Abstract
Description

最初に、10%のウシ胎児血清(FBS)及び1×ペニシリンストレプトマイシンを補足したRPMI−1640からなる通常の増殖培地中で、MCF−7細胞をプレーティングした。細胞を、96ウェル細胞培養プレート中で1スキャフォールドあたり約7,000細胞でプレーティングした。新鮮培地を播種後2日目に添加し、播種後6日目にNucBlue(登録商標)色素を使用して腫瘍オルガノイドを可視化した。健康に見える腫瘍オルガノイドの存在を確認した後に、accutase:クエン酸塩溶液(1:1)を使用して、それらをスキャフォールドから剥がし、単一細胞懸濁液のためにプロセシングした。次いで単一細胞懸濁液を生存率についてカウントし、ヒト抗CD44−APC−cy7及び抗CD24−APCにより染色した。DAPIを使用して単一細胞の集団内の生細胞を弁別し、フローサイトメトリーを使用して生存細胞からCD44+CD24−細胞集団を決定した。図1A及び1B中で参照されるように、MCF−7細胞は、通常の増殖培地を使用して、FiSSCSC上で6日後に、良く発達した第一世代SCTを形成した。重要なことには、単層MCF−7細胞中のCSC集団のパーセンテージにかかわらず、CD44+CD24−細胞集団の増加によって決定されるように、第一世代腫瘍オルガノイドはそれらのCSC集団の約3倍の増加を一貫して示した。

低酸素がCSCの維持のために要求されるように思われるので、塩化コバルト(既知の低酸素の誘導因子)を使用して、本発明の3Dモデルにおいてこのことを試験することが所望された。このために、50μMの塩化コバルトを補足した通常の増殖培地中でMCF−7細胞をプレーティングした。以前のように、腫瘍オルガノイドを播種後6日目に可視化し、次いでヒト抗CD44−APC−cy7及び抗CD24−APCを使用して、フローサイトメトリーのためにプロセシングした。DAPIを使用して単一細胞の集団内の生細胞を弁別し、フローサイトメトリーを使用してCD44+CD24−細胞集団を決定した。結果から、塩化コバルトの添加が、第一世代MCF−7腫瘍オルガノイド中のCSCのパーセンテージを変化させなかったことが示された(図3)。このCSCの増幅の欠如は、細胞培養の継続期間を通して低酸素条件を維持するように塩化コバルトを外部から添加できないことに起因し得る。塩化コバルトの頻繁な補充が、低酸素の持続を確実にするのに必要であろう。
本発明者の初期の実験が、第一世代腫瘍オルガノイド中のCSCを増加させるために、塩化コバルトを外部から添加するできないことを示したので、スキャフォールドのマトリックス内に塩化コバルトを取り込むことにした。本発明者の仮定は、スキャフォールド内に包埋された塩化コバルトが、実験の継続期間を通しての低酸素の細胞培養環境の維持を支援するであろうということであった。この目的のために、100μMの塩化コバルトを専売のポリマーの混合物と混合し、もたらされたエレクトロスピンされたスキャフォールドを使用して、MCF−7−SCTの増殖に対する効果を試験した。塩化コバルト含有するスキャフォールド上に通常の増殖培地中のMCF−7細胞をプレーティングした。以前のように、腫瘍オルガノイドを播種後6日目及びフローサイトメトリーの遂行の前にNucBlue(登録商標)を使用して可視化し、最初にスキャフォールド内の塩化コバルトが低酸素条件を維持する能力を決定した。MCF−7 SCT中の低酸素領域を、低酸素(赤色)についての蛍光発生性プローブ(それは、低酸素細胞中に存在するニトロ還元酵素活性を利用することによって、ヒドロキシルアミン(NHOH)及びアミノ(NH2)へニトロ基を変換し、蛍光プローブを放出する)を使用して検出した。6日間の培養後に、通常のスキャフォールドではなく塩化コバルトスキャフォールド上で増殖させたMCF−7 SCTのみが、赤色蛍光を示し、塩化コバルト含有するスキャフォールドが低酸素を維持する能力を実証した。次いでヒト抗CD44−APC−cy7及び抗CD24−APCを使用して、フローサイトメトリーのために細胞をプロセシングした。DAPIを使用して単一細胞の集団内の生細胞を弁別し、フローサイトメトリーを使用してCD44+CD24−細胞集団を決定した。MCF−7 SCTはCSC集団の増加を示し、それは通常のスキャフォールド上で増殖させた第一世代MCF−7 SCTにおいて観察されたものよりもわずかに高かった(図3)。
本発明者は、第一世代及び第二世代を介して連続して培養した場合に、CSC集団(CD44+CD24−細胞として定義された)は腫瘍オルガノイド中で漸進的に増加したことを以前に示した。幹細胞性の複数のマーカーが報告されているので、FiSSCSCプラットフォームが、使用されるマーカーに依存してCSCの増加を示すかどうかを確認しようとした。転写因子のファミリーのOct-4、Sox−2、及びNanog(それらはいわゆるYamanaka転写因子の一部)を検討した。Oct-4、Sox−2、及びNanogは、CSCの多分化能及び自己更新の特徴の維持において重要な役割を果たす3つの基本的な転写因子である。以前に記載されるように、FiSSCSC上で第一世代MCF−7腫瘍オルガノイドを6日間培養し、その後、それらを収穫し、2つの群へと分割した。1群にトリゾール試薬を使用するRNA抽出を行い、第2の群をFiSSCSC上でさらに培養して第二世代腫瘍オルガノイドを形成させた。6日間の終了時に、第二世代腫瘍オルガノイドを収穫し、RNA抽出を行った。単層及び第二世代腫瘍オルガノイドから抽出されたRNAをプロセシングし、Oct-4及びSox−2、及びNanogについてのプローブを使用してqRT−PCRを行った。HPRTをハウスキーピング遺伝子として使用して、遺伝子発現を正規化した。結果から、単層と比較した場合に、第二世代におけるOct−4、Sox-2、及びNanogの発現の統計的に有意な増加が示されたことが示された(図5)。単層細胞に比べて、Sox−2は比較的緩やかな増加を示したが、Oct-4、及びNanogは転写物の3〜4倍の増加を示した。このことは、CSC増加(CD44+CD24−集団中の増加によって実証されるように)は、Oct−4、Sox−2、及びNanogの遺伝子発現の増加に相関したことを実証した。
本発明の実施形態は、CSCの収率の増加のために有用である。6ウェルFiSSCSCプレート上での腫瘍オルガノイドの増殖のための条件の特徴を評価した。このスケールアップは、96ウェルプレートに対して、細胞播種の約30倍の増加、及び実験の終了時のプロセシングされたCSCの当然の増加を導いた。したがって、例えば6ウェルプレート中で播種した細胞数を増加させた培養で、すべての試験された細胞の数は、6日目の終了時に良好に形成された腫瘍オルガノイドを与えることが見出された(図6A)。CD44+CD24−細胞集団を検討した場合に、CSC数の増加が見出されたが、これは実験間でわずかに変動した。具体的には、6ウェルフォーマットでは、96ウェルフォーマットに対して、高い数のCD44+CD24−細胞を与える細胞密度があることが見出された。この細胞数は、6ウェルプレート中でウェルあたり約240,000〜約270,000細胞の間であった(図6B)。この範囲よりも低いかまたは高い細胞の数は、CD44+CD24−細胞集団の減少をもたらした。細胞の生存率は、96ウェルフォーマットにおいて得られたものに匹敵した(約80%)。したがって、本発明の一実施形態において、約240,000細胞/6ウェルをFiSSCSC上で腫瘍オルガノイドの増殖のために使用した。
CSC維持が腫瘍微小環境内で存在する細胞性及び非細胞性の構成要素からの安定した合図を要求することが報告された。この現象内で、役割を有することが示されたものとしてはCAFが挙げられる。CAFからの分泌因子がCSCの集団を増大できるかどうかを査定するために、乳癌患者からCAFを収集及び培養した。具体的には、CAFを約80%コンフルエンスまで培養し、次いでそれらを増殖培地中で48時間インキュベーションし、その終了時に培地を収集し、遠心分離し、使用するまで−80℃で保管した。使用の前に、培地を氷上で融解し、試験のために適切な希釈物をMCF−7増殖培地中で作製した。
細胞−細胞相互作用(特にがん細胞とMDSCのような免疫細胞との間の)は、CSCの誘導及び維持をインビボで促進することが示されている。共培養の効果を試験するために、正常で健康な全血液を獲得し、それを単核細胞についてプロセシングした。次いで単核細胞をHeLa細胞と共に培養して(1:100の比)、単核細胞のMDSCへの分化を支援した。6日間の培養後に、CD33+細胞を単離し、フローサイトメトリーを使用してMDSC細胞表面マーカーについての特徴を評価した。
単層細胞(0.3%)に比較して、FiSSプラットフォーム上でのLLC1の培養が少なくとも30倍のCSC活性(アルデヒド脱水素酵素(ALDH)活性に基づいて同定された)の増加を誘導することが見出された(図9A)。これらの腫瘍オルガノイドはALDH−Hi集団を示し、単層上で増殖させた細胞に比較して、CSC様集団を提示する。選別されたALDH陽性集団は、選別されないLLC1腫瘍オルガノイドよりもC57BL/6マウス中で良好な腫瘍増殖を開始することができ(図9B)、ALDH陽性集団がCSC様細胞を保持することを示唆する。加えて、FiSS上でこれらの腫瘍オルガノイドを継承して継代することは、第三世代におけるALDH陽性細胞を87.5%へエンリッチし、大多数の幹細胞性遺伝子は増大させた集団中で保存された。インビボの腫瘍から稀なCSC集団を単離及び増大する可能性を検討するために、C57BL/6マウス中にインプラントしたLLC1腫瘍を切除した。LLC1腫瘍の単一細胞懸濁液をFiSS上で6日間培養した場合に、ALDH陽性集団の10倍の増大が見出された(図9C)。
次にインビボの腫瘍から稀なCSC集団を単離及び増大する可能性を検討した。インビボの腫瘍から稀なCSC集団を単離及び増大する可能性を検討するために、C57BL/6マウス中にインプラントしたLLC1腫瘍を切除した。LLC1腫瘍の単一細胞懸濁液をFiSS上で6日間培養した場合に、ALDH陽性集団の10倍の増大が見出された。同様に、A549異種移植物をNSGマウス中にインプラントし、これらの腫瘍の単一細胞懸濁液を6日間FiSS上で培養した。CD44+CD24−集団(幹様細胞を示す)の5〜9倍の増加が、A549異種移植物(6.84%)に比較して、FiSS上で培養したA549異種移植物細胞に由来する腫瘍オルガノイド(50.9%)中で見出された(図10A)。さらに、NSGマウス中での少なくとも1,000のCD44+CD24−集団の注射は、インビボで腫瘍を開始することができ、CD44+CD24−細胞が本当にA549中でCSC様細胞を示すことを示唆した。したがって、20,000のCD44+CD24−細胞のみの注射は、3×106細胞の一次注射に比較して、同じサイズの腫瘍を誘導したという証拠は、本発明者が開発したCSC増大プロトコールが腫瘍開始細胞をエンリッチすることを指摘する(図10B)。
増大させたCSCを適切に保管きるかどうかを検討するために、マイクロビーズを使用し、CD44+細胞をプロセシングし、次いで一例としてCryostor(登録商標)培地中でこれらの細胞を冷凍した。エンリッチメントの前に、単層培養した細胞中で、細胞の約19%はCD44+であったが、スキャフォールド上で増殖された細胞の約26%はCD44+であった。さらに、腫瘍オルガノイド細胞の涸渇後に、約67%はCD44+細胞であった。細胞を冷凍及び融解した後に、細胞の約55%はCD44+であった(図11)。
Claims (48)
- a)腫瘍オルガノイドを、インビトロの細胞培養における三次元スキャフォールド上で増殖させることと;
b)前記腫瘍オルガノイドからCSCを単離することと、
を含む、がん幹細胞(CSC)を増大する方法。 - a)三次元スキャフォールドを含むインビトロの細胞培養において、がん細胞を増殖させることと;
b)前記がん細胞から腫瘍オルガノイドを前記スキャフォールド上で増殖させることと;
c)前記腫瘍オルガノイドからがん細胞(腫瘍オルガノイドがん細胞)を収穫することと;
d)前記腫瘍オルガノイドがん細胞を、三次元スキャフォールドを含む新しいインビトロの細胞培養へ移すことと;
e)前記腫瘍オルガノイドがん細胞から、腫瘍オルガノイドの後続する世代を、前記新しいインビトロの細胞培養の前記スキャフォールド上で増殖させることと、
を含む、がん幹細胞(CSC)を増大する方法。 - ステップc)〜e)が少なくとも1回反復される、請求項2に記載の方法。

- 前記の前記腫瘍オルガノイドからCSCを単離することが、前記腫瘍オルガノイドから腫瘍オルガノイドがん細胞の単一細胞懸濁液を形成することと;前記腫瘍オルガノイドがん細胞の前記単一細胞懸濁液からCSCを単離することと、を含む、請求項1または5〜7のいずれか一項に記載の方法。
