第1のポリマーの融点より低いガラス転移温度によって特徴付けられる;
c.前記片をその長手方向の伸長軸に沿って延伸し、それによって、整列した微小繊維状足場を得るステップ
を含む、上記方法。 - ステップcにおいて、前記布地片がその長手方向伸長軸に沿って175%を超えて、特に175%〜250%、さらには300%まで、さらに特に約200%延伸される、請求項1に記載の方法。
- ‐ステップcで延伸する前に、前記布地片は、前記フィラメントの第1の整列指数iA1によって特徴付けられ、
‐ステップcで延伸した後、前記整列した微小繊維状足場は、第2の整列指数iA2によって特徴付けられ、
‐前記第1の整列指数に対する前記第2の整列指数の比iA2/iA1は、2以上、特に2.5以上、さらに特に3以上である、請求項1又は2のいずれかに記載の方法。 - ‐iA1の値が≦30%、特に≦25%である、及び/又は
‐iA2の値が≧70%、特に≧75%である、及び/又は
‐比iA2/iA1の値は≧2、3、特に≧3である、
請求項3に記載の方法。 - 前記第1及び第2のポリマーがそれぞれガラス転移温度Tg及び融解温度Tmを特徴とし、ステップc.が、
‐第1のポリマーと第2のポリマーの両方のTgより高く、
‐第1のポリマーと第2のポリマーの両方のTmより低い温度で行われ、特に、ステップc.が50℃から150℃、特に60℃から80℃の間の温度で行われる、請求項1〜4のいずれか一項に記載の方法。 - a.前記第2のポリマーは生分解性及び/又は生体適合性ポリマーであり、及び/又は
b.前記フィラメントに塗布される前記第2のポリマーの量は、前記第1のポリマーの前記フィラメントの質量に対して10%から40質量%の範囲である、請求項1〜5のいずれか一項に記載の方法。 - a.第1のポリマーは、ポリエステル、ポリウレタン及びポリアニリンを含む群から選択され、特に第1のポリマーは、
‐ポリグリコリド、ポリ(乳酸)、PLGA、ポリ(トリメチレンカーボネート)、ポリヒドロキシブチレート、ポリ(4‐ヒドロキシブチレート)、ポリ(3‐ヒドロキシブチレート)、ポリ(グリコリド‐co‐カプロラクトン)、ポリ(グリコリド‐co‐トリメチレンカーボネート)、又はグリコール酸若しくは乳酸モノマーを含むコポリマーを含む第1の群から、又は
‐ポリエチレングリコール(PEG)又はPEGコポリマー;ポリアニリン;ポリエチレンテレフタレート;ポリ(2‐メトキシエチルアクリレート);ポリ(2‐メタクリロイルオキシエチルホスホリルコリン);ポリ(テトラヒドロフルフリルアクリレート);ポリスチレン、又は第2の群に含まれる2つ若しくは3つのポリマー種を含むか又はそれらから本質的になるコポリマーを含む第2の群から選択され、及び/又は
b.第2のポリマーは、前段落に記載の第1の群又は第2の群から選択されるが、第1のポリマーとは異なり;
特に、第2のポリマーはポリ‐ヒドロキシブチレート、より特にはポリ‐3‐ヒドロキシブチレート及びポリ‐4‐ヒドロキシブチレートからなる群から選択される、
請求項1〜6のいずれか一項に記載の方法。 - 前記第2のポリマーが、
a.第1のポリマーが請求項7aに記載の第1の群から選択される場合、請求項7aに記載の第1の群、又は
b.第1のポリマーが請求項7aに記載の第2の群から選択される場合、請求項7aに記載の第2の群
から選択される、請求項7に記載の方法。 - 前記コーティングステップにおいて、前記第2のポリマーが溶媒に溶解した状態で前記第1のポリマーの前記フィラメントに塗布され、前記第1のポリマー及び前記第2のポリマーが表1の1行から選択され、場合により、溶媒は表1の同じ行から選択される、請求項1〜8のいずれか一項に記載の方法。
- 前記フィラメントが、100nmから250μmの間、特に1μmから50μmの間、さらに特に10μmから20μmの間の直径を特徴とする、請求項1〜9のいずれか一項に記載の方法。
- a.請求項1のステップaで提供された布地片は、第1のポリマーのかさ密度に対して5%〜15%の足場比重を特徴とする、及び/又は
b.請求項1のステップcで得られた布地片は、第1のポリマーのかさ密度に対して10%〜20%の足場比重を特徴とする、及び/又は
c.請求項1のステップcで得られた布地片は、第1のポリマーのかさ密度に関して測定された足場比重が、請求項1のステップaで提供される布地片の足場比重よりも30%以上、特に50%以上、さらに特に100%以上高いことを特徴とする請求項1〜10のいずれか一項に記載の方法。 - 人工組織を提供するための生体外方法であって、
a.請求項1〜11のいずれか一項に記載の方法に従って整列した微小繊維状足場を提供するステップ;
b.哺乳動物細胞、特に線維芽細胞、筋芽細胞及びニューロンから選択される細胞を含む細胞培養培地中に前記整列した微小繊維状足場を懸濁するステップ
を含む上記生体外方法。 - 第2の生分解性又は生体適合性ポリマーで被覆された第1の生分解性又は生体適合性ポリマーの整列したフィラメントの微小繊維状足場であって、
a.前記足場の多孔度は80%以上、特に85%以上、さらに特に90%以上であり、
b.整列指数が50%より高い(≧50%)、特に≧65%、より特に≧80%であり、及び/又は
c.前記フィラメントの直径は100nmから250μm、特に1μmから50μm、さらに特に10μmから20μmの範囲である
ことを特徴とする上記微小繊維状足場。 - a.第1のポリマーは、
‐ポリグリコリド、ポリ(乳酸)、PLGA、ポリ(トリメチレンカーボネート)、ポリ(4‐ヒドロキシブチレート)、ポリ(3‐ヒドロキシブチレート)、ポリ(グリコリド‐co‐カプロラクトン)、ポリ(グリコリド‐co‐トリメチレンカーボネート)、又はグリコール酸若しくは乳酸モノマーを含むコポリマーを含む第1の群から、又は
‐ポリエチレングリコール(PEG)又はPEGコポリマー;ポリアニリン;ポリエチレンテレフタレート;ポリ(2‐メトキシエチルアクリレート);ポリ(2‐メタクリロイルオキシエチルホスホリルコリン);ポリ(テトラヒドロフルフリルアクリレート);ポリスチレン、又は第2の群に含まれる2つ若しくは3つのポリマー種を含むか又はそれらから本質的になるコポリマーを含む第2の群から、及び/又は
b.第2のポリマーは、前段落に記載の第1の群又は第2の群から選択されるが、第1のポリマーとは異なり、特に、第2のポリマーはポリ‐ヒドロキシブチレート、より特にはポリ‐3‐ヒドロキシブチレート及びポリ‐4‐ヒドロキシブチレートからなる群から選択される、
