Abstract
Description
本実施形態に係る美容健康用組成物が含有するケイ素の重量比を測定した。以下の表は、測定結果を示す。
本実施形態に係る美容健康用組成物の製造工程において、活性炭(または食用炭)によるフィルタリングによって、どの程度ケイ素が除去されてしまうかについて検証した。分析資料として、機能性ヤシ殻活性炭と、伊那赤松妙炭の2種類を用いた。使用機器は、ICP発光分析装置(島津製作所株式会社製ICPS−8100)とし、検証に用いるケイ素を、「和光純薬社製ICP用ケイ素標準液1000ppm」、「水溶性珪素umo超濃縮溶液」とした。試験方法は、以下の通りである。すなわち、ビーカーに水200mLを入れ、umoを20滴または40滴滴下する。次に、この溶液をろ紙にてろ過し、ろ過ブランクとする。次に、炭試料0.5gを添加し、スターラーにて3分間撹拌後、ろ紙でろ過する。次にICPにてケイ素濃度を測定した。測定結果は、以下の通りである。
検証例3では、X線回折分析により、結晶性の有無を検証した。試験日時は、「平成30年2月2日」であり、試験場所は、「滋賀県東北部工業技術センター」である。分析試料は、「(A)東海化学社製酸化ケイ素粉末、(B)umo粉末、(C)液体ケイ素(乾燥処理)、(D)液体ケイ素(半生状態)」の4種類である。ここで、「(A)東海化学社製酸化ケイ素粉末」とは、食品添加物であり、身体に吸収されない非晶質・微粒二酸化ケイ素(シリカ)である。「(B)umo粉末」とは、APAコーポレーション社の水溶性ケイ素粉末である。「(C)液体ケイ素(乾燥処理)」とは、本実施形態に係る水溶性ケイ素(メタケイ酸、またはオルトケイ酸などの含水シリカ・含水ケイ酸)の濃縮溶液のフリーズドライ粉末である。「(D)液体ケイ素(半生状態)」とは、本実施形態に係る水溶性ケイ素(メタケイ酸、またはオルトケイ酸などの含水シリカ・含水ケイ酸)の濃縮溶液の乾燥中の半生状態である。使用機器は、「株式会社リガク製 RINT2200V/PC」である。
また、以下の関係が見出される。

「メタケイ酸/ケイ素」=「78.09/28.09」=「2.78」倍

実施形態に係る美容健康用組成物は、「ハイドロキシアパタイト」と、クエン酸を含有する経口組成物(以下、「イオン化アパタイト」と呼称する。)と混合しても良い。水溶性ケイ素(水溶性イオン化ケイ素である場合も含む)とイオン化アパタイトとを混合することによって、サプリメントなどの健康 補助食品 への応用が可能となる。上述した通り、フラミンガム研究によれば、骨強化にはカルシウ ム摂取よりケイ素摂取が有効とされているため、水溶性ケイ素(水溶性イオン化ケイ素である場合も含む)とあわせて、イオ ン化アパタイトを摂取するによって、骨や歯が強化されて、骨粗しょう症や口腔ケア、ア ンチエイジングケアへの活用が期待できる。
本発明者らは、本実施形態に係るイオン化アパタイトが線虫の平均寿命に与える影響について着目し、第三者による試験結果に基づいて、検討を行なった。すなわち、抗酸化成分を含む食品を摂取することによって、老化に伴って起きる体内の活性酸素除去能力の低下を抑制することができ、老化による生活習慣病などの予防が期待できると言われているが、食品の機能性評価は主に「in vitro」で行なわれる。生体内での効果を実証するためには、「in vivo」の生物試験が必要となる。この「in vivo」試験においては、マウスなどの代わりとなる生物に、線虫の1種である「Caenorhabditis elegans」がある。この「C.elegans」は、寿命が3週間程度であり、約1mmの土壌に生息する非寄生性の線虫である。全ての遺伝子が解明されており、様々な生命現象を解析する実験のモデル生物として世界中で試験に用いられている。
本発明者らは、本実施形態に係るイオン化アパタイトが線虫の平均寿命に与える影響について着目し、第三者による試験結果に基づいて、検討を行なった。試験方法は、次の通りである。すなわち、「S-medium」に餌となる大腸菌OP−50株を懸濁させ、線虫(fer-15変異株)の培養液とした。この培養液を用いて線虫が成虫になるまで培養(20℃、100rpm)を行なった。その後、「S-buffer」にて線虫を回収、洗浄後、NaOH溶液およびハイター(登録商標)を用いて線虫の体を溶解させ、体内から卵を回収した。回収した卵は、20℃で1晩培養し、孵化させた。孵化させたL1幼虫は、24穴マイクロプレートを用い加熱処理大腸菌OP−50株(死菌体)を餌として、26.5℃で同調培養を行なった。このとき、マイクロプレートの1穴あたり線虫が約20匹となるように調整した。イオン化アパタイト(粉末)を100mg/mLの濃度で純水に溶解したものを原液とし、卵回収後、5日目に原液を0.22μmのフィルターでろ過減菌を行ない、培養液中の終濃度が、原液の10倍、100倍、1000倍希釈となるようにマイクロプレート各穴に加えた。その後、数日ごとに顕微鏡下で線虫の生存率を調べ、5日目の生存数を100%として、「Kaplan Meier法」により、生存率曲線を描き、ログランク検定によりp値を算出した。また、生存日数より平均寿命を算出した。結果は、以下の通りである。
本発明者らは、本実施形態に係るイオン化アパタイトのイオン化率を検証した。検体として、本実施形態に係るイオン化アパタイトとし、比較例1として、ホタテ貝殻由来ハイドロキシアパタイト、ホタテ貝殻由来ハイドロキシアパタイトを粉砕しナノパウダー化した分散液を検体とした。各検体を5gに水50mLを加え、30分間振とう後、0.45μmのメンブランフィルターでろ過した液について、リンとカルシウムの含有量をICP発光分析法にて測定した。ただし、検体重量から算出した。また、pHについては、1%の水溶液について、ガラス電極法によって測定した。測定結果を次の表に示す。
上記の検証の結果、本実施形態に係るイオン化アパタイトは、元素材である「ホタテ貝殻由来ハイドロキシアパタイト」や「ホタテ貝殻由来ハイドロキシアパタイトのナノ分散液」を大きく上回る数値が認められた。すなわち、イオン化アパタイトは、ハイドロキシアパタイトの配合率が、80%程度であるが、100%の元素材のホタテ貝殻由来ハイドロキシアパタイトを大幅に上回るイオン化率を有していることが分かった。さらに、イオン化アパタイトは、「ホタテ貝殻由来ハイドロキシアパタイトを、1kg当たり数万円をかけて粉砕し、<ナノレベル>にした分散液体」よりも大幅に高いイオン化率を有していることが分かった。
クルクミン(curcumin)は、ウコンなどに含まれる黄色のポリフェノール化合物であり、豆苗エキス(エンドウ芽エキス)は、アナゲイン(登録商標)という原料も販売され、エンドウ豆の新芽から抽出した水溶性エキスである。これらを本実施形態に係る水溶性ケイ素(水溶性イオン化ケイ素である場合も含む)に配合することによって、美髪効果、美肌効果、美爪効果が期待される。特に、上述したように「水溶性イオン化ケイ素」と組み合わせることによって、組織を強くする相乗効果が期待される。
Claims (4)
- 植物由来のケイ酸、富士・箱根・霧島・桜島地区の温泉・湧き水・溶岩・岩石由来のケイ酸の少なくとも一つを、活性炭または食用炭でろ過濃縮化する工程と、
前記濃縮化されたケイ酸または鉱物由来のケイ酸を、気圧が1気圧〜5気圧、温度が500℃〜2000℃の条件下で、海洋深層水を用いて溶解反応を生じさせる工程と、を少なくとも含むことを特徴とする水溶性ケイ素を含有する美容健康用組成物の製造方法。 - 前記美容健康用組成物は、重量比で5.7%〜13.8%のケイ素を含有することを特徴とする請求項1記載の水溶性ケイ素を含有する美容健康用組成物の製造方法。
- 水素による還元加工を行なった海洋深層水または活性炭フィルターによるイオン交換にてミネラルを濃縮させた海洋深層水と混合し、温度に加えて、圧力を活用して溶融反応を生じさせて水溶性ケイ素を均質化する工程をさらに含み、水溶性イオン化ケイ素を生成することを特徴とする請求項1または請求項2記載の水溶性ケイ素を含有する美容健康用組成物の製造方法。
- 植物由来のケイ酸、富士・箱根・霧島・桜島地区の温泉・湧き水・溶岩・岩石由来のケイ酸の少なくとも一つを、活性炭または食用炭を用いてろ過濃縮化を行なう工程と、
前記ろ過濃縮化されたケイ酸または鉱物由来のケイ酸を、所定の温度または所定の温度および圧力の条件下で、水素による還元加工を行なった海洋深層水または活性炭フィルターによるイオン交換にてミネラルを濃縮させた海洋深層水を用いて溶解反応を生じさせて水溶性ケイ素を生成しつつ、均質化する工程と、を少なくとも含むことを特徴とする水溶性イオン化ケイ素の調整方法。
