Abstract
Description
本発明は、とりわけ、抗AXL抗体に基づく抗体-薬物コンジュゲート(ADC)、およびがんの治療におけるそれらの使用に関する。特に、本発明は、1週用量約0.45mg/kg〜約2.0mg/kgの前記ADCを対象者に投与することを含む、該ADCの投薬レジメンに関する。
AXLは、哺乳動物の受容体型チロシンキナーゼ(RTK)のTAMサブファミリーに属する104〜140kDaの膜貫通タンパク質である。AXLは、複数の細胞型において増殖、遊走、凝集および抗炎症を含むいくつかの細胞機能に関与すると考えられており、正常細胞では弱く発現しており、主に線維芽細胞、骨髄前駆細胞、マクロファージ、神経組織、心筋および骨格筋において観察される。AXLは、そのリガンドであるビタミンK依存性のGrowth arrest-specific factor 6 (Gas6)の結合時に活性化されて、AXL二量体化、自己リン酸化およびその後の細胞内シグナル伝達経路の活性化をもたらす。
本発明の目的は、そのようながんを治療するための方法を提供することである。本発明のさらなる目的は、そのようながんを治療する方法における使用のための抗AXL抗体に基づくADCの投薬レジメンを提供することである。本発明のさらなる目的は、特定の抗AXL抗体に基づくADCのための安全で有効な新しい投薬レジメンを提供することである。

本明細書において使用される「AXL」または「Axl」という用語は、哺乳動物TAM受容体チロシンキナーゼ(RTK)のサブファミリーの一部である104〜140 kDaの分子量を有する894アミノ酸タンパク質であり、UFOまたはJTK11とも呼ばれる、AXLという名称のタンパク質を指す。分子量は、タンパク質のグリコシル化における可能性のある差のために可変である。AXLタンパク質は、タンパク質のN末端上の2つの細胞外免疫グロブリン様(Ig様)ドメイン、2つの膜近位の細胞外フィブロネクチンIII型(FNIII)ドメイン、膜貫通ドメイン、および細胞内キナーゼドメインからなる。AXLは、そのリガンドGas6の結合時に、AXL細胞外ドメイン間のリガンド非依存性同種親和性相互作用により、活性酸素種の存在下での自己リン酸化により(Korshunov et al., 2012)、またはEGFRを通したトランス活性化により(Meyer et al., 2013)活性化され、いくつかの腫瘍タイプにおいて異常に発現している。ヒトにおいて、AXLタンパク質は、SEQ ID N0:130に示されるアミノ酸配列をコードする核酸配列によってコードされている(ヒトAXLタンパク質:Swissprot P30530)。カニクイザルAXLタンパク質についてはGenbankアクセッションHB387229.1(SEQ ID N0:147)を参照されたい。

本発明の好ましい態様において、本発明の使用のためのAXL-ADCは、1週用量の単回投与が連続3週間、28日間のサイクルで実施される。いくつかの態様では、その用量を、単回の週用量として、28日間の治療サイクルの1日目、8日目、および15日目に投与する。
当業者、例えば医師は、適切な回数の治療サイクルの後に、AXL-ADCによる維持療法と、必要に応じて別の治療剤または治療剤の組み合わせによる治療を、治療サイクルに続いて行うべきであると判断するかもしれない。
したがって、本発明は、4週間治療サイクルで多数のサイクルにわたって、対象者にAXL-ADCを単回の週用量として連続3週間投与し、その後に1週間の休薬期間を設け、必要に応じて引き続き、3週間サイクルで多数のサイクルにわたって、該対象者にAXL-ADCを3週間に1回投与する維持療法を行う実施態様を包含する。
- AXLを発現するがんと診断された対象者;
- AXLを発現するがんがあると疑われる対象者;または
- 特定の治療剤に対して耐性であるか、耐性になる傾向が高いがんと診断された対象者。
本発明の別の局面において、本発明のいずれかの局面または態様に従って使用するための本明細書に記載のAXL-ADCは、薬学的組成物中に含まれる。一態様では、薬学的組成物は薬学的に許容される担体をさらに含む。特に、AXL-ADCの精製後に、それらは周知の医薬用担体または賦形剤を用いて薬学的組成物に製剤化することができる。

この実施例では、ヒトの血漿中の遊離HuMax-AXL-ADC (IgG1-1021-107-MMAE)および遊離MMAEの曝露を、以下の2つの投薬レジメンについて評価する:
- 1Q3W:3週間に1回投与(0.3、0.6、1、1.5、2、2.4mg/kg);
- 3Q4W:週1回、3週間投与、その後に無治療の1週間(0.6、0.8、1、1.2、1.4mg/kg)。
以下の表に示される、5つのインビボ前臨床試験からのデータが提供され、該データは血漿ADCレベル、血漿IgGレベル、および血漿MMAEレベル(試験5でのみ測定)の測定値を含んでいた。
個々のPKパラメータの評価は、NONMEMバージョン7.3 (Beal et al., NONMEM User's Guides. (1989-2009), Icon Development Solutions, Ellicott City, MD, USA, 2009)を用いて行った。曝露パラメータの評価ならびに他の全てのデータの準備、解析および可視化は、Rバージョン3.3.1 (Development Core Team (2008). R: A language and environment for statistical computing. R Foundation for Statistical Computing, Vienna, Austria. ISBN 3-900051-07-0)を用いて行った。
カニクイザルにおけるADC/IgG popPKモデル:
ADCおよびIgGアッセイによって定量されるADC曝露をモデル化するために、いくつかの構造モデルを試験した。標的介在性クリアランスと非標的介在性クリアランスの両方を示す2コンパートメントモデル(図1)を選択したが、それは、該モデルがより単純なモデルと比べて明らかな改善を示したためである。選択したモデルは、モノクローナル抗体への曝露を説明するために一般的に使用されるモデル構造を示し、それは、例えば、Gibianskyら(J Pharmacokinet Pharmacodyn 2008;35(5):573-91)に示されている。全ての速度および体積パラメータのアロメトリックスケーリングを前臨床モデルに含めた。
2コンパートメントモデルは、試験5からのMMAE曝露データをモデル化するのに最適であるとわかった。体重によるアロメトリックスケーリングを全ての関連パラメータに含めた。Luら(Pharm Res 2015;32:1907-19)に従って、血漿中の遊離MMAEの吸収をADCのクリアランスに比例するようにモデル化した。
1Q3Wおよび3Q4W投薬レジメンのシミュレーションを行った。その結果を以下の表および図4〜14に示す。
- 1Q3W:3週間に1回投与。0.3、0.6、1、1.5、2、2.4mg/kg;
- 3Q4W:週1回、3週間投与、その後に無治療の1週間。0.6、0.8、1、1.2、1.4mg/kg。
カニクイザルで得られた前臨床PKデータを説明することができる、線形非標的介在性クリアランスおよび非線形標的介在性クリアランスによる2コンパートメント母集団PKモデルが作成された。
抗AXL抗体へのMMAEのコンジュゲーション
抗AXL抗体を、標準的な手順にしたがってプロテインAクロマトグラフィーにより精製し、vcMMAEにコンジュゲートした。文献に記載されている手順にしたがって(Sun et al., 2005、McDonagh et al., 2006、および Alley et al., 2008 を参照されたい)、薬物-リンカーvcMMAEを、還元された抗体のシステインにアルキル化した。過剰量のN-アセチルシステインの添加によって、反応を終息させた。いずれの残りのコンジュゲートされていない薬物も、精製によって除去し、最終的な抗AXL抗体薬物コンジュゲートを、PBSにおいて製剤化した。抗AXL抗体薬物コンジュゲートを、その後、濃度(280 nmでの吸光度により)、逆相クロマトグラフィー(RP-HPLC)および疎水性相互作用クロマトグラフィー(HIC)により薬物対抗体の比(DAR)、コンジュゲートされていない薬物の量(逆相クロマトグラフィーにより)、凝集のパーセンテージ(サイズ排除クロマトグラフィー、SEC-HPLCにより)、ならびにエンドトキシンレベル(LALにより)について解析した。結果を、下記の表8に示す。
LCLC-103H細胞(ヒト大細胞肺癌)およびA431細胞(DMSZ, Braunschweig, ドイツ)を、10%(体積/体積)熱不活化Cosmic Calf Serum(Perbio;カタログ番号SH30087.03)、2 mM L-グルタミン(Cambrex;カタログ番号US17-905C)、50 IU/mLペニシリン、および50μg/mLストレプトマイシン(Cambrex;カタログ番号DE17-603E)を補給した、L-グルタミンを含むRPMI1640(Cambrex;カタログ番号BE12-115F)中で培養した。MDA-MB231細胞を、高グルコースおよびHEPESを含むDMEM(Lonza #BE12-709F)、Donor Bovine Serum with Iron(Life Technologies #10371-029)、2 mM L-グルタミン(Lonza #BE17-605E)、1 mMピルビン酸ナトリウム(Lonza #BE13-115E)、およびMEM Non-Essential Amino Acids Solution(Life Technologies #11140)中で培養した。細胞株は、37℃で、5%(体積/体積)CO2加湿インキュベーターにおいて維持した。LCLC-103H、A431、およびMDA-MB231細胞を、コンフルエント近くまで培養し、その後、細胞をトリプシン処理して、培養培地中に再懸濁し、セルストレーナー(BD Falcon、カタログ番号352340)を通過させて、単一細胞懸濁液を取得した。1×103細胞を、96ウェル培養プレートの各ウェルに播種し、細胞を、室温で30分間、その後37℃、5% CO2で5時間インキュベートして、プレートに接着させた。
MMAEコンジュゲートAXL抗体の連続希釈液(0.00015〜10μg/mLの範囲にわたる最終濃度)を培養培地中で調製し、プレートに添加した。細胞と1μMスタウロスポリン(#S6942-200, Sigma)とのインキュベーションを、100%腫瘍細胞殺傷用の参照として使用した。無処置の細胞を、100%細胞成長用の参照として使用した。プレートを、37℃、5% CO2で5日間インキュベートした。次に、CellTiter-Glo Reagent(Promega;カタログ番号G7571)をウェルに添加して(ウェルあたり20μL)、プレートを37℃、5% CO2で1.5時間インキュベートした。その後、ウェルあたり180μLを、白色96ウェルOptiplate(商標)プレート(PerkinElmer, Waltham, MA;カタログ番号6005299)に移して、室温で30分間インキュベートした。最後に、発光を、EnVisionマルチプレートリーダー(Envision, Perkin Elmer)上で測定した。
MMAEコンジュゲート抗AXL抗体のインビボ効力を、確立されたSCIDマウスにおける皮下(SC)LCLC-103H異種移植腫瘍において測定した。200μL PBS中の5×106個のLCLC-103H(大細胞肺癌)腫瘍細胞(Leibniz-Institut DSMZ-Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH (DSMZ)から取得)を、雌SCIDマウスの右側腹部の皮下に注射した。平均腫瘍サイズが100〜200 mm3よりも大きく、明瞭な腫瘍成長が観察された、腫瘍細胞接種の14〜21日後に開始して、1 mg/kg(20μg/マウス)のIgG1-AXL-vcMMAE抗体または対照(コンジュゲートされていないIgG1-b12)での単回注射を、腹腔内に与えた(100μL/マウス)。腫瘍体積を、少なくとも週あたり2回測定した。腫瘍体積(mm3)を、キャリパー(PLEXX)測定値から0.52×(長さ)×(幅)2として算出した。
IgG1-AXL-107-vcMMAE、IgG1-AXL-148-vcMMAE、およびIgG1-AXL-733-vcMMAEの抗腫瘍活性を、PAXF1657膵臓癌PDXモデルにおいて測定した(Oncotest, Freiburg, ドイツにより行われた実験)。ヒト膵臓腫瘍組織を、5〜7週齢の雌NMRI nu/nuマウスの左側腹部の皮下に移植した。動物の無作為化を、以下のように行った:50〜250 mm3、好ましくは80〜200 mm3の間の体積の腫瘍を有する動物を、群腫瘍体積の比較可能な中央値および平均値を考慮して、7種の実験群(群あたり8匹の動物)に分配した。無作為化の日(0日目)に、3種のADCを、4 mg/kgまたは2 mg/kgのいずれかで静脈内に(i.v.)投与し、対照群は、単一用量のIgG1-b12(4 mg/kg)を受けた。腫瘍体積(mm3)を、週に2回モニターして、キャリパー(PLEXX)測定値から0.52×(長さ)×(幅)2として算出した。
A431およびLCLC-103H腫瘍細胞のGas6産生
A431細胞およびLCLC-103H細胞がGas6を産生するかどうかを試験した。そのために、細胞を、完全培養培地中で3日間成長させた。上清中のGas6レベルを、Quantikine Human Gas6 ELISA(R&D Systems, Minneapolis, MN)を用い、製造業者の説明書にしたがって決定した。このアッセイは、定量的サンドイッチELISA技術を使用する。ヒトGas6に特異的なモノクローナルAbが、マイクロプレート上にプレコーティングされている。標準品および試料をウェル中にピペットで移すと、存在する任意のヒトGas6が、固定されたAbによって結合される。任意の結合していない物質を洗浄して除去した後、ヒトGas6に特異的な酵素連結ポリクローナルAbをウェルに添加する。任意の結合していないAb-酵素試薬を除去するための洗浄後に、基質をウェルに添加すると、最初の工程において結合したヒトGas6の量に比例して色が顕出する。顕色を停止して、色の強度を測定する。
いくつかのAXL-ADC(IgG1-AXL-061-vcMMAE(Ig1結合剤)、IgG1-AXL-107-vcMMAE(Ig1結合剤)、IgG1-AXL-137-vcMMAE(Ig1結合剤)、IgG1-AXL-148-vcMMAE(Ig2結合剤)、IgG1-AXL-183-vcMMAE(FN1結合剤)、およびIgG1-AXL-726-vcMMAE(FN2結合剤))のインビボ抗腫瘍活性を、高レベルのGas6を産生するA431(類表皮癌)腫瘍モデル、および、低レベルのGas6を産生するLCLC-103H(大細胞肺癌)腫瘍モデルにおいて測定した。IgG1-AXL-107は結合に関してGas6と競合しないが、IgG1-AXL-061およびIgG1-AXL-137はAXLとの結合に関してGas6と競合する。
AXLの発現を、甲状腺癌、食道癌、卵巣癌、膵臓癌、肺癌、乳癌、子宮頸癌もしくは子宮内膜癌、または悪性黒色腫を有する患者由来の組織コアを含む、新しく切ったパラフィン包埋、ホルマリン固定(FFPE)の腫瘍組織マイクロアレイ(TMA)において評定した。TMAは、US BioMaxから取得した。
IgG1-AXL-107-vcMMAE(本明細書において「HuMax-AXL-ADC」とも称される)の抗腫瘍活性を、BALB/cヌードマウスにおける皮下食道PDXモデルES0195において評定した(Crown Bioscience. Taicang Jiangsu Province, 中国により行われた実験)。患者由来の食道異種移植片を有するドナーマウスからの腫瘍断片(ES0195)を、BALB/cヌードマウス中への接種のために使用した。各マウスに、1つの腫瘍断片(直径2〜3 mm)を右側腹部の皮下に接種して、腫瘍体積が約150 mm3になるまで腫瘍を成長させた。動物の無作為化を、以下のように行った:約150 mm3の体積の腫瘍を有する動物を、群腫瘍体積の比較可能な中央値および平均値を考慮して、5種の実験群(群あたり8匹の動物)に分配した。処置群は、IgG1-b12、IgG1-b12-vcMMAE、IgG1-AXL-107、IgG1-AXL-107-vcMMAE、およびパクリタキセルであった。抗体およびADCを、無作為化の日(0日目)および7日目に、4 mg/kgで静脈内に(i.v.)投与した。パクリタキセルは、0、7、および14日目に、20 mg/kgで腹腔内に(i.p.)投与した。腫瘍体積(mm3)を、週に2回モニターして、キャリパー(PLEXX)測定値から0.52×(長さ)×(幅)2として算出した。
IgG1-AXL-183-vcMMAEおよびIgG1-AXL-726-vcMMAEの抗腫瘍活性を、NMRI nu/nuマウス(Harlan, オランダ)における患者由来の子宮頸癌異種移植CEXF 773モデルにおいて評定した。実験は、Oncotest(Freiburg, ドイツ)により行われた。
IgG1-AXL-183-vcMMAEおよびIgG1-AXL-726-vcMMAEの抗腫瘍活性を、同所MDA-MB-231 D3H2LN異種移植モデルにおいて評定した。
LU2511 PDXモデル
IgG1-AXL-107-vcMMAEの抗腫瘍活性を、BALB/cヌードマウスでの皮下エルロチニブ耐性NSCLC PDXモデルLU2511において評価した(Crown Bioscience社(Changping District, 北京, 中国)で実施した実験)。患者由来のNSCLC異種移植片(LU2511)を有するドナーマウスからの腫瘍断片を、BALB/cヌードマウスへの接種のために使用した。各マウスの右脇腹に1つの腫瘍断片(直径2〜3mm)を皮下接種し、腫瘍体積が約200mm3になるまで腫瘍を増殖させた。動物の無作為化を次のように行った:約200mm3の体積の腫瘍がある動物を、群の腫瘍体積の同等の中央値および平均値を考慮して、5つの実験群(1群8匹)に割り振った。処置群は次の通りであった:IgG1-b12、IgG1-b12-vcMMAE、IgG1-AXL-107-vcMMAE、エルロチニブ、およびエルロチニブ+IgG1-AXL-107-vcMMAE。抗体およびADCを、無作為化の日(0日目)と7日目に4mg/kgで静脈内(i.v.)投与した。エルロチニブは50mg/kg/日で2週間経口(per os)投与した。腫瘍体積(mm3)は、週に2回モニターして、キャリパー(PLEXX)測定値から0.5×(長さ)×(幅)2として算出した。

IgG1-AXL-107-vcMMAEの抗腫瘍活性を、皮下エルロチニブ耐性NSCLC PDXモデルLXFA 526において評価した(Oncotest社(Freiburg, ドイツ)で実施した実験)。4〜6週齢の雄NMRI nu/nuマウスへの腫瘍断片の接種および無作為化を上記のように実施した。
IgG1-AXL-107-vcMMAEの抗腫瘍活性を、皮下エルロチニブ耐性NSCLC PDXモデルLXFA 677およびLXFA 677_3モデル(LXFA 677モデルに由来し、エルロチニブに対する獲得耐性を有する)において評価した(Oncotest社(Freiburg, ドイツ)で実施した実験)。4〜6週齢の雄NMRI nu/nuマウスへの腫瘍断片の接種および無作為化を上記のように実施した。
IgG1-AXL-107-vcMMAEの抗腫瘍活性を、BALB/cヌードマウスの皮下子宮頸癌PDXモデルCV1664において評価した(CrownBioscience社(Changping District, 北京, 中国)で実施した実験)。BALB/cヌードマウスへの腫瘍断片の接種および無作為化は、実施例23に記載したように行った。
Claims (54)
- がんを治療する方法における使用のための、ヒトAXLに結合する抗体を含む抗体薬物コンジュゲート(ADC)であって、該方法が、それを必要とする対象者に少なくとも1サイクルで該ADCを投与することを含み、該サイクルが、連続3週間にわたる週1回の投与と、その後のADCを投与しない1週間の休薬期間とを含み、各サイクルの期間が該休薬期間を含めて28日間であり、
該抗体が、アウリスタチンまたはその機能性ペプチド類似体もしくは誘導体にリンカーを介してコンジュゲートされており、かつ
(a) それぞれSEQ ID No: 36、37および38のCDR1、CDR2およびCDR3配列を含むVH領域;ならびにそれぞれSEQ ID No: 39、GASおよび40のCDR1、CDR2およびCDR3配列を含むVL領域 [107];
(b) それぞれSEQ ID No: 46、47および48のCDR1、CDR2およびCDR3配列を含むVH領域;ならびにそれぞれSEQ ID No: 49、AASおよび50のCDR1、CDR2およびCDR3配列を含むVL領域 [148];
(c) それぞれSEQ ID No: 114、115および116のCDR1、CDR2およびCDR3配列を含むVH領域;ならびにそれぞれSEQ ID No: 117、DASおよび118のCDR1、CDR2およびCDR3配列を含むVL領域 [733];ならびに
(d) (a)〜(c)において定義される前記抗体のうちのいずれかのバリアントであって、6つのCDR配列全体にわたって、好ましくは最大で1、2、または3個のアミノ酸改変、より好ましくはアミノ酸置換、例えば保存的アミノ酸置換を有する、バリアント
からなる群より選択される重鎖可変(VH)領域および軽鎖可変(VL)領域を含む、前記使用のためのADC。 - 前記抗体が、
(a) それぞれSEQ ID No: 36、37および38のCDR1、CDR2およびCDR3配列を含むVH領域;ならびにそれぞれSEQ ID No: 39、GASおよび40のCDR1、CDR2およびCDR3配列を含むVL領域 [107];
(b) それぞれSEQ ID No: 46、47および48のCDR1、CDR2およびCDR3配列を含むVH領域;ならびにそれぞれSEQ ID No: 49、AASおよび50のCDR1、CDR2およびCDR3配列を含むVL領域 [148];

- 前記ADCの用量が、前記対象者の体重1kgあたり0.45mg〜2.0mgであり、例えば、0.45mg/kgの用量、0.5mg/kgの用量、0.6mg/kgの用量、0.7mg/kgの用量、0.8mg/kgの用量、0.9mg/kgの用量、1.0mg/kgの用量、1.1mg/kgの用量、1.2mg/kgの用量、1.3mg/kgの用量、1.4mg/kgの用量、1.5mg/kgの用量、1.6mg/kgの用量、1.7mg/kgの用量、1.8mg/kgの用量、1.9mg/kgの用量、または2.0mg/kgの用量である、前記請求項のいずれか一項に記載の使用のためのADC。
- 前記28日間のサイクルの数が、2〜48、例えば2〜36、例えば2〜24、例えば2〜15、例えば2〜12、例えば、2サイクル、3サイクル、4サイクル、5サイクル、6サイクル、7サイクル、8サイクル、9サイクル、10サイクル、11サイクル、または12サイクルである、前記請求項のいずれか一項に記載の使用のためのADC。
- 前記ADCが28日間治療サイクルで少なくとも4サイクルにわたって投与され、その際、各治療サイクルにおいて該ADCが0.45mg/kg体重の用量で週1回、連続して3週間投与され、その後に前記抗体薬物コンジュゲートが投与されない休薬週が続く、前記請求項のいずれか一項に記載の使用のためのADC。
- 前記ADCが28日間治療サイクルで少なくとも4サイクルにわたって投与され、その際、各治療サイクルにおいて該ADCが0.6mg/kg体重の用量で週1回、連続して3週間投与され、その後に前記抗体薬物コンジュゲートが投与されない休薬週が続く、請求項1〜17のいずれか一項に記載の使用のためのADC。
- 前記ADCが28日間治療サイクルで少なくとも4サイクルにわたって投与され、その際、各治療サイクルにおいて該ADCが0.8mg/kg体重の用量で週1回、連続して3週間投与され、その後に前記抗体薬物コンジュゲートが投与されない休薬週が続く、請求項1〜17のいずれか一項に記載の使用のためのADC。
- 前記ADCが28日間治療サイクルで少なくとも4サイクルにわたって投与され、その際、各治療サイクルにおいて該ADCが1.0mg/kg体重の用量で週1回、連続して3週間投与され、その後に前記抗体薬物コンジュゲートが投与されない休薬週が続く、請求項1〜17のいずれか一項に記載の使用のためのADC。
- 前記ADCが28日間治療サイクルで少なくとも4サイクルにわたって投与され、その際、各治療サイクルにおいて該ADCが1.2mg/kg体重の用量で週1回、連続して3週間投与され、その後に前記抗体薬物コンジュゲートが投与されない休薬週が続く、請求項1〜17のいずれか一項に記載の使用のためのADC。
- 前記ADCが28日間治療サイクルで少なくとも4サイクルにわたって投与され、その際、各治療サイクルにおいて該ADCが1.4mg/kg体重の用量で週1回、連続して3週間投与され、その後に前記抗体薬物コンジュゲートが投与されない休薬週が続く、請求項1〜17のいずれか一項に記載の使用のためのADC。
- 前記ADCが28日間治療サイクルで少なくとも4サイクルにわたって投与され、その際、各治療サイクルにおいて該ADCが1.6mg/kg体重の用量で週1回、連続して3週間投与され、その後に前記抗体薬物コンジュゲートが投与されない休薬週が続く、請求項1〜17のいずれか一項に記載の使用のためのADC。
- 前記ADCが28日間治療サイクルで少なくとも4サイクルにわたって投与され、その際、各治療サイクルにおいて該ADCが1.8mg/kg体重の用量で週1回、連続して3週間投与され、その後に前記抗体薬物コンジュゲートが投与されない休薬週が続く、請求項1〜17のいずれか一項に記載の使用のためのADC。
- 前記ADCが28日間治療サイクルで少なくとも4サイクルにわたって投与され、その際、各治療サイクルにおいて該ADCが2.0mg/kg体重の用量で週1回、連続して3週間投与され、その後に前記抗体薬物コンジュゲートが投与されない休薬週が続く、請求項1〜17のいずれか一項に記載の使用のためのADC。
- 前記治療サイクルの後に維持療法が実施される、前記請求項のいずれか一項に記載の使用のためのADC。
- 前記維持療法が、前記ADCを3週間に1回、例えば21日間サイクルの1日目に、投与することを含む、請求項27に記載の使用のためのADC。
- 前記維持療法のためのADCの用量が、前記対象者の体重1kgあたり0.6mg〜3.2mgであり、例えば、約0.6mg/kgの用量、約0.8mg/kgの用量、約1.0mg/kgの用量、約1.2mg/kgの用量、約1.4mg/kgの用量、約1.6mg/kgの用量、約1.8mg/kgの用量、約2.0mg/kgの用量、約2.2mg/kgの用量、約2.4mg/kgの用量、約2.6mg/kgの用量、約2.8mg/kgの用量、約3.0mg/kgの用量、または約3.2mg/kgの用量である、請求項27および28のいずれか一項に記載の使用のためのADC。
- 前記維持療法が21日間のサイクルで実施され、該サイクルの数が、2〜48、例えば2〜36、例えば2〜24、例えば2〜15、例えば2〜12、例えば、2サイクル、3サイクル、4サイクル、5サイクル、6サイクル、7サイクル、8サイクル、9サイクル、10サイクル、11サイクル、または12サイクルである、請求項27〜29のいずれか一項に記載の使用のためのADC。
- 前記がんが、AXLを発現する固形腫瘍を含むか、またはAXLを発現する血液がんである、前記請求項のいずれか一項に記載の使用のためのADC。
- 前記AXLを発現する血液がんが、白血病、例えば、慢性リンパ性白血病、骨髄性白血病、急性骨髄性白血病(AML)、および慢性骨髄性白血病;リンパ腫、例えば非ホジキンリンパ腫および多発性骨髄腫からなる群より選択される、請求項31に記載の使用のためのADC。
- 前記がんが、AXLを発現する固形腫瘍を含み、かつ肺癌、例えば、非小細胞肺癌(NSCLC)および肺扁平上皮癌;婦人科癌、例えば、卵巣癌、子宮内膜癌または子宮頸癌;甲状腺癌;皮膚癌、例えば、黒色腫;大腸癌、例えば、大腸癌腫および大腸腺癌;膀胱癌;骨がん、例えば軟骨肉腫;乳癌、例えばトリプルネガティブ乳癌;中枢神経系の癌、例えば、膠芽腫、星状細胞腫および神経芽細胞腫;結合組織癌;線維芽細胞癌;胃癌、例えば胃癌腫;頭頸部癌;腎臓癌;肝臓癌、例えば肝細胞癌;筋肉癌;神経組織癌;膵臓癌、例えば、膵管癌および膵腺癌;ならびに軟部組織肉腫からなる群より選択される、請求項31に記載の使用のためのADC。
- 前記がんが、非小細胞肺癌(NSCLC)、卵巣癌、子宮内膜癌、子宮頸癌、甲状腺癌からなる群より選択される、請求項33に記載の使用のためのADC。
- 前記がんが、チロシンキナーゼ阻害剤、セリン/トレオニンキナーゼ阻害剤および化学療法剤からなる群より選択される少なくとも1つの治療剤に対して耐性である、請求項31に記載の使用のためのADC。
- 前記チロシンキナーゼ阻害剤がEGFR阻害剤であり、前記セリン/トレオニンキナーゼ阻害剤がBRAF阻害剤およびMEK阻害剤からなる群より選択される、請求項35に記載の使用のためのADC。
- 前記がんが、NSCLC、卵巣癌、子宮頸癌、黒色腫、頭頸部扁平上皮癌(SCCHN)、乳癌、消化管間質腫瘍(GIST)、腎臓癌、前立腺癌、神経芽細胞腫、膵臓癌、食道癌、横紋筋肉腫、急性骨髄性白血病(AML)、または慢性骨髄性白血病(CML)から選択される、請求項35および36のいずれか一項に記載の使用のためのADC。
